Bioactive Compound 3,3'-Diindolylmethane Protects the Neurons Integrityand Viability Against Autoimmune Encephalomyelitis,

By Kyle J. Norton

Autoimmune encephalitis is a group of conditions caused by the body's immune system mistakenly attacks healthy brain cells.

Autoimmune encephalitis has been found to induce neurologic symptoms such as impaired memory and cognition, abnormal movements, seizures, and/or psychiatric symptoms such as aggression, inappropriate sexual behaviors, panic attacks.

Autoimmune encephalitis can be classified into
* Post-infectious encephalitis or acute disseminated encephalomyelitis (ADEM) are the conditions caused by an infection. In these cases, the immune system not only attacks the pathogens which cause infection it also destroys mistakenly the healthy cells in the brain.

Methylprednisolone is the first line of drug for the treatment of post-infectious encephalitis, if it failures, plasma exchange or intravenous immunoglobulin may be used.

* Other forms of autoimmune encephalitis are caused by the specific antibodies in the blood produced by the immune system that attack the brain cells.

According to the Autoimmune Encephalitis Alliance, a number of options are available to treat AE, the first line option include including medication that decreases the inflammatory response to antibodies, and intravenous immunoglobulin (IVIG); to increase removal of antibodies, inhibit binding of the harmful antibodies.

If the first line option failure, immune suppressive medicine may be used as a second line of treatment, including Rituximab, CellCept, and Cytoxan (Cyclophosphamide).

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

With an aim to find a potential ingredient for the treatment of autoimmune encephalomyelitis, researchers evaluated the 3,3'-Diindolylmethane (DIM) effects on the regulation of activated T cells during the development of experimental autoimmune encephalomyelitis (EAE).

During the experiment, administration of DIM 10 days after EAE was effective in ameliorating disease parameters, including inflammation and central nervous system cellular infiltration.

According to the microRNA (miRNA) microarray analysis, post-treatment of DIM alter the expression of miRNA profile in brain infiltrating CD4(+) T cells, an indication of autoimmune dysfunction on EAE mice.

Observed by the bioinformatics analysis, the efficacy of DIM-altered the miRNAs expression associated with pathways that halt cell cycle progression and promote apoptosis.

Particularly, DIM impacted these cellular processes in activated T cells.

More precisely, DIM protected the brain against the immune dysfunction in attacking the brain cell in the forming of autoimmune encephalomyelitis, by activating and suppressing the micro genes- induced immune dysfunction of EAE.

Dr. the lead scientist, after taking into account co and confounders wrote, "these studies demonstrate that DIM post-treatment leads to the amelioration of EAE development by suppressing T-cell responses through the induction of select miRNAs that control cell cycle progression and mediate apoptosis".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of autoimmune encephalomyelitis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Tomato Improves Sleep Quality by Enhancing Levels of Melatonin

By Kyle J. Norton

Sleep is a naturally recurring state of mind and body in human, In sleep, most functions of the brain are reduced including the sensory activity, voluntary muscles are inhibited.

In other words, sleep is the state in which human reduced interactions with the outside world.

The human sleep recurrently to reenergize their body so that when awake they can carry out a daily activity without feeling tired or weak.

In the brain, sleep helps to maintain the pathways associated with brain function such as learning and studying. A recent study suggested, sleep helps to remove toxins in the brain that was built up when  awake.

Believe it or not, we spend about one-third of our life sleeping.

Sleep disorders are syndromes associated negatively with the changes in the sleep pattern, including insomnia, one of the most common causes of disease disorders in the US, found in 30% of adults, including 10% of chronic insomnia is a condition of difficulty falling asleep or staying asleep throughout the night.

According to the statistics, 50-70 million US adults have a sleep disorder including 37.9% reported unintentionally falling asleep during the day.

Sleep disorders have been found to have a strong implication on economic cost. Dr. Hillman DR, the lead scientist at the Sir Charles Gairdner Hospital wrote, "The overall cost of sleep disorders in Australia in 2004 (population: 20.1 million) was dollar 7494 million".

