Herbal Butterbur may have a profound and strong effect on protecting the neuron integrity and validity against the onset of neuro diseases, some scientists suggested.
Neuroprotective effects are the activity that protects neurons of the brain and spinal cord.
A neurologic deficit is a condition of the brain, or spinal cord, leading to weaker muscles, nerves, or part of the body.
The causes of neurological deficit include genetic disorders, congenital abnormalities or disorders, infections, lifestyle or environmental health problems associated with malnutrition, and brain injury, spinal cord injury, or nerve injury.
People with a neurologic deficit may experience certain medical conditions and symptoms including partial or complete paralysis, loss of sensation, seizures, difficulty reading, writing and walking, and decreased alertness.
However, some researchers suggested the most serious symptoms are poor cognitive ability, depending on the parts of the brain or spinal cord which have been affected.
According to the statistic provided by the US government, more than 16 million people in the United States are living with cognitive impairment and disease with a tendency to increase at an alarming rate, particularly in the age group of 50 and over.
People with cognitive impairment, according to the report are more than three times as many hospital stays as individuals who are hospitalized for some other condition, causing a substantial burden to the US health care system.
Out of all neurodegenerative diseases, Alzheimer’s disease and related dementias alone are estimated to be the third most expensive disease to treat in the United States.
The Alzheimer’s Disease Facts and Figures 20106, wrote, “ The average Medicaid payment for a person aged 65 or older with Alzheimer’s or other dementias is nine times higher than that for other beneficiaries in the same age group.”
Butterbur, found in wet, marshy, damp land, including wetlands, riversides, forests, etc., is a creeping underground plant, genus Petasites of 15 -20 species, belonging to the family Asteraceae, native to the Northern Hemisphere.
The herbal medicine has been used in herbal medicine used as a pain reliever in the digestive system, including stomach, bile ducts, and duodenum, etc.
The chemical constituents of Butterbur include Petioles, petasitene and pethybrene, quercetin 3-O-beta-D-glucoside, quercetin 3-O-beta-D-6''-O-acetylglucoside, and rutin, caffeic acid, alkaloids, S-petasin and iso-S-petasin, etc.
The Second Military Medical University on finding a natural ingredient or whole food for the treatment of neurologic deficit examines the effect of the total bakkenolides, a bioactive compound isolated from the herbal Butterbur, in vitro cerebral ischemia model and oxygen-glucose deprivation of cultured nerve cells.
Oral administration of total bakkenolides showed a significant immediate effect after reperfusion at doses of 5, 10, and 20 mg/kg markedly reduced brain infarct volume and neurological deficits.
The chemical compound also was found to attenuate cell death and apoptosis in primary cultured neurons subject to 1-h hypoxia followed by 24-h reoxygenation.
The compound also protects the neurons by inhibiting the protein involved production of inflammatory cytokines through blocking the activation of the classical activation pathway due to the injury.
Further differentiation of the experiment, researchers discovered that total bakkenolides inhibit the activation of pathways associated with neuron apoptosis.
Dr. the lead scientists after taking into account of co and confounders said, "Our results provide a strong pharmacological basis for further understanding the potential therapeutic role of total bakkenolides in cerebral ischemic disease and shed new light on its neuroprotective mechanism".
Moreover, in the examine the neurologic deficit effects of 4 novel bakkenolides - bakkenolide-Ia ( 1), bakkenolide-IIa ( 2), bakkenolide-IIIa ( 3) and bakkenolide-IVa ( 4) isolated from the extract of the rhizome of Petasites tricholobus, researchers showed the compounds 1 - 4 exert a significantly neuroprotective activity in primary cultured neurons exposed to oxygen-glucose deprivation and oxidative insults by enhancing the antioxidant activity.
According to the VITRO assay, all these compounds exhibited significant neuroprotective and antioxidant activities.
