Tumor suppressor proteins are types of genetic materials that protect the body against cell proliferation by regulating the activity of a variety of cellular genes involved in growth and cell cycle progression.
In other words, tumor-suppressor genes encode proteins that slow or inhibit progression through a range of mechanisms to protect the cell integrity against mutation including inhibiting proliferation, programming cell death, and involving cell cycle arrest.
Proteins found in the cells of organisms are large, complex molecules that play many critical roles in maintaining the structure, function, and regulation of the body’s tissues and organs.
Proteins are made up of hundreds or thousands of smaller units called amino acids, attaching one to another in long chains.
The most common types of tumor-suppressor proteins are
* The retinoblastoma protein (Rb) is a tumor suppressor protein that has been found to induce dysfunction in several major cancers by preventing excessive cell growth and inhibiting the cell cycle.
* p53 is also a tumor suppressor protein that plays a critical role in the regulation of cell division through its anti-proliferative activity.
In many cancer, the loss or inactivation of the tumor suppressor genes has been shown to contribute to the abnormal proliferation of tumor cells.
Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.
In finding a potential whole for the treatment of cancer, researchers examined the effect of Indole-3-Carbinol (I3C), a dietary compound found naturally in cruciferous vegetables, including broccoli and brussels sprouts on cell arrest of human reproductive cancer cells.
According to the I3C treatment on the tested cancer cell line, the bioactive compound increased both the transcripts and protein levels of the CDK2 inhibitor p21 associated with a tumor-suppressive gene linking to DNA damage cell cycle arrest through tumor protein p53, another tumor suppressor.
In other words, ablating p53 production prevented the I3C function to induce G1 arrest and p21 expression.
In cell division assays, the antiproliferative activity of I3C was attributed to p53 protein stability and greater transactivation of p21 potential.
Based on the finding, researchers said, " I3C induced G1 arrest of human prostate cancer cells requires the induced production of the activated phosphorylated forms of p53, which stimulate transcription of the CDK2 inhibitor p21".
Taken together, broccoli processed with a high amount of Indole-3-Carbinol may be considered a functional food for the prevention and treatment of prostate cancer, pending the confirmation of the larger sample size and multicenter human study.
Intake of I3C in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthyfood #OnionExtract Exhibits Anti-Cancer Proliferative Effects Through Its Antioxidant Activity, Reserachers Suggest
Kyle J. Norton
Proliferation is a process of cancer growth associated with an abnormally high rate of cancer cell division, leading to rapid overgrowth of the tumor.
Cancer is a class of medical conditions caused by irregular cell growth that starts in the cell on the surface of the inner lining tissue in a specific organ.
Most cancers that grow in size through proliferation can be invasive. In other words, after passing the stages of colonizing the entire tissues of the organ, invasive cancer cells can travel a distance away from the primary site infecting other healthy tissue and organ via blood and fluid circulation, leading to secondary metastasis.
Cancer cell proliferation by dividing unlimitedly, compared to normal cells that die after a certain cell cycle division.
More precisely,
* The growth of normal cells
Under normal circumstances, cell growth is a natural process without even notice, every day millions of cells grow to replace millions of dead cells and to keep our body in balance and healthy.
Sometimes, acceleration of cell growth is necessary to prevent further damage to the body organ, such as cuts, injuries, etc., but after fulfilling their purposes, the growth of cells is back to a normal rate again due to our body growth inhibitors. Unfortunately, skin cells sometimes overgrow while healing, leading to scars on the skin.
*The primary tumor; size and growth
A primary tumor or original tumor that the tumor progression that yields a cancerous mass. Some cancers can begin in several places, such as lymphomas and leukemias. Most cancers can not be detected until they formed a small grape-like lump. Cancer cells need 42 doublings before they can produce enough cancer cells to cause death.
According to the statistics provided by the National Cancer Institute, In 2018, approximately 1,735,350 cases of cancer were diagnosed in the US and the disease also caused the death of 609,640 Americans.
