Ashwagandha, the Anti Bacterial Infection Functional Remedy

By Kyle J. Norton

The bacterial infection is the infection caused by bacteria. Most cases of bacterial infection are treated with antibiotics. However, bacteria are very adaptable, overuse of antibiotics has made many of them resistant to antibiotics.

There are trillions of strains of bacteria in the world, but only a few of these cause diseases in humans. Some of them live inside our gut.

The most common symptoms of bacterial infection are redness and heat, swelling, fever, pain at the site of infection.

In severe cases, the patient may also experience symptoms of swollen lymph glands.

Most cases of bacterial infection can be treated successfully if the types of bacteria that cause the infection are identified.

There are a number of risk factors associated with the onset of bacterial infection, some researchers suggested that people with a weakened immune system may increase the prevalence of bacterial infection.

Dr. van Aalst, the lead scientist wrote, "Patients with a weakened immune system due to immunosuppressive treatment are at increased risk of infection with Streptococcus pneumonia".

And, " in patients treated with immunosuppressive medication and compared to controls, the initial serologic response to the pneumococcal conjugate vaccine (PCV) and pneumococcal polysaccharide vaccine (PPSV) is impaired. Moreover, this impaired response was more profound after PCV than after PPSV".

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound for the prevention and treatment of infectious diseases, researchers examined the antibacterial effects of aqueous and alcoholic extracts of the plant (root as well as leaves) against a range of bacteria.

According to the tested differentiation, the methanolic extract sub-fractionated using various solvents and the butanoic sub-fraction showed maximum inhibitory activity against a spectrum of bacteria including Salmonella typhimurium.

The application of both aqueous and alcoholic extracts is considered safe compared to the synthetic antibiotic (viz. chloramphenicol), as these extracts did not induce lysis on incubation with human erythrocytes.

In salmonellosis in Balb/C mice, extracts isolated from plants (both root and leaves) also demonstrated a strong effect on tested salmonellosis.

Finally, researchers said, " Oral administration of the aqueous extracts successfully obliterated salmonella infection in Balb/C mice as revealed by increased survival rate as well as less bacterial load in various vital organs of the treated animals".

Taken altogether, Ashwagandha may be considered a functional herb for the treatment of bacterial infections, pending to the result of larger sample size and multicenter study

Broccoli Processes Anti-Metastatic Activities Via Innate Immune Cell Activation

By Kyle J. Norton

The immune system is biological structures of an organism that protects us against disease by identifying and killing pathogens, including bacteria, viruses, parasites, etc. and tumor cells, including irregular cell growth, cancer cells.

In most cases, the immune system does a great job to keep us healthy and preventing infections. But sometimes problems with the immune system can lead to illness and infection.

However, during the process of their functions, they may induce the production of free radicals and a chain of free radicals that can be harmful to our bodies. including the production of pro-inflammatory cytokines in the acute phase of infection.

In other words, through the expression of the lines of defense, depending on the stage of pathogenic invasion, the innate immune response protects the body via a different set of interactions.

In the acute phase of infection, the macrophages in the immune system stimulate the production of inflammatory cytokines to the site of the damage.

The inflammatory cytokines not only play a critical role in stopping the stimuli from entering into the body but also speeds up the wound healing through growth factors.

The innate immune system also can destroy the damaged cells such as cancer cells through the T cell activation of B cells which in turn stimulates the production of a protein associated with the production of antibody p53, a tumor suppressor.

Some researchers suggested immune system also can kill cancer cells by reacting to activating cytokines (IFN-α/β, IL-12, IL-15, IL-18, IL-21) produced by tumor cells or by other immune cells stimulated by tumor cells.

Metastasis is the process to induce the development of secondary malignant growths at a distance away from primary cancer.

Metastatic cancer also shares the characteristics of primary cancer. Breast cancer that spreads to the lung is called metastatic breast cancer, not lung cancer.

According to statistics, in 2018, approximately over1,700,000 cases of cancer were diagnosed worldwide. The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Believe it or not, some metastatic cancer may have an unknown primary origin or CUP, secondary cancer without a primary cancer site.

Broccoli is a mustard/cabbage plant, belong to the family Brassicaceae. The veggie has large flower heads, usually green in color and the mass of flower heads surrounded by leaves and evolved from a wild cabbage plant on the continent of Europe.

On finding a potential plant that processes anticarcinogenic activity, researchers examined the anti-metastatic activities of polysaccharides in broccoli, purified polysaccharides (BCE-I, -II, and -III) isolated by fractionation of broccoli enzyme extracts and subsequent ethanol precipitation.

