#Coffee Intake Inhibits the #OxidativeInsults Associated with the Protection of Serveral Types of #ChemicalStress, According to Studies

Coffee and coffee caffeine may have a profound and positive effect in reducing the risk of genotoxicity, caused by environmental toxins, a coffee production company opinionated.

Genotoxicity is an action that causes the alternation of cell DNA due to overwhelming toxins accumulated in the cells. In most cases, the alteration affects the cells' integrity, leading to cancers.

Substances such as radiation and chemicals that cause genotoxicity are called genotoxins.


Highly genotoxic chemicals include the known potent carcinogens benzo(a)pyrene, aflatoxin B1, N-nitroso compounds, and various alkylating agents that directly act on DNA, inducing breakage and cross-linking, and the formation of DNA adducts, abasic sites, oxidative damage, alkylation, and various other DNA lesions(4).

Interestingly, some genotoxins do not directly act on DNA but cause numerical chromosome aberrations during cell division(4).

Furthermore, certain chemicals used in pesticides, food additives, starches, shampoos, and fireproof and crease-proof fabrics may be considered genotoxins as they also cause genetic mutations.

Moreover, automobile exhaust fumes and medicine used for their antibacterial activity and in chemotherapy(5), chemicals from occupational sources, and tobacco may also induce mutations and cancer(6).

Coffee, a popular and social beverage all over the world, particularly in the West, is a drink made from roasted beans from the Coffea plant, native to tropical Africa and Madagascar.

In rat liver and primary hepatocytes, regular intake of coffee daily illustrated an enormous effect on cellular antioxidant defenses through regulating exhibited enzymes involved in mRNA and protein expression.

The study also suggested that coffee and coffee caffeine through the expression of detoxifying enzymes GSTs and AKR(in cellular detoxification) also demonstrate significant activities in reduced risk of genotoxicity and cytotoxicity caused by injection of aflatoxin B1 (AFB1).

* 3 doses of instant coffee caffeine injection displayed a decrease in genotoxicity following co-treatment of mouse lymphoma cells.

* Coffee caffeine pre-treatment or co-treatment exhibited significant antigenotoxic effects against MNNG(an environmental carcinogen that causes DNA mutation) expression.

* Both caffeinated and decaffeinated instant coffee demonstrated a similar effect against genotoxicity in the experiment samples.

Dr. Abraham SK, the lead author said, " Results from three or five independent experiments demonstrated significant protective effects of caffeinated instant coffee against MNNG-induced DNA damage in the comet assay, a mutation at the Tk locus and chromosomal damage in the cytokinesis-block micronucleus test".

In support of the above analysis, the Swiss Federal Institute of Technology (ETH), in the study of protective effects of coffee against somatic mutation and mitotic recombination induced by chemo drugs, cyclophosphamide (CPH), mitomycin C (MMC) and urethane (URE), using standard (ST) and high bioactivation (HB) crosses of the wing spot test in Drosophila melanogaster assays, suggested that in dose depending, coffee injection showed a strong effect in the genotoxicity of URE in the ST and the genetically susceptible HB cross as well as the genotoxicity caused by MMC application.

Dr. Abraham SK, the lead researcher said, "The magnitude of the protective effects of coffee against these three genotoxins was independent of the genotype of the larvae used for treatment, that is it was independent of the bioactivation capacity of these larvae".

The collective information showed definitive support for coffee and coffee caffeine used as a functional food in reduced risk against genotoxic expression, including side effects of chemo medicine used in the treatment of multi types of cancers.

Inflammation is a natural response by the immune system to protect the body against the invasion of stimuli or foreign pathogens.

Acute inflammation is a short-term condition caused by tissue injury, leading to pain, redness, swelling, and heat in the affected area. depending on the size of the injury. Some patients with acute inflammation may also experience the symptom of loss of function.

In acute inflammation, the white blood cells on the first line of defense of the immune system after sensing the danger of an infection in the body through a message from the brain stimulate the production of platelets to cover the site of injury or damage and inflammatory cytokines to kill off all pathogens before they can cause harm to the body.

Most cases of infection are stopped at the acute phase. The wound or injury is slowly recovered.

However, in people with a weakened immune system, acute inflammatory progression cannot be stopped due to the inability of the immune system to destroy all invaders, which may lead to low-grade chronic inflammation.


Lipids mediators, derived from the omega-6 polyunsaturated fatty acids (PUFA) are potent enhancers of innate and adaptive immune activation in numerous inflammatory disorders.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

In finding a potential phytochemical for the treatment of inflammatory diseases, researchers examined the effects of n3-PUFA DHA and its derived specialized pro-resolving lipid mediators (SPMs) MaR1, RvD1, and RvD2, on TNF-α inhibition of intestinal sugar and glutamine uptake.


