#Chastetreeberries Process Anti-Nociceptive in Reducing Symptoms and Improving Quality of Life in Women With PMS

Chaste tree berry is a species of Vitex agnus-castus, genus Vitex, belonging to the family Lamiaceae, native to the Mediterranean region, used in herbal medicine for thousands of years as an aphrodisiac herb and considered a Queen herb in treating menstrual problems and discomforts.

Premenstrual syndrome is a faulty function of the ovaries related to the women's menstrual cycle. It affects a woman's physical and emotional state and sometimes interferes with daily activities as a result of hormone fluctuation.

In an acetic-acid-induced writhing and metoclopramide-dihydrochloride-induced hyperprolactinemia mice study, mice treated with Chaste tree berry factions showed a significantly effective expression of anti-nociceptive and anti-hyperprolactinemia activities.

The study also found that the fraction extracted from the herb also exerted anti-nociceptive activity in a dose-dependent manner (10-50 mg/kg body weight, i.e...).

Dr. Hu Y, the lead researcher in the study said, "Our results indicate that the presence of flavonoids such as Casticin in this plant may be responsible for the (anti-nociceptive and anti-hyperprolactinemia) activity effects. Casticin has potent analgesic and anti-hyperprolactinemia properties, is likely to be one of the active components of Fructus Viticis, and may have a role in treating PMS (premenstrual syndrome)".

Further study in support of the above differentiation also insisted that the 80% ethanol extract isolated from the herbal medicine showed the highest anti-nociceptive activity in acetic acid-induced writhing test in ICR mice in comparison with other factions. The study also indicated that the anti-nociceptive activity is mediated through inhibition of pro-inflammatory activity rather than through opioid receptors.


The evaluation of petroleum ether fraction (PEF) from the aqueous ethanol extract of Chaste tree berry at doses of 12, 24, and 48 mg/kg body weight, in nociceptive mouse models induced by intraperitoneal acetic acid and sub-plantar formalin injections and on thermal nociception in the hot-plate test, produced significant dose-related inhibitions on chemical nociception, probably due to partially mediated by its anti-inflammatory activity.

Taking together, the anti-nociceptive effect of Chaste tree berries should be beneficiary in reducing symptoms and treatment of women with premenstrual syndrome and improving quality of social life.
A large amount of intake of herbal medicine can be prescribed by related field specialists, including Chaste tree berries.

#Rapeseed Protein-Derived Peptide DHNNPQIR (RAP-8) Inhibits proteins Associated with the Risk of #LiverFibrosis, Researchers Say

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing it to other parts of the body.

The liver also produces cholesterol, a waxy substance that aids digestion, builds cell membranes, and produces vitamin D and steroid hormones.

Liver disease is a class of medical conditions characterized by reducing partly or completely the function of the liver, including liver failure.


Liver fibrosis is a condition characterized by overly exuberant wound healing causing excessive connective tissue builds up in the liver. In other words, hepatic fibrosis is a portion of the liver which was affected by scar formation.

The exact causes of liver fibrosis are unknown. However, certain risk factors such as repeated liver injury caused by a virus infection, and a blockage in the bile ducts are found to elaborate on the onset of liver fibrosis.

Long-term untreated liver fibrosis can lead to cirrhosis, a condition of severe scarring of the liver, and life-threatening liver disease, including liver cancer.

The most common scale to grade the stage of liver fibrosis is the METAVIR scoring system. By examining the grade and stage of liver fibrosis of patients, their doctors can now determine the degree of severity to which patients' liver is affected.

The severity of the grades ranges from A0 to A3:
A0: No activity
A1: mild activity
A2: moderate activity
A3: severe activity

The severity of fibrosis stages ranges from F0 to F4:
F0: No fibrosis
F1: Portal fibrosis without septa
F2: portal fibrosis with few septa
F3: Numerous septa without cirrhosis
F4: cirrhosis, the most severe stage of fibrosis


Broccoli is a mustard/cabbage plant, belonging to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of liver diseases, researchers examined the effect of the potential crucial pathways involved in ameliorating liver fibrosis of the rapeseed protein-derived peptide DHNNPQIR (RAP-8).

