#Enzyme #Bromelain In the Inhibiting the Risks of Invasive #Glioma Cells, According to Scientists

A glioma is a type of brain and spinal cord tumor characterized by the alternation of DNA of the glial cells.
Believe it or not, gliomas have been found to account for 30 percent of all brain tumors and central nervous system tumors, and 80 percent of all malignant brain tumors.

Glioma can be classified into 3 types, including
* Astrocytomas are found in glial cells, star-shaped brain cells in the cerebrum called astrocytes. Most astrocytomas are localized.

* Ependymomas are cancers that grow in your brain or any part of the spine, including your neck and upper and lower back starting from the ependymal cells in the middle of your spinal cord and fluid-filled spaces in your brain.

* Oligodendrogliomas are cancers that begin on the oligodendrocytes of the brain or from a glial precursor cell.

The exact cause is unknown. Epidemiologically, age, exposure to radiation, and family history are common risk factors that cause the onset of the disease,

Out of many prevalent factors, exposure to ionizing radiation has been found in many patients with glioma.

Dr. Maricruz Rivera, the lead scientist wrote, "Glioblastoma (GBM) is the most common primary malignant brain tumor in adults with a median survival of 12–15 months with treatment consisting of surgical resection followed by ionizing radiation (IR) and chemotherapy".

And, "While IR provides the benefit of improved survival, it paradoxically promotes a selection of more malignant cellular phenotypes of GBM".


Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a potential compound for the treatment of invasive glioma cells, researchers investigated bromelain, an aqueous extract from pineapple stem against anti-invasive gliomas.

According to the tested assays, bromelain significantly and reversibly reduced glioma cell adhesion, migration, and invasion without affecting cell viability.

In other words, bromelain inhibited cancer cell proliferation and spreading without inducing cell apoptosis.

Levels of alpha 3 beta 1 integrin associated with glioma cell spreading and CD44 protein associated with cancer cell progression were also inhibited by the administration of bromelain without affecting the gene expression.

In other words, the metabolic pathways for de novo nucleotide synthesis in cancer growth were greatly attenuated by bromelain.

Moreover, according to the transactivating signaling assay, bromelain inhibited the invasive glioma cell and was also attributed to the suppression of CRE-mediated signaling processes in stimulating tumor expansion.

Taken altogether, bromelain may be considered a supplement for the prevention and treatment of invasive glioma cells. , pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Inflammation is the natural response of the immune system macrophages, the first line of defense in stimulating the production of inflammatory cytokines to protect our body against the infection caused by foreign microorganisms during the acute phase of body damage and injury.

Truly, the main focus of inflammation is to repair damage and injury.

Most cases of infection associated with inflammation are stopped at the acute phase. The wound or injury is slowly recovered.

Any inflammation that lasts more than 3 months is considered chronic inflammation.

Chronic inflammation is the result of the immune system's inability to kill off all the microorganisms that participated in the acute phase of infection, leading to the forming of autoimmune and chronic inflammatory diseases such as RA and diabetes.

Inflammation also can be caused by overexpression of free radicals compared to the antioxidant enzymes in the body. This result can lead to an imbalance of the ratio of the levels of free radicals and antioxidants, the major cause of oxidative stress.

Airway inflammation is a lung inflammatory caused by both allergens and immune mistakenly attacking the respiratory tissues.

Chronic airway disease predominantly asthma and chronic obstructive pulmonary disease (COPD) have increased at an alarming rate in the US over the past 20 years.

According to the statistics provided by the WHO, globally, approximately 65 million people suffer from chronic obstructive pulmonary disease (COPD), causing the death of 3 million each year. and 334 million people suffer from asthma, affecting 14% of all children globally.

In the US, the prevalence of chronic airway disease was found to cause a major financial burden to society, with both direct and indirect costs.

Neutrophilic airway inflammation is a severe condition that plays a role in the progression of persistent airflow limitation in asthma and raises the possibility that this progression and the development of COPD.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of respiratory diseases, researchers examined the effect of sulforaphane, an active compound of broccoli anti-airway inflammation

The study included 16 subjects that were randomized, and fifteen subjects completed all visits.

According to the tested analysis, 3 days of broccoli sprout homogenate (BSH) significantly increased levels of SFN (p = 0.001) and its major metabolites. However, the BSH supplementation group showed no significant difference in sputum neutrophilia compared to the placebo.

