#Mustards Inhibit the Risk of Chronic Diseases through Their Antioxidants and Anti-Chemical Toxicity Activity, Studies Suggest

Chronic diseases are a group of medical conditions most commonly associated with aging, including
seizures, and obesity.

Nutrigenomic activity is a scientific term that optimizes the interaction of nutrition and genes, especially in the prevention or treatment of disease.

In other words, the nutrigenomic activity provided information on how the interaction of certain foods with specific genes leads to the increased risk of chronic diseases such as type 2 diabetes, obesity, heart disease, stroke, and certain cancers.

Biologically, nutrigenomics also provided information molecularly how chemicals found in the diet altered the expression of genes that affect individual health.

More precisely, nutrigenomics is depending totally on an individual's genetic makeup.

According to the Centers for Disease Control, chronic diseases also are the major driver of healthcare costs, accounting for nearly 75 percent of aggregate healthcare spending and increasing absenteeism that impact workforce patterns.

Each chronic patient has cost health care spending $5300 annually.

Believe it or not, chronic diseases in the US, chronic diseases are responsible for 7 out of 10 deaths yearly. In other words, chronic diseases kill more than 1.7 million Americans each year.

According to the joint study led by Fordham University, chronic diseases are among the most prevalent and costly health conditions in the US, affecting over 133 million of the US population as many of them suffer from at least one chronic disease and the number is growing in a rapid rate.

Some chronic diseases such as cancer have been found to be the leading causes of hospitalization, long-term disability, reduced quality of life, and death.

The World Health Organization suggested that unhealthy diet and excessive energy intake, physical inactivity, and tobacco use are some of the most common causes of chronic diseases.

In other words, an unhealthy diet and physical inactivity raised blood pressure, raised glucose levels, abnormal blood lipids (particularly low-density lipoprotein – LDL cholesterol), and overweight (body mass index ≥25 kg/m2) and obesity (body mass index ≥30 kg/m2) are some of the most common prevalent factors found in patients in the Western World.

Other risk factors associated with the risk of chronic disease also including excessive alcohol drinking, infectious agents, and some environmental factors such as air pollution, which contribute to asthma and other chronic respiratory diseases.

Some researchers suggested by changing unhealthy lifestyle patterns, chronic diseases may be preventable.

Dr. Walter C. Willett, the lead scientist in the concern about the risk of chronic disease in poor lifestyle individuals wrote, "Several lines of evidence indicate that realistic modifications of diet and lifestyle can prevent most CAD, stroke, diabetes, colon cancer, and smoking-related cancers".

And, "Among U.S. adults, more than 90 percent of type 2 diabetes, 80 percent of CAD, 70 percent of stroke, and 70 percent of colon cancer are potentially preventable by a combination of nonsmoking, avoidance of overweight, moderate physical activity, healthy diet, and moderate alcohol consumption".


Mustard is a genus Brassica, belongings to the family Brassicaceae, native to the Himalayan region of India. The veggie has seen used in some forms in African, Italian, Indian, Chinese, Japanese, and soul food cuisine because of its popularity.

With an aim to find natural food that lowers the incidence of chronic diseases including different kinds of cancer, researchers examined the anti-cancer effects of sulfur-containing glucosinolates (GLSs)-secondary plant compounds from mustards, including isothiocyanates.

According to medical literature, ITCs promoted overall health in vitro and in Vivo. The phytochemical also exerted anti-carcinogenic and anti-inflammatory properties by activating the redox-sensitive transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2) involved in antioxidant expression. and phase II enzymes involved in drug metabolism.

Furthermore, administration ITCs also significantly ameliorate a severe inflammatory phenotype in colitic mice in vivo.

In other words, ITCs inhibited the risk of chronic disease through their antioxidants and anti-chemical toxicity activity.

Researchers after taking other factors into account wrote, "an epigenetic mode of action for Brassica-derived phytochemicals, the conduction of further studies would be recommendable to investigate if the beneficial effects of these compounds also persist during an irregular consumption pattern".

Taken together, mustards processed abundantly ITCs may be considered supplements for the prevention and treatment of chronic diseases, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Depression is a common and serious psychological illness, that affects the physical and mental functions of patients, including the patients feeling, thinking, and acting.

