#Phytochemical 3,3'-#Diindolylmethane Protects Diabetics Against Diabetes Complications by Modulating Blood Glucose and Lipid

Hyperglycemia is a condition of abnormally high blood sugar in the bloodstream associated with diabetic complications in patients with type 2 diabetes.

In order words, hyperglycemia is a result of the body which isn't properly using or doesn't make the hormone insulin.

Hyperglycemia is also considered a hallmark of patients with type 2 diabetes. Another hallmark is insulin resistance.

According to the epidemiological studies, untreated or long term persistent hyperglycemia in patients with diabetes has been found to induce serious symptoms such as shortness of breath, confusion, and coma.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

With an aim to find a natural ingredient for the treatment of hyperglycemia in patients with type 2 diabetes, researchers examined the effect of the nutritive phytochemicals, indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) on oxidative stress developed in type 2 diabetes mellitus (T2DM).

The study is carried out on the genetically modified mouse (C57BL/6J mice) similar to those of metabolic abnormalities of the human disease after the administration of a high-fat diet (HFD).

Injection of DIM and I3C positively modulated the glucose, insulin, and hemoglobin (Hb), glycated hemoglobin (HbA1c) levels, an indication of the hyperglycemically modulatory activities in the tested HFD mice.

Compared to the HFD group, treatment groups showed a decrease in the levels of thiobarbituric acid reactive substances (TBARS), lipid hydroperoxides (LOOH), conjugated dienes (CD associated with oxidative stress in lipid peroxidation.

Furthermore, DIM and I3C also improved the antioxidant enzymes produced by the host observed by the increasing SOD, CAT, and GPx levels in the erythrocyte, liver, kidney, and heart.

Levels of free radical scavengers such as vitamin C, vitamin E, and GSH levels are also increased in the plasma, liver, kidney, and heart in diabetic mice.

In other words, DIM and I3C protected diabetic mice against diabetes complications by modulating blood glucose and lipid by balancing the ratio of free radicals and antioxidants in the tested subjects.

Based on the finding, researchers wrote in the final report, "the antioxidant-scavenging action was accelerated in mice treated with DIM than the I3C treatment group which was comparable with the standard drug metformin".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of hyperglycemia, pending the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Alzheimer's disease (AD) is a neurodegenerative disease correlated with major reductions of neurons to the respective target areas, named after German physician Alois Alzheimer.
Alzheimer's destroys brain cells, causing cognitive modalities severe enough to affect language communication, memory, lifelong hobbies, or social life. Alzheimer's gets worse over time, and it is fatal.

According to statistic, over 25 million people in the world today are affected by dementia and most are suffering from Alzheimer's disease. In Canada, approximately, over half a million Canadians live with dementia accompanied by 25,000 new cases diagnosed every year.

Believe it or not, in 2016, dementia cost Canada over $10 billion, including the healthcare system and out-of-pocket caregiver costs.

Epidemiologically, cigarette smoking, midlife high blood pressure, and obesity, diabetes, cerebrovascular lesions, and psychosocial factors (eg, high education, active social engagement, physical exercise, and mentally stimulating activity) are associated with prevalent risk of AD development.

Some researchers evaluated that the increased AD in older adults in the Western world over the past few decades may be correlated to the promotion of a high-fat diet.

Dr. Peter Thériault, the lead researcher wrote, "WD promotes systemic inflammation by increasing the levels of blood-circulating monocytes and monocyte chemotactic protein-1 production, which is accompanied by an augmentation of oxidized-low density lipoprotein levels in blood circulation".

And, "At the blood-brain barrier (BBB), WD potentiates the age-induced increase of Aβ 1-40 accumulation and exacerbates the oxidative stress, specifically in cerebral microvasculature. These effects were accompanied by the dysfunction of pericytes, thus altering BBB functionality without compromising its integrity".

In other words, if you are a high-diet follow, you are at high risk of AD developing.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

On finding a potential compound for the treatment of Alzheimer's disease, researchers investigated the ability of bromelain, a plant cysteine protease derived from pineapple stems interact with synthetic Aβ42 monomers and oligomers.

