#Indicaxanthin Inhibits Pathways Associated wuth the Proliferation of the #Melanoma Cells in Vitro, According to Studies

Kyle J. Norton

The skin is the largest external organ that protects us against infection and physical trauma by guarding the underlying muscles, bones, ligaments, and internal organs.

Furthermore, the skin accompanied by liver and kidney detoxes toxins in the body
Melanoma is a type of skin cancer caused by the alternation of cell DNA in the pigment-containing cells known as melanocytes. Most cases of melanomas are found in the skin.

According to the statistics, approximately 120,000 new cases of melanoma in the US are diagnosed in a year. Skin cancer tends to spread along the skin in most cases, but also can grow down to the deeper layers of the skin.

At the advanced stage, skin melanoma can also travel a distance away to infect other healthy tissues through blood and fluid circulation.

Most common symptoms of melanoma skin cancer are
* Asymmetry
The mole on the skin is no longer symmetric as one half of the mole is different from the other half.

* Borders irregular
The border of the mole has become irregular, such as uneven or notched.

* Color
The color of the mole is different than other moles on the skin.

* Diameter
The mole is larger than other moles and evolved over time
Accompanied by pain, swelling, and bleeding in the affected skin

There are 3 main types of melanoma skin cancer, including
* Superficial spreading
This is the most common type of melanoma, accounting for over 70% of all cases. The cancer cell usually starts with a normal mole, but for whatever reason, it begins to change malignantly with irregular borders and color variations.

* Nodular type
This type of melanoma is more aggressive growth than superficial spreading and found most on the trunk, upper arms, and thighs. The nodular type of melanoma is usually detected as a lump with blue and black color.

* Acral melanoma
Acral melanoma is mostly founded on the hands, feet, and nail beds. The risk increases for people with darken-skin color.

* Lentigo malignant melanoma
Found mostly on the face of the elderly caused by great exposure to sunlight.

Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.

With an aim to find a potent ingredient for the treatment of cancer, researchers investigated the effects of Indicaxanthin, against human melanoma both in vitro and in vivo.

According to the tested analysis, the growth of A375 cells was inhibited by the injection of the phytochemical

Furthermore, Indicaxanthin also induced apoptosis and decreased the invasiveness of A375 cells.

In other words, indicaxanthin effectively inhibits the proliferation of highly metastatic and invasive, and anti-apoptotic activity of the A375 cells.

In mice, induced melanoma, oral administration of Indicaxanthin also significantly reduced tumor development.

In additional differentiation of the anti-cancer activity of Indicaxanthin, researchers found that the efficacy of the bioactive compound was attributed to the inhibition of the NF-κB pathways associated with the antiproliferative effect of indicaxanthin.

Based on the results, researchers wrote, " indicaxanthin represents a novel phytochemical able to significantly inhibit human melanoma cell proliferation in vitro and to impair tumor progression in vivo".

Taken together, indicaxanthin may be considered a remedy for the prevention and treatment of melanoma, pending the confirmation of the larger sample size and multicenter human study.

#BeetRoot Shows a Significant Cytotoxicity in Decreased the Growth Rate of #ProstateCancer Cells, Researchers Say

Kyle J. Norton

Prostate Cancer is a medical condition characterized by irregular cell growth and multiplied disorderly in the prostate tissues.
Beet, best known as the beetroot or garden beet, belonging to the amaranth family, is a perennial plant with leafy stems growing to 1–2 m tall.

Red beetroot (Beta vulgaris L.) may have a profound effect in reduced risk and treatment of prostate cancer, some studies suggested.

According to the Howard University, red beetroot (Beta vulgaris L.) extract (B), approved by Food and Drug Administration and European Union, in drinking water may be safe and used as cancer chemopreventive effect in humans.

Intake of the extract showed a significant effect in reduced synergistic antiproliferative activity against prostate (PC-3) tumor cells of human origin.

When combined anticancer drug doxorubicin (D), the combination acted synergistically in the expression of cytotoxicity against prostate cancer at a ratio of 1:5.

