#CruciferousVegetables and Celecoxib Modulates Oncogenic Activity Against the Onset of #BladderCancer, Researchers Say

Kyle J. Norton

Allyl isothiocyanate is a phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in horseradish, mustard, wasabi, etc.
The bladder is a hollow elastic organ in the center of the lower abdomen that collects urine from the kidneys and excretes it through the urethra.

Bladder cancer is the most common type of cancer that affects men twice as frequently as it affects women. Usually, it starts from the bladder lining caused by several types of malignant growths of the urinary bladder cells.

The study of AITC found cruciferous vegetables and celecoxib effects in modulating cyclooxygenase-2 (Cox-2), as its oncogenic activity has been well documented in bladder cancer and other cancers, this effectiveness is a result of associated with depletion of prostaglandin E2, a key downstream signaling molecule of Cox-2, caspase activation and downregulation of vascular endothelial growth factor in the tumor tissues.

In another study, the N-acetylcysteine conjugate (NAC-AITC) is found to be a significant inhibition of cell growth and proliferation in human bladder cancer UM-UC-3 cells or rat bladder cancer AY-27 cells with NAC-AITC at 15 μM.

#GreenPeas (Pisum Sativum) Supplementation Ameliorates the Risk and Severity of #Colitis, Researchers Say

Kyle J. Norton

Colitis is a general term associated with the inflammation of the inner lining of the colon.
The disease can be caused by infection, inflammatory bowel disease (Crohn's disease (CD), ulcerative colitis(CD)), ischemic colitis, allergic reactions, and microscopic colitis.

Most cases of colitis are involved in the damage of the intestinal mucosal barrier, particularly to those of ulcerative colitis.

Epidemiologically, damage and a reduction of gut bacteria by dietary chemicals, such as antibiotics of the protective mucus layer and the underlying tissue by the poorly inactivated digestive proteases have been found to induce both UC and CD.

The intestinal mucosal barrier is the outermost inner layer of the lining of the intestines which plays an essential role in ensuring the adequate containment of undesirable luminal contents within the intestine while preserving the ability to absorb nutrients.

Additionally, the intestinal mucosal barrier also processes various immunological mediators, including cytokines and chemokines stimulated by gut microbiota and host immune responses, thus maintaining a well-balanced relationship between gut microbes and the host immune system.

In other words, the intestinal mucosal barrier protects the colonic integrity by interacting with the gut microbiota and the immune response to prevent the inflammation of the colon that causes the onset of colitis.

Some researchers suggested the promotion of a high-fat diet over the past few decades may be one of the most prevalent factors that increase the risk of colitis.

Dr. Max Gulhane, the lead scientist wrote, "Prolonged high-fat diets (HFD) induce low-grade chronic intestinal inflammation in mice, and diets high in saturated fat are a risk factor for the development of human inflammatory bowel diseases".

And, "HFD-induced endoplasmic reticulum (ER)/oxidative stress occur in intestinal secretory goblet cells, triggering inflammatory signaling and reducing synthesis/secretion of proteins that form the protective mucus barrier".

The finding strongly suggested the mucosal barrier dysfunction induced by a high-fat diet involved in colitis.

Green peas are a species of Pisum Sativum belonging to the family Fabaceae and native to western Asia from the Mediterranean Sea to the Himalayan Mountains. The pea is a green, pod-shaped vegetable and a cool-season crop grown in many parts of the world.

In finding a potent ingredient that processes anti-inflammatory activity with oxidative stress, researchers investigated the impact of green pea (GP) supplementation on the susceptibility of a high-fat diet (HFD) in an animal model.

The study included Six-week-old C57BL/6J female mice randomly assigned into groups fed a 45% HFD or HFD supplemented with 10% GP. After 7-week dietary supplementation, colitis induced by adding 2.5% DSS in drinking water for 7 days followed by a 7-day recovery period.

According to the tested analysis, GP supplementation ameliorated the disease activity index score in HFD-fed mice by reducing the expression of pro-inflammatory cytokines.

Furthermore, inflammatory markers associated with inflammatory proteins and enzymes such as interleukin (IL)-6, cyclooxygenase-2 (COX-2), IL-17, interferon-γ (IFN-γ), and inducible nitric oxide synthase (iNOS) in HFD-fed mice were also decreased by GP supplement.

Goblet cell differentiation markers in HFD-fed mice. associated with mucus secretion and function during the stage of inflammation were also increased by the administration of GP supplementation.

