Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.
In finding a natural compound for the treatment of oxidative stress effects in the pathogenesis of Alzheimer's disease (AD), researchers examined whether lycopene or lycopene/vitamin E could exert protective effects on memory deficit and oxidative stress in tau transgenic mice expressing P301L mutation.
The experiment included P301L transgenic mice randomly assigned to three groups: P301L group (P301L), P301L+lycopene (Lyc), and P301L+lycopene/vitamin E (Lyc+VE) with age-matched C57BL/6J mice as wild type controls (Con).
The P301L group showed significant spatial and passive memory impairments observed by elevated oxidative stress markers of malondialdehyde (MDA) levels and decreased levels of antioxidant enzymes produced by the host compared to the control.
According to neuro analysis, the P301L group also exhibited an increase of tau phosphorylation at Thr231/Ser235, Ser262, and Ser396 in the brain.
Supplementations of lycopene or lycopene/vitamin E administration significantly ameliorate memory deficits, by decreasing the levels of MDA concentrations and increasing the production of the GSH-Px activities.
Markedly, the lycopene or lycopene/vitamin E treatment groups also attenuated tau hyperphosphorylation at multiple AD-related sites.
These findings strongly suggested that lycopene or lycopene/vitamin E protects the integrity of the neurons against the increase of tau phosphorylation associated with neuronal cell dysfunction caused by oxidative stress through their antioxidant activity.
Based on the results, researchers wrote, " the combination of lycopene and vitamin E antioxidants acted in a synergistic fashion to bring significant effects against oxidative stress in tauopathies".
Taken altogether, lycopene found in tomatoes used alone or combined with vitamin E may be considered supplements for the prevention and treatment of cognitive deficits, pending the confirmation of the larger sample size and multicenter human study.
Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical #Indicaxanthin Incorporates Into the Redox Machinery of Beta-Thalassemia RBC and Defends the Cell from Oxidation Beta Thalassemia Anemia

Anemia is a medical condition associated with a lack of healthy red blood cells or hemoglobin in the blood.
Iron deficiency anemia is one of the most common anemia, caused by poor iron levels in the blood, leading to the reduced delivery of oxygen throughout the body.
People with a long-term deficiency of folate and vitamin B-12 which play a critical role in the production of healthy red blood cells are also at an increased risk of Amenia.
Patients with Beta-thalassemia minor have a slight lowering of the beta hemoglobin level in the blood similar to those with mild iron-deficiency anemia.
Beta thalassemia can be classified into
* Thalassemia minor, the mild case of anemia, is the most common anemia in people of Mediterranean descent. The blood cells of patients with thalassemia minor are smaller than normal blood cells and easily die off, causing not enough new blood cells to replace the red blood cell loss.
* Thalassemia major
Thalassemia major also called Cooley's anemia is an inherited disorder that affects the production of normal hemoglobin. Thalassemia major is a deadly disease without frequent blood transfusions, people with this kind of anemia may not survive because of iron building up in the heart and other organs, which can lead to heart failure.
The most common symptoms of beta-thalassemia are fatigue, weakness, or shortness of breath, a pale appearance or yellow color to the skin (jaundice), irritability, deformities of the facial bone, slow growth, a swollen abdomen, and dark urine.
There are many risk factors involved in the onset of the condition. Blood loss, such as from heavy menstrual bleeding, ulcer, cancer, and regular use of some over-the-counter pain relievers, especially aspirin are some of the most common prevalent risk factors.
Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.
The herb has been used in traditional medicine as an antioxidant, hypoglycemic, colorant, antiseptic, and wound healing agent, and to treat flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.
Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.
In finding a potential compound for the treatment of beta-thalassemia red blood cells, researchers examined the antioxidant activity of indicaxanthin.
In vitro, human beta-thalassemia RBCs submitted to oxidative hemolysis by cumene hydroperoxide, application of indicaxanthin at 1.0-10 microM enhanced the resistance to hemolysis associated with the destruction of red blood cells in a dose-dependent manner.
Furthermore, the phytochemical also exerted a significant antioxidant activity by preventing lipid and hemoglobin (Hb) oxidation and enhancing the levels of vitamin E and GSH depletion.
Moreover, indicaxanthin also stimulated the recovery of the soluble cell compartment of the RBCs in the blood from thalassemia patients.
According to the A spectrophotometric study, the antioxidant also showed to reduce the reactive intermediate that makes an abnormal form of hemoglobin in solution caused by the oxidative stress, compared to those of Trolox or vitamin C.
Based on the findings, researchers said, "Collectively our results demonstrate that indicaxanthin can be incorporated into the redox machinery of beta-thalassemia RBC and defend the cell from oxidation".
Taken altogether, indicaxanthin may be considered a remedy for the prevention and treatment of beta-thalassemia anemia, pending the confirmation of the larger sample size and multicenter human study.

