Serum #Carotenoids Are Associated Inversely with the Risk of #BreastCancer, According to Study

Breast cancer or malignant breast neoplasm is a medical condition caused by cell growth irregularly in the tissue of the breast.
Most cases of breast cancer start in the cell on the surface of the inner lining tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.

In 2010, over 250,000 new cases of breast cancer were diagnosed in women in the U.S. alone and the risk of getting invasive breast cancer during the lifetime of women is 1/8.

Breast cancer is the most common cancer worldwide, accounting for over 24% of all cancers diagnosed every year.

The 5 years survival rate of breast cancer in the localized stage is about 100%.

Inflammatory breast cancer (IBC), is aggressive cancer that occurs in women of any age that grows in sheets instead of lumps and invades nearby skin, leading to the whole breast suddenly goes red and feeling hot, resembling an inflammation

The rare condition was found in only 1-6% of all breast cancer cases in the USA. Inflammatory breast cancer (IBC) should be treated very promptly.

The most common symptoms of IBC are
* Rapid swelling (a cup size in a few days) sometimes

* Skin changes (pink, red, or dark-colored areas)

* Nipple retraction

* Persistent itching, skin hot to touch

* Initially resembles mastitis

* Breast skin ulcers (later stage of IBC)


How dangerous is IBC? Dr. Ana M. Gonzalez-Angulo, the lead scientist wrote, " (According to the results of the study), the median follow-up was 5.8 years (range, 0.3–23.8 years). There were 238 recurrences and 236 deaths (compared to 398 patients with IBC who were treated between 1974 and 2005). The median recurrence-free survival (RFS) duration was 2.3 years and the median overall survival (OS) time was 4.2 years".

Carotenoids is a class of mainly yellow, orange, or red fat-soluble pigments, including lycopene and carotene, found abundantly in ripe tomato, pumpkins, carrots, corn, and daffodils.

In finding a potential compound for the treatment of cancer, researchers examined the protective role of carotenoids against the onset of breast cancer.

The study included a total of 521 women with breast cancer and age-matched controls (5-year interval) selected from three teaching hospitals in Guangzhou, China.

According to the chemical analysis, phytochemicals α-carotene, β-carotene, β-cryptoxanthin, lycopene, and lutein/zeaxanthin are found in serum carotenoids,
* Serum α-carotene, β-carotene, lycopene, and lutein/zeaxanthin are associated inversely with the risk of breast cancer.

* The relative odds ratio for the highest quartile of serum concentration compared with the lowest quartile were 0·44.

* More precisely, the OR for α-carotene was.44, β-carotene was 0·27 and lycopene was .41, and lutein/zeaxanthin was 0·26.

* β-cryptoxanthin, one of the major compounds was not significantly associated with the risk for breast cancer.

Based on the stratified analysis, researchers found that serum α-carotene, β-carotene, lycopene, and lutein/zeaxanthin were also inversely associated with breast cancer risk among premenopausal women and among all subtypes of ER or PR status.

The results strongly suggested that all chemical compounds in the class of carotenoids work synergistically to decrease the risk of cancer among women.

Dr. Yan B, the lead scientist, after taking other factors into account said, "The results suggest a protective role of α-carotene, β-carotene, lycopene, and lutein/zeaxanthin, but not β-cryptoxanthin, in breast cancer risk.".

Taken together, carotenoids may be considered a remedy for the prevention and treatment of breast cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of carotenoids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Vegetables and #Fruits Show Inversely Associated with the Risk of Esophageal (ESCC and EAC) Cancers, According to Studies

Esophagus, an organ in vertebrates, is the tube that leads food from the pharynx to the stomach.

Esophageal cancer is a medical condition caused by cell growth disorderly and uncontrollably in the tissue of the esophagus due to the alternation of the cell DNA.

More precisely, esophageal cancer can be a result of DNA alternation of the cells that line the upper part of the esophagus or glandular cells that line the lower part of the esophagus, connecting to the stomach.

