#Herbal #Fennel Exerts Anti-Proliferative and Apoptotic and Cytotoxic Effect on #Cancer Cells, Scientists Show

Kyle J. Norton

Scientists may have found a whole food or herb with the potential to activate the protein that programs the death of cancer cells, some studies found.

Cell death is a natural process to warrants the normal development of cells and the cell cycle maturation and maintain the normal function body in the optimized state.

However, some microorganisms are found to process the activity in triggering cell death for their own survival.

Apoptosis, or programmed cell death, is a function of the immune system that evolved as a rapid and irreversible process to efficiently eliminate dysfunctional cells, including cells that were damaged by the alternation of DNA or infection

Cancer apoptosis is an action that causes cancer cells to die off with or without affecting the nearby cells.

Some herbal medicine such as fennel was shown to induce the function of overexpression of levels of free radical in initiating a cytotoxic effect that targets cancer cells while protecting the healthy cells through its antioxidant activity.

Fennel (Foeniculum vulgare) is a plant species of the genus Foeniculum, belonging to Apiaceae (Umbelliferae), native to the Mediterranean.

Herbal medicine has been used in traditional and herbal medicine as a warming, carminative, antispasmodic, antidepressant agent and to stimulate the appetite, ease indigestion, soothe coughing, reduce intestinal spasms, regulate the menstrual cycle, and relieve PMS,...

The study to investigate several herbal plants ethanol extracts from fruits of their apoptotic activity from fruits of seven species of Apiaceae family, namely, Eryngium planum, Archangelica officinalis, Pastinaca sativa, Heracleum sibiricum, Carum carvi, Foeniculum vulgare (fennel), and Levisticum officinale against ML-1—human acute myeloblastic leukemia, J-45.01—human acute T cell leukemia, EOL—human eosinophilic leukemia, HL-60—human Caucasian promyelocytic leukemia, 1301—human T cell leukemia lymphoblast, C-8166—human T cell leukemia, U-266B1—human myeloma, WICL—human Caucasian normal B cell, and H-9—human T cell, researchers found that
* The ethanol extract from fruit F. Vulgare shows the highest mortality in the Trypan blue test for the J45 cell line with 4% of viable cells and a C8166 cell line with 100% mortality.

* The extract also shows the highest viability in other cell lines, including the HL60 at 60%, EOL at 48%, and ML1 at 42%. The normal cell line H9 and WICL showed 35% and 25% of viable cells, respectively.

Furthermore, the reaffirm the use of fennel in traditional cancer treatment, researchers at the King Abdulaziz University (KAU), launched a study to demonstrate the anti-proliferative and apoptotic effect of chloroform fraction of fennel (CFF) in MCF-7 cells.

Application of fennel was shown to inhibit the growth against MCF-7 and MDA-MB-237 in time- and concentration-dependent manners.

Fennel-induced apoptosis of around 49% at the doses of 0.5 mg/ml of CFF and PI-stained cells showed the G1 phase cell cycle arrest, according to the Annexin V/PI assay.

More importantly, the application of fennel also was found to reduce the elevated levels of ROS, disrupted mitochondrial membrane and increased levels of caspase-9 & caspase-3, and DNA fragmentation in the promotion of MCF-7 cell death. experiment

Dr. Syed FQ, the lead author in the final assessment of the experiment said, "(The results) revealed the proliferation inhibition, cell cycle arrest and apoptosis induction effect of CFF, which may help in exploring the novel anti-cancer drug for therapeutic implications".

Moreover, in the study to examine whether anethole, a major bioactive compound found in fennel, anise, and camphor processes anti-metastatic and apoptotic effects on highly-metastatic HT-1080 human fibrosarcoma tumor cells, researchers found that
* Despite weak cytotoxicity against HT-1080 cells, anethole inhibits the adhesion to Matrigel and invasion of HT-1080 cells in a dose-dependent manner.

* Anethole is also able to down-regulate the expression of matrix metalloproteinase (MMP)-2 and -9 with anti-apoptotic function and up-regulate the gene expression of tissue inhibitor of metalloproteinase (TIMP)-1 with apoptotic function.

