Gamma-Linolenic Acid, The Potential Anti-Atherosclerotic Lesions in Vivo

Atherosclerosis is a medical condition caused by plaques accumulated on the wall of the arteries, leading to narrowing the arteries in transporting oxygenated blood to the heart and other parts of the body.

Depending on the blood obstruction, atherosclerosis can induce different types of heart disease and stroke.

Plaque is the make of cholesterols, calcium, and other toxic substances, including cadmium.

Atherosclerosis is a slow, progressive disease that may begin as early as childhood, caused by damage to the inner lining of the artery. However, the actual causes of the condition are debatable.

Some researchers suggested that atherosclerosis may be caused by long term high blood pressure,
high cholesterol and triglycerides (a type of fat) in the blood. Particularly, in high levels of insulin in the blood from diabetes and in exposure to chemicals from smoking tobacco.

Patients with early stages of atherosclerosis may not experience any symptoms. However, as the disease progress, patients may have the symptoms of chest pain if the blockage occurs in an artery of the heart (coronary artery) and leg cramps during exercise or walking if the blockage occurs in the legs.

According to the statistics provided by Atherosclerosis Open Acess, 32% of deaths account for atherosclerosis. 13.8% prevalence is among adults of 20 years and above. In 2010, coronary heart disease mortality rate was about 60%.

Risk factors of atherosclerosis include high blood pressure and cholesterol, diabetes, being overweight or obese, an unhealthy diet, a family history of heart disease and physical inactivity, particularly smoking.

Some researchers suggested that since cholesterol makes up a big part in the plaque, an unhealthy diet with high saturated trans fat, red meat and processed meat may be the major cause of atherosclerosis.

Gamma-linolenic acid is a phytochemical in the group of Omega-3, 6,9 fatty acids, belongings to the class of Lipids, found abundantly in evening primrose, borage, blackcurrant, etc.

On finding a potential plant for the prevention and treatment of atherosclerosis, researchers examined the effect of the effects of D5D inhibition by a D5D inhibitor on atherosclerosis.

Where Delta-5 desaturase (D5D), encoded by fatty acid desaturase 1 (Fads1), is the rate-limiting enzyme for the conversion from dihomo-γ-linolenic acid (DGLA) to arachidonic acid (AA) in the ω-6 polyunsaturated fatty acid pathway.

According to the results of the analysis of the Western-diet fed ApoE knockout (KO) mice,
* Oral administration of compound-326 (3-10 mg/kg per day for 15 weeks) significantly inhibited the progression of atherosclerotic lesions in the aorta without affecting plasma total cholesterol and triglyceride levels.

* Compound-326 significantly decreased AA levels, while it increased DGLA levels in the liver and the blood accompanied by decreases in AA-derived eicosanoid production and increases in DGLA-derived eicosanoid production from the blood cells.

In other words, the compound-326 prevents the progression of atherosclerosis in Western-diet fed ApoE KO mice by modulating a profile of eicosanoid production.

Collectively, researchers said, "D5D inhibitors can be a novel remedy for preventing atherosclerosis and subsequent cardiovascular events".

Taken altogether, Gamma-linolenic Acid may be considered a remedy for the prevention and treatment of atherosclerotic lesions,, pending to the confirmation of the larger sample size and multicenter human study.

Lycopene Promotes Multitargeted Therapy of Cancer

Cancer is the class of diseases associated with cell growth disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Multitargeted therapy of cancer treatment, in general, to induce an imbalance between cell survival and apoptosis through interference by enhancing the cell cycle regulators and proteins involved in apoptosis. 

Dr. Mehta K, the lead scientist in the joint study led by The University of Texas MD Anderson Cancer Center, wrote, "The Society of Translational Cancer Research held its biennial meeting in the capital city of India, Delhi from February 6th through 9th, 2014 to discuss 'Multi-targeted Approach to Treatment of Cancer'"

And "Talks were presented by leaders in the cancer research field from various countries. It became clear from this conference that coupling multiple targeted-agents or using an agent that hits an individual target in several independent locations in the disease-causing pathway(s) may be the best approach to treat different cancers".

People have an eye problem at one time and another. Most eye conditions are minor and go away sometimes without even noticing.

Inflammation is a natural defense of the immune system that protects the body against the insult from the foreign invasion.

Macrophages, one the lymphocytes and the first-line defense of the immune system after sensing the damage of localized tissue, stimulate the production of proinflammatory cytokines to the site to protect the body against invasion of potential pathogens.

However, in some cases, overexpression of proinflammatory cytokines can cause the death of nearby healthy cells, leading to abnormal skin healing and the formation of irreversible scars.

Acute inflammation occurs quickly or within the hours and lasts a few days or weeks and the damaged tissues are healed themselves after all pathogens are killed off.

However, if the immune system can not completely eradicate all invaders in the acute phase of infection, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

Chronic inflammation is a low-grade inflammation caused by the inability of the immune system to eliminate the invasive pathogens that occurred in the acute phase of infection.

Any inflammation that lasts more than 8 weeks is considered chronic inflammation.

