#Herbal #Coltsfoot inhibits the Production of Inflammatory Mediators and Cytokines by Suppressing the Immune Response

The immune system formed part of the body's defense plays a critical role in the protection of the body against infectious organisms.

The immune system executes the defense strategy through a series of immune response by attacking foreign substances that invade body systems.

A variety of factors have been found to trigger immune responses including pathogens, damaged cells, and toxic compounds.

Inflammation is a natural response of the immune system which protects the body against the invasion of pathogens.

In the acute phase of tissue damage or wound, the immune white blood cells of the first line of defense stimulate the production of a blood palette to cover the wound or damage and production of pro-inflammatory cytokines to the site of infection against microbial invasion.

Most cases of microbial invasion are stopped in the acute phase of infection, and the damaged tissue is slowly healing itself. However, in this phase, the overproduction of pro-inflammatory cytokines can lead to nearby cell death and the formation of scars.

If the immune system fails to destroy all microbial invasion in the acute phase of infection within a set period in 3 to weeks, it will compromise, leading to chronic low-grade inflammation.

The continued production of low levels of pro-inflammatory cytokines overtimes regardless of the causes can induce damage to organ tissues, and irreversible chronic diseases such as cardiovascular disease, diabetes, and cancer.

The Research Center for Molecular Medicine of the Austrian Academy of Sciences (Ce-M-M) wrote, "Humans have evolved highly complex immune systems that are composed of specialized immune cells that protect the host from infection and damage. But an uncontrolled immune response such as chronic inflammation can itself inflict damage".


And, "when the immune system is compromised by a genetic mutation, individuals become more susceptible to diseases such as autoimmune disease".

Coltsfoot(Tussilago farfara) is a perennial herbaceous plant, genus Tussilago, belonging to the family Asteraceae, native to Europe and Asia. The herb has been used for thousands of years to treat asthma, various coughs, bronchial congestion, respiratory disorders, headaches and obstruction in the nasal passage, etc.

On finding a potential compound for the treatment of asthma and bronchitis, researchers examined the coltsfoot inhibitory effects on lipopolysaccharide-induced inflammation in macrophages.

In a cecal ligation and puncture (CLP)-induced mouse model of sepsis, Tussilagone suppressed the parameters associated with the production of pro-inflammation such as nitric oxide and prostaglandin E2, and the inflammatory cytokines, tumor necrosis factor-alpha (TNF-α), particularly in lipopolysaccharide-stimulated RAW 264.7 cells and peritoneal macrophages.

In other words, Tussilago inhibits the production of inflammatory mediators and cytokines by suppressing the immune response associated with the first line of defense.

Additionally, the administration of Tussilagone at doses of 1 mg/kg and 10 mg/kg) protects the lung cells by decreasing mortality and lung injury in CLP-activated septic mice.

Based on the findings, researchers said, " Tussilago exhibited protective effects against inflammation and polymicrobial sepsis by suppressing inflammatory mediators possibly via the inhibition of NF-κB activation and the MAP kinase pathway".

And, " These results suggest the possible use of Tussilago for developing novel therapeutic modalities for sepsis and other inflammatory diseases".

Taken altogether, coltsfoot may be considered a remedy for the treatment of inflammatory diseases associated with immune-inflammatory responses, pending the confirmation of the larger sample size and multicenter human study.

Carrots may have a profound and positive effect on reducing weight loss in the obese population, some scientists suggested.

Most researchers agreed that obesity is caused by a genetic mutation inherited from the parents, a long-term unhealthy diet with consumption of high amounts of energy foods, particularly fat and sugars, and physical inactivity.

Other causative factors also include certain medical conditions such as hypothyroidism and Cushing's syndrome and the long-term use of certain medications such as corticosteroids and antidepressants.

The general belief also suggested that some smokers may gain weight if they quit smoking.

According to the study conducted by the J University of Georgia, using a combined approach to promote nutrition and physical activity policies and practices in early care and education may reduce the risk of child obesity.

Estrogen-deficient animals with diet-induced obesity were examined in the study by the Obesity/Diabetes Center, Asan, Korea using black carrots to test the mechanisms of the long-term consumption of unfermented black carrot extract (BC) and its extracts fermented with Lactobacillus plantarum (BCLP) or Aspergillus oryzae (BCAO) in the prevention of menopausal symptoms including impaired energy, glucose, and lipid metabolism

During the experiment, ovariectomized (OVX) rats were fed four different high-fat diets containing 2 % dextrin (OVX-control), 2 % BC, 2 % BCLP, or 2 % BCAO for 12 weeks. Sham rats were fed high-fat diets containing 2 % dextrin.

