#Daidzein and the Garlic Enzyme Alliinase Induce #OvarianCancer Cell Apoptosis through Its Cytotoxic Effects, According to Studies

Ovarian cancer is a chronic medical condition caused by cell growth irregularly in the tissue of the ovary.
Most cases of ovarian cancer begin in the cells on the surface of the inner lining tissue of the ovary before spreading to the deeper layers to form a tumor.

In the very early stage, due to the very small size of the tumor, most patients are asymptomatic. However, as the disease progresses, patients may experience symptoms shared commonly with other types of cancer accompanied by specific symptoms of abdominal bloating or swelling, discomfort in the pelvic area, and changes in bowel habits, such as constipation.

According to the statistics provided by the American Cancer Society, ovarian cancer ranks fifth in cancer deaths among women, accounting for more deaths than any other cancer of the female reproductive system.

A woman's risk of ovarian cancer is 1 in 78. and the risk of the lifetime chance of dying from ovarian cancer is about 1 in 108.

The five-year relative survival rate for all types and stages of ovarian cancer is 47 percent, Cancer diagnosed at the early stage have a 5-year survival rate of over 72%.


The exact causes of ovarian cancer are not known. However, according to the epidemiological studies genetics preposition such as BRCA1 and BRCA2, family and history of cancer or endometriosis, aging, reproductive history, and infertility and obesity are some of the prevalent factors involved in the onset of ovarian cancer.

The Canadian Cancer Society in the evaluation of the risk of ovarian cancer in women who are never pregnant or giving birth wrote, "Women who have never been pregnant have a higher risk of developing ovarian cancer than women who have been pregnant".


And, "Researchers think that the lower risk may be because the hormones that are present during pregnancy have a protective effect. It is possible that the higher risk in women who have never been pregnant is linked to the factors that may make it difficult for her to become pregnant".


In other words, women who have at least one pregnancy are at a lower risk of ovarian cancer compared to women who do not.

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide, a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

With an aim to find a natural and potential compound for the treatment of ovarian cancer without inducing adverse effects, researchers examined the efficacy of preparation including chemical conjugate between daidzein and the garlic enzyme alliinase.

According to the tested assays, the conjugate is specifically bound to ovarian cancer cells and upon addition of the prodrug alliin by inducing the cytotoxic allicin molecules in killing the cancer cells.


In vivo, the preparation showed a targeting and antitumor effect confirmed by NIR and bioluminescence imaging in the luciferase expression of ovarian cancer cells.

Treatment of ovarian tumor-bearing mice with daidzein-alliinase and alliin effectively attenuated tumor progression during the first 12 days.

Compared to the placebo, only small individual foci of luminescence at the site of tumor cells inoculation were found in the treatment group, according to autopsy.

Interestingly, the preparation did not reveal any additional foci of carcinoma or signs of toxicity, according to the histological examination of organs and tissues.

In other words, the chemical conjugate between daidzein and the garlic enzyme alliinase induced ovarian cancer cell apoptosis through its cytotoxic effects without causing harm to nearby healthy cells.

Based on the findings researchers wrote in the final report, "the targeted alliinase conjugates in the presence of alliin, generated therapeutically effective levels of allicin which were capable of suppressing tumor progression of intraperitoneal ovarian cancer in an animal model".

Taken together, alliin used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of ovarian cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Inflammation is a natural and systematic response of the immune system in the protection of the body against infection caused by the invasion of stimuli and pathogens.

After sensing the bodily injury or damage, the white blood cells of the immune first line of defense stimulate the production of a blood clot to seal the would quickly and proinflammatory cytokine to the site of infection with an aim to kill off all microorganisms that may cause harm to our body.

In this acute phase of the battle, most of the times are won by the immune system. However, overexpression of the pro-inflammatory cytokines can lead to the death of nearby healthy cells and improper skin healing with the formation of scars.

Moreover, in order to understand the gastroprotective effects of manuka honey against ethanol-induced gastric ulcers in rats, researchers examined the joint studies led by King Abdulaziz University, including 4 our groups of rats randomly assigned to control, ethanol (ulcer), omeprazole, and manuka honey.


