#Emodin Found in #Herbal #CascaraSagrada Exihibits Anti #BreastCancer Activities by Growth Inhibition, Apoptosis Induction, and the Expression of Apoptosis-related Genes

Kyle J. Norton

Breast cancer is a medical and chronic condition characterized by cell growth irregularly due to the alternation of DNA.
Most cases of breast cancer start in the cells on the surface of the inner lining of the breast tissue, including the milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.

At the advanced, the large tumor may press on the nearby blood vessels and nerve cells, leading to bleeding and severe pain. Cancer cells from the breast can also travel a distance away to infect other healthy tissue and organs at this stage.

The most common breast cancer secondary metastasis is in the liver, brain, bones, or lungs.

According to the statistics, in 2010, over 250,000 new cases of breast cancer were diagnosed in women in the U.S. alone and the risk of getting invasive breast cancer during a lifetime of a woman is 1/8.

Researchers do not know the exact causes of breast cancer. However, family and personal history, women with dense breast tissue, use of hormone replacement therapy and oral contraceptives, genetic preposition, particularly BRCA1 and 2, and tall adult height are some prevalent risk factors of the disease.

Some researchers suggested that the increase in the prevalence of breast cancer in overweight and obesity is also a major concern of the scientists community.

Dr. Manuel Picon‐Ruiz, the lead scientist in the observation of obesity in the risk of breast cancer wrote, "Obesity is associated both with a higher risk of developing breast cancer, particularly in postmenopausal women, and with worse disease outcome for women of all ages" and "... the effects of obesity to increase local and circulating proinflammatory cytokines, promote tumor angiogenesis and stimulate the most malignant cancer stem cell population to drive cancer growth, invasion, and metastasis".

And, "The effects of recent lifestyle interventions to decrease sex steroids, insulin/insulin‐like growth factor‐1 pathway activation, and inflammatory biomarkers associated with worse breast cancer outcomes in obesity also are discussed. Although many observational studies indicate that exercise with weight loss is associated with improved breast cancer outcome".

The results strongly suggested that the risk of breast cancer is significantly elevated in the obese population compared to healthy adults.

Cascara sagrada is a species of buckthorn, genus Rhamnus, belonging to the family Rhamnaceae, native to western North America, its bark has been used in traditional medicine for gastrointestinal support. and it is thought to have a laxative and natural cleansing, etc.

The chemical constituents of Cascara Sagrada include Aloe-emodin, Anthrquinone glycosides (Barbaloin Cascarosides A, B, C and D ChrysaloinFrangul), Chrysophanol, Emodin, Heterodianthrones palmidin A, B, and C, dianthrones, Oxanthrone, Rhein, Tannins, etc.

Researchers on clarification of the effects of a bioactive ingredient emodin found abundantly in herbal Cascara Sagrada and Rhubarb. against breast cancer, examined the compound activity on estrogen receptor α (ERα).

Application of emodin demonstrated the capability in inhibiting breast cancer cell proliferation by suppressing the ER α protein levels, by blocking the ER α transcriptional activation.

Futhermore, emodin treatment led to the dissociation of heat shock protein 90 (HSP90) which plays a critical role in stabilizing the proteins against heat stress and ER α and increased ER α ubiquitination in initiation cell degradation.

Dr. Huang PH and colleagues in the final report wrote, "emodin,....specifically suppress breast cancer cell proliferation by targeting ER α protein stability through distinct mechanisms".

In order to reveal additional information on emodin in the inhibition of breast cancer, researchers investigated the anticancer effects of emodin on growth inhibition, apoptosis induction, and the expression of apoptosis-related genes in MCF-7 cells.

Emodin showed significant growth inhibitory effects on MCF-7 cells with IC₅₀ = 7.22 µg/ml (∼30 μM) and exerted a concentration-dependent inhibitory effect on the colony-forming ability of MCF-7 cells with IC₅₀ = 7.60 µg/ml (∼30 µM).