Fat mass and weight gain in tested mice were lower in descending order of OVX-control > BC and BCLP > BCAO due to increased energy expenditure and fat oxidation.

BC, BCLP and especially BCAO all normalized HOMA-IR, an indicator of insulin resistance and glucose intolerance.

Furthermore, BCAO markedly decreased hepatic triglyceride levels by increasing gene expressions of CPT-1 and PPAR-α, which are involved in fatty acid oxidation, and decreasing mRNA expressions of FAS and SREBP-1c, which are associated with fatty acid synthesis.

More importantly, Cyanidin and malvidin markedly decreased fat accumulation in 3T3-L1 adipocytes by increasing CPT-1 and decreasing FAS expression in the liver fat metabolism and SREBP-1c expression in the regulation of the genes with the function of lipid metabolism in comparison with cyanidin-3-rutinoside, an anthocyanin, and malvidin-3,5-diglycosides, another anthocyanidin.

Where 3T3-L1 is a cell line derived from 3T3 cells used in biological research on adipose tissue.

These results suggested that unfermented black carrot extract (BC) and its extracts fermented with Lactobacillus plantarum (BCLP) processed an anti-obesity mechanism by preventing the exacerbation of lipid and glucose metabolism, the major causes of weight gain.

However, further data collection on large example size and multi-centers studies performed with human consumption of whole food during the course of the disease will be necessary to complete the picture of carrot's anti-weight gain possibilities.

Moreover, LPS also exhibited overproduction of nitric oxide (NO) observed in bronchoalveolar lavage fluid in facilitated cellular cytotoxicity.


In the compared nontreatment group, the injection anethole group demonstrated a strong anti-inflammatory effect through decreased activities of proinflammatory factors without affecting the immune activity in protecting against infection.

Additional analysis also found that the treatment group reduced acute lung injury caused by the injection of LPS through its antioxidants and induced production of host natural antioxidants in the body tissue.

The inhibited LPS results according to were attributed to anethole effect in suppressing the activation of NF-κB in signaling the production of inflammatory cytokines by blocking the initiation of IκB-α degradation by inhibited TRAF2 in the induction of inflammation through TNF receptor superfamily members and NIK, a key signaling molecule of the noncanonical NF-κB.

Interestingly, further investigation of anethole as an inhibitor of TNF-induced NF-kB activation (an early response) as monitored by electrophoretic mobility shift assay, by suppression of IkBa phosphorylation and NF-kB reporter gene, researchers also indicated that

* Anethole also suppressed TNF-induced activation of the transcription factor AP-1 in regulated gene expression in response to a variety of stimuli, including LPS application.

*c-Jun N-terminal kinases (JNKs) with function in response to stress stimuli, such as cytokines,
And

* MAPKkinase in signals inflammatory response through transmitting information from a receptor on the surface of the cell to the DNA in the nucleus of the cell.


* Application of anethole also suppressed TNF-induced both lipid peroxidation and ROS generation, through the stimulated production of natural antioxidants in the host and itself, resulting in balancing the ratio of antioxidant and ROS expression.

The illustration and differentiation were elite by the experiment in the application of TNF (100 pM) with the cells pretreated for 240 and 120 min with anethole, NF-kB activation was almost completely inhibited.

However, the inhibition decreased gradually with decreased pre-incubation time.

Kimchi and Amtak Baechu Cabbage Inhibit #PancreaticCancer by Inducing Cell Apoptosis and Cell Cycle Arrest, Researchers Say

The pancreas is an oblong flattened gland in the digestive and endocrine system located in the abdomen. By secreting pancreatic juice to the small intestine, the pancreas aids the digestive system to break down carbohydrates, proteins, and lipids.

Traditional Chinese medicine suggested that the pancreas is a vital organ responsible for the absorption of foods' qi and nutrients before passing them to other organs to nourish our body.

Pancreatic cancer is a chronic condition characterized by cell growth irregularly in the tissues of the pancreas.

Most cases of pancreas cancer start in the cell on the surface of the inner lining tissue. However, as the cancer progress, the malignant cells of the pancreas, although unusual may travel a far distance away to infect other healthy tissue and organs.

The pancreas cancer was found to be the site of metastasis from renal cell cancer, lung, colon, and breast cancers.

According to statistics, every year, approximately, over 55,000 adults in the United States are diagnosed with pancreatic cancer, accounting for 3% of all cancers and 7% of all cancer deaths.

The survival rate of pancreatic cancer even if diagnosed early is very low compared to other types of cancer. The 5-year survival rate at stage IA pancreatic cancer is only14%.

The exact causes of pancreatic cancer are identified. However, epidemiological studies suggested that the increase in age, family history, race, gender, smoking, being obese or overweight and a medical condition such as diabetes is the most prevalent risk of the disease.

