#NavyBean Inhibits #Genes and #Proteins Associated with Acute-Inflammative Diseases in Vivo, According to Studies

Kyle J. Norton

Inflammation is a natural response by the immune system to protect the body against the invasion of stimuli or foreign pathogens.

Acute inflammation is a short-term condition caused by tissue injury, leading to pain, redness, swelling, and heat in the affected area. depending on the size of the injury. Some patients with acute inflammation may also experience the symptom of loss of function.

In acute inflammation, the white blood cells on the first line of defense of the immune system after sensing the danger of an infection in the body through a message from the brain stimulate the production of platelets to cover the site of injury or damage and inflammatory cytokines to kill off all pathogens before they can cause harm to the body.

Most cases of infection are stopped at the acute phase. The wound or injury is slowly recovered.

However, in people with a weakened immune system, acute inflammatory progression cannot be stopped due to the inability of the immune system to destroy all invaders, which may lead to low-grade chronic inflammation.

Anti-inflammation is the process that inhibits inflammation or swelling without affecting the immune property to curb foreign pathogens.

Conventionally, the most common use of anti-inflammatory medicines is over counter aspirin, ibuprofen (Advil, Motrin, Nuprin), and other prescription NSAIDs.
Holistically, according to the Medical University Innsbruck, the top 5 anti-inflammatory foods are
* Garlic
Recent studies also showed that garlic exhibits anti-inflammatory effects against chronic inflammatory disease through phytochemical allicin and other mechanisms.

* Ginger
Strong evidence in Western studies suggested that ginger processes anti-inflammatory, antioxidant and immune-modulatory effects.

* Turmeric
Epidemiological studies also found the efficacy of turmeric for the treatment of diabetes, microbial infection gastrointestinal diseases, and irregular cell growth such as cancer through its anti-inflammatory antioxidant and immunomodulatory activities.

* Green tea
Epidemiological studies suggested that green tea consists of many pharmaceutical properties, including anti-cancer, anti-diabetes, anti-allergy, and anti-micro-organisms infection, through its antioxidant, anti-inflammatory and immune-modulatory activities.

* Shiitake mushrooms
Recent studies showed that Shiitake mushrooms process the properties of anti-cancer, anti-HIV, and hepatitis virus, an enhanced immune system against inflammation.

The small, white navy bean, also called pea bean or haricot, popular in both dry and green forms, is very popular in Britain and the US, native to Peru. It is now grown and consumed across the world, due to its numerous health benefits.

With an aim to find a potential ingredient for diseases associated with acute inflammation, researchers examined the enriched levels of nondigestible fermentable carbohydrates and phenolic compounds found in black and navy beans' immunomodulatory effects on the severity of colitis-associated inflammatory pathology.

In an animal model, C57Bl/6 mice were pre-fed isocaloric 20% cooked navy bean (NB) or black bean (BB) diets for 3 weeks and switched to control basal diet (BD) 24 h prior to colitis induction via 5-day exposure to dextran sodium sulfate (2% w/v in drinking water)+3 days of freshwater, researchers found that* The severity of the acute colitis phenotype associated with inflammation was inhibited by bean prefeeding, observed by reduced colon tissue inflammatory transcription factor activation such as NFκB, STAT3) and inflammatory mediator levels such as IL-1β, IL-6, IL-18, and MCP-1 compared to control.

* Black and navy beans also stimulated the biomarkers that enhance wound repair responses. including colon tissue protein levels of IL-22, and IL-27.

* Furthermore, the black and navy beans diet also increases the mRNA expressions of genes involved in the normal colonic epithelial function and the promotion of epithelial barrier integrity, defense and/or restitution, and wound closure in acute colonic inflammation.

Based on the findings, researchers concluded, " bean supplementation prior to colitis induction (i.e., mimicking disease relapse) primes the colonic microenvironment to attenuate the severity of the colitis inflammatory phenotype and maintain aspects of epithelial barrier function".

Taken together, the navy bean may be considered a remedy for the prevention and treatment of acute inflammation, pending the confirmation of the larger sample size and multicenter human study.

#Phytochemical #Sinigrin Induces Genotoxicity and Proliferation Against #Cancer Cells, According to Studies

Kyle J. Norton

Sinigrin may be used as a natural and bioactive compound for killing cancer cells before they can spread, according to studies.
Cancer is a group of diseases characterized by cell growth irregularly in the tissue of the body, including glands and organs due to the alternation of cell DNA.

