Broccoli' Sulforaphane (SFN), a Natural Chemo-Preventive Phytochemical

Cancer is the class of diseases associated with the growth of the cells disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

Compared to other treatments, chemotherapy targets cancer's specific genes, proteins, or the tissue environment that contributes to cancer growth and survival.

Chemo drugs have been found to improve the survival rate of cancer patients. However, they also induce significant side effects.

The most common side effects of chemo-drugs are fatigue, hair loss, easy bruising, and bleeding. infection, anemia (low red blood cell counts) nausea and vomiting, appetite changes and constipation.

The side effects of chemo drugs are the results of damage to the healthy blood-forming cells in the bone marrow, hair follicles, and cells in the mouth, digestive tract, and reproductive system.

According to the American Cancer Society, some chemo drugs have also been found to damage cells in the heart, kidneys, bladder, lungs, and nervous system.

Chemoprevention is the use of a medication, vitamin or supplement to prevent the initiation of cancer.

On finding a potential plant for the treatment of cancers, researchers compared the effects of
Brassica family of cruciferous vegetables such as broccoli is a rich source of isothiocyanates, sulforaphane (SFN) as chemopreventive phytochemical.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

Obesity is a chronic and medical condition caused by excess body fat accumulated to some extent that impacts personal health. The hallmark of obesity is attenuated metabolism and structural cell changes in the obese fat observed by reducing metabolic activity.

According to the statistics provided by the National Center for Health Statistics (2015-2016), in the US, approximately 39.8% of adults aged 20 and over were obese accompanied by another 31.8% overweight.

Visceral fat is a layer of fat that surrounds the organs. Overexpression of visceral fat has been found to promote insulin resistance, diabetes, heart disease, and inflammation.

In obese, accumulation of visceral fat is predominant in the intra-abdominal cavity.

According to the BMI index, a person is considered overweight, if he/she has a BMI between 25 to 29.9 kg/m2. A BMI of 30 kg/m2 or more is an indication of obese.

Obesity and overweight on the U.S. economy have eclipsed $1.7 trillion, equivalent to 9.3 percent of the nation's gross domestic product, according to a new overweight report on chronic diseases.

Dr. Maximilian Tremmel, the lead scientist in a systematic literature review of the economic burden of obesity, wrote, " almost half of the world’s adult population will be overweight or obese by 2030 if the prevalence continues on the current trend [] and consequently also the costs attributable to obesity will increase"

And, "A useful example for rapidly rising costs attributable to obesity from these selected studies are the two mentioned studies from Germany [,]. The results of the two papers together show that total societal costs in Germany due to obesity have increased from €9.8 million to €12.2 million between 2002 and 2008. Therefore, public health interventions should focus on the prevention of obesity as soon as possible, ideally at a young age".


Description

Cynometra cauliflora is a species of the genus Cynometra, native to Malaysia, and found mainly in northern Peninsular Malaysia.

On finding a potential phytochemical for the prevention and treatment of chronic diseases, researchers examined the anti-obesity and lipid-lowering effects of ethanolic extract of C. cauliflora leaves and its major compound (vitexin) in C57BL/6 obese mice induced by high-fat diet (HFD).

The study included male C57BL/6 mice induced obese by feeding with HFD (60% fat) for 16 weeks followed by the treatment that starts during the last 8 weeks of HFD feeding with C. cauliflora leaf extract at 200 and 400 mg/kg/day, orlistat (10 mg/kg) and vitexin (10 mg/kg).

According to the chemical analysis,
* The oral administration of C. cauliflora (400 and 200 mg/kg) and vitexin significantly reduced body weight, adipose tissue and liver weight and lipid accumulation in the liver compared to control HFD group.

* Both doses of C. cauliflora also significantly (P ≤ 0.05) decreased serum triglyceride, LDL, lipase, IL-6, peptide YY, resistin levels, hyperglycemia, hyperinsulinemia, and hyperleptinemia compared to the control HFD group.

