Oleanolic Acid Gobbles Up Liver Cancer Cells in Vitro

By Kyle J. Norton

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing them to other parts of the body.

The liver also produces cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Liver disease is a class of medical conditions characterized by the reduction partly or completely the function of the liver, including liver failure.

Liver disease is a process of reduced function of the liver acutely or gradually.

Liver cancer is a chronic liver disease associated with irregular cell growth that starts in the cells on the surface of the liver's inner lining tissue, before penetrating into deeper layers to form tumors through cell proliferation.

Liver cancer remains the fifth most common malignancy in men and the eighth in women worldwide.
Hepatocellular carcinoma (HCC) is a common form of primary liver cancer.

According to the Canadian Liver Foundation, approximately one in ten Canadians, or more than 3 million people in Canada has some form of liver disease.

Believe it or not, 95% of deaths from liver disease are due to chronic hepatitis B and C, alcoholic liver disease, non-alcoholic fatty liver disease, and liver cancer, including 25% of obese patients.

The most common signs and symptoms of liver disease are jaundice, abdominal pain, and swelling in the legs and ankles, itchy skin, dark urine color, pale stool color, or bloody or tar-colored stool and tendency to bruise easily.

Primary liver cancer is a medical condition caused by irregular cell growth in the liver tissue as a result of the alternation of cell DNA.

According to the statistics, in 2019, approximately, 42,030 adults (29,480 men and 12,550 women) in the United States will be diagnosed with primary liver cancer.

Believe it or not, since 1980, the incidence of liver cancer has tripled. Between 2006 and 2015, the number of people diagnosed with primary liver cancer is increased by approximately 3% annually.

Oleanolic acid is a phytochemical in the subclass of Triterpenoid, belonging to the group of Terpenes found abundantly in honey mesquite, garlic, java apple, clove, etc.

On finding a potential plant that processes anti-cancer activity researchers examined the anti liver cancer effects of a natural compound oleanolic acid.

The study included human HepG2 liver cancer cells that were treated with various concentrations of oleanolic acid for 24 h.

According to the results of the analysis,
* Oleanolic acid decreased the viability of the liver cancer HepG2 cells and exhibited an IC50 of 30 µM.
* Oleanolic acid exerted very low toxic effects on normal liver cells AML12 cells and exhibited an IC50 of 120 µM.

* Furthermore, oleanolic acid also caused remarkable changes in the morphology of the HepG2 cells and inhibited their colony formation potential.

* Moreover, oleanolic acid triggered G2/M arrest and prompted apoptosis of the HepG2 cells.

* Additionally, oleanolic acid also increased the apoptotic cells from 2.2% in control to around 35% at 30 µM concentration.

* Oleanolic acid also suppressed the migration and invasion of the liver cancer cells.
Collectively, researchers said, "The results of the current research revealed that oleanolic acid can be a molecule that may be utilised in the treatment of liver cancer in the future".

Taken altogether, oleanolic acid may be considered a remedy for the prevention and treatment of liver cancer, pending to the confirmation of the larger sample size and multicenter human study.

Omega-3 Fatty Acids Demonstrates a Neuro-Protective Effect in Vivo

By Kyle J. Norton

The brain formed part of the central nervous system is the most complex organ made up of more than billions of nerve cells or neurons.

The functions of the brain are associated with the communication of trillions of connections called synapses in many specialized regions.

A neuron or nerve cell formed part of the central nervous system, made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral nerves that play an essential role in the process and transmit information.

Neurological diseases are the medical conditions of brain disorder. Anything that involves the structural, biochemical or electrical abnormalities in the brain, spinal cord or other nerves, including brain damage are considered as neuro diseases.

The most common neurodegenerative diseases in the elderly are dementia. Depending on the brain regions that are damaged or dysfunction, dementia can be Parkinson's, Alzheimer's, and Huntington's disease.

Genetic mutation inherited from parents such as the NOTCH3 gene is the most common autosomal dominant inherited cause of stroke and vascular dementia.

Poor lifestyle choices, such as smoking, overuse of alcohol, lack of physical activity and inadequate relief of chronic stress and poor diet with high in saturated and trans fat, red meat, processed foods and less in fruits and vegetable and whole-grain are some of the key contributors in the development and progression of chronic diseases, including neurological diseases.

Neuroprotection is a process to preserve the neuronal structure and/or function against the death or gradual death of neurons, including the use of medicines and herbs.

