#Phytochemical Cucurbitacin B Isolated from Cucumber Combined with Cisplatin Enhance Chemotherapy on Laryngeal Cancer

Laryngeal cancer is a medical condition characterized by abnormal cell growth in the tissues of the larynx due to the alternation of cell DNA.
Larynx, also known as the voice box is a tube above the windpipe (trachea) in the neck and in front of the food pipe.

Most cases of laryngeal cancer start in the cell on the surface of the inner lining of the larynx, before penetrating into the deeper layers to form a tumor.

At the very early of cell alternation, patients are asymptomatic. However, as the tumor continues to grow, the cancer cell may infect other healthy tissues a distance away through the blood and fluids distribution.

Laryngeal cancer shares the general symptoms with other cancer including unintended pain, weight loss, loss of appetite, fatigue, and gastrointestinal discomforts accompanied by specific symptoms of a lump or swelling in your neck. a long-lasting cough, a persistent sore throat or earache, and difficulty breathing. in severe cases.

Sometimes, patients may vomit blood due to the oversize tumor which has pressed on the nearby blood vessels.

Out of many prevalent risk factors, smoking tobacco (cigarettes, pipes, cigars) is considered a major culprit that increases the risk of developing laryngeal cancer in the Western world.


Dr. A Altieri, the lead scientist wrote, "Cancer of the larynx is extremely rare in individuals who neither smoke nor drink alcohol (Austin and Reynolds, 1996). The risk rises with increasing levels of smoking and drinking, each agent approximately multiplying the effects of the other (Elwood et al, 1984; Blot, 1992; Dosemeci et al, 1997; Talamini et al, 2002). The risk of laryngeal cancer is 10–20-fold higher in current smokers than in non-smokers".

The cucumber plant is a species of Cucumis Sativus, belongs to the family Cucurbitaceae, and native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

With an aim to find potential and natural chemotherapy for the treatment of laryngeal cancer, researchers examined whether cucurbitacin B can also enhance the sensitivity of Hep-2 laryngeal cells to cisplatin.

The study included the evaluation of the proliferation, cell cycle distribution, and apoptosis in Hep-2 cells treated with either cucurbitacin B, cisplatin, or a combination.

According to the tested results, Hep-2 cells treated with a cucurbitacin B/cisplatin combination display synergistic effects on growth inhibition, cell cycle arrest, and apoptosis induction, compared to single-agent cucurbitacin B or cisplatin-treated cells.

Moreover, Hep-2 cell lines treated with the cucurbitacin B/cisplatin combination demonstrated a significantly reduced level of p-Stat3, compared to single-agent treated cells.

Where overexpression of p-STAT3 is an indication of a poor prognosis biomarker in cancer.

Additionally, cucurbitacin B/cisplatin-treated Hep-2 cells also demonstrated a significant reduction of the protein associated with regulation in the cell cycle and anti-apoptosis, compared to single-agent cucurbitacin B or cisplatin-treated cells.

Based on the results, researchers said, "These results suggest that the cucurbitacin B/cisplatin combination treatment may be a potentially useful therapeutic option for individuals diagnosed with laryngeal cancer".

In order to reveal more information about cucumber's anti-laryngeal cancer properties, scientists evaluated the combined antitumor effect of cucurbitacin B with docetaxel on Hep-2, a human laryngeal cancer cell line.

The combination of cucurbitacin B and docetaxel produced greater efficacy in growth inhibition, cell cycle arrest at the G2/M phase, and apoptosis induction similar to those of the aforementioned study compared to single-agent treatment.


The efficacy of the combined effect of cucurbitacin B and docetaxel anti-Hep-2 was attributed to the suppression of the expression of proteins involved in tumor proliferation and migration.

In vivo, the combination of cucurbitacin B with docetaxel also synergistically inhibited tumor growth in a mouse xenograft model.

Taken together, cucumber processed with a high amount of cucurbitacin B may be considered supplements combined with conventional medicine for the treatment of laryngeal cancer, pending the confirmation of the larger sample size and multicenter human study.



Intake of cucurbitacin B in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Moreover, ANDRO stimulated neurogenesis increasing the number of newborn dentate granule neurons in various parts of the brain.

The study also found that the efficacy of reduced symptoms and protection against the early onset of AD is the result of the stimulation of neurogenesis in the hippocampus.

Taking together, Andrographis paniculata is sure consisted of a therapeutic potential to protect the development of Alzheimer's disease through various mechanisms, however, intake of large amounts daily should be taken with care.

In 3T3-L1 adipocytes, the expression of adiponectin involved in regulating glucose levels as well as fatty acid breakdown was increased by 4.4 fold whereas hormone leptin found in high levels of obese subjects was decreased by 90% by the extracts.