During the study, the Manuka honey-treated group displayed a significantly decreased ulcer index, completely protected the mucosa from lesions, and preserved gastric mucosal glycoprotein.


Manuka honey treated group also showed increased levels of antioxidants enzymes which were suppressed by the injection of ethanol to an induced ulcer.

Manuka honey also decreased gastric mucosal MDA, the oxidative stress marker, and plasma of proinflammatory cytokines.

Dr. Almasaudi SB, the lead scientist wrote in the final report, "Manuka honey likely exerted its antiulcer, effect by keeping enzymatic (GPx and SOD) and nonenzymatic (GSH and NO) antioxidants as well as inflammatory cytokines (TNF-α, IL-1β, and IL-6) in a reduced form, inhibited lipid peroxidation (MDA), and preserved mucous glycoproteins levels".

Taken together, Manuka honey may be considered a functional food for the prevention of inflammation and combined with the primary medicine for the treatment of chronic inflammatory diseases with no side effects.

Scientists may have found a bioactive ingredient for the treatment of liver toxicity with no side effects, according to studies.
Acute liver toxicity is a condition characterized by the harmful effects of a substance or organism induced by a single or short-term exposure.

Liver toxicity, on the other hand, is caused by long-term exposure to a substance or substances, including alcohol inducing the destruction and interruption of liver tissue regeneration, leading to fibrosis and cirrhosis.


The liver is the largest internal organ with self-regenerative properties. It only plays an essential role in the production of bile to aid digestive functioning, but also filters out blood from the digestive system before passing them back to the heart and other parts of the body.

The most common chronic liver diseases in the Western world are non-alcoholic fatty liver disease (NAFLD) and hepatitis C infection.

According to statistics, NAFLD affects 75 to 100 million Americans in 2017. Approximately 80% of obese and 20% of normal-weight people have NAFLD.

Believe it or not, nearly 2.4 million Americans, or 1 percent of the adult population were living with hepatitis C.

The most common cause of acute liver toxicity is Acetaminophen overdose in the United States, leading to acute liver failure.

Dr. Flores YN, the lead scientist in the examination of risk factors for liver disease among Mexican adults in the US and Mexico wrote, "Excessive alcohol consumption is a risk factor for liver disease, but only 60% correctly identified hepatitis C, being overweight or obese, or having diabetes as risk factors".

And, " Identifying the specific risk factors, levels of knowledge, and prevention activities that affect specific racial/ethnic populations is important in order to effectively target efforts to prevent liver disease".

Betanin is phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

With an aim to find a natural compound for the protection of the liver against toxicity, researchers examined the betanin (a natural pigment) mitigated effect in Organophosphates (OP)-induced hepatotoxicity in primary rat hepatocytes.

Administration of betanin (25μM) significantly increased liver cell viability, inhibited the expression of ROS formation and LPO and restored cellular antioxidant levels.

When it was used alone, betanin protected the liver against OPs-induced hepatotoxicity through inhibition of oxidative stress and mitochondrial protection against inflammation.

In order to reveal more information about betanin (from natural pigments) anti-oxidant properties against paraquat-induced liver injury in an animal model.


The study included rats randomly divided into four groups: a control group, a paraquat group, and two groups that received betanin at doses of 25 and 100mg/kg/day three days before and two days after they were administered paraquat.

Paraquat-induced liver toxicity in Sprague-Dawley rats without treatment of betanin showed significant liver toxicity observed by elevated serum aspartate aminotransferase and alanine aminotransferase levels and an increase of enzyme associated with phase I drug metabolism reactions.

In other words, the injection of paraquat led to an unbalanced ratio of free radicals and deleted antioxidant enzymes produced by the host evidenced by levels of oxidative stress.

Injection of betanin not only inhibited the historical change and the aforementioned conditions, but the phytochemical also protected the liver cells against mitochondrial damage and apoptosis-inducing factor protein levels.

Dr. Han J, the lead scientist based on the finding wrote in the final report, "Betanin had a protective effect against paraquat-induced liver damage in rats. The mechanism of the protection appears to be the inhibition of CYP 3A2 expression and protection of mitochondria".

Taken together, betanin may be considered a supplement for the treatment of acute liver toxicity, pending the confirmation of the larger sample size and multicenter human study.