In finding a potential compound for the treatment of inflammatory diseases researchers examined the effect of tomato ketchup extracts and bioactive from tomato ketchup on human monocytes and vascular endothelial cells (HUVEC).
HUVEC selected for the study were pre-treated for 1 h with either individual bioactive (7.5 µM lycopene, 1.4 µM α-tocopherol, or 55 µM ascorbic acid) or a combination of these three compounds, or with the hydrophilic or lipophilic tomato ketchup extracts or with the two extracts combined.
According to the tested assays, tomato ketchup extracts significantly reduced gene expression and pro-inflammatory cytokines TNF-α and IL-8 in HUVEC after inducing inflammation.
Furthermore, levels of anti-inflammatory release were increased observed by the expression of anti-inflammatory cytokine IL-10.
The effects were correlated with the reduction of IL-8 production in the presence of the test compounds and extracts.
In other words, lycopene found in tomato extract is one of the major bioactive compounds that induce the anti-inflammatory effect, and α-tocopherol and ascorbic acid appeared to strengthen the anti-inflammatory effect of lycopene.
In order to obtain more information about tomato anti-inflammatory activity, researchers analyzed the potential protective effects of phytochemicals in tomatoes on inflammation.
According to the nitric oxide (NO) assay, tomato extract exhibited an anti-inflammatory effect by inhibiting the NO production in LPS-stimulated RAW264 macrophages.
Levels of pro-inflammatory cytokines were significantly decreased after the injection of tomatoes.
The efficacy of tomatoes was attributed to the bioactive compound RF52 in the inhibition of the production of these proinflammatory mediators.

Hypertension is a chronic condition caused by abnormally high blood pressure.
High blood pressure is a condition of a cluster of metabolic syndrome associated with the increased risk of heart disease and stroke.
Blood pressure is the force of blood pushing against the walls of the arteries as the heart pumps out blood.
In high blood pressure individuals, due to the elevated blood pressure in your arteries, the heart has to work harder than normal to pump blood through the blood vessels.
In low blood pressure individuals, the reduced blood pressure in the arteries drops can cause the heart to pump blood at a slower rate than normal.
According to the standard guidelines, the levels of blood pressure are normal if it is more than 120 over 80 and less than 140 over 90 (120/80-140/90).
People with blood pressure lower than 120/80 are considered normal without being accompanied by symptoms of light-head or dizziness.

Twelve healthy subjects were subjected to distilling water inhalation for 10 min, and after one week they received inhalation of honey solution (60% wt/v) for 10 min. Another 12 healthy subjects received inhalation of 10% dextrose for 10 min.
Honey treatment showed a mild reduction of blood pressure compared to no change in another group.
From a time perspective, systolic and diastolic blood pressure was reduced by honey inhalation in hypertensive patients at 60 and 120 min.
After taking into account co and confounders, researchers said, "Honey inhalation was safe and effective in reducing...elevated blood pressure in hypertensive patients".
Moreover, in order to obtain more information on honey antihypertensive activity, researchers investigated the aqueous extract obtained from the mixture of the fresh leaf of Persea Americana, stems, and a fresh leaf of Cymbopogon citrus, fruits of Citrus medica, and honey on ethanol and sucrose-induced hypertension in rats.
Rats included in the study were divided into eight groups of 6 rats each and daily treated for 5 weeks. The control group received distilled water (1 mL/kg) while rats of groups 2, 3, and 4 received ethanol 40 degrees (3 g/kg/day), 10% sucrose as drinking water, and the two substances respectively.
The remaining groups received additional sucrose and ethanol, the aqueous extract (50, 100, and 150 mg/kg) and nifedipine (10 mg/kg) respectively.
The group treated with ethanol and sucrose showed a significantly (p < 0.001) increased blood pressure and heart rate compared to distilled water-treated rats.
Chronic treatment of ethanol and sucrose also exerted significantly decreased the activities of antioxidant enzymes produced by the host, observed by the levels of superoxide dismutase (SOD) and catalase (CAT) and elevated levels of oxidative stress observed by the stress marker malondialdehyde (MDA) levels.
However, administration of the aqueous extract or nifedipine prevented all changes caused by chronic ethanol and sucrose, including hemodynamic, biochemical, oxidative, and histological impairments.