According to the tested assays, bromelain protected the neuron successfully by degrading Aβ42 monomers and low and high molecular weight oligomers.

Examined the effects of bromelain preparations of 3 and 6 mU added to the cerebrospinal fluid (CSF), the levels of Aβ42 monomers and oligomers were significantly reduced, compared to that of the same standard Aβ42 preparations incubated in CSF without bromelain

In order to know more information about bromelain anti-Alzheimer's disease activity, researchers examined a beta-degrading activity of diverse alpha 2-macroglobulin (alpha 2-M)-proteinase including trypsin, alpha-chymotrypsin, and bromelain (125)I-A beta 1--42.

The complex showed a strong degradation of (125)I-A beta 1--42.

In Ex vivo, oral administration of high doses of proteinases displayed an enhanced degradation of (125)I-A beta 1--42 in the blood of volunteers.

In other words, the complexes exhibited a significant A beta catabolism, thus reducing the risk of developing AD by preventing A beta accumulation in brain and vasculature.

Taken altogether, bromelain used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of Alzheimer's disease, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Endometriosis is a condition caused by endometrial tissues which grow somewhere in the body instead of in the endometrium. Most cases of endometriosis are in the organs in the pelvic area, including ovaries and fallopian tubes.

Endometriosis also reacts to hormone signals of the monthly menstrual cycle, by building and breaking up tissues and eliminating them through the menstrual period.

* Minimal stage

Conventionally, most cases of endometriosis are treated by pain medicines such as NSAIDs (non-steroidal anti-inflammatory drugs) and hormone therapy such as birth control pills with an aim to reduce pain and improve quality of life, including daily activity.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers in the urgency to find natural compounds for the treatment of endometriosis with no side effects evaluated the efficacy and safety of acupuncture-moxibustion combined with modified Mojie tablet containing ginger in the prevention of the recurrence of endometriosis (EMS) after laparoscopic surgery.

The study included 120 cases of EMS after laparoscopic surgery, randomly assigned into an acupuncture-moxibustion group, a Western medication group, a Chinese medication group, and a combined therapy acupuncture-moxibustion and Chinese medication group (combined therapy group).

Each group containing 30 participants was separated, before treatment, after treatment, in 3 months after treatment, and in 6 months after treatment.

In the acupuncture-moxibustion group, acupuncture was applied at Xuehai (SP 10), Sanyinjiao (SP 6), and Guanyuan (CV 4); the isolated moxibustion with ginger was used at Guanyuan (CV 4), once a day, twice a week.

In the western medication group, gestrinone was prescribed for oral administration, 2.5 mg, once a day, twice a week.

In the Chinese medication group, the self-prepared Mojie tablet was prescribed for oral administration.

And, in the combined therapy group, acupuncture, moxibustion, and a modified Mojie tablet for oral administration were used in combination.

In 3 and 6 months after treatment, the positive rates of EHP-5 score were 0% (0/30) in the combined therapy group and 0% (0/30) in the acupuncture-moxibustion group, compared to the lower significantly than 13.3% (4/30) in the western medication group.

In the follow-up stage of 6 months after treatment, according to the abdominal ultrasound examination, there was no recurrence case in the combined therapy group, compared to the lower than 13.3% (4/30) in the western medication group.

According to assays, the serum CA125 detection in the combined therapy group did not find any abnormal cases, compared to the lower than 16.7% (5/30) in the western medication group (P < 0.05).

Cancer is a class of chronic and medical conditions caused by cell growth irregularly in the tissues of the body.
Cancer can be classified into

Most cases of cancer begin in the cells on the surface of the inner lining of the organs' tissues. For example, breast cancer starts in the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.

Under normal circumstances, cell growth is a natural process without even notice, every day millions of cells grow to replace millions of dead cells and to keep our body in balance and healthy.

Sometimes, acceleration of cell growth is necessary to prevent further damage to the body tissues and organs, such as cut, injuries, etc., but after fulfilling their purposes, the growth of cells are back to a normal rate again due to our body growth inhibitors.

