#Anthocyanoside (VMA) Found in #Herbal #Bilberry Shows a Neuroprotective Effect Against Retinal Damage, Researchers Say

Neuro-toxicity is a condition of brain damage caused by exposure to toxic substances.
The toxic condition was found to reduce the brain function in transmitting and processing signals to other parts of the nervous system.

Symptoms of neurotoxicity are totally depending on the degree of toxicity. Some symptoms may take some time to develop. However, in most cases, symptoms may appear immediately after exposure.

Depending on the area and location of neurons damage, weakness or numbness, loss of memory, vision, headache change of cognitive and behavioral and sexual dysfunction are the most common symptoms found in people with neurotoxicity.

In other words, out of millions of people who have neurobehavioural problems, researchers do not know how many of them are actually caused by toxicity.

Bilberry is a species of low-growing shrub in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.


The plant has been used as herb in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, psoriasis, diabetes, etc.

In the analysis of natural compounds with neuroprotective effects in patients with diabetes, researchers examined the antioxidants richly present in bilberry fruit's effects on diabetes-related brain dysfunctions.

The study included the differentiation of the expression patterns of αCaMKII in hippocampal neurons from non-diabetic, diabetic, and diabetic rats fed with an extract of bilberry fruit.

Where Ca(2+)/calmodulin-dependent protein kinase IIα (αCaMKII) plays a significant role in cognitive processes.

Diabetic rat's hippocampal neurons immunoreactive (ir) to αCaMKII were swollen and the lengths of the neuronal fibers were reduced compared to the significantly longer of αCaMKII-positive nerve fibers in diabetic rats fed with bilberry fruit.

Injection of bilberry fruit also induced significant changes in the average larger diameter of αCaMKII-in hippocampal neurons.

The results suggested that bilberry fruits exerted a neuroprotective effect on hippocampal neurons in diabetes through its antioxidants activity.

Furthermore, in order to find more information on bilberry's effect on the brain, researchers examined its Anthocyanoside (VMA) and/or its main anthocyanidin constituents (cyanidin, delphinidin, and malvidin) neuroprotective effect against retinal damage in vitro and in vivo.

In RGC cultures, VMA and all three anthocyanidins protected the brain against SIN-1 (chloride) toxicity and inhibited lipid peroxidation in mouse forebrain homogenates through the antioxidant property and NMDA-induced morphological retinal damage by allowing high levels of calcium ions (Ca2+) to enter the cells.

Moreover, VMA and all three anthocyanidins also protected the retinal damage by increasing the TUNEL-positive cells in the ganglion cell layer.

Dr. Matsunaga N, the lead scientist said, "VMA and its anthocyanidins have neuroprotective effects (exerted at least in part via an anti-oxidation mechanism) in these in vitro and in vivo models of retinal diseases".

Macular degeneration is a condition caused by the blurring of the center of the vision without affecting the side or peripheral vision.
Age-related macular degeneration (AMD) is an aging process found in older adults and is the leading cause of blindness in adults over the age of 55.

Most adults with age-related macular degeneration (AMD) are asymptomatic. However, it can be detected by professionals during annual eye check-ups.

Untreated AMD, over time, can lead to the increased size of blur area and interfere with vision, including reading.

AMD can be classified into 2 types
* The dry form is a milder form found in most patients with AMD with gradual degeneration of the central retinal tissues that make up the macula.

* The wet form is the serious form of AMD accounting for only 10% of all patients

However, the dry form can develop into a wet form over time.

Conventionally, as of today, there is no effective treatment for AMD. Most treatments such as low vision aids and lifestyle control are to delay the progression and improve quality of life.

In some cases, wet AMD may be treated with medications injected into the eye to stop leaking blood vessels, depending on the disease severity.


Besides aging and exposure to UVB, smoking has been found to increase the risk of AMD by 2-5 fold.

Dr. Sara Velilla, the lead scientist said, "Age-related macular degeneration (AMD) is one of the main socioeconomic health issues worldwide. AMD has a multifactorial etiology with a variety of risk factors. Smoking is the most important modifiable risk factor for AMD development and progression".

With an aim to find a natural treatment for eye disease, researchers examined the combinations of antioxidants including lutein+zeaxanthin (1 μM; 0.2 μM), zinc (10 µM), carnosic acid (2 µM), beta-carotene (2 µM), and standardized tomato extract containing lycopene and other tomato phytonutrients added to the culture media. against age-related macular degeneration (AMD).

