#Fennel #Extract Processes Cyto-Protection and Antitumor Activity By Inhibiting the Onset of Oxidativestress, Researchers Reveal

Fennel may have a potential and positive effect that induces cytoprotection and antitumor activity against cancer expression, scientists suggested.

The results of fennel efficacy were reported by several respectable institutes including the National Cancer Institute and the National Institute of Health.

In the study to evaluate the efficacy of fennel seed methanolic extract (FSME) for its antioxidant, cytotoxic, and antitumor activities and for its capacity to serve as a nontoxic radioprotector in Swiss albino mice through injection of FSME, 100 mg/kg intraperitoneally into mice bearing EAC before exposed to three 2-Gy doses of gamma irradiation, researchers found at after 30 days, the application exerted a strong effect at the C50 (mean standard deviation 50% inhibitory concentrations) at a dose of 50 μg/mL for the MCF7 breast cancer cell line and 48 μg/mL for the Hepg-2 liver cancer cell line.

The treatment group also displayed a decreased level of MDA and improved natural antioxidants produced by the tissues of the host compared to the elevation of MDA and significantly reduced levels of catalase and GSH contents.

These results suggested that fennel seed methanolic extract (FSME) inhibited the overproduction of ROS caused by ingestion of doses of gamma irradiation through its antioxidant and enhanced antioxidant production of the mice tissue via the ratio of ROS and Antioxidants.


In other words, the application of the fennel extract may have the potential in

* Reduced oxidative stress and protected mouse cells from damage caused by reactive oxygen species.

* Improved natural antioxidants and the oxidative stability of fatty foods during storage.

* Exhibited an antitumor effect by modulating lipid peroxidation and augmenting the antioxidant defense system in EAC-bearing mice with or without exposure to radiation.

Further illustration of the effect of herbal medicine in treatment of cancers and examine the cytoprotective effects, researchers at the National Institute of Health conducted a comparison of five herbal supplements, designated FB, FM, PP, HF, and FBL101, which contain different combinations of various natural herbs such as licorice, black cohosh, Dong Quai, false unicorn and vitex berry root against mice bearing CWR22R and PC3 prostate cancer xenografts.

According to the results of the experiment,

* All herbal medicine, FB, FM, PP, HF, and FBL101 inhibited PC3 tumor growth by 53%, 75%, 80%, 81%, and 87%, respectively.


* In CWR22R tumors, applications of supplements exerted a similar effect in suppressing the cancer cells.

* All applications did not show inhibitory activity against levels of testosterone compared to untreated mice. However, supplements PP and FBL101 exerted significantly reduced VEGF levels, a potent angiogenic factor with function in stimulated production of new blood vessels to ensure cell proliferation, in PC3 and CWR22R tumors.

* Intratumoral microvessel density was decreased in PC3 tumors treated with all five supplements but only in CWR22R tumors treated with HF.

Dr. Ng SS, the lead author said, "Herbal supplements containing the aforementioned extracts inhibit the growth of prostate tumor xenografts, possibly in part by antiangiogenic mechanisms".


The above differentiation was supported by Quaid-i-Azam University in the analysis of the antibacterial potential of some medicinal plants against multi-drug resistant (MDR) pathogens and anticancer effect against the MCF-7 cell line.

Oxidative stress is a condition caused by an imbalance of the ratio of free radicals and antioxidants in the body.
In other words, oxidative stress is either caused by overexpression of free radicals or depletion of antioxidant enzymes in the body.

Some researchers suggested that oxidative stress may also arise due to the chain reaction of free radicals in which free radicals donate or intercept an electron from other stable molecules.

Long-term oxidative stress present in the body can cause protein and lipid damage and alternation of cell DNA, leading to the onset of chronic diseases, including cardiovascular disease, diabetes, and cancer.

More precisely, exogenous factors, such as cigarette smoke, and endogenous factors, such as the oxidative burst from activated macrophages are some of the factors involved in the risk of oxidative stress.

The Liver Pathophysiology, 2017 in the explanation of the cause of oxidative stress, wrote, " (1) the presence of xenobiotics, (2) the activation of the immune system in response to invading microorganisms (inflammation), and (3) radiation, which makes oxidative stress a common denominator of toxicity or stress".

In other words, oxidative stress may be preventable by following a diet rich in antioxidants such as a diet with higher fruit and vegetable consumption accompanied by moderate exercise and reduced intake of saturated and trans fat and processed foods.

Betanin is phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

With an aim to find a potential compound for the prevention of diseases associated with oxidative stress, researchers examined the effects of natural pigment betanin on oxidative stress in the kidney of paraquat-treated rats.

