Beta-Sitosterol Protects the Pancreatic Islets functioning Against the Risk of Type 2 Diabetes

By Kyle J. Norton

The pancreas is an oblong flattened gland in the digestive and endocrine system located in the abdomen. By secreting pancreatic juice to the small intestine, the pancreas helps to break down carbohydrates, proteins, and lipids.

Traditional Chinese medicine views the pancreas, a vital organ is responsible for the absorption of foods' qi and nutrients before passing them to other organs to nourish our body.

More precisely, the functions of the pancreas including the
* Production of enzymes digest fat, protein, and
carbohydrates.

* Production of juices combined with juices from the intestines to break down protein, carbohydrates and fat, therefore, it plays an important role in maintaining good health.

*  Production of insulin, which is important in regulating the amount of sugar or glucose in the blood.

Pancreatic islets are groups of hormones producing cells, including insulin and glucagon, that work together to maintain the proper level of sugar (glucose) in the blood.

Profound distortion of islet organization in the pancreas has been found to induce account abnormal secretory behavior of the glucagon-secreting A cells in insulin-dependent juvenile-onset diabetes.

Type 1 diabetes, also known as juvenile diabetes or insulin-dependent diabetes, the most common form of diabetes found in children is a condition in which the pancreas does not produce any insulin. The most common form of type 1 diabetes is caused by genetic mutation and some viruses.

As of today, there is no cure for type 1 diabetes. Treatment is focusing on managing blood sugar levels with insulin injection, diet, and lifestyle change.

According to statistics, diabetes is one of the most common diseases in the US. Approximately 10% or 29.1 million people in the United States have diabetes, accompanied by 8.1 million that are undiagnosed and unaware of their condition.

About 1.4 million new cases of diabetes are diagnosed in the United States every year. More than one in every 10 adults who are 20 years or older has diabetes.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of infectious diseases, researchers elucidated the anti-diabetic effects of the extract of A. tenuifolia underground parts and β-sitosterol on rat pancreatic islets.

According to the results of the analysis,
* The root extract of the plant and compound β-sitosterol isolated from the root extracts showed moderate α-amylase inhibitory activity.

* Furthermore, the herb root extract was able to increase the secretion of insulin in high concentration (10 mg/mL) and improved the cell viability with no toxicity in all doses.

* The herbal extract could reduce the levels of reactive oxygen species (ROS) and lipid peroxidation (LPO) associated with the onset of oxidative stress in orchestrating damage to the pancreatic islets.

* Moreover, the plant extract also significantly decreased the enzyme activity associated with pro-apoptotic expression by increasing the antioxidant capacity of the isolated cells.

In other words, all compound found in the extract work synergistically to protect the pancreatic islets by inhibiting all parameters involved in pancreatic damage, particularly, the compound β-sitosterol

Taken altogether, beta-Sitosterol combined with other phytochemicals found in A. tenuifolia underground parts may be considered a supplement for the protection of pancreatic islets induced diabetes, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Dandelion(Taraxacum officinale) Processes Antiproliferative Activity Against Cancer

By Kyle J. Norton

Cell proliferation is a radical growth of cells to replace the damage or injury through the cell cycle division.

Cell proliferation is a process and a necessary step for wound healing.

All normal cells undergo cell cycle division to replace the old cells with new cells through a limited cell division.

On the other hand, cancer cells can multiply definitely.

In cancer, antiproliferation is a process in which the cancer cells are killed or limited to grow through many mechanisms, including the use of chemo drugs.

In the immune perspective, anti-proliferation can be carried out by activating the genes associated with cancer suppression, programming cell death,.... and cell cycle arrest.

A suppressive gene, such as pro-apoptotic p53 induces cancer cell death through cell cycle arrest. In other words, the gene inhibits tumor growth by interrupting the cancer cell cycle division.

In the programmed cell death gene, such as BCL-2 induce cell death by either inducing or inhibiting apoptosis. In other words, If cells are no longer needed or damaged, activating an intracellular death program will cause cell death.

In the phytochemicals perspective, anti-proliferation can also be carried out by antioxidants in which they stimulate the free radicals overexpression to induce toxicity to tumor cells without causing harm to the nearby healthy cells.

Some cancers can stimulate the production of a new blood vessel for cancer cells to grow and others can make enzymes that break down normal cells and tissues as they grow.