Finally, anethole significantly decreased mRNA expression of urokinase plasminogen activator (uPA), but not uPA receptor (uPAR) in the facilitated cancer cell death.

Taken all together, fennel alone or its extracts and bioactive compound anethole may be considered adjunct therapy combined with conventional medicine for the treatment of cancers

#Healthyfood #ManukaHoney Significantly Decreases the Ulcer Index and Preserves the Gastric Mucosal Glycoprotein, According to Studies

Kyle J. Norton

Scientists may have found natural alternative antibiotics for the treatment of stomach ulcers caused by a bacterial infection, some studies found.

A stomach/Gastric ulcer is a condition characterized by the breaking in the tissue lining of the stomach.

Most cases of stomach ulcers are caused by the infection with the Helicobacter pylori bacterium or long-term intake of anti-inflammatory medication.

The most common symptoms of stomach ulcers include a dull pain in the stomach and gastrointestinal discomforts such as nausea or vomiting. bloating, burping, or burning sensation in the chest.

If you are experiencing some of the above symptoms persistently, please check with your doctor to rule out the possibility of a stomach ulcer.
Unless the disease is caused by medication, most stomach ulcers can be treated successfully with the use of antibiotics and medications that reduce the overproduction of stomach acid.

Some researchers suggested that people who are obese are also susceptible to the onset of stomach ulcers due to the following reasons
* Epidemiological findings suggested that obesity is associated with inflammation in the gut, leading to the weakness of gut cells that are prone to ulcer formation.

* Obesity has been found to have an unhealthy ratio of gut bacteria.

* Overexpression of gut harmful bacteria increases the risk of toxin accumulation in the lining of the gut.

Honey is a miraculous product made by bees using nectar from flowers.

The rich golden liquid is considered one of the healthy sweet foods for replacing the use of white sugar and artificial sweeteners by many people.

Researchers on finding a natural compound for the treatment of stomach ulcers investigated the Manuka honey antiulcer activity in gastric rats.

Ethanol-induced gastric ulcers in rats selected for the study were divided into control, ethanol (ulcer), omeprazole, and manuka honey groups.

Injection of Manuka honey significantly decreased the ulcer index, completely protected the mucosa from lesions, and preserved gastric mucosal glycoprotein through the increased expression of gastric mucosal levels of nitric oxide (NO) in the inhibition of leukocyte adherence to the endothelium, and antioxidant enzymes glutathione (GSH), glutathione peroxidase (GPx), superoxide dismutase (SOD).

Furthermore, Manuka honey also decreased gastric mucosal MDA, the oxidative stress marker, and proinflammatory expression in the stomach caused by the injection of ethanol (ulcer), and omeprazole.

Dr. the lead scientist wrote, "Manuka honey likely exerted its antiulcer, effect by keeping enzymatic (GPx and SOD) and non-enzymatic (GSH and NO) antioxidants as well as inflammatory cytokines (TNF-α, IL-1β, and IL-6) in a reduced form, inhibited lipid peroxidation (MDA), and preserved mucous glycoproteins levels".

The findings clearly suggested Manuka honey inhibited stomach ulcers by modulating oxidative stress, proinflammatory cytokines, and antioxidant expression.

In order to reveal more information about the honey antiulcer activity, researchers evaluated the use of Manuka honey in the treatment of acetic acid-induced chronic gastric ulcers in rats.

The study included rats treated with three different concentrations of honey. Treatment with manuka honey was found to significantly reduce the ulcer index and maintain the glycoprotein content.

Manuka injection also reduced the mucosal myeloperoxidase activity, lipid peroxidation (MDA), and the inflammatory cytokines (TNF-α, IL-1β, and IL-6) as compared to the untreated control group.
Honey-treated groups showed a significant increase in enzymatic (GPx and SOD) and non-enzymatic (GSH) antioxidants besides levels of the anti-inflammatory cytokine IL-10, similar to that of the previous study.

More importantly, manuka honey-treated animals not only normalized cell cycle distribution but also significantly lowered apoptosis in gastric mucosa.