Paraquat injected intraperitoneally induced renal damage rats were randomly divided into four groups: a control group, a paraquat group, and two paraquat groups that were treated with betanin at 25 and 100 mg/kg/d three days before and two days after paraquat administration.

Betanin-treated rats showed strong activity in alleviating the paraquat-incurred acute kidney injury, observed by the improvement, and reduced serum and urine markers that indicate a kidney injury.

Betanin also decreased oxidative stress elicited by paraquat by increasing the expression of antioxidant enzymes produced by the host accompanied by inhibiting the levels of free radicals.

These results suggested that betanin protects the tested rats against kidney damage due to oxidative stress in the induction of chronic inflammation.

In streptozotocin (STZ) induced diabetes (DN) in rats, researchers examined the betanin anti-fibrotic mechanisms in rat kidneys.

Betanin at a dose of 25, 50 and 100mg/kg/day and lisinopril (5mg/kg/day, reference compound) orally administered for 8 weeks after the induction of DN, not only significantly inhibited proteinuria, blood glucose, serum creatinine and blood urea nitrogen (BUN) levels involved in kidney injury, but also restored antioxidant enzyme activities in kidney tissue.

Additionally, betanin treatment also protects kidney integrity by reducing the glomerular surface area, glomerulosclerosis, and tubulointerstitial fibrosis.

In other words, betanin demonstrated the kidney protective activity against streptozotocin (STZ) induced diabetes (DN) in rats through regulation of transforming growth factor beta (TGF-β) in facilitating anti-oxidative and anti-inflammatory activity.

Taken altogether, betanin may be considered a supplement for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of a larger sample size and multicenter human study.

Intake of betanin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Diabetes is a medical condition characterized by the failure of the body to utilize the insulin in converting glucose to energy, leading to high levels of glucose in the bloodstream.

Most cases of diabetes are associated with either beta cells of the pancreas died-off or insulin receptor site clogged-up by fat and cholesterol.

Diabetic complications in most cases are caused by long-term unmanaged hyperglycemia, a hallmark of diabetes.

Hyperglycemia occurs in diabetics due to uncontrolled diet, skip diabetes medication and improper use of insulin injection.

Moreover, the expression of glycation of SOD also leads to the formation of AGEs and fibrils.

In other words, alliin offered protection against elevated levels of glucose or MG-induced glycation of SOD.

Additionally, alliin efficacy against the onset of hyperglycemia was comparable with that of quercetin which is reported to be a potent natural inhibitor of glycation.


Based on the findings, researchers wrote, "Alliin has a good antiglycation effect and hence is expected to have therapeutic potential in the prevention of glycation-mediated diabetic complications".

Compared to the chemo drug doxorubicin (adriamycin), betanin, the major betacyanin constituent has been shown to exhibit a dose-dependent cytotoxic effect on human prostate cancer cells (PC-3) and in the well-established estrogen receptor-positive human breast cancer cells (MCF-7), comparable to those of doxorubicin.

In time analysis, although the cytotoxicity of the beetroot extract was significantly lower compared to doxorubicin which completely inhibits the growth of the PC-3 cells in three days, the extract continued to decrease the growth rate of the PC-3 cells (3.7% in 3 days vs. 12.5% in 7 days) at the concentration of 29 µg/ml.

Furthermore, in the normal human skin FC and liver HC cell lines, the extract showed a significantly lower cytotoxic effect compared to doxorubicin (8.6% vs. 100%, respectively, at 29 µg/ml concentration of each, three-day test period).


In chemical differentiation, the efficacy of beetroot extract was attributed to the chemical compound betanin, the major betacyanin constituent in exerting the cytotoxicity.

Based on the finding, researchers said, "Further studies are needed to evaluate the chemopreventive potentials of the beetroot extract when used alone or in combination with doxorubicin to mitigate the toxic side-effects of the latter".

Taken altogether, betacyanins processed as the major bioactive ingredient betanin may be considered a remedy for the prevention and treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.

According to the tested analysis, compared to a regular diet, the group treated with a Japanese diet (high-sodium/low-potassium) showed induced increases in oxidative stress, inflammation, and proteinuria, along with glomerular and tubular damage.

The (AMPK/UCP2) pathway associated with the inhibition of ROS production and palmitate-induced apoptosis in endothelial cells was significantly suppressed by the Japanese diet.

However, Japanese diet-induced overexpression of oxidative stress accumulation, inflammation, renal damage, and proteinuria was totally prevented by the administration of Brussels sprouts.

