#Alliin Works Synergistically with Other Antioxidants to Promote Anti-Angiogenesis and Anti Cancer Activity, Researchers Show

Angiogenesis is the process that promotes the formation of new blood vessels involving many factors including the migration, growth, and differentiation of endothelial cells.

Believe it or not, all active tissues in the body are at a distance less than a hundred micrometers from a blood capillary formed by the process of angiogenesis.

Angiogenesis is important for the cells for the diffusion exchange of nutrients and metabolites through the small blood vessels.

In normal and healthy conditions, stimulating angiogenesis has been found to have a therapeutic effect for the treatment of ischemic heart disease, peripheral arterial disease, and wound healing.


However, in cancers, stimulating angiogenesis in tumor cells has been associated with tumor proliferation and metastasis.

In other words, the angiogenesis process a function of a double sword, on one hand, the process provides an ideal condition for diseases and wound healing. On the other hand, it promotes invasive and overgrowth tumors.

Genentech in the article "Tumor Angiogenesis and the VEGF Pathway" wrote, "Angiogenesis is a necessary part of the process in the progression of cancer from small, localized neoplasms to larger, growing, and potentially metastatic tumors. To grow beyond 1 to 2 mm in diameter, a tumor needs an independent blood supply, which is acquired by the expression of growth factors that recruit new vasculature from existing blood vessels".

"This process continues even as the tumor matures. Thus, the upregulation of angiogenesis is a key step in sustained tumor growth and may also be critical for tumor metastasis".

In other words, by inhibiting the process of blood vessel formation of the tumor, cancer may be curable.

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

In the urgency to find a potential ingredient for the treatment of cancer with no side effect, scientists examined the phytochemical alliin, a compound derived from garlic anti-angiogenesis activity.

In the chick chorioallantoic membrane, the injection of alliin exerted a significant effect on the inhibition of fibroblast growth factor-2 (FGF2) associated with the angiogenesis.

In the CAM model, alliin also inhibited the vascular endothelial growth factor (VEGF)-induced angiogenesis.
Combined with antioxidant vitamins C and E significantly (P < 0.001) enhanced the function of alliin inhibitory efficacy on FGF2-induced EC tube formation and angiogenesis, particularly in the colon and fibrosarcoma tumor growth.

More precisely, alliin significantly inhibited both FGF2 and VEGF secretion from human fibrosarcoma cells compared to other types of tumors in a concentration-dependent manner

However, the anti-angiogenesis of alliin was reversed if the NO synthesis inhibitor nitro-L-arginine methyl ester (L-NAME) was injected.


The efficacy of alliin was attributed to the activation of the expression of a protein involved in immune tumor suppression.

In other words, alliin worked synergistically with other antioxidants to promote anti-angiogenesis and anticancer efficacy.

Dr. Mousa AS, the lead scientist wrote, "These data also suggest the implication of cellular NO and p53 as mediators of anti-angiogenesis and anticancer effects of alliin".

Taken together, alliin may be considered a supplement for the inhibition of angiogenesis in the promotion of tumor growth, pending the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Scientists may have found a bioactive ingredient for the treatment of liver cancer with no side effects, according to studies.
The liver is the largest internal organ in the human body which plays a critical role in maintaining the integrity of cell walls, the production of vitamin D and steroid hormones, and aiding the digestive system in the absorption of nutrients, through the production of cholesterol.

Additionally, the liver also filters out waste from the blood coming from the digestive system before passing it to the other parts of the body.

Liver cancer is a medical condition caused by irregular cell growth in the tissues of the liver.

Most cases of liver cancer start in the cells on the surface of the inner lining of the liver tissues before penetrating into deeper layers of the liver.

At the early stage due to the small size of the tumor and lack of nerve tissues, most patients are asymptomatic.

However, at the advanced stage, cancer cells can travel a distance away from the liver to infect other healthy tissues and organs, leading to secondary metastasis.

In two HCC cell lines SMMC-7721 and MHCC-97H, injection of DIM inhibited the proliferation, migration, and invasion of cancer cells.

In nude mice, DIM significantly decreased the volumes of SMMC-7721 orthotopic liver tumors and suppressed lung metastasis.

Furthermore, DIM decreased the level of phospho-FAK (Tyr397) which is found elevated in both vitro and in vivo.