According to the analysis of the fibrotic and RAP-8 treated mice by measuring cell cycle and oxidative stress in LX-2 cells and liver samples,

* RAP-8 treated mice showed a change of modules that were mainly enriched in cell division, nuclear division, and mitotic cell cycle process by alternating the Kyoto Encyclopedia of Genes and Genomes associated with enriched in the cell cycle.

* More precisely, the regulations of three core genes (Cenpp, Cyp2c55, Serpinh1) by RAP-8 were verified to target for treating liver fibrosis.

* RAP-8 induced cell cycle arrest and prevents oxidation stress in the induction of liver fibrosis

Based on the results, researchers wrote, " As a promising therapeutic candidate for hepatic fibrosis treatment, RAP-8 exhibited anti-fibrotic effects via exerting cell cycle arrest and inhibiting oxidative stress."

And "However, because variability exists in the timing and methods of detection for SFN conjugate levels, it is difficult to compare systemic SFN levels across studies".

In other words, SFN protects the integrity of the neurons against neuron damage through its anti-neuroinflammatory and neuroprotective activities.

Taken altogether, rapeseed protein may be considered a supplement for the prevention and treatment of liver fibrosis, pending the confirmation of the larger sample size and multicenter human study.

#Sinigrin Reduces Numbers Aberrant Crypt Foci Against Colon Cancer through Its Apoptotic Activity, Researchers Find

The bioactive compound Sinigrin may be used for the prevention and treatment of aberrant crypt foci (ACF) in the lining of the colon and rectum, according to studies.

Aberrant crypt foci is a condition of clusters of abnormal tube-like glands in the lining of the colon and rectum. The condition can be subclassified into dysplastic, nondysplastic, or mixed type.

Crypt cells of the colon renew the lining of the intestine and produce mucus.

Epidemiological studies suggested that aberrant crypt foci have been found to form before colorectal polyps, the main cause of colon cancer.

According to the high magnification chromoendoscopy, aberrant cryptic foci are stained darker than normal crypts.

And, dysplastic ACF is found in 25- 30% of all patients with colon cancer due to the genetic preposition of mutation of KRAS.

Where KRAS protein is associated with the regulation of cell proliferation under normal conditions.

Dr. Takayama T and colleagues in the investigation of aberrant crypt foci in gene abnormalities and risk of colon cancer, wrote, "ACF, particularly dysplastic ACF, are precursor lesions of the adenoma-carcinoma sequence in humans. In most sporadic ACF, K-ras mutations were positive, but APC mutations were negative irrespective of nondysplastic or dysplastic features".

And, "In most sporadic ACF, K-ras mutations were positive, but APC mutations were negative irrespective of nondysplastic or dysplastic features. Conversely, in most ACF from familial adenomatous polyposis patients, APC mutations were positive but K-ras mutations were negative".

The results strongly support the genetic mutation of gene KRAS involved in the initiation of dysplastic crypt foci.

Sinigrin is a phytochemical glucosinolate, that belongs to the family of glucosides found abundantly in Brussels sprouts, broccoli, the seeds of black mustard, etc.

Scientists on finding a bioactive compound for the protection of colonic mucosal crypts against the aberrant crypt foci investigated the effect of oral administration of the isolated glucosinolate sinigrin in the suppression of aberrant crypt foci in the colonic mucosa of rats treated with 1,2-dimethylhydrazine (DMH).

During the experiment, glucosinolate sinigrin found in Brussels sprout modulated crypt cell apoptosis and mitosis, and the frequency of aberrant crypt foci in the colon.

Oral administration of uncooked Brussels sprouts, whether as a juice or as a freeze-dried powder, was associated with significantly enhanced levels of apoptosis and reduced mitosis in the colonic crypts.

The findings strongly suggested that the inhibition of colonic mucosal crypts was attributed to other chemical compounds found in freeze-dried raw, microwave-cooked Brussels sprouts and sprout juice.

In-depth analysis of the activity, researchers found that the efficacy was attributed to the glucosinolate breakdown products derived from Brassica vegetables on the balance of colorectal cell proliferation and death in an animal model of precancerous colon polyp.