The results indicated that manuka honey is effective for the treatment of chronic ulcers through many mechanisms.

Taken together, Manuka honey may be considered a functioning remedy for the prevention and combined with primary medicine for the treatment of stomach ulcers with no side effects.

#Ginger's 6-#Gingerol Processes Anti-Oral Cancer Properties, Researchers Say

Kyle J. Norton

Scientists may have found a natural remedy for the prevention and treatment of oral cancer with no side effects, according to studies.

Oral cancer also known as mouth cancer is a medical condition characterized by irregular cell growth in the tissue of the oral cavity, due to the alternation of cell DNA.

Most cases of oral cancer begin in the cells on the surface of the inner lining of oral tissue.

However, at the advanced stage, oral cancer cells can travel a distance away to infect other healthy tissues and organs, leading to secondary metastasis.

According to the statistic provided by the American Cancer Society, in 2018, approximately 51,540 people were diagnosed with oral cavity or oropharyngeal cancer. The disease causes the death of an estimated 10,030 people.

The 5-year survival rate of oral cancer is 66%, including all stages.

Family history, smoking, alcohol drinking, infection by the HPV virus, poor hygiene, and genetic preposition are some of the risk factors associated with the disease.

Some researchers suggested that oral sex may be one of the major prevalent risk factors involved in the development of oral cancer. Dr. Rajiv Saini in the examination of the connection between oral sex and oral cancer, wrote, "The diverse conduit in the oral cavity that provides as a gateway of exposure of infection from the oral cavity to bloodstream include any open sores, cuts, abrasions, or bleeding gum disease such as gingivitis and periodontitis".

"Accumulating molecular and pathologic evidence has strengthened the hypothesis that HPV infection could play a role in the etiology of some oral squamous cell carcinomas.

And, "To reduce the risk of infection during unprotected oral sex, limit exposure to sexual fluids and ensure that no cuts or lesions are present in the mouth or on the genitals. Good oral health, free from bleeding gums, lip sores, cuts, broken skin, and oral epithelium, enormously reduces the chances of transmission of infection among the partners indulging in oral sex".

These results strongly suggested people who perform oral sex before, during, and after sexual intercourse may be at risk of oral cancer due to being infected by the HPV virus.

Researchers to find a natural compound for the inhibition of oral cancer examined the [6]-Shogaol ([6]-SHO), a major constituent of ginger anti-tumor activity.
[6]-SHO showed a significant effect in preventing oral squamous cell carcinoma by activating proapoptotic factors in vitro and in vivo experimental models.

In the inflammation and cell proliferation assays, injection of [6]-SHO inhibited the oral cancer cell line through cytotoxicity induced by the expression of proinflammatory cytokines, proteins associated with cell apoptosis, and anti-cell survival mechanisms.

Furthermore, in the oral cancer cells, oral administration of [6]-SHO strongly inhibited phosphorylation in stimulating the enzymes that carry energy to the cancer cells and enzymes in the degradation of cellular response to inflammation.

Dr. Annamalai G, the lead scientist said, " [6]-SHO is a novel functional agent capable of preventing DMBA-induced inflammation and cell proliferation-associated tumorigenesis by modulating multiple signaling molecules".

Additionally, in order to reveal more information about 6-Gingerol anti-oral cancer properties, researchers investigated the compound in human oral (SCC4, KB) with or without wortmannin, rapamycin, and cisplatin.

6-Gingerol showed dose-dependent cytotoxicity in the oral cell line.

Combinations of 6-gingerol with wortmannin and cisplatin showed additive effects, while rapamycin, it showed 50% cytotoxicity equivalent to that of IC50 of 6-gingerol alone.

Treatment with 6-gingerol exhibited S-phase arrest in SCC4 cells. But 6-Gingerol, wortmannin, and rapamycin treatment showed almost two-fold higher expression of proteins associated with cancer cell apoptosis.

Dr. in the final report wrote," 6-gingerol appears to be a safe and potent chemotherapeutic/chemopreventive compound acting through cell cycle arrest and induction of apoptosis in human oral....cells".

