Herbal Angelica, Processes Apoptotic and Anti Proliferative Function in Vitro

Cancer is a group of medical and chronic diseases characterized by cell growth disorderly and uncontrollably in the tissue of specific organs in the body.

Over time, the alternative cells induced tumor may become progressively worse and damage other healthy tissues, depending on the grade of the tumor.

At the later stage, the most invasive tumor can induce secondary metastasis by spreading to other organs in the body via lymph or blood.

In the immune perspective, anti-proliferation can be carried out by activating the genes associated with cancer suppression, programming cell death,.... and cell cycle arrest.

A suppressive gene, such as pro-apoptotic p53 induces cancer cell death through cell cycle arrest. In other words, the gene inhibits tumor growth by interrupting the cancer cell cycle division.

In the programmed cell death gene, such as BCL-2 induce cell death by either inducing apoptosis. In other words, If cells are no longer needed or damaged, activating an intracellular death program will cause cell death.

In phytochemicals, anti-proliferation can also be carried out by antioxidants in which they stimulate the free radicals overexpression to induce toxicity to tumor cells without causing harm to the nearby healthy cells.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

On finding a potential compound for the prevention and treatment of cancer, researchers examined the effects of Angelica phytochemical (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone from A. purpuraefolia on cell proliferation and apoptotic cell death.

In 4 breast (MDA-MB-231, MCF-7, HS578T, and T47D), three cervical (HeLa, SiHa, and C33A) cancer cell lines, and NIH 3T3 normal cells used for the experiment, researchers found that
* (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone treatment suppresses the growth and proliferation of cancer cells at a relatively low concentration (<10 μg/ml).

* The phytochemical also induces cancer cell apoptosis at a high concentration (>50 μg/ml).

* At the concentration of 10 μg/ml, the chemical compounds also exerted a strong cell cycle arrest in the S/G2 phase.

Furthermore, according to the Western blot analysis, the concentration higher than 50 μg/ml of (+)-4'-decanoyl-cis-khellactone increased the large percentage of apoptotic cells in a time-dependent manner.

Interestingly, while the (+)-4'-decanoyl-cis-khellactone using both extrinsic and intrinsic pathways to induce apoptosis, the but (+)-3'-decanoyl-cis-khellactone uses only an intrinsic pathway for the same results.

In other words, the (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone also inhibited the in vitro proliferation of six other human breast and cervical cancer cell lines with different pathways.

Based on the evidence, researchers said, "Our data strongly suggest that (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone act as an anti-cancer supplement particularly against breast and cervical cancers by inducing cell cycle arrest in the S/G2 phase and caspase-dependent apoptosis at relatively low and high concentrations, respectively".

Taken altogether, Herbal Angelica processes anti-proliferative activity against cancer, pending to larger sample size and multicenter human study.

Dandelion (Taraxacum officinale), the Best Antioxidant Functional Herbal Medicine

Antioxidants are stable atoms that inhibit oxidation caused by free radicals.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily to donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.
Antioxidants, on the other hand, are stable molecules with a unique function in the inhibition of oxidation.

Believe it or not, free radicals also play a critical role in maintaining a certain body function. The Journal of Physiology showed that free radicals act as signal substances that cause the heart to beat with the correct force.

In healthy individuals, antioxidant enzymes produced by the liver are good enough to balance the free radical produced by the cell metabolism and external sources such as the air we breathe and the food we eat.

However, in people with a weakened immune system such as infants, children and the elderly, dietary sources from supplements, fruits and vegetables may be needed.


Dandelion is a herbaceous perennial plant, genus Taraxacum, belongings to the family Asteraceae, native to temperate regions of the world.

The herbal medicine has been used in traditional and herbal medicine to treat gout, eczema, acne, gall bladder, kidney, liver and urinary disorders, hypoglycemia, dyspepsia with constipation, edema, blood pressure, and heart weakness, chronic joint, skin diseases, etc.

On finding a natural ingredient for the treatment of chronic inflammatory diseases, scientists examined in vitro and in vivo the antioxidant effects of 70% ethanol-water (v/v) leaf and root extracts of T. officinale (TOL and TOR, respectively).

The study included organ homogenate samples from Nω-nitro-L-arginine methyl ester (L-NAME)-induced hypertensive rats, treated by leaf extracts of TOL (500 mg/kg/day) and TOR (500 mg/kg/day) for 21 days.

Based on the results from the experiment, the antioxidant contents of ) polyphenol (4.35 ± 0.15 compared to 1.14 ± 0.01) and flavonoid (23.17 ± 0.14 compared to 3 ± 0.05) of TOL are significantly higher compared to TOR.

TOL exerted a significantly higher free radical scavenging activity (EC50 0.37 compared to 1.34 mg/mL) and total antioxidant capacities (82.56% compared to 61.54% ABTS, and 156 ± 5.28 compared to 40 ± 0.31 FRAP) compared to TOR.

Furthermore, both extracts inhibited the oxidative stress marker MDA levels and increased the total antioxidant capacity (TAC) without inducing toxicity with LD50 > 5000 mg/kg).

Collectively, T. officinale leaf extract possesses significant anti-oxidant effects that conferred significant in vivo antioxidant protection against free radical-mediated oxidative stress in L-NAME-induced hypertensive rats.

Taken altogether, dandelion may be considered a remedy for enhancing the antioxidant capacity against oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Oxidative stress occurs when the levels of free radical compared to the antioxidant enzymes in the body is imbalanced.

Oxidative stress is either associated with the overexpression of free radicals or reduced levels of antioxidants or both.

Free radicals can be everywhere in the environment such as the air we breathe, the foods we eat. They can also be produced by cell metabolism in the body.

