#Herbal #Turmeric Reduces Inflammatory Cell Infiltration of Myocardial Arteries Associated with the Risk of Chagas Heart Disease

Kyle J. Norton

Chagas disease (CD) is a medical condition caused by the protozoan parasite Trypanosoma cruzi.
According to the statistics, approximately 6 million people worldwide are living with Chagas disease. The majority of patients reside in Mexico, Central America, and South America,

Chagas heart disease can be classified into
* Acute Chagas heart disease is characterized by the inflammation of the heart, gastrointestinal tract (mainly esophagus and colon), meninges, and central and peripheral nervous tissues. Untreated symptomatic patients in this phase may face a roughly 1% chance of death in the acute period, usually the result of severe myocarditis or meningoencephalitis

* The indeterminate form of Chagas heart disease is characterized by small foci of mild chronic myocarditis, with interstitial edema, early fibrosis, and infiltration by lymphocytes, macrophages, and plasma cells. Most patients with the indeterminate form will progress to the chronic form of Chagas heart disease such as dilated cardiomyopathy, at a rate of 1.85% to 7% annually.

* The chronic form of Chagas heart disease is characterized by cardiomegaly and increased cardiac mass, thinning of the ventricular walls, and aneurysmal lesions in various regions of the LV and right ventricle (RV).

Dr. Edimar Alcides Bocchi, the lead scientist in the treatment of Chagas heart disease wrote, "Pharmacological treatment and antiarrhythmic therapy for ChHD is mostly based on results for other etiologies. Heart transplantation is an established, valuable therapeutic option in refractory ChHD".

And, "Implantable cardioverter-defibrillators are indicated for the prevention of secondary sudden cardiac death. Specific etiological treatments should be revisited and reserved for select patients. Understanding and management of ChHD need improvement, including the development of randomized trials".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an antioxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

With an aim to find a potential compound for the treatment of Chagas heart disease, caused by the protozoan parasite Trypanosoma cruzi., researchers examined the anti-inflammatory and cardioprotective properties of curcumin (Cur).

The study included oral therapy with Cur on T. cruzi-mediated cardio-vasculopathy in acutely infected mice and in vitro, the response of parasite-infected human microvascular endothelial cells treated with this phytochemical.

According to the tested assays, oral Cur therapy showed a reduced inflammatory cell infiltration of myocardial arteries without lowering parasite levels in the tested infected mice.

Furthermore, mice treated with Cur showed significantly decreased vessel inflammation scores (p < 0.001), and vascular permeabilities compared to mice treated with the phosphate-buffered saline-receiving group.

Moreover, levels of pro-inflammatory cytokines and ET-1 secretion involved in vascular constriction were also inhibited by the treatment of Cur.

Additionally, proteins and signaling pathways that promote the generation of inflammatory agents are also inhibited by the injection of Cur.

In other words, curcumin (Cur) exerted the anti-inflammatory and cardioprotective properties against parasite-induced the onset of Chagas heart disease.

Based on the findings researchers wrote, "These findings identify new perspectives for exploring the potential of Cur-based interventions to ameliorate Chagas heart disease".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of Chagas heart disease, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of a supplement should be taken with extreme care to prevent overdose and acute liver toxicity.

#Ursolicacid Found Inhibits Proteins and Pathways Associated with the Onset of #EndometrialCancer, According to Studies

Kyle J. Norton

Arctostaphylos uva-ursi is one of the species of Bearberry, in the genus Arctostaphylos, belonging to the family Ericaceae, native to northern North America, Asia, and Europe. The herb has been used in traditional medicine as a diuretic agent and to treat urinary problems, including bladder infections, kidney infections, kidney stones, pelvic pain, etc.

The endometrium is the inner lining of the mammalian uterus and is very susceptible to hormone change, particularly during the menstrual cycle. Endometrial cancer is a late adulthood cancer defined as a condition in which the cells of the endometrial lining of the uterus have grown uncontrollably or become cancerous as a result of the alternation of cells' DNA. It's the fourth most common cancer among women overall, after breast cancer, lung cancer, and bowel cancer.

