#Betacyanins Found in #Beetroot Induce #BladderCancer Cell Apoptosis with Cell Death Programming

Cancer is a group of medical and chronic conditions characterized by irregular cell growth in the tissue of a specific organ in the body.

Most cases of cancer start in the cell on the surface of the inner lining tissue of an organ. After penetrating deeper layers and colonizing the entire organ, the cancerous cells can spread to the healthy tissue of other organs through lymph and blood.

The bladder is a hollow elastic organ in the center of the lower abdomen that collects urine from the kidneys and excreted them through the urethra.

Bladder cancer occurs when cell mass in the inner lining of the bladder forms a malignant growth or tumor. Bladder cancer developed in the bladder lining has also been found to spread through the bladder wall to nearby organs such as the prostate gland in men or the uterus in women.

Bladder cancer can be classified depending on its proliferation
* Superficial types of bladder cancer
The most recurrent type of bladder cancer has a characteristic of noninvasive. These types of bladder cancer can be controlled or removed by regular cystoscopy.

* Invasive types of bladder cancer
Invasive types of bladder cancer tend to spread after beginning in cells in the lining of the bladder. These types of cancer often require surgery by removing part or the whole bladder.

The exact causes of bladder cancer are not identified. However, smoking has been found to induce a substantial risk of bladder cancer incidence.

Dr. Neal D Freedman, the lead scientist wrote, "...the population attributable risk of bladder cancer for tobacco smoking is 50–65% in men and 20–30% in women and that current cigarette smoking triples bladder cancer risk relative to never smoking".


And, "Compared to a pooled estimate of US data from cohorts initiated between 1963 and 1987, relative risks for smoking in the more recent NIH-AARP cohort were higher, with population attributable risks for women compared to those for men".

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In finding a natural pigment for the prevention and treatment of cancer, researchers examined Betacyanins (BC) purified from beetroot (Beta vulgaris var. Rubra L.) tested, alone or in combination with vitexin-2-O-xyloside (XVX) from Beta vulgaris var. cicla L. in the proliferation rate in T24 bladder cancer cells.

According to the tested analysis,* Combination of BC and XVX exhibited a synergistic effect that inhibits proliferation in T24 cancer cells at 24 and 48 h but not after 72 h of incubation.

* The combination-induced apoptosis of T24 bladder cancer cells was observed by means of fluorescence-activated cell sorting (FACS) analysis by increasing the expression in caspases associated with cell death programming.

The inhibition of the bladder cancer cell line was found to enhance the expression levels of the pro-apoptotic gene and inhibit anti-apoptotic and survival proteins.

Extrinsically, the administration of BC increased the activity of caspase 8, a protein associated with the induction of apoptosis.

Interestingly, the XVX, BC, and their combination induced cancer cell death without exhibiting a cytotoxic effect on normal and healthy human cells.

Based on the findings, Dr. Scarpa ES, the lead scientist said, "the efficacy and the mechanisms of the action of BC and XVX, extracted from edible plants, and suggested that a diet or a nutrition supplement, enriched with these bioactive molecules, could be used in the prevention of human bladder cancer".

Taken together, betacyanins may be considered a remedy for the prevention and treatment of bladder cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of betacyanins in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Cholesterol is a way substance produced by the liver or from food sources that plays an important role to build strong cell membranes and producing steroid hormone and vitamin D and aiding the digestive tract in absorbing the nutrients for our body needs.

Hyperlipidemia is also one of the conditions found in the cluster of metabolic syndrome.


In a healthy individual, excess cholesterol is returned to the liver after performing the aforementioned function by high-density lipoprotein or "good cholesterol".

However, a prolonged period of overexpression of abnormal blood cholesterol due to excessive consumption of fat or reduced levels of "good cholesterol" in the bloodstream can cause plaque building up on the arterial wall, leading to heart disease and stroke.

Depending on the choice of diet, your risk of high blood cholesterol can be increased or decreased.


The healthy ratio of blood cholesterol is 4 (low-density lipoprotein) to 1 (high-density lipoprotein).

If your ratio is lower than 4, you are considered to have a healthy blood-cholesterol ratio.


Hyperlipidemia is preventable. If you have high blood cholesterol, reducing bad fat intake accompanied by an increased intake of "good cholesterol" with moderate exercise can be helpful.


More precisely, by changing to a healthy diet and lifestyle with moderate exercise, you are at a lower risk of high blood cholesterol sometimes in the future (weeks or months, depending on the levels of blood cholesterol today).

In finding a dietary source for the treatment of high blood cholesterol, researchers examined whether cooked navy bean powder (NBP) and/or heat-stabilized rice bran (RB) supplementation is tolerable, improves dietary fiber intake in children, and modulates lipid profiles.

The study included children aged 8 to 13 years at risk for CVD due to abnormal lipids. In other words, all participants were diagnosed with elevated cholesterol levels including total cholesterol ≥180 mg/dL and high-density lipoprotein (HDL) <60 mg/dL; low-density lipoprotein (LDL) ≥100 mg/dL and HDL <60 mg/dL; or non-HDL >100 mg/dL and HDL <60 mg/dL.


Participants received muffins or a smoothie daily that included 0 g NBP or RB (control), 17.5 g NBP, 15 g RB, or a combination of 9 g NBP + 8 g RB. Fasting blood was collected at baseline and week 4.

Only 38 children completed the trial, including n = 9 control, n = 10 NBP, n = 9 RB, and n = 10 NBP + RB groups.

Additionally, participants in the intervention groups significantly increased intake of NBP and RB at week 4 (p≤.01), particularly increased fibers in NBP and NBP + RB groups from baseline to week 4.

Based on the tested assays, HDL-cholesterol was higher in NBP-group participants compared to the control at week 4.