The bioactive compound induced breast cancer death observed by the single-strand DNA breakage and DNA fragmentation, in emodin-treated MCF-7 cells.

Moreover, emodin efficacy in causing breast cancer cells apoptosis was attributed to the activation of the gene expression involved in the modulation of the immune response to stress and inhibition of the proteins associated with cancer cell proliferation and survival.

Dr. Li WY, the lead scientist after taking into account co and confounders said, "Emodin could induce growth inhibition and apoptosis in MCF-7 cells through the modulation of the expression of apoptosis-related genes".

Taken altogether, herbal Cascara Sagrada (Rhamnus purshiana) processed bioactive emodin may be considered a functional remedy for the prevention and combined with primary therapy for the treatment of breast cancer, pending the confirmation of a large sample size and multicenter human study.

#Greentea #Catechins Inhibit #JuvenileArthritis by Restoring Some Biomarkers of Cytokine Gene, Researchers Say

Kyle J. Norton

Green tea may have a therapeutic and positive effect in reducing the risk, progression, and treatment of juvenile arthritis, some scientists suggested.

Juvenile arthritis is an inflammatory disease affecting the synovium in children aged 16 or younger.

In other words, it is an autoimmune disorder caused by the immune system attacking some of the body's own healthy cells and tissues, in this case, the immune system attacks the joint tissue in the juvenile.

Genes that are associated with the risk of juvenile idiopathic arthritis include HLA-A. HLA-B. HLA-DPB1. HLA-DQA1. HLA-DQB1. HLA-DRB1. IL2RA. IL6, according to the National Institute of Health.

According to the statistic, the most common form of juvenile idiopathic arthritis (JIA), affects over 50,000 children in the United States alone.

Juvenile Arthritis shares the common symptoms of adult arthritis, including pain, stiffness, swelling, fever, skin rash, and in serious cases. The condition can also cause some complications such as pericarditis, hemolytic anemia, macrophage activation syndrome (MAS), and endarteritis

In the article "Juvenile Arthritis: Early Signs and Symptoms" By Susan Bernstein wrote, "Juvenile arthritis includes many different diseases, but one common thread between them is that they can have serious, even life-threatening impacts on a young child".
Green tea, a precious drink processes a number of health benefits known to almost everyone in Asia and the Western world.

In the investigation of the effect of green tea extract on the oxidative state of the liver and brain of adjuvant-induced arthritic rats, a model for human rheumatoid arthritis with daily doses of 250 mg kg(-1) (59.8 mg catechins per kg) for 23 days, researchers at the State University of Maringá, found that treatment group expressed a significant activity in reducing the protein and lipid damage in liver, brain, and plasma, caused by ROS expression in the induction of oxidation which has been found with associated to the progression of arthritis.

Injection of green tea extract also enhanced the overexpression of antioxidant defenses, found in low levels in patients with arthritis through the restoration of the glutathione (GSH) associated with the important function in preventing damage to important cellular components caused by reactive oxygen species and protein thiol levels in reducing cellular oxidation through chelation and detoxified activities and improved natural antioxidant enzymes, produced by the body.

Furthermore, green tea extract also inhibited the levels of glucose 6-phosphate dehydrogenase, which is increased in the liver in patients with arthritis, one of the major risk factors for the development of arthritis.

These results suggested that the application of green tea exerted a significantly profound effect in the liver and brain of patients suffering from rheumatoid arthritis through reduced overexpression of oxidative stress in precipitated injury to lipids and proteins.

Some researchers suggested that by modifying the metabolic function of lipid and proteins, green tea extract exhibited a strong protective effect in reducing the symptoms and progression of arthritis.

According to the study by the University of Health System of green tea effect in patients with rheumatoid
arthritis, injection of green tea extract demonstrated a therapeutic effect on a patient's synovial fibroblast of the lining tissue which surrounds the capsule of the joint by inhibiting the production of IL-1B, prototypic proinflammatory cytokines in response to acute and chronic inflammation. Overexpression of IL-1B is associated with the contribution of joint damage.