Injection of the combination protected the eye against age-related macular degeneration (AMD) by yielding an antioxidative, anti-inflammatory, and antiangiogenic effect in M1 and M2 macrophages.

The results of the effects were attributed to the upregulation of antioxidative genes (HMOX1, SOD1) expression and the inhibition of pro-angiogenic genes and pro-inflammatory genes (SDF-1, TNF-alpha, IL-6, MCP-1).

Compared to the combination, lutein monotherapy or a combination of lutein and zinc had less effect on the expression of these genes.

Dr.Rinsky B, the lead scientist said, "Combinations of supplements can modify the expression of genes and proteins that may be relevant for the involvement of macrophages in the pathogenesis of AMD".
Futhermore, in oxidative stress-induced damage to the retinal pigment epithelial (RPE) on age-related macular degeneration and neovascular retinopathies (e.g., retinopathy of prematurity, diabetic retinopathy, researchers investigated xanthophyll carotenoids, lutein, and zeaxanthin, isolated from toma in damage of retinal pigment epithelial (RPE) cells.

Also, the administration of carotenoids, lutein, and zeaxanthin showed to protect the macula (and the broader retina) via their antioxidant and photoprotective activities.

In undifferentiated ARPE-19 cells, lutein and lycopene, but not β-carotene, protected the cell integrity and viability by inhibiting the overexpression of retinal cell growth.

Moreover, pre-incubation of ARPE-19 cells with lutein or lycopene protected against tBHP-induced cell loss and cell co-exposure of lutein or lycopene with tBHP essentially neutralized tBHP-dependent cell death at tBHP concentrations up to 500 μM.


Where tert-Butyl hydroperoxide (tBHP) is an organic peroxide used to induce oxidation processes.

In other words, lutein and lycopene protected the RPE against oxidative stress-induced cell loss

Taken altogether, tomatoes may be considered a natural remedy for the prevention and treatment of age-related macular degeneration (AMD), pending the confirmation of a larger sample size and multicenter human study.

Intake of lutein and lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Alzheimer's disease (AD) is a neurodegenerative disease found mostly in the aging population characterized by impaired memory, thinking, and behavior. caused by the gradual loss or death of neurons. These symptoms are slowly getting worse over time and becoming severe enough to interfere with daily activity.
60 percent to 80 percent of dementia found in the US is involved in Alzheimer's disease (AD).

According to the statistics, the number of patients living with AD is growing fast, affecting over 5.5 million people aged 65 and older. AD is the sixth leading cause of death in the US. Most patients with AD live four to eight years after diagnosis. However, the years of survival rate is totally depending on other factors.

In contrast to general belief, getting old is a risk factor for AD but AD is not a disease of aging. Approximately 200,000 Americans under the age of 65 have younger-onset Alzheimer’s disease.

As of today, there is no cure for AD, the aim of treatment is to slow down the severity of the symptoms and delay the progression of the disease.

Selected mice induced free radical toxicity and apoptosis in human SH-SY5Y neuroblastoma cells overexpressing APP751.

APdE9 mice fed with anthocyanin-rich bilberry showed decreased Amyloid Precursor Protein (APP)APP C-terminal fragment levels, an indication of reduced death of neurons in the cerebral cortex compared to APdE9 mice fed with a control diet.

Furthermore, soluble Aβ40 and Aβ42 levels, the plaque in the brain associated with increased risk for AD, mild cognitive impairment, and cognitive decline were significantly decreased in bilberry-fed mice, compared to blackcurrant-fed mice.

Moreover, the bilberry diet protected the spatial working memory of aged APdE9 mice, compared to mice on the control diet, thus reducing the risk of deficits.


The results clearly suggested bilberry diet protects the integrity and viability of the neuron in the brain against the risk of AD.

Dr. Vepsäläinen S, the lead scientist said, "These data suggest that anthocyanin-rich bilberry and blackcurrant diets favorably modulate APP processing and alleviate behavioral abnormalities in a mouse model of AD".

In order to reveal more information on bilberry anti-AD activity, researchers examined the anthocyanoside extracts (Vaccinium myrtillus anthocyanoside (VMA)) obtained from bilberry on the aggregation and deposition of the amyloid beta (Aβ1-42) peptide in fibrillar form.

According to the tested assays, injection of VMA-protected Neuro2a cells by inhibiting the formation of Aβ peptide fibrils and reducing the toxicity of these aggregates in vitro.

A diet containing 1% VMA was found to exhibit significant activity in the prevention of cognitive degeneration in AD-tested mice.