Some patients with depression may feel sadness and/or a loss of interest in activities once enjoyed.

There is no exact cause of depression. Changes in hormone levels, medical conditions, stress, grief, or difficult life circumstances that initiate the change of brain chemistry in inducing depressive syndrome are some of the most prevalent factors found in patients with depression.

The most common symptoms of depression include sadness, hopelessness, guilt, change of mood, and loss of interest in friends, family, favorite activities, and sex.

In severe cases, some patients with depression also experience cognitive symptoms of trouble concentrating, making decisions, remembering, and thoughts of harming themselves.

According to the statistics provided by the Anxiety and Depression Association of Americans, major depressive disorder is the leading cause of disability in the U.S. for ages 15 to 44.3, affecting more than 16.1 million American adults, or about 6.7% of the U.S. population age 18 and older.

Conventionally, selective serotonin reuptake inhibitors (SSRIs) are the primary medication for the treatment of depression due to their relatively lower side effects compared to depressants. Other types of antidepression medication may include serotonin-norepinephrine reuptake inhibitors (SNRIs), typical antidepressants, and monoamine oxidase inhibitors (MAOIs).

Dr. Steven S. Clevenger and colleagues in examining the role of selective serotonin reuptake inhibitors in the relapse risk of major depressive disorder wrote, "According to the included studies, the relapse risk in adults was lower when SSRIs were combined with psychotherapy".

And, " the combination of SSRIs and cognitive behavioral therapy may effectively reduce relapse. Escitalopram appeared to yield better results and fewer side effects than did other SSRIs or SNRIs".
On finding a potent compound that processes depressant-like activity, researchers examined the effects of Sulforaphane (SFN), an isothiocyanate compound derived from broccoli on inflammation-related depressive symptoms.

In lipopolysaccharide, (LPS, 0.5 mg/kg) induced depression-like behavior in mice, pretreatment SFN showed a significant effect in the blockage of the increase in the production of pro-inflammatory cytokine (tumor necrosis factor-α (TNF-α)) level and an increase in microglial activation of the brain associated with the inflammatory response by increasing the expression anti-inflammatory cytokine IL-10.

Activation of microglia is a hallmark of brain diseases.

In the tail suspension test and forced swimming test, immotility caused by the LPS of the tested mice was also attenuated by the injection of SFN.


Degeneration of cognition in the brain caused by the alternation of the density of the proteins associated with the brain functioning due to the LPS was also restored by the SFN.

More importantly, dietary intake of 0.1% glucoraphanin (a glucosinolate precursor of SFN) food in the juveniles and adolescents has been found to prevent the risk of LPS-induced depression-like behaviors and dendritic spine changes in the brain regions in adulthood.


In other words, SFN isolated from the rich broccoli sprout exhibited depressant-like activity that was attributed to the anti-inflammatory property.

Based on the findings, researchers said, "supplementation of SFN-rich broccoli sprout could be a prophylactic vegetable to prevent or minimize the relapse by inflammation in the remission state of depressed patients".

Taken together, broccoli sprouts rich in SFN may be considered a functional food for the treatment of depression, pending the confirmation of the larger sample size and multicenter human study.

The condition can also be caused by athletic hypertrophy (a condition related to exercise, valve disease, hypertrophic cardiomyopathy (HOCM), and congenital heart disease.

In finding a potential compound that attenuates cardiac H9c2 cell hypertrophy, researchers examined the effects of 3,3'-Diindolylmethane (DIM) is the major product of the acid-catalyzed condensation of indole-3-carbinol (I3C), a component of extracts of Brassica food plants.


The study was conducted using an in vitro model with 1 µM angiotensin II (Ang II) in cultured rat cardiac H9c2 cells, to observe the effects of DIM on cardiac hypertrophy.

According to the tested analysis, the optimal concentration of DIM for 6, 12, and 24 h. showed a significant effect on the atrial natriuretic peptide, brain natriuretic peptide, and myosin heavy chain β mRNA expression induced by Ang II in H9c2 cells.