Epidemiologically, the odds of cancer incidence is 439.2 per 100,000 men and women per year based on 2011–2015 statistics.
The onion is a plant in the genus Allium, belonging to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.
Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild, or sweet.
In finding a potential compound for the treatment of cancer, researchers examined the effects of ethyl acetate extract of onion (EEO) had potent inhibitory effects on the FAS gene involved in the over-expressing human breast cancer MDA-MB-231 cells.
According to the tested analysis, EEO exhibited MDA-MB-231 cells. apoptosis through the inhibition of FAS, the apoptosis antigen.
Furthermore, according to the differentiation of 3T3-L1 adipocytes used in vitro model of white adipocytes, EEO also suppressed the lipid accumulation via inhibition of FAS, a condition found in obese patients.
These results strongly suggested that EEO not only prevents the risk of breast cancer in obese but also has a strong impact on the reduced risk of other types of cancer associated with the FAS gene.
Dr. Wang Y, the lead scientist wrote in the final report, "Since obesity is closely related to breast cancer and obese patients are at elevated risk of developing various cancers, these findings suggested that onion might be useful for preventing obesity-related malignancy".
Additionally, in the examination of the anticancer effects on five onion varieties grown in Ontario against human adenocarcinoma (Caco-2) cells, the University of Chinese Academy of Sciences suggested
* Onion extracts and pure extracts of flavonoids exhibit antiproliferative effects against the tested cancer cell through its major phytochemical quercetin, myricetin, and kaempferol.
* Onion varieties also exhibited antiproliferative activity similar to purified flavonoids, according to the lactate dehydrogenase (LDH) assays.
* The Safrane extracts showed the weakest antiproliferative activity compared to other variety.
Taken together, onion may be considered a remedy for the prevention and treatment of cancer via antiproliferative activity, pending the confirmation of the larger sample size and multicenter human study.

#Herbal Comfrey Extract Exerts A Significant Wound Healing Effect on #Pediatry Uses
Kyle J. Norton
Wound healing is the natural process by covering up the damage and injury of the body,Skin wound healing is a process to repair skin damage and injury.
The process involves 4 phases
* The first phase of wound healing is to cover up the wound. In this phase, the immune first line of defense white blood cells stimulates the emergency repair system in the production of blood clotting to block drainage.
* In phase 2, the white blood cells activate the proteins associated with the inflammatory cytokines to destroy the pathogens before they can enter the body through the wound.
In most cases, due to the overproduction of proinflammatory cytokines, patients may experience symptoms of fever, localized pain, redness, and swelling.
Furthermore, the production of pro-inflammatory cytokines in some cases can cause the death of nearby healthy cells, leading to the formation of irreversible scars, as seen in the liver.
* In phase 3, new blood vessels and tissues are formed in the damage and injury area.
* The recovery phase.
The final phase of wound healing is involved in the remodeling of collagen from type III to type I cellular activity reduces.
However, the healing process is easier said than done, the complex process in many cases is interrupted by local and systemic factors, including moisture, infection, age, and nutritional status, leading to abnormal cell membrane repair, a major cause of skin scars.
Comfrey is a perennial plant in the genus of Symphytum, belonging to the family Boraginaceae, native to Europe. It has been used in herbal and traditional medicine to ease excessive bleeding or on open wounds, relieve pain and swelling, treat circulation issues, reduce cholesterol, etc.
In finding a potential compound to speed up injury recovery, researchers investigated the wound healing effects of a topically applied preparation (Traumaplant) containing a concentrate (10 % active ingredient) from the aerial parts of medicinal comfrey (Symphytum x uplandicum NYMAN).
A randomized clinical double-blind study including 278 patients with fresh abrasions (verum n = 137), among them, 64 patients of up to 20 years of age (verum n = 29, reference product n = 35)with an otherwise identical low-dose preparation (1 % active ingredient; n = 141) used as a control.
