#Phytochemical 3,3'-Diindolylmethane In the Promotion of Enzymes and Antioxidant Activity Associated with Anti- Hematopoietic Injury, Researchers Find

Scientists may have found a bioactive ingredient for the protection of hematopoiesis with no side effects, according to studies.
Hematopoiesis is the process by which blood cells are formed, including the formation of all of the cellular components of blood and blood plasma.
1. Red blood cells with a function to transport oxygen and hemoglobin to the body.
2. White blood cells that form parts of the immune system, including
* T cells and B cells, that fight viruses, and tumors.
* Neutrophils that fight bacteria and fungi.
* Eosinophils that fight some parasites and initiate an inflammatory response.
* Basophils that release the histamines necessary for the inflammatory response.
* Macrophages, the first line of the immune response in the acute phase of infection.
3. Platelets (thrombocytes) for coagulation.
Diseases of the bone marrow, spleen, lymph nodes, and thymus may affect the process of hematopoiesis, leading to various diseases of the hematopoietic system.
3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, kale, etc.
On finding a natural compound for the treatment of hematopoietic injury, researchers examined the 3,3'-Diindolylmethane (DIM) effect on the hematopoietic system injury induced by total body irradiation (TBI) that leads to the generation of intracellular reactive oxygen species (ROS) and DNA damage.
In TBI-induced lethality mice, DIM ameliorated TBI-induced hematopoietic injury by inhibiting the production of ROS production in hematopoietic stem cells (HSCs).
Furthermore, DIM improved the bone marrow (BM) HSC frequency, hematopoietic progenitor cell (HPC) clonogenic function, and multilineage engraftment after transplantation in tested mice.
In an additional analysis of DIM anti-hematopoietic injury, researchers found that DIM promotes the genes associated with the increased production of antioxidant enzymes (nuclear factor erythroid 2-related factor 2 (Nrf2)) in the hematopoietic stem cells (HSCs) and decreased the production of free radicals (NADPH oxidase 4 (NOX4)).
DIM also protected the cells against apoptosis by increasing the expression of the anti-apoptotic protein (Bcl-2) and decreasing the expression of the pro-apoptotic protein (Bax).
The results strongly suggested that DIM attenuates TBI-induced hematopoietic injury through the inhibition of both oxidative stress in HSCs and hematopoietic cell apoptosis.
Dr. Lu L and colleagues wrote in the final report, "DIM protected BM hematopoietic cells against ionizing radiation and led to increased clonogenicity in vitro. Therefore, DIM has the potential to be used as an effective radioprotectant to ameliorate TBI-induced hematopoietic injury".
Taken together, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the protection and treatment of hematopoietic injury, pending the confirmation of the larger sample size and multicenter human study.

Additionally, in an animal model of dextran sodium sulfate (DSS)-induced colitis, a specific, curcumin preparation containing essential turmeric oils (ETO-curcumin) compared to standard curcumin at three specific doses (0, 5, 25, or 50 mg/kg), both ETO and standard curcumin treatments provided protection against DSS-induced inflammation.
However, according to the tested assays, ETO-curcumin improved disease activity index (DAI) dose-dependently, compared to the anti-inflammatory efficacy of standard curcumin remained constant.
In other words, ETO-curcumin was superior in the expression of anti-inflammatory efficacy compared to standard curcumin.
Moreover, ETO-curcumin displayed anti-colitis activity in the colon by exerting anti-inflammatory cytokines including IL-10 and IL-11 as well as FOXP3.
Collectively, Dr. Toden S., the lead scientist, wrote in the final report, " The combined treatment of curcumin and essential turmeric oils provides superior protection from DSS-induced colitis than curcumin alone, highlighting the anti-inflammatory potential of turmeric".
Taken altogether, turmeric plus essential turmeric oils may be considered supplements for the prevention of colitis, pending the confirmation of the larger sample size and multicenter human study.
Intake of turmeric in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

There are many causes associated with the onset of depression, including genetics, changes in hormone levels, certain medical conditions, stress, grief, or difficult life circumstances that initiate the change of brain chemistry in inducing depressive syndrome.
The most common symptoms of depression include sadness, hopelessness, guilt, change of mood, and loss of interest in friends, family, favorite activities, and sex.
Some patients with depression also experience cognitive symptoms of trouble concentrating, making decisions, remembering, and thoughts of harming themselves.

8 studies that satisfied the standard criteria and guidelines were chosen, including 2 RCTs of human subjects and 6 animal studies.
The randomized studies suggested that depressed elderly treated with a mix of honey and barely exhibited a significant decrease in depression, stress, and mood disturbances scores compared to the control group.
In menopausal women, treatment with honey or a mix of honey and pollens exerted a significant activity in the improvement of the three subscales of the Menopause Rating Scale (MRS) by 68.3% and 70.9%, respectively.
The results also suggested that posttreatment demonstrates a better effect compared to pretreatment when the mixture of honey was applied.
Furthermore, the application showed similar effects on the serum levels of cholesterol, such as total, HDL, or LDL cholesterol), triglycerides, or oestradiol in both groups.
According to the survey returned, participants reported that they observed improvements 3-4 days after the initiation of treatment. On a 10-point taste scale, (1 = very good and 10 = very bad) honey received an average score of 2.3, whereas a honey and pollen mix received an average score of 6.0.

Cognitive deficit is a medical condition associated with the impairment of a person's mental processes and the ability to acquire information and knowledge that interact with the outside world.

Based on the behavioral tests, D-galactose (D-gal)-induced neurotoxicity in mice treated with betacyanins showed a strong effect on the improvement of learning and memory impairments.
Additionally, the levels of brain antioxidant levels such as superoxide dismutases (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) in D-gal-treated mice were significantly increased by the injection of betacyanins.
Furthermore, lipid peroxidation product found in the brain tissues and neurons such as malondialdehyde (MDA) was decreased by betacyanin administration.
Moreover, levels of the activities of SOD, CAT GR, and GPx in the mouse brain, had a significant negative correlation compared to the positive correlation of the level of MDA with the mouse cognitive impairment.
In other words, oxidative stress caused by D-galactose (D-gal)-induced neurotoxicity induced low levels of antioxidants enzymes such as SOD, CAT GR and GPx in the mouse brain were restored by the injection of betacyanins.
Based on the finding, researchers wrote in the final report, "These results suggest that the neuroprotective effect of betacyanins against D-gal-induced neurotoxicity might be caused, at least in part, by an increase in the activities of antioxidant enzymes with a reduction in lipid peroxidation" and "the betacyanins had a more pronounced effect on ameliorating cognition deficits in mice".
Taken together, betacyanins may be considered a remedy for the prevention and treatment of cognition deficits, pending to the confirmation of the larger sample size and multicenter human study.
Intake of betacyanins in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