The findings strongly suggested total bakkenolides and their derivatives demonstrate neurologic deficits protective effects through blocking the mechanisms involved in neuron cell apoptosis through their antioxidant property.
Taken together, herbal Butterbur processed bioactive compounds such as bakkenolides may be considered a functional herb for the prevention and combined with primary medicine for the treatment of neurologic deficits.

#Herbal #Comfrey Inhibits Symptoms of Painful Osteoarthritis of the Knee, Researchers Say
Kyle J. Norton
Osteoarthritis (OA) one of the most common forms of arthritis that affects over 25 25 million people in the United States, is a chronic inflammatory disease of the joints due to wear and tear over time.
The characteristics of osteoarthritis are aching pain, stiffness, or difficulty in moving the joint or joints. The pain usually gets worse in a change of weather, at night, and in advanced diseases, the pain can occur even at rest.
Conventionally, treatment of osteoarthritis (OA) focuses on pain relief and improved physical function through pharmacological, nonpharmacological, and surgical treatments.
The most common risk factors associated with the onset of OA include genetic predisposition, being overweight or obese, previous joint injury and repetitive movements, or injuries to joints (such as a fracture, surgery, or ligament tears).
the most common diseases associated with OA are
* Hypertension
According to the University Health Network Research Institute, the economic burden incurred by RA significantly exceeds that related to OA and HBP(76)(77).
* Cardiovascular disease
According to Erasmus University Medical Centre, there are associations of atherosclerosis with osteoarthritis of the knee and hand joints in women(72)(73)(78).
* Peripheral vascular disease
According to Himchan Hospital, patients with osteoarthritis are associated with a risk of asymptomatic peripheral vascular disease(80). The average vessel wall thickness of the popliteal artery was 1.09 mm in patients with generalized OA, and 0.96 mm in the matched normal reference population(79).
* Congestive heart failure
Hypertension, diabetes, and older age have been shown to increase the risk of Congestive heart failure modestly(81). Etoricoxib, an anti-inflammatory painkillers showed statistically significant inhibition at 90 mg in patients with osteoarthritis and rheumatoid arthritis(82).
* Renal function impairment
According to the Southampton University Hospitals NHS Trust, pain pharmacotherapies such as osteoarthritis (OA) or mixed OA and rheumatoid arthritis may increase the risk of adverse events in patients with concurrent cardiovascular (CV) or renal disease(83). Median excretion of pyridinoline and deoxypyridinoline in the OA patients as a whole was raised above values found in a healthy control population(84).
* Diabetes
Several epidemiological and experimental data support the hypothesis that diabetes could be an independent risk factor for osteoarthritis (OA)(76)(85).
Comfrey is a perennial plant in the genus of Symphytum, belonging to the family Boraginaceae, native to Europe. It has been used in herbal and traditional medicine to ease excessive bleeding or on open wounds, relieve pain and swelling, treat circulation issues, reduce cholesterol, etc.
In finding a potential compound for the treatment of osteoarthritis, researchers investigated the effect of Kytta-Salbe f (3 x 2 g), a topical including comfrey over a 3-week period with patients suffering from painful osteoarthritis of the knee.
The randomized, double-blind, bi-center, placebo-controlled clinical trial including 220 patients was conducted by Merck Selbstmedikation GmbH.
In the course of the trial, the visual analog scale (VAS) total score (primary target value) in the verum group dropped by 51.6 mm (54.7%) and in the placebo group by 10.1 mm (10.7%).
The average difference between the groups of 41.5 mm (95% confidence interval=34.8 to 48.2 mm) or 44.0% is significant (p<0.001).
The WOMAC (Western Ontario and McMaster Universities) total score (secondary target value) also improved similarly to the VAS total score.
At the end of the trial, a reduction of 60.4 mm (58.0%) was recorded for the verum group and a reduction of 14.7 mm (14.1%) for the placebo group.
The average group difference of 45.7 mm (95% confidence interval=37.1 to 54.3 mm) or 43.9% is significant (p<0.001).