Interestingly, in the study of adolescent male Wistar rats with 12-week exposure to Cd and Pb (7mg Cd and 50mg Pb in 1kg of the diet randomly assigned to 12 to each group, as positive control received without Cd, Pb, and teas, a negative control group received Cd and Pb, and groups supplemented additionally with green (GT), black (BT), red (RT), and white tea (WT), researchers indicated that application of tea significant inhibits the decreased levels of the geometric(shape) and densitometric(density) parameters and a total thickness of articular cartilage caused by Cd and Pb in compared to the negative control group.

The tea injection group also displayed a huge improvement in mechanical endurance, growth plate thickness, and trabecular histomorphometry depending on the tea type.

Due to the limitation of the study, Dr. Tomaszewska E, the lead researcher said, " It is difficult to indicate which tea has the best protective effects on bone and hyaline cartilage against heavy metal action".

More importantly, in the examine some major factors contributing to juvenile arthritis, green tea rich in catechins (Healthy green tea) in animal studies, green tea injection restored these biomarkers of cytokine genes (IL-1A, IL-2, IFN-alpha, FGF-alpha, TNF-alpha) to near normal levels, thus preventing or attenuating the development of a certain type of inflammatory arthritis.

Taken all together, green tea and its bioactive polyphenols may be considered a functional food for reduction of the early onset, and progression and combined with standard therapy for the treatment of juvenile and adult arthritis.

However, the intake of green tea supplements should be taken with exceptional care as acute liver toxicity has been reported in a number of medical literature.

#Avocado May Be Considered a Novel Lipophilic Antioxidants in a Food Matrix, researchers Show

Kyle J. Norton

In the search for natural therapy for the prevention and treatment of diseases that involved oxidative stress, scientists may have found a dietary whole fruit with efficacy compared to that of conventional medicine, according to studies.
Antioxidants are stable atoms that inhibit free radicals and stop the free radicals chain reaction.

Free radicals on the other hand are unstable and highly reactive molecules with a single or unpaired electron on the outer ring. In order to maintain the balance, free radicals are ready either to donate or intercept an electron from another stable atom, leading to domino effect which can not be stopped until the electron is paired or inhibited by an antioxidant.

Overexpression of free radicals and suppressed levels of antioxidants cause oxidative stress, leading to the damage of cell structure, protein, and lipid, and alternation of cell DNA.

Every day, the human body is bombarded by the invasion of free radicals. Some foods we eat, the polluted air we breathe, the use of toxic chemical substances, and the body's metabolism all contribute to the expression of free radicals in our body.

Luckily, for a healthy individual, our body's antioxidant enzymes are good enough to maintain the balance ratio, regardless of the levels of free radicals produced.

However, for people with suppressed antioxidant enzymes in the body, dietary sources of antioxidants from fruits and vegetables can be very helpful.

Researchers at the Karolinska Institutet suggested that a high intake of fruit and vegetables is related to low oxidative stress.

Dr. Rikard Åsgård, the lead scientist said, "Fruit and vegetable intake was inversely related to oxidative stress" and " An increased intake of fruit and vegetables can, therefore, be beneficial for patients with type 2 diabetes since these patients are documented to have raised oxidative stress and inflammation".

The results clearly encouraged that people who suppress antioxidants produced by the body should add some portions of fruits and vegetables into their diet.
Avocado is a commercially valuable fruit cultivated in tropical climates throughout the world. It is a green-skinned, pear-shaped fruit that ripens after harvesting and is native to the Caribbean, Mexico, South America, and Central America, belonging to the flowering plant family Lauraceae.

