Lycopene Protects the Cerebral Vessels and Neurons Induced by Hyperlipidemia

By Kyle J. Norton

The liver is the largest internal organ located in the upper right portion of the abdomen, beneath your diaphragm and above your stomach.

The liver not only plays a critical role to filter the blood coming from the intestine before passing them to other parts of the body but also protects our body against toxin by detoxifying chemicals and metabolizes drugs.

Cholesterol is a waxy substance produced by the liver. In a moderate amount, it helps to improve the gastrointestinal tract in food digestion, builds strong cell membranes, produces vitamin D and steroid hormones.

The healthy ratio of low-density lipoprotein (LDL)/high-density lipoprotein (HDL) is ant numbers that are less than 4.

Hyperlipidemia is a medical condition of abnormally high levels of cholesterol and fat in the bloodstream.

High blood cholesterol is the most common cause of hyperlipidemia. According to statistics provided by the CDC, more than 12% of adults age 20 and older are living with high blood cholesterol with a total cholesterol higher than 240 mg/dL. and more than 18% had high-density lipoprotein (HDL, or “good”) cholesterol levels less than 40 mg/dL.

In other words, hyperlipidemia has a high prevalent ratio that affects over 100 million Americans and the condition is increasing at an alarming rate due to overweight and obesity.

Cerebral vessels or brain vessels include anterior and posterior segments that supply blood to the brain.

A neuron or nerve cell made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral nerves that play an essential role to process and transmit information.

Tomato is red, edible fruit, genus Solanum, belonging to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

On finding a potential compound for the treatment of diseases associated with the vessel and neuron damage, researchers examined the effects of lycopene isolated from tomato on vessel and neuron damage in the brain against hyperlipidemia.

Rats selected for the study were fed with hypercholesterolemic feed and treated with lycopene orally by gavage at the dose of 5, 25, 45, 65, 85, and 105, 125 mg/kg/bw-1/d-1 for 4 weeks.

Based on the results at the end of the experiment, lycopene doses and serum lycopene concentration showed an inverse U-shape curve.

In other words, long term intake of lycopene increases the serum of lycopene in the blood plasma linearly.

Serum lycopene concentration was negatively correlated with the levels of serum TC, TG, LDL-C, and cerebral LDL-C and pro-inflammatory cytokines in the brain such as VEGF, and VCAM-1.

Furthermore, the serum lycopene concentration was positively correlated with the expression of Claudin-5 associated with the tight junctions that provide one mechanism of cell to cell adhesion and the number of neurons in hippocampal CA1 and CA3.

In other words, long-term intake of lycopene protects the vessels and neurons against the pathologic change caused by hyperlipidemia.

Collectively, researchers said, "These results suggested an inverse U-shape relation between dose and serum concentration of lycopene, and intermediate doses were most effective to protect cerebral vessels and neurons from being damaged by hyperlipidemia.".

Taken altogether, lycopene found abundantly may be considered supplements for the prevention and treatment of hyperlipidemia induced cerebral vessel and neuron impairment, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Cranberry in the Promotion of Metabolic Health

By Kyle J. Norton

Metabolic health refers to ideal levels of blood sugar, triglycerides, high-density lipoprotein (HDL) cholesterol, blood pressure, and waist circumference, without using medications.

Metabolic syndrome is a cluster of conditions associated with the increased risk of cardiovascular disease and diabetes.

The health problems found in the group include too much fat around the waist, elevated blood pressure, high triglycerides, elevated blood sugar, and low HDL cholesterol.

In other words, if you have 3 of the conditions in the group, your risk of CVD and diabetes are increased substantially, compared to those who have less.

According to the statistics, approximately 30% of people in the US are living with metabolic syndrome.

Believe it or not, among US adults aged 18 years or older, the prevalence of metabolic syndrome rose by more than 35% from 1988–1994 to 2007–2012, increasing from 25.3% to 34.2%.

Metabolic dysfunction is a condition caused by any interruption of the aforementioned processes.
In other words, metabolic dysfunction induces the production of excess or a reduced amount of substances needed to stay healthy.

A metabolic disorder can be caused by genetic proposition induced inborn errors of metabolism, or acquired by an individual in the lifetime

There are many risk factors associated with the onset of metabolic dysfunction depending on the types of disruption such as abnormal blood glucose metabolism induced type 2 diabetes.

