#Healthyfood #OnionExtract Shows A Protective Effects Cyto and Geno-Toxicity, Reserachers Say

Kyle J. Norotn

Cytotoxicity is the process that kills cells by inducing toxic substances.
Cancer cytotoxicity is the stage of killing cancer cells by toxicity with or without inducing side effects.

Most cases of cytotoxicity of a substance are to determine the potential toxicity of the quality of the substance, including plant extracts or biologically active compounds isolated from plants.

In other words, the aim of cytotoxic studies is to find the minimal quality that is essential for the successful development of pharmaceutical or cosmetic preparation that induces cellular toxicity for the treatment of cancer.

Before the preparation can be used in humans, bioassays associated with a number of different cell lines in vitro and vivo have been used to assess the cytotoxicity of many herbal plants, including those of African medicinal plants.

Free radicals have also been found to induce cytotoxicity to cause cancer death by oxidative stress.

Dr. Clifford Fong wrote, "An analysis of the cytotoxicity data for a wide range of anti-cancer drugs applied to triple-negative breast cancer (TNBC) cells and ovarian cancer cells under normoxic, hypoxic and anoxic conditions have shown that free radical forms of the drugs are involved".

And " This finding is consistent with the known involvement of oxidative stress in the TNBC and ovarian cancer cells. A mechanism to explain the formation of these drug-free radicals and how they can be involved in oxidative stress is proposed".

Genotoxicity is the term associated with chemical agents that damages the genetic information within a cell, causing mutations of cell DNA, a major cause of cancer.

Not all genotoxicity is mutagenicity, However, all mutagens are genotoxic.

The onion is a plant in the genus Allium, belonging to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild, or sweet.

In finding a potential compound for the treatment of cancer, researchers examined the effect of an onion extract on cyto- and genotoxicity in human lymphocytes treated with bleomycin (BLM), a radiomimetic agent.

In radiation exposure -induced oxidative DNA damage in the increased risk of cancer, the risk the richest sources of dietary flavonoids found in onion inhibited the frequency of micronuclei (MN) and DNA damage following treatment with BLM, according to the cytokinesis-blocked micronucleus assay and a single cell gel electrophoresis assay.

Phytochemicals in the onion showed a significant increase in cell viability in lymphocytes exposed to BLM compared to cells treated with BLM alone.

Onion also suppressed the increased frequency of BLM-induced MN and DNA damage in a dose-dependent manner evidenced by lymphocyte viability pretreated with onion extract (10 and 20 μL/mL).

Moreover, the frequency of BLM-induced MN was decreased at all doses of BLM, and DNA damage was decreased by the administration at 3 μg/mL of BLM.

Based on the findings, researchers said, "onion extract may have protective effects against BLM-induced cyto- and genotoxicity in human lymphocytes".

Taken altogether, onion-processed abundantly bioactive compounds may be considered a remedy for the prevention and treatment of cytotoxicity and genotoxicity, pending the confirmation of the larger sample size and multicenter human study.

#PintoBean's Lectin Inhibits Proteins Associated with Cancer Cell Proliferative Activity, According to Studies

Kyle J. Norton

Proliferation is a process of cancer growth associated with an abnormally high rate of cancer cell division, leading to rapid overgrowth of the tumor.

Cancer is a class of diseases associated with the growth of cells disorderly and uncontrollably due to the alternation of DNA.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Cancer cell proliferation by dividing unlimitedly, compared to normal cells that die after a certain cell cycle division.

More precisely,
* The growth of normal cells

Under normal circumstances, cell growth is a natural process without even notice, every day millions of cells grow to replace millions of dead cells and to keep our body in balance and healthy.

Sometimes, acceleration of cell growth is necessary to prevent further damage to the body organ, such as cuts, injuries, etc., but after fulfilling their purposes, the growth of cells is back to a normal rate again due to our body growth inhibitors.

Unfortunately, skin cells sometimes overgrow while healing, leading to scars to the skin.

*The primary tumor; size and growth

A primary tumor or original tumor that the tumor progression that yields a cancerous mass.

Some cancers can begin in several places, such as lymphomas and leukemias. Most cancers can not be detected until they formed a small grape-like lump.

Cancer cells need 42 doublings before they can produce enough cancer cells to cause death.