Chemical constituents
The fruit contains campesterol, high amounts of β-sitosterol (average 76.4 mg/100 g); fatty acids (approximately 60% monounsaturated, 20% saturated, and 20% unsaturated); high amounts of glutathione (27.7 mg/100 g); approximately 2% protein; 6–9% carbohydrates and sugars (glucose, fructose, d-mannoheptulose, a taloheptulose, and an alloheptulose); two bitter substances (1-acetoxy-2,4-dihydroxyheptadeca-16-ene and 1,2,4-trihydroxyheptadeca-16-ene); carnitine; proanthocyanidins; persenones A and B.

With an aim to find natural ingredients with significant antioxidant effects against bacterial infection in the induction of oxidative stress, researchers examine the thyl acetate, containing 70% acetone, and 70% methanol extracts of the peel, pulp, and seed from two avocadi (Persea Americana Mill) against Gram-positive bacteria.

Peels and seeds were found to possess higher amounts of phenolics and had a more intense in vitro antioxidant potential, compared to the pulp.

The efficacy of the peels and seeds against Gram-positive bacteria was associated with the rich in catechins, procyanidins, and hydroxycinnamic acids, whereas the pulp was particularly rich in hydroxybenzoic and hydroxycinnamic acids and procyanidins.

The results indicated that avocado displays moderate antimicrobial effects against Gram-positive bacteria.

Furthermore, injection of the extracts (70% acetone) from avocado peels and seeds as inhibitors of oxidative reactions in meat patties, researchers also found that avocado extracts protected meat lipids and proteins against oxidation with the effect on lipids being dependent on the avocado variety.

Moreover, in the examine the antioxidant activity of other compounds such as monounsaturated fatty acids, tocopherols, carotenes, acetogenins and sterols found in avocado researchers once again confirmed the antioxidant property by comparing the lipophilic AOC of the pulp and the fruit hydrophilic AOC.

According to the chemical assay of oxygen radical absorbance capacity (ORAC), the highest antioxidant activity of lipophilic AOC was attributed to the presence of high levels of acetogenins, compared to other compounds.

The findings showed that the presence of emulsifying agents which are soluble in both fat and water could serve as novel lipophilic antioxidants in a food matrix.

Interestingly, the University of California in the study "The effect of harvest date on nutritional compounds and antioxidant activity (AOC) in avocado (Persea Americana Mill. cv Hass) fruit", suggested that AOC in early harvested fruit after storage for 35 days was much higher antioxidant contents than that in late-harvested fruit after storage for 21 days.

Taken together, there is no doubt that whole fruit avocado processed strong antioxidant properties may be considered a functional food for the prevention and treatment of oxidative stress-related diseases

Decreased intake of Maternal Coffee Intake Linearly Is Associated with Reduced Risk of #Miscarriage, Scientists Suggest

Kyle J. Norton

The risk of miscarrying is increased substantially in excessive (over 4 cups/day) coffee drinkers during pregnancy, an Asian study suggested.
A miscarriage, according to the definition is the loss of a fetus before the 20th week of pregnancy.

Believe it or not, according to medical literature, as many as 50% of all pregnancies end in miscarriage and over 80% of miscarriages are occurring within the first three months of pregnancy.

Some researchers suggested that women who carry mutation genes associated with chromosome gains and losses during embryonic development can lead to miscarriage or the failure of IVF treatments.

Other risk factors associated with an increased risk of miscarriage include aging Age. Women who are older than age 35, have previous miscarriages. smoking and excessive alcohol drinking before and during pregnancy have a higher risk of miscarriage compared to younger women,

Chronic medical health conditions such as obesity and uterine or cervical problems can also substantially increase the miscarriage risk.

Coffee, one of the most popular and social beverages all over the world, particularly in the West, is a drink made from roasted beans from the Coffea plant, native to tropical Africa and Madagascar.

In a case-controlled study conducted in Milan of 561 (72%) cases of spontaneous abortion and 877 (57%) controls, coffee drinking during the first trimester of the index pregnancy with healthy women who gave birth at term (> 37 weeks gestation) to healthy infants as control, researchers reported that the risk of miscarrying is classified as follows
* Coffee intake of 1, 2 or 3, and 4 or more cups of coffee per day are associated with an increased risk of 1.2, 1.8, and 4.0, respectively, compared to non-drinkers.