Additionally, in order to reveal more information about the sinigrin effects on the colonic mucosal crypts, researchers examined the sinigrin on the intestinal mucosa of rats treated with dimethylhydrazine (DMH).

Sinigrin was shown to inhibit the aberrant crypt foci at 18, 24, 38, 48, and 72 h after injection, compared to control groups that were given DMH only, or a sham injection.

Before the injection of sinigrin, tested rats showed higher numbers of aberrant crypt foci in colonic tissue from both groups of DMH-treated rats and the controls.

Moreover, the experiment of rats given sinigrin (400 microg/g diet) 22 h after the second of two injections of DMH, showed that sinigrin reduced numbers of ACF by increasing the levels of apoptosis compared to those of higher DMH and controls.

The findings clearly support the use of sinigrin for the protection of colonic mucosal crypts.


Taken altogether, sinigrin in the form of glucosinolates, the sulfur compounds may be considered a supplement for the prevention and treatment of aberrant crypt foci, pending the confirmation of a larger sample size and multicenter human study.

#Cranberry Powders, a Potential Hyperglycemia Management Agent Against Diabetics Complications

Diabetes is a medical and chronic disease characterized by insufficient insulin entering the bloodstream to regulate glucose.

The disease is either caused by cells in the pancreas that died off or receptor sites clogged up by fat and cholesterol, such as type 2 diabetes.

Most cases of type 1 diabetes are associated with a gene mutation that causes the immune to attack the insulin-producing beta cells in the pancreas.

Acquired diabetes in most cases is caused by unhealthy lifestyle choices, including a high-fat diet and physical inactivity.

According to the statistics provided by the CDC, approximately 23.1 million people in the United States are living with diagnosed diabetes, costing the healthcare system more than $245 billion per year.

Sadly, there is an estimation that another 7.2 million people have diabetes but remain undiagnosed, accompanied by another 84.1 million adults 18 years and older with prediabetes.

Hyperglycemia, a hallmark of diabetes is a condition of abnormally high blood glucose in the blood.

Most cases of hyperglycemia are a result of poor management of blood glucose including uncontrolled diet, physical inactivity, miss-medication, and improper use of insulin injection.

Uncontrolled diabetes or hyperglycemia over the long term can lead to diabetic complications, including coronary heart disease, stroke, hypertension, depression, pain, polypharmacy, and functional disability.

Long-term elevated glucose values above 180 to 200 milligrams per deciliter (mg/dL), or 10 to 11 millimoles per liter (mmol/L) have been found to induce symptoms of frequent urination, increased thirst, blurred vision, fatigue, and headache.

In severe cases, some patients may also experience life-threatening complications such as fruity-smelling breath, shortness of breath, weakness, confusion, and coma.

Dr in the study of hyperglycemia in the aging population wrote, "Higher levels of Sirt1 and PGC-1α and increased expression of gluconeogenesis pathway genes are found in the hyperglycemic NOD mice. Increased Sirt6 abundance is found in the hyperglycemic NOD mice, which might increase DNA damage repair".

And, "With aging, lower Sirt1 abundance and activity increased acetylated histone modifications and Sirt7 levels, and NOR methylation is found. Thus, whereas in normal aging cell metabolism is reduced, in the diabetic mice a compensatory mechanism may elevate Sirt1 and Sirt6 levels, increasing gluconeogenesis and DNA repair from the oxidative damage caused by hyperglycemia".

Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of diabetes complications, researchers examined the effect of aqueous solutions of two different cranberry powders (CP and CP-SAB) on hyperglycemia.

In vitro, in vitro CP and CP-SAB administration showed a significant inhibition of α-amylase, α-glucosidase, and angiotensin I-converting enzyme (ACE) associated with the management of hyperglycemia linked to type 2 diabetes.


Low levels of α-amylase have been found in patients with diabetes.

α-glucosidase is associated with the digestion of carbohydrates and is elevated in patients with diabetes

According to the chemical analysis, the efficacy of CP and CP-SAB that inhibits hyperglycemia was probably attributed to the chemical constituents p-Coumaric acid and quercetin derivatives, the main phenolic compounds identified in the cranberry powders.


