Chaste tree berry is a species of Vitex agnus-castus, genus Vitex, belonging to the family Lamiaceae, native to the Mediterranean region, and used in herbal medicine for thousands of years as an aphrodisiac herb. In a self-administered questionnaire, containing 15 questions, sent to all gynecologists in private practice in Germany to evaluate the complementary and alternative medicine (CAM) in the treatment of climacteric disorders, 98% of the returnees are experienced with therapies of black cohosh (Cimicifuga racemosa), chaste tree (Vitex agnus castus) and St. John's wort(1).
According to the 1Royal Melbourne Institute of Technology (RMIT) University, Bundoora, although evidence from rigorous randomized controlled trials is lacking for the individual herb emerging pharmacological evidence supports a role for V. agnus-castus in the alleviation of menopausal symptoms(2).
Dr. Chopin Lucks B. in the trial of two essential oils (derived separately from leaf and fruit) of Vitex agnus castus indicated that both essential oils show a strong symptomatic relief of common menopausal symptoms(3).
Unfortunately, according to the University Hospital Hamburg-Eppendorf, Agnus castus showed no significant difference in the treatment of climacteric complaints(4) with a combination of Hypericum perforatum (St. John's wort) and Vitex agnus-castus (Chaste tree/berry) also showed the same(5).

#Quercetin Aglycone and PAC DP-9 From #Cranberry Flavonoids Inhibit the Onset of #OvarianCancer, Researchers Suggest

The ovaries formed part of the female reproductive system are the oval-shaped ovum that connects to the fallopian tubes and attaches to the outer layer of the uterus via the ovarian ligaments.
Normally, the left and right ovary take a turn to release an egg every month. The ovaries also play a critical role in the production of estrogen and progesterone in the regulation of the process of women menstrual cycle including the release of an egg that migrates down the fallopian tube.
The most common disease associated with ovary dysfunction is polycystic ovary syndrome (PCOS), a hormonal disorder among women of reproductive age.
Ovarian cancer is a chronic and medical condition characterized by abnormal cells growth of ovarian cells which start on the surface of the inner lining of the ovarian tissue. Over time, after penetrating into deeper layers in the ovary, the cancer cell may travel a distance away to infect other nearby healthy tissues or organs, causing secondary metastasis.
Ovarian cancer is one of the most common cancers in the US. According to the statistics adapted from the American Cancer Society's publication, Cancer Facts & Figures 2010, approximately 21,880 women in the United States were diagnosed with ovarian cancer. In the same year, ovarian cancer also causes the death of 13,850 American women.
The exact causes of ovarian cancer are not identified. However, epidemiologically, aging, genetic preposition, race, never pregnant women, family history, long-term chemical exposure, and menopausal hormone therapy MHT are some prevalent factors found in patients with the disease.
Dr. Prof. Rod Baber wrote, "Increased risk persisted after stopping the treatment for some years among women who had used MHT for more than 5 years".
And, "Overall, this increased risk was similar for European and North American perspective studies and for both estrogen-only and estrogen plus progestogen preparations but differed across the subtypes of epithelial ovarian cancers, being increased only in serious and endometrioid subtypes and decreased in mucinous and clear cell subtypes".
Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.
In finding a potential compound for the treatment of ovarian cancer researchers examined the effect of cranberry flavonoids (flavonols and flavan-3-ols) SKOV-3 and OVCAR-8 ovarian cancer.
According to the tested assays, treatment with quercetin aglycone and PAC DP-9 from the cranberry flavonoids exhibited the strongest activity that induces apoptosis compared to other bioactive compounds.
The combination also inhibited the caspase-3 activation and PARP deactivation associated with apoptosis and increased the cisplatin sensitivity against the tested cell lines.
In SKOV-3 cells, PAC DP-9 also reduced the expression and activation of the epidermal growth factor receptor (EGFR) associated with cancer cell differentiation and proliferation.
Furthermore, quercetin aglycone and PAC DP-9 deactivated MAPK-ERK pathway and the overexpression of proteins associated with the promotion of cancer cell growth.
Moreover, the combination also promotes the gene expression of cancer suppression and arrested cell cycle progression.
Based on the findings, researchers said, "Overall, this study demonstrates promising in vitro cytotoxic and anti-proliferative properties of two newly characterized cranberry flavonoids, quercetin aglycone, and PAC DP-9, against ovarian cancer cells".
Taken together, cranberry-processed abundantly quercetin aglycone and PAC DP-9 may be considered a remedy for the prevention and treatment of ovarian cancer, pending to the confirmation of the larger sample size and multicenter human study.

