#Hydroethanolic (HME) and #Acetone (AcE) Found in #Tomato Induces Cytotoxicity Against Cancer Cells through Their Antioxidant Activity, Scientists Suggest

Cytotoxicity is the process of the induction of cell death through toxins.
In other words, cell cytotoxicity is the process to use certain chemicals or mediator cells to destroy living cells.

In normal, cytotoxicity can cause the death of healthy cells

In cancers, cytotoxicity can be beneficial to humans by destroying the cancer cells and preventing them from proliferation.

For example, cytotoxicity induced by oxidative stress due to the imbalanced ratio of free radicals and antioxidant enzymes produced by the body can cause protein, lipid, and cellular damage that leads to chronic low-grade inflammatory diseases and cancer.

However, stimulation of ROS and antioxidants production to induce cytotoxicity to the tumors can also cause cancer cell death without damage to nearby healthy cells.

Dr. Shikha Srivastava, the lead scientist in the evaluation of the antioxidant and cytotoxic properties in Quercetin, wrote, "Quercetin interacts with DNA directly, and could be one of the mechanisms for inducing apoptosis in both, cancer cell lines and tumor tissues by activating the intrinsic pathway".


And, " quercetin-induced cytotoxicity in leukemic cells in a dose-dependent manner, while ellagic acid showed only limited toxicity. Besides leukemic cells, quercetin also induced cytotoxicity in breast cancer cells, however, its effect on normal cells was limited or none".

Scientifically, most cases of cancer cells cytotoxicity are associated with the stimulation of proteins and pathways involved in either gene suppression or cell death programming.

Tomato is a red, edible fruit, genus Solanum, belonging to the family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

In the urgency to find a natural ingredient that kills cancer without inducing adverse effects, researchers examined the toxicity against human gastric adenocarcinoma cells (AGS) and the antioxidant activity of hydroethanolic (HME), acetone (AcE), an alkaloid (AE) extracts prepared from the leaves of the tomato plant.

Both HME and AcE extracts showed a significant effect in scavenging •NO (IC50 values ranged from 0.87 ± 0.12 to 1.54 ± 0.23 and from 0.90 ± 0.01 to 2.23 ± 0.14 mg mL-1, respectively), compared to HMEs that only scavenge O2•- showing IC50 values from 0.12 ± ≤0.01 to 0.43 ± 0.08 mg mL-1.


In other words, tomato extracts demonstrated a strong effect against human gastric adenocarcinoma cells (AGS) through their cytotoxicity without causing harm to normal cells through the antioxidant activity.

Furthermore, in the study to assess different varieties of tomato seed extracts potential antioxidant and/or antiproliferative activity, researchers found that San Marzano Rosso seed extract displays the highest free radical-scavenging activity with 68% of 2,2-Diphenyl-1-picrylhydrazyl radical inhibition.


Moreover, according to the brine shrimp test (LD50: 23,198 ppm), San Marzano Rosso seed extract also demonstrated the highest cytotoxicity against the A375 cell line, compared to other varieties.


Taken altogether, tomato and its leaves may be considered supplements for the prevention and treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.


Intake of tomato in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Ovarian cancer is a medical condition caused by irregular cell growth in the lining tissues of the ovary.
Most cases of ovarian cancer begin in the cells on the surface of the inner lining of the ovary tissue before penetrating into deeper layers and colonizing the entire organ to induce secondary metastasis through blood and fluid circulation.

Ovarian cancer can be divided into the types below
* Epithelial Ovarian Cancer
The epithelial ovarian tumor also is known as a surface epithelial-stromal tumor, caused by the alternation of DNA of the cell on the surface of the ovary. Epithelial ovarian tumor is the most common form of ovarian cancer accounting for over 90% of all cases, including serious tumors, endometrioid tumors, and mucinous cystadenocarcinoma.

* Germ cell ovarian cancer
Germ cell ovarian tumor is a cancer of the egg-producing cells within the ovary and can be cancerous or non-cancerous tumors. The condition tends to occur in children and teens as a result of birth defects.