In other words, neuroprotection protects the neuronal integrity against the reduction in the rate of neuronal loss over time inducing neurological disease, including neurodegenerative disease.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of neuro-diseases, researchers examined the effects of omega-3 polyunsaturated fatty acids (ω-3 PUFAs) on alleviating microinfarct burdens and functional deficits.

According to the results of Fat-1 transgenic mice and fish oil supplements, ω-3 PUFAs showed strong protective activity against microinfarcts in a multiple diffuse microinfarcts model.

In other words, ω-3 PUFAs significantly attenuated cell apoptosis.

Furthermore, ω-3 PUFAs alleviated the diffuse microinfarct burdens and remarkably improved the functional deficits by reducing spontaneous anxiety, increased preference for the novel object and improved hippocampal-based learning and short-term memory.

Collectively, researchers said, " enriched brain ω-3 PUFAs are effective for reducing microinfarct burdens and improving the function deficits, which support the clinical research and application of ω-3 PUFAs in the treatment or prophylaxis in vascular dementia".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a neuro-protective functional food, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol May Alleviates the Progression of Fatty Liver

By Kyle J. Norton

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing them to other parts of the body.

The liver also produces cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Liver disease is a class of medical conditions characterized by reducing partly or completely the function of the liver, including liver failure.

A fatty liver is a liver condition associated with the accumulation of excess fat in the liver.

There are two main types of fatty liver disease:
* Nonalcoholic fatty liver disease is a medical condition characterized by fat accumulated in the liver tissue other than excessive consumption of alcohol.

According to the American liver foundation, Nonalcoholic Fatty Liver Disease is one of the most common causes of liver disease and approximately 30% (100 million) individuals in the United States are estimated to have nonalcoholic fatty liver disease.

The causes of Nonalcoholic Fatty Liver Disease are unknown. Also, as of today researchers do not understand why some people are susceptible to liver fat accumulation while others are not.

The disease can be classified into the types of non-inflammatory fatty liver (NAFL) and inflammatory nonalcoholic steatohepatitis (NASH) and acute fatty liver of pregnancy (AFLP),  a relatively rare disorder that happens only during pregnancy.

* Alcoholic fatty liver diseases is a condition caused by lifelong heavy alcohol use.

In other words, substances found in the alcohol damage liver cells, promote inflammation and weaken your body's natural defenses. More precisely, the more alcohol that you drink, the more you damage your liver.

Alcoholic fatty liver disease is the earliest stage of alcohol-related liver disease.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of liver diseases, researchers elucidated the effects of stigmasterol and β-sitosterol on a high-fat Western diet (HFWD)-induced nonalcoholic fatty liver disease (NAFLD).

According to the results after 33 weeks of treatment
* stigmasterol and β-sitosterol were effective in protecting the liver against HFWD-induced NAFLD.

* The parameters that contributed to the onset of NAFLD are triacylglycerols (TGs), phosphatidylcholines, cholesteryl esters, diacylglycerols, and free fatty acids (FFAs).

* The efficacy of stigmasterol and β-sitosterol int he alleviation of NAFLD was mainly associated with decreases in hepatic cholesterol, TGs with polyunsaturated fatty acids, and alterations of free hepatic FFA.

Based on the findings, researchers said, "phytosterols, at a dose comparable to that suggested for humans by the FDA for the reduction of plasma cholesterol levels, are shown to protect against NAFLD in this long-term (33-week) study".
Beta-Sitosterol may be considered a remedy for the prevention and treatment of NAFLD, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Tocopherols Show to Inhibits the Risk of Mammary Cancer in Female Mice

By Kyle J. Norton

Cancer is the class of diseases associated with the growth of the cells disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.
The mammary gland located in the breasts of females is responsible for lactation, or the production of milk.

Both males and females have glandular tissue within the breasts, however, in females, the glandular tissue begins to develop after puberty in response to estrogen release.
A mammary tumor is a medical and chronic condition originated from the milk-producing gland.

Most cases of the mammary cancer are first found in the cell on the surface of the inner lining of the milk-producing gland, before penetrating deeper into the layers.

At the advanced stage, the malignant cancer cells can also travel a distance away from the original site to infect other healthy tissues and organs, leading to secondary metastasis.

Epidemiologically, mammary cancer has been found to spread to the colon and the liver.

Most common risk factors of mammary cancer are similar to those of breast cancer, including, aging, physical inactivity, being obese or overweight, family and personal history, having a dense breast, radiation therapy, pregnancy history, genetic proposition, particularly in women who had used drug diethylstilbestrol (DES).