The difference between the treatment groups increased systematically and significantly, in parallel with the duration of the treatment.
Based on the results, researchers said, "the superiority of the treatment with Kytta-Salbe f over that with the placebo is proven, even through the multi-factorial multivariate analysis for repetitive measurements. In respect of the explorative secondary target values SF-36 (quality of life), angle measurement (mobility of the knee), CGI (clinical global impression), and global assessment of efficacy by the physician and the patient, a significant superiority (p<0.001 each) of the verum group over the placebo group was also proven".
Taken together, Comfrey combined with other herbal medicines may be considered for the treatment of a remedy for painful osteoarthritis, pending the confirmation of the larger sample size and multicenter human study.

3,3'-#Diindolylmethane Inhibits Enzymes, Proteins and Expressions Associated with the Proliferation of #ProstateCancer, According to Researchers
Kyle J. Norton
Scientists may have found a plan based bioactive compound for the treatment of prostate cancer with no side effects, according to studies.
Prostate cancer is a medical condition caused by cell growth disorderly in the tissue of the prostate glands. Prostate cancer is one of the most slow-growing cancers that take years to develop.
The disease often is discovered during the rectum examine annually. Most cases of prostate cancer start in the cells on the surface of the inner lining of prostate tissue.
In the early stage, patients may not experience any symptoms. However, at the advanced stage, prostate cancer can induce symptoms of blood in the urine and semen, disruption of urinary passing, frequent and urged urinary.
In severe cases, prostate cancer can also cause erectile dysfunction.
According to statistics, 1 man in 9 will be diagnosed with prostate cancer during his lifetime.
Most cases ( about 60%) of prostate cancer are found in older men, particularly in the African-American. The 5-year survival rate of local or regional prostate cancer is nearly 100.
Out of many risk factors, the unhealthy diet may be one of the major risk factors associated with the onset of the disease.
Some researchers in the study of the preventive diet against prostate cancer wrote, " Although the available data are not entirely consistent, possible preventative behavioral factors include increased physical activity, intakes of tomatoes, cruciferous vegetables, and soy. Factors that may enhance prostate cancer risk include frequent consumption of dairy products and, possibly, meat".
" By comparison, alcohol probably exerts no important influence on prostate cancer development. Similarly, dietary supplements are unlikely to protect against the onset of prostate cancer in healthy men".
"Several factors, such as smoking and obesity, show a weak association with prostate cancer incidence but a positive relation with prostate cancer mortality. Other factors, such as fish intake, also appear to be unassociated with incident prostate cancer but show an inverse relation with fatal prostate cancer".
3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.
Researchers on finding a natural compound for the treatment of prostate cancer examined the effect of 3,3'-Diindolylmethane (DIM) as tumor inhibitors in multiple malignant tumors on microRNAs (miRNAs).
Tetrachlorodibenzo-p-dioxin (TCDD) and 3,3'-Diindolylmethane (DIM) application showed a significant effect in the inhibition of proliferation and invasion of prostate cancer cells.
Furthermore, Tetrachlorodibenzo-p-dioxin (TCDD) and 3,3'-Diindolylmethane (DIM) treatment induced cancer death by inhibiting the expression level of CYP1A1, an enzyme associated with the activation of the compounds with carcinogenic properties.
In other words, Tetrachlorodibenzo-p-dioxin (TCDD) and 3,3'-Diindolylmethane (DIM) treatment activated the expression of MAP3K12 in the promotion of cancer cells by targeting the mRNA of miR-150-5p in prostate cancer cells.
Moreover, in order to reveal more information about DIM anti-prostate cancer activity, researchers examine the modulation of the immune system by cancer-protective food bioactives, indole-3-carbinol (I3C) and 3,3'-diindolylmethane (DIM).
