#Broccoli's Sulforaphane (SFN) Suppresses the Proteins and Genes Associated with the Onset of Diabetic Lipotoxicity and Cardiomyopathy in Diabetes

Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream to regulate glucose.

Diabetic cardiomyopathy is a complication found in patients with diabetes associated with reducing the heart function in circulating blood through the body, leading to heart failure and increased accumulation of fluid in the lungs.

Diabetic cardiomyopathy is classified into different types, depending on the damage that has been affected, including
* Microangiopathy is a disorder that affects small blood vessels, leading to impaired myocardial blood flow reserve.

* Autonomic neuropathy is a disorder that affects the nerves that manage everyday body functions, leading to abnormal blood pressure, and heart rate.

* Endothelial dysfunction is a disorder associated with the damage of vascular endothelium. The condition is considered an early event of cardiovascular disease.

The most common causes of diabetic cardiomyopathy are myocardial dysfunction in the absence of overt clinical coronary artery disease, valvular disease, and other conventional cardiovascular risk factors, such as hypertension and dyslipidemia.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of diabetic complications, researchers examined the effect of sulforaphane (SFN) on T2DM-induced cardiac lipid accumulation and subsequent cardiomyopathy.

The study included male C57BL/6J mice fed a high-fat diet for 3months to induce insulin resistance, followed by treatment with 100mg/kg body-weight streptozotocin to induce hyperglycemia as the T2DM mouse model compared to age-matched mice fed a normal diet as control.

According to the tested analysis,
* SFN treatment significantly attenuated cardiac remodeling and dysfunction induced by T2DM.

* SFN treatment also significantly inhibited cardiac lipid accumulation, measured by Oil Red O staining, and improved cardiac inflammation oxidative stress and fibrosis, shown by down-regulating diabetes-induced PAI-1, TNF-α, CTGF, TGF-β, 3-NT, and 4-HNE expression involved in oxidation.

* SFN upregulated the expression of the gene associated with the promotion of antioxidant enzymes and decreased the expression in enhancing the initiation of oxidative stress.

* SFN also reversed diabetes-induced inhibition of LKB1/AMPK and its downstream targets associated with the regulation of lipid, cholesterol and glucose metabolism.

Based on the results, researchers wrote, " SFN treatment to T2DM mice may attenuate the cardiac oxidative stress-induced inhibition of LKB1/AMPK signaling pathway, thereby preventing T2DM-induced lipotoxicity and cardiomyopathy".

Taken altogether, broccoli processed abundantly SFN may be considered supplements for the prevention of diabetic cardiomyopathy, pending to the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Coffee Intake Daily and Regularly Is Associated with A Reduced Risk of Stroke, Researchers Say

Coffee, second to green tea is a popular and social beverage all over the world, particularly in the West, made from roast beans from the Coffea plant, native to tropical Africa and Madagascar.
A stroke is a medical condition characterized by interrupted blood supply, thus reducing oxygen and nutrient delivery to the brain, resulting in the death of brain cells.


Intake of coffee and coffee caffeine may have a profound effect in reducing the risk of stroke incidence, a renowned institute study suggested.

In a third National Health and Nutrition Examination Survey (1988-1994; NHANES III) data to examine coffee consumption and stroke with participants aged ≥17 years old, researchers found that
coffee intake of this population showed a significant inverse association to stroke risk incidence, regardless of smoking status.


Interestingly, the study also discovered that frequent coffee drinkers displayed a less occurrence of heart failure, diabetes, and hypertension but the risk of high cholesterol for those consuming ≥3 cups per day.


Dr. Liebeskind DS, the lead author said, "Heavier daily coffee consumption is associated with decreased stroke prevalence" and ". Multivariate analyses revealed an independent effect of heavier coffee consumption (≥3 cups/day) on reduced stroke".

Furthermore, in an analyzed data from a prospective cohort of 83,076 women in the Nurses' Health Study without a history of stroke, coronary heart disease, diabetes, or cancer at baseline, with coffee consumption assessed first in 1980, then repeatedly every 2 to 4 years, and follow-up through 2004, researchers at the Harvard School of Public Health filed the following results

1. The relative risk ratio of stroke in participants coffee consumption (<1 cup per month, 1 per month to 4 per week, 5 to 7 per week, 2 to 3 per day, and >or=4 per day) were 1, 0.98, 0.88, 0.81, and 0.80 repetitively.
2. Intake of coffee and coffee caffeine daily and regularly showed a significantly positive effect in reduced stroke risk after taking to count of other favors, including high blood pressure, hypercholesterolemia, and type 2 diabetes.
3. Risk of stroke is also associated stronger correlation among never and past smokers (RR for >or=4 cups a day versus <1 cup a month, 0.57) than among current smokers (RR for >or=4 cups a day versus <1 cup a month, 0.97).