* The risk of pregnancy loss also is associated with the duration of years of coffee drinking. The estimated multivariate odds ratios of spontaneous abortion were 1.1 in comparison to the non-coffee intake group.

Furthermore, in the evaluated effect of caffeine in the influence of miscarriage researchers at the Boston University School of Public Health, conducted a prospective cohort study that included 5132 Danish women planning pregnancy, enrolled from 2007 to 2010, after carefully assessing the returned beverage consumption on questionnaires, a researcher found that a significantly miscarried risk is 1.62 higher in women consuming more than 300 mg/day of caffeine per day, in compared to 1.09 in women who consume <100 mg/day. Specifically, women with a daily intake of caffeine of
* <100 mg/day showed a materially associated SAB risk of 1.09.

* 100-199mg/day showed a materially associated with SAB risk of 0,89.

* 200-299mg/day showed a materially associated with SAB risk of 1.33

Additionally, in prospective cohort study included 15,590 pregnancies from 11,072 women with no history of SAB in the Nurses' Health Study II (1991-2009) with beverage intake assessed every 4 years using a validated questionnaire in case pregnancies lost spontaneously at <20 weeks gestation, total coffee intake had a positive, linear association with SAB.

In comparison to non-pre-pregnancy coffee intake, the risk of SAB increased by 20% for women consuming ≥4 servings/day.

More importantly, Dr. Gaskins AJ, the lead author said, "Pre-pregnancy coffee consumption at levels ≥4 servings/day is associated with the increased risk of SAB, particularly at weeks 8-19".

Taking together, there is no doubt that coffee may contribute to a relative risk of miscarrying for women consuming more than 4 cups per day.

Women who are pregnant between 12 to 28 weeks of gestation with excessive coffee intake habits are recommended to consult with their doctors.

#Fennel Reduces the Risk of Diseases Caused by High Blood Cholesterols, Researchers Find

Kyle J. Norton

Scientists may have found a natural whole food with the potential to treat hyperlipidemia without inducing any side effects, some studies suggested.

Cholesterol, the natural ingredient produced in the liver plays an important role in stimulating the production of vitamin D, steroid hormones, and bile acids and maintaining healthy cell membranes.
Hyperlipidemia is a condition characterized by abnormally elevated levels of cholesterol in the plasma of the blood.

The causes of hyperlipidemia are unknown. However, an unhealthy diet with a lot of unsaturated fat and trans fat is associated with the early onset of the disease. Foods such as cheese egg yolks fried and processed foods and red meat have been found to increase blood cholesterol, and long-term intake of such foods in great amount have been found to increase the risk of hyperlipidemia, epidemiological studies found.

The Beni-Suef University, in the study to determine the development of obesity in response to a high-fat diet (HFD) an Egyptian Herbal mixture formulation (HMF), including fennel, showed, "feeding an HFD diet significantly increased final body weight, triglycerides (TG), total cholesterol, & LDL concentration compared with controls, while significantly decreasing HD".

Hyperlipidemia is a lifelong condition that is treatable.

Dr. Robert B. Kelly at the Fairview Hospital/Cleveland Clinic Family Medicine Residency Program in the article "The most beneficial changes result from reducing intake of saturated and trans fats; increasing intake of polyunsaturated and monounsaturated fats; fortifying foods with plant stanols or sterols; isocalorically adding tree nuts to the diet; consuming one or two alcoholic drinks per day; and adopting a Portfolio, Mediterranean, low-carbohydrate, or low-fat diet. Smaller but still beneficial effects result from reducing intake of dietary cholesterol, increasing intake of soluble fiber and soy protein, and eating fatty marine fish or taking marine-derived omega-3 fatty acid supplements".

Therefore changing the diet pattern and engaging in regular physical exercise are the fundamental
options that not only can prevent the onset of the condition, particularly in people who are at a higher risk of hyperlipidemia, and also treat high blood cholesterol in patients who already have the condition.