More precisely, BCE-I mainly consisted of galactose and arabinose, whereas BCE-II mainly consisted of galacturonic acid and rhamnose, and BCE-III mainly consisted of rhamnose and galactose.

According to the results of the analysis, compared to other compounds, BCE-I showed the greatest enhancement of tumor necrosis factor-α, interleukin (IL)-12, and IL-6 secretion in the induction of cytotoxicity on stimulation of murine peritoneal macrophages.

Furthermore, intravenous (i.v.) administration of BCE-I enhanced the lethal activity of natural killer (NK) cells on YAC-1 tumor cells significantly and dose-dependently in an ex vivo experiment.

Moreover, Ii an experimental model using lung metastasis of Colon26-M3.1 carcinoma cells, prophylactic i.v., oral administration of BCE-I significantly and dose-dependently inhibited lung metastatic activity.

Based on the evidence, researchers said, "These results indicated that BCE-I has potent antitumor metastatic activity and that its anti-metastatic activity has relevance to the stimulation of NK and other immune cells".

Taken altogether,  broccoli processes anti-metastatic activities via innate immune cell activation, pending to the confirmation of the larger sample size and multicenter human study.

Cranberry Modulates the Expression of Oxidative Stress

By Kyle J. Norton

Oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissues.

Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

There is no single cause of oxidative stress. Some researchers suggested that a number of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Sadly, a prolonged period of oxidative stress presented in the human body can cause oxidative damage to biomolecules, (lipids, proteins, DNA) and the death of healthy cells through cytotoxicity.

Certain chronic diseases are found to be associated with oxidative stress, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid and cell damage.

Antioxidants can either produced by the liver or intake from dietary sources.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of diseases associated with oxidative stress, researchers investigated the effects of cranberry polyphenolic fractions in oxidative stress (OxS), on intestinal Caco-2/15 cells.

According to the tested analysis, cranberry extracts pre-treatment in Caco-2/15 cells not only showed a strong activity iron/ascorbate lipid peroxidation but also inhibited lipopolysaccharide in the mediation of the production of inflammation in pro-inflammatory cytokines such as TNF-α and interleukin-6.

Furthermore, cranberry extracts pre-treatment also inhibited the inflammatory symptoms such as pain associated with the overexpression of enzyme cyclo-oxygenase-2 and hormone prostaglandin E2.

Moreover, Cranberry polyphenols (CP) fractions regulated the levels of antioxidant enzymes Nrf2 and levels of a pro-inflammatory cytokine such as nuclear factor κB activation.

Taken altogether, cranberry may be considered a remedy for the management of oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Oleanolic Acid, A Promising Remedy for the Treatment of Chronic Diseases

By Kyle J. Norton

Chronic diseases are medical conditions that last more than 3 months, depending on the type of diseases, including arthritis, cardiovascular diseases such as heart attacks and stroke, cancer, diabetes, epilepsy and seizures, and obesity.

The exact causes of chronic diseases are not identified. Aging, unhealthy lifestyle such as excessive alcohol drinking smoking, poor diet, lack of physical activity are the most common risk factors associated with the onset of the disease.

According to the statistics, chronic diseases affect millions of people worldwide, leading to the death of 35 million people in 2005, including many young people and those in middle age.

According to the joint study led by Fordham University, chronic diseases are among the most prevalent and costly health conditions in the US, affecting over 133 million of the US population as many of them suffer from at least one chronic disease and the number is growing in the rapid rate.

Some chronic diseases such as cancer have been found to be the leading causes of hospitalization, long-term disability, reduced quality of life, and death.

In other words, chronic diseases generally cannot be prevented or cured by medication, nor do they just disappear.

In an inflammatory perspective, chronic inflammatory diseases are the results of the immune system response inability to kill off all infectious pathogens during the acute phase of infection, leading to the immune adaptation to the new change.

In some cases, chronic diseases also are caused by the immune system mistakenly attacking the bodily tissue, leading to low-grade inflammation such as rheumatoid arthritis.

Oleanolic acid is a phytochemical in the subclass of Triterpenoid, belonging to the group of Terpenes found abundantly in honey mesquite, garlic, java apple, clove, etc.

On finding a potential plant that processes anti-inflammatory activity researchers examined the potential of oleanolic acid (OA) in the prevention and management of chronic diseases.

According to the results of the analysis of other studies,
* OA exerted a therapeutic effect on different chronic diseases and symptoms.

* Oleanolic acid also serves as a framework for the development of novel semi-synthetic triterpenoids for finding therapeutic modalities for various ailments.

* Synthesis of chemical derivatives of OA to enhance its solubility, bioavailability, and potency.
Collectively, researchers said, "This review suggests that oleanolic acid and its derivatives are important candidates in the search for alternative therapy in the treatment and management of chronic diseases".