* Sex cord-stromal ovarian cancer
Sex cord-stromal ovarian tumor is a cancer of the tissue of the ovary that accounts for 8% of ovarian cancers. It is believed that the cancer is caused by excess estrogen produced by sex cord-stromal ovarian tumors. Sex cord-stromal ovarian cancer tends to develop in the older age group.

The most common symptoms of ovarian cancer are abdominal pain, bloating or swelling, quickly feeling full when eating, discomfort in the pelvis area, and change of bowel movement accompanied by the general symptoms shared by other cancers, such as unintended weight loss, gastrointestinal discomfort, loss of appetite and persistent fatigue.


Some researchers suggested that the widespread obesity caused by the promotion of a high-fat diet in the Western world may correlate to the increased risk of ovarian cancer.

Dr. Foong KW, wrote, " " 14 studies found a statistically significant positive association between ovarian cancer risk and higher body mass index, 26 studies found no significant association, and 3 studies found a negative association between ovarian cancer risk and higher body mass index".

With an aim to find a potential compound for the treatment of ovarian cancer, researchers investigated the combined effect of reduced graphene oxide-silver nanoparticles (rGO-AgNPs, with biomolecule called lycopene,) and trichostatin A (TSA) in human ovarian cancer cells (SKOV3).

According to the tested assays, AgNPs uniformly distributed on the surface of the graphene sheet with an average size between 10 and 50 nm, rGO-Ag, and TSA demonstrated significant inhibition of the cell viability in a dose-dependent manner.

At low concentrations, the combination of rGO-Ag and TSA not only had a significant effect on cell viability, and increased cytotoxicity but also inhibited the level of malondialdehyde (oxidative stress) and decreased the level of glutathione (antioxidants) that induced cancer cell mitochondrial dysfunction.

Based on the findings, researchers suggested, " This combination chemotherapy could be possibly used in advanced cancers that are not suitable for radiation therapy or surgical treatment and facilitate overcoming tumor resistance and disease progression".

Furthermore, the combination of rGO-Ag and TSA-induced cancer cell apoptosis was attributed to their pronounced effect on DNA fragmentation and double-strand breaks.

In order to reveal more information about lycopene, and anti-ovarian cancer activity scientists examined the protective role of dietary lycopene against the risk of ovarian cancer.

The search of the meta-analysis. identified 10 studies selected from the PubMed database, and final risk estimates, using a random-effects model showed that lycopene showed an effect on the risk of ovarian cancer among postmenopausal women, compared to an insignificant reverse association between dietary lycopene and ovarian cancer risk to other groups.

In other words, rGO-AgNPs, with a biomolecule called lycopene, are effective in reducing the risk and treatment of ovarian cancer in all groups compared to dietary lycopene in postmenopausal women.


Taken altogether, tomatoes processed with abundantly bioactive compound lycopene may be considered supplements for the prevention and treatment of ovarian cancer, pending the confirmation of the larger sample size and multicenter human study.


Intake of lycopene in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Cancer is a group of chronic diseases characterized by cell growth disorderly and uncontrollably in the body tissue of a specific organ.

More precisely, curcumin inhibits tumor cell migration and invasion by reducing the expression and activity of MMP-2/9 associated with tumors proliferation via the suppression of the ERK signaling pathway in stimulating cancer cell growth.


Based on the findings and after taking other factors into account, Dr. QIAN CHEN, the lead scientist wrote in the final report, "Curcumin is a potential therapeutic agent for EC".

Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream. Insulin resistance and hyperglycemia are hallmarks of diabetes.

In other words, insulin produced by the pancreas is either blocked by the clogging-up of the insulin receptors by fat and cholesterol or the death of beta cells in the pancreas. In some cases, diabetes can also be associated with autoimmune disease, attacking the beta cells.

There are some major types of diabetes, including the best known, type 1 and 2.

With an aim to find a potent compound for the treatment of diabetes, researchers examined the effects of Withania somnifera (WS) on lipid peroxidation (LPO) and pancreas in type 2 diabetic rats.