Furthermore, endoplasmic reticulum (er) stress-induced production of pro-inflammatory cytokines in HFD-fed mice was also inhibited by GP supplement.

Based on the findings, researchers wrote, "GP supplementation ameliorated the severity of DSS-induced colitis in HFD-fed mice, which was associated with the suppression of inflammation, mucin depletion, and ER stress in the colon".

Taken together, green peas may be considered a functional food for the prevention and treatment of colitis, pending the confirmation of the larger sample size and multicenter human study.

Regular Intake of #Onion Is Associated with a Reduced Risk of #Cancer, According to Studies

Kyle J. Norton

Cancer is a group of medical conditions caused by irregular cell growth in the tissues and organs due to the alternation of cell DNA.
Most cases of cancer start in the cells on the surface of the inner lining of the infected tissue or argan before penetrating into the deeper layers to form a tumor.

Certain factors have been found to affect prognosis including
* The type of cancer and where it is in your body

*The stage of cancer, most often refers to the size and metastasis of cancer.

** Stage 0
Cancer is classified as Stage 0 if the cancerous cell has not penetrated deeper tissue but on the surface of the inner lining.
** Stage I
In stage I, The cancerous cells are no longer in the surface but have invaded the deep lung lining, but still completely inside the lung.
***Stage IA1
The cancer is ≤ 3 mm (1/8 inch) deep and ≤ 7 mm (1/4 inch) wide.
**** Stage IA1: The spreading is < 3mm(1/8 inch) deep and & less than 7mm (1/4 inch) wide.
**** Stage IA2: The invasion area is ≥ 3 mm but ≤ 5 mm (about 1/5 inch) deep and & less than 7 mm (about 1/4 inch) wide.
***. Stage IB: cancer in this stage has invaded the connective tissue, & less than 5mm (1/5 inch).
**** Stage IB1: Cancer is 4 cm large (1 3/4 inches).
**** Stage IB2: Cancer is ≥ 4 cm (1 3/4 inches) but & less than 5cm (1/5 inch)

**Stage II
In stage II, the cancerous cells have spread to distant tissues but are still within the organ.

**Stage III
At this stage, cancerous cells have spread to the tissues immediately surrounding the organ.

**Stage IV
The stage, IV is the most advanced stage of cancer as cancerous cells have spread to distant parts of the body

In this stage, cancerous cells have spread to the tissues immediately surrounding the organ.

* Cancer’s grade, low-grade cancers usually grow more slowly and are less likely to spread while high-grade cancer indicates otherwise.

* Certain traits of cancer cells include
** How cancer cell stimulates their own growth

** How cancer cells resist inhibitory signals that stop their growth

** How cancer cells inhibit programmed cell death signaling.

Some researchers also include the age, health of patients, and how patients responded to previous cancer treatment

The onion is a plant in the genus Allium, belonging to the family Alliaceae, closely related to garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

To find a potential compound for the treatment of cancer, researchers examined the effect of garlic and onion on certain types of cancer.

Intake of onion is found inversely associated with the risk of onions or garlic and cancers of the oral cavity/pharynx, larynx, renal cells, breast, ovary, and endometrium.

More precisely, people who eat onions ≥7 times/week have the OR of oral cavity/pharyngeal cancers of 0.16, laryngeal cancer of 0.17, and ovarian cancer of 0.27 compared to those who did not eat onions.

Furthermore, according to the study of cases from the Hawaii Tumor Registry and controls randomly selected from a list of Oahu residents, intake of onion was associated with decreased risk of lung cancer with the OR = 0.5 in the highest quartile of intake compared to the lowest.

In prostate cancer, the Italian study suggested that both onions and garlic did not have significantly decreased OR of prostate cancer compared to those with the lowest intake, However, increasing intake showed a trend of decreasing the ORs.

In esophageal cancer, the intake of raw garlic/onion at least once per week has the adjusted OR of esophageal cancer of 0.2.

In colorectal cancer, the Italian and Swiss case-control studies found that both onions show a protective effect against colon cancer regardless of the amount of intake onion intake compared to non-consumers of onion.

In stomach cancer, according to the meta-analysis, consumption of large amounts of total Allium vegetables, such as garlic and onion reduced the risk of gastric cancer by comparing the highest and lowest consumption groups.

Taken together, onion may be considered a remedy for cancer prevention, pending confirmation of the larger sample size and multicenter human study.