At the advanced stage, most cases of esophageal cancer after penetrating the wall of the esophagus tend to spread to the lymph node around the bottom of the esophagus, stomach, and chest, then to the distant parts of the body.

According to the statistics provided by the American Cancer Society, in 2019, approximately 17,650 new esophageal cancer cases will be diagnosed, including 13,750 in men and 3,900 in women.

Also, in 2019, esophageal cancer will also cause the death of 16,080 Americans, including 13,020 in men and 3,060 in women.

The 5-year survival rate of people diagnosed in the local stage of esophageal cancer is 45%, much less than other cancers, including 100% of breast cancer.

Believe it or not, esophageal cancer is more common among men than among women. The lifetime risk of esophageal cancer in the US is 1 in 132 in men and about 1 in 455 in women.

Patients with esophageal cancer at a very early stage due to the very small size of the tumor are asymptomatic. However, at the advanced stage, esophageal cancer tends to travel a distance away to infect other healthy tissues and organs, leading to secondary metastasis.

The most common areas of metastasis of esophageal cancer are lymph nodes around the bottom of the esophagus, stomach, and chest.

* Total vegetable consumption was nonsignificantly inversely associated with EAC and ESCC risk, but not with GCA and GNCA risk/

* There were significant inverse associations between raw vegetables and EAC risk [RR per 25 g/day: 0.81, 95% confidence interval (CI) 0.68-0.98], and Brassica vegetables and GCA risk (RR per 25 g/day: 0.72, 95% CI 0.54-0.95).

* Total fruit consumption was associated with a nonsignificantly decreased EAC risk. Citrus fruits were inversely associated with EAC and GCA risk (RRs for highest vs. lowest intake: 0.55, 95% CI 0.31-0.98 and 0.38, 95% CI 0.21-0.69, respectively).

* Particularly, current smokers, vegetables, and possibly fruit intake were inversely associated with ESCC and EAC risk.

Based on the evidence, scientists said, "Consumption of (specific groups of) vegetables and fruits may protect against subtypes of esophageal and gastric cancer:.

Taken together, vegetables and fruits may be considered functional foods for the prevention and treatment of esophageal cancer, pending the confirmation of the larger sample size and multicenter human study.

#Green Tea and its Bioactive Polyphenols Decrease Cortical and Trabecular Bone Loss and Increase Bone Density, Studies Find

Green tea may have a therapeutic and positive effect in reducing the onset, progression, and treatment of osteoporosis, some scientists suggested.

Osteoporosis is a chronic condition characterized by progression in the reduction of bone mass and deterioration of bone tissue and is considered a major health problem in both elderly men and women.

The disease also affects all bone in the body, including the bones of the spine, hip, and wrist

Scientists as of today do not know the exact causes of osteoporosis. However, they do know that certain factors are associated with the onset and progression of the disease.

Osteoporosis is a common disease among the elderly, probably due to the aging effect that induces nutrition deficiency such as
* Vitamin D insufficiency and reduced calcium absorption,

* Reduced hormone production, the drop of estrogen levels after menopause, can speed up bone loss.

* Reduced production of bone mass and increased bone reabsorption.

These results suggested that osteoporosis can make the bone more fragile and easier to break. In the elderly, osteoporosis can be dangerous, as fractures can cause prolonged immobility, and require a longer healing process.

Some researchers suggested that fractures in the older population are the most common cause of blood clots or pneumonia, both of which can have fatal consequences.

Long-term use of certain medicines, such as anti-seizure drug Glucocorticoids and medical conditions such as kidney disease, Cushing's syndrome, and an overactive thyroid or parathyroid, can also increase the risk of osteoporosis.


Dr. Stephen P Tuck, the lead author in the study Osteoporosis in men said, "The incidence of osteoporotic fractures in elderly men, just as in aged women, increases exponentially with age; the rise in men, however, is some 5–10 years later than in women. Up to 50% of male osteoporosis have no identifiable etiology; however elderly males have a much higher likelihood of having an identifiable secondary cause than younger men".

