#Bromelain Inhibits Free Radicals Associated with a Reduced Risk of #Oxidativestress, According to Studies

Kyle J. Norton

Antioxidants are stable molecules that play an essential role in the inhibition of oxidation caused by free radicals.

Free radicals are unstable and highly reactive molecules with a single electron in the outermost ring. Therefore, in order to maintain stability, free radicals either donate or intercept an electron from other stable molecules, leading to a chain of reaction that can not be stopped until all single electron in the outermost ring is paired or inhibited by antioxidants.

Overexpression of free radicals and reduced production of antioxidant enzymes by the body induce oxidative stress. Over time, oxidative stress can cause protein, lipid, and cell damage, leading to chronic diseases, including cardiovascular disease and diabetes.

There are 2 types of antioxidants. antioxidants enzymes produced by the liver and the dietary sources, including
* Antioxidant enzymes from dietary protein and minerals, including superoxide dismutase (SOD), glutathione peroxidase, glutathione reductase, and catalases.

* Antioxidant vitamins found in supplements or daily intake of food. such as vitamins A, C, E, folic acid, and beta-carotene. Most antioxidant vitamins process a free radical scavenging activity.

In other words, they gobble up free radicals before they can cause a chain of reaction.

* Antioxidant phytochemicals are found mostly in the plant's second metabolites including fruits and vegetables, carotenoids, flavonoids, allyl sulfides, and polyphenols.

Believe it or not, in a healthy individual, antioxidants produced by the liver are more than enough to balance the production of free radicals. However, in people with a weakened immune system, intake of antioxidants from dietary sources and supplements may be necessary.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a potential ingredient for the treatment of disease associated with oxidative stress, researchers evaluated the ameliorative efficacy of bacoside A and bromelain on oxidative stress biomarkers in the serum of dichlorvos-intoxicated mice.

The experiment included mice were allocated into six groups. The first group received saline as a vehicle; the second group was administered with dichlorvos (40 mg/kg b.w.); the third group was administered with bromelain (70 mg/kg b.w.), the fourth group received a dose of bacoside A (5 mg/kg b.w.), the fifth group was given concomitant exposure of bacoside A and bromelain both and mice of the sixth group were exposed to bacoside A, bromelain and dichlorvos for 21 days consecutively.

The group treated with dichlorvos showed a significant increase in oxidative stress biomarkers thiobarbituric acid reactive substances (TBARS) and protein carbonyl content (PCC).

The results suggested that dichlorvos stimulates the production of free radicals in mice.

Bromelain and dichlorvos treated group showed a strong effect in the inhibition of the free radical expression induced by dichlorvos.

Furthermore, co-administration of bromelain and bacoside A in dichlorvos-exposed mice also displayed a significant effect in reversing the level of CAT, SOD, GPx, and GSH (p < 0.05) which was decreased due to the dichlorvos administration.

Based on the findings, researchers said, "Findings of research conclude that concomitant exposure of bacoside A and bromelain was much effective in combating oxidative stress induced by dichlorvos".

Taken together, bromelain may be considered a supplement for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Herbal #Angelica Sinensis Inhibits #Liverstress Maekers Associated with LiverToxicity and Injury, Researchers Show

Kyle J. Norton

The liver is the largest internal organ in the human body and the only organ that can regenerate itself. The liver is the first line of the bodily toxin defense by filtering the blood that comes from the digestive tract, before passing it to the rest of the body.

The liver also produces cholesterol that plays a critical role in building strong cell membranes, producing steroid hormones and vitamin D, and aiding the digestive function in food absorption and detoxifying chemicals, and metabolizing drugs.

The hepatoprotective or liver protective effect is an action that maintains the liver's healthy functioning against any harmful toxins, including detoxification which is a natural process of our body to maintain the proper function and protect our organs and tissues against toxicity by eliminating toxins accumulated.

When they fail, the toxic accumulations cause a disruption of hormone production, damage the body's detoxification organs, and weaken the immune system, leading to hormone imbalance and chronic illness consequently.

Symptoms of liver diseases include jaundice, abdominal pain, swelling, swelling in the legs and ankles, itchy skin, dark urine, pale stool color, and a tendency to bruise easily.

There are no exact causes of liver disease. viruses, toxins, genetics, alcohol, and unknown causes are some prevalent factors found in patients with chronic liver disease, including cancer.