Contact dermatitis is a skin condition with symptoms of a red, itchy rash caused by direct contact with a substance or an immune overreaction against contact allergens.
Many chemicals and nonchemical substances have been found to induce such abnormal reactions, including cosmetics, fragrances, jewelry, and plants.

Most cases of contact dermatitis are nonlife-threatening, however, the condition can induce certain discomforts, particularly in people with an allergic reaction to other conditions.

Contact dermatitis can be healed within a week or 2. The condition can be preventable if the cause is identified.

Besides the common symptoms of a red, itchy rash on the skin, some people may experience more serious symptoms including crusting swelling, burning, or tenderness skin.

Dr. Imbesi S, the lead scientist in the explanation of the immune involvement of allergic contact dermatitis wrote, " an inflammatory skin condition due to pro-inflammatory and toxic factors able to activate the skin innate immunity (irritant contact dermatitis) or caused by a T-cell- mediated hypersensitivity reaction (allergic contact dermatitis)".

Bilberry is a species of low-growing shrub in the genus Vaccinium, belongings to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, psoriasis, diabetes, etc.

In the urgency to find a natural ingredient for the treatment of chronic allergic contact dermatitis with no side effects, researchers examined the bilberry extract (Bilberon-25) in the alleviation of pruritus in a mouse model of chronic allergic contact dermatitis.

The study involved BALB/c mice with chronic allergic contact dermatitis induced by 3 weeks of repeated application of 2,4,6-trinitro-1-chlorobenzene (TNCB) followed by the administration of Bilberon-25 orally for 3 weeks after sensitization with TNCB.

During the experiment, administered Bilberon-25 orally significantly attenuated the TNCB-induced increase in scratching behavior, compared to that of dexamethasone.

Both Bilberon-25 and dexamethasone inhibited the ear swelling.

Additional differentiation of the Bilberon-25 efficacy, the action of the herbal remedy inhibited symptoms of chronic allergic contact dermatitis by alleviating T helper cell type 2 (Th2) predominance of the lesional skin.

Based on the findings, researchers wrote in the final report, "Anthocyanins from bilberry might be beneficial for the treatment of chronic pruritus which can occur in patients with inflammatory skin diseases such as atopic dermatitis".

In order to reveal more information about anthocyanins' activity against chronic allergic contact dermatitis, researchers at Chiba Univ compared the anti-pruritic effect of anthocyanin-rich quality-controlled bilberry extract and anthocyanidin-rich degraded extract in a mouse model of allergic contact dermatitis.

Once again, BALB/c mice with allergic contact dermatitis were induced by 4 weeks of repeated application of 2,4,6-trinitro-1-chlorobenzene (TNCB) followed by administration of Bilberon-25 orally for 4 weeks.

Compared with nonheated Bilberon-25, the proportion of anthocyanins in heated Bilberon-25 decreased, and the proportion of anthocyanidins was increased in a heated-time-dependent manner.

Treatment with non-heated Bilberon-25 significantly attenuated the TNCB-induced increased scratching behavior, whereas treatment with 2 h-heated Bilberon-25 did not.

Particularly, 300 μg/mL nonheated Bilberon-25 showed significant inhibition of degranulation in RBL-2H3 mast cells.

The results suggested that the anthocyanin-rich but not anthocyanidin-rich bilberry extract inhibited skin diseases involving pruritic symptoms, such as chronic allergic contact dermatitis, atopic dermatitis, and rhinitis.

Taken altogether, bilberry-processed abundantly bioactive compound anthocyanoside may be considered a functioning remedy for the treatment of symptoms in patients with chronic allergic contact dermatitis, pending the validation of a larger sample size and multicenter human study.

#Enzyme #Bromelain Suppresses Symptoms of #Osteoarthritis (OA) Compared to Those of #Diclofenac, Researchers Say

Osteoarthritis (OA), a form of arthritis, is a condition associated with aging-induced wear and tear on a joint or joint.
The condition is very common in older adults, affecting over 25 million people in the US with no sign of stopping.