Injection of indole-3-carbinol (I3C) and 3,3'-diindolylmethane (DIM), exerted prostate cell protective effect by the regulation of inflammatory responses in monocytes and macrophages of the immune system.
In other words, I3C and DIM inhibited the lipopolysaccharide (LPS), the major component of the outer membrane of Gram-negative bacteria in the induction of IL-1β mRNA associated with the immune response to infection and protein in the monocyte form of the white blood cells.
I3C and DIM also exerted anti-inflammatory activity by inhibiting the expression of gene CD84 mRNA in signaling T-cell activation by regulating the proteins associated with the expression of the gene.
Dr. Wang TTY and colleagues after taking into account co and confounders, wrote, "cross-talk between AHR(protein associated with regulating gene expression) and inflammation-mediated pathways, but not CD84-mediated pathways, in monocytes but not macrophages may contribute to the modulation of tumor environments by I3C and DIM in prostate cancer'.
The results strongly suggested that DIM modulated the immune function against the process of prostate cancer development and progression.
Taken together, DIM may be considered a supplement for the prevention and combined with primary medicine for the treatment of prostate cancer, pending to the confirmation of larger sample size and multicenter human study.
Intake of DIM in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

The central system is the complex of nerve tissues including the brain and spinal cord with functions to respond to the integrating sensory information.
Neuroinflammation is a process of the immune system's response to harmful pathogens that induce oxidative stress in the brain.
Neuroinflammation is also the inflammation of nervous tissue associated with the immune response in the first phase of infection including infection, traumatic brain injury, toxic metabolites, or autoimmunity.
Neurological disorders are a class of diseases associated with the brain, and the spinal cord including the nerves that connect them.
The most common disease of neurological disorders is dementia, including ALS. PD, Alzheimer's disease, birth defects of the brain and spinal cord, brain injury and tumor.
Back pain associated with related nerve damage is one of the common complaints in the US that send many patients to seek help from their doctor. Believe it or not, 80% of the US population will experience back pain at some time in their lives.
There is no single cause of neuro-dysfunction. Lifestyle, infections, genetics, and nutrition-related disease, environment, and physical injuries have been found to some major prevalent factors that induce the onset of neurological abnormalities.
Furthermore, neurological disorders have been found to facilitate psychological symptoms such as anxiety and depression and physical symptoms, depending totally on the neurons affected, including partial or complete paralysis muscle weakness seen in post-stroke patients, difficulty reading and writing, and poor cognitive abilities such as patients with dementia.
According to the American Addiction Center, if you experience some of the below symptoms persistently, inclduing headaches, blurry vision, persistent fatigue, changes in behavior, numbness in the legs or arms, changes in coordination or balance weakness, slurred speech and tremors, you should seek help from the professionals immediately, as these may be the first signs of neurological disorders.
Neurodegenerative diseases are the most common forms of low-grade chronic neuroinflammation, including Alzheimer's disease, vascular dementia, and dementia with Lewy bodies.
Dr. WEI-WEI CHEN, the lead scientist at the Xuzhou Central Hospital in the investigation of inflammatory cytokines and chronic neuroinflammation wrote, "Accordingly, pro-inflammatory cytokines are important in the pathophysiology of depression and dementia"
And, "These data suggest that the role of neuroinflammation in neurodegeneration must be fully elucidated, since pro-inflammatory agents, which are the causative effects of neuroinflammation, occur widely, particularly in the elderly in whom inflammatory mechanisms are linked to the pathogenesis of functional and mental impairments.".
Ashwagandha also known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, the Middle East, and the Mediterranean.
The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine for over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered an anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.
In finding a potent compound for enhancing brain performance, researchers examined the ashwagandha effects on the neurons.
Epidemiologically, environmental stressors have been found to cause global concerns in modern society, by rapidly increasing the incidence of a variety of pathological conditions including brain tumors, and neurodegenerative & neuropsychiatric disorders.