Diseases associated with metabolic dysfunction can be moderate or severe, depending on the organs involved, including liver or respiratory failure, cancer, chronic obstructive pulmonary disease (COPD, includes emphysema and chronic bronchitis), and HIV/AIDS.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the promotion of metabolic health, researchers evaluated the effects of isomalto-oligosaccharides (IMOs; sweet-tasting non-digestible oligosaccharides) with cranberry extract (CRX) on a high fat diet (HFD)-induced metabolic alterations in a mice model.

More precisely, Male Swiss albino mice included in the study were fed normal chow or HFD (58% fat kcal) and were administered either CRX (200 mg/kg) alone or in combination with IMOs (1 g/kg).

Based on the tested analysis, the co-supplementation of CRX and IMOs showed a significant improvement of cecal short-chain fatty acids, SCFAs, especially butyrate-producing bacteria (clostridial cluster XIVa bacteria) and butyrate kinase expression in HFD-fed mice.

In other words, the co-supplementation of CRX and IMOs promotes the growth of the population of good bacteria in balancing the ratio of gut bacteria.

Furthermore, compared to individual compounds, the combination also significantly improved gut beneficial bacterial abundance, gut histology and related changes in the tested mice.

More precisely, the combination not only prevented HFD-induced systemic and tissue inflammation, glucose intolerance but also reduced the systemic obesity-associated metabolic changes in adipose tissue and liver.

The combination of CRX and IMOs appeared more effective in the prevention of HFD-induced gut derangements.

Collectively, researchers said, "Combination of CRX and IMOs could be advantageous for normalization of metabolic alterations seen in diet-induced obesity via beneficial modulation of gastrointestinal health".

Taken altogether, cranberry combined with other herbal medicine may be considered a remedy for the management of metabolic health, pending to the confirmation of the larger sample size and multicenter human study.

Broccoli Exerted a Significant Pulmonary Injury Protection

By Kyle J. Norton

The lung is one of the vital organs in humans located near the backbone on either side of the heart with functions of inhaling oxygen from the air then transporting them to the bloodstream to nourish the body cells' need and exhaling carbon dioxide from the bloodstream.

Lung diseases are medical conditions, affecting the upper respiratory tract, trachea, bronchi, bronchioles, alveoli, pleura and pleural cavity, and the nerves and muscles of breathing.

People with lung diseases have breathing problems, preventing them to get enough oxygen for the body's needs.

According to the statistic from the World Health Organization, respiratory diseases are leading causes of death and disability in the world. Chronic obstructive pulmonary disease (COPD) and asthma are 2 of the most common respiratory disease in the world, affecting 65 million and 334 million people, respectively.

3 million people die from chronic obstructive pulmonary disease (COPD) each year, making it the third leading cause of death worldwide. And 14% of 334 million people suffer from asthma are children.

Long-term smoking is considered one of the leading risk factors of lung diseases. Other risk factors associated with the onset of lung diseases include radon, asbestos, second-hand smoke, and air pollution.

According to Dr. Elizabeth R. Johnson, the lead scientists in the study of "Acute Lung Injury: Epidemiology, Pathogenesis, and Treatment", wrote, "Acute lung injury (ALI) remains a significant source of morbidity and mortality in the critically ill patient population"

And, " Defined by a constellation of clinical criteria (acute onset of bilateral pulmonary infiltrates with hypoxemia without evidence of hydrostatic pulmonary edema), ALI has a high incidence (200,000 per year in the US) and overall mortality remains high".

Recently, some researchers suggested, " E-cigarette, or vaping, product use associated lung injury (EVALI, also called vaping associated pulmonary injury [VAPI]), initially described in 2019, is an acute or subacute respiratory illness that can be severe and life-threatening".

Broccoli is a mustard/cabbage plant, belongs to the family Brassicaceae. The veggie has large flower heads, usually green in color and the mass of flower heads surrounded by leaves and evolved from a wild cabbage plant on the continent of Europe.

On finding a potential plant for the treatment of lung diseases, researchers examined the effects of Sulforaphane (SFN) is a phytochemical antioxidant on the pulmonary injury.

In wild-type (Nrf2+/+) and Nrf2-deficient (Nrf2-/-) mice induced hyperoxia or air exposure-induced pulmonary injury and oxidation indices, pretreatment with SFN orally or as standardized broccoli sprout extract diet (SBE), SFN or SBE upregulated a large cluster of basal lung genes involved in mitochondrial oxidative phosphorylation, energy metabolism, and cardiovascular protection only in Nrf2+/+ mice.in Nrf2+/+ mice but not in Nrf2-/- mice by elucidating the ARE genes.