According to the University of Porto Medical School, the differences in prevalence and incidence estimates of osteoarthritis (OA), according to the case definition, are in the knee, hip, and hand joints(3).

The characteristics of osteoarthritis are aching pain(5), stiffness(6), or difficulty in moving the joint or joints(7). The pain usually gets worse in a change of weather, at night.

Today management of osteoarthritis (OA) focuses on pain relief and improved physical function through pharmacological, nonpharmacological, and surgical treatments(4).

Some researchers suggested that OA is not simply a disease related to aging or mechanical stress of joints but rather a “metabolic disorder(70)(71)” interrelated to lipid, metabolic, and humoral mediators(70).

Indeed, OA has been linked not only to obesity(46)(47) but also to other cardiovascular risk factors(72)(73), namely, diabetes(74), dyslipidemia, hypertension, and insulin resistance(75).

More precisely, the majority of patients with OA also suffer from one or more of the below conditions
* Hypertension
According to the University Health Network Research Institute, the economic burden incurred by RA significantly exceeds that related to OA and HBP(76)(77).

* Cardiovascular disease
According to Erasmus University Medical Centre, there is an association of atherosclerosis with osteoarthritis of the knee and hand joints in women(72)(73)(78).

*Peripheral vascular disease
According to Himchan Hospital, patients with osteoarthritis are associated with a risk of asymptomatic peripheral vascular disease(80). The average vessel wall thickness of the popliteal artery was 1.09 mm in patients with generalized OA, and 0.96 mm in the matched normal reference population(79).

* Congestive heart failure
Hypertension, diabetes, and older age have been shown to increase the risk of Congestive heart failure modestly(81).

* Renal function impairment
According to the Southampton University Hospitals NHS Trust, pain pharmacotherapies such as osteoarthritis (OA) or mixed OA and rheumatoid arthritis may increase the risk of adverse events in patients with concurrent cardiovascular (CV) or renal disease(83).

6. Diabetes
Several epidemiological and experimental data support the hypothesis that diabetes could be an independent risk factor for osteoarthritis (OA)(76)(85).

* Respiratory disease
People with respiratory disease are also at a higher risk to develop osteoarthritis (OA), including chronic cough and bilateral infiltrates on chest roentgenogram(86).

* High serum of cholesterol
There is an association between high serum cholesterol levels and both knee and generalized OA, according to the study of 113 females and 133 males with average ages 46 14.2 and 51.54 16.0 years by King Khalid University Hospital(87).

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

On finding a natural compound from healthy food for the treatment of inflammatory diseases, researchers examined the effects of bromelain on patients with osteoarthritis of the knee.

The randomized, double-blind placebo-controlled trial included 47 patients with a confirmed diagnosis of moderate to severe knee OA randomized to 12 weeks of bromelain 800 mg/day or placebo, with a 4-week follow-up.

According to the results from 31 patients who completed the trial, including 14 in the bromelain group and 17 in the placebo group, both treatment groups showed clinically relevant improvement in the WOMAC disability subscale only with a mild adverse outcome.

In order to reveal and confirm the effects of bromelain on OA, a joint randomized, single-blind, active-controlled pilot study. led by Mahidol University was conducted in patients with OA for 16 weeks.
40 knee OA patients enrolled in the study were randomized to receive oral bromelain (500 mg/day) or diclofenac (100 mg/day).

According to the tested assays,
* At week 4, the improvement of total WOMAC and pain subscales from baseline was observed in both groups, however, two patients in the diclofenac treatment group showed adverse effects that lead to discontinuation of diclofenac.

* At week 16, the bromelain treatment group showed a significant improvement in all aspects, including total WOMAC scores (12.2 versus 25.5), pain subscales (2.4 versus 5.6), stiffness subscales (0.8 versus 2.0), and function subscales (9.1 versus 17.9), and physical component of SF-36 (73.3 versus 65.4), compared to baseline values.

* Also compared to the based line, the bromelain-treated group also showed a decrease in plasma MDA and LPS-stimulated PGE2 production,

* Bromelain exerted a similar effect to those of diclofenac after 4 weeks.