In the US, most cases of liver disease are a result of hepatitis viral infection and autoimmune diseases. Infectious viral hepatitis can produce toxins to protect them against the antioxidant enzymes produced by the host body and prevent the attack of the systematic immune response.

Furthermore, in recent years, excessive alcohol drinking has been found to induce alcoholic liver disease in many parts of Asia and America.

In finding a natural ingredient for the prevention and treatment of liver diseases, scientists examined the hepatoprotective effects of polysaccharides-enriched fractions from the root of Angelica sinensis in rodents.

Angelica or Dang Gui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen the heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the tested analysis,
* Intra-gastric administration of Angelica sinensis polysaccharides-enriched fraction (AP) at the doses of 50 or 75 mg/kg dose-dependently prevented liver toxicity induced by acetaminophen in mice but did not affect the serum acetaminophen concentration.
* AP inhibited the liver stress markers alanine transferase (ALT), malondialdehyde (MDA) hepatic nitric oxide synthase (NOS) activities, and glutathione level in the liver.

* In the carbon tetrachloride (CCl4) induced liver damage animals, AP showed weak protective activity.

* However, a higher dose of AP inhibited the MDA level without affecting the serum ALT level, the hepatic glutathione level instead while the NOS activity in CCl4 induced liver damage in rats.

Based on the results, researchers suggest, "These findings suggest that the pathogenic mechanisms of both acetaminophen and CCl4 are different. AP is more effective in the protection against liver damage induced by acetaminophen, which is associated with glutathione depletion and nitric oxide synthase activation in the liver".

Taken together, Dandelion may be considered a remedy for the protection against liver damage, pending the confirmation of the larger sample size and multicenter human study.

#Herbal #Angelica Sinensis Inhibits Genes Associated With Weight Gain, According to Studies

Kyle J. Norton

Obesity is a medical condition caused by excess body fat accumulated to some extent that impacts personal health. The hallmark of obesity is attenuated metabolism and structural cell changes in the obese fat observed by reducing metabolic activity.

According to the Body Mass Index(BMI), people who have a BMI between 25 to 29.9 are considered overweight and a BMI of over 30 is an indication of obesity.

The formula of the BMI indexBMI= weight (kg)/ height (m2)

According to the statistics, 68% of the American population is either overweight or obese. However,
In 2016, more than 1.9 billion adults, 18 years and older, were overweight, including over 650 million who were obese worldwide.

Visceral fat obesity is associated with 90% of patients with ischemic heart disease. In obese, accumulation of visceral fat is predominant in the intra-abdominal cavity.

Visceral fat is a layer of fat that surrounds the organs. Overexpression of visceral fat has been found to promote insulin resistance, diabetes, heart disease, and inflammation.

However, in non-obese subjects, visceral fat accumulation is correlated with glucose intolerance, hyperlipidemia, and hypertension.

Obesity rates have increased for all age groups in the US over the last several decades, coincidently with the periods that promoted the Western diet.

Weight gain is an increase in body weight associated with some parameters, including increased muscle mass, fat deposits, excess fluids such as water, or other factors.

However, weight gain has also been found to be induced by serious medical conditions.

In finding a natural ingredient for the prevention and treatment of obesity, scientists examined the radix Angelica sinensis (RAS) suppressed body weight by altering the expression of the fat mass and obesity-associated (FTO) gene in an animal model.

According to tested analysis in mice fed with high-fat diet (HFD)-induced obesity,
* 36 differentially expressed genes (DEGs) associated with the increased onset of obesity were found in adipose tissues from the RAS supplementation group (DH) and control group (HC).

* Six out of the 36 DEGs identified to be involved in lipid metabolism, with the APOA2 gene a potential anti-obesogenic influence were inhibited by the injection of RAS compared to control.

In other words, RAS supplementation in HFD-induced obese mice was associated with an anti-obesogenic global transcriptomic response.

Based on the results, researchers suggest, "This study provides insight into potential applications of RAS in obesity therapy".

Taken altogether, Dandelion may be considered a remedy for the treatment of weight gain in obese, pending the confirmation of the larger sample size and multicenter human study.