However, pretreatment of SFN showed no significant activity against the transcript abundance of the mitochondrial machinery genes in hyperoxia exposure in SFN-treated Nrf2+/+ mice, compared to SFN-treated Nrf2-/- mice.

Furthermore, SFN also exerted a strong effect on a central molecule in transcriptome networks associated with anti-cell apoptosis.

In other words, SFN can only exert a minor improvement of hyperoxia-caused lung histopathology and neutrophilia in Nrf2-/- mice compared to Nrf2+/+.

Based on the evidence, researchers said, ", SFN is suggested to be a preventive intervention in a preclinical model of acute lung injury by linking mitochondria and Nrf2. Administration of SFN alleviated acute lung injury-like pathogenesis in an Nrf2-dependent manner".

Taken altogether,  broccoli may be used as a pulmonary injury protection functional food, pending to the confirmation of the larger sample size and multicenter human study.

Tocopherols Process Cytotoxicity Against Cancer Cells

By Kyle J. Norton

Cytotoxicity is the process in the induction of cell death through toxins.

In other words, cell cytotoxicity is the process in which certain chemicals or mediator cells are used to destroy living cells.

In cancers, cytotoxicity can be benefits to humans by destroying the cancer cells and preventing them from proliferation.

Most cases of cytotoxicity used by a substance are to determine the potential toxicity of the quality of the substance, including plant extracts or biologically active compounds isolated from plants.

But, before the preparation can be used in humans, bioassays associated with a number of different cell lines in vitro and Vivo have been used to assess the cytotoxicity of many herbal plants, including those of African medicinal plants.

In other words, the aim of the cytotoxic studies is to find the minimal quality that is essential for the successful development of pharmaceutical or cosmetic preparation that induces cellular toxicity for the treatment of cancer.

Some of the cancer drugs with cytotoxic activity have been found to induce cancer cell death accompanied by serious health problems, including weakened immune functioning such as low blood counts cause an increased possibility of developing infection or anemia, tiredness, mouth soreness, nausea, vomiting, loss of appetites, constipation or diarrhea, skin changes or reactions. particularly hair loss.

However, the combination used of chemo drug and herbal medicine and plant-based antioxidants that can also stimulate the  ROS and antioxidants production to induce cytotoxicity to the tumors can also cause cancer cell death without damage to nearby healthy cells.

For example, cytotoxicity induced by oxidative stress due to the imbalanced ratio of free radicals and antioxidant enzymes produced by the body can cause protein, lipid and cellular damage that leads to chronic low-grade inflammatory diseases and cancer.

Tocopherols are phytochemicals of which many have vitamin E activity, belongings to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of cancer, researchers examined the cytotoxicity of tocopherols and their quinones in drug-sensitive and multidrug-resistant leukemia cells.

According to the tested analysis,
* Among tocopherols, only delta-T exhibited cytotoxicity.

* Among para quinones, alpha-TQ showed no cytotoxicity, while gamma-TQ and delta-TQ were highly cytotoxic in both CEM and CEM/VLB100 cell lines (LD50 < 10 muM).

* Delta-TQ and gamma-TQ were more cytotoxic than the widely studied chemotherapeutic agent doxorubicin, which also showed selective cytotoxicity to CEM cells.

* Compared to doxorubicin, the orthoquinone Tocored was less cytotoxic than doxorubicin in drug-sensitive cells but more cytotoxic than doxorubicin in multidrug-resistant cells.

* Cytotoxicity was not a function of the phytyl side-chain since both TMCQ and PR were cytotoxic in leukemia cells.

* Cytotoxicity was enhanced when the glutathione pool was depleted by preincubation with buthionine-[S,R]-sulfoximine, but cytotoxicity was diminished by the addition of N-acetylcysteine to cultures. alpha-T also diminished the cytotoxicity of para- and orthoquinones.

Collectively, researchers said, "tocopheryl quinones represent a new class of alkylating electrophilic quinones that function as highly cytotoxic agents and escape multidrug resistance in acute lymphoblastic leukemia cell lines".

