#Herbal #Ashwagandha Inhibits #ProstateCancer through Immuno-modulation and Cell Cycle Arrest In Vitro

Kyle J. Norton

Cancer is a group of chronic diseases caused by irregular cell growth in the specific tissue of an organ due to DNA alternation.
Most cases of cancer start in the cell on the surface of the inner lining tissue before penetrating into deeper layers to form a tumor.

However, as cancer continued proliferation, at certain stages it can damage other healthy tissues. Sometimes cancer cells can spread to other organs in the body via lymph or blood.

Prostate cancer is a medical condition caused by the alternation of the cell on the surface of the inner lining of the prostate tissue.

Most prostate cancers are slow-growing and enlarged prostate and prostate cancer may be detected during physical (rectum) exams.

According to the statistic, approximately, 1 man in 9 will be diagnosed with prostate cancer during his lifetime and 6 cases in 10 are diagnosed in men aged 65 or older.

Prostate cancer shared common symptoms of an enlarged prostate, including frequent urination particularly at night, weak or interrupted urine flow and erectile dysfunction, blood in the urine, and blood in the seminal fluid.

The causes of prostate cancer are unknown. The increases with age, especially after age 50. family history. ethnicity, hereditary breast, and ovarian cancer (HBOC) syndrome, and genetic proposition are some prevalent factors found in patients with the condition.

Conventionally, treatments for prostate cancer are totally depending on the type, stage, and grade accompanied by the patient's overall health, age, and life expectancy.

Ashwagandha also known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, the Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine for over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

Researchers on finding a potent and natural ingredient for the treatment of prostate cancer investigated the effects of Withania extracts on highly metastatic, androgen-independent prostate cancer cell line (PC3).

According to the results obtained from tested differentiation, the extracts demonstrated strong inhibitory activity in decreasing the cancer cell viability with a half-maximal inhibitory concentration (IC50) of 10 μg/mL.

The sample of prostate tissue and PC3 cells also exhibited the predominantly high expression levels of IL-8 associated with the production of chemokine produced by macrophages and COX-2 involved in the contribution of immune evasion and resistance to cancer immunotherapy was also suppressed by the lowest dose of W. somnifera in 24 hours.

Furthermore, the extract-induced cycle arrest of the cancer cell line at the cell cycle in the G2/M phase, was observed from the rapid accumulation of PC3 cells significantly.

Moreover, W. somnifera extracts suppressed the PC3 inherent metastatic activity.

Based on the results, researchers wrote, "W. somnifera may be a good therapeutic agent in addition to the existing drugs for PC".

Taken together, Ashwagandha may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of prostate cancer, pending the confirmation of a larger size and multicenter human study.

#Bilberry Modulates Gene Expression Against the Onset of #LungCancer, Scientists Say

Kyle J. Norton

Bilberry may have a strong effect in reducing the risk and treatment of lung cancer, according to studies.
Lung cancer is a chronic condition characterized by cell growth irregularly and disorderly in the lung tissue.

Most cases of lung cancer caused by the alternation of calls DNA begin in the cells on the surface of the inner lining of the lung.

At the advanced stage, the cancer cell from the lung can travel a distance away to infect other healthy tissue and organs through the circulation of fluid and blood, leading to secondary metastasis.

According to statistics, in 2017, 28,600 Canadians were diagnosed with lung cancer, representing 14% of all new cancer cases. Also in 2017, lung cancer caused the death of 21,100 Canadians.

Smoking although it is a major cause of lung cancer, however, not all lung cancer patients with lung cancer are smokers. Nonsmokers' lung cancer may be caused by exposure to radon, secondhand smoke, air pollution, or other factors.

Some researchers suggested that lung cancer in many incidences is associated with oxidative stress as found in advanced lung cancer patients.

Dr. the lead scientist in the examination of the oxidative stress in the risk lung cancer said, "Oxidative stress also plays important roles in the pathogenesis of many diseases, including aging, degenerative disease, and cancer. Among cancers, lung cancer is the leading cause of cancer in the Western world".