Dandelion is a herbaceous perennial plant, genus Taraxacum, belongings to the family Asteraceae, native to temperate regions of the world. It has been used in traditional and herbal medicine to treat gout, eczema, acne, gall bladder, kidney, liver and urinary disorders, hypoglycemia, dyspepsia with constipation, edema, blood pressure, and heart weakness, chronic joint, skin diseases, etc.

On finding a natural ingredient for the prevention and treatment of chronic diseases, scientists compare five different common South European species of Taraxacum (T. obovatum (Willd.) DC., T. marginellum H. Lindb., T. hispanicum H. Lindb., T. lambinonii Soest and T. lacistrum Sahlin) antioxidant and anti-proliferative activity.

According to the dissimilarities between species in LC/MS patterns, T. marginellum demonstrated a strong intracellular antioxidant activity and the cytotoxicity by reducing the expression of ROS.

Compared to the other 3 species, T. marginellum showed the most efficient against the induction of free radicals overexpression in vitro.

Furthermore, in free radical theory, compared to T. marginellum, T. obovatum was the best free radical scavenger and a relevant cytotoxic effect was found in T. lacistrum extract over HeLa and HepG2 cell lines.

Taken altogether, Dandelion may be considered an antioxidant remedy that processes antioxidant and anti-proliferative activity, in a type-dependent manner, pending to the confirmation of the larger sample size and multicenter human study.

Cramp Bark(Viburnum opulus) in the Prevention and Treatment of Ureteral Stones

By Kyle J. Norton

The kidneys formed part of the secretary system are two bean-shaped organs located against the back muscles in the upper abdominal area.

The kidneys play a critical role in filtering wastes from the breakdown of active muscle and the food you eat and removing extra water from the blood through urinary secretion.

Furthermore, the kidneys also maintain the fluid in the body by secreting extra water through urinary secretion.

Kidney stones are composed of mineral salts formed in the kidneys. Men account for 80%  kidney stones incidence and are at risk of the formings between 30 and 40 years of age. About 75% of kidney stones are calcium stones.

According to statistics, kidney stones affect over 10% of the world population.

The causes of kidney stones are debatable. However, some researchers suggested that dehydration, food restriction, Rotavirus infection, over intake of calcium and supplements,... and high fat intake are associated with increased risk of forming kidney stones.

Ureteral stones are kidney stones that stuck in one or both ureters (the tubes that carry urine from the kidneys to the bladder), leading to the obstruction of the urinary secretion.

Most people with kidney stones are asymptomatic. However, some may experience symptoms of renal colic,  an excruciating intermittent pain, usually in the flank (the area between the ribs and hip), that spreads across the abdomen, often to the genital area and inner thigh.

The pain tends to come in waves, gradually increasing to peak intensity, then fading, over about 20 to 60 minutes. The pain may radiate down the abdomen toward the groin or testis or vulva.

Other symptoms include nausea and vomiting, restlessness, sweating, and blood in the urine. A person may have an urge to urinate frequently, particularly as the stone passes down the ureter. Chills, fever, and abdominal distention.

Cramp Bark is an ornamental plant, genus Viburnum, belongings to the family Adoxaceae, native to Europe and Asia. T herbal medicine has been used in traditional and herbal medicine as anti-spasmodic, anti-inflammatory, nervine. hypotensive agent and to treat menstrual cramps and pain, afterbirth, postpartum pains, excessive blood loss in periods, bleeding associated with the menopause, etc.

On finding a potential herbal medicine for the prevention and treatment of kidney stones, researchers examined the efficacy of Gilaburu (Viburnum opulus) extract in the treatment of distal ureteral stones <10 mm.

The study included data of a total of 103 patients retrospectively analyzed. divided into two groups: those given V. opulus 1000 mg peroral 3×2 and diclofenac 50 mg peroral on-demand (n=53) and those given only diclofenac sodium 50 mg peroral on-demand (n=50).

According to the expulsion rate assay, group treated with V. opulus 1000 mg peroral 3×2 and diclofenac 50 mg peroral on-demand showed a rate of stone expulsion that is significantly higher of 82% compared to the diclofenac 50 mg peroral on-demand treatment group of 66%.

Furthermore, elapsed time to stone expulsion was significantly shorter in the V. opulus 1000 mg peroral 3×2 and diclofenac 50 mg peroral on-demand of 9±1.8, compared to another group of 14±2.3 days.

