#Casticin Found in #Chastetreeberry Induces Cytotoxicity Agaisnt #Leukemic Cells, Researchers Find

Oral Administrated ripe fruit of Vitex agnus-castus (Vitex) may reduce the risk of various types of solid tumor cells, particularly leukemia.

Leukemia is a condition of abnormal increase of white blood cells produced by the bone marrow and/or the lymphatic system. Depending on the malignant granulocytes or lymphocytes, leukemia is classified into myelogenous or lymphoblastic leukemia.

Chaste tree berry is a species of Vitex agnus-castus, genus Vitex, belonging to the family Lamiaceae, native to the Mediterranean region, used in herbal medicine for thousands of years as an aphrodisiac herb and considered as Queen herb in treating menstrual problems and discomforts.

According to the Tokyo University of Pharmacy and Life Sciences, casticin,, the major chemical compound found in extract from the ripe fruit of Vitex agnus-castus (Vitex) exhibited strong cytotoxic activities against leukemia cell lines, HL-60 and U-937 in dose depending manner.


Additionally, the study also expressed an interesting result as the levels of cytotoxicity of Vitex/Casticin were largely attributed to the degree of differentiation of leukemia cells depending on cancer cell line maturity. compared to the HL-60 cell line with a less mature U-937 cell line.


Dr. Kikuchi H, the lead author said, "Casticin accounted for approximately 1% weight of Vitex. Dose-dependent cytotoxicity of Vitex and casticin was observed in both cell lines, and HL-60 cells were more sensitive to the cytotoxicity of Vitex/casticin compared to U-937 cells".

In a further study, casticin, also displayed significant anti-leukemia activity in cells through an expression molecular mechanism. apoptosis and cell cycle arrest were observed through different assays.


The application of casticin yielded a result of cancer cell death at 75.3% at 12 in the G2 phase in tumor cell cycle division, increased and subsequently declined to 27% at 48 h which was an indication of leukemic cell death via apoptosis and mitotic catastrophe.


Another study in support of the above differentiation suggested that the extract from the ripe fruit of Vitex angus-cactus (Vitex), inhibited the leukemia cell lines probably due to other mechanism such as significant decrease levels of intracellular reactive oxygen species (ROS) and level of gp91(phox), an important component of NADPH oxidase, in production of oxidation, and mediated cytotoxicity in HL-60 cells by regulating ROS generation...


Taken together, Chaste tree berry has a significant implication in reduced risk and treatment of leukemia through a number of mechanisms, but intake of large amounts should be taken with care to prevent toxicity.

#Nishamalaki (NA), a Combination of Curcuma Longa and Emblica Officinalis Effectively Prevents the Development of Diabetic Neuropathy

Diabetic neuropathy is a complication of patients with diabetes associated with nerve damage, ranging from pain and numbness in your legs and feet to problems with your digestive system.
Most cases of diabetic neuropathy are involved in diabetics who have a history of hyperglycemia caused by long-term high levels of glucose in the bloodstream, urinary tract, blood vessels, and heart.


Diabetic neuropathy can be classified into 4 main types, including.

* Peripheral neuropathy is the most common type of diabetic neuropathy, affecting the nerves that control the feet and legs first, followed by the hands and arms.

* The autonomic neuropathy that affects the nerves that control the heart, bladder, stomach, intestines, sex organs, and eyes.

* Radiculoplexus neuropathy found mostly in older patients affects nerves that control the thighs, hips, buttocks, or legs.

* Mononeuropathy or focal neuropathy also found commonly in older patients affects the nerves that control the face, middle of the body (torso), or leg.

The exact causes of diabetic neuropathy are totally depending on the type of neuropathy. However, damage of nerves and blood vessels in the affected area has been found to induce over time, uncontrolled high blood sugar.

In other words, diabetic neuropathy is preventable by keeping the blood glucose in the healthy range.

Dr. Michael Greger M.D. FACLM in the article "the best diet for diabetic neuropathy" wrote, "

There are blood vessels within our nerves that can get clogged up too. The oxygen levels in the nerves of diabetics were found to be lower than even the levels of oxygenated blood".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.
In the urgency to find a potential ingredient for the treatment of diabetic neuropathy researchers examined the turmeric's antioxidant properties, anti-inflammatory, CNS depressant effect, and antibacterial and antifungal properties.

According to the results of the experiment, turmeric inhibited the pathogenesis of diabetic neuropathy by improving the antioxidant in the reduction of the level of oxidative stress associated with the occurrence of the present complication.