The Ingredients It contains arbutin, ursolic acid, tannic acid, gallic acid, some essential oil and resin, hydroquinones tannins, phenolic glycosides, and flavonoids.

Ursolic acid, a pentacyclic triterpene acid, extracted from bearberry is found to be effective in inhibiting the growth of endometrial cancer cell HEC108 line, in a dose- and time-dependent manner. As exposure to Ursolic acid decreased the anti-apoptotic Bcl-2 protein levels (member of the Bcl-2 family of regulator proteins that regulate cell death) but increased Bax expression (upregulated by the tumor suppressor protein p53,) through a mitochondrial pathway, that induced apoptosis of endometrial cancer HEC108 line. In the study of the same but within endometrial cancer SNG-II cells, ursolic acid strongly inhibited the growth of SNG-II cells in a dose- and time-dependent manner. The induced apoptotic processes in the endometrial cancer SNG-II cells also involve mitochondrial pathways and Bcl-2 family proteins.

#Chastetreeberry Fraction Inhibits Chemical #Nociception Compared to Those of #Naloxone, Researchers Suggest

Kyle J. Norton

A recent study suggested that essential oil isolated from Chaste tree berries may have a potential effect in reduced neuro-pain through antinociceptive function in tested subjects.

The antinociceptive effect is the activity of reduced pain through interference with information transmitted and received by brain neurons.

Chaste tree berry is a species of Vitex agnus-castus, genus Vitex, belonging to the family Lamiaceae, native to the Mediterranean region, used in herbal medicine for thousands of years as an aphrodisiac herb and considered a Queen herb in treating menstrual problems and discomforts.

In the study of three behavioral models of nociception in adult male Wistar rats conducted by the University of Tabriz, the topical essential oil extracted from Vitex Agnus-cactus (EOVAC) was applied to animals expressed a reduced pain response in tail immersion tests, inhibited the acetic acid-induced writhing response with little adverse effects.

Dr. Khalilzadeh E, the lead author said, " major components of the EOVAC were α-pinene (14.83%), limonene (10.29%), β-caryophyllene (6.9%), sabinene (5.27%), and β-farnesene (5.9%), (may have direct influence to the) endogenous opioidergic system, as well as muscarinergic receptors of the cholinergic system, may be involved in the antinociceptive activity of Vitex agnus-castus essential oil in these models of pain in rats".

Interestingly, the extract of herbal medicine also displayed a strong effect against chronic inflammation in induced pain.

The joint study led by the Federal University of Santa Maria suggested that crude extract caused antinociceptive/antidepressant-like effects in the CFA-induced chronic inflammation model incomparable to naloxone, without altered inflammatory parameters at doses of 10mg/kg,

This result indicated that The VmE extract demonstrated pain relief by inducing opioid-dependent antinociception and antidepressant-like effects, without anti-inflammatory activity.

Furthermore, in the investigated petroleum ether fraction (PEF) from the aqueous ethanol extract of Chaste tree berry in several nociceptive mouse models, mouses treated with the fraction produced significant dose-related inhibitions on chemical nociception incomparable to Naloxone (1 mg/kg BW subcutaneously), a non-selected opioid receptor antagonist.

In the xylene-induced ear edema test, the fraction also exerted strong analgesic effects partially mediated by its anti-inflammatory activity.

Taking together, Chaste tree berries may have a therapeutic potential to be used as an antinociceptive functional food in reduced pain by blocking the information transfer by neurotransmitters.

#Herbal Turmeric's #Curcumin Capsules Inhibit Parameters Associated with Toxicity in β-thalassemia, Researchers Say

Kyle J. Norton

Anemia is a health condition in which our body do not have enough red blood cells or less than the normal quantity of hemoglobin in the blood that carries the function of delivering oxygen and nutrient to the tissues.

Patients with Beta-thalassemia minor have a slight lowering of the beta hemoglobin level in the blood similar to those with mild iron-deficiency anemia.