#Carotenoid Intake Regularly and Daily Is Associated with Beneficial Skeletal Effects in Women with Osteoporosis, Researchers Suggest

Carrots may have a profound and positive effect on improving bone health, some scientists suggested.
Osteoporosis is a bone disease characterized by the inability of the body to produce bone to make up for bone loss, leading to the loss of bone density over a long period of time.
People with low bone density are susceptible to fracture and break from a fall due to bone weakening.
The exact causes of osteoporosis are unknown. However, researchers do know that certain preventable risk factors such as smoking, low calcium intake, physical inactivity, excessive alcohol consumption, and unpreventable risk factors such as aging, family history, gender, race, body frame size,... and sedentary works are associated to increase the onset of the disease.
Some researchers also suggested that loss of bone density due to age, medications used to treat breast cancer, arthritis, asthma, and certain chronic diseases such as Crohn's disease and Addison's disease and hyperthyroidism, hypopituaritism and eating disorders are also considered as some major risk factors of osteoporosis.
According to the statistic provided by the National Osteoporosis Foundation, over 10 million Americans are affected by osteoporosis and 34 million more have low bone mass, called osteopenia.
The most common symptoms of osteoporosis include back pain, as a result of a fractured or collapsed vertebra, loss of height, and a stooped posture.
Many patients with osteoporosis also experience symptoms of a bone fracture.
If you experience some of the symptoms above, you may have osteoporosis.
Conventional medicine such as Bisphosphonates: Fosamax, Actonel, Boniva, and Zoledronic Acid Bisphosphonates may be effective in reducing the risk of fracture, decelerating bone loss and ameliorating pain. In a total of 59 postmenopausal women participated, researchers found that
* Intake of carotenoids scores a significant effect of protecting the bone density, observed by the dual-energy x-ray absorptiometry, and osteoporosis diagnosis was based upon T-scores.
* Serum lycopene and beta-cryptoxanthin measured by 0.03 and 0.006, respectively in participants were correlated to the total intake of fruits and vegetables.
* However, serum alpha-carotene concentration was strongly associated with carrot intake.
Researchers, after taking into account other co and confounders suggested that carotenoid intake may have beneficial skeletal effects in women with osteoporosis.
Additionally, in vitro and in vivo studies suggest that carotenoids may have a strong protective effect of inhibited bone resorption, according to researchers at the Jean Mayer USDA HNRCA at Tufts University in differentiation of associations of total and individual carotenoid intake (alpha-carotene, beta-carotene, beta-cryptoxanthin, lycopene, lutein + zeaxanthin) with incident hip fracture and nonvertebral osteoporotic fracture.
Furthermore, a total of 370 men and 576 women (mean age, 75 +/- 5 yr) from the Framingham Osteoporosis Study completed a food frequency questionnaire (FFQ) in 1988-1989 and followed for hip fracture until 2005 and nonvertebral fracture until 2003, researchers showed that
* Highest tertile of total carotenoid intake had a lower risk of hip fracture (p = 0.02).
* Higher lycopene intake had lower risk of hip fracture (p =0.01) and nonvertebral fracture (p = 0.02).
* However, total beta-carotene was associated with a weak protective trend for hip fracture alone, and NO significant associations were observed with alpha-carotene, beta-cryptoxanthin, or lutein + zeaxanthin.
These results suggested that only lycopene was found to have a clinical effect on the protection of bone health in all aspects compared to other bioactive compounds isolated from carrots.
And several members of carotenoids may play a protective role in bone health in older adults.
Moreover, in the reassessed the carotenoids bone protective effect in inhibited bone resorption and stimulate proliferation and differentiation of osteoblasts, in 334 men and 540 women (mean +/- SD age: 75 +/- 5 y) in the Framingham Osteoporosis Study, researchers at the Tufts University showed that intake of lycopene may protect the mean of bone density in both genders
* Carotenoid and lycopene intake exerted a 4-y change in lumbar spine BMD for women, and
* Total carotenoids, beta-carotene, lycopene, and lutein+zeaxanthin with 4-y change in trochanter BMD in men.
DR. Sahni S, the lead scientist said, "Carotenoids showed protective associations against 4-y loss in trochanter BMD in men and in the lumbar spine in women" and " These results support a protective role of carotenoids for BMD in older men and women".
Taken together, carrot and its bioactive compound lycopene may be considered as functional foods in reducing the risk of osteoporosis, including inhibited bone resorption and proliferation and differentiation of osteoblasts,... in both older men and women.

The herbal plant has been used as foods and herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.
Researchers on finding a replacement for the treatment of lung cancer with no side effects examined the berries' bioactive compound anthocyanidins and its derivative cyanidin, malvidin, peonidin, petunidin, and delphinidin anticancer effects.
A mixture, including blueberry, bilberry and Indian blackberry ('Jamun') was selected to test in two aggressive non-small-cell lung cancer cell lines.