Taken altogether, oleanolic acid may be considered a remedy for the prevention and treatment of chronic diseases, pending to the confirmation of the larger sample size and multicenter human study.

Cruciferous Vegetables Reduce the Risk of Diabetic Nephropathy in Diabetes

By Kyle J. Norton

Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream to regulate the glucose.

In other words, diabetes occurs when cells in the pancreas dying off or receptor sites clogged up by fat and cholesterol. In some cases, diabetes is also caused by allergic reactions of cells in the immune system.

There are 2 major types of diabetes
* Type 1 diabetes, also known as juvenile diabetes or insulin-dependent diabetes, the most common form diabetes found in children is a condition in which the pancreas does not produce any insulin. The most common form of type 1 diabetes is caused by genetic mutation and some viral infection.

* Type 2 diabetes is a medical condition characterized by insufficient insulin entering the bloodstream to regulate the glucose due to either cell in pancreas dying off or receptor sites clogged up by fat and cholesterol.

Diabetic nephropathy (diabetic kidney disease) is a complication of kidney damage caused by un uncontrolled diabetes.

Long-term unmanaged hyperglycemia can damage the part of the kidneys thus reducing the kidney function in filter waste from the blood filters, leading to 'leaky' syndrome that allows protein into your urine.

Diabetic nephropathy is the 3 stages of kidney change in patients with diabetes. In this stage, patients may experience symptoms of abnormally elevated urinary albumin excretion, compared to the 15 and 300 μg/min in the baseline situation.

The progression of diabetic nephropathy that leads to an increased rate in albumin excretion is higher in patients with increased blood pressure.

More importantly, if high blood pressure is left untreated, diabetic nephropathy can induce persistent proteinuria >0.5 g/ 24 h that furtherly declines the renal function with the mean fall rate being around 1 ml/min/mo.

In other words, by treating hypertension and hyperglycemia in patients with diabetes, the progression of diabetics kidney disease may be slowed or preventable.

Cruciferous vegetables are a group of vegetables, belongings to the family Brassicaceae (also called Cruciferae) with many genera, species, and cultivated all over the world in suitable climate for commercial profits, including cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts, and green leafy vegetables.

On finding a potential remedy for reducing diabetics complications, researchers investigated the effects of 3,3'-diindolylmethane (DIM)-a natural compound produced from indole-3-carbinol, found in cruciferous vegetables on diabetic nephropathy.

Selected mice included in the study were induced diabetes by streptozotocin (STZ)-induced hyperglycemia.

According to the experimental results,
* DIM decreased STZ-increased high blood glucose levels and food and water intake in diabetic mice.

* DIM also improved diabetic nephropathy by inhibiting the expression of PKC-α, the marker of albuminuria and kidney damage, and TGF-β1, an indicator of renal hypertrophy, in diabetic mice.

Based on the evidence, researchers said, " DIM may ameliorate hyperglycemia and diabetic nephropathy through the inhibition of PKC-α and TGF-β1 signaling".

Taken altogether, cruciferous vegetables may be considered functional remedies for the prevention of hyperglycemia-induced diabetic nephropathy, pending to the confirmation of the larger sample size and multicenter human study.

Angelica sinensis Processes Antiplatelet Therapy

By Kyle J. Norton

Antiplatelet activity is a process that prevents and treats blood clots.

Platelets are tiny blood cells that protect the other to prevent injury and infection by forming clots to stop bleeding.

Platelets are made in the bone marrow that also produces white and red blood cells.

Platelet aggregation refers to the platelet-to-platelet adhesion in the blood that forms a clot.

Platelet aggregation plays a critical role in the protection of the body against blood loss due to injury or tissue damage.

However, activated platelets that stimulate blood clot formation have also been found to induce atherothrombotic disease.

Antiplatelet aggregation medicines, also known as a platelet agglutination inhibitor are the types of medicine that process blood thinning property.

In other words, antiplatelet aggregation medicines reduce the risk of blood clotting used in patients who have a high risk of cardiovascular disease such as aspirin.

Coagulation disorders or bleeding disorders are medical conditions that affect blood clotting activities.

The most common diseases associated with coagulation disorders are Hemophilia, Von Willebrand disease, clotting factor deficiencies, hypercoagulable states, and deep venous thrombosis.

The most common causes of bleeding disorders are found in patients with a lower red blood cell count and vitamin K deficiency.

However, long-term use of certain medications has also been found in the side effects of bleeding disorder.