In other words, DIM lower the blood pressure, promotes diuresis, natriuresis, vasodilation of the systemic and pulmonary vasculature, and reduction of circulating levels of endothelin and aldosterone and cardiac muscle contraction induced by Ang II in H9c2 cells.

Furthermore, H9c2 cells treated with DIM for 0, 15, 30, and 60 min induced by Ang II also improve the function of 5'-AMP-activated protein kinase catalytic subunit alpha-1 that regulates the activities of a number of key metabolic enzymes in protecting the cardiac cells from stresses in response to hypertrophic stimuli.

Based on the findings, Brassica food plants consist of DIM that may be used for the prevention of cardiac hypertrophy, pending the confirmation of the larger sample size and multicenter human study.

Antioxidants can be either produced by the body or injected into the body from outside sources such as the consumption of antioxidant-rich fruits and vegetables.

Free radicals are everywhere, including the air we breathe, the food we eat and the body daily functioning such as cell metabolism.

In other words, any chemical compound which has an oxygen molecule can become a free radical after entering our body.

Most healthy individuals can produce enough amounts of antioxidants endogenously to counter the bombards of free radicals. However, people with a weakened immune system such as the elderly, and immature immune systems such as the baby may need the injection of antioxidants from outside sources.

More precisely, people with a weakened immune system do not produce enough antioxidant enzymes to inhibit the chain reaction of free radicals which occurs daily in the body due to the unstable nature of the molecules.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, kale, etc.

With an aim to find the natural ingredient for the treatment of diseases associated with oxidative stress, researchers investigated the effect of indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) in type 2 diabetes mellitus (T2DM).
The study included selected genetically modified mice (C57BL/6J mice) similar to that of metabolic abnormalities of the human disease after the administration of a high-fat diet (HFD).

Including indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) into the diet showed positively modulated glucose, insulin, and Hb and HbA1c levels in HFD mice against the risk of diabetes complications.

Compared to the HFD group, injection of I3C and DIM protected the infected rat against the formation of lipid peroxidation observed by levels of thiobarbituric acid reactive substances (TBARS), lipid hydroperoxides (LOOH), conjugated dienes (CD).

Furthermore, levels of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were also increased, an indication of the balance of the ratio of antioxidants and free radicals.

Interestingly, I3C and DIM also improved the free radical scavengers such as vitamin C, and vitamin E, and improved the reduced glutathione (GSH) levels in treated mice.

Based on the findings, researchers summarized, "From the study, it was clear that the antioxidant-scavenging action was accelerated in mice treated with DIM than the I3C treatment group which was comparable with the standard drug metformin".

Taken together, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and combined with primary medicine for the treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

The expression of oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.
Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radicals before they can cause a chain reaction in facilitating protein, lipid, and cell damage.

Antioxidants can either be produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.


Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.

Due to its nature to become stable, free radicals readily donate or steal an electron from other molecules, leading to the chain of domino effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

There is no single cause of oxidative stress. Some researchers suggested that a number of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Sadly, a prolonged period of oxidative stress present in the human body can cause oxidative damage to biomolecules, (lipids, proteins, DNA) and the death of healthy cells through cytotoxicity.


Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, and buckwheat. etc.

In finding a potential phytochemical for the prevention and treatment of diseases associated with oxidative stress, researchers examined beta-sitosterol (BS), a plant sterol antioxidant activity.

In vitro, HT22 cells and primarily cultured hippocampal cells induced oxidative stress by glucose oxidase (GOX) incorporate BS into the plasma membrane, preventing GOX-induced oxidative stress and lipid peroxidation.

Furthermore, the incorporation of BS into the cell membrane increased the PI3K activity in the promotion of cellular function and recruitment of PI3K to lipid rafts.

Additionally, the levels of p-GSK3beta also in the promotion of cell integrity were also enhanced by the application of BS against the onset of oxidative stress.

Based on the evidence, researchers said, " ...the results provide a better understanding of the beneficial effects of these BS-enriched nutrients on neurodegenerative diseases such as AD".

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of diseases associated with oxidative stress, including AD, pending the confirmation of the larger sample size and multicenter human study.
