#Chronic Injection of Dietary Phenylethyl ITCs Isolated From #CruciferousVegetables Inhibit #Cancer Cell Colony-Forming Capability and Induced Apoptosis, According to Studies

Kyle J. Norton

The liver is one of the organs with a lack of nerve tissue, therefore liver cancer does not induce pain until the later stage.
Liver cancer is a medical condition characterized by cell growth irregularly in the liver tissue.

The causes of liver cancer are associated with the alternation of cells' DNA that normally begins in the cells on the surface of the inner lining.

Liver cancer at the early stage no not cause any symptoms. However, at the advanced stage, liver cancer cells can cause a wide range of symptoms including jaundice, abdominal swelling and pain, and dark urine and white stool accompanied by the general symptoms of cancers such as unintended weight loss, loss of appetite, chronic fatigue and tiredness and gastrointestinal discomforts.

The most common cause of liver cancer is infected by the hepatitis virus. However, in North America, due to the overexpression of the overweight and obese population, liver cancer may slowly progress from nonalcoholic fatty liver disease through cirrhosis to liver cancer.

There are a number of risk factors other than those mentioned above associated with the onset of liver cancer such as male, ethnicity, and hepatitis viral carriers.

According to the statistics provided by the Canadian Cancer Society, every year, approximately, 1,900 men and 580 women are diagnosed with liver cancer, causing the death of 950 men and 270 women.

In 2017, 2,500 Canadians were diagnosed with liver cancer and 1,200 Canadians died from liver cancer.

Compared to the early stage of other cancer, the 5 years survival rate in the early stage diagnosis is only 31%, and 43% of people with liver cancer are diagnosed at stages 0 and 1.

Cruciferous vegetables are a group of vegetables, belongings to the family Brassicaceae (also called Cruciferae) with many genera, and species, and cultivated all over the world in suitable climates for commercial profits, including cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts, and green leafy vegetables.

On finding a potential compound for the treatment of cancer, researchers examined the effects of Isothiocyanates (ITCs), the phytochemicals derived from the breakdown of glucosinolates that exist in cruciferous vegetables against liver cancer cells.

In HepG2 cells, all ITCs reduced metabolic activity in each cell line, particularly in the compound phenylethyl-ITC.

Both dietary and synthetic ITCs were found to suppress the migratory and invasive potential of all cell lines.

Furthermore, chronic injection of dietary and synthetic ITCs also inhibited the cancer cell colony-forming capability and induced apoptosis.

Phenylethyl-ITC exposure resulted in a significant generation of reactive oxygen species to stimulate the expression of cytotoxicity in the cancer cell lines.

Collectively, researchers said, "These data highlight the potential advantages of utilizing ITCs to delay the carcinogenic process and the potential for dietary and synthetic ITCs to act as anticancer agents".

Taken together, Cruciferous vegetables may be considered functional foods for the prevention of liver cancer, pending the confirmation of the larger sample size and multicenter human study.

#Sesquiterpenoids Isolated From Herbal #Coltsfoot Exerts Neuro-Protective Activity by Inhibiting Overexpression of Oxidativestress, Researchers Say

Kyle J. Norton

Neuroprotection is a process to preserve the neuronal structure and/or function against the death or gradual death of neurons, including the use of medicines and herbs.

In other words, neuroprotection protects the neuronal integrity against the reduction in the rate of neuronal loss over time inducing neurological disease, including neurodegenerative disease.

Neurodegenerative disease is the class of conditions associated with the progressive loss of neurons that occurs mostly in older adults, including dementia. The most common dementia in the US is Alzheimer`s disease (AD).

Alzheimer's disease is an irreversible, progressive brain disorder caused by the slow loss of neuron in the brain that affects the memory and thinking skills and, eventually, the ability to carry out the simplest tasks as the disease progress into an advanced stage.

According to the statistics, dementia affects over 47.5 million people worldwide, and approximately 58 percent are living in low and middle-income countries.

Believe it or not, 5.8 million Americans of all ages are living with Alzheimer's dementia in 2019 and two-thirds of people with Alzheimer's disease are women.

Conventionally, as of today, there is no cure for dementia. Treatment of dementia focuses on improving dementia symptoms and slowing down the progression of the disease.