In normal cells, through the process of mitosis, new cells are born through the division of one cell into two with an aim to replace cells that have been lost, damaged, or worn out. The cell division cycle is closely regulated and they can give rise to only a limited array of cells.

The characteristics of cancer cell growth are completely different compared to normal cells. The cancer cell can divide indefinitely; They are out of control by the immune system. They don't follow the signals of stop division,.... and they do not have a specific function.

How cancer grows? Most cancers stimulate the production of a new blood vessel for nutrition and the function of producing enzymes that break down normal cells and tissues.

All cancers share the general symptoms of unintended weight loss, loss of appetite, fatigue, tiredness, and gastrointestinal discomforts accompanied by specific symptoms depending on the location of the cancer or the organ that was affected.

Out of a number of risk factors, some researchers suggested the creation of obesity in the modern world may be one of the major risk factors associated with the incidence.

Dr. Nathan A. Berger in the evaluation of obesity and risk of cancer wrote, "Overweight and obesity have reached pandemic levels on a worldwide basis and are associated with increased risk and a worse prognosis for many but not all malignancies".

And, " lessons learned from the use of murine models to study the association, the impact of obesity on pancreatic cancer, the effect of dietary fats and cholesterol as cancer promoters, and the mechanisms by which the intestinal microbiome affects obesity and cancer".

The results showed a relationship between the unhealthy diet risk of obesity in the induction of cancer.

Sinigrin is a phytochemical glucosinolate, that belongs to the family of glucosides found abundantly in Brussels sprouts, broccoli, the seeds of black mustard, etc.

Researchers on finding a natural compound for the treatment of cancers examined sinigrin, the bioactive ingredient in cruciferous vegetables for their health benefits and cancer prevention properties in vitro against HL60 (human promyelocytic leukemia cell line) systems.

According to the HL60 assays, sinigrin showed the tumoricidal activity of plant samples (IC50 = 0.28 mg·mL(-1)) by breaking down the products of sinigrin hydrolysis.

Sinigrin also exerted a significant effect in the induction of tumor genotoxicity and proliferation causing harm to the nearby healthy cells.

The finding strongly suggested that the efficacy of cruciferous vegetables in inducing cancer cell death and proliferation was attributed to the presence of the bioactive ingredient sinigrin.

Furthermore, in a model of carcinogen-induced hepatotoxicity in rats, injection of sinigrin significantly inhibited the proliferation of liver tumor cells without inducing liver toxicity.

Injection of the compound for a period of 3 months exhibited pro-apoptotic activity against liver cancer by activating the tumor-suppressive gene in the rats.

The active bioactive compound also inhibited cancer cell proliferation by deactivating the production of proteins associated with liver cancer cell differentiation.

The effect of sinigrin against liver cancer is also found to cancer cell cycle arrest in the G0/G1 phase.

The results clearly suggest that sinigrin processes important anti-proliferative activities in carcinogen-induced hepatocarcinogenesis in rats.

Taken altogether, sinigrin a bioactive ingredient in cruciferous vegetables may be considered a functional supplement for the prevention and treatment of cancer, pending to the confirmation of a large sample size and multicenter human study.

However, intake of bioactive in the form of supplements should be taken with extreme care, as liver toxicity may be induced if overdosed.

#Cranberry Supplementation Reduces Aβ toxicity induced Brain Cell Stress Agaisnt the Onset of #Alzheimer's disease, Reseracher Say

Kyle J. Norton

Amyloid beta (Aβ or Abeta) is the main component of the amyloid plaques that cause the gradual death of neurons, leading to the onset of Alzheimer's disease.
Alzheimer's disease is a common form of dementia or neurodegenerative disease found in the elderly.

Alzheimer's disease is a type of dementia correlated with major reductions of neurons to the respective target areas, named after German physician Alois Alzheimer.

According to the statistics, the disease is most prevalent in the elderly. Approximately 5.7 million Americans of all ages are living with Alzheimer's. 15.9 million family caregivers provide an estimated 18.2 billion hours and $230 billion to people with dementia worldwide.

Alzheimer's disease is caused by a combination of genetic, lifestyle and environmental factors that affect the neurons of the brain over time. However, less than 10% of patients with AD are associated with a genetic preposition. Patients with such mutated genes on chromosomes 21, 14, and 1are virtually guaranteed to develop the disease.

More precisely, Alzheimer's disease is caused by the gradual loss of neuron functioning that usually starts with the early symptom of the difficulty of remembering recent events and worsens over time.