According to the results of the examination, after 2-3 days of applied medication exerted a highly significant and clinically relevantly faster initial reduction of wound size of 49 + or - 19 % versus 29 + or - 13 % per day in favor of verum (p < 5x10(-21))compared to control.
Furthermore, the linear regression time to complete healing was 2.97 days faster with verum compared to the control reference.
The physicians rated efficacy as good to very good in 93.4 % of cases, as compared to only 61.7 % in the control.
Moreover, on a scale of 0-100, verum was rated with 84.4 + or - 10.1 points by the patients themselves, compared to the rate of 65.5 in the control.
No adverse effects or problems with drug tolerability occurred. Specifically, the cutaneous reactions throughout the 10-day observation phase.
Based on the findings, researchers said, "Symphytum herb extract can be attributed to distinct wound healing effects, effects that can explicitly be used in paediatry.".
Taken altogether, Comfrey may be considered a remedy for speeding up wound healing, pending the confirmation of the larger sample size and multicenter human study.

#Tomatoes and #BetaCarotene Improve the Skin Health through Their Antioxidant Activity, Researchers Say
Kyle J. Norton
Tomatoes may have a profound and positive effect on improved skin health, some scientists suggested.
The results of analysis were carried out by several universities and research companies, including IUF - Leibniz Research Institute for Environmental Medicine and published on numbers of online literature.
Tomato is red, edible fruit, genus Solanum, belongings to the family Solanaceae, native to South America. Because of its health benefits, tomato is grown world wide for commercial purpose
and often in the green house.
In the aim to reaffirm tomatoes' effect in reduced negative impact against ultraviolet (UV)A/B and UVA1 radiation at a molecular level, scientists at the IUF - Leibniz Research Institute for Environmental Medicine launched a study of the tomato bioactive compounds lutein and lycopene photoprotective effect on human skin health.
The purpose of the study was to assess the two active treatments containing either lycopene (TNC) or lutein to test for their capacity in decreased expression of UVA1 the radiation-inducible genes HO1, ICAM1, and MMP1
Gene heme oxygenase 1 (HO1) is an enzyme with function to to protect cells from oxidative damage, in this case, it was the UVA1
Gene intercellular adhesion molecule 1 (ICAM1) also known as CD54 (Cluster of Differentiation 54) is a protein with function in adhesion phenomena involved in the immune response.
Matrix metalloproteinase-1 (MMP-1) also known as interstitial collagenase and fibroblast collagenase is an enzyme with function to break down collagens located in the ECM.
The placebo-controlled, double-blinded, randomized, crossover study included 65 healthy volunteers allocated to four treatment groups and subjected to a 2-week washout phase, followed by two 12-week treatment phases separated by another 2 weeks of washout.
Volunteers started either with active treatment and were then switched to placebo, or vice versa.
24 hours after the irradiation phase skin, biopsies were taken from untreated, UVA/B- and UVA1-irradiated skin for subsequent reverse transcriptase polymerase chain reaction analysis of gene expression.
Interestingly, after taking into account of other co and con founders, researchers found that lycopene isolated from tomato exerts a strong activities in completely inhibited UVA1- and UVA/B-induced upregulation of heme-oxygenase 1, intercellular adhesion molecule 1, and matrix metallopeptidase 1, through restricting the transformation of mRNA in initiated cell changes.
Blood samples taken from lycopene treatment group, showed a clinically relative protective effect of skin cells through its inhibition of levels of oxidative stress induced by UVA1- and UVA/B exposure in compared to other groups.
Compared to lycopene active group, a group treated with lutein injection provided complete protection taken in the first period but showed insignificantly effects in the second sequence.
These results suggested that there must be some unknown mechanisms to cause the reduced levels of
lutein in the second sequence compared to TNC.
Dr. Grether-Beck S, the lead author said, " the role of these genes as indicators of oxidative stress, photodermatoses, and photoageing" and " these results might indicate that TNC and lutein could protect against solar radiation-induced health damage".