And, "The direct and indirect costs of sleep disorders are high. The total financial costs (independent of the cost of suffering) of dollar 4524 million represents 0.8% of Australian gross domestic product. The cost of suffering of dollar 2970 million is 1.4% of the total burden of disease in Australia".

Tomato is red, edible fruit, genus Solanum, belongs to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

On finding a natural and potential compound for the improvement of sleep quality, researchers investigated the effect of tomato intake before bed on obese postmenopausal women's urinary 6-sulphatoxymelatonin (aMT6s) level, a reliable surrogate biomarker reflecting the blood melatonin concentration and sleep quality.

The study is an 8-week open-label, randomized controlled dietary intervention trial included 36 subjects completed the entire trial.

In the tomato group, participants ate 250 g of beefsteak tomatoes 2 h before sleep for 8 weeks.

After 8 weeks, by comparing the blood and urine samples collected at the baseline and in the 8th week, researchers found at 8 weeks of the beefsteak intervention, improved all components of all components the PSQI compared to the Pittsburgh Sleep Quality Index (PSQI) before treatment.

Furthermore, tomatoes and beefsteak intervention also improved the levels of aMT6s level by 10-fold compared that of the control group.

In other words,  tomato combined with beefsteak improved the sleep quality of the participants.

These results strongly suggested that tomato enhanced the levels of melatonin in the beef substantially in the improvement of sleep quality.

Dr. Yang TH, the lead scientist at the Taipei Medical University wrote in the final report, "supplementation with beefsteak tomato before sleep can increase circulating melatonin and improve sleep quality in obese postmenopausal women".

Taken altogether, tomatoes combined with beefsteak may be considered treatment to improve sleep quality, pending to the confirmation of the larger sample size and multicenter human study.

The Easy Way to Prevent Osteoporosis That No One Wants to Tell --Eat Your Cooked Tomato, Scientists Say

By Kyle J. Norton

Osteoporosis is a condition associated with low bone mass and deterioration of bone tissue.

The disease is most common in older adults as the result of the bone made by the body can not cover the bone loss due to an increased serum of osteoclast mediator involved in bone resorption.

Consequently, the bone in osteoporotic patients become weak and breaking easily from a fall.
Believe it or not, in serious cases, bone can also be broken due to sneezings or minor bumps.

According to the statistics provided by the International Osteoporosis Foundation, approximately 44 million U.S. women and men aged 50 and older suffer from osteoporosis and low bone mass.

There are over 200 million women worldwide affected by osteoporosis, including one-tenth of women aged 60, one-fifth of women aged 70, two-fifths of women aged 80 and two-thirds of women aged 90.

The most common cause of osteoporosis is the reduced production of hormones, particularly estrogen in women and androgen in men due to aging or other conditions such as medical condition.

(Diseases associated with osteoporosis include rheumatoid arthritis and rheumatological conditions, steroid hormone Deficiency, primary hyperparathyroidism, diabetes, and chronic kidney and liver disease).

In older women, menopause accompanied by lower estrogen levels are found in patients with osteoporosis.

According to the epidemiological studies, being a woman and being older, family history with osteoporosis, a small frame, and structure, ethnicity, follow a long-term calcium deficient diet are some of the prevalent factors of disease.

However, out of the aforementioned risk factors, some researchers suggested smoking may be one of the major culprits in inducing the risk of the condition in the Southeast Asain.

The research team at the University of Pennsylvania in the study of how smoking causes osteoporosis, said, ""It has been known from many epidemiological studies that smokers usually show bone loss, and their bone density is much lower than non-smokers' bone density,"

and "What we show here is that excessive formation of osteoclasts by cigarette chemicals causes bone loss".

In other words, if you are a smoker, your risk of osteoporosis is increased substantially compared to a non-smoked individual.