In a total of 24 patients (mean age 68+/-8 years) with hormone-resistant and metastatic prostate cancer treated with taxan chemotherapy, given twice 10 g daily for 1 month of natural table beet products come from commercial service, intake of beet juice showed a significant effect in control of the progression of prostate cancer in promoted life expectancy and reduced toxicity side effect induced by conventional chemo-drug.

The efficacy may be contributed by the mediated activities of phytochemicals in the juice in the inhibition of inflammation through the expression of the production of antioxidants.

The study also suggested that moderate and permanent consumption of table beet products may favorably affect the life expectancy of prostate cancer patients.

Furthermore, in the testing extract of red beetroot (Beta vulgaris L.), the FDA-approved red food color E162, against various cancer cell lines, researchers at Howard University found that the extract showed significant cytotoxicity in decreased the growth rate of the PC-3 cells, although lesser in compared to doxorubicin, a chemo medicine( (8.6% vs. 100%, respectively, at 29 µg/ml concentration of each, three-day test period).

The study also suggested that betanin, the major betacyanin constituent found in beetroot, may be phytochemicals responsible for the cytotoxicity.

These findings showed that beetroot may be used as a functional food to a reduced risk of early-onset and treatment of prostate cancer. And reduced adverse toxicity when used in combination with chemo drugs.

#Andrographis Inhibits Pathways and Metastasis Associated with an Increased Risk of #BreastCancer

Kyle J. Norton

Herbal Andrographis paniculata may be used as a therapeutic agent for treatment and drug resistance to breast cancer, a study by the renowned institute suggested.

Andrographis paniculata also known as Andrographis is a herbaceous plant, genus Andrographis, belonging to the family Acanthaceae, native to India and Sri Lanka, used in traditional Siddha and Ayurvedic in treating the common cold, flu, and upper respiratory infections, infectious diseases, diarrhea, dysentery, cholera, pneumonia,...

Breast cancer (malignant breast neoplasm) is a condition characterized by abnormal cells growing in the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma).

In 2010, over 250,000 new cases of breast cancer were expected to be diagnosed in women in the U.S. alone and the risk of getting invasive breast cancer during the lifetime of a woman is 1/8.

According to the joint study led by the University of Kalyani, Andrographolide showed significant inhibition of MDA-MB-231 breast cancer cell proliferation in a time- and concentration-dependent manner.

The study also found that the inhibition of breast cancer cell line proliferation with no effect on normal breast epithelial cells, through reduced oxygen species (ROS) production and increased the population of apoptotic cells.

Others, in the study of the cytotoxic effect of a chemical compound, 14-Deoxy-11,12-didehydroandrographolide (14-DDA), a major diterpenoid isolated from Andrographis paniculata (Burm.f.) Nees, application of the products suppressed the T-47D breast carcinoma cells caused cytotoxicity, and elicited a non-apoptotic cell death.

Also in the MDA-MB-231 breast cancer cell, Andrographolide, a major chemical compound found in Andrographis exhibited strong anti-breast cancer cell growth activities in regulating breast cancer cell proliferation, survival and progression, and breast cancer metastasis through various mechanisms involving pro-inflammatory pathways.

Taking together, there is no doubt that Andrographis may be used as a possible chemopreventive and chemotherapeutic agent for human breast cancers, But a large amount of intake should only be prescribed by an herbalist.

#AqueousExtract of Leaves From #Beetroot Exerts Anti Inflammatory Activity by Regulating the Inflammatory Mediator

Kyle J. Norton

Beet may be one functional food, having an anti-inflammatory potential in reducing pain in damaged tissues or local infection.
Inflammation is a natural reaction of the immune system to protect of our body during tissues damage, injure, and infection

Beet, best known as the beetroot or garden beet and belonging to the amaranth family, is a perennial plant with leafy stems growing to 1–2 m tall.

According to the Meerut Institute of Engineering & Technology, in the carrageenan-induced paw edema method for acute inflammation and cotton pellet granuloma for chronic inflammation in a rat model, injected aqueous extract of leaves of Beta vulgaris exhibited anti-inflammatory effects at a dose level of 1000 mg/kg in comparison to those of indomethacin, a group of nonsteroidal anti-inflammatory drugs (10 mg/kg).