#Phytochemical #Betacyanins Reduce Chemo-Drug Toxicity and Overcome Multidrug Resistance of Cancer Cells, Researchers Say

Kyle J. Norton

Cancer is a class of disease characterized by cell growth disorderly and uncontrollably in the tissues and organs.
The most common cancers are
* Breast cancer, accounted for 25.4% of the total number of new cases diagnosed in 2018.

Breast cancer starts in the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk. there are also rare cases of breast cancer that start in other areas of the breast. The risk of getting invasive breast cancer during the lifetime of women is 1/8.

Luckily, the 5 years survival rate of localized breast cancer is about 100%.

* Colorectal cancer and lung cancers are the second and third most common cancers, accounting for 43.9% of all cancers, excluding non-melanoma skin cancer.

Colorectal cancer is cancer in the colon, rectum, or vermiform appendix. Colorectal cancer is a relatively very common and slowly growing and progressing cancer.

The 5-year survival rate of the localized stage of colorectal cancer is about 90%.

Lung cancer is a condition of the abnormal growth of the cells in the lung's tissue. The most common form of primary lung cancer is derived from epithelial cells.

Lung cancer can also be classified into
* Non-small cell lung cancer
Non-small cell lung cancer is the most common type of lung cancer. It usually grows and spreads methodically and predictably.

* Small cell lung cancer
Small cell lung cancer is a fast-growing type of lung cancer, it can spread to distant parts of the body in a relatively early stage.

The overall 5-year survival rate of lung cancer is about 60%.

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In the finding, of the health benefits of Betacyanins, researchers compared the effect of green beet (Beta vulgaris var. cicla L.) and red beetroot (B. vulgaris var. Rubra L.) phytochemicals that benefit human health.

According to the chemical analysis, green beet contains apigenin, vitexin, vitexin-2-O-xyloside, and vitexin-2-O-rhamnoside, compared to the red beetroot as a source of betaxanthins and betacyanins.

All phytochemicals found in green and red beet show considerable antioxidant, anti-inflammatory, and antiproliferative activities.

From a cancer perspective, vitexin-2-O-xyloside combined with betaxanthins and betacyanins, exerts a significant antiproliferative activity in breast, liver, colon, and bladder cancer cell lines, through the induction of cancer cell apoptosis.

Additionally, betacyanins and other phytochemicals found in the green and red beet inhibited the pro-survival and anti-apoptotic genes, and modulated proteins associated with angiogenesis.

Moreover, betaxanthins and betacyanins used in combination with conventional anticancer drugs were found to reduce chemo-drug toxicity and overcome the multidrug resistance of cancer cells.

Based on the findings, researchers concluded betacyanins may be used alone or combined with other phytochemicals or chemo drugs to block the onset of the proliferation of tumor cells.

Taken together, betacyanins may be considered a remedy for the prevention and treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of betacyanins in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthyfood #Papaya Controls Glucose and Reduces the Risk of Complications in Diabetics, Researchers Say

Kyle J. Norton

Papaya may be the next generation of whole foods for the prevention and treatment of type II diabetes with no side effects, some studies suggested.
Diabetes is a condition characterized by insufficient insulin entering the bloodstream to regulate glucose.

Most cases of type II diabetes are either caused by cells in the pancreas that died off or receptor sites clogged up by fat and cholesterol.

In some cases, diabetes is also caused by allergic reactions of cells in the immune system.

Epidemiological studies suggested that obesity, family history, ethnicity, the increase in age, gestation diabetes, and polycystic ovary syndrome. and high blood pressure is the most common risk factor associated with the onset of prediabetes and type II diabetes.

Dr. Stephen S. Rich, in the article "the genetics of Diabetes and Its complications", suggested that type II diabetes may be a genetically related disease. Unlike type 1 diabetes, in which there seems to be an autoimmune process, type 2 diabetes is a disease of relative rather than absolute insulin deficiency. Genes involved in type 2 diabetes, with the most promising, replicated findings on chromosomes are 1q21-q24, 2q37, 12q24, and 20.

Untreated and unmanageable type II diabetes may induce complications of the onset of cardiovascular disease, and nerve damage (neuropathy). kidney damage (nephropathy), eye damage (retinopathy). foot damage and skin conditions.

Medication used for the treatment of type II diabetes is focused on alleviating the symptoms improving quality of life and slowing down the progression of the disease.

If you currently intake medicine for the treatment of diabetes, please sure you know all the side effects for the preventive measure.

Papaya is a species of Carica Papaya, belonging to the family Caricaceae and native to the tropics of the Americas, and was first cultivated in Mexico several centuries

Unripe pawpaw fruit (Carica papaya) not only inhibited α-amylase, and α-glucosidase but also reduced lipid peroxidation in a dose-dependent manner, in rat pancreas(1).