The study included diabetic neuropathy in type 2 diabetic Wistar rats under treatment for 12 weeks.

Selected rats were randomly divided into group I (Control) vehicle-treated consisting of 6 rats and
group II-VII Diabetic Control, NA low dose, NA High dose, Glibenclamide, Pioglitazone, and Epalrestat respectively.

Diabetes-induced in 36 Wistar rats with Streptozotocin (STZ) (35mg/kg) intraperitoneally followed by High Fat High Fructose diet. After confirmation of the development of diabetes; rats were divided into six groups as the aforementioned division.

Compared to other treatment groups, Nishamalaki (NA) not only controlled hyperglycemia and reduced lipid levels but also effectively prevented the development of diabetic neuropathy.

Taken together, turmeric used alone or combined with other herbal medicines may be considered supplements for the prevention of diabetic neuropathy, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Fibrinolysis is the condition associated with the enzymatic breakdown of the fibrin in blood clots.
Where fibrin plays an essential role in hemostasis as the primary product of the coagulation cascade of fibrinolysis.

In other words, fibrinolysis is a process that prevents blood clots from growing to induce certain health problems such as cardiovascular disease.

Platelet aggregation is a condition characterized by platelets clumping together in the blood. The process also is necessary for the cover of vascular injury. However, overexpression of platelet counts in the blood is problematic to human health.

In other words, the production of blood platelets is to protect tissue injury against infection.
Furthermore, platelet disorders can lead to abnormal bleeding. The most common types of the genetic proposition of platelet disorders including

* Disorder of platelet adhesion, also known as Bernard-Soulier Syndrome is a condition in which the platelets fail to adhere together, leading to frequent bruises, nose bleeds, and bleeding in the mouth and gums.

The study included blood samples taken from the antecubital vein of 10 healthy male non-smokers.

According to the tested observation, platelet count decreased after incubation with 2.5 and 5 mg bromelain/ml.


The levels of ADP and TRAP-6 associated with platelet aggregation were inhibited after incubation with 2.5 mg or 5 mg compared to control.

Interestingly, according to the flow cytometry, the percentage of the total number of platelets or unstimulated CD62P positive platelets.


In other words, under normal conditions, bromelain does not have any effect on unstimulated CD62P positive platelets even with higher incubation with 5 mg bromelain/ml compared to control.


However, after TRAP-6 stimulation with platelet aggregation, the incubation with 5 mg bromelain/ml led to a remarkable decrease, compared to the untreated control.

The changes of CD62P (TRAP-stimulated) and the results of platelet aggregation after incubation with bromelain in vitro may demonstrate the potential of bromelain as a substance for platelet inhibition.

Based on the findings, researchers said, "The changes of CD62P (TRAP-stimulated) and the results of platelet aggregation after incubation with bromelain in vitro may demonstrate the potential of bromelain as a substance for platelet inhibition".

These results suggested that bromelain has a strong effect on the prevention and treatment of cardiovascular disease associated with platelet aggregation.


Taken together, bromelain may be considered a remedy for the prevention and treatment of fibrinolysis,, pending the confirmation of the larger sample size and multicenter human study.

Cardiovascular diseases (CVD) are a group of conditions associated with blood vessels and the heart.
According to the statistics provided by the World Health Organization, cardiovascular diseases are the number 1 cause of death globally. Approximately, 17.9 million people died from CVDs in 2016, representing 31% of all global deaths.

Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream to regulate glucose.

In other words, diabetes occurs when cells in the pancreas die off or receptor sites clogged up by fat and cholesterol. In some cases, diabetes is also caused by allergic reactions of cells in the immune system.

The most common types of diabetes are
* Type 1 diabetes, also known as juvenile diabetes or insulin-dependent diabetes, the most common form of diabetes found in children is a condition in which the pancreas does not produce any insulin. The most common form of type 1 diabetes is caused by genetic mutation and some viruses.

As of today, there is no cure for type 1 diabetes. Treatment focuses on managing blood sugar levels with insulin injection, diet, and lifestyle changes.

* Type 2 diabetes is a medical condition characterized by insufficient insulin entering the bloodstream to regulate the glucose due to either cells in the pancreas dying off or receptor sites clogged up by fat and cholesterol.

The most common symptoms of type 2 diabetes are an increased thirst that won't stop with water intake, increased hunger (especially after eating), dry mouth, frequent urination, and unintentional weight loss.

* Latent autoimmune diabetes in adults (LADA), similar to type 1 diabetes, is a slow-progressing form of autoimmune diabetes. The disease is caused by the pancreas's inability to produce enough insulin, because of the slow die-off of insulin-producing cells in the pancreas.