Although other lipid parameters also showed signs of decreasing, they are insignificantly due to the
shorter duration of the experiment (4 weeks).


In other words, higher daily intakes are needed for a longer duration to induce favorable changes across multiple serum lipid parameters.

Taken together, navy bean may be considered a remedy for the prevention and treatment of high blood cholesterol, pending the confirmation of the larger sample size and multicenter human study.

Prostate Cancer is a medical condition characterized by irregular cell growth and multiplied disorderly in the prostate tissues.
Beet, best known as the beetroot or garden beet, belonging to the amaranth family, is a perennial plant with leafy stems growing to 1–2 m tall.

Red beetroot (Beta vulgaris L.) may have a profound effect in reduced risk and treatment of prostate cancer, some studies suggested.

According to the Howard University, red beetroot (Beta vulgaris L.) extract (B), approved by Food and Drug Administration and European Union, in drinking water may be safe and used as cancer chemopreventive effect in humans.

Intake of the extract showed a significant effect in reduced synergistic antiproliferative activity against prostate (PC-3) tumor cells of human origin.

When combined anticancer drug doxorubicin (D), the combination acted synergistically in the expression of cytotoxicity against prostate cancer at a ratio of 1:5.

In a total of 24 patients (mean age 68+/-8 years) with hormone-resistant and metastatic prostate cancer treated with taxan chemotherapy, given twice 10 g daily for 1 month of natural table beet products come from commercial service, intake of beet juice showed a significant effect in control of the progression of prostate cancer in promoted life expectancy and reduced toxicity side effect induced by conventional chemo-drug.

The efficacy may be contributed by the mediated activities of phytochemicals in the juice in the inhibition of inflammation through the expression of the production of antioxidants.

The study also suggested that moderate and permanent consumption of table beet products may favorably affect the life expectancy of prostate cancer patients.

Furthermore, in the testing extract of red beetroot (Beta vulgaris L.), the FDA-approved red food color E162, against various cancer cell lines, researchers at Howard University found that the extract showed significant cytotoxicity in decreased the growth rate of the PC-3 cells, although lesser in compared to doxorubicin, a chemo medicine( (8.6% vs. 100%, respectively, at 29 µg/ml concentration of each, three-day test period).

The study also suggested that betanin, the major betacyanin constituent found in beetroot, may be phytochemicals responsible for the cytotoxicity.


These findings showed that beetroot may be used as a functional food to a reduced risk of early-onset and treatment of prostate cancer. And reduced adverse toxicity when used in combination with chemo drugs.

Nees, application of the products suppressed the T-47D breast carcinoma cells caused cytotoxicity, and elicited a non-apoptotic cell death.

Also in the MDA-MB-231 breast cancer cell, Andrographolide, a major chemical compound found in Andrographis exhibited strong anti-breast cancer cell growth activities in regulating breast cancer cell proliferation, survival and progression, and breast cancer metastasis through various mechanisms involving pro-inflammatory pathways.

Taking together, there is no doubt that Andrographis may be used as a possible chemopreventive and chemotherapeutic agent for human breast cancers, But a large amount of intake should only be prescribed by an herbalist.

According to the Meerut Institute of Engineering & Technology, in the carrageenan-induced paw edema method for acute inflammation and cotton pellet granuloma for chronic inflammation in a rat model, injected aqueous extract of leaves of Beta vulgaris exhibited anti-inflammatory effects at a dose level of 1000 mg/kg in comparison to those of indomethacin, a group of nonsteroidal anti-inflammatory drugs (10 mg/kg).

Also carrageenan (100 µg/paw)-induced paw edema and neutrophil migration to the paw skin tissues. treatment with betalain (100 mg/kg, i.p.) significantly inhibited the expression of paw edema. caused by carrageenan, a common food additive and complete Freund's adjuvant (10 µl/paw) used in animal experiments to induce infection.

Neutrophils are white blood cells that form an important role in defense against infection and inflammation caused by injury and microorganisms invasion and neutrophil migration is a rapid movement of white blood cells to the location of infection or inflammation.

#Asparagus Processes Antidiarrhoeal Activity by Regulating Vascular Smooth Muscle Contraction and Vasoconstriction

Asparagus is a flowering plant belonging to species the genus Asparagus, native to the western coasts of northern Spain, north to Ireland, Great Britain, northwest Germany, northern Africa, and western Asia, used from early times as a vegetable and medicine because its delicate flavor and diuretic properties.
Diarrhea is a condition of frequent discharge of feces in liquid form.

According to the K.P. College of Pharmacy, Asparagus racemosus wild root extract used in traditional herbal medicine exerted the antidiarrhoeal potency against several experimental models of diarrhea in model animals, similar to those of conventional medicine such as loperamide.


This efficacy is attributed to the root extracts in the mediation of Prostaglandin E2 (PGE2), a principal hormone in the expression of inflammatory diseases.
Dr. Venkatesan N., the lead researcher said, "The plant extracts showed significant (P < 0.05) inhibitory activity against castor oil-induced diarrhea and PGE2 induced enter polling in rats when tested at 200 mg/kg. Both extracts also showed significant (P < 0.001) reduction in gastrointestinal motility ".

In support of the above study, the test of the animal model on Alpha-2 blocker (see alpha(2)-adrenoceptor) experimentally-induced diarrhea and ulceration, showed a positive effect of methanolic extract of Asparagus pubescens root in reducing significantly intestinal propulsive movement and intestinal fluid accumulation in (500-1500 mg/kg) in a dose-dependent manner.

In fact, the antidiarrhoeal effect of asparagus may be a result of modulating the expression of alpha(2)-adrenoceptor in regulated vascular smooth muscle contraction and vasoconstriction through Prostaglandin E2 (PGE2) modification. But further studies may be necessary to validate this potency.