The key enzymes of α-amylase, α-glucosidase, involved in the digestion of carbohydrates, have been linked to the onset of type 2 diabetes. Inhibition of these enzymes can significantly reduce the postprandial increase of blood glucose and has been used for the management of blood glucose levels in type 2 diabetic and borderline patients by therapists (2).

In the streptozotocin-induced diabetic rats study, the aqueous extract of leaves of C. papaya administered strongly controlled blood glucose levels(3), through the prevention of hepatocyte disruption, as well as accumulation of glycogen and lipids via its hypoglycemic and antioxidant effect(4)

In a new study, conducted by researchers at the Davis Heart and Lung Research Institute, fermented papaya preparation was tested whether it improves dermal wound healing outcomes in adult diabetic mice by preventing the risk of infections(5).

The result was astonishing, the fermentation not only up-regulated the Sp1 DNA binding activity(modulating the cellular response to DNA damage) in Peripheral blood mononuclear cells (PBMC) but also the protein and mRNA expression of Rac2(regulates several cellular functions)(5).

A randomized controlled clinical trial conducted at the Cardiac Centre, ANDI Centre of Excellence for Biomedical and Biomaterials Research and Department of the Biosciences University of Mauritius, suggested that oral administration of 6g FPP®/day for 14 weeks could improve the general health status of several organs targeted by oxidative stress during diabetes and the supplement may be used in the management the complications of diabetes such as cardiovascular disease, neurological disease and other conditions worsened by overt inflammation and oxidative stress(6).

Taken together, unripe, fermented papaya and its' leaves may be effective in diabetic management by controlling the levels of glucose and reducing the risk of complications.

Some Western researchers insisted that large sample sizes and multicenter studies are necessary to validate its effectiveness by identifying the ingredients that are comparable to recent pharmaceutical medicine in use because geography, environment, time of harvest, etc. may influence the concentration of the food cultivation.

In fact, herbalists have long acknowledged that the external factors (mentioned above) may alter the quality of the herbs and foods by classifying these differences either through price or formula differentiation.

Indeed, the effectiveness with little or no adverse side effects of the formula is not the concentration of individual herbs and the herb itself but the interaction of all ingredients in an herb and herbs in the same formula. Extracting only one ingredient of the herb for the treatment of certain diseases may be effective but enhance complicated consequences.

#Cucumber Prevents Colonic Inflammation and Improves Digestive Functioning, Scientists Show

Kyle J. Norton

The gut microbiome made up of trillions of microorganisms is the bacterial composition found in the gut of the digestive tract.
In a healthy individual, the ratio of the bacterial composition is balanced. In other words, the bad bacteria in the digestive tract is totally countered by the presence of good bacteria.

However, the imbalanced ratio of the composition of the gut microbiota has been associated with chronic diseases ranging from inflammatory bowel disease (IBD), obesity, and allergic disorders, in both human and animal models.

Believe it or not, gut microbes have been found to influence many aspects of human health including immune, metabolic, and neurobehavioural traits.

In immunity, the balance of the gut microbes is shown to stimulate the immune function against inflammatory immune disorders.

Digestively, balanced gut microbes protect the gut against gastrointestinal diseases and non-gastrointestinal diseases. caused by overexpression of bad bacteria.

In metabolism, gut microbes influence what the human host is able to extract from its diet, such as nutrients, thus reducing the risk of obesity and obesity complications.

Neurologically, gut microbiota can control brain insulin signaling and metabolite levels that play a critical role in neurobehaviors.

Some researchers suggested, that there are huge benefits that influence the function of the brain, immunity, heart, and digestive system and reduce the risk of chronic diseases and conditions such as diabetes and metabolic syndrome by maintaining a good ratio of the gut microbiome.

The cucumber plant is a species of Cucumis Sativus, belonging to the family Cucurbitaceae and native to Western Asia. It is a creeping vine with roots in the ground and grows with the support of frames.

To discover a natural ingredient for balancing the ratio of microbiota composition, researchers investigated and evaluated cucumber phytochemical compositions along with their potential gut microbiota modulating, free radical scavenging, and anti-inflammatory capacities.

Seed flour extracts from cucumber enhanced the total number of gut bacteria by altering the abundance of specific bacterial phylum or genus in vitro.

In other words, cucumbers improved digestive functioning by enhancing the ratio of microbiota compositions in the gut.