Beta thalassemia can be classified into
* Thalassemia minor, a mild case of anemia. is the most common anemia in people of Mediterranean descent. The blood cells of patients with thalassemia minor are smaller than normal blood cells and easily die off, causing not enough new blood cells to replace the loss of red blood cells.

* Thalassemia major
Thalassemia major also called Cooley's anemia is an inherited disorder that affects the production of normal hemoglobin. Thalassemia major is a deadly disease without frequent blood transfusions, people with this kind of anemia may not survive because of iron building up in the heart and other organs, resulting in heart failure.

The most common symptoms of beta thalassemia are fatigue, weakness, or shortness of breath, a pale appearance or yellow color to the skin (jaundice), irritability, deformities of the facial bone, slow growth, a swollen abdomen, and dark urine.

If you have experienced persistently the aforementioned symptoms, please make sure that you check with your doctor for preventive measures.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

In finding a natural treatment for anemia, researchers examined the effects of curcumin, the active polyphenol in turmeric, on iron overload, hepcidin level, and liver function in β-thalassemia major patients.

The double-blind randomized controlled clinical trial was conducted on 68 β-thalassemia major patients, randomly divided into 2 groups to receive either 500 mg curcumin capsules (total: 1,000 mg) twice daily or a placebo for 12 weeks.

Based on the observation of the tested assays, the curcumin group showed a significant decrease in levels of nontransferrin bound iron (NTBI), aminotransferase (ALT), and Aspartate aminotransferase (AST) compared to the placebo group.

In other words, turmeric protects the integrity of the red blood by reducing cellular oxidative stress and inhibiting iron toxicity in β-thalassemia major patients without affecting the levels of hepcidin and other variables.

Based on the results, researchers wrote, " Curcumin administration alleviated the iron burden and liver dysfunction by reducing NTBI, ALT, and AST levels in patients with β-thalassemia major".

Additionally, according to the evaluation of the Mahidol University on 10 β-thalassemia/Hb E patients treated with 500 mg curcuminoids daily for 12 months, injection of the supplement showed significantly decreased intensity in thalassemic plasma, compared to those of normal subjects, including increased levels of plasma levels of proteins associated with the condition and reduced levels of oxidative damage.

More precisely, serum non-transferrin-bound iron associated with iron overload, platelet factor-3-like activity, oxidative stress parameters, and antioxidant enzymes were also improved after curcuminoid treatment.

Taken altogether, turmeric-processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of β-thalassemia, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of a supplement should be taken with extreme care to prevent overdose and acute liver toxicity.

#Antioxidants Found in #CabbageExtract Inhibit the Onset of Oxidative Stress, According to Studies

Kyle J. Norton

Oxidative stress is a condition caused by an imbalance of the ratio of free radicals and antioxidants in the body. In other words, oxidative stress is either caused by overexpression of free radicals or depletion of antioxidant enzymes in the body.

Some researchers suggested that oxidative stress may also arise due to the chain reaction of free radicals in which free radicals donate or intercept an electron from other stable molecules.

Long-term oxidative stress in the body can cause protein and lipid damage and alternation of cell DNA, leading to chronic diseases, including cardiovascular disease, diabetes, and cancer.

More precisely, exogenous factors, such as cigarette smoke, and endogenous factors, such as the oxidative burst from activated macrophages are some of the factors involved in the risk of oxidative stress.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.

Due to its nature to become stable, free radicals readily donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibit the onset of free radicals before they can cause a chain reaction in facilitating protein, lipid, and cell damage.

Antioxidants can either be produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

Cabbage is a species of Brassica oleracea belonging to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem with a crowded mass of leaves, usually green but in some varieties, it may be red or purplish.

In findings of a natural whole for preventing and treating diseases associated with oxidative stress, researchers investigated the effects of red cabbage anthocyanin (RCA)-enriched extract on H₂O₂ expression on HepG2 cells.