Dr. Kausar H and colleagues said, in the final report "Our results thus demonstrate the therapeutic potential of berries rich in this mixture of diverse anthocyanidins for non-small-cell lung cancer treatment and to prevent its future recurrence and metastasis".
In order to reveal more information about bilberry anti-lung cancer activity, researchers assessed the involvement of oxidative stress in lung adenocarcinogenesis, using mice deficient in the 8-hydroxyguanine DNA glycosylase 1 (Ogg1) gene and wild-type mice (C57BL6/J origin) administered 4-(N-hydroxymethylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK).
Where deficiency of 8-hydroxyguanine DNA glycosylase 1 (Ogg1) gene was found to associate with the advanced stage of cancer.

#Bromelain Inhibits Inflammation and Destructive Arthritis-Associated Changes in Rheumatoid Arthritis, Researchers Say

Rheumatoid Arthritis (RA) a chronic disorder as a result of inflammation, affecting mostly the flexible (synovial) joints and tissues and organs in the body.
The disease affects more women than men and generally occurs after the ages of 40, causing the diminished quality of life of many elders(1).
According to CDC, Musculoskeletal disorders (MSDs) affect over 52 million of adults in the US alone, including 294,000 children under age 18 with some form of arthritis or rheumatic conditions(2).
Rheumatoid Arthritis can induce bone loss through elevating bone resorption without increasing bone formation(4). A cross-sectional population-based study of 1042 patients with rheumatoid arthritis showed that RA patients had an increased risk of death from various causes(4a).
The most common symptoms of RA are
* The feet and joints
Patients with rheumatoid arthritis (RA) often associate with valgus deformities of the feet and gait due to body function in an effort to support the collapsing longitudinal arch of the foot.
* The gait
Rheumatoid arthritis (RA) causes a significant reduction in joint motions, joint moments, such as decreased hip flexion-extension range, hip abduction, knee flexion-extension range.

With an aim to find a natural agent for the treatment of adjuvant rheumatic diseases, researchers examined the anti-inflammatory property of both enzyme therapy and combined therapy with cyclosporin in an animal model.
The study included rats with adjuvant-induced arthritis administered either cyclosporin A (2.5 or 5.0 mg/kg/day per os), a mixture of enzymes (Phlogenzym (PHL); 45 mg/kg twice daily intrarectally), or a combination of 2.5 mg cyclosporin A and 90 mg PHL for a period of 40 days.
According to the tested assays, the combination therapy with cyclosporin A plus PHLshowed a significantly inhibited both the inflammation and destructive arthritis-associated changes, such as serum albumin, changes in hind paw swelling and bone erosions.
Compared to the higher dose, 2.5 mg of cyclosporin A and PHL alone inhibited the disease markers to a lesser extent and a later stage of arthritis development.
In other words, the combination inhibited all parameters associated with the onset of adjuvant rheumatic diseases in a dose-dependent manner.
Based on the findings, researchers wrote, "The results show the inhibitory effect of enzyme therapy on rat adjuvant arthritis, as well as the efficacy of a low dose of cyclosporin A given in combination with enzyme therapy, which may be useful in the treatment of rheumatoid arthritis".
Taken altogether, bromelain used alone or combined with conventional medicine may be considered a remedy for the prevention and treatment of adjuvant rheumatic diseases, pending to the confirmation of the larger sample size and multicenter human study.
Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

In the study of gastric ulcers induced by ethanol, aspirin, pylorus ligation, and cold restraint stress models, rats treated with rats pretreated with HAEAP (100,200,500mg/kg b. wt for 30 days) of Andrographis paniculata expressed a significant ulcer reduction, in a dose-dependent manner, according to the Bharathi Women's College.
Dr. Panneerselvam S, the lead author said, "The ulcer-preventing effect of HAEAP may partly be due to its regulating effect on H(+)K(+)ATPase activity and /or mucin preserving effects" and "The flavonoids present in the HAEAP might be responsible for the gastroprotective action probably by maintaining the antioxidants and thiol status in the gastrointestinal tract".
In support of the above differentiation, the review of a period of 1 year from 2014-2015, using semi-structured questionnaires from different traditional Bengali people having knowledge of medicinal plants, out of 49 plants under 46 genera belonging to 33 families, Andrographis paniculata is one of the most prominent plants used among the people of Southern Assam for treating digestive system disorders (DSD).
Furthermore, seed ethyl acetate, acetone, and methanol extracts of Andrographis paniculata (Burm.f.) Wall. ex Nees has drawn a lot of scientists' attention due to the limited success of chemical anthelmintics, a group of antiparasitic drugs (levamisole, flubendazole, and thiabendazole) in reducing gastrointestinal diseases.
In animal controls, ED(50), an ethanol extract of A. paniculata at a dose of 2.90 mg/ml, and in larval development assay at ED(50) of dose 4.26 mg/ml, showed significant effects against sheep parasite, Haemonchus contortus.
Taken together, there is no doubt that Andrographis paniculata consisted of a strong gastroprotective effect against the activities of harmful organisms, including parasites, and may be used as a functional food in improving digestive functions.