Ashwagandha used in Indian traditional home medicine has been shown to process the anti-stress and remedial potential for the treatment of aging and neurodegenerative pathologies, including environmental toxins.
Furthermore, Ashwagandha extracts and their active principles also exerted the underlying molecular mechanism(s) that inhibits the brain pathologies through the herb nootropic potency.
Moreover, in the examing the Ashwagandha leaf extracts effects, researchers found that the Ashwagandha inhibited neuroblastoma and glioma cell initiation, and reversal of Alzheimer's and Parkinson's pathologies, protection against environmental neurotoxins and enhancement of memory.
Based on the findings, researchers said, "Ashwagandha supplementation is associated with significant increases in muscle mass and strength and suggests that ashwagandha supplementation may be useful in conjunction with a resistance training program".
Taken altogether, Ashwagandha may be considered a functional herb that increases body mass and strength in resistance training, pending the result of a larger sample size and multicenter study

#Herbal #Turmeric Inhibits Proteins, TNF-Alpha Levels and Mean Clinical and Histopathological Grade Associated with the Onset of #Uveitis, According to Studies
Kyle J. Norton
Uveitis is a type of eye inflammation, affecting the middle layer of tissue in the eye wall (uvea).
The condition has been found to onset suddenly and get worse quickly. The most common symptoms of uveitis are eye redness, pain, and blurred vision that can affect one or both eyes.
In some cases, patients with the condition also experience symptoms of light sensitivity, floaters, and decreased vision.
If you have some of the aforementioned symptoms, please check with your doctor to rule out the possibility.
The exact cause of uveitis is unknown. However, some researchers suggested that people with a compromised immune system associated with inflammatory disease and infected diseases are at an increased risk of uveitis.
Other risk factors, epidemiologically such as eye injury, and diseases that affect the eye such as diabetes are also found in patients with uveitis.
Most cases of uveitis are treated by prescription eye drops in combination with anti-inflammatory medications. If all fail, surgery may be necessary.
Untreated uveitis has been found to induce complications of glaucoma, cataracts, optic nerve damage and retinal detachment.
Out of the prevalent factors of uveitis, some researchers suggested that aging associated with the weakened immune system may be the major factor that causes uveitis.
Dr, Marwan R. Abdulaal, the lead scientist said, "Uveitis is a vision-threatening inflammation of the eye that carries considerable morbidity. It is responsible for 10% of legal blindness in the United States and up to 25% in the developing world".
And. "The aging body has a changing response of the immune system, which might reflect a different pattern of uveitis in the elderly population".
Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.
The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.
On finding a natural compound for the treatment of uveitis, researchers examined the effect on turmeric administered 4 h after induction of E. coli lipopolysaccharide-induced uveitis in rats.
Compared to the control group, turmeric showed significantly suppressed inflammation with a significantly lower mean clinical grade, histopathological grade, and aqueous humor (AH) protein level, second to the conventional medicine prednisolone.
The turmeric administered 4 h did not affect proinflammatory cytokines levels of TNF-alpha levels.
However, when the aqueous extract was administered starting 3 days before induction of uveitis, the treatment group not only showed a similar effect on the mean clinical and histopathological grade, as well as AH protein but also reduced TNF-alpha levels, comparable to the treatment group.
Based on the findings, researchers said, "topically applied a standardized aqueous extract of C. longa suppresses endotoxin-induced uveitis in rats by reducing TNF-alpha activity".
To reveal more information about turmeric's anti-uveitis properties, researchers examined the anterior uveitis induced in rabbits by intravitreal injection of lipopolysaccharide from Escherichia coli after pretreatment with C. longa and B. aristata aqueous extracts.
According to the clinical and histopathologic grading, anterior segment inflammation in the control group was significantly higher compared to both the extract-treated groups.
More precisely, the inflammatory cell count in the control group was 30.75 cells/mL compared to 2.39 and 11.56 cells/mL in the C. longa- and B. aristata-treated groups, respectively.