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of chronic diseases, researchers examined the synergistic effect of lycopene and tocopherol against oxidative stress and mammary tumorigenesis.

According to the analysis from the administration of lycopene (Lyco) alone or combined with tocopherol (Lyco + Toco) for 90 days against a single oral dose of (50 mg/kg body weight) 7,12-dimethyl[a], benzanthracene (DMBA)-induced oxidative stress and mammary carcinogenesis in female rats,
* There is a significant elevation in the levels of malondialdehyde (MDA) and nitric oxide (NO) in serum and breast tissues of DMBA-injected rats.

* The combined treatment (Lyco + Toco) group showed a potential reduction of these parameters more than Lyco alone group.

* Furthermore, Lyco alone showed significantly higher activities of superoxide dismutase, catalase, and glutathione peroxidase.

In other words, the formation of tumor and angiogenesis in DMBA-induced rats and these abnormal changes were ameliorated by combined treatment with Lyco + Toco.

After talking about other factors into account, researchers said, "these results suggested that supplementation of the diet with Lyco and Toco provided antioxidant defense, with strong chemopreventive activity against DMBA-induced mammary tumors".

Taken altogether, tocopherols used alone or combined with lycopene may be considered adjunct therapies in the treatment of breast cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Gamma-Linolenic Acid, The Potential Anti-Atherosclerotic Lesions in Vivo

By Kyle J. Norton

Atherosclerosis is a medical condition caused by plaques accumulated on the wall of the arteries, leading to narrowing the arteries in transporting oxygenated blood to the heart and other parts of the body.

Depending on the blood obstruction, atherosclerosis can induce different types of heart disease and stroke.

Plaque is the make of cholesterols, calcium, and other toxic substances, including cadmium.

Atherosclerosis is a slow, progressive disease that may begin as early as childhood, caused by damage to the inner lining of the artery. However, the actual causes of the condition are debatable.

Some researchers suggested that atherosclerosis may be caused by long term high blood pressure,
high cholesterol and triglycerides (a type of fat) in the blood. Particularly, in high levels of insulin in the blood from diabetes and in exposure to chemicals from smoking tobacco.

Patients with early stages of atherosclerosis may not experience any symptoms. However, as the disease progress, patients may have the symptoms of chest pain if the blockage occurs in an artery of the heart (coronary artery) and leg cramps during exercise or walking if the blockage occurs in the legs.

According to the statistics provided by Atherosclerosis Open Acess, 32% of deaths account for atherosclerosis. 13.8% prevalence is among adults of 20 years and above. In 2010, coronary heart disease mortality rate was about 60%.

Risk factors of atherosclerosis include high blood pressure and cholesterol, diabetes, being overweight or obese, an unhealthy diet, a family history of heart disease and physical inactivity, particularly smoking.

Some researchers suggested that since cholesterol makes up a big part in the plaque, an unhealthy diet with high saturated trans fat, red meat and processed meat may be the major cause of atherosclerosis.

Gamma-linolenic acid is a phytochemical in the group of Omega-3, 6,9 fatty acids, belongings to the class of Lipids, found abundantly in evening primrose, borage, blackcurrant, etc.

On finding a potential plant for the prevention and treatment of atherosclerosis, researchers examined the effect of the effects of D5D inhibition by a D5D inhibitor on atherosclerosis.

Where Delta-5 desaturase (D5D), encoded by fatty acid desaturase 1 (Fads1), is the rate-limiting enzyme for the conversion from dihomo-γ-linolenic acid (DGLA) to arachidonic acid (AA) in the ω-6 polyunsaturated fatty acid pathway.

According to the results of the analysis of the Western-diet fed ApoE knockout (KO) mice,
* Oral administration of compound-326 (3-10 mg/kg per day for 15 weeks) significantly inhibited the progression of atherosclerotic lesions in the aorta without affecting plasma total cholesterol and triglyceride levels.

* Compound-326 significantly decreased AA levels, while it increased DGLA levels in the liver and the blood accompanied by decreases in AA-derived eicosanoid production and increases in DGLA-derived eicosanoid production from the blood cells.

In other words, the compound-326 prevents the progression of atherosclerosis in Western-diet fed ApoE KO mice by modulating a profile of eicosanoid production.

Collectively, researchers said, "D5D inhibitors can be a novel remedy for preventing atherosclerosis and subsequent cardiovascular events".