Based on the differentiation,* DHA blocked TNF-α-induced inhibition of α-methyl-D-glucose (αMG) uptake, an analog of glucose, and SGLT1, a glucose transporter, expression in the apical membrane of Caco-2 cells.

* Compared to DHA, SPMs showed the same preventive effect at concentrations 1000 times lower.

* In diet-induced obese (DIO) mice, oral gavage of SPMs, MaR1 reversed the up-regulation of pro-inflammatory cytokines found in the intestinal mucosa of these mice without inducing a reduced intestinal transport of αMG and SGLT1 expression in the DIO mice.

* In Caco-2 cells, EPA, DHA, and the DHA-derived SPMs. also prevent the TNF-α in the inhibition of glutamine uptake.

Collectively, scientists said, " DHA and its SPMs as promising biomolecules to restore intestinal nutrients transport during intestinal inflammation".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the treatment of diseases associated with impaired lipid inflammatory mediators, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose and acute liver toxicity.

#Greentea Inhibts #BladderCancer by Regulating Angiogenetic Effects on Cancer Cells Proliferation, Metastatic Spread, Researchers Say

Green tea may have a therapeutic and sustainable effect in reduced risk and treatment of bladder cancer, some scientists suggested.

In the review of medical literature published online, green tea major catechin EGCG in reduced risk and treatment may be attributed to a number of mechanisms.

In the T24 human bladder cancer cell line, application of the chemopreventive/antiproliferative potential of (-)-epigallocatechin gallate (EGCG, the major phytochemical in green tea) induced anti-malignant cell proliferation by modulating the expression of phosphatidylinositol 3' kinase/Akt in activated Bcl-2 family proteins with a function in the regulation of cell death, thus enhancing apoptosis of T24 bladder cancer cell line in dose- and time-dependent manner.

The phosphatidylinositol 3'-kinase/AKT pathway is a crucial regulator of cellular processes including cell proliferation and apoptosis.

Bcl-2 family proteins are a modulator responsible for the regulation of cell apoptosis.

In C3H/HeN mice subcutaneously implanted with MBT-2 murine bladder tumor cells, administration of EGCGs physically attached to the surface of nanogold particles pNG(EGCG-adsorbed pNG) exerted a significant anti-cancer proliferation through induction of apoptosis.

This action of anti-cancer effect was attributed to the mechanism of EGCG-pNG in the activation of caspases which plays an important role in induced cell apoptosis and inflammation through Bcl-family proteins with a function in control apoptosis by activating caspase proteases in governing the space between outer and inner mitochondrial membrane.

Under stress, the mitochondrial pathway or the intrinsic pathway is activated depending on the release of proteins from the intermembrane space of mitochondria.

Caspase cascade, a family of protease enzymes with function in programmed cell death and inflammation.

Additionally, injecting EGCG-pNG directly into the tumor site inhibited tumor expansion through ameliorated the function of vascular endothelial growth factor (VEGF) in stimulating the formation of the new blood vessels in provided nutrients and oxygen for the survival of the cancer site compared to
oral administration of EGCG in stimulated immune function in suppression of cancer progression.

A vascular endothelial growth factor is a signal protein produced by cells in the stimulated formation of blood vessels for cell division and site proliferation.

Based on the information collected,, the prepared EGCG-pNG was more effective than free EGCG in inhibiting bladder tumors in model mice.

More interestingly, bladder tumor induced by N-butyl-(-4-hydroxybutyl) nitrosaminre (BBN) 0.05% solution on 8-week-old female C3H/He mice for 14 or 24 weeks on treatment with or without green tea polyphenols (GTP) expressed a significantly lower rate of tumor invasive frequency of 23.8%; in the group treated with both GTP and BBN in compared to 65.2% in the group treated with BBN alone.

The tumor volume and microvascular density (MVD) as an independent predictor within a tumor. and the connective, functionally supportive framework of a biological cell, tissue, or organ region in the BBN+GTP group was also significantly lower than in BBN-treated mice.

Unbelievably, green tea polyphenols inhibited tumor growth and invasion in mice with established bladder cancer through regulation of angiogenetic effect in suppressing the forming of new blood vessels for the tumor cells proliferation, metastatic spread, as well as providing oxygen and nutrients and removal of waste products of cancer cells without exhibiting anti-carcinogenic effect.