#Bromelain Processes Cytotoxic and Antiproliferative Activity Against Peritoneal #Mesothelioma, Researchers Say

Peritoneal mesothelioma is a medical condition characterized by irregular cell growth in the tissues of the abdomen (the peritoneum).

Most cases of peritoneal mesothelioma begin in the cell on the surface of the inner lining of the peritoneum, before penetrating into the deeper layers.


At the early stage, due to the small size of the tumor, patients with peritoneal mesothelioma are asymptomatic.

However, at the later stage, patients may experience symptoms of abdominal pain or swelling due to the larger size tumor pressed on the nearby nerve tissues and blood vessels, changes in bowel habits, feeling of fullness and night sweat accompanied by the general symptoms shared by other types of cancer, such as unexplained weight loss, gastrointestinal discomforts, and loss of appetite.


According to the statistics provided by the American Cancer Society, compared to other types of cancer mesothelioma is rare, with approximately 2,000 to 3,000 new cases of mesothelioma in the US each year. The asbestos-related mesothelioma only accounts for less than 0.3 percent of all cancer diagnosed in the country.

Peritoneal mesothelioma accounts for less than 20 percent of all mesothelioma cases.

Dr. Rodolfo Fonte, the lead scientist in the examination of the risk of asbestos-induced peritoneal mesothelioma in a construction worker, wrote, "Peritoneal mesothelioma is a rare and aggressive asbestos-related malignancy. We present the case of a 65-year-old man who developed recurrent ascites after having been exposed to asbestos in the building industry for > 40 years".


And, "Occupational history and immunohistochemistry are helpful for the correct diagnosis, which, in turn, is important in relation to prognosis and treatment (adoption of new integrated procedures that seem to promise prolonged survival and increased quality of life), and in relation to medico-legal issues and occupation-related compensation claims following asbestos exposure".

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural ingredient for the treatment of peritoneal mesothelioma, researchers examined the effects of Bromelain (cysteine proteinase) effects on Mucin 1 (MUC1), a glycosylation-dependent protein that confers tumors with invasiveness, metastasis, and chemoresistance.

In the study in which researchers compared the efficacy of the application of various concentrations of bromelain after 4 and 72 h and combinations of bromelain and cytotoxic drugs (cisplatin or 5-FU) at 72 h, MUC1-expressing MPM cell lines found that bromelain increased the cytotoxicity of cisplatin significantly in cell lines.

Compared to the aforementioned inhibition, combining fluorouracil with bromelain did not show a significant increase in cytotoxicity.

In other words, bromelain improves chemotherapy depending totally on the types of drugs used for the treatment of peritoneal mesothelioma.

Furthermore, bromelain used alone also showed a significant effect in inducing cell death by apoptosis and autophagy.

On pancreatic and breast cancer cells, injection of bromelain also adversely affected the viability of MUC1 expressing on both cell lines.


Based on the findings, Bromelain used alone or combined with other chemo drugs might have a strong effect on the barrier function of MUC1.

Collectively, bromelain inhibited the progression and proliferation of peritoneal mesothelioma by blocking the activity of gene MUC1.


Dr. Pillai K, the lead scientist said, "Current evidence indicates that bromelain may have the potential of being developed into an effective anticancer agent for MPM".

Taken together, bromelain may be considered a supplement for the prevention and treatment of peritoneal mesothelioma, pending the confirmation of the larger sample size and multicenter human study.

Scientists may have found a natural whole food that processes a significant activity in reducing the risk and treatment of acute coughing in children with no side effects, some studies suggested.
Coughing can be a natural and reflex reaction to expel phlegm from the airway or a result of viral and bacterial infection of the upper airway.

A cough that lasts less than three weeks is considered an acute cough.


A chronic cough is a medical condition that lasts more than 3 weeks. In children, the condition can be caused by allergies and sinusitis, asthma, whooping cough, cystic fibrosis, aspiration, acid reflux, and a blockage in the airway.

The most common causes of acute cough are cold and flu. However, mild adverse events were found in seven children (9.3%) from the honey group and two (2.7%) from the dextromethorphan group.