Truly, both males and females share the same conditions of the onset of osteoporosis, but the onset of the disease in men is about 10 years later, compared to a woman of the same age.


Interestingly, in the observation of hip fracture in the aging population, Dr. Eric Orwoll, a physician-researcher who studies osteoporosis at Oregon Health and Science University wrote, “A third of all hip fractures occur in men, yet the problem of osteoporosis in men is frequently downplayed or ignored.”

Green tea is a precious drink that processes a number of health benefits known to almost everyone in Asia and the Western world.

In the study to evaluate the effects of ingestion of green tea and green tea bioactive compounds in mitigating bone loss and risk of osteoporotic fractures in both elderly women and men, researchers found that the applications exert significant activities in reducing the onset of osteoporosis, through their antioxidants and initiation of the production of natural antioxidant in the body such as glutathione and enzymes (e.g., catalase and superoxide dismutase) in inhibiting the levels of ROS that facilitate the chain reaction.

Truly, the overexpression of free radicals has been associated with the change in the balance between the activities of osteoblasts and osteoclasts, leading to osteoclasts destroying the calcified tissue and inducing bone remodeling.

These results demonstrated that green tea and its bioactive components exhibit a significant decrease in the risk of fracture by improving bone mineral density and preventing free radicals from damaging the bone-forming osteoblasts while suppressing osteoclastic activities.

Moreover, in the review of the effect of green tea and its bioactive components on bone health, researchers indicated that the ingestion displays an impressive osteoprotective impact on bone mass and microarchitecture in various bone loss models associated with aging, sex hormone deficiency, and chronic inflammation.

The implication of reduced onset and progression of osteoporosis in the aging population due to sex hormone deficiency may be attributed to green tea and its bioactive compound in increasing the levels of osteocalcin, a bone formation marker, decreasing serum of tartrate-resistant acid phosphatase (TRAP), a bone resorption marker and improving the body in the absorption of calcium.

In fact, the serum levels of osteocalcin (OC) and tartrate-resistant acid phosphatase (TRAP) are considered effective measurements of the rate of bone turnover in patients with osteoporosis.

Interestingly, the application of green tea and its bioactive compound also is found to modulate the expression of proinflammatory cytokines in the regulation of osteoblasts and osteoclasts, by inhibiting the production of C-reactive protein (CRP) in the liver in initiating inflammation and reducing levels of interleukin-1 and 6(IL-1-6) and tumor necrosis factor-alpha (TNF-α), the sensitive markers of systemic inflammation.

This differentiation was supported by the study in the evaluation of the efficacy of green tea polyphenols (GTPs) at mitigating bone loss and microstructure deterioration along with related mechanisms in 40 androgen-deficient aged male rats, a model of male osteoporosis, randomly assigned to received A 2 (sham vs. orchidectomy) × 2 (no GTP and 0.5% GTP in drinking water) for 16 weeks.

At the end of the experiment, researchers found that rats treated with GTP demonstrated increased serum osteocalcin concentrations, bone mineral density, and trabecular volume, number, and strength of femur; trabecular volume and thickness and bone formation in both the proximal tibia and periosteal tibial shaft, and decreased eroded surface in the proximal tibia and endocortical tibial shaft, and increased liver glutathione peroxidase activity.

The findings indicated a strong implication of green tea polyphenols in decreasing cortical and trabecular bone loss and increasing both bone density by stimulating the overexpression of bone formation and reducing bone resorption via antioxidant capacity.

Taken together, green tea and its bioactive polyphenols may be considered functional foods in ameliorating early-onset, progression, and combined with standard medicine for the treatment of osteoporosis.

However, intake of green tea supplements should be taken with extreme care due to recent reports of acute liver toxicity.