Additionally, injection of Brussels sprouts also promoted the expression of antioxidant enzymes produced by the host observed by the increased level of a gene associated with drug toxicity without affecting the systolic blood pressure (SBP).


Taken together, Brussels sprouts may be considered supplements for the prevention of renal damage progression in hypertension, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Scientists may have found a natural alternative for the treatment of bad breath, according to studies. Bad breath also known as halitosis, is a condition caused by an unpleasant odor of the mouth.

The causes of bad breath may result from a combination of food, tobacco products, poor dental hygiene, health problems, dry mouth, oral infections, dental problems, or medications.

However, chemicals produced by some diseases, such as some cancers, and metabolic disorders, can cause a distinctive breath odor.

Chronic reflux of stomach acids (gastroesophageal reflux disease, or GERD) may be considered a major cause of bad breath in older adults.

The most common treatment for bad breath in the Western world is the use of mouth rinses and toothpaste if the bad breath is the result of bacterial infection.


People with bad breath due to loss dentures may need to see their dentists for correction.

However, if bad breath is associated with dental diseases such as gum disease then seeing a related field specialist may be necessary.

Some researchers suggested that obesity is also tied to bad breath. The Tel Aviv University in examining the risk of bad breath in obese wrote, " the more overweight you are, the more likely your breath will smell unpleasant to those around you".

Honey is a miraculous product made by bees using nectar from flowers.

The rich golden liquid is considered one of the healthy sweet foods for replacing the use of white sugar and artificial sweeteners by many people.

Researchers with an aim to find a natural remedy for the treatment of bad breath examined the Manuka honey anti-oral halitosis activity.

Manuka honey contains the typical antibacterial substance methylglyoxal (MGO) which has been found to process a strong antibacterial activity against oral bacteria.

The study to compare the before and after intake of Manuka honey or Acacia honey and their halitosis effect included 10 healthy subjects (average age: 36 years old, 5 men and 5 women).


The concentrations of volatile sulfur compounds (VSCs) produced by the oral bacteria and anaerobic bacteria in the mouth decreased after intake of both honey, but faster in the Manuka honey compared to those of Acacia honey.

Manuka honey showed a 127 times higher amount of MGO than acacia honey, according to analysis.

These results clearly suggested that the efficacy of honey in the inhibition of bad breath was attributed to the strong antibacterial activity of MGO.

In order to reveal more information about honey's anti-bad breath activity, researchers investigated the effect of honey in preventing gingivitis and dental caries.

In a total of 20 female orthodontic patients between the ages from12–and 18 participated in this randomized controlled study, Researchers compared the effect of honey with either 10% sucrose or 10% sorbitol that served as positive and negative controls.

Honey injection showed significant differences in pH compared to the pH observed in the sorbitol group. The maximum pH drop occurred at 5 min in both the honey and sucrose groups and the pH following sucrose exposure fell <5.5 and was associated with a 30 min recovery time.

Furthermore, bacterial counts were in the honey treatment group significantly reduced compared to the other treatment groups.


Moreover, honey also significantly inhibited the growth of all studied strains compared to the inhibition observed with antibiotics.

Taken together, honey, particularly, Manuka honey or Acacia honey may be considered functional remedies for the prevention and treatment of bad breath, pending the confirmation of a larger sample size and multicenter human study.

#Tomato Balances Gut Microbiota For the Improvement of #DigestiveHealth, Researchers Show

Tomato is a red, edible fruit, genus Solanum, belonging to the family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

In the urgency to discover healthy foods for enhancing the ratio of dut microbiota composition researchers examined the effect of consuming tomato products and lycopene on chronic inflammatory disorders associated with intestinal bacteria in an animal model.

The study included Winnie mice as a model of spontaneous ulcerative colitis treated with or without chow enriched with 1% Bronze tomato.

At week 2, the Bronze-enriched chow-treated group showed no macroscopic effect on the IBD symptoms.

However, levels of pro-inflammatory cytokines found in the treated mice were significantly reduced, as observed by the levels of IL17A and IFNγ.


According to the analysis of the microbial composition, two weeks of Bronze enriched diet demonstrated a strong effect in the improvement of the gut microbiota ratio in the tested mice.


In additional differentiation of the results, researchers found that the efficacy of Bronze tomato in the promotion of gut health was attributed to the phytochemical polyphenols presented in the tomato.

Taken altogether, tomatoes processed with a high amount of lycopene may be considered a dietary supplement for gut health, pending to large sample size and multicenter human study.

Intake of polyphenols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
