#Epigallocatechin Gallate (EGCG) Found in #Elecampane #Extract Inhibits Liver Stress Markers Through Antioxidant Activites, Researchers Say

Kyle J. Norton

The liver is the largest internal organ in the human body and the only organ which can regenerate itself. The liver is the first line of the bodily toxin defense by filtering the blood that comes from the digestive tract, before passing it to the rest of the body.

The liver also produces cholesterol that plays a critical role in building strong cell membranes, producing steroid hormones and vitamin D and aiding the digestive function in food absorption and detoxifying chemicals and metabolizing drugs.

Furthermore, the liver also processes a function of detoxifying chemicals and metabolizes drugs.

The hepatoprotective or liver protective effect is an action that maintains the liver's healthy functioning against any harmful toxins.

Symptoms of liver diseases include jaundice, abdominal pain, and swelling, swelling in the legs and ankles, itchy skin, dark urine, and pale stool color, and a tendency to bruise easily.

There are no exact causes of liver disease. Viruses, toxins, genetics, alcohol, and unknown causes are some prevalent factors found in patients with chronic liver disease, including cancer.

Liver damage and disease can also be caused by long-term intake of certain medications, such as paracetamol, and Illegal drugs such as cocaine.

In the US, most cases of liver disease are a result of hepatitis viral infection and autoimmune diseases. Infectious viral hepatitis can produce toxins to protect them against the antioxidant enzymes produced by the host body and prevent the attack of the systematic immune response.

Furthermore, in recent years, excessive alcohol drinking has been found to induce alcoholic liver disease in many parts of Asia and America.

Some researchers suggested overweight and obese and congenital diseases such as Hemochromatosis and Wilson’s disease also are prevalent risk factors for liver damage and diseases.

In finding a natural ingredient for the prevention and treatment of liver diseases, scientists examined the hepatoprotective effect of Inula Britannica using fermentation on HepG2 cells.

Elecampane, also known as Horse-heal (Inula helenium) is a perennial composite plant, genus nulla, belonging to Asteraceae, native to Southern Europe, and Asia.

The plant has been used in traditional and herbal medicine in treating bronchitis or emphysema, night sweats, pulmonary tuberculosis, irritating cough, catarrhal discharges, etc.
According to the tested analysis,
* The Epigallocatechin gallate (EGCG) found I. Britannica extract was enhanced from 2.06 to 10.28 μg/mg during fermentation (p < 0.001).

* The fermentation also increased the DPPH radical-scavenging ABTS radical-scavenging, and superoxide anion-scavenging abilities to 92.65%, 694.25 μM Trolox/mL, and 86.38%, respectively.

* At 500 μg/mL (p < 0.05). Cupric-ion-reducing capacity with formation of the Cu+-neocuproine complex increased by 5.88%, 6.38%, 3.24%, and 8.55% at 62.5 to 500 μg/mL. Ferric-ion-reducing capacity of the fermented extract increased by 20%, 7.16%, 3.85%, and 5.45% at each concentration (p < 0.05).

* Furthermore, compared to the unfermented extracts yielded cell viability of 91.42%, 90.59%, 88.38%, and 79.17%, the fermented extract yielded 100.28%, 99.66%, 96.15%, and 89.90%, respectively.

* Moreover, the fermented extract decreased the liver stress marker of alanine transaminase activity from 117.2 to 61.7 U/mL in the ethanol-damaged HepG2 cell line (p < 0.01).

Based on the results, researchers suggest, " Overall, both antioxidant and hepatoprotective effect increased by fermentation in I. Britannica extract.

Taken together, Elecampane may be considered a liver protective remedy, pending the confirmation of the larger sample size and multicenter human study.

#Leeks Process Anti Liver Triglycerides (TG) Accumulation Induced by High-Fat Diet, Researchers Reveal

Kyle J. Norton

Cholesterol is a waxy substance that is produced either by the liver or injected from animal food sources.

Cholesterol found in our body is needed for our body to build cell walls, hormones, and vitamin D, and create bile salts that aid digestion. However, too much of them, in most cases due to the intake of animal protein can cause plaques to build up on the arterial wall, the major cause of heart disease and stroke.

More precisely, if levels of low-density lipoprotein are high compared to high-density lipoprotein which returns the accessed cholesterol back to the liver, you have high blood cholesterol.

