#Extract Polyphenols from #Eggplant Inhibit Peroxidation of Cerebral and Hepatic Lipids through Their Antioxidant Activity

Scientists may have found a whole vegetable that processes the function of balancing the ratio of free radicals and antioxidants, some studies suggested.

Free radicals are unstable molecules with an unpaired electron in the outer ring. In order for them to become stable, free radicals always seek other atoms or molecules to bond to leading to a domino's effect that cannot stop until they are either paired or inhibited by antioxidants

The continuation of this domino's effects is known to begin a process called oxidative stress. A prolonged period of overexpression of free radicals can cause oxidative stress, a major cause of some chronic diseases, including diabetes.

Truly, free radicals and oxidants play a dual role as both toxic and beneficial compounds. On the benefit side, according to Wanjek wrote in 2006, "The body's ability to turn air and food into chemical energy depends on a chain reaction of free radicals. Free radicals are also a crucial part of the immune system, floating through the veins and attacking foreign invaders".

Free radicals can promote toxicity to cancer cells, leading to antiproliferative and apoptotic effects without harming the nearby healthy cell if administered with dietary antioxidants or stimulated the production of antioxidants enzymes in the host through healthy food sources.

On the other hand, overexpression of free radicals can be very harmful, Dr. Lien Ai Pham-Huy, the lead author in the study "Free Radicals, Antioxidants in Disease and Health" said, "When an overload of free radicals cannot gradually be destroyed, their accumulation in the body generates a phenomenon called oxidative stress. This process plays a major part in the development of chronic and degenerative illnesses such as cancer, autoimmune disorders, aging, cataract, rheumatoid arthritis, cardiovascular and neurodegenerative diseases".

"The human body has several mechanisms to counteract oxidative stress by producing antioxidants, which are either naturally produced in situ or externally supplied through foods and/or supplements".

These analyses suggested that a healthy diet and lifestyle play a critical role in reducing the risk of imbalance of the ratio of free radicals and antioxidants by increasing the expression of antioxidants and decreasing the risk of overload of free radicals.

Eggplant is a species of S. melongena, belonging to the family Solanaceae and native to India. The plant has been cultivated in southern and eastern Asia since prehistory.

It is famous for its deep purple and the best eggplant can be found from August through October when it is in season.

The vegetable chemical compounds include sterols (i.e. typical plant sterols, androstane, pregnant, and cholestane derivatives, steroidal alkaloids, and sapogenins), phytosterols, triterpenes, δ-amyrin, Chlorogenic acid, Nasunin, and other Polyphenolic compounds.

In reaffirming the effects of eggplant-rich source of polyphenol compounds endowed with antioxidant properties, researchers launched an evaluation to compare the antioxidant activity of the extract polyphenols from eggplant entire fruit, pulp, or skin, both fresh and dry and conventional extraction and microwave-assisted extraction (MAE).

* SAG exhibited potent and concentration-dependent free radical-scavenging activity (IC50/DPPH=275.03±7.8 μg/mL).

* Reductive and iron chelation abilities also increase with an increase in SAG concentration.

* SAG also inhibited peroxidation of cerebral and hepatic lipids subjected to iron oxidative assault.

The effect of eggplant in reducing the expression of free radicals was attributed to the presence of major bioactive compounds, including gallic acid [(17.54±0.04) mg/g], chlorogenic acid (21.90±0.02 mg/g), caffeic acid (16.64±0.01 mg/g), rutin [(14.71±0.03) mg/g] and quercetin [(7.39±0.05) mg/g].


Dr. Elekofehinti OO, the lead scientist said, "SAG extract is a potential source of natural antioxidants that may be used not only in the pharmaceutical and food industry but also in the treatment of diseases associated with oxidative stress".

Taken together, there is no doubt that eggplant processes a powerful antioxidants activity in the prevention of overexpression of free radicals in the initiation of oxidative stress and combined with a primary theory for the treatment of oxidative stress-related diseases, depending on the confirmation of further data collection from the large example size and multi-center studies in human.