LADA is also known as type 1.5 diabetes because the disease has characteristics of both type 1 (autoimmune in nature) and type 2 diabetes.

In finding a potential plant that processes chronic diseases, researchers compared the effects of broccoli sprout powder, as an alternative and complementary treatment, to those of standard triple therapy, as a common medical treatment in cardiovascular diseases (CVD) risk in patients with type 2 diabetes.

The study included 86 type 2 diabetic patients with positive H.pylori stool antigen test (HpSAg) who were randomized to receive one of the three following regimens:
* STT) Standard triple therapy (omeprazole 20 mg, clarithromycin 500 mg, amoxicillin 1000 mg, twice a day for 14 days),

* BSP) 6 g/d broccoli sprouts powder for 28 days, and

* Combination of these as STT + BSP. After 4 weeks of treatment.

Based on the results of the analysis of 77 participants who completed the study [STT (n = 28), BSP (n = 25), STT + BSP (n = 24)],

* Before treatment, the H.pylori eradication rates were 89.3%, 56.0%, and 91.7% in STT, BSP, and STT + BSP groups, respectively.

* After the treatment, both systolic and diastolic blood pressure significantly decreased in the STT + BSP group (P < 0.05).

* Serum triglycerides and TG/HDL-C ratio increased in the STT patients group (<0.05).

* Serum hs-CRP levels significantly decreased in the patients who were treated with BSP per se (3.0 ± 2.5 at baseline vs. 2.3 ± 2.1 after the treatment, P < 0.05).

Collectively, researchers said, "Compared to standard triple therapy, BSP regimen in addition to considerable effects on H.pylori eradication had also favorable properties on cardiovascular risk factors following the H.pylori eradication".

Taken together, broccoli may be considered a functional food for reducing the risk of CVD in diabetes, pending the confirmation of the larger sample size and multicenter human study.

The redox status refers to the balance between the levels of free radicals and other reactive species and antioxidants.
Oxidants or ROS species can be produced by the body's cell metabolism or from external sources such as the air we breathe and the foods we eat.

Free radicals in moderate amounts are necessary for the body's functioning, inclduing NO for vascular relaxation.

Free radicals are atoms, molecules, or ions with unpaired electrons through chemical bonds with other atoms or molecules during a chemical reaction.

Antioxidants are stable molecules that play a critical role in protecting our body against oxidation.

There are 2 types of antioxidants, antioxidants produced by the host are called antioxidant enzymes and antioxidants come from dietary intake.

Under normal conditions, antioxidants produced by the host are good enough to keep the levels of free radicals in check.

Imbalanced levels of free radicals and antioxidants induced oxidative stress, a major cause of many chronic ailments. In other words, free radicals overexpression caused by chain oxidative reaction that induces oxidative stress causes protein, lipid, and DNA, leading to chronic diseases including cardiovascular disease, diabetes, and cancer.

According to Dr. Parvin Mirmiran, and colleagues in the joint study led by the Beheshti University of Medical Sciences, on oxidative stress in patients with metabolic syndrome, wrote, "(By) following a healthy pattern filled with fruits and vegetables ameliorates oxidative stress status and on the contrary, attachment to an unhealthy pattern, characterized by higher intakes of fast foods and processed foods, aggravated the oxidative stress levels in Tehranian individuals suffering from the metabolic syndrome".

Therefore, by enhancing antioxidant status in the body through regular physical activity and intake of fruits and vegetables which contain a high amount of dietary antioxidants to offset the free-radical toxic effects, the risk of oxidative stress can be reduced substantially.

Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains found abundantly in beets, chard, etc.

In finding a potential compound that processes chemo-activity, researchers examined the effect of Indicaxanthin as a natural chemoactive and chemopreventive agent.

The randomized, double-blind, placebo-controlled, and crossover design included 8 women (23.25±2.95 years old, weight of 54.13±9.05 kg, height of 157.75±0.66 cm, and BMI of 21.69±0.66 kg/m2) randomly divided into 2 groups and each group was supplied with either 50 ml OFI, diluted to 170 ml with water, or 170 ml Placebo containing the same concentration of fruit juice ingredients except for Vitamin C and indicaxanthin.

According to the blood samples from women taken before and after the effort test without supplementation (baseline), pre-and post-test, 24 h and 48 h post-test with OFI or Placebo supplementation every day for 3 days before of effort test on a cycle ergometer and continued to take it for 2 consecutive days after testing, free radical H2O2 levels were significantly decreased and total antioxidant capacity (PAT) significantly increase in the OFI group comapred to placebo.