The findings suggested that long-term coffee consumption was not associated with the risk of stroke but modestly reduce the risk of stroke.


Promisingly, an Asian study of data obtained from the Health Examinees (HEXA) study, involving 146,830 individuals aged 40-69 years demonstrated that high coffee consumption was associated with a 38% lower odds ratio for stroke in women compared to (none drinkers vs. ≥ 3 cups/day drinkers in women but not in men.

The expression of coffee intake in reduced risk of stroke was also found greater in women who are healthy, younger, non-obese, non-hypertensive, non-diabetic, non-smokers, and non-alcohol drinkers.

Taking together, there is no doubt that the intake of coffee daily and regularly has a remarkable effect on the reduced risk of stroke, particularly in women.

Sepsis is a medical condition caused by the body's over-response to infection by releasing a large number of chemicals into the bloodstream, leading to a chemical imbalance that can damage multiple organ systems.

The most common sites of infection leading to sepsis are found in older patients with the lungs, urinary tract, tummy (abdomen), and pelvis.

The causes of sepsis, in most cases, are involved in people with a weakened immune system or a severe infection that allows the inflammation to spread to the entire body.

In order to reveal more information about ginger antisepsis activity, researchers examined zingerone (ZGR), a bioactive compound isolated from ginger on TGFBIp-mediated septic responses.

Similar to that of the above, ZGR reduced the TGFBIp-mediated severe inflammatory responses in human endothelial cells and mice.

Injection of ZGR suppressed TGFBIp-induced sepsis lethality and pulmonary injury.

Myocardial infarction (MI) is a medical condition known as a heart attack caused by the blood flow decreasing or stopping to a part of the heart, leading to damage to the heart muscle.

The study included rats-induced myocardial infarction by subcutaneous injection of isoproterenol (ISO) (150 mg kg(-1)) at an interval of 24 h for 2 days.

Before treatment of SACS, ISO-treated rats showed a significant increase in the levels of serum iron, uric acid, and blood glucose, Na(+) and Ca(2+) associated with electrical abnormalities in the heart.

ISO-treated rats showed a significant decrease in the levels of plasma iron-binding capacity, an indication of inflammation, serum total protein, and albumin/globulin ratio similar to those of patients with abnormal heart rhythm, heart K(+), and heart glycogen involved in weak heart muscles.

In other words, ISO has caused significant damage to the heart functioning physically in myocardial infarction rats.

Furthermore, ISO also induced significant production of proinflammatory cytokines observed by the significant increase in red blood cells, hemoglobin, packed cell volume, white blood cells, neutrophils, platelet count, and fibrinogen level.

Additionally, the heart failure marker of glycoproteins in serum and the heart were increased in both levels/concentrations.

However, oral pretreatment with SACS (40 and 80 mg kg(-1)) daily for a period of 35 days reversed all the biochemical parameters studied in ISO-induced rats.

Collectively, SACS showed a protective effect against ISO-induced myocardial infarction in tested rats.

Taken altogether, alliin may be considered a supplement for the prevention and treatment of myocardial infarction with no side effects, responding to the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Atherosclerosis is a medical condition induced by arterial narrowing due to the accumulation of plague that weakens the arterial wall.
Atherosclerosis is the most common cause of heart disease and stroke in the Western world, depending on the location of oxygenated rich blood flow blockage.

A plague is made up of fat, cholesterol, calcium, and other substances in the blood.

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

With an aim to find potential treatment for atherosclerosis, a leading cause of cardiovascular disease, researchers evaluated the capacity of Allium ursinum (wild garlic) leaf lyophilisate (WGLL; alliin content: 0.261%) to mitigate cardiovascular damage in hypercholesterolemic rabbits.

Selected tested New Zealand rabbits included in the study were divided into three groups: (i) cholesterol-free rabbit chow (control); (ii) rabbit chow containing 2% cholesterol (hypercholesterolemic, HC); (iii) rabbit chow containing 2% cholesterol + 2% WGLL (hypercholesterolemic treated, HCT); for eight weeks.

According to the differentiation at the zero- and eight-week time points, WGLL treatment protects the cardio functioning by mediating the increase in fractional shortening, correct ventricular function; peak systolic velocity; tricuspid annular systolic velocity in live animals.

Furthermore, WGLL improved aortic and coronary flow.

In other words, WGLL exerted cardiovascular protective activity by promoting the function of ventricles in blood circulation via regulating blood pressure.

Moreover, the Western blot analysis revealed WGLL-associated increases in HO-1 protein associated with reduced cardio oxidative stress and decreases in SOD-1 protein involved oxygen metabolism.