Furthermore, increased with increased doses (1-5 mg/mL) of CP-SAB exerted the increased α-glucosidase inhibitory activity from 60% to 100%.

At 5 mg/mL, CP-SAB exerted a maximum of 40% inhibition against α-amylase.

Levels of ACE were also significantly inhibited by CP-SAB at the concentration of 100 and 200 mg/mL sample concentrations.

Based on the results, researchers wrote, "These in vitro results indicate the potential of cranberry powders as a dietary supplement and food-based strategies for potential hyperglycemia management".

Taken altogether, cranberry may be considered a remedy for the management of hyperglycemia, pending the confirmation of the larger sample size and multicenter human study.

The expression of oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

In some cases, this reaction also causes the long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

There is no single cause of oxidative stress. Some researchers suggested that a number of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Sadly, a prolonged period of oxidative stress present in the human body can cause oxidative damage to biomolecules, (lipids, proteins, DNA) and the death of healthy cells through cytotoxicity including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily to donate or steal an electron from other molecules, leading to the chain of domino effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibit the onset of free radicals before they can cause a chain reaction in facilitating protein, lipid, and cell damage.

Apoptosis refers to the death of cells which occurs as a normal and controlled part of an organism's growth or development associated with the limited cycle division.

In finding a natural compound for the treatment of malignant tumors, the researchers examined the effects of Angelica Sinensis' polysaccharide (ASP) anti-apoptotic activity.

Based on the experimental differentiation, IN H9c2 cell, treated with hydrogen peroxide (H2O2), A. Sinensis exerted protective effects against oxidative injury through its various active ingredients on the regulation of multiple pathways associated with pro-inflammatory cytokines induced by oxidative stress.

Furthermore, ASP pretreatment significantly decreased H2O2-induced cytotoxicity and apoptosis in H9c2 cells.

Moreover, H2O2-induced oxidative stress observed by the expression of oxidative stress and ER stress markers such as malondialdehyde production and lactic dehydrogenase release was also inhibited by the antioxidant activity of ASP.


Collectively, researchers said, " Conclusions Our findings indicate that ASP, a major water-soluble component of A. Sinensis, exerts protective effects against H2O2-induced injury in H9c2 cells by activating the ATF6 pathway, thus ameliorating ER and oxidative stress".


Taken altogether, Herbal Angelica may be considered a remedy that protects the cell against oxidative stress, pending to larger sample size and multicenter human study.

A neuroprotective effect is an event that protects the central nervous system (CNS) by relatively preserving the neuronal structure and/or function.

Neurological diseases are the medical conditions of CNS disorders. Anything that involves structural, biochemical, or electrical abnormalities in the brain, spinal cord, or other nerves, including brain damage is considered as neuro disease.

The most common neurodegenerative diseases in the elderly are dementia. Depending on the brain regions that are damaged or dysfunctional, dementia can be Parkinson's, Alzheimer's, and Huntington's disease.

There are many risk factors involved in the incidence of neurological disorders, including unhealthy lifestyle, infections, genetic preposition, and physical injury.

Genetic mutation inherited from parents such as the NOTCH3 gene is the most common autosomal dominant inherited that causes stroke and vascular dementia.

Oxidative stress is a result of an imbalance of the ratio of levels of free radicals and antioxidants caused either by overexpression of free radicals depletion of antioxidant enzymes or both.

Free radicals can be produced internally by normal cellular metabolism or from external sources such as the air we breathe, and smoking. Free radicals are unstable molecules with unpaired electrons on the outer ring.

In order for them to maintain stability, free radicals must either donate an electron or intercept an electron from other atoms, leading to a chain reaction that can not be stopped until the outer ring electron is paired or inhibited by antioxidants.

Oxidative stress has been found to be a major risk for the onset of neurological disorders in the Western world, particularly in North America.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen the heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

In finding a natural compound for the treatment of neurological diseases, the researchers examined the effects of Ligustilide (LIG) a principal active ingredient of Radix Angelica Sinensis on neuroprotective activity/

The aging model mice induced by subcutaneous (S.C.) injection of d-gal once daily for 8 weeks followed by LIG (80 mg/kg) were simultaneously administered orally.

