#Cucumber's #Cucurbitacins Promote Neuroprotection by Increasing the Antioxidant Activty in the Brain Tissues, Researchers Show

The central nervous system containing the brain and the spinal cord is the complex system of nerve tissues that controls the activities of the human body.

Dr. the lead scientist wrote, "This increase in glial responses was associated with increased plaque burden in the hippocampus. Interestingly, given recent studies highlighting the importance of TREM2 in microglia/monocytes in AD susceptibility and progression, B6 and APP/PS1 mice fed the western diet showed significant increases in TREM2+ microglia/monocytes".

And, " Therefore, an increase in TREM2+ microglia/monocytes may underlie the increased risk from a western diet to age-related neurodegenerative diseases such as Alzheimer’s disease".

In finding a potential ingredient that processes neuroprotective effects, researchers evaluated the protective effect of Cucumis melo var. flexuosus (C. Melo var. flexuosus) leaf extract on the brains of diabetic rats.

The study included dietetic male albino rats induced by a single intraperitoneal injection of streptozotocin (STZ; 60 mg/kg bw).divided into 5 groups of 6 rats each, a normal control group and four diabetic groups.

Additionally, 3/4 other groups were treated with C. Melo var. flexuosus (cucumber) leaf extract at the doses of 30, 60, and 120 mg/kg bw for a period of 30 days compared to the nontreatment of a diabetic group.

Compared to non-treated diabetic rats, treated rats protected the brain neurons by markedly increasing the plasma dopamine, melatonin, and brain vascular endothelial growth factor-A levels, particularly the antioxidant activity in the brain tissues.

In other words, cucumber leaf extract processes a neuroprotective effect by preventing neuron oxidative damage associated with diabetes.

Based on the results, Dr. Ibrahim DS, at Benha University wrote, "It can be concluded that the C. Melo var. flexuosus leaf extract exerts a neuroprotective effect against oxidative damage associated with diabetes".

In order to reveal more information about cucumber neuroprotective property, researchers examined the effect of Cucurbitacins (CuB), a major bioactive compound from cucumber on neuroinflammatory responses that are involved in neuronal injury.

According to the tested results, pretreatment with Cucurbitacins significantly reduced the production of pro-inflammatory cytokines (TNF-α, IL-1β, and IL-6 iNOS and COX-2 expression) in microglia, a type of neuroglia (glial cell) located throughout the brain and spinal cord.

Furthermore, CuB also inhibited the proteins and pathways associated with the stimulation of immune response in the activated production of inflammation.

Moreover, the neuroprotective of CuB was also attributed to the bioactive compound in the activation of (Nrf2/ARE pathway) pathways involved in significantly greater resistance to oxidative stress and protein associated with the production of antioxidants and anti-inflammatory cytokines.


Dr. Park SY, the lead scientist said, "Cucurbitacins is a potent activator of the Nrf2/ARE pathway and is therapeutically relevant not only to neuroinflammatory responses of microglia but also neuroinflammation mediated neuronal injury".

Taken altogether, cucumber processed with high levels of CuB may be considered a neuroprotective remedy, pending the confirmation of the larger sample size and multicenter human study.

Intake of CuB in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Antioxidants are stable molecules that inhibited free radicals that cause oxidative stress if overexpressed.
Oxidative stress-related conditions are the class of diseases associated with cellular damage, including the alternation of cell DNA, and damage of lipid and protein.

Oxidative stress can be acute or chronic. Acute oxidative stress caused by a suddenly burst of free radicals due to stress or external sources, in most cases can be inhibited by increasing antioxidant enzymes in the lung.

Chronic oxidative stress is caused by the unbalanced ratio of free radicals and antioxidant enzymes produced by our bodies. The condition is either caused by oner expression of free radicals or depletion of antioxidant enzymes or both.

There are many risk factors that cause oxidative stress. However, in the concerns of the prevalence of chronic diseases associated with the Western diet, some researchers suggested that the link of the oxidative diseases in the Western world may be resulted by the promotion of the Western diet over the past few decades.

The list of chronic diseases caused by cell oxidation is very long, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

The most common causes of oxidative stress are overexpression of free radicals in the induction of a chain reaction.

Due to the unstable nature of free radicals with unpaired electrons in the outer ring, free radicals are ready to donate or intercept the electrons from other atoms, leading to a chain reaction that can not be stopped until the outring electron are either paired or inhibited by antioxidants.

In contrast to the general belief, the damage caused by free radicals are overly exaggerated, under normal condition, free radicals produced by internal or external source are needed by our body on cellular responses and immune function.

* Most importantly, levels of postprandial triglycerides associated with oxidative stress and antioxidant activity were significantly lower in the CJ group compared to. the high fructose corn syrup (HFCS) and ascorbate group.

Based on the findings, researchers aid, "Cranberries are an excellent source of high-quality antioxidants and should be examined in human supplementation studies".