Alzheimer's destroys brain cells, causing cognitive modalities severe enough to affect language communication, memory, lifelong hobbies or social life. Alzheimer's gets worse over time, and it is fatal.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia.

Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of mental disorders, researchers examined the effect of cranberry supplementation on β-amyloid aggregation.

According to tested analysis, the cranberry protected the neurons by mediating Aβ toxicity alleviation required heat shock transcription factor (HSF)-1 rather than DAF-16 and SKN-1 associated with cellular stress.

In other words, cranberry supplementation reduced Aβ toxicity induced brain cell stress by reducing the transactivity of HSF-1 in an IIS-dependent manner.

More precisely, HSF-1 plays a pivotal role in maintaining proteostasis, a process that regulates proteins within the cell in order to maintain the neuron cellular health involved in oxidative stress.

Based on the findings, researchers aid, "(cranberry) elucidated the mechanism whereby cranberry attenuated Aβ toxicity in C elegans and stressed the significance of proteostasis in the prevention of age-related diseases from a practical point of view.".

Taken altogether, cranberry may be considered a remedy for the treatment of AD, pending to the confirmation of the larger sample size and multicenter human study.

#Phytochemical #Alliin Supplement Exhibits Neuro-Protection through Antioxidant Effects in Vitro, According to Researchers

Kyle J. Norton

Neuroprotection is an action induced by the preservation of neuronal structure and/or function, including the use of medicines and herbs.

In other words, neuroprotection protects neuronal integrity against the reduction in the rate of neuronal loss over time, particularly in neurodegenerative diseases.

Neurodegenerative disease is the class of conditions associated with the progressive loss of neurons occurred mostly in older adults, including dementia, with the most common being Alzheimer`s disease (AD).

According to the statistic, dementia affects over 47.5 million people worldwide, approximately 58 percent are living in low and middle-income countries.

Believe it or not, 5.8 million Americans of all ages are living with Alzheimer's dementia in 2019 and two-thirds of people with Alzheimer's disease are women.

Conventionally, there is no cure for dementia. The purpose of the medications that are used for the treatment of dementia is to improve dementia symptoms and slow down the progression of the disease.

Some researchers are concern that Alzheimer's disease in people younger than the age of 60 was up 5 percent over the past few decades may be correlated to the promotion of a high-fat diet in the US.

Dr. Ruan Y, the lead scientist wrote, "A higher dietary saturated fat intake was significantly associated with an increased risk of 39% and 105% for AD (RR: 1.39; 95% CI: 1.00, 1.94) and dementia (RR: 2.05; 95% CI: 1.06, 3.98), respectively".

And, "Dose-response analysis indicated a 4 g/day increment of saturated fat intake was related to 15% higher risk of AD (RR: 1.15; 95% CI: 1.01, 1.31)".

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide, a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

In the urgency to discover a natural and potential compound for the treatment of neurodegenerative disease, scientists examined the alliin neuroprotective activity.

In vitro, alliin exerted a significant effect to protect neuronal cell integrity and viability against beta-amyloid (A) toxicity and apoptosis.

Furthermore, also in vitro, alliin inhibited oxidative stress caused by overexpression of reactive oxygen species (ROS) in cells in facilitating several neurodegenerative diseases including Alzheimer's disease (AD).

In lipopolysaccharide- (LPS-) stimulated 3T3-L1 used in biological research on adipose tissue, incubation in a period of 24 h together with 100 μmol/L alliin, alliin prevented the increase of proinflammatory cytokines in the liver, muscle, heart, and brain cells through its antioxidant activity.

The inhibition of inflammation of alliin was attributed to the decreased expression of the ERK1/2 pathway involved in the expression of oxidative stress and inflammation.

In other words, alliin protects the brain cells by suppressing the LPS inflammatory signals by generating an anti-inflammatory gene expression.

Taken together, alliin may be considered a supplement for neuroprotection, depending on the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthyfood #BlackBeans Exhibit Anti-Proliferative Activity Against Cancers through Their Antioxidant Effects, Researchers Suggest

Kyle J. Norton

Intake of black(common) beans and other common beans may associate with reduced risk and treatment of various types of cancers, a study of renowned institute study suggested.

Cancer is a class of chronic conditions characterized by irregular cell growth in the body's organs. Sometimes, the cancer cells can spread to other tissues or organs depending on the stages of cancer.

Black bean, a small roughly ovoid legume with glossy black shells, genus Phaseolus, belongings to the family Fabaceae can be bought in most grocery stores all around the year in dried and canned forms. It is believed that the black bean was the first domesticated growth in South America.