In other words, by comparing the effect on OFI and placebo groups, the antioxidants vitamin C and indicaxanthin may be the major constituents in the induction of the efficacy of OFI in restoring redox balance.

Based on the results, researchers said, " OFI supplementation might be used to restore redox balance after intense exercises in moderately trained women".

Taken together, indicaxanthin may be considered a remedy for restoring redox balance after intense exercises in moderately trained women, pending the confirmation of the larger sample size and multicenter human study.

#Herbal #Turmeric Inhibits the Risk of Chronic Inflammatory Diseases through Its Antioxidant Activity, Researchers Suggest

Inflammation is a process of the immune system that responds to harmful pathogens.
Inflammation can be acute or chronic. In the acute phase of infection, the immune system after receiving information from the neurons, macrophages, and the largest white blood cells in the first line of defense stimulates the production of blood palette to cover the wound and the production of inflammatory cytokines to kill off micro-organisms, leading to symptoms of fever, redness, and swelling.

However, in some cases, the overproduction of proinflammatory cytokines may damage nearby healthy cells and tissues, causing the formation of scars.

In severe cases, a protein with duo functions in the production of pro and anti-inflammatory cytokines which produce anti-inflammatory cytokines under normal conditions also switch to produce pro-inflammatory cytokines.

Most cases of acute inflammation are resolved within a few days without causing damage to the wound tissues.

However, if the immune first line of defense can not completely destroy the invaders within a period of 3 to 8 weeks, it will compromise, leading to low-grade chronic inflammation.

Diseases associated with low-grade chronic inflammation include diabetes, cardiovascular disease, and cancer.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

In finding a potential compound for the treatment of chronic diseases, researchers examined the efficacy of turmeric on continued oxidative stress and oxidative damage that lead to chronic inflammation in the mediation of most chronic diseases.

At the molecular level, turmeric showed significant anti-inflammatory activity by suppressing the proteins associated with the production of pro-inflammatory cytokines, including the NF-κB and COX-2.

Furthermore, the proteins associated with the response to cytokines and growth factors such as signal transducer and activator of transcription 3 were also inhibited by the injection of turmeric.


Moreover, the administration of turmeric also inhibited the production of ROS by promoting the production of antioxidant enzymes such as nrf2, a protein that regulates the expression of antioxidant proteins that protect against oxidative damage.

Furthermore, numerous studies also indicated that curcumin not only is highly potent against antimicrobial disease but also inhibits the onset of various chronic diseases including cancers, diabetes, obesity, cardiovascular, pulmonary, neurological, and autoimmune diseases.


As of today, over 100 different clinical trials also showed that curcumin is safe, tolerable and effective against various chronic diseases in humans.

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of chronic inflammatory diseases pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Gastric cancer is a medical condition characterized by irregular and uncontrollable cell growth in the stomach lining in the beginning. At the advanced stage, the malignant cells can travel a distance away through blood and fluid circulation to infect other healthy tissues and organs.

Researchers do not know why some people with the same health condition are susceptible to the risk of gastric cancer, while others do not. The exact causes of gastric cancer are also unknown. In finding a natural ingredient for the prevention and treatment of gastric diseases, scientists examined the efficacy of Xiao Wei Yan Powder (XWYP) decoction on intestinal metaplasia (IM) and 104 cases of atypical hyperplasia (AH).

XWYP is a traditional Chinese Medicine formula containing Smilax glabrate, Hedyotis diffusae, Taraxacum mongolicum, Caesalpinia sappan, Paeonia alba, Cyperus rotundus, Bletilla striata, Glycyrrhiza uralensis, etc.

According to the results of 138 cases of intestinal metaplasia (IM) and 104 cases of atypical hyperplasia (AH) of the gastric mucosa of chronic gastritis that was treated by Xiao Wei Yan Powder (XWYP) orally 5-7g, for a period from 2-4 months, compared to control, the decoction showed a significant effect rate of IM was 91.3% and that of the AH was 92.16% compared to the control group of 21.3% and 14.46% respectively.

More precisely, XWYP not only had marked therapeutic effects for IM and AH but also showed no toxic effects in the animal experiments revealed no toxic effect,

These results strongly suggested that a safety guarantee was provided for its clinical application.

Taken together, Dandelion in combination with other Chinese medicines may be considered a remedy for the treatment of intestinal metaplasia and atypical hyperplasia (AH), pending the confirmation of the larger sample size and multicenter human study.