#Phytochemical #Betanin Inhibits Intestinal Mucosal Inflammation in the Formation of Colitis, Researchers Say

Colitis is a general term associated with the inflammation of the inner lining of the colon.

The disease can be caused by infection, inflammatory bowel disease (Crohn's disease (CD), ulcerative colitis(CD)), ischemic colitis, allergic reactions, and microscopic colitis.

Most cases of colitis are involved in the damage of the intestinal mucosal barrier, particularly to those of ulcerative colitis.

Epidemiologically, damage and a reduction of gut bacteria by dietary chemicals, such as antibiotics of the protective mucus layer and the underlying tissue by the poorly inactivated digestive proteases have been found to induce both UC and CD.


The intestinal mucosal barrier is the outermost layer of the lining of the intestines which plays an essential role in ensuring the adequate containment of undesirable luminal contents within the intestine while preserving the ability to absorb nutrients.


Additionally, the intestinal mucosal barrier also processes various immunological mediators, including cytokines and chemokines stimulated by gut microbiota and host immune responses, thus maintaining a well-balanced relationship between gut microbes and the host immune system.

In other words, the intestinal mucosal barrier protects the colonic integrity by interacting with the gut microbiota and the immune response to prevent the inflammation of the colon that causes the onset of colitis.

Some researchers suggested disruption of the inaction of the immune system and gut microbiota caused by dysfunction of the intestinal mucosal barrier due to trauma, infection, neoplasia, and inflammation has been found to induce colitis in the experimental models.

Dr. K R Gardiner, the lead scientist in the examination of the mucosal barrier dysfunction as a feature of colitis irrespective of etiology or species wrote, "There were significantly increased systemic endotoxin concentrations in rats with acetic acid (7.5 (1.7-119.5) pg/ml), ethanol (13.7 (0-111.2) pg/ml), and hapten-induced (14.4 (5-31.1) pg/ml) colitis compared with saline controls (3.3 (0-13.7) pg/ml).


And, "There were significant correlations between the systemic endotoxin concentration and both the severity of colitis and of illness in acetic acid-induced colitis".

The finding strongly suggested that mucosal barrier dysfunction induces a significant increase in the chemical toxins involved in colitis.

Betanin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In finding a natural and bioactive compound with autophagy and anti-inflammation properties against the onset of colitis, researchers examined the effects of phytochemicals isolated from beetroot in an animal model.


The study included mice treated with intrarectal administration of 2,4,6-trinitrobenzenesulfonic acid (TNBS) (3.5 mg·20 g-1 ) induced colitis 2 or 4 days after treatment. Among the treated mice, some mice received trehalose (3% in drinking water 3 weeks before TNBS administration) or a daily administration of rapamycin (1.25 mg·kg-1, i.p.), betanin (1 g·kg-1, i.p.) or betanin + 3-methyladenine (3MA) (10 mg·kg-1, i.p.).

TNBS-treated mice alone showed impaired autophagy associated with body weight loss and intestinal damage.

Administration of trehalose, rapamycin, or betanin not only prevented the impaired autophagic flux induced by TNBS but also reduced the expression of pro-inflammatory cytokines in the intestinal mucosa.
The anti-inflammatory activity of trehalose, rapamycin, or betanin was also observed by a decrease of M1 macrophage markers.

The results suggested that betanin processes pharmacological stimulation of mucosal autophagy in reduced intestinal inflammation and improved murine colitis.

Furthermore, in patients with ulcerative colitis (UC) and Crohn's disease (CD). betanin also protected the colitic cells against oxidative DNA damage and induced apoptosis in neutrophils isolated from blood, through its antioxidant and anti-inflammatory properties.

The finding suggested that in normal intestinal cells, betanin protects the integrity of the cell against intestinal mucosal inflammation in the formation of colitis by inducing autophagy and anti-inflammatory effects.

Taken together, betanin used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of colitis, pending the confirmation of the larger sample size and multicenter human study.


Intake of betanin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