Taken altogether, bromelain used alone may be considered supplements for the prevention and treatment of osteoarthritis, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

#Yoga Intervention Is Associated with a Reduced Risk of Complications of Preeclampsia in High Risk #Pregnancy Women, Clinical Trials Suggest

Yoga participation may have profound and strong implications in reduced risk of complications and improvement of pregnancy outcomes in high-risk pregnancies, some studies suggested

Preeclampsia is a medical condition characterized by abnormally high blood pressure during pregnancy which is one major risk factor in interference with pregnancy outcome, negatively.


According to a study of 68 high-risk pregnant women recruited from two maternity hospitals in Bengaluru, India randomly assigned into yoga and control groups with yoga group (n=30) received standard care plus one-hour yoga sessions, three times a week, from the 12th to the 28th week of gestation and the control group (n=38) received standard care plus conventional antenatal exercises (walking) during the same period, women in yoga + standard care showed a significant fewer pregnancy-induced hypertension (PIH)

The risk of pre-eclampsia in the yoga group showed a ratio of 0,042 compared to the control.


Furthermore, high-risk pregnant women who participated in maternal yoga also expressed significantly lower syndromes of preeclampsia, ...... and improved pregnancy outcomes.


In support of the above differentiation, the study of 121 women attending the antenatal clinic at Gunasheela Surgical and Maternity Hospital (GSMH) in Bangalore, India, with abnormal Doppler study of umbilical and uterine arteries and between 18-20 weeks of pregnancy. Out of 121 participants, 68 women were randomly assigned to the yoga group and 53 women to the control group, after taking into account other confounders, researchers suggested that women practiced physical postures, breathing, and meditation showed a strong improvement in the occurrence of complications of pregnancy, including hypertension.


Further analysis also indicated that yoga scores a higher outcome in all aspects including high-risk pregnancy-induced intrauterine growth retardation, pre-term delivery, particularly hypertension, compared to the non-yoga group.

Indeed, maternal and regular yoga exercise improved outcome of high-risk pregnancy was attributed to the function in ameliorated metabolic syndrome risk, which has been found to associate with the development of pre-eclampsia risk and exaggerated pre-eclampsia complications through enhancing the production of antioxidants in inhibited reactive oxygen species in initiated oxidative stress which can cause cellular dysfunction.

Additionally, yoga also has an implication in improved cellular immunomodulating activity, thus reducing the risk of autoimmune diseases caused by overexpression of pro-inflammatory cytokines which play an important role in the control of cell growth, cell proliferation, cell differentiation, and apoptosis.


More interestingly, in the study of a group of women to investigate the correlation between high risk of complications of impaired placentation and inadequate trophoblast invasion in first-trimester uterine artery resistance, randomly assigned 59 high-risk pregnant women into yoga (n = 27) and control (n = 32) groups, scientists showed that yoga group showed significantly higher values in the (28th week) for biparietal diameter, head circumference, femur length, and estimated fetal weight measured by RM-ANOVA assay in compared to control.

Repeated Measures (RM)ANOVA is a technique used to test the equality of means in comparison to three or more groups, with the same number of participants.

Additionally, researchers also indicated that women in the yoga group demonstrated a strong improvement of the resistance index in the right uterine artery, umbilical artery, and fetal middle cerebral artery and lower impedance.

In fact, the improved blood circulation, strengthen fetus artery, and prevented initiated pre-eclampsia and pre-eclampsia complications of regular yoga practice showed a significantly reduced risk of impaired placentation and cytotrophoblastic stem cell differentiation into extravillous cytotrophoblasts (EVCT) with invasive properties.


According to statistics, 7% of impaired placentation and inadequate trophoblast invasion in first-trimester uterine artery resistance were associated with the condition of pre-eclampsia and pre-eclampsia complications in high-risk pregnant women.

Taking it all together, there is no doubt that yoga may be considered an adjunct therapy used combined with standard treatment in the attenuated risk of complications and improved favorable pregnancy outcomes in high-risk pregnant women.