Taken together, cranberry may be considered a remedy for the enhancement of the antioxidant activity, pending to the confirmation of the larger sample size and multicenter human study.

Based on the findings, researchers said, " the consumption of CFB improves gingival and plaque indices without posing a risk of caries development. Thus CFB can be recommended as a safe adjunct for nonsurgical PT in patients with gingivitis".

Chemotherapy is a drug treatment that kills cancer cells by inducing cytotoxicity. Most cases of chemotherapy have been found to cause severe side effects on the patient, including fatigue, loss of appetite, nausea, bowel issues such as constipation or diarrhea, hair loss, mouth sores, skin and nail problems.

On finding a potential plant that processes anti-kidney toxicity induced by chemo-drugs, researchers examined the effects of sulforaphane (SF), a natural constituent of cruciferous vegetables such as broccoli, on Cisplatin (cis-diamminedichloroplatinum II, CIS) nephropathy.

In rodents, before treatment with FN, cisplatin therapy triggered marked activation of stress signaling pathways to promote cancer cell death in the kidney by increasing the DNA fragmentation, apoptotic proteins,

Furthermore, CIS also exhibited the terminal associated with attenuation of various prosurvival signaling pathways and increased levels of the pro-inflammatory cytokines in the kidney.

In other words, the injection of CIS not only induced kidney injury but also caused damage to the death of the kidney cells, through several aforementioned mechanisms.


After the administration of SF, all these effects were significantly attenuated by the pretreatment of rodents with SFN.

Based on the evidence, researchers wrote, " SFN is able to prevent the CIS-induced renal injury by modulating these pathways, providing a novel approach for preventing this devastating complication of chemotherapy".

Taken together, the consequences of these results suggest that the broccoli process the potential anti-side effect of CIS in the induction of kidney toxicity, pending to the confirmation of the larger sample size and multicenter human study.

Esophageal cancer is a chronic medical condition that arises from the esophagus due to the alternation of cell DNA.

Most cases of esophageal cancer start in the cell on the surface of the inner lining of the esophagus between the throat and the stomach. At the early stage, due to the very small size of the tumor, most patients are asymptomatic.

As the tumor continues to grow, patients may experience symptoms of difficulty swallowing, persistent chest pain and pressure, pain-burning sensation, coughing or hoarseness, and heartburn.

Some patients may also vomit blood if the tumor has pressed onto the nearby blood vessels.

At the advanced stage, the primary cancer cell can also travel a distance away to infect other healthy tissues and organs, leading to secondary metastasis.

According to the statistics, in the US, approximately 17,650 adults (13,750 men and 3,900 women) in the United States will be diagnosed with esophageal cancer this year, accounting for 1% of cancers diagnosed.

Although there are many risk factors associated with the onset of the disease, some researchers suggested patients with chronic gastroesophageal reflux disease (GERD) are found to have a strong implication on the disease onset.

Dr. Shaheen N, the lead scientist at the University of North Carolina-Chapel Hil, wrote, "Gastroesophageal reflux disease (GERD) is a risk factor for adenocarcinoma of the esophagus, rare cancer whose incidence is increasing".

However, "Strong evidence supports the association of GERD and adenocarcinoma of the esophagus; however, the risk of cancer in any given individual with GERD is low".

The cucumber plant is a species of Cucumis Sativus, belongs to the family Cucurbitaceae, and is native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

With an aim to discover a potential ingredient for the treatment of esophageal cancer, researchers investigated the possible cytotoxic effect of freeze-dried aqueous extract of E. elaterium fruit on KYSE30 (human esophageal squamous cell carcinoma).

The study included the free dried aqueous extract of the fruits of E. elaterium incubated for 24, 48, and 72 h. on KYSE30 cancer cell lines.

The extracts showed a significant effect in the inhibition of the KYSE30 cell line at IC(50) values at concentrations of 500, 150, and 125 μg/ml after 24, 48, and 72 h, respectively.

According to the EB/AO staining, the extract showed an increase in apoptotic cells of the KYSE30 cell line.

Furthermore, the free-dried aqueous extract of the fruits of E. elaterium exerted a significant increase in cell density at the G2/M phase before inducing cell cycle arrest, according to the cell cycle analysis.

Based on the findings, researchers suggested that the freeze-dried aqueous extract of E. elaterium fruit has a cytotoxic effect on esophageal cancer cell lines via the induction of apoptosis.


Dr. Bohlooli S, the lead scientist wrote, " It seems that several compounds are possibly responsible for the cytotoxic effect of the extract".

In other words, several compounds in the extract worked synergistically to exhibit cytotoxicity against the tested cancer cell line.

Taken altogether, cucumber may be considered supplements for the prevention and treatment of esophageal cancer, pending the confirmation of the larger sample size and multicenter human study.