Taken altogether, tocopherols may process cytotoxic activity against cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Ashwagandha Reduces the Neuons' Damage After Cerebral Infarction

By Kyle J. Norton

Cardiovascular disease is a chronic medical condition characterized by narrowed or blocked blood vessels that can lead to a heart attack including chest pain (angina) or stroke.

Stroke is a result of a blockage in the blood vessels (ischemic stroke) or bleeding (hemorrhagic stroke) that are associated with ischemic stroke, transient ischemic attack, or non-traumatic intracranial hemorrhage.

According to the statistics, stroke is the leading cause of serious, long-term disability in the United States. Every year, almost 795,000 Americans suffer a stroke, including 600,000 of first attacks, and 185,000 of recurrent attacks.

Nearly three-quarters of all strokes occur in people over the age of 65.

Brain ischemia is a condition caused by insufficient blood flow to the brain to meet metabolic demand, leading to irreversible brain damage in severe cases.

In other words, brain ischemia-induced the death of neurons of the affected tissues.

A neuron or nerve cell made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral nerves that play an essential role to process and transmit information.

Brain ischemia can be classified into
* Focal ischemia is a condition caused by damage to a specific region of the brain; and

* Global ischemia is a condition of damage that affects the wide areas of brain tissue, leading to symptoms of impairments in vision, body movement, and speaking.

A cerebral infarction is an area of the brain caused by a blockage or narrowing in the arteries supplying blood and oxygen to the brain.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

In the urgency to discover a natural ingredient for the treatment of stroke, researchers examined the effects of Withaferin A (WA), a steroidal lactone derived from the plant Withania somnifera, on experimentally induced cerebral infarction.

According to the results of the experiment,
WA treatment (25, 50 or 100mg/kg body weight) significantly reduced the infarct area in a carotid ligation model.

Furthermore, the application of WA reduced intimal hyperplasia and proliferating cell nuclear antigen (PCNA) associated with cell replication by suppressing PI3K/Akt signaling involved neuron's apoptosis following cerebral ischemia/reperfusion injury.

Moreover, besides inhibiting the expression of proteins associated with brain cell apoptosis following cerebral ischemia/reperfusion injury, WA also elevated the genes that suppress the production of apoptotic cytokines and the promotion of cell cycle division.

More importantly, in vitro, WA exposure severely downregulated matrix metalloproteinases (MMP)-2 and -9 that play a critical role in the degradation of the components of the extracellular matrix and inhibited migration of DGF-stimulated A7r5 cells and A7r5 cells proliferation significantly.

Based on the evidence, researchers said, WA exerted neuroprotective effects by activating the PI3K/Akt pathway, modulating the expression of MMPs, and inhibiting the migration of VSMCs.".

Taken altogether, Ashwagandha may be considered a neuroprotective remedy, pending to the confirmation of the larger sample size and multicenter human study.

Cabbage Protects the Neuron Integrity Against Neurodegenerative Diseases

By Kyle J. Norton

Neuroprotection an action involved the use of a natural substance to protect the integrity of neurons against damage or death.

In other words, neuroprotection is the process to protect brain functioning.

Neurodegenerative diseases are a class of conditions associated with the progressive loss of neurons in the brain caused by several mechanisms, including toxicity and oxygen deficiency.

The most common diseases associated with neurodegenerative diseases are dementia, a cluster of conditions included Alzheimer’s disease, leading to a negative influence on the body's functioning, including balance, movement, talking, breathing, and heart function.

Dementia is a neuropsychiatric disorder as a result of cognitive impairment and behavioral disturbances. The behavioral and psychological symptoms of dementia (BPSD) are common, contributing to caregiver burden and premature institutionalization.

The true symptoms of dementia are a progressive loss of memory and other mental abilities that can cause impairment of a person's ability to perform usual tasks in everyday life.

According to the statistics, about 5-8% of all people over the age of 65 have some form of dementia, and this number doubles every five years above that age.

Cabbage is a species of Brassica oleracea belongings to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem of which is crowded a mass of leaves, usually green but in some varieties, it may be red or purplish.

With an aim to find a natural food for the treatment of age-related diseases, researchers examined the effect of cabbage consumption in the risks of neurodegenerative disease.

According to the tested differentiation,  isothiocyanates (ITCs), the sulfur- and nitrogen-containing phytochemicals found in cabbage reduced the risk for developing neurodegenerative diseases (NDDs) and some other chronic disorders including but not limited to cardiovascular diseases, various forms of cancer and inflammatory disorders among others.