Also carrageenan (100 µg/paw)-induced paw edema and neutrophil migration to the paw skin tissues. treatment with betalain (100 mg/kg, i.p.) significantly inhibited the expression of paw edema. caused by carrageenan, a common food additive and complete Freund's adjuvant (10 µl/paw) used in animal experiments to induce infection.

Neutrophils are white blood cells that form an important role in defense against infection and inflammation caused by injury and microorganisms invasion and neutrophil migration is a rapid movement of white blood cells to the location of infection or inflammation.

According to the study, the anti-inflammatory effect of the aqueous extract of leaves of Beta vulgaris probably was attributed to phytochemicals found in the extract such as flavonoids, carbohydrates, pentose, amino acids, saponins, tannins, and steroids in promoted and increased movement of plasma and leukocytes from the blood into the injured tissues to counteract the invasion of foreign substances and disease. and exhibited the release of histamine in the generation of mass and white blood cells and serotonin-induced platelets binding to a clot, then ameliorating inflammation through regulation of inflammatory mediators including leukotrienes and prostaglandins,

Please note that
1. Hormone prostaglandins play a key role in the generation of the inflammatory response and stimulation of constriction and clotting of platelets.

2. Leukotrienes (LTs) is a family of lipid and inflammatory mediators with a function in response to pathogenesis of inflammation

The study emphasized that in compared to a group of nonsteroidal anti-inflammatory drugs, an aqueous extract of leaves of Beta vulgaris at a specific dose exhibited a stronger and higher inhibition of acute inflammation induced by prostaglandin synthesis, including the inflammatory response to diverse stimuli in an induced temporary increase of physiological activity.

Further analysis also suggested that in chronic inflammatory diseases induced by cotton pellet granuloma, such as arthritis, aqueous extract of leaves of Beta vulgaris although less effective than the conventional drug of indomethacin, had a profound effect in reduced symptoms and prevented inflammatory process of cells at the site of inflammation by healing of the tissue through the initiated cells shift to induced apoptosis.

In other words, an aqueous extract of leaves of Beta vulgaris ameliorated risk of chronic inflammation by healing the affected tissue, thus preventing cell being damage caused by prolonged inflammation.

Furthermore, the application of betalain (100 mg/kg), isolated from beetroot also suppressed free radical production and inflammatory activity through enhanced antioxidant activity and inhibited the overexpression of pro-inflammatory proteins such as

TNF-α, a tumor necrosis factor, a protein with function in regulating the production of inflammatory cytokines in the acute phase of invasion by foreign substances, and interleukin (IL)-1β have a duo function in the activated either pro-inflammatory cytokine or anti-inflammatory myokines, and promoted
IL-10 expression in the production of an anti-inflammatory cytokine

In healthy adults, with inflammatory bowel diseases (IBD), betanin, the major component of beet (Beta vulgaris var. rubra) also demonstrated a significant anti-inflammatory property in modulated reactive oxygen species (ROS) production and oxidative stress cause of DNA damage as well as increasing apoptosis in neutrophils by analyzing the blood samples of tested subjects.

Dr. Zielińska-Przyjemska M the lead author said, "Betanin treatment at the concentration of 100 μM for 24 h increased DNA damage assessed by comet assay in IBD patients' neutrophils. A similar effect although less pronounced was observed in Caco-2 cells(colon cancer cell line)".

The findings suggested that beet with plenty of phytochemicals may have a therapeutic potential to be utilized in the treatment of inflammation-associated diseases.

#Asparagus Processes Antidiarrhoeal Activity by Regulating Vascular Smooth Muscle Contraction and Vasoconstriction

Kyle J. Norton

Asparagus is a flowering plant belonging to species the genus Asparagus, native to the western coasts of northern Spain, north to Ireland, Great Britain, northwest Germany, northern Africa, and western Asia, used from early times as a vegetable and medicine because its delicate flavor and diuretic properties.
Diarrhea is a condition of frequent discharge of feces in liquid form.