In other words, healthy blood cholesterol is any level with either less than or equal to 4 (low-density lipoprotein/ 1 (high-density lipoprotein).

Low-density lipoprotein is needed for our body to build cell walls, make hormones and vitamin D, and create bile salts that help you digest fat. However, overexpression of LDL can induce plaque accumulated on the arterial wall, leading to atherosclerosis, a major cause of cardiovascular disease.

Low-density lipoprotein is produced by the liver or from dietary sources. High-density lipoprotein neutralizes the levels of LDL by returning them to the liver.

Some researchers suggested that diets high in trans fats, saturated fat, sugar, smoking, and medical conditions such as diabetes or high blood pressure are associated with an increased risk of high blood cholesterol.

According to the statistics, in 2015-2016, by age and gender, a total of 12.4 percent of adults aged 20 years and over had high cholesterol in the United States.

Although there are many risk factors such as obesity, large waist circumference, physically inactive, smoking, and diabetes associated with the onset of high blood cholesterol, researchers suggested that a poor diet rich in saturated and trans fat, and processed foods is the major cause of high blood cholesterol in the Western world.

Epidemiological studies indicated a healthy diet that contains a high amount of fruits and vegetables and whole-grain have a positive on blood cholesterol. In other words, hyperlipidemia can be prevented by a healthy diet.

Leek is a genius of Allium, belonging to the vegetable family vegetable Alliacaea, the same as onion and garlic. Leeks are easy to grow from seed and usually reach maturity in the autumn months, and have few pest or disease problems.

In finding a potential plant that processes anti-high blood cholesterol, researchers examined the effects of oral administration of Persian leek on plasma and liver lipids.

The study included male Syrian hamsters that were randomly divided into three groups: control (standard diet), high-fat control (high-fat/high-cholesterol diet), Persian leek (high-fat/high-cholesterol diet + 1% per weight of diet from dried powdered Persian leek) for 14 weeks.

Before treatment, a high-fat diet showed an increase in plasma and liver lipids compared to a standard diet.

Administration of Persian leek to the high-fat/high-cholesterol diet resulted in no significant changes in the concentration of the plasma lipids or liver cholesterol.

Liver triglycerides (TG), plasma Alanine aminotransferase, and gene expression of tumor necrosis factor- α associated with liver stress marker and pro-inflammatory cytokines, compared to high-fat diet only.

Collectively, researchers said, "Persian leek might be considered as a herbal food that can reduce liver TG accumulation induced by high-fat diets".

Taken together, leeks may be considered a functional food for the prevention and treatment of high blood cholesterol, pending the confirmation of the larger sample size and multicenter human study.

#Omega 3 Fatty Acids Inhibits the Onset of #ColorectalCancer by Regulating the DNA Methylation Process, Reserachers Find

Kyle J. Norton

Cancer is a class of diseases associated with irregular cell growth in the tissue of organs.
The colon-rectum formed as a part of the digestive system is involved in the absorption of water from the small bowel contents and broken down of certain materials in the feces into substances of which some of them to be reabsorbed and reused by the body.

Colorectal cancer is a medical condition caused by cell growth irregularly in the tissue of the colon or rectum due to the alternation of cells' DNA, including

*Rectal cancer is the slowest-growing cancer. Therefore, most cases of rectal cancer are diagnosed at the advanced stage in the older population.
* Colon cancer is relatively very common and slowly growing and progressing in a predictable way.

Most cases of primary colon cancer start in the cell on the surface of the inner lining of the colon. At the early stage. most patients with colon cancer do not experience any symptoms.

The common type of colorectal cancer is adenocarcinomas, originating in epithelium tissues and squamous cells that make up the bowel lining together with the gland cells,

According to the statistics provided by the American Cancer Society, in the US, colorectal cancer is the third most common cancer diagnosed in both men and women compared to other types of cancer.

The lifetime risk of colorectal cancer is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

Approximately, 140,000 cases of colorectal cancer were diagnosed in the US in 2018. Colorectal cancer also causes the death of over 50, 000 people, representing 8.3% of total cancer death.

Being overweight or obese. smoking, physical inactivity, excessive alcohol drinking, increased in age, family and personal history, and genetic preposition are some of the prevalent risks of the disease.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

In finding a potential phytochemical for the treatment of clonic diseases, researchers examined omega-3 PUFAs influences on DNA methylation levels in the colorectal cancer (CRC) rat model.