#Xuebijing Supplemention Injection (XBJ) Including #Angelica Reduces the Mortality Rate in Patients with Sepsis, Researchers Find

Kyle J. Norton

Sepsis is a condition caused by chemicals released into the bloodstream to fight the infection that triggers inflammation throughout the body as a result of severe infection(a)(b). According to a study by the University of Utah, sepsis is the commonest cause of admission to medical ICUs across the world.

Mortality from sepsis continues to be high. Besides shock and multi-organ dysfunction occurring following the intense inflammatory reaction to sepsis, complications arising from sepsis-related immunoparalysis contribute to the morbidity and mortality from sepsis(c).

The most common risk factors associated with the prevalent factors of sepsis, term infants, while very low birth weight (VLBW) and preterm infants, obesity, operative vaginal delivery and age <25 years, elderly, patients in intensive care and with the weakened immune system, invasive devices, and alcoholism.

Diseases associated with Sepsis are
* Staphylococcal infections
Preterm infants are especially susceptible to late-onset sepsis which is often due to Gram-positive bacterial infections resulting in substantial morbidity and mortality(24).

* Elevated central venous pressure
Elevated central venous pressure is associated with impairment of microcirculatory blood flow in sepsis(25).

* Neonatal cellulitis
There is a report of a case of late-onset neonatal invasive group A streptococcal disease characterized by rapidly progressing cellulitis and development of sepsis(26).

* Methicillin-resistant Staphylococcus aureus
There is a report of the spectrum of community-acquired S. aureus infections and to compare the patients infected with methicillin-susceptible or methicillin-resistant strains among patients aged <20 years. Overall, 90 cases of community-acquired S. aureus were detected in an 11-year period(27).

* Bladder infection
Bladder bacteria is a common but unique cause of sepsis(28).

* Urinary tract infection
In the study to evaluate the effectiveness of Lactobacillus GG supplementation in reducing the incidence of urinary tract infections (UTIs), bacterial sepsis, and necrotizing enterocolitis (NEC) in preterm infants, found that seven days of Lactobacillus GG supplementation starting with the first feed is not effective in reducing the incidence of UTIs, NEC, and sepsis in preterm infants. Further studies are required to confirm our results in lower birth weight populations(29).

In finding a natural ingredient for the prevention and treatment of infectious diseases, scientists examined the effects of Xuebijing injection (XBJ), a Chinese patent medicine on sepsis.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the search of medical literature in Embase, PubMed, the Cochrane Central Register of Controlled Trials, China National Knowledge Infrastructure (CNKI), Chinese Science and Technology Journal Database (VIP), and the Wanfang database from inception to June 6, 2017, including relevant RCTs comparing XBJ combined with RT and RT alone for sepsis, * 1602 records and 16 RCTs (1144 patients) identified suggested that the combined therapy reduced 28-day mortality and body temperature.

* WBC count improved with combined medication at high dosages (one study included, 40/1144, MD -8.00, 95% CI -10.18 to -5.82) with no reduction at moderate dosages.

* However, moderate-quality evidence indicated positive results with low dosages.

Based on the results, researchers suggest, "This study suggested that supplementation with XBJ in addition to regular treatment may improve the 28-day mortality rate, APACHE Ⅱ scores, WBC count, and body temperature of sepsis patients without serious adverse events".

Taken together, angelica used combined with other herbal medicines may be considered a remedy for the treatment of sepsis, pending the confirmation of the larger sample size and multicenter human study.

#Dried Cranberries Inhibit Parameters Associated a Reduced Symptoms of #InflammatoryBowelDiseases (IBD), Researchers Reveal

Kyle J. Norton

Colitis is a medical condition characterized by the inflammation of the large intestine, including the colon, caecum, and rectum.

Most cases of colitis are caused by infection, such as bacteria like C. difficile, viruses, and parasites, and inflammatory bowel diseases (IBD) such as Crohn's disease and ulcerative colitis (UC).

However, according to the study by the Catholic University of the Sacred Heart, depending on the causes, colitis can be classified into microscopic colitis, ischemic colitis, segmental colitis associated with diverticula, radiation colitis, diversion colitis, eosinophilic colitis, and Behcet's colitis.

The most common symptoms of colitis are abdominal pain and diarrhea, bleeding per rectum, distention with bloody diarrhea, abdominal bloating and flatulence, and changes in bowel habits.