The DIM anti-cancer cells' efficacy on proliferation, migration, and invasion is probably due to the inhibition of the expression of matrixes (MMP2-9) associated with the promotion of cancer cell growth and invasion.

Dr. Li WX, the lead scientist wrote, "These results demonstrated that DIM blocks HCC cell metastasis by suppressing tumor cell migration and invasion".

Moreover, in order to reveal more information about DIM anti-liver cancer activity, researchers investigated whether diindolylmethane affects HA59T cells.

At concentrations of 10-75 μM, diindolylmethane killed cells in a concentration-dependent manner, particularly, in diindolylmethane (25-50 μM) induced apoptosis in a concentration-dependent manner.
The cytotoxic effect of diindolylmethane against the tested liver cancer cell line was not reversed by chelating cytosolic Ca(2+)


Where Calcium (Ca2+) ions are important for cellular signaling. Its flow plays an essential role in regulating several secondary messenger systems in neural adaptation for the visual, auditory, and olfactory systems.


The results suggested that DIM inhibited the onset and progression of liver cancer through a number of mechanisms, including proteins involved in anti-cell proliferation and migration, and the promotion of apoptosis.

Taken together, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of liver cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Gastric ulcers or stomach ulcers are conditions characterized by painful sores in the stomach lining. Unlike the gastric ulcers, peptic ulcers are sores found in the stomach and small intestines.

The exact causes of gastric ulcers are not clearly identified. However, according to epidemiological studies, a bacterial infection caused by H. pylori infection has been found in the range of 30 to 50% of people with stomach ulcers.

Other risk factors associated with gastric ulcers include long-term use of certain medications, such as NSAID which has been found to cause more than 50% of peptic ulcers and stress.

Treatment of gastric ulcers is depended totally on the underlying causes, including antacids for the reduced acid in the stomach, and antibiotics for the elimination of Helicobacter pylori.

Unfortunately, people with gastric ulcers infected by Helicobacter pylori treated with antibiotics have a higher risk of recurrence. In severe cases, surgery may be necessary.

Untreated or uncovered stomach ulcers may induce some complications, including stomach bleeding penetration, and cancer.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

With an aim to find a potential ingredient for the treatment of gastric ulcers with no side effects, researchers examined the possible mechanism of the protective effect of ginger on aspirin-induced gastric ulcers.

The study included 40 adult male albino rats randomized and divided into four groups (10 animals per each group) and orally received the following once daily for 5 days:

Group I: 3 ml of 1% carboxymethyl cellulose;

Group II: ginger powder (200 mg/kg body weight) suspended in 3 mL of 1% carboxymethylcellulose;

Group III: aspirin (400 mg/kg body weight) suspended in 3 ml of 1% carboxymethylcellulose in water.

Group IV: ginger and 30 minutes later, received aspirin suspended in 1% carboxymethylcellulose, in similar doses as received in groups II and III.

Before the treatment of ginger, aspirin-induced a significant increase in the macroscopic ulcer score, and gastric damage was observed by the shed and disrupted epithelium, mucosal hemorrhage, submucosal edema and leukocyte infiltration, loss of the mucus of the mucosal surface.

Furthermore, in the protein's perspective, Aspirin significantly increased the expression of apoptosis regulator Bax, inducible nitric oxide synthase (iNOS) associated with the production of proinflammatory cytokines and heat shock protein 70 (HSP70) involved stress response.


Injection of ginger protected the gastric integrity by restoring all damage caused by aspirin in the induction of gastric ulcer.

Dr. Salah Khalil M, said, "Ginger ameliorated the histological changes by reducing Bax and iNOS and increasing HSP70 expressions".

In other words, the overexpression of proteins associated with the onset of gastric ulcer also was significantly reduced by ginger administration.

Additionally, in a cross-sectional population-based survey of 8,821 adults (51.4% female) conducted in Krakow, Poland. to evaluate the coffee and tea consumption and risk of metabolic syndrome, using food frequency questionnaires, observation of the questionnaire returned from participants expressed an interesting association between high coffee consumption of 3 cups and the clusters of lower BMI, waist circumference, systolic and diastolic blood pressure, triglycerides, and higher HDL cholesterol than those drinking less than 1 cup/day.

The study apparently indicated that people drinking 3 cups of coffee daily and regularly have a favorably ameliorated risk of metabolic syndrome even after adjusting for potential confounding factors.