According to the K.P. College of Pharmacy, Asparagus racemosus wild root extract used in traditional herbal medicine exerted the antidiarrhoeal potency against several experimental models of diarrhea in model animals, similar to those of conventional medicine such as loperamide.

This efficacy is attributed to the root extracts in the mediation of Prostaglandin E2 (PGE2), a principal hormone in the expression of inflammatory diseases.
Dr. Venkatesan N., the lead researcher said, "The plant extracts showed significant (P < 0.05) inhibitory activity against castor oil-induced diarrhea and PGE2 induced enter polling in rats when tested at 200 mg/kg. Both extracts also showed significant (P < 0.001) reduction in gastrointestinal motility ".

In support of the above study, the test of the animal model on Alpha-2 blocker (see alpha(2)-adrenoceptor) experimentally-induced diarrhea and ulceration, showed a positive effect of methanolic extract of Asparagus pubescens root in reducing significantly intestinal propulsive movement and intestinal fluid accumulation in (500-1500 mg/kg) in a dose-dependent manner.

In fact, the antidiarrhoeal effect of asparagus may be a result of modulating the expression of alpha(2)-adrenoceptor in regulated vascular smooth muscle contraction and vasoconstriction through Prostaglandin E2 (PGE2) modification. But further studies may be necessary to validate this potency.

Liquid Extracts From #Artichoke Improves Liver Functioning by Enhancing Liver Cell Regeneration, According to Studies

Kyle J. Norton

In seeking natural therapy with no side effects, scientists may have found a herbal remedy for the improvement of liver function, according to studies.

The liver function plays a critical role in filtering blood that comes from the intestinal before passing them to other organs and tissues. The reddish-brown organ also detoxes toxic chemicals and metabolizes medicine and secrete bile to the intestine for breaking down food and absorbing nutrients.

Additionally, the liver is found to process the ability to secrete proteins such as albumin, fibrinogens, and apolipoproteins into the blood for the regulation of oncotic pressure, blood clotting, and lipoprotein metabolism, respectively.

There are many factors that can disrupt liver function including viral infection, alcohol use, or obesity, leading to scar formation in the liver tissue.

Long-term untreated chronic liver infection that causes the replacement of normal liver tissue by scar tissues eventually leads to cirrhosis, a non-reversible, dangerous condition that can lead to liver failure and cancer.

At the early stage of liver disease, most patients are asymptomatic. However, as the disease progresses patients may experience unspecific symptoms of loss of appetite, fatigue, general malaise, gastrointestinal discomforts, or weight loss.

At the advanced stage severe on the right side of the abdomen, jaundice, pale stool, harden urine color, spiderlike blood vessels on the skin, and redness in the palms of the hands.

Liver damage at this stage may cause loss of period in some women and loss of sex drive, breast enlargement, or testicular atrophy in men.

In severe cases, patients also have symptoms of confusion, drowsiness, and slurred speech.

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine as a liver protective and detoxified agent, and to treat digestive disorders, abdominal pain gas and bloating, etc.

In finding a natural compound for enhancing liver function, researchers at the Laboratoire de pharmacology examined the liver-protective effects of artichoke (Cynara scolymus) leaf extract.

Injection of artichoke extract improved the liver function in the production of bile and liver lipid production and lipid recycling by lowering blood cholesterol.

In animal models, the liquid extracts of the roots and leaves of artichokes were also found to demonstrate an ability to protect the liver, possibly in enhancing liver cell regeneration.

Furthermore, the administration of artichoke extract reduced the risk of cardiovascular disease by lowering risk factors associated with liver function in regulating cholesterol and triglycerides.

According to the therapeutic effects of artichoke on biochemical and liver biomarkers in patients with nonalcoholic steatohepatitis (NASH), artichoke improved the function of the liver by improving liver enzymes which were significantly reduced in NASH.

Taken altogether, artichoke may have a strong implication in improving liver function in many aspects, pending the validation of larger sample sizes and multicenter studies.

