The exact causes of esophageal cancer are unidentified. However, according to epidemiological studies, aging, male gender, smoking, excessive alcohol drinking, genetic preposition, obesity, and thermal injury to the esophagus are some prevalent factors that increase the risk of the disease.
Some researchers suggested that the increased incidences of esophageal cancer in the Western world over the past few decades may be correlated to the promotion of a high-fat diet.
Dr. Silvano Gallus wrote, "Over the last 30 years, the incidence of squamous-cell carcinoma has decreased in several areas of the world, but adenocarcinoma has risen by over fourfold in the US as well as in Australia and several areas of Europe; its incidence has particularly increased in men".
And, " diets high in saturated fat and cholesterol have been associated with an increased risk (of esophageal cancer)".
Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.
In finding a potential compound for the treatment of gastrointestinal tumors, researchers examined the effect of a purified cranberry-derived proanthocyanidin extract (C-PAC) on esophageal adenocarcinoma (EAC) cell lines, including acid-sensitive JHAD1 and OE33 cells, and esophageal tumor xenografts in athymic NU/NU mice.
According to the tested assays, C-PAC shared a strong activity in inducing the expression of a protein associated with cell death programming mainly in JHAD1 and OE33 cells.
Autophagy that promotes cancer cell growth and anti-apoptosis of JHAD1 and OE33 cells were also inhibited by the injection of C-PAC.
In vitro, C-PAC-induced cell death was attributed to the deactivation of the phosphatidylinositol-3-kinase (PI3K)/Akt and the mammalian target of rapamycin (mTOR) signaling pathways expression in the promotion of cancer cellular processes, including cell survival, growth, and differentiation.
Additionally, C-PAC not only inhibited the levels of anti-apoptotic proteins such as BCL-xL, Cytochrome C, PARP), pathways involved in the occurrence and progression of cancer but also exhibited the expression of proteins that induce cell cycle arrest.
Similarly, in vivo, oral delivery of C-PAC significantly inhibited OE19 tumor xenograft growth as the aforementioned activities.
In other words, in vitro and vivo purified cranberry-derived proanthocyanidin extract (C-PAC) eradicates the esophageal adenocarcinoma (EAC) cells through several mechanisms involved in pro-apoptosis, anti-anti-apoptosis, and blocking cellular division.
Based on the finding, researchers after taking other factors into account wrote, " C-PAC is a potent inducer of EAC cell death and is efficacious in vivo at non-toxic behaviorally achievable concentrations, holding promise for preventive or therapeutic interventions in cohorts at increased risk for EAC, a rapidly rising and extremely deadly malignancy".
Taken altogether, cranberry extract may be considered a remedy for the prevention and treatment of esophageal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Other, in a cross-sectional study of a community-based sample of 243 women with vasomotor symptoms to evaluate the menopause symptoms of hot flashes (HF) and night sweats (NS) in symptomatic middle-aged women, intake of coffee showed to elevate symptom of hot flashes (HF) and night sweats (NS) in compared to nondrinkers.
And caffeine intake also is found as one of the predictors in exhibited both hot flashes (HF) and night sweats (NS) in the same study group.
But in the animal model, according to the behavioral tests and oxidative stress parameters in ovariectomized mice, caffeine intake also showed a significant in reduced symptoms of anxiety induced by oxidative stress.
Administration of low doses of caffeine, for a short period of time, maybe a new therapeutic approach to modulate the oxidative stress and anxiety in menopause, the "Iuliu Hatieganu" University of Medicine and Pharmacy postulated.
Taken together, coffee and coffee caffeine may have a strong effect in reduced and elevated menopause symptoms, in types depending manner.