According to chemical analysis, all derivatives of cyanidin (268 ± 2 μg/mg) found in RCA, mainly with 19% nonacylated, 51% monoacylated, and 31% diacylated structures with ferulic, sinapic, p-coumaric, and caffeic acids exerted antioxidant activity against the oxidative stress induced by the injection of H₂O₂.

Furthermore, the application of RCA extracts markedly ameliorated cell apoptosis and improved viability.

Moreover, in another study, cabbage extract protected against H2O2-induced cell death and did not elicit any cytotoxic effects by suppressing ROS production and increasing the expression of antioxidant proteins (SOD-1, catalase, and GPx) produced by the host.

In oxidative stress-exposed H9c2 cells, the protective effects of cabbage extract also inhibited apoptotic responses and activation of MAPK proteins (ERK1/2, JNK, and p-38) involved in the apoptosis resistance by preserving mitochondrial functions.

Based on the results, researchers wrote, "Cabbage extract protects against oxidative stress and suggests that it can be used as an alternative therapeutic strategy to prevent the oxidative stress in the heart".

Taken altogether, cabbage may be used as a functional food for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

#Herbal #Turmeric #Extract Inhibits Parameters Associated with the Acute and Chronic Vascular Inflammation, Researchers Reveal

Kyle J. Norton

Inflammation is a natural reaction of the immune system's self-protection to remove harmful stimuli, including damaged cells, irritants, pathogens... from our body.

Acute inflammatory inflammation is a result of the over-response of the immune system after sensing the injury or damage to the body by activating the protein associated with the production of pro-inflammatory cytokines.

The aim of the proinflammatory cytokines is to kill the invasive pathogens, which try to enter the body through the wound.

In the acute phase of infection, the immune system in the stimulating production of proinflammatory cytokines may induce overproduction of these cytokines can cause death or damage to the nearby cells, leading to scar formation and symptoms of localized swelling, redness, and fever.

Chronic inflammation is a serious condition caused by the immune system to adapt to the new change due to its inability to kill all the invasions within a set of times.

Vasoprotection is the process of improving the function of the blood vessels, including the use of herbal and natural remedies.

Blood vessels are the tubular structure including a vein, artery, or capillary which carries the blood through the tissues and organs pumped by the heart.

The 3 main types of blood vessels are arteries that deliver oxygen-rich blood from the heart to the tissues and organs of the body. Damage of arteries may induce coronary artery disease associated with the narrowing or blockage of the coronary arteries due to plaques accumulated in the arterial wall.

The veins that carry blood toward the heart. Chronic venous insufficiency (CVI) is a common condition associated with a disease of the veins that affects up to 20 percent of adults.

The capillaries or the smallest blood vessels that have been found to induce systemic capillary leak syndrome (SCLS) is a rare acquired disorder.

Vascular inflammation is a condition associated with the inflammation of the blood vessels caused by the overexpression of pro-inflammatory cytokines involved in vascular inflammation. Atherosclerosis is characterized by chronic inflammation of the vascular wall.

Chronic vascular inflammation has been found to cause historical changes in the blood vessel walls, leading to blood vessel wall thickening, weakening, narrowing or scarring on orchestrating blood flow restriction, resulting in organ and tissue damage.

The most common symptoms of vascular inflammation are associated with the reduced blood flow to supply nutrients and fluid to the body tissues, including fever, headache, fatigue, general aches, and pains, night sweats, rash, numbness, and weakness.

In findings, a potential compound for the treatment of acute vascular inflammation researchers evaluated the anti-inflammatory activity, and the cytotoxic effect of a novel chemically modified, non-carbonyl compound enriched Curcuma longa L. (C. longa) extract (CMCE).

According to the tested analysis, CMCE at the concentration of 1 or 10 μg/mL at 14 h, significantly decreased LPS (50-100 ng/mL) induced production of pro-inflammatory cytokines including TLR4 expression in THP-1 cells, human, and mouse whole blood.

In THP-1 cells, CMCE inhibited the expression of IRAK1, which plays a critical role in initiating an innate immune response against foreign pathogens, MAPK is involved in directing cellular responses to a diverse array of stimuli.