#Ginger Inhibits Cervical Cancer by Inducing DNA Damage of the Tumor Cell and G2/M Cell Cycle Arrest, According to Studies

Scientists may have found a natural and pungent spice for the prevention and treatment of cervical cancer, according to the studies.
Cervical cancer is a medical condition characterized by cell growth disorderly and uncontrollably in the cervix tissue.

According to the statistics provided by the American Cancer Society, in 2018, approximately, 13,240 new cases of invasive cervical cancer were diagnosed. The disease causes the death of 4,170 women in the same year.

The 5-year survival rate for women with invasive cervical cancer is 92% if the cancer is detected at an early stage. About 46% of women with cervical cancer are diagnosed at an early stage.
Most cases of cervical cancer are detected during the annual physical exam during the Pap test.

The exact cause of cervical cancer is due to the alternation of the cells on the surface of the inner lining of the cervical tissue. Researchers do not know, Why? some women are more susceptible to the onset of cervical cancer while others do not, even have the same health conditions and family history in the same family.

However, researchers do know that certain risk factors such as human papillomavirus (HPV) infection, the increase in age, herpes infection, smoking are associated with the onset of cervical cancer.

Ethnicity also plays a critical role in cervical cancer, the American Cancer Society wrote, "In the United States, Hispanic women are most likely to get cervical cancer, followed by African-Americans, Asians, and Pacific Islanders, and whites. American Indians and Alaskan natives have the lowest risk of cervical cancer in this country".

Furthermore, some researchers suggested that oral sex which refers to sexual activities involving the stimulation of the genitalia by the use of the mouth, tongue, teeth, or throat also has a strong linkage to cervical cancer. Dr. Rajiv Saini examined the link between oral sex and cervical cancer and wrote, "Current research suggests that oral sex may transmit human papillomavirus (HPV), the virus implicated in the majority of cervical cancers. HPV is ubiquitous, with 120 strains isolated from human beings, about 40 of which are in the mouth and genital tracts".

Researchers on finding a natural compound for the treatment of cervical cancer examined the antitumor activities of 6-Gingerol, a potent nutraceutical isolated from the ginger on human cervical cancer (HeLa) cell lines with or without wortmannin, rapamycin, and cisplatin.

Injection of 6-Gingerol showed dose-dependent cytotoxicity on human cervical cancer (HeLa) cell lines.

Combinations of 6-gingerol with wortmannin and cisplatin showed additive effects, while rapamycin, showed 50% cytotoxicity that was equivalent to IC50 of 6-gingerol alone.

Treatment with 6-gingerol resulted in G2-phase arrest in HeLa cells. 6-Gingerol, wortmannin, and rapamycin treatment showed almost two-fold higher expression of a pro-apoptotic protein in the tested cell line.

The finding strongly suggested that 6-gingerol may be a safe and potent chemotherapeutic/chemopreventive compound acting through cell cycle arrest and induction of apoptosis in cervical tumor cells.

In order to reveal more information about ginger Anti-cervical cancer properties, researchers examined the polyphenolic alkanone, 6-Gingerol (6G), presented in the pungent extracts of ginger (Zingiber officinale Roscoe) anti-tumorigenic and pro-apoptotic activities against cervical cancer.

In vivo. 6G potently inhibited the proliferation of the HPV positive cervical cancer cells.

Additionally, 6G was found to inhibit the chymotrypsin activity associated with the induced degradation of tumor suppressor proteins, reactivate the gene involved in regulating the cell cycle and tumor suppression, and suppress the protein involved in cellular response to cytokines, chemokines, and stimuli.

More interestingly, injection 6G also expressed an inhibitory activity against cervical cancer cell lines through inducing DNA damage of the tumor cell and G2/M cell cycle arrest and potentiate the cytotoxicity of cisplatin

6G treatment induced a significant reduction of tumor volume, tumor weight, inhibition of proteasome that breaks down protein, and tumor-suppressive gene accumulation in HeLa xenograft tumor cells in vivo.

Dr. Rastogi N and colleagues said, "Our data underscore the therapeutic and chemosensitizing effects of 6G in the management and treatment of cervical cancer".

Taken together, ginger with abundantly bioactive compound 6-gingerol may be considered a functional remedy for the prevention and combined with primary medicine for the treatment of cervical cancer, pending the confirmation of large sample size and multicenter human study.