Taken together, green tea extract polyphenols may be considered as an adjunct therapy for reduced risk and used in combination with standard medicine for the treatment of bladder cancer.

The diseases are most common in the aging population.

These results suggested that intentional carrot wounds may have a potential effect as an inexpensive rich source of phenolic antioxidants for the regular diet (with some resistance from the public), as Dr. Surjadinata BB, the lead author said, " By simply increasing wounding stress intensity, it is possible to enhance the biosynthesis of phenolic antioxidants".

Moreover, according to the investigation of the polyphenolic profile and biological properties of a black carrot crude extract (BCCE) through an in-depth analysis of the main polyphenolic effect in antioxidant and cytoprotective properties.

Black carrot (Daucus carota L. ssp. sativus var. atrorubens Alef.) is a valuable source of carbohydrates, minerals, and vitamins.

The root vegetable contains also high amounts of anthocyanins in the expression of the characteristic of deep-purple color.

Anthocyanins are known as natural dyes used in the food and pharmaceutical industry.

According to the experiment, the application of black carrot crude extract (BCCE) demonstrated a strong antioxidant through free radical scavenging activity, observed in the hydrogen transfer-based assays (ORAC and β-carotene bleaching) and a significant increase in the cell viability.

The increase in the cell viability may be attributed to the extract in stimulated production of antioxidants of itself and antioxidants found in the body tissue of the host inhibited the overexpression of ROS, resulting in balancing the antioxidants and free radical ratio.

Furthermore, BCCE exhibited strong anti-angiogenic activity against free radicals in alternated DNA of healthy cells in the initiation of tumor proliferation, at the highest concentration assayed on the chick chorioallantoic membrane (50μg/egg).


Interestingly, ingestion of black carrot crude extract (BCCE) also promoted a cytoprotective effect against ROS in causing healthy cell damage and apoptosis.


The analysis of the above studies was supported by the joint investigation of purple carrot (PC) as a valuable source of antioxidants in modulating the reactive oxygen species (ROS) level in the gastrointestinal tract conducted by a number of institutes, led by the Poznań University of Life Sciences.


According to the end experiment, the application of purple carrot (PC) showed a strong effect in inhibited colonic cells in situ after allowing passage to the gastrointestinal tract.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite, and fatigue but also localized symptoms depending on the affected tissues and organs.

Mouth/oral cancer is a medical condition associated with irregular cell growth on the tissue of the mouth caused by genetic alteration of the cell DNA.

Most cases of mouth cancer start in the flat, thin cells (squamous cells) of the inner lining tissue that line the lips and the inside of the mouth.

According to the statistics provided by the American cancer society, in 2018, approximately, over 50,000 people will get oral cavity or oropharyngeal cancer. An estimated over 10,000 people will die of these cancers.in the United States.

The exact causes of mouth cancer are unknown. However, the increase in age, gender, exposure to radioactive, long-term use of tobacco use and excessive alcohol drinking, viral infection, and genetic mutation are the most common prevalent risk factors found in people with mouth cancer.

Risk factors associated with the onset of mouth cancer are
* Smoking
In the studies, Cancer Prevention Study (CPS) I and II, sponsored by the American Cancer Society (ACS), researchers found that the mortality risk for oral cancer in cigarette smokers is substantially greater than that observed among lifelong never smokers.


* Alcohol drinking

* Chewing tobacco

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, seed, or nut oils, etc.

In finding a potential phytochemical for the treatment of chronic diseases, researchers investigated associations between retinol, α-tocopherol, and β-carotene with the risk of oral cancer.

The comparative cross-sectional study included a total of 240 involving oral cancer patients and healthy individuals in matched cases and controls where subjects were selected from the Malaysian Oral Cancer Database and Tissue Bank System (MOCDTBS).

According to the analysis,
* Serum levels of retinol and α-tocopherol among cases (0.177±0.081, 1.649±1.670μg/ml) were significantly lower than in controls (0.264±0.137, 3.225±2.054μg/ml) (p<0.005) compared to treatment group.

* Serum levels of β-carotene among cases (0.106±0.159 μg/ml) were lower compared to controls (0.134±0.131μg/ml)

These results suggested that high serum levels of retinol (OR=0.501, 95% CI=0.254-0.992, p<0.05) and α-tocopherol (OR=0.184, 95% CI=0.091-0.370, p<0.05) was significantly related to lower risk of oral cancer.


After taking other factors into account, researchers said, "High serum levels of retinol and α-tocopherol confer protection against oral cancer risk".

Taken altogether, tocopherols may be considered an adjunct therapy in the treatment of oral cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose and acute liver toxicity.