According to the tested analysis of N-methyl phosphite nitrourea and omega-3, PUFAs fed to the rats during tumor induction, tumor incidence in omega-3 treated rats was much lower than in CRC model rats.
In other words, omega-3 PUFAs process anti-colorectal cancer properties.

More precisely, 6 PUFA members categorized into omega-3 and omega-6 families showed similar activity in lowering the ratio of omega-6/omega-3 PUFAs in the omega-3 PUFAs treatment group than in the CRC model group.

Interestingly, the normal cytosine nucleotide DNA (5 mC) content, in omega-3 PUFAs treated rats was higher than in CRC model rats. These results suggested that omega-3 PUFAs promoted 5 mC synthesis.

Where 5mC oxidation reaction is mutated in human tumors.

Based on the findings, researchers said, "omega-3 PUFAs probably inhibited tumor growth via regulating DNA methylation process, which provided a novel anticancer mechanism of omega-3 PUFAs from epigenetic view".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the prevention and treatment of colorectal cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#UrsolicAcid UA-Liposomes, The Potential Tumor-Mediated Immune-Suppressive Agent, Researchers Suggest

Kyle j. Norton

Cancer is a class of diseases associated with the growth of cells disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite, and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.
The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Compared to all cancers, lung, breast, prostate, and colorectal cancers are the most common cancer in the US.

The causes of cancer are unknown. age, excessive alcohol drinking, exposure to cancer causing-substances, diseases associated with chronic inflammation, unhealthy diet, hormone therapy, and use of immunosuppression are some of the prevalent factors involved in the onset of cancer.
Cancer immunotherapy is a type of treatment used to treat cancer patients that involve or uses components of the immune system.

Depending on the types of cancer immunotherapies, some may consist of antibodies that bind or inhibit the expression of proteins in the cancer cells.

Ursolic acid is a phytochemical in the subclass of Triterpenoid, belongings to the group of Terpenes found abundantly in apples, basil, bilberries, cranberries, peppermint, lavender, oregano, hawthorn, prunes., etc.

In finding a potential phytochemical for the treatment of chronic diseases, researchers examined the effect of ursolic acid (UA), a potent triterpenoid compound found in plants and fruits with activities modulating key cell-signaling pathways in cancer.

According to the analysis of the development of UA-liposomes made by HPβCD assisted active loading,
* Compared to lipid suspensions of UA (Lipid-UA) with similar lipid composition, the novel process enabled the formation of UA-Ca crystalline structures inside the liposomes and therefore sustained the release of UA in vivo.

* UA-liposomes were not generally toxic towards 4T1 triple-negative breast cancer cells, they could effectively modulate CD4+CD25+Foxp3+ in regulating T cells from 4T1 tumor-bearing mice by inhibiting the inflammatory cytokines.

* In vivo, the administration of UA-liposomes at 10 mg/kg dose led to reduced numbers of myeloid-derived suppressor cells (MDSCs) and regulatory T cells (Tregs) residing in tumor tissues

Collectively, researchers said, "These changes signified the correction of the tumor-mediated immune-suppressive microenvironment. The UA-liposomes treatment alone was already effective in deterring tumor growth. Such a formulation may be highly promising as an immunotherapy agent and be combined with chemotherapeutics or targeted drugs",

Taken together, Ursolic acid may be considered adjunct immunotherapy. therapies in the treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of Ursolic acid in the form of supplements should be taken with extreme care to prevent overdose and acute liver toxicity.

#SodiumFerulate Found in #Herbal #Angelica Inhibits Pathways Associated with the Onset of #Osteoarthritis, According to Studies

Kyle J. Norton

Osteoarthritis (OA) is a form of arthritis, found in the general term of musculoskeletal disorders (MSDs) characterized by wear and tear on joints.

The chronic and medical condition affects over 25 million people in the United States alone.

The characteristics of osteoarthritis are aching pain, stiffness, or difficulty in moving the joint or joints. Therefore one must understand the differences in prevalence and incidence estimates of osteoarthritis (OA), compared to other types of arthritis.

Furthermore, the pain usually gets worse in a change of weather, at night, and in advanced diseases, and the pain can occur even at rest.

In contrast to the general belief, osteoarthritis only damages the joints of hands, knees, but also cause inflammation of the joints in your hips and spine.

The condition is a slow development disease, often starting in the 40s and 50s, and affects almost all people to some degree by age 80.