Based on the findings, researchers said, " the analyzed extract may be a promising source of bioactive compounds that support long-term weight maintenance and promote the effective management of obesity".

Taken together, cranberry combined with other herbal medicine may be considered a remedy for the management of obesity, pending the confirmation of the larger sample size and multicenter human study.

Memory impairment can be acute or chronic.

In acute impairment, memory is a temporary loss in most cases associated with physical impact on the brain that causes injury of neurons.

The results clearly stated if you are a Western diet follower, you are at a higher risk of memory impairment compared to healthy diet individuals.

Ashwagandha also known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, the Middle East, and the Mediterranean. The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine for over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered an anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

In finding a potent compound for the treatment of brain health, researchers examined the efficacy and safety of ashwagandha (Withania somnifera (L.) Dunal) on memory and cognitive functioning in adults with mild cognitive impairment (MCI).

The prospective, randomized, double-blind, placebo-controlled study included 50 adults assigned to the treatment of either ashwagandha-root extract (300 mg twice daily) or placebo for eight weeks.

According to the analysis of Wechsler Memory Scale III subtest scores for logical memory, at week 8, the ashwagandha treatment group exerted significant improvements in both immediate and general memory, compared with the placebo group.

Futhermore, verbal paired associates I (p = 0.042), faces I (p = 0.020), family pictures I (p = 0.006), and logical memory II (p = 0.006) scores were also significantly increased compared to placebo.
According to the scores of Eriksen Flanker task (p = 0.002), Wisconsin Card Sort test (p = 0.014), Trail-Making test part A (p = 0.006), and the Mackworth Clock test (p = 0.009), ashwagandha treatment group also displayed a significantly greater improvement in executive function, sustained attention, and information-processing speed also compared to placebo.

Based on the findings, researchers said, "Ashwagandha may be effective in enhancing both immediate and general memory in people with MCI as well as improving executive function, attention, and information processing speed.".

Taken altogether, Ashwagandha may be considered a functional herb for the improvement of brain health, pending the result of larger sample size and multicenter study

Oral administration of alliin enhanced glucose homeostasis, increased insulin sensitivity, and improved the lipid profile in the DIO mic without affecting the body weight, adiposity, or energy balance.

In other words, alliin inhibited hyperglycemia selectively without normalizing the adiposity tissues' inappropriate responses of energy intake and energy expenditure.


Based on the tested results, researchers found that the efficacy of alliin in the inhibition of hyperglycemia is partly associated with the modulation of the intestinal microbiota composition, typically decreased Lachnospiraceae and increased Ruminococcaceae.


In order to reveal more information about alliin antihyperglycemic activity, researchers examined the production and therapeutic efficiency of alliin extracted from garlic leaves of plants grown under ex-situ and in-situ conditions.

According to the results of alliin aqueous leaf extracts (from ex-situ and in situ grown plants) were given to normal and alloxan-induced diabetic rats for five weeks, under in situ conditions, alliin extract lowered the serum glucose, triglycerides, total lipids, and total cholesterol, low-density lipoprotein (LDL)-, and very-low-density lipoprotein (VLDL)-cholesterol in diabetic rats by ~54%, 15%, 14%, 20%, 24%, and 15% compared to 35%, 14%, 10%, 12%, 17% and 11% from the ex-situ grown plants.

Furthermore, compared to glibenclamide, the leaf extract of plants lowered serum enzyme levels (alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase) associated with liver oxidative stress toward the normal.

Moreover, The histopathological alteration in the pancreas caused by alloxan was also inhibited markedly by leaf extract.


Based on the findings, Dr. Nasim SA, the lead scientist wrote, "These findings demonstrate leaf extract obtained from plants grown under in situ conditions possess higher therapeutic efficiency in comparison with leaf extract obtained from plants grown under ex-situ condition".

In other words, geographic growth may influence the therapeutic potential of phytochemicals in plants.

Taken together, alliin may be considered a supplement for the prevention and treatment of hyperglycemia, depending on the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Thyroid disease is a medical condition that causes a malfunction of the thyroid.

Hyperthyroidism is a condition in which the thyroid gland is overactive and produces too much thyroid hormones.

* Sulforaphane did not affect the serum levels of thyroid-stimulating hormone, free thyroxine, and thyroglobulin.

* More importantly, sulforaphane does not affect thyroid autoimmunity status.

Based on the findings, researchers said, "These results provide evidence in favor of the safety of chemoprevention strategies that target the activation of Nrf2 to protect against environmental exposures and other oxidative stress-related pathologies".

Taken altogether, Broccoli sprouts may is safe for the prevention and treatment of oxidative stress-related diseases without causing thyroidal toxicity, pending to the confirmation of the larger sample size and multicenter human study.