Interestingly, VMA promoted the cognitive ability accompanied not by a reduction in an aggregate deposition in brains, but rather an increase in insoluble deposits compared to control diet mice.

In other words, bilberry protected the brain neuro against the risk of AD by diverting Aβ aggregation to insoluble deposits, an alternate, non-toxic form.

Taken altogether, bilberry-processed abundantly bioactive compound Anthocyanoside may be considered a functioning remedy for the prevention and treatment of AD, pending the validation of larger sample size and multicenter human study.

Hypothermia is a medical emergency due to the loss of heat faster than the body can produce, leading to a temperature that lowers the normal low body temperature.
In other words, if your body temperature falls below 95 F (35 C) with no signs of reverse, you may have hypothermia.

Most case of hypothermia in North America is associated with long-term exposure to cold temperature due to inadequate clothing. According to the statistics, from 1999 to 2011, hypothermia caused the death of a total of 16,911 people in the United States associated with exposure to excessive natural cold.

Orally administered GE/SD (30 mg of GE/kg) inhibited ethanol-evoked hypothermia by improving oral absorption of 6G and 8G.

Storages of ginger were found slightly reduced levels of 6G and 8G by degradation without affecting the functions of 6G and 8G against the hypothermic activity.

Oral administration of GE (300 mg/kg) and GE/SD (100 mg of GE/kg), significantly elevated the relative bioavailabilities of 6G and 8G in GE/SD by 5.0- and 5.8-fold compared those in GE against the hypothermia in tested subjects.

The results suggested that solid dispersion (SD) may be used to improve the GE function of hypothermia by enhancing the levels of bioactive compounds.


Taken all together, ginger-processed bioactive compound 6-gingerol (6G) and 8-gingerol (8G) with or without employing a solid dispersion (SD) approach may be considered a functional remedy for the prevention and treatment of hypothermia with no side effects, pending the confirmation of a large sample size and multicenter human study.

Furthermore, according to the investigation of the antiobesity benefits of stingless bee honey (SBH) from H. itama in 48 male Sprague Dawley (SD) induced obesity by a high-fat diet, 3 dosages of SBH showed a higher reduction in body mass index (BMI), percentage of body weight gain, adiposity index, and relative organ weight (ROW) compared to typical obesity treatment methods.

#Gingers Intake Daily Attenuate the Progression of MGO-Associated with Patients' #DiabeticComplications, Researchers Say

Advanced glycation end is a product formed by protein or fat combined with sugar caused by cooking meat at high temperatures.
The products have been found to affect metabolic health.

Most common causes of AGEs are from the diet, particularly in people who follow the Western diet with high saturated and trans fat, a lot of red meats, and processed foods.

Low levels of AGEs may not be a problem, however, overexpression of AGEs can induce oxidative stress, leading to inflammation and damage to proteins, lipids,s and sometimes alternation of cell's DNA.

In other words, AGEs initiate the production and free radicals and free radicals a chain reaction that can not be stopped until all electrons in the outer ring are paired or inhibited by antioxidants.

Accelerated formation and accumulation of advanced glycation end-products have been found to increase hyperglycemia or oxidative stress, leading to age-related and chronic diseases such as diabetes mellitus, chronic renal failure, neurodegenerative diseases, osteoarthritis and also non-diabetic atherosclerosis, and chronic heart failure.

Some researchers suggested that modern life byproducts AGEs are the results of the promotion of the processed food diet.

Dr. Chetan Sharma, and Amarjeet Kaur, the lead scientist in the assessment of AGEs, wrote, "As modern diets are largely heat processed, they are more prone to contain high levels of advanced glycation end products (AGEs)".

And, "The chemistry of the AGEs formation, absorption and bioavailability and their path-biochemistry particularly in relation to different complications like diabetes and aging,... The emerging evidence about the adverse effects of AGEs makes it necessary to investigate the different therapies to inhibit AGEs".

The results strongly suggested the link of a modern diet with highly processed food is a leading cause of chronic diseases.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers on finding a medical remedy in the inhibition of advanced glycation end (AGE) compared tea flavonoid (-)-epicatechin and [6]-Shogaol, a bioactive compound found in ginger against methylglyoxal (MGO) induced AGE formation.

Where Methylglyoxal (MGO) is a reactive dicarbonyl species considered a precursor of advanced glycation end products (AGEs).

Application of tea flavonoid (-)-epicatechin and [6]-Shogaol showed a strong effect in decreasing about 41% of MGO at a concentration of 10 μM within 24 h.

Both compounds also exerted a similar effect in vivo.