Fennel (Foeniculum vulgare) is a plant species of a genus, belonging to Apiaceae (Umbelliferae), native to the Mediterranean.

The plant has been used in traditional and herbal medicine as a warming, carminative, antispasmodic, and depressant agent and to stimulate the appetite, ease indigestion, soothe coughing, reduce intestinal spasms, to regulate the menstrual cycle, and relieve PMS.

In the study to examine the effect of the aqueous extract of fennel on the reduced risk of hyperlipidemia, researchers conducted an experiment in Triton WR-1339-induced hyperlipidemia in mice.

Mice treated with the extract at different doses display a significant reduction of cholesterol, triglycerides, LDL-cholesterol, and apolipoprotein-B decreased by 40%, 23%, 61%, and 61%, respectively.

Further analysis, the application also increased the levels of HDL-cholesterol and apolipoprotein A1 by 85% and 58%, respectively

The Mashhad University of Medical Sciences also conducted a study to assess the effects of Fennel (Foeniculum vulgare Mill.) on lipid profiles, in sixty eligible postmenopausal women, randomly assigned into the fennel and placebo groups.

Measurement of total blood cholesterol, cholesterol fractions, and triglycerides was tested at the baseline, and after a three-month follow-up.

At the end of 3 months, blood profiles withdrawn from the women suggested
* There are no significant difference in triglyceride (P = 0.679), total cholesterol (P = 0.103), low-density lipoprotein cholesterol(LDL-C; P = 0.146) and high-density lipoprotein cholesterol (HDL-C; P = 0.266) levels between the two groups.

* Fennel treated group demonstrates a significant borderline improvement for HDL-C and triglyceride, compared to the placebo group.

In additional differentiation of the process of the double-blind, randomized, and placebo-controlled trial, researchers suggested that longer durations, higher doses, and larger sample sizes are recommended to validate the results.

These results indicated that the intake of foods for the treatment of conditions or diseases must be on a long-term basis. The time of 3 months may be too short for foods to exert their potential effects.

Taken together, fennel may be considered a functional food for the prevention and treatment of hyperlipidemia. Intake of fennel should be increased gradually for the food to reach its antihyperlipidemic stage, a longer duration may be needed. In other words, please be patient.

#GreenTea Inhibits #GoputyArthritis Through Its Effects on Acute Phrase of Inflammation, Researchers Reveal

Kyle J. Norton

Green tea may have a therapeutic and positive effect in blocking the onset and treatment of gout and recurrent gout, some scientists suggested.

Gout is a condition of recurrently inflammatory arthritis characterized by a red, tender, hot, and swollen joint caused by overexpression of purine in the body.

Gout is caused by an excess of uric acid in the blood, or hyperuricemia. Uric acid is a by-product that is produced during the digestive process in the breaking down of chemical ingredients purines.

Purines are natural nitrogen-containing compounds found in the cell DNA and RNA, including adenine (6-aminopurine) and guanine (2-amino, 6-oxypurine), which are made inside the cells of body.

Under normal conditions, uric acid is eliminated by the kidney through urination. However, a high uric acid level can occur when the kidneys don't eliminate uric acid efficiently. People with kidney deficiency are associated with high levels of uric acid if foods with high levels of purines are consumed in large amounts, including red meat, seafood, and mushroom.

Some researchers suggested that people who are obese, overweight with diabetes, long-term use of diuretics, and excessive alcohol drinking are associated with the risk of high levels of uric acid in the blood.

Additionally, genetic mutations inherited from the parents such as Q141K polymorphism and CFTR ΔF508 certain medical conditions such as tumor lysis syndrome, hypothyroidism, and psoriasis are also some major risk factors for causing excessive levels of uric acid in blood.