The genetic and acquired genetic mutation of p16 and TP53 and KRAS, BRAF, and DPC4 (SMAD4) were also found in patients with pancreatic cancer.

In finding a potential plant that processes anti-cancer property activity researchers investigated the effects of kimchi prepared using the standard Baechu cabbage and Amtak Baechu cabbage on hepatic cancer cells.

The study included 4 different kimchi samples prepared for this study which are t(1) the standard Baechu cabbage and dry salt treatment method (SK-D); (2) the Amtak Baechu cabbage and dry salt treatment method (AK-D); (3) the standard Baechu cabbage and brine solution treatment method (SK-B); and (4) the Amtak Baechu cabbage and brine solution treatment method (AK-B).


Based on the results of the analysis,
* The liver cancer cells showed a significant increase in cell growth inhibition when exposed to AK-D and AK-B compared with SK-D and SK-B (p < 0.05). At 2.0 mg/mL,

* AK-B and AK-D treatment exerted 2.0 times against the Capan-2 cells, compared with SK-B and SK-D.

* AK-D and AK-B treatment compared with SK-D and SK-B demonstrated a significant increase of tumor-suppressive p21 levels associated with apoptosis and cell cycle arrest.

* AK-B displayed higher ongoing cytotoxic effects compared with AK-D.


Based on the evidence, researchers said,. "Overall, these results indicate that kimchi made with Amtak Baechu cabbage and treated with brine solution (AK-B) had a superior anticancer potential against both types of cancer cells, with the effects being greater in pancreatic cancer cells compared with liver cancer cells".

Taken together, cabbage may be considered a functional food for the prevention and treatment of
pancreatic cancer, pending the confirmation of the larger sample size and multicenter human study.

#Cucumber Enhances the Ratio of Gut Microbiota in the Gut Associated with the Improvement of Insulin Sensitivity, According to Studies

Insulin Resistance, a hallmark in patients with type II diabetes is the condition in which the cells in tissues that start to resist or ignore the signal of the hormone insulin.

In other words, the insulin produced by the body no longer can convert glucose to energy even with the presence of the hormone insulin, leading to high levels of glucose in the bloodstream.

According to the statistics provided by the CDC, approximately, 1 in 3 people in the United States have prediabetes, including half of those aged 60 and older.

People with insulin resistance have been found to increase the risk for prediabetes, type 2 diabetes and other serious health problems, such as cardiovascular disease and cancer.

Although researchers do not know the exact cause of insulin resistance, they do not certain risk factors such as genetic preposition, aging, and ethnicity are associated with the onset of the condition.

However, an unhealthy lifestyle that causes excess body weight, too much belly fat, smoking and lack of exercise are the most prevalent factors in the Western world.

Dr. Martin O. Weickert wrote, "Insulin resistance has been proposed as the strongest single predictor for the development of Type 2 Diabetes (T2DM). Chronic oversupply of energy from food, together with inadequate physical activity, has been recognized as the most relevant factor leading to overweight, abdominal adiposity, insulin resistance, and finally T2DM".

And, "Conversely, energy-reduced diets almost invariably facilitate weight loss and reduce abdominal fat mass and insulin resistance. However, sustained weight loss is generally difficult to achieve, and distinct metabolic characteristics in patients with T2DM further compromise success".

The findings strongly suggested that by maintaining a healthy lifestyle including a healthy diet and moderate exercise, insulin resistance is preventable.

The cucumber plant is a species of Cucumis Sativus, belongs to the family Cucurbitaceae and native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

In finding a potential compound for the treatment of insulin resistance, researchers investigated the effect of the effects of Cucumis melo L. (Cucumis) on obesity-induced insulin resistance (IR) in genetically leptin-deficient Lepob/Lepob mice.

According to the results of the tested assays, Cucumis showed significant activity in the improvement of IR in obese mice and inhibition of inflammation in adipose tissue and blood.

The efficacy of Cucumis against insulin resistance of the tested mice was also attributed to the enhancement of the ratio of gut microbiota in the gut-associated with the improvement of insulin sensitivity.

Based on the findings, researchers wrote in the final report, " administration of Cucumis improved IR by reducing inflammation, thereby changing the gut microbiota composition. Cucumis is thus a promising treatment for obesity-induced insulin resistance and the inflammatory state".

Lund University in the study of glycemic and insulinemic responses to fermentation products suggested that adding pickled cucumber to breakfast with a high-glycemic-index bread significantly lowered postprandial glycemia and insulinemia compared to no metabolic response of regular milk and fresh cucumber.

In other words, fermentation products such as Prickle play an essential role in reducing the risk of insulin resistance.