Compared to broccoli and carrot seed flour extracts, cucumber seed extract showed the least effect in DPPH radical scavenging capacity, oxygen radical absorbing capacity, and hydrdoxyl radical (HO•) scavenging capacity values of 85, 634, and 270 μmol Trolox equivalent (TE)/g.

In the tested J774A.1 cells, cucumber suppressed the LPS-induced production of pro-inflammatory cytokines.

The results strongly suggested that the administration of Cucumis prevented colonic inflammation and improved digestive functioning by changing the gut microbiota composition.

To reveal more information about cumber in the improvement of gut microbiota, composition researchers examined Cucumis melo L. (Cucumis) on obesity-induced IR in genetically leptin-deficient Lepob/Lepob mice.

According to the tested assays, Cucumis improved IR and relieved inflammation in adipose tissue and blood which led to the balance of the ratio of microbiota composition.

Dr. Lee D, the lead scientist after taking into account co and confounders wrote, "administration of Cucumis improved IR by reducing inflammation, thereby changing the gut microbiota composition".

Taken together, fermented cucumber may be used for the treatment of digestive dysfunction by improving the ratio of gut microbiota composition, pending the confirmation of the larger sample size and multicenter human study.

#Onion's #Quercetin inhibits Parameters Associated with the Risk of #Hypertension In the Induction of #Cardiovascular Disease, Researchers Say

Kyle J. Norton

Hypertension is a condition of abnormally high blood pressure in the bloodstream. Hypertension a member of the cluster conditions of metabolic syndrome associated with an increased risk of cardiovascular disease and diabetes.
Blood pressure is the force of blood pushing against the walls of the arteries as the heart pumps out blood.

According to the statistics provided by the American Heart Association, approximately, 103 million U.S. adults have high blood pressure, or 1 in 2 adults in the US have the syndrome.

Obesity is a medical and chronic condition caused by long-term excessive body fat accumulated. Overweight is a condition of excess body weight relative to height.

According to the Body Mass Index(BMI), people who have a BMI between 25 to 29.9 are considered overweight and a BMI of over 30 is an indication of obesity.

The formula of the BMI indexBMI= weight (kg)/ height (m2)

According to the statistics, 68% of the American population is either overweight or obese. However,
in 2016, more than 1.9 billion adults, 18 years and older, were overweight, including over 650 million who were obese worldwide.

Bad eating habits, unhealthy lifestyles, and certain medical conditions such as kidney disease, diabetes, and sleep apnea not only induce overweight and obesity but also increase the risk of high blood pressure.

Dr. Shu-Zhong Jiang, the lead scientist wrote, "The imbalance between energy intake and expenditure is the main cause of excessive overweight and obesity. Technically, obesity is defined as the abnormal accumulation of ≥20% of body fat, over the individual's ideal body weight".

And, "Excessive overweight remains among the most neglected public health issues worldwide, while obesity is associated with increased risks of disability, illness, and death. Cardiovascular diseases, the leading cause of mortality worldwide, particularly hypertension and diabetes, are the main illnesses associated with obesity".

The results strongly suggested that obesity not only is associated with hypertension but also is a major contributor to the risk of other chronic diseases.

In finding a potential compound for the inhibition of hypertension, researchers examined the effects of polyphenol quercetin in pre-hypertension and stage I hypertension in overweight and obese individuals.

The double-blinded, placebo-controlled cross-over trial included 70 Subjects randomized to receive 162 mg/d quercetin from onion skin extract powder or placebo for 6 weeks separated by a 6-week washout period.

According to tested assays and compared to the baseline,
* The group treated with quercetin did not significantly show an effect on 24-hour ambulatory blood pressure (ABP) parameters and office BP in the hypertensives subgroup.

* However, quercetin decreased 24 h systolic BP by -3·6 mmHg (P=0·022) compared to placebo.

* Furthermore, quercetin treatment significantly decreased day-time and night-time systolic BP in hypertensives without alternating vasoactive biomarkers such as endothelial function, parameters of oxidation, inflammation, lipid, and glucose metabolism in the total and hypertensive subgroup.

Based on the findings, researchers said, " supplementation with 162 mg/d quercetin from onion skin extract lowers ABP in patients with hypertension, suggesting a cardioprotective effect of quercetin".

In other words, supplementation with 162 mg/d quercetin from onion skin extract protects the heart against CVD by normalizing the blood pressure in hypertensive patients through unknown mechanisms.

Taken together, onion may be considered a remedy for the prevention and treatment of liver alcoholic cytotoxicity pending the confirmation of the larger sample size and multicenter human study.