Overexpression of free radicals is harmful to the body. However, in a moderate amount, free radicals are necessary for our body to function, including NO for muscle relaxation.

Epidemiologically, oxidative stress has been found to induce lipid and protein oxidation and cell DNA damage, a leading cause of various chronic diseases such as atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, stroke.

The heart formed part of the circulatory system is a muscular organ that pumps blood through the blood vessels by providing the body with oxygen and nutrients and removal of metabolic wastes.

In the heart, cardio-oxidative stress causes heart muscle damage, leading to dilated cardiomyopathy and heart failure.

Dr. Dhalla NS, the lead scientist, in the examination of the risk of oxidative stress in cardiovascular disease, wrote, "Oxidative stress in cardiac and vascular myocytes describes the injury caused to cells resulting from the increased formation of ROS and/or decreased antioxidant reserve. Furthermore, the anti cardio oxidative stress induced by Dox was also inhibited by the injection curcumin by activating Bad (S112) phosphorylation against cell death and, reducing the heart rate, ST-segment, low-density lipoprotein (LDH) and creatine kinase (CK) activities found in the serum.

In other words, curcumin decreased cardiotoxicity by protecting the heart muscle cells' integrity through many mechanisms, including proteins associated with oxidative stress in the induction of apoptosis.

Based on the findings, researchers said, "Cur protected the myocardium against Dox-induced injury via upregulating 14-3-3γ expression, thereby promoting the translocation of Bcl-2 to mitochondria, suppressing oxidative stress, and improving mitochondrial function".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of heart diseases associated with cardio-oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

* Proctosigmoiditis is an Inflammation involved in the rectum and sigmoid colon (lower end of the colon).

The exact causes of UC are not cleared. Some researchers suggested there are a complex interaction of genetics, immune response, and environmental triggers that cause the onset of the disease.

On finding a potential compound for the prevention of colitis, researchers examined the decursin and decursinol angelate-rich Angelica gigas Nakai (AGNE) on dextran sulfate sodium (DSS)-induced murine ulcerative colitis (UC) in a mouse model.

In mice with DSS-induced UC, treatment with the AGNE at 10, 20, and 40 mg/kg exerted strong colonic protection and activity by alleviating decreased disease activity index scores, and reduced colon shortening and weight loss.

Levels of pro-inflammatory cytokines were also decreased in serum and colon tissue with the administration of AGNE.  

Furthermore, AGNE also improved the colon movement by increasing the production of PGE2 in colon tissue and suppressed the increased expression of COX-2 and HIF-1α in the increased intestinal mucosa of UC mouse.

Finally, the histological damage of the colon was also alleviated by AGNE treatment.

Collectively, researchers said, "The findings of this study verified that AGNE significantly improves clinical symptoms and reduces the activity of various inflammatory mediators. These results indicate the AGNE has the therapeutic potential in mice with DSS-induced UC".

Taken altogether, herbal Angelica may be considered a  remedy dor the treatment of UC, pending to larger sample size and multicenter human study.

Broccoli In the Prevention of Colitis

Colitis is a medical condition of inflammation of the large intestine, including the colon, caecum, and rectum.

Types of colitis according to the Catholic University of the Sacred Heart, are including
microscopic colitis, ischemic colitis, segmental colitis associated with diverticula, radiation colitis, diversion colitis, eosinophilic colitis, and Behcet's colitis.

There is no exact cause of colitis. Certain risk factors such as colon infection, inflammatory bowel disease (Crohn's disease, ulcerative colitis), ischemic colitis, allergic reactions, and microscopic colitis have been found to increase the risk of the onset of the condition.

The most common symptoms of colitis are totally depending on the types, including abdominal pain, cramping, diarrhea, with or without blood in the stool.

The most common risk factors associated with the onset of colitis are Medical conditions such as hypertension, diabetes, hyperlipidemia, and atrial fibrillation aging, genders, smoking, depression, and psychosocial stress and family history.

Other risk factors may also contribute to the incidence of colitis, according to the study by Service d'Epidémiologie et de Santé Publique, in a multivariate model are familial history of inflammatory bowel disease (odds ratio (OR) 4.3 (95% confidence interval 2.3-8)), breastfeeding (OR 2.1 (1.3-3.4)), bacille Calmette-Guerin vaccination (OR 3.6 (1.1-11.9)), and history of eczema (OR 2.1 (1-4.5)), disease during pregnancy (OR 8.9 (1.5-52)), and bedroom sharing (OR 7.1 (1.9-27.4)).

Conventionally, besides treating colitis with antibiotics, certain medications also are prescribed to improve the symptoms and quality of life of the patients, including anti-diarrheal medications.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The vegetable has large flower heads, usually green in color and the mass of flower heads is surrounded by leaves and evolved from a wild cabbage plant on the continent of Europe.

On finding a potential plant that process anti-colitis activity researchers compared the effects of broccoli-derived nanoparticle (BDN) and SFN on three mouse colitis models.

According to the data collected from the experiment, orally given nanoparticles isolated from broccoli extracts protected mice against colitis by-mediating the activation of 5'-adenylic acid-activated protein kinase (AMPK) associated with cellular energy homeostasis.

In the immune analysis, BDN prevented the dendritic cells (DCs) to act as messengers between the innate and the adaptive immune systems in dextran sulfate sodium (DSS)-induced colitis in C57BL/6 (B6) mice by targeting dendritic cells (DCs) activation and inducing DC tolerance.

Compared to BND, DCs pre-pulsed with SFN also prevented DSS-induced colitis in B6 mice,, similar to those of BND.

Taken altogether, broccoli derived nanoparticle (BDN) or SFN isolated from broccoli may be considered a functional food for the prevention and treatment of colitis, pending to the confirmation of the larger sample size and multicenter human study.