Tomato is red, edible fruit, genus Solanum, belongs to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find a potential compound for the treatment of osteoporosis against bone resorption, researchers examined the lycopene a carotenoid commonly found in tomatoes and tomato products against osteoporosis risk.

The study included 23 healthy postmenopausal women, 50-60 years old, provided blood samples at baseline and following a one-month lycopene-depletion period according to the St. Michael 's Hospital, Toronto, ON, Canada. PARTICIPANTS AND STUDY DESIGN.

According to the results of sample analysis, one-month lycopene-depletion period diet showed a significantly decreased serum lycopene, lutein/zeaxanthin and levels of antioxidants glutathione peroxidase (GPx).

Furthermore, the lycopene restriction diet not only increased lipid and protein oxidation but also depression of levels of antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), significantly.

These results lead to significant increased of the levels cross-linked N-telopeptide of type I collagen (NTx), a marker of osteoclast-mediated bone resorption.

In other words, tomatoes restrictive diet increased the risk of osteoporosis in postmenopausal women.

In order to reveal more information about tomato effect in osteoporosis, researchers conducted a study to determine whether lycopene would act as an antioxidant to decrease oxidative stress parameters, resulting in decreased bone turnover markers, thus reducing the risk of osteoporosis in postmenopausal women.

The study included 60 postmenopausal women, 50-60 years old, following a 1-month washout without lycopene consumption, randomly divided into group consumed either (N = 15/group): (1) regular tomato juice, (2) lycopene-rich tomato juice, (3) tomato Lyc-O-Mato lycopene capsules, or (4) placebo capsules, twice daily for total lycopene intakes of 30, 70, 30, and 0 mg/day respectively for 4 months.

According to the blood sample collected at the end of the experiment, consumed juice or lycopene capsules treated groups for 4 months significantly increased serum lycopene compared to placebo.

Moreover, consumed juice or lycopene capsules treated groups for 4 months exerted a similar effect as the aforementioned study in significantly increasing the total serum antioxidant capacity (TAC) and decreasing lipid and protein peroxidation and NTx compared to the placebo group.

Based on the findings, researchers said, "the daily consumption of lycopene may be important as it acts as an antioxidant to decrease bone resorption in postmenopausal women and may, therefore, be beneficial in reducing the risk of osteoporosis".

Taken altogether, tomatoes and its major bioactive compound lycopene may be considered remedies for the prevention and treatment of osteoporosis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Turmeric, the Hepatoprotective Functional Remedy

By Kyle J. Norton

Hepatoprotection is the action including the use of a substance such as herbal remedy to protect the liver against damage and injury.

Antihepatotoxicity is the ability that protects the liver against toxins.

Acute liver diseases in most cases are caused by drugs and long-term intake of certain medication. Most acute liver disease can be cured if the cause is found.

Chronic liver diseases (CLD) are any liver diseases that last more than 6 months. Most chronic liver diseases are caused by a genetic preposition or a variety of factors that damage the liver, including viruses and alcohol use.

Non-alcoholic fatty liver disease (NAFLD) is one of the most common liver diseases in the world affecting 25% of the general population most of them in the Western world.

Some researchers suggested that the use of substances to protect the liver may involve several mechanisms, including strengthening the liver, gastrointestinal and immune system.

Dr. C. Girish, the lead scientists in the discussion of hepatoprotective effect, wrote, " The regulation of gastrointestinal system may improve the general well-being of the patients, as well as improvement of the constipation, may prevent the absorption of harmful substances and indirectly decrease ascites (Langmead and Rampton, 2001). The protection of liver cells against toxic materials including drugs, lipid peroxidation, and free radical injury may decrease inflammation, improve liver blood flow, and ultimately help in reduction of ascites and blood pressure (Yang et al., 2000)".

In other words, herbal medicine expressed liver protection not only by improving the blood flow to the liver but also through regating the gastrointestinal system against the building up toxins in the induction of free radicals and suppressing the normal function of the liver and improving the immune system against inflammation induced the lipid oxidation.

