Beetroot Juices BRJ Inhibits Oxidative Stress and Inflammation Associated with Cardiac Tissue Damage, Scientists Suggest

Inflammation is a natural response of the immune system that tries to protect the body against invasive pathogens.
In the acute phase of infection, after receiving the information transmitted by the central nervous system, the white blood cells of the first line of defense stimulate the production of blood platelets to cover the site of damage and pro-inflammatory cytokines, killing off the foreign invaders.

During the acute phase of infection, overproduction of proinflammatory cytokines may cause damage to nearby healthy cells, leading to the formation of scars.

Most cases of infection are stopped at the acute phase of infection. The wound is slowly healed within a few days or weeks.

However, if the immune system can not destroy all pathogens within a period in the range of 3 to 8 weeks, it will adapt to the new change, leading to chronic inflammation.

In other words, chronic inflammation happens when the immune response lingers, causing low-grade inflammation. Chronic inflammation over time may damage the body tissues and organs.

In some cases, the infection may be latent in which the pathogen remains within the body for a long time without producing any symptoms, such as the hepatitis virus.

Diseases associated with chronic inflammation include cancer, heart disease, rheumatoid arthritis, type 2 diabetes, and obesity.

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, chard, etc.

On finding a natural pigment for the prevention and treatment of disease associated with chronic inflammation, researchers examined the effects of Beetroot (Beta vulgaris L.) juice (BRJ, betacyanins, and betaxanthin) anti-inflammatory activity in vitro and in vivo.

The study included rats-induced myocardial injury by isoproterenol (ISP) at 24 h intervals, followed by oral administration of BRJ for 28 days at doses of 150 and 300 mg/kg.


According to the tested assays, pretreatment with BRJ (150 and 300 mg/kg) not only reduced oxidative/nitrosative stress but also restored the cardiac endogenous antioxidants in myocardial injury rats.

ISP-induced cardiac inflammatory cytokines such as NF-α, IL-6, and IL-10 and biomarkers of inflammatory promotion such as NF-κB also inhibited by the pretreatment of BRJ.

Oxidative stress-induced hyperlipidemia level was significantly reduced by the BRJ pretreatment.

The results strongly suggested that BRJ pretreatment mitigates cardiac dysfunction and structural damage by decreasing oxidative stress, and inflammation in cardiac tissues.

Dr. Raish M, the lead scientist said, "These results further support the use of BRJ in traditional medicine against cardiovascular diseases".

Furthermore, in rat-induced renal damage by Paraquat injected intraperitoneally randomly divided into four groups: a control group, a paraquat group, and two paraquat groups that were treated with betanin at 25 and 100 mg/kg/d three days before and two days after paraquat administration.

According to the differentiation of the proteins associated with pro-inflammatory cytokines and pathways that promote inflammatory stimulation, betanin administration found in the family of betacyanins alleviated the paraquat-incurred acute kidney injury by decreasing oxidative stress associated with the inflammatory expression.


Taken altogether, betacyanins processed abundantly bioactive ingredient betanin in BRJ may be considered a remedy for the prevention and treatment of disease associated with inflammation, pending the confirmation of a larger sample size and multicenter human study.

Intake of betacyanins in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Inflammation is a natural response of the immune system which tries to protect our body by destroying harmful pathogens before they cause damage.

In the acute phase of infection, the immune system white blood cells in the first line of defense stimulate the production of a blood palette to cover the wound and the production of inflammatory cytokines to kill off micro-organisms, leading to symptoms of fever, redness, and swelling.

Overproduction of proinflammatory cytokines in some cases has been found to damage nearby healthy cells and tissues, causing the formation of scars.

In severe cases, a protein with duo functions in the production of pro and anti-inflammatory cytokines which produce anti-inflammatory cytokines under normal conditions also switch to produce pro-inflammatory cytokines.

Most harmful pathogens are stopped at the acute phase of infection. However, if the immune first line of defense can not completely destroy the invaders within a period of 3 to 8 weeks, it will compromise, leading to low-grade chronic inflammation.

Diseases associated with low-grade chronic inflammation include diabetes, cardiovascular disease, and cancer.

Conventional treatment of acute phase inflammation is to manage inflammation and prevent the risk of long-term damage, including anti-inflammatory nonsteroidal anti-inflammatory drugs (NSAIDs) which can be bought without a prescription.


3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, kale, etc.