Most importantly, the V. opulus group showed no need for additional treatment (URS and ESWL) and analgesic was less requirement compared to another group.

Based on the findings, researchers said, "V. opulus is an herbal treatment alternative that facilitates the passage of ureteral stones <10 mm. Prospective, randomized studies are needed to support these results".

Taken altogether, cramp bark(Viburnum opulus) may be considered a functional remedy for the treatment of ureteral stones, pending to the confirmation of the larger sample size and multicenter human study.

Ashwagandha May Cause Thyrotoxicosis, Study Suggests

By Kyle J. Norton

Thyroid disease is a condition or malfunction of the thyroid. Hyperthyroidism is a condition in which the thyroid gland is overactive and produces too much thyroid hormones. Hypothyroidism is a condition in which the thyroid gland is underactive and produces very little thyroid hormones.

The thyroid gland is an endocrine gland located in the neck, consisting of two lobes connected by an isthmus with functions to secrete thyroid hormones (thyroxine/T4 and triiodothyronine/T3), and calcitonin.

Thyroid hormones play an important role in metabolism by stimulating almost every tissue in the body to produce proteins and increasing the amount of oxygen to the cells.

Abnormal thyroid hormone production can be diagnosed by examining whether the thyroid gland is enlarged or by taking a blood test or/and a radioactive iodine uptake test.

Thyrotoxicosis is a medical condition caused by an excess of thyroid hormone in the body.

More precisely, in thyrotoxicosis, patients experience a low level of thyroid stimulating hormone, TSH in the bloodstream.

The most common treatment of thyrotoxicosis is the use of radioactive iodine and antithyroid medication, following by careful follow-up.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

The Sectie Geneesmiddelenbewaking in Postbus in examining the case of a 32-year-old healthy woman developed thyrotoxicosis induced symptoms chronic fatigue while taking capsules that contained ashwagandha herbal extract suggested
* Taking ashwagandha herbal extract in reduced doses or occasionally do not cause thyrotoxicosis

* However, overdoses of ashwagandha herbal extract can induce clinical symptoms indicative of thyrotoxicosis.

* Discontinued ashwagandha herbal extract can resolve the symptoms spontaneously, according to the laboratory values.

The incidence also has been proved in the animal studies as ashwagandha can increase serum concentrations of thyroid hormones.

Based on the findings, researchers said, "thyrotoxicosis is a potentially serious side effect of ashwagandha".

Taken altogether, Ashwagandha overdose may induce thyrotoxicosis, pending to the result of larger sample size and multicenter study

Turmeric Protects the Cells integrity Against Cancer In Vitro

By Kyle J. Norton

Scientists may have found a bioactive ingredient for the treatment of cancer with no side effects, according to studies.

Cancer is a class of medical diseases caused by the alternation of cell DNA.

Most cases of primary cancer begin in the cells on the surface of the inner lining of tissues. However, at the later stage, the malignant cancer cells can travel a distance away from the original site to infect other healthy tissues and organs, leading to secondary metastasis through blood and fluids circulation.

The 5 years survival of early diagnosed cancer are high, some of them have a statistic of 100% such as breast cancer. However, the actual 5-year survival rate is totally depending on the types of cancer.

According to the statistics provided by the National Cancer Institute, in the US, 1,735,350 new cases of cancer were diagnosed and the disease causes the death of 609,640 people in 2018.

The most common cancer in the US, 2018 are breast cancer followed by lung and bronchus cancer, prostate cancer, colon and rectum cancer.

Believe it or not, over 1/3 of men and women will be diagnosed with cancer at some point during their lifetimes.

Most cancers share the common symptoms of gastrointestinal discomforts, such as nausea vomiting,  fatigue unintended weight loss and loss of appetite accompanied by specific symptoms depending on the types and grade of cancer.

The exact causes of cancer are not identified. However, according to the epidemiological studies most common risk factors associated with cancer onset are aging, lifestyle, family history, and race.

Some researchers suggested that the introduction of Western diet over the past few decades may be one of the major factors that cause the rises of cancer.

The Global News in the examined the risk of cancer in people who follow the Western diet wrote, "Published in the journal JAMA Oncology, the study followed more than 120,000 adults over 26 years and found that inflammation is a potential link between diet and cancer development. Researchers found a 44-per-cent risk increase for men, and 22-per-cent for women who consumed a pro-inflammatory diet (even after adjusting for other factors, such as high body mass index and decreased physical activity)".