Chronic inflammation can also be caused by an autoimmune disorder in which the immune system continuously attacks normal and healthy tissue, by mistaking them for the causative pathogens.

Low-grade inflammation is a different type of chronic inflammation characterized by a production of the inflammatory cytokines in a low-grade state.

* Interestingly, the interaction of OAA with IκB kinase α/β (IKKα/β) strongly attenuated the production of certain proteins and inflammatory cytokines in the TLR3 signaling pathway.

Collectively, researchers said, "These results reveal new insight into the understanding of the regulatory mechanisms of the downstream TLR3 signaling pathway and consequent inflammatory responses that are involved in the development and progression of inflammatory diseases".

Dandelion(Taraxacum officinale), the Potential Anti-Gastric/Stomach Cancer Herbal Medicine

The stomach located between the esophagus and the small intestine is a muscular, hollow and important organ of the digestive tract.

The stomach plays a critical role to assist the digestive system in digesting foods by secreting acid and enzymes.

Cancer is a class of chronic diseases associated with irregular cell growth in the tissues of the affected organ.

Stomach cancer is a condition of abnormal growth of the mucus-producing cells of the inside lining of the stomach. Adenocarcinoma is the most common type of stomach cancer.

According to the statistics, most cases of gastric/stomach cancer are found in older people. The average age of people when they are diagnosed is 68. The risk of developing gastric/stomach cancer in a man lifetime is about 1 in 95.

Reactive oxygen species (ROS) or free radicals are highly reactive chemical species containing oxygen molecules. In other words, free radicals are unstable atoms with an unpaired electron in the outer ring.

Therefore, in order to become stable, free radicals must either donate or steal an electron from other atoms, leading to a chain reaction that can not be stopped until all electrons are paired or inhibited by antioxidants.

Believe it or not, free radicals in moderate amounts are needed for our body for cell signaling and activation of several transcriptional factors, apoptosis, immunity, and differentiation. However, overexpression of free radicles can cause damage to protein, lipid and cell alternation, leading to a number of chronic illnesses, including cancer.

Free radicals are generated as metabolic by-products by biological systems. Environmental stressors (i.e., the air we breathe, UV, ionizing radiations, pollutants, and heavy metals) and xenochemicals are also found to contribute to the great increase of ROS production.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid and cell damage.

Antioxidants can either produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

However, in people with a weakened immune system, intake dietary sources of antioxidants may be necessary.

Devil's claw (Harpagophytum procumbens) Suppresses the Symptoms of Osteoarthritis

Osteoarthritis is a type of musculoskeletal disorders (MSDs) and medical condition mostly caused by work-related occupations and working environment, affecting patients’ muscles, joints, tendons, ligaments and nerves and developing over time.

More precisely, Osteoarthritis (OA), a form of arthritis, is a condition of aging causes of wear and tear on a joint, affecting over 25 million people in the United States alone. 

University of Porto Medical School indicated that one must understand the differences in prevalence and incidence estimates of osteoarthritis (OA), according to case definition, in the knee, hip and hand joints(3).

The most common symptoms associated with OA are aching pain, stiffness, or difficulty of moving the joint or joints. 

The pain usually gets worse in a change of weather, at night and in the advanced diseases, the pain can occur even at rest. 

Today management of osteoarthritis (OA) focuses on pain relief and improved physical function through pharmacological, nonpharmacological, and surgical treatments.

Osteoarthritis getting worse overtime can induce complications of
* Gout
The forming of sodium urate crystals forming in and around your joints, due to high levels of urate can result from inflammatory arthritis or gout.

* Chondrocalcinosis

Devil's claw also known as the wood spider, is a plant of genus Harpagophytum in the family of Pedaliaceae, native to South Africa. 

The plant has been used as an herb in traditional and herbal medicine to treat fever, rheumatoid arthritis, skin, gallbladder, pancreas, stomach and kidneys conditions, etc.

On finding a natural ingredient for the prevention and treatment of chronic inflammatory diseases, scientists investigated the effectiveness of a dietary supplement including hydrolyzed gelatin, chondroitin sulfate, glucosamine sulfate, and devil's claw and bamboo extracts for pain reduction and improvement of functional capacities in patients with osteoarthritis (OA) of the knee and/or hip (REDART study).

According to the results of a total of 130 patients with OA who were recruited from Spanish hospitals received the dietary supplement for 6 months,
* Patients (N = 78) showed a reduction of pain of 3.77 ± 1.77 points after 6 months (p < .0001) in the VAS.

* The total reduction in the Lequesne Functional Index was 6.30 ± 4.08 points after 6 months (p < .0001), with significant reductions in all subindexes of the scale.

* A similar pattern was found for the WOMAC index, with an overall reduction of 22.49 ± 14.03 points after 6 months (p < .0001) and significant reductions in all subindexes.

Based on the results, researchers suggest, "No major adverse events were noted during the treatment. This exploratory study shows that treatment with the dietary supplement significantly reduces pain and improves locomotor function in patients with OA of the knee and/or hip".

Taken altogether, Devil's claw (Harpagophytum procumbens) may be considered a remedy for the treatment of osteoarthritis, pending to the confirmation of the larger sample size and multicenter human study.