#AloeEmodin Inhibits High Blood Pressure by Decreasing Vascular Smooth Muscle Stiffness, Scientists Show

Kyle J. Norton

Good news to people with hypertension, Aloe Vera may be a potential therapeutic agent for lowering high blood pressure a renowned University study suggested.

Hypertension is a condition of abnormally high blood pressure, leading to symptoms of headaches, shortness of breath, or nosebleeds......

Hypertension is a measurement with home systolic/ diastolic BP > or = 135/85 mmHg.

According to statistics, the prevalence of hypertension affects over 39.4% of men and 29.3% of women in the Western World.

Prolong untreated hypertension is found to induce complications of strokes, heart attacks, heart failure, or kidney disease.

Some researchers suggested that patients who are actively in modulated daily high blood pressure may be benefits from reduced risk of stroke by 35%, heart attack by 20%, and heart failure by 50%.

According to the WHO-based Healthy Diet Indicator (HDI), the American Heart Association 2006 Diet and Lifestyle Recommendations(4),
* The prevalence of untreated hypertension and treated hypertension was 22.4% and 8.5%.

* Patients with untreated hypertension had significantly lower HDI and AI-84 scores compared with non-hypertension.

* After adjusting for age, energy intake, smoking habit, alcohol drinking, physical activity and salt intake, a low adherence to HDI and a lowest quartile of AI-84 score were associated with a significantly higher prevalence of untreated hypertension, with an OR of 3.33 (95% CI 1.39 to 7.94, p=0.007) and 2.23 (1.09 to 4.53, p=0.027), respectively.

These results are agreed to a general and popular belief that an unhealthy diet plays an important role in the induction of high blood pressure. Furthermore, the lower dietary quality was associated with an increased prevalence of untreated hypertension.

People who eat more fruits and vegetables compared to average daily intake are associated with a significantly lower risk of hypertension.

Dr. Utsugi MT in the comparison of fruits and vegetable intake daily said, "the highest-tertile consumptions of fruits, vegetables, potassium, and vitamin C were associated with a significantly lower risk of hypertension (45%, 38%, 46%, and 43% lower risk of home hypertension, respectively(5)".

Aloe Vera is species of succulent plant in the genus Aloe, belonging to the Family Xanthorrhoeaceae, native to Sudan. It has become very popular for commercial cultivation due to its enormous health benefits. Aloe vera has also been used in herbal medicine in treating many kinds of disease, including wound, burn healing, minor skin infections, sebaceous cysts, diabetes, and elevated cholesterol,

According to the Hamdard University, aloeemodin, aloin A, a chemical compound found in Aloe Vera showed to express hypotensive effects by lower mean arterial blood pressure by 26 %, 52 %, and 79 % in the corresponding doses of 0.5, 1, and 3 mg/kg in an animal study.

Dr. Saleem R, the lead author of the study suggested that Aloeemodin has emerged as a potent hypotensive agent in the current pharmacological investigation.

In a total of In a double-blind, placebo-controlled, crossover study, healthy, Aloe-emodin, another major chemical compound\ inhibited vascular smooth muscle stiffness, a condition in the development of hypertension, involved cells in supporting structural and biochemical and the interior surface of blood vessels.

In hypertension, increased large-artery stiffness is partly due to the intrinsic mechanical properties of vascular smooth muscle cells.

More importantly, for smooth muscle cells derived from hypertensive and control rats studies, treatment of Aloe-emodin also significantly reduced hypertension by improving the elastic value of vascular smooth muscle cells, through its effects on vascular smooth muscle cell cytoskeletal proteins in the regulated function of cell division.

Other researchers suggested that the antihypertensive properties of Aloe-emodin may be a result of the inhibition of tonic tension and regulated apoptosis, pro- and anti-apoptotic activities on vascular smooth muscle cells.

With all the information collected, Aloe Vera may have an antihypertensive effect in regulated protein in cell production on vascular smooth muscle which is the main cause of high blood pressure, but more precise understanding may be necessary to reassess this pharmaceutical value.