Based on the experimental results,* LIG administration markedly improved the behavioral performance of d-gal-treated mice.

* LIG reduced the level of neuron stress markers MDA and increased the activity of Na(+)-K(+)-ATPase in the promotion of neurophysiology in the brain of d-gal-induced aging mice.


* Moreover, LIG significantly raised the expression of GAP-43 associated with the development and axonal regeneration and reduced cleaved caspase-3 to promote cell death and GFAP levels in brain injury in the brain of d-gal-treated mice.

Collectively, researchers said, "These results demonstrated that LIG improves d-gal-induced cognitive dysfunction and brain toxicity, which suggests that LIG may be developed as a new medicine for the treatment of age-related conditions".

Taken altogether, Herbal Angelica may be considered a neuroprotective remedy, pending a larger sample size and multicenter human study.

#Cranberry Improves Metabolic Syndrome throught its Antioxidant Activity, Researchers Find

Homocysteine produced in the bloodstream is a common amino acid in the blood from eating meat.
High levels of homocysteine in the blood have increased the risk of heart disease and stroke.

In other words, homocysteine induced atherosclerosis by causing arterial damage, leading to the accumulation and formation of the plaque and blood clot.

Believe it or not, overexpression of homocysteine accompanied by the low levels of vitamins B6, B12, and folate, also have been found in some patients with renal disease.


Dr. Bradley A. Maron, the lead scientist in the examination of the disease associated with hyperhomocysteinemia wrote, "The unique biochemical profile of homocysteine is characterized by chemical reactivity supporting a wide range of molecular effects, and a tendency to promote oxidant stress-induced cellular toxicity".


And, "Numerous epidemiological reports have established hyperhomocysteinemia as an independent risk factor for cardiovascular disease, cerebrovascular disease, dementia-type disorders, and osteoporosis-associated fractures".


Adiponectin is a type of protein associated with the function that regulates glucose levels and fatty acid breakdown.

The protein also plays a critical role in the development of insulin resistance and atherosclerosis. Low levels of adiponectin levels increase the risk of insulin resistance and type 2 diabetes.

In type 2 diabetes, and administration of adiponectin not only lowers plasma glucose levels but also increases insulin sensitivity.

More precisely, in patients with type 2 diabetes, increased levels of adiponectin involved in the enhancement of insulin sensitivity were attributed to the increase of fatty acid oxidation and inhibition of hepatic glucose production.

Dr. Lihn AS in the examining the effects of administration of adiponectin in type 2 diabetes wrote, "adiponectin in addition to possible anti-inflammatory and anti-atherogenic effects appears to be an insulin enhancer, with potential as a new pharmacologic treatment modality of the metabolic syndrome and type 2 diabetes".

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the inhibition of cardiovascular risk factors, researchers examined the effects of cranberry juice concomitantly on homocysteine and adiponectin levels in patients with the MetS.

The study included a total of 36 individuals with the MetS divided into two groups: the control group (n 36) and cranberry-treated group(n 20) received cranberry juice (0·7 liters/d) containing 0·4mg folic acid for 60 days.

According to tested analysis, the cranberry-treated group showed an increase in adiponectin involved in regulating glucose levels as well as fatty acid breakdown and a decrease in homocysteine associated with a risk for heart disease comapred to baseline values and the controls.

However, the cranberry-treated group improved MetS without affecting the production of pro-inflammatory cytokines TNF-a, IL-1, and IL-6.

According to the measurement of oxidative stress, the cranberry-treated group also showed decreased lipoperoxidation and protein oxidation levels, compared to the control group.

The results strongly suggested that chronic consumption of cranberry juice for 60 d improves some cardiovascular risk factors.

Based on the findings, researchers concluded. "The present data reinforce the importance of the inverse association between homocysteine and adiponectin and the need for more specifically designed studies on MetS patients.".

Taken altogether, cranberry may be considered a remedy for the prevention and treatment of MetS, pending the confirmation of the larger sample size and multicenter human study.