According to the statistics provided by the National Cancer Institute, In 2018, approximately 1,735,350 cases of cancer were diagnosed in the US and the disease also caused the death of 609,640 Americans.

In finding a natural ingredient for the prevention and treatment of cancers, scientists examined the antiproliferative effects of Lectins (hemagglutinins) from the common edible bean Phaseolus vulgaris cv. Hokkaido large pinto bean.

The pinto bean is also known as the common bean, genus Phaseolus and the family Fabaceae. It has a mottled appearance of darker reddish-brown spots on a lighter brownish-beige background, thus appearing painted or spotted.

According to the tested analysis of a 60 kDa dimeric hemagglutinin with a blocked N-terminus was isolated in large yield (190 mg/60 g) from the pinto beans,

* The administration of 60 kDa showed an inhibitory proliferation of nasopharyngeal carcinoma (CNE2), human breast cancer (MCF7), and hepatoma (HepG2) cells.

* The IC50 values were considered relatively low among lectins of different P. vulgaris cultivars toward HepG2, MCF7, and CNE2 cells after treatment for 48 h were 8.1, 6.07, and 7.49 μM, respectively,

In other words, the lectin found in the pinto bean as sugar-binding proteins or glycoproteins with various biological activities exerted a promising remedy for the treatment of cancers in vitro.

Taken altogether, Pinto beans may be considered a promising anti-proliferative functional food, pending the confirmation of the larger sample size and multicenter human study.

#Healthyfood #GreenLeafyVeggies Inhibit Levels of Cholesterol through Their Bile Acid Binding Activity, Researchers Suggest

Kyle J. Norton

Bile acids are synthesized by oxidation from cholesterol in the liver, flowing to the gallbladder to aid digestion.
Cholesterol is a waxy substance that is produced either by the liver or injected from animal food sources.

Cholesterol found in our body is needed for our body to build cell walls, hormones, and vitamin D, and create bile salts that aid digestion. However, too much of them, in most cases due to the intake of animal protein can cause plaques to build up on the arterial wall, the major cause of heart disease and stroke.

Bile acid-binding is a process that interferes with normal fat digestion and absorption including the prevention of absorption of fat-soluble vitamins such as A, D, E, and K.

In other words, the binding of bile acids (BAs) may have a strong effect to lower blood cholesterol levels by increasing the excretion of bile acids in the stool and reducing the amount of bile acid returning to the liver.

By inhibiting the levels of bile acid caused by a certain type of liver/bile duct disease such as partial biliary obstruction, levels of cholesterol can be controlled.

Conventionally, the effects of bile acid sequestrant are limited. Low doses of 8 grams/day of Cholestyramine, a bile acid sequestrant, which binds bile in the gastrointestinal tract to prevent its reabsorption can lower LDL cholesterol by 10%-15% compared to high doses of 24 grams/day of only 25%.

In other words, bile acid-binding medicine such as Cholestyramine, used alone is less effective, compared to statins in lowering LDL cholesterol.

The most common symptoms associated with the intake of Cholestyramine are constipation, diarrhea, stomach/abdominal pain, bloating, nausea, vomiting, and loss of appetite.

In finding a potential plant that processes anti-high blood cholesterol activity, researchers compared the bile acid-binding potential that lowers the risk of heart disease and cancer of various vegetables.

Compared to Cholestyramine (a cholesterol-lowering, bile acid-binding drug) that was used as the positive control treatment and cellulose as the negative control, researchers found that* Relative to cholestyramine, in vitro bile acid binding on dry matter basis was for the collard greens, kale, and mustard greens, 13%; broccoli, 10%; Brussels sprouts and spinach, 8%; green bell pepper, 7%; and cabbage, 5%.

In other words, collard greens = kale = mustard greens showed the strongest healthy-promoting potential in bile acid-binding compared to the following lesser extent of > broccoli > Brussels sprouts = spinach = green bell pepper > cabbage.

More importantly, steam cooking significantly improved the in vitro bile acid binding of collard greens, kale, mustard greens, broccoli, green bell pepper, and cabbage compared to vegetables raw (uncooked).