And, "Distinct measures of primary and secondary prevention have been investigated to reduce the risk of morbidity and mortality caused by lung cancer. Among them, it seems that physical activity and nutrition have some beneficial effects".

The results clearly suggested that the external sources of dietary antioxidants from the food sources may be helpful in reducing the risk of lung cancer by suppressing the overexpression of free radicals before they can induce oxidative stress.

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The herbal plant has been used as foods and herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

Researchers on finding a replacement for the treatment of lung cancer with no side effects examined the berries' bioactive compound anthocyanidins and its derivative cyanidin, malvidin, peonidin, petunidin, and delphinidin anticancer effects.

A mixture, including blueberry, bilberry and Indian blackberry ('Jamun') was selected to test in two aggressive non-small-cell lung cancer cell lines.

Injection of the mixture exerted a higher induction of cell-cycle arrest, apoptosis, and suppression of NSCLC cell invasion and migration compared to individual anthocyanidins.

According to tested assays, the mixture inhibited lung cancer cells apoptosis by decreasing the expression of the genes associated with cell cycle division and cell survival and proliferation.

Furthermore, the mixture also elevated the protein involved in cancer cell death programming.

Interestingly, in vivo, both the mixture of anthocyanidins from bilberry (0.5mg/mouse) and the most potent anthocyanidin, delphinidin (1.5mg/mouse) significantly inhibited the growth of H1299 xenografts in nude mice by 60%.

Dr. Kausar H and colleagues said, in the final report "Our results thus demonstrate the therapeutic potential of berries rich in this mixture of diverse anthocyanidins for non-small-cell lung cancer treatment and to prevent its future recurrence and metastasis".

In order to reveal more information about bilberry anti-lung cancer activity, researchers assessed the involvement of oxidative stress in lung adenocarcinogenesis, using mice deficient in the 8-hydroxyguanine DNA glycosylase 1 (Ogg1) gene and wild-type mice (C57BL6/J origin) administered 4-(N-hydroxymethylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK).

Where deficiency of 8-hydroxyguanine DNA glycosylase 1 (Ogg1) gene was found to associate with the advanced stage of cancer.

On week after induced lung cancer by administered 4-(N-hydroxymethylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), selected mice were treated with one of 4 antioxidants (phenyl N-tert-butyl nitrone 0.13% in drinking water, resveratrol 20 ppm in the diet, lactoferrin 2% in diet and bilberry powder 2% in diet) or no supplement for 33 weeks.

All antioxidant applications demonstrated a strong inhibition against lung adenocarcinogenesis, through modulating the gene expression of gene deficiency such as 8-hydroxylamine DNA glycosylase 1 (Ogg1) and gene mutation such as Egfr and K-ras.

Taken altogether, bilberry processed abundantly bioactive compounds me be considered a functional remedy for the prevention and treatment of lung cancer, pending the confirmation of a large sample size and multicenter human study.

#Bromelain Inhibits Inflammation and Destructive Arthritis-Associated Changes in Rheumatoid Arthritis, Researchers Say

Kyle J. Norton

Rheumatoid Arthritis (RA) a chronic disorder as a result of inflammation, affecting mostly the flexible (synovial) joints and tissues and organs in the body.

The disease affects more women than men and generally occurs after the ages of 40, causing the diminished quality of life of many elders(1).

According to CDC, Musculoskeletal disorders (MSDs) affect over 52 million of adults in the US alone, including 294,000 children under age 18 with some form of arthritis or rheumatic conditions(2).

Rheumatoid Arthritis can induce bone loss through elevating bone resorption without increasing bone formation(4). A cross-sectional population-based study of 1042 patients with rheumatoid arthritis showed that RA patients had an increased risk of death from various causes(4a).

The most common symptoms of RA are
* The feet and joints
Patients with rheumatoid arthritis (RA) often associate with valgus deformities of the feet and gait due to body function in an effort to support the collapsing longitudinal arch of the foot.

* The gait
Rheumatoid arthritis (RA) causes a significant reduction in joint motions, joint moments, such as decreased hip flexion-extension range, hip abduction, knee flexion-extension range.