Despite significantly increased plasma levels of SFN in the BSH group, post-O3 gene expression of NRF2 and phase II antioxidant defense genes in NECs and peripheral blood were not significantly different from placebo.

In other words, SFN with broccoli sprout homogenate in healthy human subjects did not induce the expression of antioxidant genes, NRF2.

Furthermore, BSH-induced high levels of SFN do not protect against neutrophilic airway inflammation in an ozone-exposure model.

Based on the results, researchers wrote, " These contradictory results may be due to different systemic levels of SFN achieved with dietary supplementation with BSH, which is not standardized"

And "However, because variability exists in the timing and methods of detection for SFN conjugate levels, it is difficult to compare systemic SFN levels across studies".

In other words, SFN protected the integrity of the neurons against neuron damage through its anti-neuroinflammatory and neuroprotective activities.

Taken altogether, broccoli sprouts processed abundantly SFN may not be considered supplements for the prevention of neutrophilic airway inflammation, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Eggplant may have a potential and substantial effect in reduced risk and treatment of hypercholesterolemia, some scientists suggested.

Eggplant is a species of S. melongena with deep purple color, belongings to the family Solanaceae and native to India, cultivated in southern and eastern Asia since prehistory.

Hypercholesterolemia is a common genetic disorder of cholesterol metabolism characterized by abnormal high low-density lipoprotein cholesterol (LDL-C) levels, one of the leading causes of cardiovascular disease.

Hyperlipidemia is a chronic condition of abnormally high levels of blood cholesterol.

According to statistics, approximately 78 million U.S. adults (nearly 37%) had abnormally high blood cholesterol levels in the US in 2012.


In the investigation of the blood lipid effect of four eggplant varieties on lipid indices in 108 adult albino rats for 4 weeks (30 days), assigned to 2 major groups (fed with 10% and 20% eggplant supplementation) with four subgroups (Groups A-D) and remainder rat assigned to a control group fed with commercial feed, at the end of the experiment, researchers found that all groups fed with eggplant supplement regardless to doses exhibit significantly lower blood cholesterol in compared to the control group.


Further analysis indicated that the administration of diet supplemented at a dose of 10% and 20% of S. aetheopicum and S. gilo in the rat group showed a highest of 9.92 and 9.53 and lowest of 7.65 and 5.95g weight gain respectively.

Levels of cholesterol concentration of (60.76±1.7mg/dl) also was found significantly lower in the 20% S. gilo supplemented diet rat group in compared to control and levels of HDL concentration significantly increased in rats placed on 10% and 20% S. macrocarpon (oval) and S. gilo supplemented diet respectively relative to the control group.

The results finding indicated that all 4 varieties of eggplant exerted s strong effect in displayed weight reduction and improved hypolipidemic levels.

Although Solanum gilo and S. macrocarpon (oval) displayed higher hypocholesterolemia than the other 2 groups, overall, all groups increased HDL and reduced weight gain in tested subjects.


Dr. OSSAMULU, the lead author, after taking into account other confounders said, "(The findings suggested that eggplant) can be employed in the management of overweight and several cardiovascular diseases".

In the study to test the efficacy of dried powdered fruits of eggplant (Solanum melongena) used in traditional medicine in Brazil for the treatment of human hyperlipidemia, researchers at the joint study lead by the Instituto de Endocrinologia de Maringá conducted a double-blind placebo-controlled study to test the effectiveness of oral Solanum melongena (SM) in a total of 41 hyperlipidemic volunteers allocated to active treatment (n= 21, received two capsules containing SM (450 mg) or placebo (n= 20, 450 mg) twice a day for 3 months, indicated that there was no significant difference of levels of serum triglycerides, HDL-c, VLDL-c, AST, ALT, gGT, glucose, and body mass index, in the group treated with SM in compared to placebo.

And both groups expressed the same levels of decreased levels of serum total cholesterol, LDL-c, and LDL-c/HDL-c.


The result of the findings prompted Dr. Silvard Fruits), to require further clinical trials before being recommended to treat hyperlipidemia".

Contrast to the above study, oral application of infusion of a powdered preparation of the eggplant fruit of 38 hypercholesterolemic human volunteers, receiving either S. Melongena infusion (N = 19) or placebo (N = 19) participated in two clinical experiments, exhibited a strong effect in significantly reduced the blood levels of total and LDL cholesterol and of apolipoprotein B in compared to control, according to the Intraindividual analysis.