* C. cauliflora significantly up-regulated the expression of adiponectin, Glut4, Mtor, IRS-1 and InsR genes associated with the sustaining weight loss and significantly decreased the expression of Lepr in white adipose tissue involved in stimulating weight gain.

Based on the findings, researchers said, "This study shows interesting outcomes of C. cauliflora against HFD-induced obesity and associated metabolic abnormalities".

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of bacterial infectious diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Cystic fibrosis (CF) is the most common fatal genetic disease affecting mostly the lung in children and young adults, leading to difficulty breathing and coughing up mucus due to frequent lung infections.

However, the condition has also been found in the pancreas, liver, kidneys, and intestine.

According to the Cystic Fibrosis Foundation Patient Registry, more than 30,000 people are living with cystic fibrosis in the US.

Believe it or not, most cases of CF are diagnosed by age 2.

The most common symptoms are depending on the location of cystic fibrosis including
sinus infections, poor growth, fatty stool, clubbing of the fingers and toes.

In male adults, the condition also induces infertility as a result of the blockage or absence of the sperm canal.

Conventionally, as of today, there is no cure for CF. Most cases of CF caused by lung infection are treated intravenously, inhaled, or by mouth.

In severe cases, a lung transplant may be the only option.

Although most cases of CF are associated with a genetic preposition, environment, lifestyle, and age are also 2 main factors that increase the risk of CF.

According to statistics provided by the Canadian Cystic Fibrosis Foundation, approximately 1 in every 3,600 children born in Canada has CF.

In age perspective,

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

In the urgency to discover a natural ingredient for the treatment of diseases associated with genetic disorders, researchers examined the effects of Withaferin A (WFA), a steroidal lactone from the plant Withania Somnifera L. Dunal on cystic fibrosis.

According to the tested analysis, WFA blocks platelet-activating factor (PAF)-induced activation of NFκB in the induction of inflammation through the innate immune cells and inflammatory T cells.

More precisely, WFA inhibited the NFκB activation that leads to a reduction of the increased expression of IL-8 and NFκB subunit p65 translocation involved in inflammation.

In other words, WFA demonstrated anti-inflammatory activity in the cystic fibrosis model.

Based on the evidence, researchers said, "the airways of CF patients can be specifically targeted for delivery of therapeutics, we propose that WFA should be further studied as an anti-inflammatory agent in models of CF-related inflammation mediated by NFκB".

Cardiovascular diseases refer to heart diseases and stroke, affecting millions of people in the world.
According to statistic, over 17.7 million people died from CVDs in 2015, Hyperlipidaemia and hypertension are the risk factors of the diseases.

In other words, stroke can be caused by either ischemia, due to lack of blood flow or hemorrhagic, due to bleeding.

A stroke is a medical emergency. Early intervention is most cases can minimize brain damage and potential complications.

Not all strokes can lead to permanent brain damage. Some people with a temporary disruption of blood flow to the brain do not result in permanent damage.

Besides aging, smoking, medical conditions such as unmanaged high blood pressure and cholesterol and diabetes, obesity, physical inactivity are some of the prevalent factors found in stroke patients.

There is a general belief that stroke is an accumulation over a long period of the unhealthy diet with red meat and processed meats in the induction of artery-clogging.

Some researchers suggested that increased intake of fruits and vegetables such as carrot accompanied with good fats such as salmon, tuna, and mackerel may have a substantial effect in reduced inflammation in the arteries, improved blood flow and decreased the chance of blood clots.

Sadly, stroke is one of the most common causes of death worldwide.

The study included aged (18-month-old) mice which were administered n-3 PUFA-enriched fish oil in daily chow for 3 months before and up to 8 weeks after 45 minutes of transient middle cerebral artery occlusion (tMCAO).

According to the results of differentiation,
* Mice receiving dietary supplementation of n-3 PUFAs for 3 months showed significant increases in brain ratio of n-3/n-6 PUFA contents.