Taken together, ginger processed bioactive ingredient 6-gingerol and [6]-Shogaol may be considered a functional remedy for the prevention and combined with primary medicine for the treatment of oral cancer, pending the confirmation of large sample size and multicenter human study.

#Phytochemical #Betanin Inhibits Diseases Associated with #Oxidativestress, According to Studies

Kyle J. Norton

Antioxidants are stable atoms that inhibit oxidation caused by free radicals.
Free radical scavengers are types of antioxidants that inhibit free radicals before they can orchestrate a chain reaction that can not be stopped until all electrons in the outermost ring are paired or inhibited by antioxidants.

Oxidative stress is associated with chronic disease due to the imbalanced ratio of free radicals and antioxidants produced by the host including heart disease, stroke, and diabetes.

Antioxidants besides being produced by the body, they can also come into the body's dietary sources.

In a healthy individual, an antioxidant produced by the body is good enough to counter the expression of free radicals induced by cell metabolism and free radicals induced by the environment such as the air we breathe and the food we eat.

However, for people with a weakened immune system, intake of antioxidants from outside sources such as fruits and vegetables can be helpful.

Believe it or not. moderate free radicals in the body are necessary for body functioning, including the promotion of heart function in blood circulation.

Most common types of free radical scavengers include
* Catalases, the very important enzyme is produced by the host that protects the cell from oxidative damage by free radicals.

*Glutathione peroxidase, is a cytosolic enzyme produced by the host that dissolves hydrogen peroxide to water and oxygen and peroxide radicals to alcohols and oxygen.

*Superoxide dismutase (SOD), is also produced by the host that dissolves superoxide radicals into either ordinary molecular oxygen or hydrogen peroxide.

*Vitamin E, Vitamin C, and vitamin D.

Betanin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In the urgency to find a natural compound for the treatment of diseases associated with overexpression of free radicals, researchers examined the betanin free radical scavenging activity.

According to the Trolox equivalent antioxidant capacity (TEAC) assay, at a pH > 4, betanin is about 1.5-2.0-fold more active to scavenge free radicals compared to those of anthocyanins.

More precisely, the exceptionally high antioxidant activity of betanin was associated with an increase of its H-donation and electron-donation ability, totally depending on the expression of cationic state to mono-, di- and tri-deprotonated states present at basic solutions.

In other words, the efficacy of betanin's high free radical scavenging activity was dependent on the levels of pH.

Taken together, betanin may be considered supplements for the prevention and treatment of diseases associated with oxidative stress, pending to the confirmation of larger sample size and multicenter human study.

Intake of betanin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Honey Is Effective to Shortens the Duration of Bacterial Diarrhea, Researchers Say

Kyle J. Norton

Scientists may have found a natural alternative for the treatment of diarrhea, according to studies.
Diarrhea is a condition of loose, watery stools. Acute diarrhea is a common problem in North America that lasts for a very short time.

However, chronic diarrhea can be worrisome. The most common causes of chronic diarrhea are diseases associated with the gastrointestinal tract, including irritable bowel syndrome (IBS), inflammatory bowel disease (Crohn's disease and ulcerative colitis), and malabsorption syndromes.

According to the statistics, acute diarrhea is one of the most common causes of death among children younger than 5 years old worldwide.

The disease is associated mostly in countries with poor environmental sanitation, inadequate water supplies, and poverty.

There are many risk factors involved in the onset of the syndrome, including demography, socio-economy, poor water supply, poor sanitation, poor hygiene practices, breastfeeding, and malnutrition.

Some researchers suggested that a severely weakened immune system such as the use of immune-suppressive medicine and the elderly may also be at a higher risk of diarrhea.

Dr. Bouchaud O in examining the risk of immune deficiency and diarrhea wrote, "With a prevalence of approximatively 50%, diarrhea is a frequent event in an immune deficiency of any cause".

And "Gram-negative infections and Clostridium difficile colitis have to be diagnosed because efficient treatment is available. In these patients usually in bad general condition, because diarrhea is often of unknown etiology or noncurable, the diagnostic and therapeutic strategy must be pragmatic to control the symptoms without an excess of invasive procedures".