According to the joint study led by the University of Nagasaki, green tea and its bioactive compounds Epigallo catechin-O-gallate (EGCG) may be considered as a putative curative drug for various diseases of gout, through its properties in reducing the vascular endothelial growth factor receptor 2 (VEGFR2) in response to vascular endothelial growth factor signal (VGFG) for the formation of gout through an expression of levels of uric acid.

The study also emphasized that green tea with high levels of antioxidants and function in the stimulated production of natural antioxidants in the body may also be used to reduce symptoms of recurrently inflammatory arthritis and inflammatory arthritis such as gout, through inhibiting the oxidative stress in the induced production of inflammatory cytokines to facilitate symptoms of inflammation.

The above differentiation was supported by the study of the effect of EGCG on monosodium urate monohydrate (MSU)-induced inflammation and cryopyrin (NLRP3 inflammasome) activation in C57BL/6 mice received a subcutaneous injection or oral gavage of EGCG before the intraperitoneal injection of MSU.

Truly, the application of green tea EGCG decreased the expression of gouty arthritis caused by the injection of MSU-induced neutrophil infiltration with a function to initiate inflammation in the acute phase of infection and IL-1β secretion, in response and resistance to pathogens, but can also exacerbate damage during chronic disease and acute tissue injury.

The injection of green EGCG also decreased MSU-triggered neutrophil cytosolic factor 1, in the activated inflammatory response to optimize healing and reduce injury to the body but the proteins have been found to cause severe damage to the infectious site and NLRP3 protein expression in initiated systematic inflammatory expression.

Moreover, the effect of green tea in protection against gout occurrence also limited pro-inflammatory mediator secretion interleukin 1 beta (IL1β), a mediator of inflammation, in response to acute phase infection, interleukin 6 (IL-6) produced by large amounts at sites of acute and chronic inflammation, monocyte chemoattractant protein-1 in regulated monocytes, memory T cells, and dendritic cells to the sites of inflammation, and serum amyloid A, the serum was found to increase substantially in patients of chronic inflammatory disease, without limiting the function of the mediators in protect the infectious site against pathogens invasion.

More profoundly, the efficacy of green tea in reduced recurrent gout also extended to suppress NLRP3 inflammasome in stimulated caspase-1 in the promoted secretion of other proinflammatory cytokines activation.

Dr. Jhang JJ, the lead author said, "EGCG treatment ameliorates MSU-induced inflammation, suggesting that EGCG exerts anti-inflammatory effect against MSU-induced acute gout attack."

Additionally, the University of Pretoria suggested that polyphenols extracted from green tea also exhibited an antioxidant effect in reducing the pronounce of oxidative stress by their antioxidant properties.

Catechin (C) (Ki = 303.95, uncompetitive), epicatechin (EC) (Ki = 20.48, mixed), epigallocatechin (EGC) (Ki = 10.66, mixed), epicatechin gallate (ECg) (Ki = 2.86, mixed) and epigallocatechin gallate (EGCg) (Ki = 0.76, competitive) were found to have similar efficacy as allopurinol (Ki = 0.30, mixed), the drug used to inhibit the levels of xanthine oxidase (XO) in gout patients, through reducing reactive oxygen species in the initiation of oxidative stress that facilitates the inflammatory symptoms.

Taken together, green tea and its bioactive polyphenols may be considered a functional food for reduced onset and combined with conventional medicine for the treatment of gout with no adverse effects.

Herbal Cascara sagrada processed with a bioactive compound emodin may have a significant and potential effect in reducing the risk and treatment of liver cancer, according to studies.
Liver cancer is a medical and chronic condition caused by irregular cell growth in the liver tissues, due to the alternation of cell DNA.

The liver organ is one of the organs with a lack of nerve tissue, therefore liver cancer does not induce pain until the later stage.

At the advanced stage of liver cancer, the cancer cell may travel a distance away to infect other tissues and organs, leading to secondary metastasis.

According to the statistics provided by the Canadian Cancer Society, every year, approximately, 1,900 men and 580 women are diagnosed with liver cancer, causing the death of 950 men and 270 women.