Scientists may have found a bioactive compound for the prevention and treatment of pancreatic cancer, according to studies.
Cancer of the pancreas or pancreatic cancer is a condition of a malignant neoplasm of the pancreas.
Most cases of primary pancreatic cancer start in the cells on the surface of the inner lining of the pancreas as a result of irregular cell growth caused by the alternation of DNA.
The pancreatic cancers start in the ducts (an exocrine component of the pancreas) carried pancreatic juices with cancer of an epithelium that originates in glandular tissue and accounts for over 75 - 90% of all cases of exocrine cancers.
According to the statistics, pancreatic cancer is the fourth leading cause of cancer death in both men and women in the US. Cancer has the poorest prognosis compared to other major types of cancer, with a 5-year survival rate of approximately 5%.
More precisely, in the US, the age-adjusted incidence of pancreatic cancer is higher in men compared to women regardless of ethnicity.
Epidemiologically, the increase in age, gender, ethnicity, smoking, being obese and obese, and certain medical conditions such as diabetes, and family history are associated with an increased risk of pancreatic cancer.

Where miR-21 simultaneously regulates multiple programs that enhance cell proliferation, apoptosis, and tumor invasiveness by targeting the genes associated with cancer proliferation.
In human pancreatic cancer cells (Panc-1), injection of 13C inhibited the expression of miR-21 positively correlated in a time- and dose-dependent manner.
13C and gemcitabine combination therapy increased cytotoxicity in Panc-1 cells.
The efficacy of 13C was attributed to its activity in promoting cancer cell apoptosis by attenuating miR-21 transfection.
The finding suggested that I3C would be effective in enhancing the sensitivity of pancreatic cancer cells to gemcitabine via the inhibition of the overexpression of miR-21.
An additional experiment demonstrating the ability of a dietary agent, indole-3-carbinol (I3C), to lower the LD(50) of gemcitabine (Gemzar) in decreasing growth of both male (MiaPaca2) and female (SU86.86) pancreatic cancer cells, researchers found that* Without an injection of 13C, female pancreatic cancer cells were more resistant to gemcitabine.
* In male MiaPaca2 cells application of 1, 10, or 100 μM of I3C induced apoptosis by reactivating the tumor suppressor gene in pancreatic cancer cells.
* 13C also protects the healthy pancreas cells by enhancing the activity of the tumor suppressor gene.
Moreover, by examining the DNA methylation researchers suggested suppressor tumor gene (p16INK4a) inactivation in the promotion of hypermethylation is considered an early event in pancreatic carcinogenesis.
DNA methylation is a process by which methyl groups are added to the DNA molecule.
These results clearly indicated, that I3C, a possible non-toxic hypomethylating agent, combined with the anticancer agent, gemcitabine, is a powerful strategy for treating pancreatic cancer.

Renal fibrosis is a progressive condition caused by excessive accumulation of extracellular matrix including collagen, enzymes, and glycoproteins which provided structural and biochemical support of surrounding the renal cells during aging and chronic kidney disease (CKD).

Most cases of renal fibrosis ultimately lead to end-stage renal failure and a need for dialysis or kidney transplantation.
At the early stage of renal fibrosis, patients may experience symptoms of dull pain in the abdomen or back, pain on one side between your upper abdomen and back, leg pain. discoloration swelling in one or both legs.

Furthermore, DIM also inhibited the expression of Smad2 phosphorylation in the formation of renal scar tissue.
Based on the results, researchers concluded that DIM protects renal cell integrity by inhibiting TGF-β/Smad2/3 signaling against the formation of renal fibrosis.

In the urgency to discover the potential compound for the treatment of renal fibrosis, researchers examined the effects of 3,3'-Diindolylmethane (DIM) a natural acid condensation extracted from cruciferous plants, on renal fibrosis.