These results clearly suggested that if you have a severely weakened immune system, you are at higher risk of bacterial-infected diarrhea.

Honey is a miraculous product made by bees using nectar from flowers.

The rich golden liquid is considered one healthy sweet food for replacing the use of white sugar and artificial sweeteners by many people.

Researchers searching for a natural compound to treat diarrhea evaluated the honey gastroprotective effect against diarrhea.

Injection of honey induced a significant reduction in the duration of bacterial diarrhea, and it worked well as a substitute for glucose in an oral rehydration solution containing electrolytes.

In nonbacterial gastroenteritis, honey exerted a similar effect as glucose on the duration of diarrhea.

In an array of gram-positive and gram-negative bacteria, most pathogenic bacteria failed to grow in honey at a concentration of 40% and above, similar to those studies of honey on Staphylococcus aureus, Proteus mirabilis, and Candida albicans failed to grow in the presence of undiluted honey in vitro experiments.

Honey-agar all intestinal bacterial pathogens failed to grow at a concentration of 40% and above of pure honey, compared to those of bacteria in control plates and different growth media.

Scientists wrote in the final report, "Findings from studies on honey have thus shown that when given with an ORS it shortens the duration of bacterial diarrhea and may safely be used as a substitute for glucose, provided that the solution contains electrolytes".

In order to reveal more information about honey anti-diarrhea activity, researchers examined the honey on diarrhea and fecal microbiota in critically ill patients.

The double-blind, randomized controlled single-center study, included 32 patients randomly selected to receive a high protein kitchen enteral diet or honey as 10% of their carbohydrate intake.

The treatment group showed an insignificant increase in the frequency of bifidobacterium DNA by study day 7 in comparison with the control group and a considerable reduction in diarrhea.

Honey group also reduced the length of the Intensive Care Unit and the development of organ failure in critically ill patients.

Taken together, honey may be considered a functional remedy for the prevention and treatment of diarrhea, pending the confirmation of a large sample size and multicenter human study.

#Fennel Extract Exhibits Antipyretic Activity against #Hyperpyrexia, Researchers Suggest

Kyle J. Norton

Scientists may have found food and plant processes the antipyretic activity comparable to those of acetaminophen in patients with fever, some studies suggested.

Fever is a natural reaction caused by the production of pro-inflammatory cytokines in the immune system to protect our body from damage and injury against foreign pathogens during the acute phase of infection.

According to medical literature, a person is not considered to have a significant fever until the temperature is above 100.4 F (38.0 C).

Although fever is a common syndrome, however, if you or your kid are experiencing fever accompanied by symptoms of a severe headache, unusual skin rash, sensitivity to bright light, persistent stiff neck and pain, mental confusion, particularly persistent vomiting, and difficulty breathing or chest pain, you or your kid should go to the hospital right the way.

Fennel (Foeniculum vulgare) is a plant species of the genus Foeniculum, belonging to Apiaceae (Umbelliferae), native to the Mediterranean.

The plant and food have been used in traditional and herbal medicine as warming, carminative, antispasmodic, and antidepression agents and to stimulate the appetite, ease indigestion, soothe coughing, reduce intestinal spasms, regulate the menstrual cycle, and relieve PMS,...

In the study to evaluate the antipyretic effect of the aqueous extract of an herbal coded formulation containing an equal amount of Salix alba, Emblica Officinalis, Glycyrrhiza glabra, Adhatoda vasica, Viola odorata, in a yeast-induced pyrexia model in rabbits, researchers found that after recording the results immediately before the administration of the extractor paracetamol and again at 1 hour, application of the extract at 240mg/kg dose significant reduces elevated temperature as compared with that of standard drug paracetamol (150mg/kg).

Fennel extract applied alone showed antipyretic activity against hyperpyrexia in mice, induced by S.C. administration of 2 mL/100 g of a 20% aqueous suspension of brewer's yeast.