However, after taking into account dietary orientation, researchers suggested that no intra- or intergroup differences were seen for any of the parameters analyzed.

Dr. Guimarães PR, the lead scientist at the Instituto de Ciências Biológicas said, " S. melongena infusion had a modest and transitory effect, which was not different from that obtained with standard orientation for dyslipidemia patients (diet and physical activities)".

Taking it all together, although there is no universal confirmation, eggplant may have potential and therapeutic effects in reduced risk and treatment of hyperlipidemia pending further investigation.

Cytotoxicity is a process or action that destroys living cells, through activating the production of pro-inflammatory cytokines and proteins associated with cell apoptosis and programmed cell death.
Free radical cytotoxicity is the ability of ROS in exerting oxidative stress in the cancer cells, leading to apoptosis.

Cancer is a class of diseases characterized by irregular cell growth due to DNA alternation.

Most cancers do not cause any symptoms at an early stage. However, at the later stage, a larger size of tumor can press on the nearby blood vessels and nerve cells, causing localized bleeding and severe pain.

The most common symptoms of cancer are unintended weight loss, loss of appetite, fatigue or tiredness, and gastrointestinal discomforts and symptoms depending on the location of the cancer.


If you experience some of the above symptoms, please check with your doctor to rule out the p[ossibility of the disease.

The causes of cancer are unknown. Researchers can not explain why people with the same health conditions, and family history in the same family, are susceptible to cancer, while others do not.

In normal cells, overproduction of free radicals or oxidative stress can affect the cell lipid, and protein, leading to cell damage or alteration of cell DNA.

Most cases of natural remedies initiating the cytotoxic effects against cancer without causing nearby healthy cell death probably are associated with the stimulation of the production of antioxidants produced by the host and the presence of antioxidants in the remedy.


Bilberry is a species of low-growing shrub in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The herbal plant has been used as foods and herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, psoriasis, diabetes, etc.


Researchers on finding a natural compound with cytotoxic potency examined the bilberry chemical compound in facilitating the onset of cytotoxicity against cancer and neutralizing the neuro cytotoxicity against the formation of plague on K562 Human Erythroleukemic cancer cells and Alzheimer's diseases (AD), respectively.

GroES self-assembled into amyloid fibrils similar to those observed in various neurodegenerative diseases was found to induce cell toxicity.

Before the application of bilberry anthocyanins, injection of soluble intermediate aggregates of GroES exhibited neuronal cell death, rather than forming mature fibrils.

Pretreatment with anthocyanins isolated from bilberry, not only inhibited both fibril formation but also neutralized the toxicity of intermediates.

The findings strongly suggested anthocyanins process an inhibitory effect on the amyloid fibril formation of various conformational disease-causing proteins.

Oppositely, from a cancer perspective, anthocyanins stimulated the tumor suppressor genes to induce apoptosis in cancer cells.


Dr. Seth P. Thibado and colleagues in the study to enhance the chemical compound bioavailability, observed the cytotoxicity of anthocyanins in an encapsulated in 5.5 nm diameter liposomal micelles, called NutraNanoSpheres (NNS), at a concentration of 2.5 mg/50 µl [25% (w/w) anthocyanins].


Bilberry NNS induced the apoptotic/cytotoxic effects on K562 Human Erythroleukemic cancer cells.

The concentrations of bilberry demonstrated the greatest levels of percentage inhibition of 60–70% at 0.018–1.14 mg/ml (n=6) and 60% inhibition at 4 mg/ml, compared to the control.

And, the lowest percentage of inhibition (30%) occurred at 2 mg/ml. The lethal dose 50 was determined to be 0.01–0.04 mg/ml of Bilberry per 105 K562 cells at 72 h of cell culture exposure.

At 48 h incubation, the highest percentage of inhibition was only 27%,.

The findings clearly suggested that NutraNanoSpheres (NNS) demonstrates direct cytotoxic effects, at a concentration that is 8–40 times lower than the levels required for bilberry if not encapsulated.

Taken altogether, bilberry-processed abundantly bioactive ingredient anthocyanins may be considered a functional remedy in the modulation of the cytotoxic activity against cancer, pending the confirmation of a large sample size and multicenter human study.