In the urgency of finding a potential compound that processes a strong anti-inflammatory activity, researchers examined the effects of 3,3'-Diindolylmethane (DIM), a condensation product of indole-3-carbinol, and a glucosinolate naturally occurring in Brassica genus vegetables.

The study was carried out by differentiating the 3T3-L1 adipocytes in the co-cultured with RAW 264.7 macrophages and exposed to 20 µM, 40 µM, and 60 µM DIM for 24 h followed by 100 nM insulin for 20 min.

According to the tested assays, DIM significantly (p<0.05) reduced the production and mRNA expression of proinflammatory cytokines (MCP-1, IL-6, and TNFα) in a concentration-dependent manner.

Injection of DIM was found to increase the expression of proteins involved in the positive regulation of metabolism and proliferation (IRS-1 pY612 and Akt-1/PKB pT308) through its anti-inflammatory properties.

In other words, DIM influences the glucose metabolic pathway by exerting an anti-inflammatory effect.

Dr.Lopez-Vazquez A, the lead scientist said, "The potential therapeutic benefits of DIM in the treatment of glucose metabolic disorders deserve further studies".

In order to reveal more information about DIM anti-inflammatory effects, researchers examined the dietary components found in cruciferous vegetables, on brain inflammation.


Administration of DIM suppressed lipopolysaccharide (LPS)-induced expression of reactive oxygen species and promoted the production of antioxidant enzymes in the host by inhibiting the microglial hyperactivation by attenuating inflammatory transcription factor NF-κB.

Furthermore, DIM protected primary cortical neurons from inflammatory toxicity induced by LPS.

In vivo model of neuroinflammation, DIM also suppressed LPS-induced brain inflammation in the mouse hippocampus.

Based on the findings, Dr. Kim HW, the lead researcher said, "DIM may have beneficial potential against brain inflammation and neurodegenerative diseases through the negative regulation of the NF-κB signal pathway in microglia".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of diseases associated with chronic inflammation, pending the confirmation of the larger sample size and multicenter human study.

NADPH oxidases, a multisubunit enzyme comprising membrane and cytosolic components plays a critical role to protect our body against pathogenic microorganism by maintaining immune function, cell growth, and apoptosis.
NOX family of NADPH oxidases deficiency can cause immunosuppression, leading to the overexpression of intracellular ROS that results in various lethal diseases.

Epidemiologically, diseases associated with the NOX family of NADPH oxidases deficiency-induced immunosuppression are frequent and recurrent pneumonia, bronchitis, sinus infections, ear infections, meningitis or skin infections, inflammation, and infection of internal organs, blood disorders, such as low platelet counts or anemia, digestive problems, such as cramping, loss of appetite, nausea and diarrhea and delayed growth and development, particularly in cancer developing.

The NOX family of NADPH oxidases overexpression caused by a defect of its different subunits produced high levels of ROS in the gastrointestinal tract that causes chronic inflammatory stress; cytokine induction, leading to the formation of digestive cancer such as colorectal cancer.

A recent joint study led by the National Cancer Institute suggested that lycopene may be used for the treatment of cancer that targets the expression of NADPH oxidase 4-5 (NOX4-5).

Cancer is a class of diseases caused by cell growth irregularly and uncontrollably due to the alternation of cell DNA. Over time, cancer cells induced tumor formation to become progressively worse, leading to secondary metastasis.

Lycopene is a phytochemical in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas.

Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

In patients with chronic pancreatitis, overexpression of free radicals has been found to orchestrate pancreatic cancer.

Furthermore, NOX5 overexpression increased the pre-malignant fibrotic states that lead to carcinomas of the lung and liver.

High expression of NOX5 is also found in patients with malignant melanomas, prostate cancer, and Barrett's esophagus-associated adenocarcinomas,

NOX4 and 5 are members of the NOX family of NADPH oxidases.

Furthermore, lycopene administration in vitro and Vivo significantly inhibited NOX4 protein expression at 2.5 μM, compared to other concentrations.

The efficacy of lycopene ameliorated the expression of NOX4 protein was attributed to the significant inhibition of the NOX4 mRNA expression associated with the function of NOX in the production of reactive oxygen species.

Dr. Roy K, the lead scientist in the investigation of the risk of cancer in the NOX family of NADPH oxidases wrote, "Over-expression of functional NOX proteins in many tissues helps to explain tissue injury and DNA damage from ROS that accompany pre-malignant conditions, as well as elucidating the potential mechanisms of NOX-related damage that contribute to both the initiation and the progression of a wide range of solid and hematopoietic malignancies".