As an antipyretic agent, ethanolic extract of F. vulgare fruit showed a moderate antipyretic activity that was statistically significant after 30 and 90 min.

These results showed that herbal coded medicine at a dose of 240mg/kg and fennel extract have marked an antipyretic activity in animal models and this was strongly supported by the ethnopharmacological uses of medicinal plants.

Furthermore, in the examine the hepatotoxicity induced by acetaminophen caused by the excessive production of N-acetyl-para-benzoquinone imine (NAPQI) in the induction of symptoms such as fever due to inflammation, researchers showed that the application of trans anethole, an aromatic compounds isolated from fennel exerted a significant effect in the inhibited pro-inflammatory mediators such as prostaglandin (PGE2), cytokines (TNF, IL-1) and nitric oxide (NO)thus decreasing the risk of fever caused by an overreaction of the immune system.

Application of AN at doses of 125 and 250mg/kg once a day for seven days prevented the changes caused by the APAP overdose.

Additionally, researchers also discovered, that AN infection prevents the overexpression of proinflammatory mediators such as NO, TNF, IL-1α, MIP-1α, and MCP-1 and induced the up-regulation of anti-inflammatory cytokines (IL-4 and IL-10), thus reducing the onset of fever.

Taken together, there is no doubt that fennel may be considered a functional food for the treatment of inflammatory induced fever.

#Alliin Found in Garlic Powder Inhibits N-nitrosodimethylamine (NDMA) -Induced DNA Alteration Associated with the Onset of #LiverCancer, Scientists Say

 Kyle J. Norton

The liver is the largest detoxed internal organ with a function to filter out blood coming from the intestines before passing it to other parts of the body,

cancer is a chronic condition caused by irregular cell growth due to the alternation of liver cell DNA.

Additionally, the liver also produces cholesterol to aid digestion, the production of steroid hormones, vitamin D, and the building of strong cell membranes.

Liver pared to normal liver cell growth which is stopped after a certain stage of division, liver cancer cells can grow indefinitely to form a tumor, leading to reduced function of the liver.

According to the statistics provided by the American Cancer Society, in 2019, about 42,030 new cases (29,480 in men and 12,550 in women) will be diagnosed.

Primary liver cancer and intrahepatic bile duct cancer also caused the death of 31,780 people (21,600 men and 10,180 women) in 2019.

In the Western world, non-alcoholic fatty liver disease(NAFLD), a spectrum of hepatic disorders induced by obesity and metabolic syndrome, is a recognized risk factor for liver cancer.

Dr. György Baffy at the Harvard Medical School wrote, "Hepatocellular cancer (HCC) is the fifth most prevalent cancer worldwide and the third leading cause of cancer-related deaths. Non-alcoholic fatty liver disease (NAFLD), a spectrum of hepatic disorders associated with obesity and metabolic syndrome, is a recognized risk factor for HCC".

And, "NAFLD that is advanced to cirrhosis carries the highest risk for HCC, but there is an increasing concern that NAFLD-associated HCC may also occur in the non-cirrhotic liver".

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

With an aim to find a potential compound for the treatment of liver cancer, researchers examined the bioactive compound alliin isolated from garlic in an animal model, against liver cell DNA damage.
The study included rats induced with liver cancer by N-nitrosodimethylamine (NDMA) treated by administration of garlic powders for 2 weeks (5% of the diet).

According to the liver comet assay, injection of different garlic powders demonstrated a significant liver protective effect by a 35-60% reduction in DNA damage induced by N-nitrosodimethylamine (NDMA) in rat liver.

Additional injection of alliin content of the garlic powder showed a strongly proportional decrease in NDMA-induced DNA alteration.

Three garlic powders with or without alliin also protected the liver cell against DNA damage induced by aflatoxin B1 in the liver.

However, these decreases were not correlated to the allicin content of the garlic powders.

In other words, additional alliin increased the garlic powder function in the inhibition of liver cancer initiation caused by NDMA while alliin found in the garlic must work with other compounds synergically to induce the same effects.

Taken together, alliin may be considered a supplement for the prevention and treatment of liver cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