NADPH oxidases, a multisubunit enzyme comprising membrane and cytosolic components plays a critical role to protect our body against pathogenic microorganism by maintaining the immune function, cell growth, and apoptosis.

NOX family of NADPH oxidases deficiency can cause immunosuppression, leading to the overexpression of intracellular ROS that results in various lethal diseases.

Epidemiologically, diseases associated with the NOX family of NADPH oxidases deficiency-induced immunosuppression are frequent and recurrent pneumonia, bronchitis, sinus infections, ear infections, meningitis or skin infections, inflammation, and infection of internal organs, blood disorders, such as low platelet counts or anemia, digestive problems, such as cramping, loss of appetite, nausea and diarrhea and delayed growth and development, particularly in cancer developing.

The NOX family of NADPH oxidases overexpression caused by a defect of its different subunits produced high levels of ROS in the gastrointestinal tract that causes chronic inflammatory stress; cytokine induction, leading to the formation of digestive cancer such as colorectal cancer.

A recent joint study led by the National Cancer Institute suggested that lycopene may be used for the treatment of cancer that targets the expression of NADPH oxidase 4-5 (NOX4-5).

Cancer is a class of diseases caused by cell growth irregularly and uncontrollably due to the alternation of cell DNA. Over time, cancer cells induced tumor formation to become progressively worse, leading to secondary metastasis.

Lycopene is a phytochemical in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas.

Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

In patients with chronic pancreatitis, overexpression of free radicals has been found to orchestrate pancreatic cancer.

Furthermore, NOX4 overexpression increased the pre-malignant fibrotic states that lead to carcinomas of the lung and liver.

High expression of NOX5 is also found in patients with malignant melanomas, prostate cancer, and Barrett's esophagus-associated adenocarcinomas,

NOX4 and 5 are members of the NOX family of NADPH oxidases.

Furthermore, lycopene administration in vitro and Vivo significantly inhibited NOX4 protein expression at 2.5 μM, compared to other concentrations.

The efficacy of lycopene ameliorated the expression of NOX4 protein was attributed to the significant inhibition of the NOX4 mRNA expression associated with the function of NOX in the production of reactive oxygen species.

Dr. Roy K, the lead scientist in the investigation of the risk of cancer in the NOX family of NADPH oxidases wrote, "Over-expression of functional NOX proteins in many tissues helps to explain tissue injury and DNA damage from ROS that accompany pre-malignant conditions, as well as elucidating the potential mechanisms of NOX-related damage that contribute to both the initiation and the progression of a wide range of solid and hematopoietic malignancies".

In SK-Hep-1 cells, intracellular ROS levels were also decreased by the injection of lycopene.

Moreover, the levels of NOX4 protein expression, NOX activity, and the intracellular ROS associated with transforming growth factor β (TGF-β)-induced cancer cell metastasis also were eradicated by the activity of lycopene upon injection.

Other mechanisms such as proteins involved in anti-apoptosis and enhanced cancer cell migration, invasion, and adhesion were also inhibited by the expression of lycopene.

Taken altogether, lycopene may be considered a natural remedy for the prevention of cancer cell metastasis associated with the overexpression of NADPH oxidase, pending to the confirmation of larger sample size and multicenter studies.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

The Effects of #Herbal #Ashwagandha Cytostatic Activities on #MultipleMyeloma, Scientists Say

Multiple myeloma (Myeloma) is a type of cancer that originated from plasma cells in the bone marrow.

The disease is a condition of DNA alternation of white blood cells normally responsible for the production of antibodies to protect our body against infection with some characteristics.

According to the statistics provided by the American Cancer Society, multiple myeloma is a relatively uncommon cancer. In the United States, the lifetime risk of getting multiple myeloma is 1 in 132 (0.76%).

Every year, approximately, 32,110 new cases are diagnosed, including 18,130 in men and 13,980 in women. The disease also causes the death of 12,960 Americans including 6,990 in men and 5,970 in women.
The Grades of multiple myeloma are depending on the tendency of spreading. Low-grade cancers usually grow more slowly and are less likely to spread while high-grade cancer indicates otherwise.

The exact causes of MS are unknown. Epidemiologically, the increase of age (If you are between 50 and 70 years old you are at higher risk to develop Multiple Myeloma), gender (men are more likely to develop multiple myeloma than women), race (Cancer tends to affects blacks more often than whites), previous multiple myeloma, exposed to certain chemicals, family history, and radiation
Most common symptoms of MS are* Bone pain
Abnormal growth of plasma cells can cause a collapse of the bone.