In other words, the risk of cancer is many folds higher in people who follow the long-term Western diet.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

In the urgency to find a natural compound for the treatment of cancer with no side effects, researchers evaluated the anticancer activity of Curcuma caesia Roxb. (Black turmeric), used as a spice, food preservative and coloring agent commonly in the Indian subcontinent.

According to the total antioxidant activity (TAO) method, among all the extracts of dried C. caesia rhizome viz- hexane, ethyl acetate, methanol and water, hexane Rhizome Extract (HRE) was found to possess remarkable antioxidant activity (1200mg ascorbic acid equivalent/100g) compared to other extracts.

In MTT assay, HRE exhibited a dose-dependent inhibition of cancer cells, with notable activity in HepG2 cell lines (IC50: 0976µg/mL).

In Western blotting and flow cytometry assay, HRE induces cell arrest at the G2/M phase along with cellular apoptosis by stimulating the production of a protein associated with cell death programming.

According to the analysis of Gas Chromatography-Mass Spectrophotometry (GC-MS) assays, the anti-cancer cells efficacy of hexane Rhizome Extract (HRE) was attributed to over 20 compounds that inhibit the mutation of epidermal growth factor receptor EGFR (1XKK) in the induction of cancer.

Dr. Mukunthan KS, the lead scientist said, " Our results provided significant evidence of the toxicity mechanisms of HRE that may be beneficial for more rational applications of drug discovery for slowing down cancer progression".

Taken altogether, turmeric may be considered a supplement for the prevention and treatment of cancer with on side effects, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Coltsfoot (Tussilago farfara) Protects the Neurons in Focal Cerebral Ischemia in Vivo

By Kyle J. Norton

Acute brain infarction is a severe and medical emergency characterized by the obstruction of the blood supply to the brain, causing the local death of the neurons. Prompt treatment can minimize brain damage and potential complications.

The occurrence and extent of brain infarction are associated with three basic factors, including the site of arterial occlusion, the rapidity of arterial occlusion, and the presence or absence of collateral circulation.

In other words, brain infarction can be classified into myocardial infarction due to plaque rupture and myocardial oxygen supply-demand imbalance secondary to other acute illnesses.

The exact causes of a blood clot to induce acute brain infarction are unknown. However, the trouble with speaking and understanding, confusion, sudden numbness, weakness or paralysis in your face, arm or leg, headache and the trouble with walking are some of the most common symptoms found in patients with post-myocardial infarction.

Depending on the severity of the acute infarction, in serious cases, patients may also experience symptoms, paralysis of the face, arm or leg, trouble with seeing in one or both eyes.

Although there are many factors associated with the risk of acute infarction, some researchers suggested an unhealthy diet may have a strong and negative implication on the onset of the condition.

Dr. Guo J, the lead scientist at the Peking Union Medical College & Chinese Academy of Medical Sciences wrote, "Some dietary patterns are risk factors for acute myocardial infarction (AMI)".
And, "Several dietary frequencies (serves per week) including vegetables, fruits and tofu were closely associated with a decrease of AMI risk. Carbohydrate pattern showed a weak relationship with AMI. We observed a U-shaped association between frequencies of fat and protein pattern and AMI risk. Excessive fat intake increased the AMI risk.".

Coltsfoot(Tussilago farfara) is a perennial herbaceous plant, genus Tussilago, belonging to the family Asteraceae, native to Europe and Asia. The herb has been used over thousands of year to treat asthma, various coughs, bronchial congestion, respiratory disorders, headaches and obstruction in the nasal passage, etc.

On finding a potential compound for the treatment of brain ischemia, researchers examined the effects of flower buds extract of Tussilago farfara Linné (Farfarae Flos; FF)on brain ischemia was induced in Sprague-Dawley rats by a transient middle cerebral artery occlusion (tMCAO) for 90 min and reperfusion for 24 h.

Twenty rats included in the study were randomly divided into 4 groups (n=5 per group): normal, tMCAO-induced ischemic control, tMCAO plus FF extract 300 mg/kg-treated, and tMCAO plus MK-801 1 mg/kg-treated as reference drug. FF extract (300 mg/kg, p.o.) or MK-801 (1 mg/kg, i.p.) was administered after reperfusion.

According to the results of the differentiation, researchers found that
* FF protects the neurons by observing the rate of neuronal death and the microglia/astrocytes activation in ischemic brains.