* The hands
The effects of the deformation of RA's hands can be described through the defects of bone ankylosis as a result of bone damage to the prolonged onset of the disease(12).

* The Muscle strength
The effects of muscle strength in a patient with RA may contribute to the prevalence of functional limitations physically of that can affect daily living and quality of life.

* Rheumatoid Nodules
Rheumatoid nodules, lump on the skin, closed to the joint have been found to affect the joints of patients with rheumatoid nodules.

* Conjunctival nodule
Although is rare in patients with rheumatoid arthritis, the appearance of the nodules may exhibit the
the severity of the underlying disease.

* Morning stiffness
Morning stiffness, as a marker of inflammatory activity that reflects functional disability and pain, is a very common symptom of patients with early rheumatoid arthritis.

Conventionally, there is no cure for RA. Medication is to relieve symptoms such as pain, slow the progression and improve quality of life, including disease-modifying antirheumatic drugs (DMARDs) and nonsteroidal anti-inflammatory drugs (NSAIDs), and corticosteroids.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural agent for the treatment of adjuvant rheumatic diseases, researchers examined the anti-inflammatory property of both enzyme therapy and combined therapy with cyclosporin in an animal model.

The study included rats with adjuvant-induced arthritis administered either cyclosporin A (2.5 or 5.0 mg/kg/day per os), a mixture of enzymes (Phlogenzym (PHL); 45 mg/kg twice daily intrarectally), or a combination of 2.5 mg cyclosporin A and 90 mg PHL for a period of 40 days.

According to the tested assays, the combination therapy with cyclosporin A plus PHLshowed a significantly inhibited both the inflammation and destructive arthritis-associated changes, such as serum albumin, changes in hind paw swelling and bone erosions.

Compared to the higher dose, 2.5 mg of cyclosporin A and PHL alone inhibited the disease markers to a lesser extent and a later stage of arthritis development.

In other words, the combination inhibited all parameters associated with the onset of adjuvant rheumatic diseases in a dose-dependent manner.

Based on the findings, researchers wrote, "The results show the inhibitory effect of enzyme therapy on rat adjuvant arthritis, as well as the efficacy of a low dose of cyclosporin A given in combination with enzyme therapy, which may be useful in the treatment of rheumatoid arthritis".

Taken altogether, bromelain used alone or combined with conventional medicine may be considered a remedy for the prevention and treatment of adjuvant rheumatic diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Andrographis #HydroalcoholicExtract Exerts a Strong Gastroprotective Property, Researchers Suggest

Kyle J. Norton

Andrographis paniculata showed to improve gastric tone and prevent gastric dysfunction through the protection of its antioxidants, the renowned institute suggested.

Andrographis paniculata also known as Andrographis is a herbaceous plant, genus Andrographis, belonging to the family Acanthaceae, native to India and Sri Lanka. It has been used in traditional Siddha and Ayurvedic in treating the common cold, flu, upper respiratory infections, infectious diseases, diarrhea, dysentery, cholera, pneumonia,...

The gastroprotective effect is a condition involved that enhances the function of the digestive system against harmful organisms in induced chronic indigestion.

In the study of gastric ulcers induced by ethanol, aspirin, pylorus ligation, and cold restraint stress models, rats treated with rats pretreated with HAEAP (100,200,500mg/kg b. wt for 30 days) of Andrographis paniculata expressed a significant ulcer reduction, in a dose-dependent manner, according to the Bharathi Women's College.

Dr. Panneerselvam S, the lead author said, "The ulcer-preventing effect of HAEAP may partly be due to its regulating effect on H(+)K(+)ATPase activity and /or mucin preserving effects" and "The flavonoids present in the HAEAP might be responsible for the gastroprotective action probably by maintaining the antioxidants and thiol status in the gastrointestinal tract".

In support of the above differentiation, the review of a period of 1 year from 2014-2015, using semi-structured questionnaires from different traditional Bengali people having knowledge of medicinal plants, out of 49 plants under 46 genera belonging to 33 families, Andrographis paniculata is one of the most prominent plants used among the people of Southern Assam for treating digestive system disorders (DSD).