More importantly, the study also pointed out that high coffee consumption was negatively associated with waist circumference, hypertension, and triglycerides in women but not in men.

Furthermore, in the animal evaluation of the effect of coffee drinking on clinical markers of diabetes and metabolic syndrome in Zucker rats including Diabetic Zucker rats with metabolic syndrome and control Zucker rats, researchers at the Fluminense Federal Institute postulated that after animals received daily doses of coffee drink by gavage for 30 days, coffee consumption expressed a decreased risk of metabolic syndrome in reduced serum glucose, total cholesterol and triglycerides in compared to non treatment group.

Dr. Abrahão SA, the lead researcher said, "The results demonstrate that treatment with roasted coffee drink, because of its hypoglycemic and hypolipidemic effect, is efficient in the protection of animals with metabolic syndrome and diabetes mellitus type 2".

The findings from the above studies suggested that coffee and coffee caffeine drinking daily and regularly have a favorably profound effect in decreasing the risk of metabolic syndrome, particularly in women.

Osteoarthritis (OA), a form of arthritis, is a condition characterized by aging progression of wear and tear on a joint, affecting over 25 million people in the United States alone.

In a randomized open-label active-controlled clinical trial of 50 adults with osteoarthritis of the knee allocated to receive the green tea extract (in dosage form of the tablet) plus diclofenac tablet as "intervention group"; or: diclofenac tablet alone as "control group" for a period of four weeks, combination-treated group exerted a significant improvement of physical function in compared to control.

#Ginger Exerts Gastroprotective Effects through Its Antioxidant Activity, According to Studies

Ginger may have a significant effect on the prevention and treatment of gastrointestinal disorders, some scientists suggested.
Gastrointestinal disorders are medical conditions, involving the esophagus, stomach, small intestine, large intestine, and rectum.

Most common gastrointestinal disorders are very mildly associated with digestive discomforts that can be resolved in a few days.

The gastroprotective effect is an action that protects the gastrointestinal tract against the onset of any disease, including infection and inflammation.

The causes of digestive disorders can be multiple. Most gastrointestinal disorders are caused directly by the disorders of the digestive system such as poor diet patterns. However, long-term use of certain medications can also induce gastrointestinal disorders, including Manticholinergic medication, non-steroidal anti-inflammatory drugs, and antibiotics.

In the search for a diet therapy that can reverse and protect the digestive system against gastrointestinal disorders, researchers at the University of Florida, wrote, "Diet therapy through the elimination of offending foods such as wheat gluten or lactose, or inclusion of specialized products such as medium-chain triglycerides or elemental formulas, can sustain nutritional status".

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

With an aim to find a natural whole food that processes gastrointestinal protective effects, researchers looked into rhizomes of Zingiber officinale Roscoe (Zingiberaceae), commonly known as ginger, an important kitchen spice, possessing myriad health benefits.

In laboratory animals., ginger was shown to be effective in preventing gastric ulcers induced by nonsteroidal anti-inflammatory drugs [NSAIDs like indomethacin, aspirin], reserpine, ethanol, and acetic acid and Helicobacter pylori-induced gastric ulcerations.

The gastroprotective efficacy of ginger was attributed to its free radical scavenging, antioxidant; inhibition of lipid peroxidation activities.


Furthermore, in the evaluation of the gastroprotective effect to examine the turmeric (Curcuma longa) and ginger (Zingiber officianale), researchers conducted a study in the ethanol-induced ulcers animal model.

Selected rats were given 100, 500, and 1000 mg/kg body weight orally.


Application of TEO and GEO inhibited ulcers caused by the injection of ethanol by 84.7% and 85.1%, respectively, as seen from the ulcer index.

The reduced antioxidant enzymes such as GPx, SOD, catalase, and GSH produced by alcohol administration were significantly (p<0.001) increased by the simultaneous administration of TEO and GEO.

Ethanol-induced lesions such as necrosis, erosion, and hemorrhage of the stomach wall were significantly reduced after the oral administration of essential oils.


These results strongly suggested turmeric and ginger exerted a significant gastroprotective effect against gastrointestinal lesions with no side effects.

Taken together, ginger may be considered functional for the prevention and combined with primary medicine for the treatment of gastrointestinal disorders,