Furthermore, in re evaluate the effect of Carotenoids in skin photoprotection against UV radiation through analysis of the collection of dermal biopsies of healthy humans (N=27) and blood samples, researchers reconfirmed that lycopene and beta-carotene made up the majority of carotenoids in both skin and plasma followed by beta-cryptoxanthin.
These expressions suggested that lycopene and beta-carotene may play important role in improved skin health through its antioxidant activity in compared to other members of carotenoids such as
dihydroxy carotenoids, lutein, and zeaxanthin.
In other words, skin of human is relatively enriched in lycopene and beta-carotene, compared to lutein and zeaxanthin, possibly reflecting a specific function of hydrocarbon carotenoids in human skin photoprotection.
Additionally, in the examine the skin collected from 74 men and women with diverse skin pigmentation using the Resonance Raman spectroscopy (RRS) to measure the palm, inner arm, and outer arm were obtained at baseline, 1 wk, 2 wk, 1 mo, 3 mo, and 6 mo (to maximize seasonal variation), researchers with no surprise found that total carotenoids across time are 0.97 (palm), 0.95 (inner arm), and 0.93 (outer arm).
Similarly, the assessment of lycopene by RRS was significantly correlated with lycopene assessed by HPLC of dermal biopsies (r = 0.74, P < 0.0001).
These finding once again indicated the protective effect of tomato and bioactive ingredient lycopene in skin health in compared to other compounds.
Taken together, there is no doubt that tomato with its major bioactive phytochemical may be considered as functional food for improved skin health against skin damage caused by number of negative implications including oxidative stress, photodermatoses and photoageing

#Antioxidant #OleanolicAcid Inhibits #GastricCancer by Enhancing Apoptosis and #eversing Multidrug Resistance (MDR) of Cancer Cells, According to Studies
Kyle J. Norton
The stomach located between the esophagus and the small intestine is a muscular, hollow, and important organ of the digestive tract.
Gastric/Stomach cancer is a chronic condition characterized by irregular cell growth in the tissues of the stomach.
Most cases of gastric cancer start in the cells on the surface of the inner lining of stomach tissue.
Due to the small size at the early stage, gastric cancer is asymptomatic.
However, at the later stage, after penetrating into the deeper layers of the stomach, the cancer cell may travel a distance away to infect other healthy tissues and organs, leading to secondary metastasis.
At this stage, most patients with gastric cancer may experience symptoms of severe abdominal pain caused by the suppression of the oversize tumor on the nerve cells, or vague discomfort in the abdomen, usually above the navel, particularly heartburn and indigestion, accompanied by general symptoms shared with other types of cancer.
According to the statistics provided by the American Cancer Society, in 2019, approximately, 27,510 cases of stomach cancer will be diagnosed (17,230 in men and 10,280 in women) and 11,140 people will die from gastric cancer (6,800 men and 4,340 women).
Overall, the 5-year survival rate for people with gastric cancer is 31%. Cancer diagnosed at the localized stage have a 5 years survival rate of 67%.
Common sites metastases of gastric cancer are liver (48%), peritoneum (32%), lung (15%), and bone (12%).
The most common causative risk factor of gastric cancer is infection with a common bacteria, H. pylori, which was found to cause ulcers and Inflammatory gastritis,.
Oleanolic acid is a phytochemical in the subclass of Triterpenoid, belonging to the group of Terpenes found abundantly in honey mesquite, garlic, java apple, clove, etc.
In finding a potential plant that processes anti-cancer activity researchers examined the effect of a
synergism of cisplatin-oleanolic acid on colon cancer.
According to the results of the analysis of cancer cell membrane (CM) decorated calcium carbonate (CC) hybrid nanoparticles (HN) for the co-delivery of cisplatin (CDDP) and oleanolic acid (OA),* The -prepared DDS was core-shell structured and well-dispersed nanoparticles with a size around 100 nm.
* The HN/CDDP/OA showed high stability and biocompatibility with pH-responsive drug release.