Furthermore, seed ethyl acetate, acetone, and methanol extracts of Andrographis paniculata (Burm.f.) Wall. ex Nees has drawn a lot of scientists' attention due to the limited success of chemical anthelmintics, a group of antiparasitic drugs (levamisole, flubendazole, and thiabendazole) in reducing gastrointestinal diseases.

In animal controls, ED(50), an ethanol extract of A. paniculata at a dose of 2.90 mg/ml, and in larval development assay at ED(50) of dose 4.26 mg/ml, showed significant effects against sheep parasite, Haemonchus contortus.

Taken together, there is no doubt that Andrographis paniculata consisted of a strong gastroprotective effect against the activities of harmful organisms, including parasites, and may be used as a functional food in improving digestive functions.

#Aloe Vera Lel Extract Restores Normal Levels of Cholesterol in Diabetes, Researchers Say

Kyle J. Norton

Dietary Aloe vera improved plasma lipid profile, through enhancing antioxidant and hepatoprotective capacities, a study by a renowned institute suggested.

Aloe Vera is species of succulent plant in the genus Aloe, belonging to the Family Xanthorrhoeaceae, native to Sudan. It has become very popular for commercial cultivation due to its health benefits. Aloe vera has been used in herbal medicine in treating many kinds of diseases, including wound, burn healing, minor skin infections, sebaceous cysts, diabetes, and elevated cholesterol, etc.

Cholesterol is needed for our body to build cell walls, make hormones and vitamin D, and create bile salts for digested fat. However too much can be dangerous because cholesterol cannot dissolve in your blood.

According to the joint study led by the Nanjing Agricultural University, oral administration of less than or equal to 1.88, 1.86, and 2.79 %/kg in test animals showed improved plasma lipid profile status, antioxidant, and hepatoprotective enzyme activities.

Dr. Gabriel NN, the lead author suggested, animals fed with Aloe Vera exhibited a significantly elevated high-density lipoprotein in 0.5 and 1 % Aloe/kg with no significant changes in low-density lipoprotein. After 8 weeks of treatment, the group fed dietary Aloe at 4 %/kg expressed a significantly reduced total cholesterol and triacylglycerol.

Also in an animal study of 5 treatment groups of rats (n = 7) fed with experimental diets: a normal control diet, a hypercholesterolemic diet (HD), hypercholesterolemic diet (HD) + Lactobacillus rhamnosus GG (LGG), hypercholesterolemic diet (HD) + AV gel, and hypercholesterolemic diet (HD) + Lactobacillus rhamnosus GG (LGG) + AV gel, the group fed with a combination of LGG and AV gel exerted a therapeutic potential to decrease cholesterol levels and the risk of cardiovascular diseases

Furthermore, the efficacy of aloe vera in inhibited of high blood cholesterol is the result of the complex functions in restoring normal levels of high-density lipoprotein-cholesterol, low-density lipoprotein-and very low-density lipoprotein-cholesterol, through blocking altered fatty acid composition in the liver and kidney.

The most effective dose of oral administration of Aloe vera gel extract is a dose of 300 mg/kg body weight per day to STZ-induced diabetic rats.

Dietary Aloe Vera has been shown to inhibit lipid peroxidation and restore normal ratio levels of blood cholesterol in some animal studies, human trials are necessary to validate this claim.

#Celery Suppresses Symptoms of #ArthritisDiseases by Inhibiting the Production of Pro-Inflammatory Cytokines

Kyle J. Norton

Celery may be used as natural food therapy in reducing symptoms of acute and chronic painful or inflammatory conditions, such as arthritis.
According to a recent study, Celery Seed Extract (CSE) has been found to be more effective in comparison to aspirin, ibuprofen, and naproxen in suppressing arthritis symptoms in a model of polyarthritis without interaction with compared medication intake.

Dr. Powanda MC, the lead researcher said, "Unlike many dietary supplements, the available data suggest that CSE does not significantly affect the p450 enzyme systems( function in interaction with medication intake) and thus is less likely to alter the metabolism of drugs the individual may be taking"

Celery is a species of Apium graveolens, belonging to the family Apiaceae, cultivated all around the globe as a vegetable. Celery can grow to 1/2 m tall with stalks (leaf on the top) arranged in a conical shape joined at a common base.