The common risk factors of osteoarthritis are aging, gender, improper healing of injured joints at a young age, obesity, and bone deformity. However, people who have a job with tasks that place repetitive stress on a particular joint are associated with the onset of osteoarthritis in most industrial countries.

According to the statistic from the NIH, osteoarthritis (OA) is the most common joint disorder in the US. Among adults, 60 years of age or older the prevalence of symptomatic knee and sadly OA affects approximately 10% of men and 13% of women.

Epidemiological strongly suggested that obese and overweight have a strong impact on the weight-bearing joints, the genetic preposition of a patient can make such patients more susceptible to the disease compared to others who do not. People with a joint injury that did heal properly at a younger age and people with occupations that require standing for long periods of time, repetitive bending, heavy lifting, or other movements, and the increase in age are associated with the increased risk of the onset of the disease.

In finding a natural ingredient for the prevention and treatment of aging-related diseases, scientists examined the angelica phytochemical, sodium ferulate, and a polysaccharide fraction on osteoarthritis animal models or in human chondrocytes stimulated by the proinflammatory cytokine, Interleukine-1β.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen the heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the results of the experiment,* Sodium ferulate found in angelica exhibited marked anti-inflammatory and anti-apoptotic properties by inhibiting the TNF/TNFR signal transduction pathway associated with inflammation and autoimmunity.

* On another hand, the polysaccharide fraction which contains a mixture of various carbohydrates was found to promote proteoglycan biosynthesis in the cartilage matrix by stimulating the activity of the UDP-glycosyltransferases that synthesize the chondroitin sulfate chains of aggrecans.

In other words, both chemical compounds demonstrated strong protective effects against osteoarthritis through different mechanisms.

based on the findings, researchers said, "the combined action of sodium ferulate and polysaccharides fraction would prevent cartilage destruction in osteoarthritis and favor cartilage repair".

Taken altogether, herbal angelica may be considered an antioxidant remedy for the treatment of osteoarthritis, pending the confirmation of the larger sample size and multicenter human study.

#Bromelain Shows a Significant Effect on Decreasing the Incidence and Severity of Spontaneous and Chronic Colitis, According to Researchers

Kyle J. Norton

Inflammatory bowel disease (IBD) is a digestive condition characterized by an inflammation of the large intestine. The disease affects over 1.6 million Americans today.
The most common types of IBD are
*Crohn’s disease (CD) is an inflammation of any part of the digestive tract. Most cases of CD affect the tail end of the small intestine.

* Ulcerative colitis is an inflammation of the large intestine.

The exact causes of inflammatory bowel disease are unknown. However, some researchers do know that
* The disease may be caused by the weakened immune system that fails to kill off the invasion of infectious micro-organisms in the acute phase of digestive infection caused by tissue injury and damage.

* IBD may be a result of the immune dysfunction mistakenly attacking the digestive tract, a case of autoimmune disease.

* Imbalance of ratio of good and bad bacteria in the colon, caused by long-term intake of certain medicines.

* IBD is also associated with age, family history, ethnicity, parasite, and smoking.

* Genetic mutation of NOD2, ATG16L1and IL23R involved in innate immunity, autophagy, and inflammatory response may also elevate the risk of IBD.

Believe it or not, if you are living in the Western world, you are also at a higher risk of the disease. For example, people who live in Western countries such as the United States and Europe have a higher IBD risk than those in other parts of the world.

Some researchers suggested an individual diet pattern may influence the risk of IBD.

Dr. Donjete Statovci, the lead scientist wrote, "Westernized diet, defined as high dietary intake of saturated fats and sucrose and low intake of fiber, represent a growing health risk contributing to the increased occurrence of metabolic diseases, e.g., diabetes and obesity in countries adopting a westernized lifestyle. Inflammatory bowel diseases (IBD) and asthma are chronic mucosal inflammatory conditions of unknown etiology with increasing prevalence worldwide".

And, "Recent reports indicate that the gut microbiota and modifications thereof, due to the consumption of a diet high in saturated fats and low in fibers, can trigger factors regulating the development and/or progression of both conditions".

In other words, by pursuing a healthy diet with a higher in fruits and vegetables and reducing intake of red meat, saturated and trans fat, and processed foods, IBD is preventable.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

In an aim to find the potential compound for the treatment of inflammatory bowel disease, researchers examined the efficacy of bromelain in colitis in an animal study.