Taken altogether, Gamma-linolenic Acid may be considered a remedy for the prevention and treatment of atherosclerotic lesions,, pending to the confirmation of the larger sample size and multicenter human study.

Lycopene Promotes Multitargeted Therapy of Cancer

By Kyle J. Norton

Cancer is the class of diseases associated with cell growth disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Multitargeted therapy of cancer treatment, in general, to induce an imbalance between cell survival and apoptosis through interference by enhancing the cell cycle regulators and proteins involved in apoptosis.

Dr. Mehta K, the lead scientist in the joint study led by The University of Texas MD Anderson Cancer Center, wrote, "The Society of Translational Cancer Research held its biennial meeting in the capital city of India, Delhi from February 6th through 9th, 2014 to discuss 'Multi-targeted Approach to Treatment of Cancer'"

And "Talks were presented by leaders in the cancer research field from various countries. It became clear from this conference that coupling multiple targeted-agents or using an agent that hits an individual target in several independent locations in the disease-causing pathway(s) may be the best approach to treat different cancers".

Lycopene is a phytochemical found in tomato in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

On finding a potential compound for the treatment of cancer, researchers examined the role of lycopene (L)  as a multitargeted therapy of cancer.

In vitro, lycopene exerted many mechanisms of chemoprevention, including the induction of apoptosis and antiproliferation in cancer cells, anti-metastatic activity.

Furthermore, lycopene also enhanced and promoted an antioxidant response in the synthesis of cytoprotective enzymes.

Moreover, lycopene, a substrate for carotene-9',10'-monooxygenase (CMO2) and can be converted to apo-10'-carotenal plays a critical to inhibit cancer cell migration and angiogenesis.

Taken altogether, lycopene found in tomato may be considered supplements for the prevention and treatment of cancer, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Oleic Acid, A Potential Remedy for the Treatment of Dry Eye

By Kyle J. Norton

Eye diseases are the condition characterized by the change of vision, including a gradual loss of cells in the retina and neurons associated with the loss of vision.

Most eye diseases at the early stage are asymptomatic and painless with no change in the vision until the disease has become quite advanced.

People have an eye problem at one time and another. Most eye conditions are minor and go away sometimes without even noticing.

The most common causes of eye diseases are inflammation as a result of microbial infection, including bacterial, viral, parasitic or fungal infection, eye structure damage due to physical impact, genetic preposition due to mutation of certain associated with the vision, medical conditions such as diabetes and allergens.

According to statistics, approximately one person in three has some form of vision-reducing eye disease by the age of 65 in the US. Age-related macular degeneration (AMD), glaucoma, cataract are the most prevalent eye diseases found in the elderly.

Dry eye is a condition characterized by a lack of quality tears to lubricate and nourish the eye for clear vision.

In other words, dry eye is a condition caused by an imbalance of tear production and drainage or low-quality tear production. The condition is very common in the elderly.

There are certain risk factors that may affect the levels of quality of tears, such as aging (most senior age 65 and over have experience of dry eye), gender (the risk of dry eye is higher in women due to the change of hormone as a result of pregnancy and menopause), medications and long term of use of contact lens.

According to the statistics, in the United States, dry eye affects over 4.88 million Americans age 50 and older. The syndrome is most common in the population of middle-aged or older.

Although it is rare, untreated dry eye or serious dry eye syndrome has been associated with the risk of infection, leading to scarring, thinning, and even perforation of the cornea.

Oleic acid is a phytochemical in the group of Omega-3, 6,9 fatty acids, belongings to the class of Lipids, found abundantly in evening primrose, borage, blackcurrant, etc.

On finding a potential plant that improves eye vision, researchers examined the effect of essential fatty acids (EFAs) as dietary supplements in treating dry-eye for reducing inflammation at the ocular surface.

According to the results of the analysis of the topical application in eye drops to deliver fatty acid (FA, (LA-linoleic acid, ALA-alpha-linolenic acid), OA (oleic acid), and GLA (gamma-linolenic acid)) directly to the ocular surface.
* EFAs or OA in topical ophthalmic preparations can affect the spread and stability of the tear film lipid layer.

* The omega-6 LA (not omega-3 FA) at low concentrations (20 mol%) can be beneficial for enhancing tear stability in dry eye patients.

In other words, out 3 phytochemicals that showed an impact in the treatment of dry eye, only oleic acid exerted a similar effect to those of EFAs.

Taken altogether, OA may be considered a functional fatty acid for the prevention and treatment of dry eye, pending to the confirmation of the larger sample size and multicenter human study.