Interestingly, WGLL also expressed a significant decrease in aortic atherosclerotic plaque coverage, plasma ApoB, an important component of many lipoproteins involved in atherosclerosis and cardiovascular disease. and the activity of LDH and CK (creatine kinase) in plasma.

Particularly, the chemical compound also inhibited the plasma LDL.

Based on the findings, researchers wrote in the final report, "The results clearly demonstrate that WGLL has complex cardioprotective effects, suggesting future strategies for its use in prevention and therapy for atherosclerotic disorders".

#RedBeetroot, the FDA-Approved Red Food Color E162 and Its Extract Inhibits Breast Cancer Cells Through Its Cytotoxicity

The breasts of women are made of specialized tissue that produces milk (glandular tissue) and fatty tissue. The size of a woman's breast is totally depending on the amount of fat in the breast. Breast cancer is a medical condition caused by cell growth irregularly due to the alternation of cell DNA.

Most cases of breast cancer start in the cells on the surface of the inner lining of the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk, before colonizing the deeper layer tissues to form a tumor through proliferation.

According to the statistics, in the US, the lifetime risk of getting invasive breast cancer of a woman is 1/8. In 2019, approximately 268,600 new cases of invasive breast cancer will be diagnosed, along with 62,930 new cases of non-invasive (in situ) breast cancer.

The 5 years survival rate of breast cancer diagnosed in the localized stage is 100%.

The exact causes of breast cancer are not identified.

The stages of breast cancer can be classified depending on
* The size of the cancer
* Invasive or not
* Whether cancer has spread to the lymph nodes
* Whether cancer has spread to distant parts or organs of the body.

There are 4 stages of breast cancer in TNM rating (where T is referred for tumor size, N is referred for Lymph Node status, and M is referred for invasive or not)
# T: Tumor Size is divided into four classes: T-1 is between 0 - 2 cms, T-2 is between 2 - 5 cms, T-3 is > 5cms, and T-4 is a tumor of any size growing into the chest wall or skin, including inflammatory breast cancer.
#N: No swollen nodes: N-0, some swelling but negative node (not cancerous): N-1a, swelling nodes and positive node (cancerous): N-1b, quite swollen and bunched together: N-2, quite swollen and near the collarbone: N-3.
# M: if no cancer cell is found: M-0, cancer cells found: M-1.

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belongings to the group of Betalains, found abundantly in beets, chard, etc.

In finding a potential compound for the treatment of breast cancer, researchers examined the effect of an extract of red beetroot (Beta vulgaris L.), the FDA-approved red food color E162 on estrogen receptor-positive human breast cancer cells (MCF-7).

According to the tested differentiation, both doxorubicin and the beetroot extract exhibited a dose-dependent cytotoxic effect in MCF-7.

Compared to doxorubicin, the cytotoxicity of beetroot extract was significantly lower.

In other words, at the same doses, doxorubicin is more effective to induce the death of breast cancer cells compared to the beetroot extract.

Due to the limitation of the experimental selection, side effects induced by both treatments cannot be observed.

After taking other factors into account, researchers suggested, the principal betacyanin constituent may play an important role in the cytotoxicity exhibited by the red beetroot extract.

Taken altogether, betacyanins may be considered a remedy for preventing and treating breast cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of betacyanins in supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Antioxidants are stable molecules that inhibit oxidation. Antioxidants are either produced by the host or from dietary sources.

Under normal conditions, antioxidants produced by the body are good enough to counter the presence of free radicals in the body of a healthy individual.

However, in people with a weakened immune system or medical condition, intake of dietary antioxidants may be necessary due to the depletion of the antioxidant enzymes.

Free radicals are unstable molecules with a single electron in the outer ring. Therefore for them to maintain stability, they must either donate or intercept an electron from other molecules, leading to a chain reaction that can not be stopped until they are either inhibited by antioxidants or electrons are paired.

Oxidative stress is an imbalance of the ratio of free radicals and antioxidants in the body caused by either overexpression of free radicals or depletion of antioxidant enzymes produced by the host.

Epidemiological studies suggested that oxidative stress has been found to cause lipid and protein and cellular damage, leading to the onset of chronic diseases, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

Out of many risk factors associated with the onset of oxidative stress, widespread obesity may be one of the significant factors that contribute to the severity of the disease due to oxidative stress.

Mitochondrial dysfunction is found to interfere with cells' function in producing energy for cell or organ needs.

Oxidative balance impairment is another term for oxidative stress.

More precisely, it is caused by either overexpression of reactive oxygen species and/or depletion of antioxidant enzymes produced by the host.

Taken altogether, sinigrin a bioactive ingredient in cruciferous vegetables may be considered a functional supplement for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of a large sample size and multicenter human study.

However, intake of bioactive in the form of supplements should be taken with extreme care, as liver toxicity may be induced if overdosed.