Fermentation products of soybean, black bean, and green bean mixture (BN999), showed a significant effect in inhibiting the growth of human breast cancer AU565 cells and prostate adenocarcinoma PC-3 cells without harming the normal human cells.

The study also found that in syngenic BALB/c mice, the mixture(BN999) inhibited the growth of mouse CT-26 colon cancer xenografts, without causing adverse effects.

Furthermore, BN999, also induced cancer cell death in other mechanisms, such as activating the protein gene involved in pro-cancer cell apoptosis via cell mediatory activities, including a network of membranous anatomical structure and double membrane-bound organelle.in energy conversion.

Additionally, according to the joint study led by the Universidad Autónoma de Querétaro, the efficacy of common bean in attenuated cancer risk factors is shown in animal models and in cell cultures. may also be attributed to other parameters of non-digested compounds like fiber, phenolics, peptides, and phytochemicals.

In fact, black (common) bean whole and non-digestible fraction also exhibited anti-proliferative activity and induced apoptosis in vitro and in vivo against colon cancer.

Dr. Rocio Campos-Vega lead author in the joint study also raised the important role of bean fiber and protein of the bean in ameliorated cancer risks, saying, " the potential health benefits and bioactivity of beans on tumor inhibition, highlighting studies involving functional compounds, mainly non-digestible fractions that modulate genes and proteins, thereby, unraveling their preventive role against the development of cancer".

Other researchers in the evaluation of phytochemicals of black beans against various types of cancers showed that flavonoids and saponins isolated from black beans exerted a strong antioxidant capacity and antiproliferative activity against numbers of cancer cell lines in a concentration-dependent manner.

The study also insisted that depending on days of germination, total phenolic content extracted from seed coats at the 3 days exhibited the strongest effect in inhibiting all cancer cell lines proliferation with no cytotoxicity against control cells.

Specifically, the chemical compound genistein showed the most effectiveness against mammary cancer cells compared to more related to hepatic and colon cancers in flavonols and saponins.

The findings suggested that black beans may be considered a functional food as well as an integrated form of injection for reduced risk and treatment of various types of malignant tumors

#Bromelain Inhibits Alzheimer's disease by Preventing A Beta Accumulation in the Brain and #Vasculature, According to Studies

Kyle J. Norton

Alzheimer's disease (AD) is a neurodegenerative disease correlated with major reductions of neurons to the respective target areas, named after German physician Alois Alzheimer.

Alzheimer's destroys brain cells, causing cognitive modalities severe enough to affect language communication, memory, lifelong hobbies, or social life. Alzheimer's gets worse over time, and it is fatal.

According to statistics, over 25 million people in the world today are affected by dementia and most are suffering from Alzheimer's disease. In Canada, approximately, over half a million Canadians live with dementia accompanied by the 25,000 new cases diagnosed every year.

Believe it or not, in 2016, dementia cost Canada over $10 billion, including the healthcare system and out-of-pocket caregiver costs.

Epidemiologically, cigarette smoking, midlife high blood pressure, and obesity, diabetes, cerebrovascular lesions, and psychosocial factors (eg, high education, active social engagement, physical exercise, and mentally stimulating activity) are associated with prevalent risk of AD development.

Some researchers in evaluated that the increased AD in older adults in the Western world over the past few decades may be correlated to the promotion of a high-fat diet.

Dr. Peter Thériault, the lead researcher wrote, "WD promotes systemic inflammation by increasing the levels of blood-circulating monocytes and monocyte chemotactic protein-1 production, which is accompanied by an augmentation of oxidized-low density lipoprotein levels in blood circulation".

And, "At the blood-brain barrier (BBB), WD potentiates the age-induced increase of Aβ 1-40 accumulation and exacerbates the oxidative stress, specifically in cerebral microvasculature. These effects were accompanied by the dysfunction of pericytes, thus altering BBB functionality without compromising its integrity".

In other words, if you are a high-diet follower, you are at high risk of AD developing.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

In finding a potential compound for the treatment of Alzheimer's disease, researchers investigated the ability of bromelain, a plant cysteine protease derived from pineapple stems interact with synthetic Aβ42 monomers and oligomers.

According to the tested assays, bromelain protected the neuron successfully by degrading Aβ42 monomers and low and high-molecular-weight oligomers.