More precisely, the Inclusion of steam-cooked collard greens, kale, mustard greens, broccoli, green bell pepper, and cabbage in our daily diet may have the potential in improving healthy levels of blood cholesterol.

Collectively, researchers said, "These green/leafy vegetables, when consumed regularly after steam cooking, would lower the risk of cardiovascular disease and cancer, advance human nutrition research, and improve public health".

Taken altogether, green leafy veggies may be considered functional foods for lowering blood cholesterol, pending the confirmation of the larger sample size and multicenter human study.

#Healthyfood #Tomatoes Inhibit Parameters Associated with #Hypercholesterolemia, Researchers Suggest

Kyle J. Norton

Hypercholesterolemia is a condition of high blood cholesterol. A condition is a form of hyperlipidemia or high blood lipids.

The cause of hypercholesterolemia is a result of overexpression of low-density lipoprotein with reduced levels of high-density lipoprotein in the blood.

Believe it or not, cholesterol produced by the liver or from outside sources is essential for the building of strong cell membranes, the production of steroid hormone and vitamin D.

However, overexpression of blood cholesterol-induced hypercholesterolemia has been found to exhibit the risk of cardiovascular disease and diabetes.

Compared to hypercholesterolemia, healthy levels of LDL/HDL are any numbers that are less than 4.

There is no single cause of hypercholesterolemia. Epidemiologically, diet, obesity, genetic preposition and medical conditions such as diseases such as type 2 diabetes and an underactive thyroid are some the prevalent factors associated with the risk of the condition.

According to the statistics, over 34 million American adults are living with elevated blood cholesterol levels (higher than 240 mg/dL). ...

The most common inherited form of high blood cholesterol is called familial hypercholesterolemia found in 1/500 people.

By adopting a healthy lifestyle and moderate exercise, you may do just fine without medication.

Tomato a is red, edible fruit, genus Solanum, belongs to the family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find a natural ingredient or whole food for the treatment of hypercholesterolemia, researchers compared the effect of dried tomato, grape and apple pomace (5% in diet) on the cholesterol levels in serum in an animal model

The study included selected male Wistar rats fed a cholesterol diet (0.3%) shortly after weaning followed by administration of individual fruits contained 47.2, 67.0 and 46.2 g of total dietary fiber per 100 g of dry matter for tomato, grape and apple pomace, respectively.

Compared to apple pomace reduced the liver cholesterol by 11% tomato showed a better-reduced cholesterol level in the liver by 15%

Tomatoes and other tested pomace species reduced cholesterol content in the heart by 18-21%.

Only tomatoes exerted a significant effect on the levels of triacylglycerol content in the heart compared to other tested pomaces.

All products strongly reduced (up to half of the original value) the activity of enzymes HMG-CoA reductase associated with the production of cholesterol in the liver and increased the fractional catabolic rate of plasma cholesterol by 60-120%.

In rats fed with hypercholesterolemic feed and treated with lycopene orally by gavage at the dose of 5, 25, 45, 65, 85, and 105, 125 mg/kg/bw-1/d-1, researchers found that injection of all doses of lycopene exerted a statistical significance in reducing the levels of cholesterol observed by inverse U shape at the 4 weeks of the experiment.

Furthermore, serum lycopene concentration was negatively correlated with the levels of serum TC, and LDL-C, particularly in the cerebral LDL-C.

In other words, serum lycopene found in the tested model showed a strong effect against brain cholesterol in facilitating neuron damage.

Dr. Wang W, the lead scientist said, "These results suggested an inverse U-shape relation between dose and serum concentration of lycopene, and intermediate doses were most effective to protect cerebral vessels and neurons from being damaged by hyperlipidemia".

Taken together, tomatoes processed with high amounts of lycopene may be considered supplements for the prevention and treatment of hypercholesterolemia, pending the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Cranberry#Extract #Oximacro(®) and Purified A-Type Proanthocyanidins (PACs-A) Inhibit #Herpes Simplex Virus 1 and 2 (HSV-1, 2) Infection

Kyle J. norton

Herpes simplex virus 1 (HSV-1) primarily causes oral herpes, including cold sores and fever blisters around the mouth and on the face.