According to the urine samples collected from epicatechin and [6]-school + epicatechin treated mice, both compounds also demonstrated significant activity of adduction against mono-MGO.

Dr. Huang Q, the lead scientist in the joint study at the Nanjing Normal University, said, "This is the first study demonstrating the additive MGO trapping efficacy of [6]-shogaol and epicatechin and that [6]-shogaol and epicatechin retained their MGO trapping capacity in mice".

The results suggested that [6]-shogaol + epicatechin prevents the production of AGEs by inhibiting the precursor (MGO).

Additionally, in order to reveal more information about the ginger effect on AGEs, researchers investigated [6]-shogaol (6S) and [6]-gingerol (6G) on Methylglyoxal (MGO) which has been found the contributes to the protein glycation and formation of advanced glycation end products (AGEs).

Similar to the above study, both [6]-shogaol (6S) and [6]-gingerol (6G) markedly trapped MGO in vitro.

In the depth analysis, researchers found that 6S or 6G trapping MGO. was associated with the α-carbon of the carbonyl group in the side chain of the compounds in a time-dependent manner.

The findings showed the potential effects of 6S and 6G on the prevention of protein glycation by inhibiting the MGO.

Dr. Zhu Y, the lead scientist said, "...regular consumption of ginger root extract may attenuate the progression of MGO-associated diabetic complications in patients.

Taken all together, ginger used alone or combined with green tea may be considered a functional remedy for the production of advanced glycation end products, pending the confirmation of large and multicenter human studies.

Diabetic renal inflammation is a condition associated with inflammation in the development of diabetic nephropathy.
Diabetic nephropathy (Dn) is a chronic kidney disease in diabetics, a leading cause of end-stage renal failure.

Epidemiologically, diabetic nephropathy is also associated with cardiovascular disease and increases the mortality of diabetic patients.

Metabolic and hemodynamic alterations, oxidative stress, and activation of the renin-angiotensin system are some of the risk factors that have been found in patients with Dn.

Inflammation is a natural defense of the immune system that protects the body against the insult from foreign invasion.

Acute inflammation occurs quickly or within hours and lasts a few days or weeks and the damaged tissues are healed itself after all pathogens are killed off.


However, if the immune system can not completely destroy all invaders in the acute phase of infection, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

In diabetes, the presence of chronic low-grade inflammation activated by the innate immune system induced production of pro-Inflammatory cytokines, mainly IL-1, IL-6, and IL-18, as well as TNF-α, are some of the major factors that involved in the development and progression of diabetic nephropathy.

Dr. Juan F. Navarro-González, the lead scientist in the examined the effect of inflammatory cytokines in the risk of Dn wrote, "When considering the role of cytokines in pathophysiological processes underlying disease, it is necessary to take into account the fact that the activities of these molecules are very complex, as reflected by important features, including their pleiotropic actions—and thus a cytokine may trigger several different cellular responses depending on diverse factors, such as cell type, timing, and context".

And, "Cytokines share receptor subunits; act synergistically in many contexts, and, therefore, the association of two cytokines can markedly amplify their effects; cytokines stimulate the cells that produce them, or adjacent cells, or even can intervene through direct cell-cell interaction; and, finally, cytokines induce the expression of other cytokines and cytokine receptors".

In other words, diabetic renal inflammation is the main culprit that causes the development of Dn.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, and wound healing agent, and for the treatment of flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

With an aim to find a potential compound for the treatment of renal inflammation, researchers examined the effect of Curcumolide, a novel sesquiterpenoid isolated from Curcuma wenyujin for the control of diabetic retinopathy.

According to the experiment results, treatment with curcumolide alleviated retinal inflammatory activities both in vitro and in vivo in an STZ-induced diabetic rat model and in TNF-α-stimulated HUVECs.

Furthermore, curcumolide alleviated retinal vascular permeability by blocking the quality of a material or membrane that causes it to allow liquids or gases to pass through it, and leukostasis that causes elevated blast cell count and symptoms of decreased tissue perfusion in the retinas.

Moreover, curcumolide attenuated the overexpression of TNF-α and ICAM-1associaed with the production of pro-inflammatory cytokines in diabetic retinas through activation of NF-κB a protein complex and p38 MAPK.

Based on the findings, researchers wrote, "These data suggest potential treatment strategies against diabetic retinopathy, particularly in the early stages of the disease".

Taken altogether, turmeric processed abundantly bioactive compound curcumolide may be considered supplements for the prevention and treatment of renal inflammation, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.