In the UUO model, DIM inhibited the expression of fibronectin, and collagen-1 associated with the formation of scars but retained E-cadherin involved in the formation of adherens junctions to bind cells with each other.
In other words, DIM protects the renal cells against the induction of renal fibrosis.
In vivo and in vitro, the administration of DIM is also found to suppress local fibroblast activation observed by the levels of myofibroblast markers α-SMA and vimentin.
Most importantly, DIM significantly inhibited the levels of pro-inflammatory cytokine-induced abnormal proliferation of NRK49F cells, in a time- and dose-dependent manner.
Furthermore, DIM also inhibited the expression of Smad2 phosphorylation in the formation of renal scar tissue.
Based on the results, researchers concluded that DIM protects renal cell integrity by inhibiting TGF-β/Smad2/3 signaling against the formation of renal fibrosis.
Taken together, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of renal fibrosis, pending the confirmation of the larger sample size and multicenter human study.
Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytic acid (Inositol hexaphosphate) may have a potential effect in reducing the risk and treatment of cancers, some scientists suggested.
Cancer is a class of chronic diseases associated with the growth of irregular cells that start in the cells of the inner lining of a tissue or organ caused by DNA alternation.
At the advanced stage, cells of the invasive tumor may travel a distance away from the primary site to infect other healthy tissue of the organs, through lymph and blood, leading to secondary metastasis.
Some cancers can be slow-growth such as prostate and colon cancer.
Primary cancer at stage 0 and 1 do not induce any symptoms. However, at the later stage, a larger size tumor can suppress the nearby tissue and nerve cells leading to localized and severe symptoms and pain, depending on the type of cancer.
According to the statistics, in the US, the number of new cases of cancer(cancer incidence) is 439.2 per 100,000 men and women per year (based on 2011–2015 cases). The number of cancer deaths (cancer mortality) is 163.5 per 100,000 men and women per year (based on 2011–2015 deaths).
Cancers who are diagnosed in the early stage have a 5 years survival rate close to 100%. Therefore, the annual examination may be necessary for people who are classified as having a higher risk of cancer development.
The causes of cancer are unknown. researchers cannot explain why people with the same healthy condition, and family history in the same family, are susceptible to the onset of the disease while others do not.
Either they can explain why people who carry some of the risk factors associated with the development of cancer never have cancer, while others having no risk factors are diagnosed with cancer.
Conventionally, the treatment of cancers completely depends on the types and grades of cancer. Some people with cancer at an early stage may only need one treatment. However most people with cancer in the later stage have a combination of treatments, including surgery with chemotherapy and/or radiation therapy.
Phytic acids (Inositol hexaphosphate) are phytochemicals of the organic acid found abundantly in nuts, sesame seeds, soybeans, wheat, pumpkin, beans, almonds, etc.
With an aim to find a natural ingredient for the prevention and treatment of cancers without inducing side effects, researchers examine the anticancer effects of phytic acid (IP6).
In vitro and in vivo phytic acid (IP6) exerted a strong anticancer activity by inhibiting the tumor growth and cell proliferation in a series of cancer cell lines, including colon, nipple, breast, prostate, cervix, liver, pancreas, melanoma, and glioblastoma.
The efficacy of IP6 was probably associated with the promoted antioxidant production, improved signal transduction, enhanced functions of the NK-cells, and exhibited cell cycle arrest and activated proteins associated with cell death programming.
Furthermore, in the findings, the proper role for the treatment of cancer researchers looked into the concentration of IP6 and its lower phosphorylated forms (IP(1-5)),
At the 10 microM to 1 mM concentrations, IP6 and its lower phosphorylated forms (IP(1-5)) showed a potential activity in regulating vital cellular functions such as signal transduction, cell proliferation, and differentiation, similar to those of the previous study.
In vivo and in vitro, exogenously administered IP6 and internalized, IP6 and IP(1-5) inhibited cell growth.
Moreover, researchers found that the application of IP6 not only decreased cellular proliferation but also caused the differentiation of malignant cells often resulting in a reversion to a normal phenotype.
The results strongly indicated that IP6 inhibits the cancer cell lines through a number of mechanisms, including cell cycle regulatory genes, differentiation genes, and tumor suppressor genes.
Taken together, phytic acid (IP6) may be considered an anti-cancer bioactive compound for the prevention and treatment of cancer with no side effects, pending the confirmation of a large sample size and multicenter human study.