* The CM modification in HN also demonstrated a highly elevated tumor-homing nature than barely decorated calcium carbonate (CC).
* HN/CDDP/OA in the treatment of gastric cancer (MGC-803 cell line) showed better performance than mono systems with enhanced apoptosis and capable of reversing multidrug resistance (MDR) of cancer cells.
Taken together, Oleanolic acid may be considered a remedy or the prevention and treatment of gastric cancer, pending the confirmation of the larger sample size and multicenter human study.

#PintoBeans Inhibit α-Amylase Associated with the Causes of Hyperglycemia and Development of Type 2 Diabetes, According to Study
Kyle J. Norton
Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream to regulate glucose.
In other words, diabetes occurs when cells in the pancreas die off or receptor sites clogged up by fat and cholesterol. In some cases, diabetes is also caused by allergic reactions of cells in the immune system.
Type 2 diabetes is a medical condition characterized by insufficient insulin entering the bloodstream to regulate glucose due to either cells in the pancreas dying off or receptor sites clogged up by fat and cholesterol.
Researchers do not know what causes the pancreas to die off or receptor sites to clog up. However, they do know obesity or overweight, gender, ethnicity, age, family history, and gestation diabetes, are associated with an increased risk of type 2 diabetes.
Some researchers also suggested that people with certain medical conditions such as polycystic ovary syndrome. and high blood pressure is more prone to the onset of the disease.
The most common symptoms of diabetes are excessive thirst and hunger, frequent urination, unintended weight loss or gain, frequent fatigue, irritability, and gastrointestinal symptoms such as nausea.
In severe cases, patients may also experience symptoms of blurred vision and slow wound healing or ulcer.
Hyperglycemia, a medical condition of abnormally high blood glucose, is a major concern, in people with both type 1 and type 2 diabetes.
Most cases of hyperglycemia occurred in patients who do not manage their diabetes properly.
Hyperglycemia, a hallmark of diabetes has been found to induce diabetics complications, including neuropathy, nephropathy, and retinopathy). ...
On finding a natural ingredient for the prevention and treatment of chronic diseases associated with oxidative stress, scientists examined the effects of bioactive peptide fractions from de-hulled hard-to-cook (HTC) beans on enzyme targets of type-2 diabetes and oxidative stress.
The tested Protein isolated from Pinto Durango and Negro 8025 beans were hydrolyzed (120min) with either alcalase® or bromelain and separated into five peptide fractions.
The pinto bean is also known as the common bean, genus Phaseolus and the family Fabaceae. It has a mottled appearance of darker reddish-brown spots on a lighter brownish-beige background, thus appearing painted or spotted.
According to tested analysis,
* The <1kDa pinto Durango-bromelain fraction showed the best inhibition of α-amylase (49.9±1.4%) associated with the causes of hyperglycemia and development of type 2 diabetes.
* Furthermore, the <1kDa pinto Durango-alcalase fraction inhibited both, α-glucosidase (76.4±0.5%), and dipeptidyl peptidase-IV (DPP-IV, 55.3±1.6%) involved in the elevated blood glucose levels.
* Hydrolysates and peptide fractions showed antioxidant capacity (ORAC: 159.6±2.9 to 932.6±1.1mmolTE/g) and nitric oxide inhibition (57.5±0.9 to 68.3±4.2%) that inhibit the overexpression of oxidative stress found in type 2 diabetics.
* Moreover, the hydrolysates and fractions <1 and 1-3kDa were able to increase glucose-stimulated insulin secretion from iNS-1E cells up to 57%, compared to glucose control.
Based on the results, researchers suggest, " Hydrolysates from HTC beans inhibited enzymes related to diabetes management, being the smallest peptides (<1kDa) the most potent. HTC bean could be a source of protein to produce bioactive peptides with potential antidiabetic properties".
Taken together, Pinto beans may process antioxidant and anti-diabetes properties, pending the confirmation of the larger sample size and multicenter human study.