Similarly, C. longa- and B. aristata-treated groups also exerted strong effects on the protein content and TNF-alpha level as of that in the aforementioned study.
Taken together, turmeric may be considered a supplement for the prevention and treatment of uveitis, pending the confirmation of the larger sample size and multicenter human study.
Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthyfood #Cranberry Exhibits Anti-Cell Proliferation, #Angiogenesis and Metastasis in #Cancer, Scientists Reveal
Kyle J. Norton
Cancer is a class of diseases characterized by abnormal cell growth that begins in the cells on the surface of the inner lining of the tissue of an organ caused by the alternation of cell DNA with unknown nature.
Under normal circumstances, cell growth is a natural process that happens daily to replace millions of dead cells. In some cases, acceleration of cell growth is necessary to prevent further damage to the body organ, such as cut, injure, etc., but after fulfilling their purposes, the growth of cells are back to a normal rate again regulated growth inhibitors.
In other words, healthy cell division is limited to several cycles. Comapred to normal cells, cells with an alternation of DNA or cancer cells can grow indefinitely.
Unfortunately, skin cells sometimes overgrow while healing, leading to scar to the skin.
At the early, most people with cancer are asymptomatic, however, later, due to the size and the tumor, patients may experience general cancer symptoms such as unintended weight loss, fatigue, gastrointestinal discomfort, and loss of appetite and localized symptoms such as severe pain and bleeding.
Cancer can be classified by the types of cells that have become malignant.
* Carcinoma
Carcinoma is a malignant tumor found in anywhere of epithelial tissues lining the cavities, surfaces of structures and glands.
* Sarcoma
Sarcoma is a type of cancer that arises from malignantly connective tissue cells that originated from embryonic germ cells or bone, cartilage, and fat tissues and cells.
* Lymphoma and leukemia
Hematopoietic cells are multipotent stem cells that give rise to all types of blood cells, including red blood cells, white blood cells, and platelets become cancerous.
* Germ cell tumor
Cancer is derived from pluripotent cells. In adults, these are most often found in the testicle and ovary but are more common in babies and young children.
* Blastoma
Blastoma is cancer derived from immature "precursor" or embryonic tissue, mostly in children.
The causes of cancer are not identified. However, most prevalent factors associated with the cancer onset include age, family history, long-term exposure to certain cancer-induced chemicals, use of tobacco and excessive alcohol drinking,..... the list goes on and on, depending on the types of cancer.
Some researchers suggested that the most common risk factor that causes cancer in the Western world may be associated with widespread obesity.
Dr. Karen Basen-Engquist, in the evaluation of the obesity risk of cancer, said, "In the United States, approximately 85,000 new cancer cases per year are related to obesity. Recent research has found that as the body mass index increases by 5 kg/m2, cancer mortality increases by 10%".And, "High priorities for future research involve additional work on the underlying mechanisms, and trials to examine the effect of lifestyle behavior change and weight loss interventions on cancer and intermediate biomarkers".
The findings strongly indicated the risk of cancer is increased substantially in obese.
Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.
In finding a potential compound for the treatment of invasive tumors, researchers examined the potential effects of cranberry on cancer prevention and treatment.
According to the review of the medical literature by searching the database of PubMed, cranberry and/or its components may act as chemopreventive agents, that diminish the parameters associated with the onset of cancer, including the inhibition of cells oxidation and inflammatory-related processes, in vitro.
Cell proliferation angiogenesis and metastasis involved in cancer cell invasion were also attenuated by the administration of cranberry's apoptotic activity.
Dr. Mantzorou M, the lead scientist, after taking other factors into account wrote, " Cranberry could be considered as a conglomeration of potential effective anticancer drug-like compounds".
Additionally, in chemical constituents analysis, cranberry showed inhibitory action against cancers of the esophagus, stomach, colon, bladder, prostate, glioblastoma, and lymphoma.