* Furthermore, the administration of dietary supplementation of n-3 PUFAs for 3 months also markedly reduced long-term sensorimotor deficits and chronic ischemic brain tissue loss after tMCAO.

* Moreover, n-3 PUFAs robustly promoted post-ischemic angiogenesis and neurogenesis to enhance the brian neuron activity and enhanced white matter integrity after tMCAO.

* Most importantly, the enhancement of neurogenesis and white matter integrity were both correlated positively with the improvement of sensorimotor activities after tMCAO.

Collectively, researchers said, "prophylactic dietary supplementation of n-3 PUFAs effectively improves long-term stroke outcomes in aged mice, perhaps by promoting post-stroke brain repair processes such as angiogenesis, neurogenesis, and white matter restoration".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the improvement of long-term stroke outcomes, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Cruciferous Vegetables Gobbles Up Melanoma Cells in Vitro

Skin diseases are conditions of skin damage and classified according to each pathogenic cause.

Most cases of skin disease are involved in allergens, irritants. However certain medical conditions such as irritable bowel disease, autoimmune diseases, and genetic preposition can also cause r skin diseases.

Recent studies suggested that a poor diet is also associated with the onset of skin diseases such as the dermatological disease may be linked to systemic disease, and dietary changes may affect health outcomes, as in psoriasis.

Skin melanoma is a skin cancer characterized by the alternation of the pigment-containing cells known as melanocytes. However, in rare cases, melanoma can also occur in the mouth, intestines, or eye.

Most cases of melanoma start in melanocytes on skin before spreading to the nearby tissue. In the advanced stage, the cancer cells can also travel a distance away from the originated site to infect other tissues and organs, leading to secondary metastasis, through the circulation of blood and fluids.

The most common sites of melanoma metastasis are the lung and breast.

According to the statistic, melanoma skin cancer is increasing and anticipated to continue to rise. In the United States, approximately 70,230 new cases of melanoma were diagnosed, causing the death of 8,790.

The causes of melanoma skin cancer are associated with the alternation of the pigment cells in the skin. Why some people are susceptible to the onset of the disease while others do not even share similar health conditions, family history, and ethnicity.

However, according to the epidemiological studies, genetic mutation inherited from the parent, exposure to the environmental toxins and sun UV rays are the most common associated with the increased risk of melanoma skin cancer. 

On finding a potential plant that processes anti-cancer activity researchers investigated the effects of indole-3-carbinol (I3C), a natural anti-cancer phytochemical derived from cruciferous vegetables such as cabbage and broccoli, as a new chemical scaffold of small molecule enzymatic inhibitors of NEDD4-1 on human cancer cell lines.

Where NEDD4-1 is a proto-oncogenic ubiquitin ligase for PTEN, a tumor suppressor that plays a critical regulator for multiple cellular processes. The gene is mutated or deleted frequently in various human cancers.

According to the tested assays,
*  I3C's stable synthetic derivative 1-benzyl-I3C showed to directly inhibit NEDD4-1 ubiquitination activity.

* Furthermore, compared to I3C, which has an IC50 of 284μM, 1-benzyl-I3C was a significantly more potent NEDD4-1 enzymatic inhibitor with an IC50 of 12.3μM.

* The compounds 2242 and 2243, (the two indolecarbinol analogs with added methyl groups) result in a more nucleophilic benzene ring π system, further enhanced potency with IC50s of 2.71μM and 7.59μM, respectively.

* Most importantly, the indolecarbinol compounds inhibited human melanoma cell proliferation in a manner that generally correlated with their effectiveness as NEDD4-1 enzymatic inhibitors.


Based on the findings, researchers said, " I3C analogs represent a novel set of anti-cancer compounds for the treatment of human melanomas and other cancers that express indolecarbinol-sensitive target enzymes".


Taken altogether, cruciferous vegetables consisting of indole-3-carbinol (I3C) and five novel synthetic analogs may be considered a functional food for the prevention and treatment of melanoma, pending to the confirmation of the larger sample size and multicenter human study.