CMCE (30, 100, and 300 mg/kg; 10 days p.o.) pre-treated and LPS (10 mg/kg) in Swiss mice exhibited attenuated plasma TNF-α, IL-1β, nitrite, aortic iNOS expression, and vascular dysfunction associated with the production of pro-inflammatory cytokines.
In HL-60 cells., CMCE. exerted cytotoxic effects through cell cycle arrest on the Sub-G1 phase, increased apoptotic cells, loss of mitochondrial membrane in the cancer cells, and activation of cascade associated with pro-apoptosis.

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of vascular inflammation, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Cranberry #Extract Disrupts Biofilm Structural Architecture That Causes Candida Albicans, Researchers Say

Kyle J. Norton

The fungus is a class of primitive organisms, including mushrooms, mold, and mildew, found in the air, in the soil, on plants, and in water.

Fungi reproduce through tiny spores in the air if you inhale the spores, it may spread to cause fungal infection, often starting in the lungs or on the skin.

Similar to other bacteria, there are millions of different fungal species on Earth, but only about 300 are known to infect people.

Conventionally, fungal infection is treated with antibiotics, including topical antifungal ointments or oral medications, depending on the severity of the disease.

Candida is a class of yeasts that cause fungal infections worldwide.

Candida albicans is a member of a large group of an organism whose cells contain complex structures enclosed within the membranes, including yeast and mold that live among the gut flora in the human mouth and gastrointestinal tract.

Under normal circumstances, Candida albicans do not cause harm to humans, but overgrowth results in candidiasis.

Oral candidiasis is an oral condition infected by the fungal C. Albicans found mostly in people with a weakened immune system including patients who are taking the immune-suppressive medicine.

Biofilms are microbial communities in which cells stick to each other and often also to a surface.

Symptoms of oral Candida overgrowth are worse on a damp day, including persistent drowsiness, lack of coordination, headaches, mood swings, loss of balance, rashes, bad breath, dry mouth, and postnasal drip.

There are many risk factors associated with Candida overgrowth including medical conditions that cause a weakened immune system, inhalant medicine, low levels of stomach acids, and pregnancy.

Some researchers suggested aging may be one of the major risk factors that cause Candida overgrowth

Dr. Aikaterini Flevari, the lead scientist said, "Candida species remain the most important cause of opportunistic infections worldwide, affecting predominantly patients over 65 years old, while they are considered to be the fourth most common cause of nosocomial bloodstream infections".

And, "The rapidly growing elderly population has specific physiological characteristics, which makes it susceptible to colonization and subsequent infection due to Candida species"

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of Candida albicans, researchers examined the effects of polyphenol-rich cranberry extracts on dual-species Streptococcus mutans-Candida. albicans biofilms.

On S. mutans-C. albicans biofilms grown on saliva-coated hydroxyapatite discs (s-HA) mounted on the high-throughput Amsterdam Active Attachment model, cranberry extracts(ACE)/vehicle control was applied at the five minutes just before biofilm growth and subsequently, for similar exposure times, after 12 hours and 24 hours of biofilm growth.

Based on the analysis, treatment of ACE at doses of 500 to 1,000 μg/mL showed significant reductions in acidogenicity and metabolic activity, compared to the control.

Biovolumes of the EPS (P=0.003) and microbial biofilm components associated with the overgrowth of Candida albicans were significantly decreased by the application of ACE.

According to the qualitative assessment of confocal biofilm images, cranberry extract disrupted biofilm structural architecture that causes Candida albicans.

Moreover, ACE showed significantly fewer S. mutans (P=0.006) and C. Albicans (P=0.036) compared to the control.

Based on the findings, researchers aid, "Cranberry extracts inhibited cariogenic virulence properties of S. mutans-C. albicansdual-species biofilms in an in vitro model".

Taken together, cranberry may be considered a remedy for the treatment of Candida albicans,, pending the confirmation of the larger sample size and multicenter human study.