The over-the-counter (OTC) oral remedies for arthritis and rheumatism classified celery seed extracts have an effective efficacy as ibuprofen in the anti-arthritic assay without causing gastric bleeding, but the treatment ranged from almost zero to highly effective depending on a certain class of product.

In support of the above differentiation, A. graveolens (AGE) against liver oxidative stress in an adjuvant-induced arthritic rat model study found that oral administration of AGE (250, 500, and 1,000 mg/kg) for 24 consecutive days in the rat model, reduced symptoms of arthritis through reduced liver oxidative stress and enhanced liver antioxidant properties.

Taking it all together, the intake of celery may be effective in ameliorated symptoms of arthritis through binding to the expression of proinflammation cytokines. But oral administrated a large number of celery supplements such as celery seed should be consulted with related field specialists.

#Herbal #AshwagandaRoot Extract Inhibits Stress Associated with Levels of Serum Cortisol, Researcher Show

Kyle J. Norton

Chronic stress is the response to emotional pressure in demanding situations over a period of time, leading to the release of the hormone cortisol and adrenaline.
The most common factors associated with stress incidence include psychological pressures, such as work deadlines, and exams, high-pressure jobs, and financial difficulties.

Over time unresolved chronic stress can cause a wide range of symptoms and increase the risk of anxiety.

Anxiety disorder is a class of medical conditions associated with chronic worries and fear, leading to symptoms of the fast heart rate and shakiness.

Most people with anxiety may experience the feeling of nervousness in responding to events such as a problem at work, before taking a test, or before making an important decision.

According to the statistics, anxiety disorders are the most common mental illness in Canada and approximately 1 in every 8 Canadians is affected by the illness. In other words, over 250,000 people in Canada suffer from anxiety.

The most common prevalent factors that induce the risk of the condition are, family history, gender (women are more likely diagnosed with the condition compared to men), chronic stress, people with traumatic life experiences and the use of substances are some prevalent risk factors of anxiety.

However, some researchers suggested moderate physical activity, mind-body practices of breathing exercises, yoga, and meditation have been found to reduce stress, a strong positive impact on anxiety.

The Anxiety and Depression Association of America wrote, "Whether, in good times or bad, most people say that stress interferes at least moderately with their lives. Chronic stress can affect your health, causing symptoms from headaches, high blood pressure, and chest pain to heart palpitations, skin rashes, and loss of sleep".

"Physical activity is a proven way to reduce stress. Regular participation in aerobic exercise has been shown to decrease overall levels of tension, elevate and stabilize mood, and improve sleep and self-esteem. Other effective methods include mind-body practices of breathing exercises, yoga, and meditation".

Ashwagandha also known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, the Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered an anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

In finding a potent compound for the treatment of symptoms associated with chronic stress, researchers examined the efficacy of a high-concentration full-spectrum extract of Ashwagandha roots in reducing stress and anxiety in adults who were under stress.

A total of 64 subjects with a history of chronic stress included were randomized to either drug treatment group and were asked to take one capsule containing 300 mg of high-concentration full-spectrum extract from the root of the Ashwagandha plant or the placebo control group. twice a day for a period of 60 days.

Based on the results of the tested analysis, the treatment group received a high-concentration full-spectrum Ashwagandha root extract and showed a significant reduction in scores on all the stress-assessment scales on Day 60 compared to the placebo.

The serum cortisol levels were substantially reduced in the Ashwagandha group, compared to the placebo group.

According to the patient's reports, there are no serious adverse events reported during the experimental period.

In other words, the short-term application of the Ashwagandha plant at a high concentration not only reduced the levels of c stress but also stress-induced hormone expression of anxiety.

Based on the findings, researchers said, "a high-concentration full-spectrum Ashwagandha root extract safely and effectively improves an individual's resistance towards stress and thereby improves self-assessed quality of life".

Taken together, Ashwagandha may be considered a functional herb for the prevention of anxiety induced by chronic stress, pending to the result of larger sample size and multicenter study