On finding a natural ingredient for the prevention and treatment of cardiovascular diseases, scientists examined the antiplatelet activity of 4 traditional Chinese medicine (TCM) herbs effective for prevention cardiovascular diseases, namely Radix Salvia Miltiorrhiza (Red sage root, Danshen), Radix Pueraria Lobata (Kudzu root, Gegen), Radix Angelica Sinensis (Angelica root, Danggui), and Rhizoma Ligusticum chuanxiong (Szechuan lovage rhizome, Chuanxiong).

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

Selected Sprague-Dawley rats treatment groups which were received a bolus oral dose of DAPT on day 1 followed by DAPT for consecutive 13 days in absence and presence of orally co-administered four TCM herbs (Danshen, Gegen, Danggui and Chuanxiong) at their low and high doses.

According to the experimental results
* Co-administration of Gegen and Danggui significantly altered the pharmacokinetics of ASA and CLP in DAPT with increased systemic exposure of ASA and CLP respectively.

* All co-administered herbs, significant inhibition on rCyp2c11 and carboxylesterase activities associated with the hydrolysis of some drugs to inactive metabolites, particularly, for DAPT with Danshen, Gegen and Danggui co-treatment.

* All the treatments did not cause detectable changes in COX (associated with inflammation_ and P2Y12 mRNA (associated with platelet formation and protein expressions.

Based on the results, researchers suggest, "Among the four studied TCMs, it was demonstrated that co-administration of Gegen and Danggui could lead to altered pharmacokinetics of DAPT with significant inhibition on rCyp2c11 and carboxylesterase activities. Although Gegen, Danggui, and Chuanxiong might potentially offset the anticoagulant activity of DAPT, the overall pharmacodynamics outcome was not considered to be harmful due to a lack of risk in bleeding".

Taken altogether, Dandelion may be considered an anti-platelet activity, pending to the confirmation of the larger sample size and multicenter human study.

Ursolic acid Ameliorates the Risk of Muscle Atrophy in Vivo

By Kyle J. Norton

Muscle atrophy is a condition associated with a lack of physical activity, inducing loss of muscle mass due to the muscles weakening and shrinking.

A neuron or nerve cell made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral nerves that play an essential role to process and transmit information.

The function of neurons can be classified into 3 types
* The sensory neurons found within the nervous system that convert external stimuli from the organism's environment into internal electrical impulses.

*The motor neurons transmit impulses from the spinal cord to skeletal and smooth muscles,. In other words, motor neurons directly control all of the muscle movements.

* The interneurons connect the motor and sensory neurons. In other words, the interneurons transfer signals between sensory and motor neurons.

The most common diseases associated with neuron dysfunction are amyotrophic lateral sclerosis (ALS). Lou Gehrig's disease or classical motor neuron disease that disrupts signals to all voluntary muscles on both sides of the body.

The most common disease involved in muscle atrophy is amyotrophic lateral sclerosis (ALS), also known as Lou Gehrig's disease.

Amyotrophic lateral sclerosis (ALS) is a type of dementia associated with neurodegeneration syndrome among elders, affecting memory, thinking, orientation, comprehension, calculation, learning capacity, language, and judgment.

Over 47 million of worldwide populations are affected by the disease, mostly in the West.

Ursolic acid is a phytochemical in the subclass of Triterpenoid, belongings to the group of Terpenes found abundantly in apples, basil, bilberries, cranberries, peppermint, lavender, oregano, hawthorn, prunes., etc.

On finding a potential phytochemical for the treatment of neurodegenerative diseases, researchers examined the ursolic acid (UA) effects on the alleviation of the deleterious effect of skeletal muscle atrophy on bone health.

According to the analysis of the immobilization (IM) method used on the left hindlimb of Sprague Dawley (SD) rats for 10 days to induce muscle atrophy, divided into two groups, SED (sedentary, n=6) and UA (n=6) to demonstrate the effect of UA on atrophic skeletal muscles.
* Before UA treatment, muscle atrophy was induced and bone mineral density (BMD) decreased significantly in experimental rats.

* The 8-week UA treatment significantly increased the gastrocnemius muscle mass compared to the SED group.

*  UA treatment showed negative results of a decrease in BMD, trabecular bone volume fraction, and trabecular number, and an increase in trabecular separation in the SED group.

* No significant difference was observed in atrophic hindlimbs between SED and UA groups.

After talking about other factors into account, researchers said, "These results alone are insufficient to suggest that UA is a potential resistance exercise mimetic for atrophic skeletal muscle and weakened bone. However, this study will help determine the potential of UA as a resistance exercise mimetic".

Taken altogether, ursolic acid,  may have an effect on muscle atrophy, pending to the confirmation of the larger sample size and multicenter human study.

Intake of ursolic acid in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