The study included type 2 diabetes induced by a single intraperitoneal injection of STZ (100 mg/kg) to 2 days old rat pups pretreated by WS.

According to the tested analysis, untreated rats group showed significant levels of lipid peroxidation (LPO) observed by reducing levels of antioxidant enzymes produced by the host including glutathione (GSH) contents and by enzymatic activities of glutathione peroxidase (GPx), glutathione reductase (GR), glutathione-S-transferase (GST), superoxide dismutase (SOD) and catalase (CAT).

However, injection of WS not only caused a significant decrease in blood glucose and tissue LPO levels caused by the application of STZ but also a significant increase in GSH contents, compared to type 2 diabetic control rats.

Furthermore, antioxidant enzymes reduced in the untreated rat such as GPx, GR, GST, SOD, and CAT were also increased compared to type 2 diabetic control rats.

Most importantly, the significant reduction in the number and size of pancreatic beta-cells in STZ rats was normalized upon the administration of WS, evident from the histopathological examination.

In other words, WS protects the cells in the pancreas against oxidation and maintains the integrity of beta cells in insulin production in type 2 diabetes rats.

Based on the findings, researchers said, "WS has shown strong free radical scavenging activity and helped in improving the non-enzymatic and enzymatic antioxidants in type 2 diabetic rats".


Taken altogether, Ashwagandha WS may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of type 2 diabetes, pending to the results of large example size and multicenter study.

The liver is the largest internal organ in the human body, and plays an essential role in filtering blood from the intestine before passing it to other parts of the body.

The organ also produces cholesterol, a waxy substance that is needed to aid digestion and build strong cell membranes, production of steroid hormones, and vitamins.

Liver injury may be a result caused by physical impact or penetrating trauma. In some cases, liver injury can also be induced by toxicity, leading to acute liver injury.

Liver injury can be mild or severe. In a mild case, liver injury can recover itself. However, in severe cases. surgery to stop the bleeding and remove the part of the injury may be necessary.

According to the statistics, liver injury has been found to constitute 5% of all traumas, making it the most common abdominal injury.


Other most common symptoms associated with liver injury are shocks due to severe bleeding inducing a rapid heart rate, rapid breathing, and cold, clammy, pale, or bluish skin.

In some cases, blood accumulated in the abdomen may irritate the abdominal tissue, leading to symptoms of abdominal pain swelling, and tenderness.

The severity of a liver injury can be diagnosed by computed tomography (CT) or ultrasonography.

Depending on the size of the wound, most cases of liver injury are healed by themselves. However, in severe cases, hospitalization may be necessary if the bleeding does not stop.

The most common cause of liver injury is an overdose of medicine which has been found to be the number 1 factor.


Cabbage is a species of Brassica oleracea belonging to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem with a crowded mass of leaves, usually green but in some varieties, it may be red or purplish.

* Bocfal EO contained organic polysulphides (OPSs), such as dimethyl trisulphide (DMTS) 65.43 ± 4.92% and dimethyl disulfidefood (DMDS) 19.29 ± 2.16% as major constituents.

* Bocfal EO and DMTS were found to be potent TBARS, a byproduct of lipid peroxidation inhibitors with IC50 values of 0.51 and 3 mg/L, respectively.


* Bocfal EO demonstrated better hepatoprotective properties than did DADS (p < 0.05), although both slightly affected the hepatic parenchyma per se.

Nonalcoholic fatty liver disease (NAFLD) is a chronic condition induced by the onset of cirrhosis and liver cancer caused by fat accumulated in the liver over time, with no influence of excessive alcohol use. The disease can be classified into the types of non-inflammatory fatty liver (NAFL) and inflammatory nonalcoholic steatohepatitis (NASH).

Then, rats from the ND·4w group continued for 4 weeks with the same diet (ND·8w), and rats from HFD were fed during 4 weeks with an ND (HFD·4w+ND·4w) or an ND plus LYC (HFD·4w+ND+LYC·4w).

Lycopene is a phytochemical found in tomatoes in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

Based on the tested analysis, ND plus LYC showed a significant effect on NAFLD observed by the normalizing liver weight, hepatic TC, and serum LDL.