Herpes simplex virus-2 (HSV-2) primarily causes genital herpes and genital herpes outbreaks

Infection can be acute or chronic. In the acute phase of infection, after receiving the messages from the central nervous system related to the danger of microbial infection, the immune macrophages in the first line of defense stimulates the production of blood palette to the cover the wound and pro-inflammatory cytokines to the site of the infection to kill off pathogens which try to enter the body.

However, overproduction of pro-inflammatory cytokines in the acute phase of infection has also killed off or damage to the nearby healthy cells, leading to the formation of scars.

In most cases in a healthy individual, the immune system can easily eradicate all foreign invaders and the wound is recovered within days and weeks.

Symptoms of the acute infection include gastrointestinal discomforts, such as congestion vomiting or nausea, diarrhea, short-term joint or muscle pain, fever, headache, skin rash, and cough, Some patients with acute infection also experience symptoms of swollen lymph nodes, an indication of the body is fighting infection.

In chronic infection, the symptoms are most vague or no specific symptoms, including bronchitis and asthma, persistent low-grade chronic joint and muscle pain, or swelling, and the alternations between diarrhea and constipation, bloating, or heartburn, In other words, the symptoms are totally depended on the location of the low-grade inflammation.

Most cases of infection except those of viral infection are treated by topical antibiotic cream and/or oral antibiotics.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of skin infection, researchers investigated the effects of cranberry (Vaccinium macrocarpon Ait.) extracts Oximacro(®) against Herpes simplex type 1 (HSV-1) and type 2 (HSV-2) infections.

In vitro, the novel cranberry inhibited HSV-1 and HSV-2 replication.

Furthermore, Oximacro(®) prevented the adsorption of HSV-1 and HSV-2 to target cells.

Moreover, Oximacro(®) and its PACs-A targeted the viral envelope glycoproteins gD and gB to enter cells, thus resulting in the loss of infectivity of HSV particles.

Most importantly, Oximacro(®) completely exerted continuously its anti-HSV activity even at acidic pHs (3.0 and 4.0) and in the presence of 10% human serum proteins, and conditions that mimic the physiological properties of the vagina.

Based on the findings, researchers said, "Oximacro(®) as an attractive candidate for the development of novel microbicides of natural origin for the prevention of HSV infections".

Taken together, cranberry may be considered an immune-modulatory remedy, pending to the confirmation of the larger sample size and multicenter human study.

#CrushedGarlic's Allicin Inhibits and Pathways Associated with the Risk of #BowelCancer, Researcher Find

Kyle J. Norton

Studies suggested that garlic may have the potential in killing bowel cancer cells before they can spread to form colonization.
Bowel cancer also known as colorectal cancer, is a chronic medical condition caused by the abnormal proliferation of cells in the colon, rectum, or vermiform appendix.

Bowel cancer is a relatively very common and slowly growing cancer. The progression of cancer is predictable.

The exact causes of bowel cancer are unknown. researchers as of today do not know why people in the same health condition and age in the same family and similar dietary patterns, are susceptible to the onset of bowel cancer, while others do not.

However, researchers do know that certain risk factors are associated with the development of the disease, including age, family history

Some researchers suggested that the mutated gene inherited from the parents, and certain medical conditions such as inflammatory bowel disease, polyps, and obesity are also some of the bowel cancer prevalent risk factors.

A recent study also showed people who smoke and consume high volumes of red meat are also at elevated risk of the onset of the disease.

Truly, following a healthy diet and active lifestyle can decrease the risk of bowel cancer by 10 folds. Dr. IAIN ROBERTSON, the lead author at Monash University, in the study "Colorectal cancer, diet, and lifestyle factors: opportunities for prevention" wrote, " The potential for colorectal cancer risk reduction by decreasing consumption of animal fats, red meat, alcohol, and cigarettes, by increasing physical activity levels, and by increasing intake of fruits, vegetables, and dietary fiber".

If you are experiencing symptoms of a change of bowel habits that last longer than one month, persistent rectal bleeding, abdominal pain that does not stop,.and blood in the stool, please check with your doctor to rule out the possibility of bowel cancer.

Garlic (Allium sativum) is a species in the onion genus, belonging to the family Amaryllidaceae, native to central Asia.

The pungent spice has been used popularly in traditional and Chinese medicine in treating the common cold and flu to the Plague, blood pressure cholesterol levels, natural antibiotics, etc.