In 2017, 2,500 Canadians were diagnosed with liver cancer and 1,200 Canadians died from liver cancer.

Compared to the early stage of other cancer, the 5 years survival rate in the early stage diagnosis is only 31%, and 43% of people with liver cancer are diagnosed at stages 0 and 1.

The exact causes of liver cancer are unknown. However, chronic infection with hepatitis virus, cirrhosis, genetic preposition, a medical condition of diabetes, nonalcoholic fatty liver disease, and excessive alcohol consumption are some of the most prevalent risk factors of liver cancer, epidemiologically.

Dr. Chuang SC, the lead scientist in the concern of excessive alcohol drinking in risk of liver cancer, wrote, "Compared to individuals who never drank or drank at very low frequencies, the meta-relative risk (mRR) forever drinkers was 1.29 (95% confidence interval, CI 1.16-1.42) and 1.46 (95% CI 1.27-1.65) for case-control studies".

And, "The dose-response relation between alcohol and liver cancer was apparent with RR = 1.08 (95% CI 1.04-1.11) for 12 g/day (~1 drink), 1.54 (95% CI 1.36-1.74) for 50 g/day, 2.14 (95% CI 1.74-2.62) for 75 g/day, 3.21 (95% CI 2.34-4.40) for 100 g/day, and 5.20 (95% CI 3.25-8.29) for 125 g/day of alcohol consumption".

The finding clearly suggested, one alcoholic drink per day (~12 g/day) may be associated with 1.1 times higher liver cancer risk and the risk of liver cancer increases proportionally to the volume intake per day.

Cascara sagrada is a species of buckthorn, genus Rhamnus, belonging to the family Rhamnaceae, native to western North America. The herbal bark has been used in traditional medicine for gastrointestinal support. and it is thought to have a laxative and natural cleansing, etc.

The chemical constituents of Cascara Sagrada include Aloe-emodin, Anthrquinone glycosides (Barbaloin Cascarosides A, B, C and D ChrysaloinFrangul), Chrysophanol, Emodin, Heterodianthrones palmidin A, B, and C, dianthrones, Oxanthrone, Rhein, Tannins, etc.

Researchers on finding a natural compound for the treatment of liver cancer examined the effect of emodin, a bioactive ingredient isolated from herbal Cascara Sagrada against the CXCL12-CXCR4 signaling cascade which plays an important role in the process of invasion and metastasis of liver cancer.

Where The CXCL12/CXCR4 axis is a pathway involved in tumor progression, angiogenesis, metastasis, and survival.

Injection of emodin inhibited the expression of CXCR4 in a dose and time-dependent manner in HCC cells.

Treatment of emodin demonstrated a better effect compared to the pharmacological proteasome and lysosomal inhibitors on the decrease in CXCR4 expression.

The suppression of CXCR4 expression was attributed to several mechanisms, including the inhibition of cancer cell's mRNA expression, a protein associated with cancer cell proliferation, and the production of antigens by innate immunity.

Furthermore, emodin treatment significantly suppressed metastasis to the lungs in an orthotopic HCC mice model and CXCR4 expression in tumor tissues.

In other human cancer cell lines, emodin-induced apoptosis in HepG2 cells through cell cycle arrest.

According to the Western blot data, emodin inhibited cancer cell growth through a set of mechanisms, including cell degradation and protein associated with cell death programming.

Moreover, the application of emodin increased the expression in the initiation of cytotoxicity against HepG2 without causing harm to nearby healthy cells.

Additionally, emodin increased the protein p53 associated with tumor suppression and decreased the protein involved in cancer cell proliferation in HepG2 cells.

These results suggest that emodin induces apoptosis in the liver cancer cell lines through a multifaceted complex cascade of events.

Taken altogether, herbal Cascara Sagrada processed a major bioactive compound emodin may be considered a functional remedy for the prevention and treatment of liver cancer, pending the confirmation of a large sample size and multicenter human study.