Additionally, the ND plus LYC fed group demonstrated the reversal of some macroscopic and microscopic images of livers.

However, lycopene failed to inhibit the liver stress markers such as malondialdehyde level and serum aspartate aminotransferase activity during the period of study.


Dr. Piña-Zentella RM, the lead scientist after taking other factors into account wrote, "The results from this study suggest the potentially protective role of LYC against NAFLD".

Taken altogether, lycopene found in tomato may be considered a supplement for the prevention and treatment of Nonalcoholic Fatty Liver Disease, pending the confirmation of the larger sample size and multicenter human study.

Fennel may have a potential and positive effect in reducing stress implications, some scientists suggested.

Society and environment impact stress conditions, over a prolonged period of time, which may induce a change of behavior, leading to disturbances in normal physiological functioning due to increased free radicals generation in the initiated onset of hypertension, neurosis, immune suppression, and other physical and mental disorders.

In the study to evaluate the anti-stress and memory-enhancing properties of F. Vulgare extract in experimentally stress rats at doses of 50, 100, and 200 mg/kg body weight administered orally with an orogastric tube researchers found that rats treated with all doses exert a significant effect in reduced stress implication linearly, observed by Urinary levels of vanillylmandelic acid (VMA) assay in 24 hours.

Further analysis of the rat group treated fennel with an additional injection of ascorbic acid also found that the secretion of urinary ascorbic acid levels increased from 65.74 to 78.59, 108.41, and 125.82 μg/kg within the same period on doses of 50, 100, and 200 mg/kg body weight respectively in compared to the untreated control group.

These results indicated that fennel has a stronger impact on reduced chronic stress connotation compared to both ascorbic acid and control groups.

Moreover, the group treatment group also displayed a reduced expression of lipid peroxidation in both rat liver and brain homogenates catalyzed by the stress in induction of ROS production and reduced natural antioxidant defense compared to the rat group treated with ascorbic acid.



Dr. S Koppula,, the lead author, after taking into account other confounders said, "The (Fennel) extract also exhibited potent antioxidant effect by inhibition of lipid peroxidation in both rat liver and brain homogenates to a greater extent than the standard antioxidant, ascorbic acid".


Additionally, in the review literature of a number of such drugs mostly in the form of their extracts (holistic approach) or in some, as active principles isolated from them, have been evaluated for their antistress activity by a number of tests which include open-field behavior, researchers found that
* Fennel may have a strong effect in reduced stress in tested mice through observation of swimming endurance and hexobarbitone sleeping time in association with stress in induction of anxiety.

* Fennel inhibited the production of the level of stress hormone corticosterone produced in the cortex of the adrenal gland.

* Fennel increased the production of antioxidants and enhanced the antioxidant defense of hosts such as Superoxide Dismutase (SOD) and sorbitol dehydrogenase (SDH), in inhibiting overexpression of ROS-in initiated chain reactions caused by chronic stress.

* Fennel inhibited levels of MAO in association with alternation of mood and mental conditions.

Finally, the application of fennel also reduced the tension of the central nervous system in response to stress impact, thus reducing neurotransmitter levels in stimulated production of stress hormones in facilitated ROS production, depression, anxiety,.......

The above illustration was supported by the application of fennel oil in exerted significant anti-psychiatric disorders caused by chronic stress such as depression and anxiety in tested mice respectively treated with water, fluoxetine (0.3 mg/kg), and fennel oil (0.5 ml/day) conducted by the Aga Khan University

According to the study, repeated administration of fennel oil for 3 weeks exerted significant antidepressant- and anxiolytic-like effects, observed by struggling time in forced swim test (FST) and time spent in open arm in the elevated plus maze (EPM) comparable to fluoxetine treatment group.

Interestingly, the rats treated group also exhibited high levels of antioxidants in inhibited ROS in facilitated production of free radicals through lipid peroxidation.

Taken together, fennel with abundant phytochemicals may be considered a functional food in reduced stress implication in the initiation of related diseases.