#Phytochemical #Lycopene Protects the Liver Cells Against the Chronic or Excessive Alcohol Intake-Related Liver Injury
Kyle J. Norton
Nonalcoholic fatty liver disease (NAFLD) is a medical and chronic liver condition associated with fat accumulated over time, without the presence of excessive alcohol use.
NAFLD can be classified into non-inflammatory fatty liver (NAFL) and inflammatory nonalcoholic steatohepatitis (NASH).
Untreated Nonalcoholic fatty liver disease (NAFLD) has been found to induce cirrhosis, a major cause of liver cancer.
According to world statistics, nonalcoholic fatty liver disease (NAFLD) is normally known as a disease of the Western world. However, today, due to the economic prosperity of Southeast Asia over the past few decades,, the disease also was found in a large number of populations in the cities, causing concerns of many scientists in the region.
The prevalence rate of non-alcoholic fatty liver disease (NAFLD) among the more affluent regions of China, is approximately 15%. The number may decrease substantially if the poor rural populations where obesity is non-existence are also taken into account.
In the US, over 100 million people have non-alcoholic fatty liver disease and the condition is more doubled over the past 20 years. Today, the disease also is also prevalent in liver disease in children.
Most people at the early stage of NAFLD are asymptomatic. However, as the disease progresses into the later stage, most patients experience symptoms of right upper abdominal discomfort, fatigue, and/or malaise, and jaundice(20) with yellowing of the skin and eyes.
The exact causes of NAFLD are not identified. Some researchers suggested that certain risk factors such as long-term use of certain medications, genetic preposition, insulin resistance, high cholesterol and triglycerides, polycystic ovary syndrome, metabolic syndrome, obesity, and type 2 diabetes are associated with the onset of the disease.
Some researchers suggested that an unhealthy diet such as the promotion of a high-fat diet in the Western world over the last few decades may also have a strong implication on NAFLD.
Dr. Jensen VS, the lead scientist in the study of high-fat diet-induced non-alcoholic fatty liver disease, wrote, "In humans and animal models, excessive intake of dietary fat, fructose, and cholesterol have been linked to the development of non-alcoholic fatty liver disease (NAFLD)".
And, " Only HFr-fed rats developed dyslipidemia as characterized by higher levels of plasma triglycerides compared to all other groups (p < 0.0001). Hepatic dysfunction and inflammation were confirmed in HFD-fed rats by elevated levels of hepatic MCP-1 (p < 0.0001), TNF-alpha (p < 0.001), and plasma β-hydroxybutyrate (p < 0.0001), and in NASH-fed rats by elevated levels of hepatic MCP-1 (p < 0.01), increased hepatic macrophage infiltration (p < 0.001), and higher plasma levels of alanine aminotransferase (p < 0.0001) aspartate aminotransferase (p < 0.05), haptoglobin (p < 0.001) and TIMP-1 (p < 0.01) compared to Control".
Lycopene is a phytochemical in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,
Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.
In finding a potential compound for the prevention of alcoholic fatty liver disease, researchers evaluated the effect of lycopene (LYC) on chronic or excessive alcohol intake-related liver injury.
Lycopene inhibited the expression of cytochrome P450 2E1 (CYP2E1) enzyme associated with chronic and excessive alcoholic drinking.
Where cytochrome P450 2E1(CYP2E1) not only represents the major enzymes in the CNS that catalyze ethanol oxidation but also plays a vital role in alcohol, drug, toxin, lipid, and carcinogen metabolism associated with reactive oxygen species-induced oxidative stress that promotes hepatocyte injury and death.
In other words, excessive alcohol drinking induced the production of free radicals that lead to liver damage in the initiation of inflammation, fibrosis, and lipid peroxidation.
More precisely, lycopene prevented liver injury in the induction of alcoholic fatty liver disease by targeting the expression of CYP2E1 induction.
Taken together, lycopene found in tomatoes may be considered supplements for the prevention and treatment of alcoholic fatty liver disease, pending the confirmation of the larger sample size and multicenter human study.
Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