Coltsfoot(Tussilago farfara) is a perennial herbaceous plant, genus Tussilago, belonging to the family Asteraceae, native to Europe and Asia. The herb has been used for thousands of years to treat asthma, various coughs, bronchial congestion, respiratory disorders, headaches obstruction in the nasal passage, etc.

In finding a potential compound for the treatment of the neurological disorder, researchers examined the neuroprotective effect of Tussilagone (TSL) on LPS-induced neuronal cell death.

According to the tested assays, 4 sesquiterpenoids isolated from the TSL showed a significant effect against the production of nitric oxide, prostaglandin E2, and tumor necrosis factor-α in LPS-treated BV-2 cells.

In other words, the sesquiterpenoids inhibited the production of free radicals and proinflammatory expression associated with the BV-2 cells oxidative stress induced by LPS.

More precisely, the inhibition against the initiation of oxidative stress was attributed to the sesquiterpenoids activity in blocking the activation of the proinflammatory (NF-κB) pathway

Furthermore, the sesquiterpenoids also inhibited LPS-induced neuronal cell death in the co-culture system by inhibiting the expression of NF-κB pathway and scavenging of ROS.

Based on the findings, researchers said, "sesquiterpenoids from Tussilago farfara may have beneficial therapeutic potential for the treatment of neurodegenerative diseases through inhibition of microglial activation".

Taken together, coltsfoot may be considered a remedy for the treatment of neurological disease, pending to the confirmation of the larger sample size and multicenter human study.

#Herbal #Angelica, Processes Anti -Apoptotic, Anti-Proliferative and Cell Cycle Arrest Activities Against the Risk of #Cancer, Researchers Show

Kyle J. Norton

Cancer is a group of medical and chronic diseases characterized by cell growth disorderly and uncontrollably in the tissue of specific organs in the body.

Over time, the alternative cells-induced tumor may become progressively worse and damage other healthy tissues, depending on the grade of the tumor.

At the later stage, the most invasive tumor can induce secondary metastasis by spreading to other organs in the body via lymph or blood.

According to the National Cancer Institute, approximately 38.4% of men and women will be diagnosed with cancer at some point during their lifetimes, according to the data from 2013–2015.

In 2017, an estimated 15,270 children and adolescents ages 0 to 19 were diagnosed with cancer, and 1,790 died of the disease.

In cancer, antiproliferation is a process in which the cancer cells are killed or limited from growing through many mechanisms, including the use of chemo drugs.

In the immune perspective, anti-proliferation can be carried out by activating the genes associated with cancer suppression, programming cell death,.... and cell cycle arrest.

A suppressive gene, such as pro-apoptotic p53 induces cancer cell death through cell cycle arrest. In other words, the gene inhibits tumor growth by interrupting the cancer cell cycle division.

The programmed cell death gene, such as BCL-2 induces cell death by either inducing apoptosis. In other words, If cells are no longer needed or damaged, activating an intracellular death program will cause cell death.

In phytochemicals, anti-proliferation can also be carried out by antioxidants which stimulate the free radicals overexpression to induce toxicity to tumor cells without causing harm to the nearby healthy cells.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen the heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

On finding a potential compound for the prevention and treatment of cancer, researchers examined the effects of Angelica phytochemical (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone from A. purpuraefolia on cell proliferation and apoptotic cell death.

In 4 breast (MDA-MB-231, MCF-7, HS578T, and T47D), three cervical (HeLa, SiHa, and C33A) cancer cell lines, and NIH 3T3 normal cells used for the experiment, researchers found that
* (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone treatment suppresses the growth and proliferation of cancer cells at a relatively low concentration (<10 μg/ml).

* The phytochemical also induces cancer cell apoptosis at a high concentration (>50 μg/ml).

* At the concentration of 10 μg/ml, the chemical compounds also exerted a strong cell cycle arrest in the S/G2 phase.

Furthermore, according to the Western blot analysis, the concentration higher than 50 μg/ml of (+)-4'-decanoyl-cis-khellactone increased the large percentage of apoptotic cells in a time-dependent manner.

Interestingly, while the (+)-4'-decanoyl-cis-khellactone uses both extrinsic and intrinsic pathways to induce apoptosis, the but (+)-3'-decanoyl-cis-khellactone uses only an intrinsic pathway for the same results.