* Infection
Overcrowding of abnormal plasma cells reduces the functions of white blood cells produced by the bone marrow, leading to a weakened immune system. The most common infections found to be pneumonia and pyelonephritis.

* Anemia
As a result, malignant plasma cells have to interfere with the normal production of red blood cells.

* Bleeding
Due to the lower production of the platelet.

* Renal failure
Abnormal plasma cells can also affect the function of the kidney in getting rid of waste, causing urea and creatinine to accumulate in the bloodstream.

* Neurological symptoms
Due to hypercalcemia, as a result of the breakdown of the bone caused by the release of calcium into the bloodstream.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.
The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered an anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound for the treatment of tumor of the bone marrow, researchers examined the growth inhibitory effects of withanolide D (WND), a steroidal lactone isolated from Withania somnifera on drug-resistant subpopulation known as cancer stem cells (CSCs).

According to tested assays, WND exhibited similar cytostatic effects by inhibiting cancer cell growth and division. on drug-resistant and drug-sensitive cell lines in the nM range.

In cancer cell lines MM-CSCs and RPMI 8226, the administration of WND also induced cancer cell death and apoptosis.

However, WND did not increase the accumulation of the fluorescent P-gp substrate rhodamine 123 in MM-CSCs associated with cancer cells' drug resistance. More precisely, the result suggested that WND at the tested doses may not inhibit P-gp accumulation involved in the development of resistance of cancer cells to chemotherapeutic drugs.

Based on the findings, researchers said, " the WND-induced cytostatic effect may be independent of P-gp efflux".

Taken altogether, Ashwagandha may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of myeloma, pending the result of larger sample size and multicenter study

#Antioxidant #Carotenoids Inhibit the OxidativeStress that Causes #CysticFibrosis (#CF)

Cystic fibrosis (CF) is the most common fatal genetic disease affecting mostly the lung in Canadian children and young adults, leading to difficulty breathing and coughing up mucus and frequent lung infections.

However, the condition has also been found in the pancreas, liver, kidneys, and intestine.
According to the Cystic Fibrosis Foundation Patient Registry, more than 30,000 people are living with cystic fibrosis in the US.

Believe it or not, most cases of CF are diagnosed by age 2.

The most common symptoms are depending on the location of cystic fibrosis including
sinus infections, poor growth, fatty stool, and clubbing of the fingers and toes.

In male adults, the condition also induces infertility as a result of the blockage or absence of the sperm canal.

Conventionally, as of today, there is no cure for CF. Most cases of CF caused by lung infection are treated intravenously, inhaled, or by mouth.

In severe cases, a lung transplant may be the only option.

Although most cases of CF are associated with a genetic preposition, environment, lifestyle, and age are also 2 main factors that increase the risk of CF.

Cystic Fibrosis Canada in the investigation of the risk of CF in older adults wrote, "Today, the estimated median age of survival of CF patients in Canada is over 50 years, and the majority of patients are over 18 years of age. CF patients enjoy an improved quality of life, pursuing careers, and starting their own families".

Lycopene is a phytochemical found in tomatoes in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

In finding a potential compound for the treatment of lung disease researchers examined the effects of carotenoids, the potent antioxidants on singlet oxygen and other toxic oxygen species in patients with cystic fibrosis (CF).

The study included a total of 17 patients with cystic fibrosis (CF) and 10 normal children,

According to the assessment of plasma levels of four carotenoids, beta-carotene, alpha-carotene, lutein, and lycopene, levels of carotenoids in children with CF were significantly lower compared to regular control participants.

There was no significant difference in total cholesterol and total carotene between groups. Furthermore, there was no correlation between fat absorption or measures of adiposity in children with CF compared to control.

However, levels of selected carotenoids were correlated negatively with serum IgG levels associated with the measurement of inflammation.

More importantly, researchers also found that the differences in plasma carotenoid levels between children with CF and normal children were involved in the rapid turnover of carotenoids, by quenching of toxic oxygen species in inflammatory states of CF.


The results strongly suggested that improving the concentration of carotenoids that reduces the implication of the toxic ROS expression may have an impact on the prevention and treatment of CF.


Intake of lycopene in supplements should be taken with extreme care to prevent overdose acute liver toxicity.