Allicin (diallyl thiosulfinate), is the best-known biologically active component in freshly crushed garlic, found to reduce the risk of bowel cancer through inhibiting bowel cancer cell lines HCT-116, LS174T, HT-29, and Caco-2, via the activity of Nrf2, according to The Hebrew University of Jerusalem.

Nrf2 is a protein that regulates the expression of antioxidant proteins to protect the cells against damage and injury caused by oxidative stress triggered by inflammation.

However, in the colon cancer cell lines, overexpression of Nsf2 can initiate cancer cell apoptosis.

The Chungbuk National University study that uses colon cancer cells (SW620 and HCT116) to investigate whether sulfur compounds thiacremonone isolated from garlic could modulate NF-kappaB activity and thereby induce apoptotic cell death of colon cancer cells, suggested, the application of different concentrations ranging from 30 - 150 microg/ml of thiacremonone for the periods from 0 to 48 hour inhibited colon cancer cell (SW620 and HCT116) growth followed by induction of apoptosis in a dose-dependent manner.

Further analysis showed that the efficacy of the novel sulfur compound against colon cancer cell growth and induction of apoptotic cell death was attributed to the compound's function in modulating NF-kappaB, a protein that controls transcription of DNA, cytokine production, and cell survival.

In vitro, intake of garlic and its lipid-based extracts, induced cell cycle arrest, through a fourfold increase in caspase activity (apoptosis) in PC-3 cells when treated at a dose of 0.5 or 1 μg/m and may be considered a potential as promising anticancer agents, including colon cancer.

Dr. Baena R and Dr, Salinas P said, " Dietary factors and lifestyle may contribute to the increasing colorectal cancer (CRC) incidence... Fish consumption may decrease CRC risk by 12% whereas garlic intake is not significantly associated with reduced CRC risk".

In fact, diet represents 30-35% of risk factors that contribute to the onset of cancer, adding garlic in your diet together with the reduction of meat intake can reduce the risk of various cancers, including colorectal cancer

However, in order to confirm garlic isolated compounds' anti bowel cancer efficacy, further data collection on large example size and multi-center studies performed with human consumption of whole food during the course of the disease will be necessary.

Extracts from #GymnemaLeave and Cinnamomum Bark Inhibit #CervicalCancer by Inducing Cytotoxicity, and Apoptosis, According to Studies

Kyle J. Norton

Cinnamon may be the Best Wholefood Medicine for Prevention, management, and Treatment of Cervical Cancer. According to the Manipal University, Eugenol, a potential chemopreventive agent, one of the components of cinnamon possesses antiseptic, analgesic, antibacterial, and anticancer properties in the expression of apoptosis and inflammation against cervical cancer cell lines.

Cinnamon is a spice derived from the inner bark of tree, native to South East Asia, of over 300 species of the genus Cinnamomum, belonging to the familyLauraceae, use in herbal and traditional medicine as anti-fungal and bacterial levels, to improve reproductive organ, prevent flatulence and intestinal cramping, and to treat indigestion, diarrhea, bad breath, headache, migraine, etc.

In the SiHa cancer cell line, established from cancerous tissues of the cervix uteri, methanol extracts of Gymnema sylvestre (GS) leave and Cinnamomum zeylanicum (CZ) bark exhibited dose-dependent cytotoxicity, inducing apoptosis.

Other researchers suggested that the effective use of cinnamon against tumor activity, particularly in cervical cancer cell line SiHa may result in a reduced number of colonies, risk of migration potential through down-regulated normal physiological processes.

Dr. Kwon HK, the lead researcher at the Gwangju Institute of Science and Technology, insisted, Cinnamomum cassia bark,.. containing several active components such as essential oils, expressed anti-tumor effect via enhanced pro-apoptotic and inhibition of pro-inflammatory activities, in vitro, and in vivo mouse melanoma model.

In fact, the potential anti-tumor effects of cinnamon may be due to increased anti-tumor activities in the suppression of progression by increasing the levels of cytolytic molecules and cytotoxic activity.

Although the whole herb medicine may be a potential agent for reduced risk and treatment of cervical cancer, oral use of cinnamon should be taken with care and only prescribed by an herbalist.