In other words, the (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone also inhibited the in vitro proliferation of six other human breast and cervical cancer cell lines with different pathways.

Based on the evidence, researchers said, "Our data strongly suggest that (+)-4'-decanoyl-cis-khellactone and (+)-3'-decanoyl-cis-khellactone act as an anti-cancer supplement particularly against breast and cervical cancers by inducing cell cycle arrest in the S/G2 phase and caspase-dependent apoptosis at relatively low and high concentrations, respectively".

Taken together, Herbal Angelica processes anti-proliferative activity against cancer, pending a larger sample size and multicenter human study.

#Phytochemical #Indicaxanthin Inhibits the Onset of Oxidative Stress induced by 7-KC, Researchers Find

Kyle J. Norton

Oxidative stress is a condition associated with the long-term imbalanced ratio of free radicals and antioxidants in the body, leading to protein, lipid, and cell damage that cause chronic illness such as heart disease, stroke, and diabetes.

In other words, the condition is caused either by overexpression of free radicals depletion of antioxidant enzymes produced by the host, or both.

Antioxidants are either produced by the body or injected into the body from outside sources such as the consumption of antioxidant-rich fruits and vegetables.

Free radicals are everywhere, including the air we breathe, the food we eat, and the body's daily functioning such as cell metabolism.

More precisely, any chemical compound that has an oxygen molecule can become a free radical after entering our body.

In healthy individuals, the antioxidant produced by the host is more than enough to counter the bombards of free radicals. However, people with a weakened immune system such as the elderly and immature immune systems such as the baby may need the injection of antioxidants from outside sources.

Particularly, people with a suppressive immune system due to chemotherapy may be required to intake large amounts from antioxidant-rich food regularly.

Dr. Biller-Takahashi JD and colleagues wrote, "Reactive oxygen species (ROS) are reactive molecules containing oxygen, that form as byproducts of aerobic metabolism, including immune system processes. Too much ROS may cause oxidative stress".

And, " This study shows a relationship between oxidative stress and immunity, and, from a practical perspective, shows that improving immunity and health in pacu through the administration of selenium could improve their growth performance".

However, free radicals in moderate amounts are needed for body functioning, including lowering blood pressure and relaxing blood vessels to move blood, such as nitric oxide NO.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, kale, etc.

Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains found abundantly in beets, chard, etc.

In finding a potential ingredient for the prevention of disease associated with oxidative stress, researchers investigated the effect of indicaxanthin (Ind) on the 7-KC-induced apoptosis of human monocyte/macrophage THP-1 cells.

Where 7-Ketocholesterol (7-KC) has been found to induce apoptosis of macrophages, a key event in the development of human atheromas.

Based on the tested analysis, 7-KC within 24 hours of injection showed the pro-apoptotic activity observed by the early formation of intracellular reactive oxygen species over basal levels, NADPH oxidase-4 (NOX-4) over-expression, and elevation of cytosolic Ca²⁺, with progressive depletion of total thiols.

In other words, 7-KC induced oxidative stress in the human monocyte/macrophage THP-1 cells, leading to cytotoxicity.

Interestingly, 7-KC also inhibited the process of the redox-sensitive NF-κB against oxidation in the tested cells.

Co-incubation of 2·5 μm of indicaxanthin completely prevented 7-KC-induced pro-apoptotic events.

The efficacy of indicaxanthin was attributed to the inhibition of NOX-4 basal activity and over-expression and inhibition of NF-κB activation.

More precisely indicaxanthin promoted the THP-1 cells' redox balance and Ca homeostasis, thus preventing mitochondrial damage-induced apoptosis.

Based on the findings, researchers said, " Ind, a highly bioavailable dietary phytochemical, may exert protective effects against atherogenic toxicity of 7-KC at a concentration of nutritional interest".

Taken together, indicaxanthin may be considered a remedy for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

#Anthocyanin-Rich Red Cabbage Extract (ARCE) Reduces the Risk of Cell Apoptosis and Cytotoxicity in Induced Myocardial Infarction

Kyle J. Norton

Myocardial infarction is a severe medical emergency characterized by the obstruction of the blood supply to the brain, causing local brain neurons to die. Prompt treatment can minimize brain damage and potential post-disease complications.