Examined the effects of bromelain preparations of 3 and 6 mU added to the cerebrospinal fluid (CSF), the levels of Aβ42 monomers and oligomers were significantly reduced, compared to that of the same standard Aβ42 preparations incubated in CSF without bromelain

Furthermore, the concentration of bromelain incubations with 0.1, 0.5, and 1 mU/ml was not toxic to the central nervous system.

Based on the findings, researchers said, "These results represent an important insight into the action of bromelain on Aβ42 oligomers, suggesting its potential use in the therapy of AD".

In order to know more information about bromelain anti-Alzheimer's disease activity, researchers examined a beta-degrading activity of diverse alpha 2-macroglobulin (alpha 2-M)-proteinase including trypsin, alpha-chymotrypsin, and bromelain (125)I-A beta 1--42.

The complex showed a strong degradation of (125)I-A beta 1--42.

In Ex vivo, oral administration of high doses of proteinases displayed an enhanced degradation of (125)I-A beta 1--42 in the blood of volunteers.

In other words, the complexes exhibited a significant A beta catabolism, thus reducing the risk of developing AD by preventing A beta accumulation in the brain and vasculature.

Taken altogether, bromelain used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of Alzheimer's disease, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Betanin Processes #LungCancer Inhibitory Effects Involving #Apoptosis and #Angiogenesis, According to Studies

Kyle J. Norton

The lungs are a pair of spongy, air-filled organs located on either side of the chest with the main function to process gas exchange during breathing.

In other words, the lung transfer life-giving oxygen into the blood supply and remove carbon dioxide produced by cells metabolism from the blood.

Additionally, the lung is also the first-line defense against inhaled particles and prevent the circulation of emboli such as blood clot (thrombus), fat globule (fat embolism), a bubble of air or other gas (gas embolism), or foreign material.

Lung cancer is a medical condition characterized by irregular cell growth in the lung as a result of the alternation of cells in the lung tissue.

Most cases of lung cancer start in the cell on the surface of the inner lining of the lung tissue, before proliferating into deeper layers to induce a tumor.

At the early stage, due to the very small size of the tumor, most patients are asymptomatic. However, as the proliferation continues over time, as the later stage, the primary lung cancer not only can elevate the general symptoms shared by all cancers accompanied by localized specific symptoms such as severe pain and bleeding but also travel a distance away from the lung to cause secondary metastasis via circulation of blood and fluids.

The most common secondary metastatic cancers from primary lung cancer are the adrenal gland, bones, brain, and liver.

Out of many risk factors associated with lung cancer, smoking has been found to have a strongly negative implication to induce the early onset of the disease.

Dr. Ozlü T, the lead scientist said, "Nowadays, around one-third of adults are known to be smokers, and smoking rates are increasing among the female population. It is estimated that deaths attributable to tobacco use will rise to 10 million by 2025, and one-third of all adult deaths are expected to be related to cigarette smoking".

And, " The association between cigarettes and lung cancer has been proven by large cohort studies. Tobacco use has been reported to be the main cause of 90% of male and 79% of female lung cancers. 90% of deaths from lung cancer are estimated to be due to smoking. The risk of lung cancer development is 20-40 times higher in lifelong smokers compared to non-smokers".

The finding strongly suggested the risk of lung cancer in smokers is substantially higher compared to non-smokers.

Betanin is a Phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In the urgency to find a natural compound for the treatment of lung cancer, researchers examined the chemopreventive activity of betanin by oral consumption was investigated in two mouse lung tumor models.

The study included Vinyl carbamate (VC) and benzo(a)pyrene (B(a)P) induced lung tumors in female A/J mice, followed by the treatment of betanin in drinking water.

Betanin significantly decreased tumor multiplicity and tumor load induced by Vinyl carbamate (VC) and benzo(a)pyrene (B(a)P).

Precisely, tumor multiplicity and tumor load were decreased by 20% and 39% in the VC lung model, and by 46% and 65% in the B(a)P lung model, respectively.

Betanin inhibited the lung tumor by reducing the number of microvessels formation and increasing the expression of a protein associated with programmed cell death.

In other words, the lung tumor inhibitory effects were involved in the induction of apoptosis and angiogenesis.

The findings strongly suggested that betanin significantly inhibits lung tumorigenesis in vivo.

Moreover, compared to capsanthin, cranberry, red onion skin, and short and long red bell peppers, betanin showed a vitro inhibitory effect on Epstein-Barr virus early antigen (EBV-EA) induction using Raji cells.

According to the results from the lung bioassays, betanin also revealed a significant lung tumor inhibitory effect.

Taken together, betanin may be considered a supplement for the prevention and treatment of lung cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of betanin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