In SK-Hep-1 cells, intracellular ROS levels were also decreased by the injection of lycopene.

Moreover, the levels of NOX4 protein expression, and NOX activity and the intracellular ROS associated with transforming growth factor β (TGF-β)-induced cancer cell metastasis also were eradicated by the activity of lycopene upon injection.

Other mechanisms such as proteins involved in anti-apoptosis and enhanced cancer cell migration, invasion, and adhesion were also inhibited by the expression of lycopene.

Taken altogether, lycopene may be considered a natural remedy for the prevention of cancer cell metastasis associated with the overexpression of NADPH oxidase, pending to the confirmation of larger sample size and multicenter studies.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radicals before they can cause a chain reaction in facilitating oxidative stress that causes damage to proteins, lipids, and cell DNA.

Oxidative stress is a result of an imbalance in the ratio of free radicals and antioxidants in the body.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.

Due to their nature to become stable, free radicals readily donate or steal an electron from other molecules, leading to the chain of domino effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.
Antioxidants, on the other hand, are stable molecules with a unique function in the inhibition of oxidation.

Antioxidants can either be produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

However, in people with a weakened immune system, intake of dietary sources of antioxidants may be necessary.

Rapeseed, also known as rape, or oilseed rape, is a member of the family Brassicaceae (mustard or cabbage family), cultivated in suitable climate regions for its oil-rich seed for commercial profits.

In finding a potential compound for the prevention of heart disease, researchers at the University of Applied Sciences investigated the effects of a hypo energetic diet with low energy density (ED) enriched in rapeseed oil derived n-3 fatty acid α-linolenic acid (ALA), in patients with metabolic syndrome.

The human trial included eighty-one patients with metabolic syndrome randomly assigned to receive either rapeseed oil derived with high MUFA content and an ALA intake of 3.5 g/d on body weight or olive oil diet rich in MUFA but with a low ALA content.

According to the results, after a 6-month dietary intervention, the rapeseed oil and olive oil groups showed a significant in reduced body weight and metabolic factors associated with cardiovascular risk.

In other words, intervention groups with rapeseed oil not only significantly decrease the systolic blood pressure, but also reduce total cholesterol and LDL-cholesterol, and insulin levels in patients with metabolic syndrome.


More interestingly, serum triglycerides (TAG) associated with cardiovascular and nonfatty liver disease risk factors were significantly reduced only in the high ALA intake, compared to no effect on the low ALA group.

Based on the findings, researchers said, "Our dietary food pattern with a low ED and high intakes of MUFA and ALA may be a practical approach for long-term dietary treatment in patients with the metabolic syndrome".

Taken altogether, rapeseed oil may be considered a supplement for the prevention and treatment of metabolic syndrome, pending the confirmation of the larger sample size and multicenter human study.

Furthermore, at 16 μg/ml of naringenin, treatment organisms showed reduced expression of α-toxin significantly compared to the untreated group.

Moreover, in vivo, a mouse model of S. aureus pneumonia treated with naringenin demonstrated a significant effect in attenuating symptoms of lung injury and inflammation against infection.


In other words, naringenin acted as a natural antibiotic in preventing the overexpression of α-toxin produced by Staphylococcus aureus through its antibacterial activity.


Based on the findings, Dr. Zhang Y, the lead scientist after taking into account co and confounders wrote in the final report, "These results suggest that naringenin is a promising agent for the treatment of S. aureus infection".

Taken together, tomatoes may be considered supplements for the prevention and treatment of Staphylococcus aureus pneumonia, pending the confirmation of the larger sample size and multicenter human study.

Intake of naringenin in the form of a supplement should be taken with extreme care to prevent overdose and acute liver toxicity.

* LVLPGE and LAK, two novel peptides exhibited the highest ACE inhibitory activity associated with lowering hypertension with IC50 values of 13.5 and 48.0 μM, respectively.

* Moreover, the hypotensive effect of the peptides was assessed after oral administration to spontaneous hypertensive rats (SHRs).

Collectively, researchers said, "LVLPGE and LAK demonstrated a significant hypotensive effect in vivo. Protein from broccoli may thus constitute a potential antihypertensive peptide source".

Taken together, broccoli may be considered a functional food for the prevention and treatment of hypertension, pending the confirmation of the larger sample size and multicenter human study.