In other words, myocardial infarction (MI) occurs due to the imbalance in oxygen supply and demand caused by plaque rupture with thrombus formation in an artery, leading to reduced blood flow to the heart.

The occurrence and extent of brain infarction are associated with three basic factors, including the site of arterial occlusion, the rapidity of arterial occlusion, and the presence or absence of collateral circulation.

In other words, brain infarction can be classified into myocardial infarction due to plaque rupture and myocardial oxygen supply-demand imbalance secondary to other acute illnesses.

The exact causes of a blood clot to induce acute brain infarction are unknown. Plaque accumulation in the arteries associated with hardening and stiffing arterial walls is a leading cause of cardiovascular disease, including stroke.

According to the statistics provided by the CDC, every year, approximately, about 790,000 Americans have a heart attack, including 580,000 cases of a first heart attack and 210,000 people who have already had a first heart attack.

The most common symptoms of MI are pressure, tightness, pain, or a squeezing or aching sensation in your chest, digestive discomforts such as nausea, indigestion, heartburn, abdominal pain, shortness of breath, and dizziness.

Conventionally, treatment of myocardial infarction is a focus to reduce the damage of the heart muscle accomplished through medical or mechanical means.

Cabbage is a species of Brassica oleracea belonging to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem with a crowded mass of leaves, usually green but in some varieties, it may be red or purplish.

In findings of natural whole food for the treatment of post-stroke, researchers examined the effects of anthocyanin-rich red cabbage extract (ARCE) on H2O2-treated rat neonatal cardiomyoblasts (H9c2 cells) and isoproterenol (ISO) induced rodent model of myocardial infarction.

According to the tested analysis,
* H2O2 treated H9c2 cells recorded cytotoxicity (48-50%) and apoptosis (57.3%), compared to the reduction in the presence of ARCE (7-10% & 12.3% respectively).

* Rats pretreated with ARCE for 30 days followed by ISO treatment recorded a favorable heart: body weight ratio as compared to ISO treated group.

* Furthermore, the mRNA levels of enzymatic antioxidants (sod and catalase) and apoptotic genes (Bax and BCL-2) in the ARCE+ISO treated group were similar to the control group suggesting that ARCE pretreatment prevents ISO-induced depletion of enzymatic antioxidants and apoptosis.

* Moreover, ARCE+ISO treated group (4.5%) recorded results comparable to control (0%) in infracted areas (10%) and derangement of the myocardium.

* The ARCE+ISO treated group accounted for promoted the caveolin-3 associated with the heart muscle and SERCA2a expression involved in the reduced risk of heart failure, compared to the ISO treated group an indication of ARCE in the mediated reduction in membrane damage and calcium imbalance.

Based on the results, researchers wrote, "Overall this study elucidates the mechanism of ARCE mediated prevention of experimentally induced myocardial damage".

Taken together, cabbage may be used for the treatment of myocardial infarction, pending the confirmation of the larger sample size and multicenter human study.

#Honey Inhibits the Risk of #RefluxEsophagitis RE through Its Monperoxide Antibacterial Activity, According to Studies

Kyle J. Norton

Scientists may have found a natural alternative for the treatment of reflux esophagitis, some studies found.
Acid Reflux or heartburn is a condition characterized by the going back up of acidic stomach contents to the esophagus. Most cases of acid reflux are happened immediately after eating causing a burning sensation or pain in the lower chest area.

According to the statistics, in the US, approximately, 20% of people with acid reflux will develop gastroesophageal reflux disease (GERD). More than 65% of people who suffer from acid reflux are 40 years old or older. And, 15 to 30 5 of the U.S. population has GERD.

Too much tea, coffee, and alcohol smoking pregnancy, obesity, and eating habits such as eating close to bedtime, eating a large meal, and eating fatty and fried foods are some of the risk factors that initiate the early onset of acid reflux, according to epidemiological studies.

Believe it or not, long-term intake of certain medications such as bronchodilators for asthma, progestin for birth control or to treat abnormal menstrual bleeding, sedatives for insomnia and anxiety, calcium channel blockers for high blood pressure.