Herbal Bilberry, Protects your Lung Against Mycobacterium tuberculosis

By Kyle J. Norton

Mycobacterium avium complex (MAC) is a group of bacteria that cause tuberculosis through infection. Most cases Mycobacterium tuberculosis are found in the lungs.

Mycobacterium tuberculosis is pathogens that can cause tuberculosis (TB) in humans discovered in 1882 by Robert Koch.

According to statistics, 8 million new cases of TB were diagnosed annually. The disease causes the death of 2 million people worldwide, mostly in the third world.

Most people infected by MAC are asymptomatic as the bacteria remaining in your body in the inactive state. Latent TB is not contagious.

In the active stage, TB can induce symptoms of persistent coughing, unintentional weight loss, and fatigue.

In severe cases, TB can cause or pain with breathing or coughing, coughing up blood and chest pain.

Active TB is very contagious, it can spread through tiny droplets released into the air via coughs and sneezes.

All cases of active tuberculosis are treated with antibiotics that lasts six to nine months.

However, the length of treatment is totally depending on patients age and overall health, the infected location.

Some researchers suggested People with a weakened immune system are prone to the infection of Mycobacterium tuberculosis.

Dr. S H E Kaufmann said, "Mycobacterium tuberculosis. Of these, fewer than 10% ever develop the disease, although the pathogen is not eradicated but rather contained in discrete lesions. Hence, the immune system is highly effective in containing the pathogen but fails to eradicate it".

And, "Disease typically develops through reactivation once the immune system is weakened"

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

In the urgency to find a natural source for the treatment of Mycobacterium aurum tuberculosis for the third world countries, where the Western medications are scarce, researchers examined the efficacy of Vaccinium myrtillus (MIC = 8 microg/mL) extract.

Compared to the ethanol extracts of Xerocomus badius, Chalciporus piperatus, Suillus luteus and of endophytes isolated from C. vulgaris, E. nigrum, Vaccinium Vitis-idaea, V. myrtillus respectively at  90 to 96% inhibition at 100 microg/mL exerted a potent antitubercular activity.

Compared to herbal C. arbuscula, Empetrum nigrum, J. communis roots, Calluna vulgaris aerial parts, Myrica gale the roots and stems at 93 to 99% inhibition at 100 microg/mL V. myrtillus showed a lesser efficacy.

These results provided some scientific basis for the traditional use of V. myrtillus for the treatment of tuberculosis.

Thes results also strongly indicated that fungal endophytes recovered from Scottish plants are a source of antimycobacterial agents

Taken altogether, bilberry may be considered a function remedy for the treatment of TB, pending to the validation of larger sample size and multicenter human study.

Herbal Artichoke Health Benefit

By Kyle J. Norton

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine for as liver protective and detoxified agent, and to treat digestive disorders, abdominal pain gas and bloating, etc.

Fructooligosaccharides (FOS), are oligosaccharide, a molecule made up of a small group of these sugar,

Due to its nature as a natural source of sugar and low in calories,  FOS may be used as an alternative sweetener as intake of FOS does not cause a spike in blood sugar.

FOS are considered prebiotics. Intake of FOS can increase the levels of good bacteria in the intestine, leading to a healthy ratio of gut microbiota.

Digestively, FOS contains high levels of soluble fiber which has been found to improve bowel function. Consumption of FOS also increased fecal bolus and the frequency of depositions, while a dose of 4-15 g/day given to healthy subjects has been found to reduce constipation.

According to the study, FOS also showed anti toxicity, carcinogenicity, and genotoxic effects.

FOS improved mineral absorption by decreasing levels of  pH in the colon, thus facilitating the absorption of mineral ions from the intestine, mainly calcium and magnesium

Furthermore, FOS also found in studies decrease levels of serum cholesterol, triacylglycerols, the lead causes of cardiovascular diseases.

