#Diphenylhydantoin and #Valproicacid Isolated From Chaste Tree Berry Process Anticonvulsant Activity, Researchers Show

Chaste tree berry is a species of Vitex agnus-castus, genus Vitex, belonging to the family Lamiaceae, native to the Mediterranean region, used in herbal medicine for thousands of years as an aphrodisiac herb and considered a Queen herb in treating menstrual problems and discomforts.

In a study using an animal model of maximal electroshock seizures (MES) and induced seizures (MES) by administrated pentylenetetrazole (PTZ), intake of ethanolic leaf extract of Vitex-negundo, at doses of 250, 500, and 1000 mg/kg p.o, exerted a significant of postictal depression, an altered state of consciousness after an epileptic seizure in the dose of 1000 mg/kg body weight in comparison to control but no effects in other applications.

At doses of (1000 mg/kg, PO), the ethanolic leaf extract exhibited a 50% half-life protection in chronic seizures after a 24-hour injection against PTZ-induced seizures. Researchers also found that the efficacy of the extract at that dose is the result of valproic acid, a major ingredient found in Chaste tree berries.

Dr. Tandon VR and co-author Dr. Gupta RK. of the study said, " The anticonvulsant activity of Vitex-negundo has not been found quite effective with standard drugs".


Furthermore, because the potentiation of diphenylhydantoin and valproic acid isolated Vitex-negundo displayed a significant effect in treating animal models, the extract may be used as an adjuvant therapy along with standard anticonvulsants.

Additionally, according to the Baqiyatallah University of Medical Sciences, hydrophilic extract of Vitex agnus castus fruit (Vitex) administered doses (60, 120, or 180 mg/kg) showed a strong anticonvulsant activity even at low doses (60 mg/kg) of Vitex by increasing the afterdischarge(AD) threshold and decrease the afterdischarge duration ( ADD) and stage 5 duration (S5D).


Promisingly, at the highest dose, the extract inhibited significantly the seizure stage (P<0.01) and reduced or prevented epileptic activity as demonstrated by the reduction of ADD and S5D (length of convulsion) in a dose-dependent manner.

More importantly, methanolic extract (ME) of the leaves of V. negundo also showed to process the analgesic properties and potentiated analgesia demonstrated by morphine and pethidine in protection against strychnine and leptazole-induced convulsions.

Taking together, Chaste Tree Berry may be one of the next-generation therapeutic herbal medicines that have a strong effect on the prevention and treatment of seizures as a result of abundant phytochemicals, but intake of the large amount should be taken with care to reduce the risk of toxicity.

Mice selected for the study were divided into nondiabetic and diabetic mice induced by a single injection of streptozotocin (STZ, 200 mg/kg, i.p.) orally administered HemoHIM (100 mg/kg) once a day for 4 weeks.
Diabetic mice with HemoHIM showed improved hyperglycemia and glucose tolerance as well as elevated plasma insulin levels with the preservation of insulin staining in pancreatic β-cells, compared to the diabetic control group.

Furthermore, HemoHIM treatment restored parameters associated with the onset of diabetes including thymus weight, white blood cells, lymphocyte numbers, and splenic lymphocyte populations (CD4(+) T and CD8(+) T), which were reduced in diabetic mice.

Moreover, HemoHIM also animalized the IFN-γ production associated with innate and adaptive immunity in response to Con A stimulation in facilitating mediated T cell proliferation.

Based on the results, researchers said, "HemoHIM may have potential as a glucose-lowering and immunomodulatory agent by enhancing the immune function of pancreatic β-cells in STZ-induced diabetic mice".

Taken altogether, Herbal Angelica is used in combination with other herbal medicines for the treatment of diabetes, pending to larger sample size and multicenter human study.

The green-plant membrane in rats, oral fat tolerance tests, and increased serum triglyceride levels were significantly reduced.


Moreover, fecal total lipid and bile acid volumes were significantly increased in rats that were administered 200 mg/mL green-plant membrane.

Based on the evidence, researchers said, "...green-plant membrane with dietary oil inhibits dietary fat absorption via promotion of bile acid excretion in feces and the effectiveness of eating green-plant membrane, such as green leafy vegetables, with meals".

Taken altogether, the green-plant membrane may be considered a functional food for the prevention of hypertriglyceridemia, pending the confirmation of the larger sample size and multicenter human study.

The liver protective activity of PC-1 was attributed to the activation of the levels of antioxidant enzymes in the host and the improvement of liver histology, blood chemistry parameters.

Moreover, the osmotic fragility test (OFT) also addressed that PC-1 extract inhibited the destruction of red blood cells in carbon tetrachloride intoxicated rats.

Based on the findings, researchers said, "These data support the strategy of in-vitro plant selection for developing efficacious tissue-protective photocomposition".

Cancer is a group of diseases characterized by the cell growth uncontrollably in the tissue of certain organs, due to the alternation of cell DNA.

Scientists concerned about the side effects of chemotherapy for the treatment of cancer examined the effects of I3C and its dimeric product, 3, 3'-diindolylmethane (DIM) anti-proliferative and anticancer activities against human cancer cells.

Application of I3C and its dimeric product, 3, 3'-diindolylmethane (DIM) exerted a strong activity in the inhibition of altered expression and exhibition of the functions of proteins associated with cell cycle arrest.

I3C and its dimeric product, 3, 3'-diindolylmethane (DIM) deactivated the signal transduction pathways in the stimulation of cancer cell growth.

Furthermore, the phytochemicals suppressed the genes associated with cancer cell survival and proliferation and the protein involved in the generation of new blood vessels for the tumor.

Furthermore, the tested bioactive compound also activated the protein associated with the pro-apoptosis against the gene involved in anti-apoptosis of the cancer cells.


Additionally, I3C and its dimeric product, 3, 3'-diindolylmethane (DIM) also overexpressed the gene involved in cell cycle arrest against the progression of cancer.

Dr.Noroozi MK and colleagues wrote at the end of the experiment, "I3C and its dimeric product, 3, 3'-diindolylmethane (DIM), have been widely investigated for their importance against a number of human cancers both in vitro and in vivo".

In the microRNA-21 analysis, researchers found that DIM inhibits the microRNA (miRNAs) expression against colon cancer (CAC) pathogenesis.

MicroRNAs (miRNAs) are a class of small noncoding RNAs of ∼22nt in length involved in the regulation of gene expression including inflammation and angiogenesis.

In mesothelioma, injection of DIM exerted anti-mesothelioma potentials, by lowering the expression of microRNA regulatory activities by suppressing macrophage infiltration and pro-inflammatory cytokines.

In other words, DIM modulated the immune response against the onset and progression of mesothelioma in stimulating the production of oxidative stress due to the overexpression of inflammatory cytokines in the initiation of the disease.

Taken altogether, DIM may be considered a supplement for the prevention and combined with primary medicine for the treatment of cancer, pending the confirmation of a large sample size and multicenter human study.