Furthermore, Sulforaphane(SFN), derived from glucoraphanin EXERTED its ability not only arrest many target proteins but also to control various metabolic pathways in neuronal cells by targeting amyloid β (Aβ)-induced oxidative damage and apoptosis in a human neuroblastoma cell line (SH-SY5Y).

Moreover, SFN inhibited the progression of Parkinson’s disease (PD) through the interception of pro-inflammatory and apoptotic pathways in vivo, induced by intrastriatal injection of 6-hydroxydopamine (6-OHDA) for 28 days.

Additionally, in the examing the chemical compound moringin (MG) isolated from the leaf extracts of cabbage, researchers found that the administration in different doses improved spatial memory and reduced neuronal degeneration to the minimum level in the hippocampal region of the animal disease model.

Based on the results, researchers wrote, "This review summarizes the most significant and up-to-date in vitro and in vivo neuroprotective actions of sulforaphane (SFN), moringin (MG), .... in neurodegenerative diseases".

Taken altogether, cabbage processed abundantly bioactive compounds may be considered a remedy for the prevention and treatment of neurodegenerative diseases, pending to the confirmation of the larger sample size and multicenter human study.

Omega -3 Fatty Acids Might Block the Onset of Breast Cancer

By Kyle J. Norton

The woman breast is made up of specialized tissue that produces milk (glandular tissue) and fatty tissue. The size of a woman's breast is totally determined by the amount of fat.

Cancer is a class of chronic condition characterized by irregular cell growth associated with the alternation of cell DNA.

Breast cancer is a medical condition characterized by cell growth irregularly and disorderly in the breast tissue.

Most cases of breast cancer start in the cell on the surface of the inner lining of the breast tissue before spreading into deeper layers of the breast.

In the early stage, most patients with breast cancer may experience lumps on the breast or surrounding the breast.

However, breast skin irritation or dimpling, nipple pain, nipple retraction (turning inward), redness and scaliness that cannot be healed within 3 weeks are also the signs and symptoms of breast cancer.

At the later stage, after colonizing most parts of the breast, the breast cancer cells may travel a distance far the way to infect other healthy cells and tissue.

According to the statistics, the lifetime risk of invasive breast cancer developing is 12%.

In 2019, approximately 268,600 new cases of invasive breast cancer will be diagnosed in the U.S., along with 62,930 new cases of non-invasive (in situ) breast cancer.

Just about 20% of breast cancers are diagnosed in women under 50, while almost 30% are diagnosed in women over 70 years of age.

The 5 years survival rate of breast cancer diagnosed at the early stage is closed to 100%.

Epidemiologically, there are several factors that can reduce the risk of breast cancer including breastfeeding, early age at first full-term pregnancy, an increasing number of births, preeclampsia, longer duration of breastfeeding.

The stages of breast cancer can be classified depending on
* The size of the cancer
* Invasive or not
* Whether cancer has spread to the lymph nodes
* Whether cancer has spread to the distance parts or organs of the body.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of malignant tumors, researchers compared the effect of DHA, Delta-T3, and DHA plus Delta-T3 co-treatment on triple-negative breast cancer cell line MDA-MB-231.

Based on the results of cells treated by 50 μM DHA and/or 5 μM Delta-T3,
* DHA can trigger an increase in lipid droplet (LD) biogenesis and co-treatment with Delta-T3 was able to reduce this LD biogenesis.

Please note that a higher cytoplasmic LD content is associated with a higher breast cancer cell malignancy and proliferation.

* The inhibition of the cytoplasmic LD content by silencing ADRP (adipose differentiation-related protein), also decreased cell proliferation in MDA-MB-231 cells.

* Treatment with DHA and Delta-T3 alone or co-treatment did not reduce cell viability

* More importantly, DHA can trigger lipophagy associated with the autophagic degradation of lipid droplets in MDA-MB-231 cells compared to DHA plus Delta-T3 co-treatment that was able to enhance this lipophagy process.

Based on the findings, researchers said, " co-treatment with DHA plus Delta-T3 in MDA-MB-231 cells could reduce LD biogenesis and potentiate lipophagy in these cells, possibly having a positive impact to inhibit breast cancer malignancy".

Taken altogether, Omega-3 fatty acids together with Delta-T3 vitamin E isoform may be considered a supplementation for the prevention and treatment of breast cancer,  pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