According to the tested assays, daily treatment with oral bromelain at the age of 5 weeks showed a significant effect on decreasing the incidence and severity of spontaneous colitis in C57BL/6 IL-10-/- mice.

Furthermore, bromelain also significantly decreased the severity of colonic inflammation by inhibiting the production of the pro-inflammatory cytokine in mice colitis induced by piroxicam exposure.

Moreover, bromelain anti-inflammatory effects were associated with the exertion of proteolytical activity.

In wild-type mice treated orally with up to 1000 mg bromelain/kg/day for 18 weeks, researchers do not observe any adverse effects such as dermatitis, hair loss, and weight loss due to mucositis.

Also in the animal model, short-term oral treatment with bromelain was found to decrease the severity of colonic inflammation in C57BL/6 Il10(-/-) mice with chronic colitis.

Mice treated with fresh pineapple juice at a level equivalent to 14 mg bromelain purified from the stem decreased histologic colon inflammation scores and a lower incidence of inflammation-associated colonic neoplasia (35% versus 66%; P < 0.02), with fewer neoplastic lesions/colon.

According to the flow cytometric analysis, murine splenocytes exposed to fresh pineapple juice in vitro affect leukocyte trafficking and activation in facilitating the production of pro-inflammatory cytokines associated with immune response.

Dr. Hale LP, the lead scientist after taking into account co and confounders wrote, "Long-term dietary supplementation with fresh or unpasteurized frozen pineapple juice with proteolytically active bromelain enzymes is safe and decreases inflammation severity and the incidence and multiplicity of inflammation-associated colonic neoplasia in this commonly used murine model of inflammatory bowel disease".

Taken together, bromelain may be considered a remedy for the prevention and treatment of colitis, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Sulforaphane (SFN), an Isothiocyanate Found in #CruciferousVegetables Exerts Cytotoxicity Against Non-Small Cell Lung Cells

Kyle J. Norton

Cancer is a class of diseases associated with the growth of cells disorderly and uncontrollably due to the alternation of DNA.
The lung is a vital organ of humans located near the backbone on either side of the heart with functions of inhaling oxygen from the air then transporting them to the bloodstream to nourish the body cells' needs and exhaling carbon dioxide from the bloodstream.

Lung cancer is a condition of the abnormal growth of the cells in the lung's tissue. The most common form of primary lung cancer is derived from epithelial cells. In the US.

The most common types of lung cancers include
* Squamous-cell carcinoma is often found in an outer area of the lung,

* Large-cell carcinoma found in the center of the lung by an air tube

* Adenocarcinoma is found in any part of the lung.

Non-small cell lung cancer, the most common type of lung cancer, usually grows and spreads methodically and predictably.

The most common risk factors of non-small cell lung cancer (NSCLC) are long-term smoking and people exposed to long-term second-hand smoke.

According to the statistics, in 2019, approximately 228,150 adults including 116,440 men and 111,710 women in the United States will be diagnosed with non-small cell lung cancer (NSCLC). Lung cancer makes up about 13% of all new cancer diagnoses. Black men are about 20% more likely to get lung cancer than white men.

Cruciferous vegetables are a group of vegetables, belongings to the family Brassicaceae (also called Cruciferae) with many genera, and species, and cultivated all over the world in suitable climates for commercial profits, including cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts, and green leafy vegetables.

In finding a potential compound for the treatment of lung cancer, researchers examined the anti-metastatic activity of Sulforaphane (SFN), an isothiocyanate found in cruciferous vegetables on two non-small cell lung cancer (NSCLC) cell lines, A549, and CL1-5 cells.

SFN exhibited more potent cytotoxicity against suspending rather than adherent cancer cells.

The inhibition of SFN was attributed to the induction of anoikis. evidenced by a loss of normal cell-matrix interactions, markers of apoptosis, and activation of the mitochondrial apoptotic pathway.

Furthermore, SFN also inhibited NSCLC cells to form spherical colonies, to induce proliferation.

Moreover, SFN also suppressed the activation of signaling pathways to induce cancer cell adhesion.

Based on the findings, researchers said, "These results provide new insight into mechanisms underlying the chemopreventive ability of SFN and suggest a potential benefit of SFN to interfere with tumor metastasis".

Taken together, Cruciferous vegetables may be considered functional foods for the prevention and treatment of non-small cell lung cancer, pending the confirmation of the larger sample size and multicenter human study.