Dr. Zeynel Mungan and colleagues in examing the medication risk of GERD wrote, "Gastroesophageal reflux disease (GERD), which is common in many communities, is associated with structural factors, eating habits, and the use of certain drugs".

And, "The use of such drugs can lead to the emergence of GERD and can also exacerbate existing reflux symptoms. These drugs can contribute to GERD by directly causing mucosal damage, by reducing lower esophageal sphincter pressure (LESP), or by affecting esophagogastric motility".

Therefore, if you currently taking some of the aforementioned medication, please make sure that you know all the side effects for preventive measure.
Honey is the miraculous product made by bees using nectar from flowers.

The rich golden liquid is considered of healthy sweet food for replacing the use of white sugar and artificial sweetener by many people.

Scientists finding a natural remedy for the treatment of acid reflux investigated the honey effect on reflux oesophagitis (RE).

Injection honey displayed a strong effect on reflux oesophagitis by inhibiting the free radical expression which has been found to be the mediator in the generation of RE.

Furthermore, besides inhibiting the overexpression of free radicals, honey also promotes the production of antioxidant enzymes in the host, leading to a balanced ratio of free radicals and antioxidants in the body.

Moreover, honey injection also reduced the levels of inflammation by inhibiting the nitric oxide and prostaglandin E2 production associated with free radicals and inflammation, respectively.

The effects of honey against RE are also attributed to the nature of high density, high viscosity, and low surface tension, which makes them stay longer in the esophagus as a coating on the mucous membrane.

Additionally, researchers also found that the anti-RE activity of honey was also due to its nonperoxide antibacterial activity.

The findings suggested honey inhibits the onset of progression RE through a number of mechanisms, including proteins associated with the production of pro-inflammatory cytokines and antioxidant enzymes produced by the honey and the host.

Taken altogether, honey may be considered a functional remedy for the prevention and combined with the primary therapy for the treatment of reflux esophagitis.

#Broccoli' #Sulforaphane (SFN) Exerts Chemo-Preventive Activity by Controlling CancerCell #Carcinogenesis, Studies Find

Kyle J. Norton

Cancer is the class of diseases associated with the growth of cells disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite, and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American Cancer Society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Chemo-drugs are a class of conventional medicine used for the treatment of cancers. including 5-fluorouracil, 6-mercaptopurine, cytarabine, gemcitabine, and methotrexate.

Compared to other treatments, chemotherapy targets cancer's specific genes, proteins, or the tissue environment that contributes to cancer growth and survival.

Chemo drugs have been found to improve the survival rate of cancer patients. However, they also induce significant side effects.

The most common side effects of chemo-drugs are fatigue, hair loss, easy bruising, and bleeding. infection, anemia (low red blood cell counts) nausea and vomiting, appetite changes, and constipation.

The side effects of chemo drugs are the results of damage to the healthy blood-forming cells in the bone marrow, hair follicles, and cells in the mouth, digestive tract, and reproductive system.

According to the American Cancer Society, some chemo drugs have also been found to damage cells in the heart, kidneys, bladder, lungs, and nervous system.

Chemoprevention is the use of a medication, vitamin, or supplement to prevent the initiation of cancer.

In finding a potential plant for the treatment of cancers, researchers compared the effects of
The Brassica family of cruciferous vegetables such as broccoli is a rich source of isothiocyanates and sulforaphane (SFN) as a chemopreventive phytochemical.

Broccoli is a mustard/cabbage plant, belonging to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

According to the differentiation, sulforaphane (SFN) exerted its potent chemopreventive agent based on its ability to target multiple mechanisms within the cell to control carcinogenesis.

More precisely, anti-inflammatory, pro-apoptotic, and modulation of histones are some of the more important and known mechanisms by which SFN demonstrates chemoprevention.

Furthermore, The effect of SFN on cancer stem cells is another area of interest that has been explored in recent years and may contribute to its chemopreventive properties.

Based on the results, researchers said, "In this paper, we briefly review structure, pharmacology, and preclinical studies highlighting chemopreventive effects of SFN".

Taken together, broccoli consisting of SFN may be considered a chemopreventive functional food, pending the confirmation of the larger sample size and multicenter human study.