The randomized, placebo-controlled, double-blind 12-week study included patients with schizophrenia who received 1,000 mg of standardized WSE. conducted by the University of Pittsburgh Medical Center.

Based on the 66 patients with depression symptoms, the Positive and Negative Syndrome Scale. showed the WSE exerted medium effect sizes of 0.683 (95% confidence interval [CI], 0.16 to 1.21) compared to placebo.

More importantly, the adverse effects were mild and transient, according to patient reports.

Collectively, researchers wrote after taking other factors into account, "WSE may hold promise in the treatment of depression... symptoms in schizophrenia".

* More precisely, prudent dietary pattern and carotenoid component scores were negatively associated with serum hs-CRP concentration (unstandardized β for a prudent component: -0·053, 95% CI -0·102, -0·003; carotenoid component: -0·183, 95% CI -0·233, -0·134).

Where, a prudent dietary pattern (which reflects a diet high in the intakes of fish, yogurt, pulses, rice, pasta, and wine, in addition to a fruit and vegetable-rich with carotenoids diet.

In other words, a prudent diet rich in carotenoids is associated with a reduced risk of chronic inflammatory diseases.

Taken together, carotenoids may be considered an adjunct therapy in the treatment of diseases associated with chronic low-grade inflammation, pending the confirmation of the larger sample size and multicenter human study.

Intake of carotenoids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Based on the results, researchers suggested, consumption of large amounts of tomato products daily and regularly induced high levels of lycopene in the blood is associated with a reduced risk of gastric cancer.

Taken together, lycopene found in tomatoes may be considered supplements for the prevention and treatment of gastric cancer, pending the confirmation of the larger sample size and multicenter human study.

* Pretreatment with 50 mg/kg of each compound induced a significant antihyperglycemic effect after glucose and sucrose loading (2 g/kg) compared with the control group (p<0.05).

* Moreover, in vitro studies showed that compounds 1 and 2 induced inhibition of 11β-HSD 1 associated with hepatic glucose output activity at 10 μM.


Based on the findings, researchers said, "moronlic and moronic acids have shown sustained antidiabetic and antihyperglycemic action possibly mediated by an insulin sensitization with consequent changes of glucose, cholesterol, and triglycerides, in part mediated by inhibition of 11β-HSD 1".

Taken together, moronic acids may be considered supplements for the treatment of hyperglycemia, pending the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Scientists may have found a natural remedy for the prevention and treatment of oral cancer with no side effects, according to studies.

Oral cancer also known as mouth cancer is a medical condition characterized by irregular cell growth in the tissue of the oral cavity, due to the alternation of cell DNA.

Most cases of oral cancer begin in the cells on the surface of the inner lining of oral tissue.

However, at the advanced stage, oral cancer cells can travel a distance away to infect other healthy tissue and organs, leading to secondary metastasis.

According to the statistic provided by the American Cancer Society, in 2018, approximately 51,540 people were diagnosed with oral cavity or oropharyngeal cancer. The disease causes the death of an estimated 10,030 people.

The 5-year survival rate of oral cancer is 66%, including all stages.

Family history, smoking, alcohol drinking, being infected by the HPV virus, poor hygiene, and genetic preposition are some of the risk factors associated with the disease.

These results strongly suggested people who perform oral sex before, during, and after sexual intercourse may be at risk of oral cancer due to being infected by the HPV virus.

Researchers on finding a natural compound for the inhibition of oral cancer examined the [6]-Shogaol ([6]-SHO), a major constituent of ginger anti-tumor activity.

[6]-SHO showed a significant effect in preventing oral squamous cell carcinoma by activating proapoptotic factors in vitro and in vivo experimental models.

In the inflammation and cell proliferation assays, injection of [6]-SHO inhibited the oral cancer cell line through cytotoxicity induced by the expression of proinflammatory cytokines, proteins associated with cell apoptosis, and anti-cell survival mechanisms.

Furthermore, in the oral cancer cells, oral administration of [6]-SHO strongly inhibited phosphorylation in stimulating the enzymes which carry energy to the cancer cells and enzymes in the degradation of cellular response to inflammation.