* The extract. FF extract also suppressed the productions of the pro-inflammatory cytokines such as
iNOS, TNF-α, and HIF-1α, IL-1β, and IL-6 expression in ischemic brains.

* Furthermore, the FF extract (0.2 and 0.5 mg/mL, P<0.01) and Tussilago 20 and 50 μmol/L, P<0.01) also significantly inhibits the iNOS mRNA and protein expression associated with the production of NO production.

* Moreover, the FF extract and Tussilago significantly suppressed the protein kinase intracellular signaling molecules that are involved in functions including the regulation of cell division functions in a dose-dependent manner.

Based on the findings, researchers said, "FF extract has a neuroprotective effect in ischemic stroke by the decrease of brain infarction, and the inhibition of neuronal death and microglial activation-mediated inflammatory responses".

Taken altogether, coltsfoot may be considered a remedy for the protection of brain cells in the post of focal cerebral ischemia, pending to the confirmation of the larger sample size and multicenter human study.

Comfrey(Symphytum officinale L.). a Anti-inflammatory Herbal Medicine

By Kyle J. Norton

Inflammation is the natural response of the immune system macrophages, the first line of defense in stimulating the production of inflammatory cytokines to protect our body against the infection caused by foreign microorganisms during the acute phase of body damage and injury.

Truly, the main focus of inflammation is to repair damage and injury.

Most cases of infection associated with inflammation are stopped at the acute phase. The wound or injury then is slowly recovered.

However, overexpression of inflammatory cytokines in most cases can cause improper healing with scar formation or the death of nearby cells.

The most common symptoms of acute inflammation occurred with a short period of time such as hours is pain, redness, immobility, swelling, and heat.

Conventionally, depending on the types of acute inflammation, non-steroidal anti-inflammatory drugs (NSAIDs) may be prescribed to alleviate the pain by counteracting an enzyme that contributes to inflammation.

Any inflammation that lasts more than 3 months is considered chronic inflammation.

Chronic inflammation is the result of the immune system's inability to kill off all the microorganisms participated in the acute phase of infection, leading to the forming of autoimmune and chronic inflammatory diseases such as RA and diabetes.

Inflammation also can be caused by overexpression of free radicals compared to the antioxidant enzymes in the body. This result can lead to an imbalance of the ratio of the levels of free radicals and antioxidants, the major cause of oxidative stress.

The chronic inflammation statistics, by Roma Pahwa, Anand Singh, and Ishwarlal Jialal, wrote, "Chronic inflammatory diseases are the most significant cause of death in the world. The World Health Organization (WHO) ranks chronic diseases as the greatest threat to human health. The prevalence of diseases associated with chronic inflammation is anticipated to increase persistently for the next 30 years in the United States. in 2000, nearly 125 million Americans were living with chronic conditions and 61 million (21%) had more than one".

And, "In recent estimates by Rand Corporation, in 2014 nearly 60% of Americans had at least one chronic condition, 42% had more than one and 12% of adults had 5 or more chronic conditions".

Comfrey is a perennial plant in the genus of Symphytum, belonging to the family Boraginaceae, native to Europe. It has been used in herbal and traditional medicine to ease excessive bleeding or on open wounds, relief pain and swollen, treat circulation issues, reduce cholesterol, etc.

On finding a potential compound for the treatment of diseases associated with chronic diseases, researchers compared the efficacy of Symphytum officinale, commonly known as comfrey in chronic inflammation.

According to the study,
* Topical use of comfrey, may process a strong anti analgesic and anti-inflammatory property for the treatment of diseases involved in the overexpression of pro-inflammatory activity.

* Furthermore, a hydroalcoholic extract of comfrey root impairs the development of a pro-inflammatory scenario in primary human endothelial cells in a dose-dependent manner.

* Moreover, the extracts also exhibited anti-inflammatory properties by impairing the pro-inflammatory genes interleukin-1 (IL-1) in activating the expression of pro-inflammatory markers including E-selectin, VCAM1, ICAM1, and COX-2.

* Additionally, the NF-κB, a transcription factor of central importance for the expression of these and other pro-inflammatory genes were also inhibited by the extracts through 2 different stages, including the cytokines IKK1/2 and IκBα degradation.

Taken altogether, Comfrey may be considered a remedy for the treatment of inflammatory diseases, pending to the confirmation of the larger sample size and multicenter human study.