The efficacy of anti-cancer activity was attributed to the chemical compounds that work synergistically to result in cellular death induction via apoptosis, necrosis, and autophagy, reduction of cellular proliferation, the elevation of reactive oxygen species, and modification of cytokine and signal transduction pathways, similar to those of aforementioned study.
Taken together, cranberry may be considered a remedy for the prevention and treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.

#Phytochemical #Allicin Inhibits Liver Oxidativestress by Preventing Lipid Peroxidation of Liver Homogenate, Researchers Say
Kyle J. Norton
Phytochemicals may be the next potential source of new medications for the treatment of diseases with little or no side effects.
Scientists may have found a dietary and bioactive ingredient for the prevention and treatment of diseases associated with oxidative stress.
Allicin is a phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in onion and garlic.
Oxidative stress is a condition characterized by the overexpression of free radicals in the host body.
Most cases of oxidative stress are caused by the inability of the host liver to stimulate the production of antioxidant enzymes which play a critical role in inhibiting the free radical expression, causing oxidative stress.
In a healthy individual, the ratio of free radicals and antioxidant enzymes is always balanced.
However, an individual whose antioxidant enzymes were suppressed due to damage to the antioxidant enzymes' productive system or other sources may have a greater risk of the onset of oxidative stress.
Dietary antioxidant intake such as fruits and vegetables may be helpful to keep the free radicals in check.
Oxidative stress has been found to cause damage to cell lipids, proteins, and DNA, leading to many chronic diseases, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.
The oxidative stress that causes chronic ailment is a result of a chain reaction started by free radical donating or stealing an electron from another atom, leading to a domino effect that can not be stopped until all outer ring electron of free radicals is paired or inhibited by antioxidants.
A free radical is any atom or molecule that has a single unpaired electron in an outer shell and is highly reactive to react with another cell.
Most free radicals and other ROS are derived internally either from normal essential metabolic processes in the human body or from external sources such as exposure to X-rays, ozone, cigarette smoking, air pollutants, and industrial chemicals.
Antioxidants are biomolecules that inhibit oxidation.
On finding a natural ingredient and whole food for the prevention and treatment of diseases associated with oxidative stress, researchers investigated the effect of allicin (the active ingredient of garlic) contained in the commercial preparation Garlicin against hydroxyl radicals (.OH) using high-pressure liquid chromatographic (HPLC) method.
Allicin equivalent in Garlicin (1.8, 3.6, 7.2, 14.4, 21.6, 28.8, and 36 micrograms) produced concentration-dependent decreases in the formation of 2,3-DHBA and 2,5-DHBA, the OH adduct products.
The inhibition of the formation of 2,3-DHBA and 2,5-DHBA with 1.8 micrograms/ml was 32.36% and 43.2% respectively while with 36.0 micrograms/ml the inhibition was approximately 94.0% and 90.0% respectively.
These results suggested that allicin is highly active against the OH adduct products through its free radical scavenging activity.
Furthermore, Allicin also exerted a significant antioxidant activity by preventing the lipid peroxidation of liver homogenate in a concentration-dependent manner.
Additionally, by examining the antioxidant effects of four main chemical classes, alliin, allyl cysteine, allyl disulfide, and allicin, prepared by chemical synthesis or purification, researchers discovered
* Alliin scavenged superoxide, while allyl cysteine and allyl disulfide did not react with superoxide.
* Allicin suppressed the formation of superoxide by the xanthine/xanthine oxidase system, probably via a thiol exchange mechanism.
* Allicin did not exhibit hydroxyl radical scavenging activity in this study, probably due to some unknown mechanisms.
The finding suggested that allyl disulfide, alliin, allicin, and allyl cysteine exhibit different patterns of antioxidant activities as protective compounds against free radical damage.
Taken altogether, allicin-processed high levels of free radicals scavenging activity may be used for the prevention of the overexpression of ROS in the initiation of oxidative stress.