Dr. Annamalai G, the lead scientist said, " [6]-SHO is a novel functional agent capable of preventing DMBA-induced inflammation and cell proliferation-associated tumorigenesis by modulating multiple signaling molecules".

Additionally, in order to reveal more information about 6-Gingerol anti-oral cancer properties, researchers investigated the compound in human oral (SCC4, KB) with or without wortmannin, rapamycin, and cisplatin.

6-Gingrol showed dose-dependent cytotoxicity in the oral cell line.

Combinations of 6-gingerol with wortmannin and cisplatin showed additive effects, while rapamycin, it showed 50% cytotoxicity equivalent to that of IC50 of 6-gingerol alone.

Treatment with 6-gingerol exhibited S-phase arrest in SCC4 cells. But 6-Gingerol, wortmannin, and rapamycin treatment showed almost two-fold higher expression of proteins associated with cancer cell apoptosis.

Dr. in the final report wrote," 6-gingerol appears to be a safe and potent chemotherapeutic/chemopreventive compound acting through cell cycle arrest and induction of apoptosis in human oral....cells".

Taken together, ginger processed bioactive ingredient 6-gingerol and [6]-Shogaol may be considered a functional remedy for the prevention and combined with primary medicine for the treatment of oral cancer, pending the confirmation of a large sample size and multicenter human study.

#Bromelain Reduces Neutrophil Migration Against the Initiation of Inflammation, According to Studies

Acute inflammation is a natural response of the immune system to protect the human body against harmful stimuli and pathogens.

Acute information is the first phase of injection. At this phase, after sensing the body injury by the central nervous system, the immune first line of defense stimulates the production of blood platelets to cover the site of damage and pro-inflammatory to kill off pathogens that try to enter the body through the wound or damaged tissue.

Acute inflammation usually receded after a few days or weeks depending on the size and damage. However, if all pathogens were not eradicated after 3 to 8 weeks, the immune system will compromise, leading to chronic low-grade inflammation, a leading cause of chronic illness.

The most common symptoms of acute inflammation occurred within a short period of time such as hours pain, redness, immobility, swelling, and heat.

Conventionally, depending on the types of acute inflammation, non-steroidal anti-inflammatory drugs (NSAIDs) may be prescribed to alleviate the pain by counteracting an enzyme that contributes to inflammation.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

In finding a potential compound for the treatment of acute inflammation, researchers examined the effect of bromelain, a mixture of proteases derived from pineapple stem on inflammatory diseases, including murine inflammatory bowel disease.

According to the tested assays,* In vitro, bromelain modulated immune activity by removing a number of cell surface molecules that are vital to leukocyte trafficking.
* Bromelain inhibited neutrophil migration to IL-8 and decreased acute responses to inflammatory stimuli in the initiation of the production of pro-inflammatory cytokines.

* In vivo, bromelain treatment generated a 50-85% reduction in neutrophil migration in 3 different murine models of leukocyte migration into the inflamed peritoneal cavity through its anti-inflammatory properties.

In other words, through its anti-inflammatory activity, bromelain inhibited the primary long-term effect on the abrogation of firm adhesion of leukocytes to blood vessels at the site of inflammation in the induction of inflammation.

Additionally, in an ovalbumin (OVA)-induced murine model of acute allergic airway disease (AAD) with female C57BL/6J mice sensitized with intraperitoneal (i.p.) OVA/alum and then challenged with OVA aerosols for 3 days, administration of 200 mg/kg bromelain twice daily for four consecutive days, exerted a significant anti-inflammatory activity against the overexpression of pro-inflammatory cytokines caused by the injection of ovalbumin (OVA).

Interestingly, bromelain also inhibited allergic inflammation by modulating the immune response against ovalbumin (OVA).

Based on the findings, researchers wrote, " these studies demonstrate that bromelain can effectively decrease neutrophil migration to sites of acute inflammation".

Taken altogether, bromelain may be considered supplements for the prevention and treatment of acute inflammation, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