In severe cases, patients may also experience symptoms of elevated liver enzymes, leukopenia, thrombocytopenia, severe peripheral arthralgias, and chronic watery diarrhea.

Believe it or not, fifty-five percent of patients stated that UC symptoms over the past year had affected their quality of life, while physicians and nurses estimated that 35% to 37% of patients would have a reduced quality of life.

Conventionally, treatments for colitis are totally dependent on the type and severity of the condition. For example, in patients with inflammatory bowel disease (IBD), Self-care at home through home tele-management, Web-guided therapy, and pain management are the most common approaches accompanied by biological therapy, if necessary.

Some researchers suggested the promotion of the Western diet over the past few decades may be the leading cause of colitis in patients with IBD.

Dr. Jimmy K. Limdi wrote, "The parallelism between “westernization” and particularly high dietary intakes of total fat (particularly animal fats, ω-6 polyunsaturated fatty acids (PUFA), milk fats), refined sugars, meat and lower intakes of fruit and vegetables, implicates diet in the great risk of developing IBD".

And, "Diet may influence intestinal inflammation through several biologically plausible mechanisms including dietary antigen presentation, alterations in the gut microbiome, the mucosal immune system and epithelial barrier function among others".

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry is cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine for the treatment of wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of IBD, researchers examined the effects of cranberry products on preventing dextran sulfate sodium-induced murine colitis.

According to the tested differentiation, both cranberry extract and dried cranberries-fed groups had significant activity against the disease index.

However, dried cranberries were more effective in preventing colitis compared to that of cranberry extract.

Moreover, both cranberry applications also showed a significant effect in preventing the shortening of colon length,

Biomarkers associated with inflammatory bowel diseases (IBD) such as colonic myeloperoxidase activity were attenuated in animals fed dried cranberries compared to the controls.

Additionally, dried cranberries also inhibited much better results against the production of pro-inflammatory cytokines caused by the injection of dextran sulfate sodium, compared to the controls.

Based on the findings, researchers wrote, "The current report suggests that cranberries can be applied to prevent and reduce the symptoms of IBD".

Taken altogether, cranberry may be considered a remedy for the prevention and treatment of colitis in IBD patients, pending the confirmation of the larger sample size and multicenter human study.

#Carotenoids Inhibit the Risk of #Liverdiseases through Its Antioxidant Activity, Researchers Find

Kyle J. Norton

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing them to other parts of the body.
The liver also produces cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Liver disease is a class of medical conditions characterized by the partial or complete function of the liver, including liver failure.

The most common types of liver disease in the US are
* Hepatitis C (26%) is a liver disease caused by liver inflammation caused by viral Hepatitis C, leading to irreversible liver scars. Over time, the condition can also induce liver cancer.

* Alcoholic liver disease (21%) is a condition that causes long-term excessive alcohol drinking. It is known that years of alcohol abuse can cause the liver to become inflamed and swollen. Over time, it has been found to induce the formation of liver scars, known as cirrhosis. Cirrhosis is the final stage of liver disease.

* Hepatitis C plus alcoholic liver disease (15%)

* Cryptogenic causes (18%) is a condition with no apparent underlying reason. However, most researchers suggested that many cases of cryptogenic cirrhosis are likely caused by non-alcoholic fatty liver disease (NAFLD), a condition of fat accumulated in the liver.

* Hepatitis B and D (15%) are liver diseases caused by liver inflammation caused by viral Hepatitis C, leading to irreversible liver scars. Over time, the condition can also induce liver cancer.

The most common symptoms of liver disease are jaundice, abdominal pain, and swelling, swelling in the legs and ankles, itchy skin, dark urine color, pale stool color, bloody or tar-colored stool, chronic fatigue, and nausea or vomiting.

Carotenoids are a class of mainly yellow, orange, or red fat-soluble pigments, including lycopene and carotene, found abundantly in ripe tomato, pumpkins, carrots, corn, and daffodils.

On finding a natural ingredient for the treatment of chronic liver diseases, researchers examined the effects of carotenoids against reactive oxygen species and oxidative damage associated with the onset of liver damage.

Overexpression of free radical-induced oxidative stress in the liver not only induced lipid and protein oxidation but also caused the alternation of the liver cell DNA, leading to reduced function of the liver.