By modulating the levels of phospholipids, FOS lower the risk of coronary heart disease, inflammation or cancer, particularly in colon cancer.

In foods, FOS are increasingly included in food products and infant formulas for their prebiotic effect in stimulating the growth of nonpathogenic intestinal microflora.

In the examined the effects of chronic ingestion of short-chain fructooligosaccharides (FOS), an indigestible carbohydrate, on hepatic glucose production, insulin-mediated glucose metabolism, erythrocyte insulin binding, and blood lipids in 12 healthy volunteers selected received either 20 g FOS/d or sucrose for 4 wk in a double-blind crossover design, researchers found that the mean (+/- SEM) basal hepatic glucose production was lower after FOS than after sucrose consumption, through the effects on insulin-stimulated glucose metabolism in healthy subjects.

Taken altogether, artichoke processed many health benefits may be used as a functional remedy for diseases prevention in healthy subjects, pending to the validation of larger sample size and multicenter human study.

Herbal Ginger, the Natural Treatment for Gastric/Stomach Ulcers

By Kyle J. Norton

Gastric ulcers or stomach ulcers are conditions characterized by the painful sores in the stomach lining. Unlike the gastric ulcers, peptic ulcers are sores found in the stomach and small intestines.

The exact causes of gastric ulcer are not clearly identified. However, according to the epidemiological studies, a bacterial infection caused by the H. pylori infection has been found in the range of 30 to 50% of people with stomach ulcers.

Gastric ulcers caused by H. pylori infection are more likely to develop in patients over 40 in the US

Other risk factors associated with gastric ulcer include long-term use of certain medications, such as NSAID which has been found to cause more than 50% of peptic ulcers and stress.

Treatment of gastric ulcers is depended totally on the underlying causes, including antacids for the reduced acid in the stomach, and antibiotics for the elimination of Helicobacter pylori.

Unfortunately, people with gastric ulcer infected by the Helicobacter pylori treated with antibiotics have a higher risk of recurrence. In severe cases, surgery may be necessary.

Untreated or uncovered stomach ulcers may induce some complications, including stomach bleeding penetration,  and cancer.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

With an aim to find a potential ingredient for the treatment of gastric ulcers with no side effects, researchers examined the possible mechanism of the protective effect of ginger on the aspirin-induced gastric ulcer.

The study included 40 adult male albino rats randomized divided  into four groups (10 animal per each group) and orally received the followings once daily for 5 days:

Group I: 3 ml of 1% carboxymethyl cellulose;

Group II: ginger powder (200 mg/kg body weight) suspended in 3 mL of 1% carboxymethylcellulose;

Group III: aspirin (400 mg/kg body weight) suspended in 3 ml of 1% carboxymethylcellulose in water.

Group IV: ginger and 30 minutes later, received aspirin suspended in 1% carboxymethylcellulose, in similar doses as received in groups II and III.

Before treatment of ginger, Aspirin-induced a significant increase of the macroscopic ulcer score, and gastric damage observed by the shed and disrupted epithelium, mucosal hemorrhage, submucosal edema and leukocyte infiltration, loss of the mucus of the mucosal surface.

Furthermore, in the proteins perspective, Aspirin significantly increased expression of apoptosis regulator Bax, inducible nitric oxide synthase (iNOS) associated with the production of proinflammatory cytokines and heat shock protein 70 (HSP70) involved stress response.

Injection of ginger protected the gastric integrity by restoring all damage caused by aspirin in the induction of gastric ulcer.

Dr. Salah Khalil M, said, "Ginger ameliorated the histological changes by reducing Bax and iNOS and increasing HSP70 expressions".

In other words, the overexpression of proteins associated with the onset of gastric ulcer also was significantly reduced by the ginger administration.

Take all together, ginger may be considered a functional alternative for the treatment of gastric ulcers, pending to the confirmation of large sample size and multicenter human study.