Dietary carotenoids as antioxidants not only protected the liver's cellular metabolism but also balance the ratio of free radicals in the liver, thus preventing the onset of liver oxidative stress, the major cause of chronic liver disease, including cancer.

In other words, carotenoids enhance liver functioning by inactivating reactive oxygen species and obstructing lipid and protein oxidative damage, the vital prophylactic strategic molecules.

In chemical differentiation,
* β-Carotene supplementation enhanced the levels of antioxidants in the tested subject that act as free radical scavengers such as vitamin C, glutathione, and glutathione-related enzymes against liver toxicity caused by the administration of aflatoxin-B1.

Furthermore, β-Carotene also inhibited liver disease associated with both HBV and HCV by suppressing the elevation of inflammatory and oxidative conditions in hepatocytes.

* Lycopene found abundantly in tomato and products of tomato possesses antioxidant, and detoxification abilities in many epidemiological and animal studies, thus preventing liver oxidative stress-induced chronic diseases.

* Lutein, another phytochemical and bioactive compound found in carotenoids has been found epidemiologically to possess a strong antioxidant activity by promoting the production liver antioxidant enzymes such as superoxide dismutase, catalase, glutathione peroxidase, and glutathione, against diseases associated with oxidative stress including antiviral activity against hepatitis B.

* Other carotenoids such as β-Cryptoxanthin, α-carotene, fucoxanthin, and zeaxanthin also show promising effects against chronic liver injury through their antioxidant activity.

Taken together, betacyanins used alone or combined with conventional medicine may be considered a remedy for the prevention and treatment of adjuvant rheumatic diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of carotenoids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Turmeric's #Curcuminoid (CUR) and #Extracts Exert #AntiAging Effects through Their Antioxidant Activities, Researchers Say

Kyle J. Norton

Aging is a natural and biological process that cannot be prevented. Skin aging is one of the most visible processes that occurs constantly in our skin organs.

However, actively engaging healthy living lifestyle by increasing the intake of good healthy food such as whole grains, fruits, vegetables, beans, and legumes, etc., and reducing the consumption of harmful foods, such as saturated fat, trans fat, artificial ingredients, etc. accompanied with moderate exercise has been found to delay aging progression.

In other words, it is possible that a person is physiologically younger than his or her biological age.

There are many theory that explains the aging process. According to the free radical theory, oxidative damage to the enzymes, other proteins, unsaturated fatty acids, phospholipids, DNA and RNA, etc., caused by free radical chain reaction can lead to aging of the organisms by attacking the lipid bilayers of the cell walls.

You can view other theories of aging by clicking this link.

There are many risk factors associated with aging and premature aging processes. Dr. Bhatt N in the assessment of the risk factors and self-perception for facial aging among the Nepalese population wrote, "Around 50% of attendees were conscious about facial health, 53% perceived their facial age as their chronological age, 39% used some facial care products, and 68% were aware of the risk factors for facial aging".

"The major perceived risk factors were unhealthy eating habits, stress, less exercise, dehydration, diseased state, and sleeping habits. A significant association was found between increasing age, place of birth in hills or mountains, and sun exposure with high wrinkling score".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

In finding a better compound for the treatment of skin aging, researchers determined the antiaging activity of Curcuma heyneana to prove its traditional use as skincare, for cosmetic and body freshness.

In vitro, according to the tested assay, the extracts showed a dose-dependent manner in the final concentration range from 62.5 to 500 µg/mL in the inhibition of free radical expression and 31.25 to 250 µg/mL for tyrosinase and collagenase.

The extract containing a high amount of curcuminoid (CUR), the active principle of Curcuma is the best antioxidant, that acts as a tyrosinase inhibitor, and collagenase inhibitor, compared to those with a low concentration of CUR.

The topical application of the crude extract also induced significant improvement effects on the UV-induced skin structure damage.

By examining the efficacy of constituents of the extracts, researchers found that total CUR content was correlated with the anti-aging activity.

Based on the findings, researchers said, "The results show that C. heyneana contains antioxidant compounds and has potent anti-aging activity, indicating that it can be used as an anti-aging drug candidate or as a photo-cosmeceutical".

Taken altogether, turmeric in the form of an extract may be considered a supplement used for the treatment of skin aging, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
