Bioactive Compound #Vitexin and #EthanolicExtract From the Cynometra Cauliflora Leaves Inhibit Parameters Associated with Obesity, According to Studies

Kyle J. Norton

Obesity is a chronic medical condition caused by excess body fat accumulated to some extent that impacts personal health. The hallmark of obesity is attenuated metabolism and structural cell changes in the obese fat observed by reducing metabolic activity.

According to the statistics provided by the National Center for Health Statistics (2015-2016), in the US, approximately 39.8% of adults aged 20 and over were obese accompanied by another 31.8% overweight.

Visceral fat is a layer of fat that surrounds the organs. Overexpression of visceral fat has been found to promote insulin resistance, diabetes, heart disease, and inflammation.

In obese, accumulation of visceral fat is predominant in the intra-abdominal cavity.

According to the BMI index, a person is considered overweight, if he/she has a BMI between 25 to 29.9 kg/m2. A BMI of 30 kg/m2 or more is an indication of obesity.

In other words, obesity is a progressive chronic disease characterized by abnormal fat accumulation in the body tissues that may impair health.

Similar to other chronic medical conditions including diabetes, cardiovascular disease or high blood pressure, obesity has recently become a serious health problem in the US as nearly 35% of Americans are obese accompanied by a similar percentage of overweight adults.

Obesity and overweight in the U.S. economy have eclipsed $1.7 trillion, equivalent to 9.3 percent of the nation's gross domestic product, according to a new overweight report on chronic diseases.

Dr. Maximilian Tremmel, the lead scientist in a systematic literature review of the economic burden of obesity, wrote, "Almost half of the world’s adult population will be overweight or obese by 2030 if the prevalence continues on the current trend [6] and consequently also the costs attributable to obesity will increase".

And, "A useful example for rapidly rising costs attributable to obesity from these selected studies are the two mentioned studies from Germany [44,46]. The results of the two papers together show that total societal costs in Germany due to obesity have increased from €9.8 million to €12.2 million between 2002 and 2008. Therefore, public health interventions should focus on the prevention of obesity as soon as possible, ideally at a young age".

Cynometra cauliflora is a species of the genus Cynometra, native to Malaysia, and found mainly in northern Peninsular Malaysia.

In finding a potential phytochemical for the prevention and treatment of chronic diseases, researchers examined the anti-obesity and lipid-lowering effects of ethanolic extract of C. cauliflora leaves and its major compound (vitexin) in C57BL/6 obese mice induced by a high-fat diet (HFD).

The study included male C57BL/6 mice induced obesity by feeding with HFD (60% fat) for 16 weeks followed by the treatment that starts during the last 8 weeks of HFD feeding with C. cauliflora leaf extract at 200 and 400 mg/kg/day, orlistat (10 mg/kg) and vitexin (10 mg/kg).

According to the chemical analysis,
* The oral administration of C. cauliflora (400 and 200 mg/kg) and vitexin significantly reduced body weight, adipose tissue and liver weight, and lipid accumulation in the liver compared to the control HFD group.

* Both doses of C. cauliflora also significantly (P ≤ 0.05) decreased serum triglyceride, LDL, lipase, IL-6, peptide YY, resistin levels, hyperglycemia, hyperinsulinemia, and hyperleptinemia compared to the control HFD group.

* C. cauliflora significantly up-regulated the expression of adiponectin, Glut4, Mtor, IRS-1 and InsR genes associated with sustaining weight loss and significantly decreased the expression of Lepr in white adipose tissue involved in stimulating weight gain.

Based on the findings, researchers said, "This study shows interesting outcomes of C. cauliflora against HFD-induced obesity and associated metabolic abnormalities".

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of bacterial infectious diseases, pending the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthy 3,3'‑#Diindolylmethane (DIM) Protects the Kidney Against Injury Caused by Parameters Associated with Oxidativestress

Kyle J. Norton

Kidneys, a pair of bean-shaped organ in the human body not only filter out blood but also balances body fluids by eliminating them and removing them through urinary secretion.
Additionally, the kidney accompanied by the liver and skin form also detoxes the toxins in the body.

Kidney disease is a medical condition associated with a gradually reduced function of the kidney.

Kidney diseases can be classified into 2 types
* Acute kidney disease is a sudden episode of kidney failure or kidney damage associated with reduced kidney function in keeping the right balance of fluid in the body, that lasts a few hours or a few days.

* Chronic kidney disease (CKD) is a condition of kidney damage, leading to a decreased level of kidney function that lasts more than three months.

Depending on the severity of renal damage, patients may experience symptoms of decreased urine output, and fluid retention, that lead to swelling in the legs, ankles, or feet, fatigue, nausea, and physical weakness

Renal protection is a strategy that protects the kidney against injury or damage. In other words, renal protection is a process that maintains the function of the kidneys.

Renal toxicity or nephrotoxicity in most cases is toxicity in the kidneys caused by toxic chemicals and medications.

The most common symptoms of renal toxicity may cause reduced kidney function in eliminating toxins, leading to a reduced amount of urine. swelling of your legs, ankles, and feet from the retention of fluids.

In a serious case, the condition can also result in unexplained shortness of breath. excessive drowsiness or fatigue. persistent nausea. confusion. pain or pressure in your chest.

Depending on the severity of kidney damage, kidney disease can range from mild to severe, including kidney failure.

With an aim to find a natural food for the prevention and treatment of renal diseases, researchers examined the effect of 3,3'‑Diindolylmethane (DIM), a naturally derived indole compound found in the Brassica family of vegetables on kidney injury.

The study included male BALB/c mouse models of AKI that were established using intraperitoneal injections of 10 mg/kg LPS followed by the DIM (40 mg/kg) administered intraperitoneally 24 and 2 h before LPS exposure.
According to the tested analysis,
* DIM significantly mitigated histopathological changes in the kidneys and improved the levels of blood urea nitrogen and serum creatinine.

* DIM also suppressed the LPS‑induced production of reactive oxygen species in the induction of cell apoptosis.

* DIM treatment also significantly decreased the expression of NADPH oxidase 2 (NOX2) and NOX4 which results in oxidative stress in LPS‑treated mice.

More precisely, DIM protected the kidney cell integrity against all parameters expressed by the injection of LPS.

Based on the results, researchers wrote, " Therefore, DIM may exert its renoprotective actions by inhibiting NOX‑mediated oxidative stress and the apoptosis of renal tubular epithelial cells"

Taken altogether, Brassica rapa vegetables may be considered supplements for the prevention of kidney injury, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

3,3'-Diindolylmethane (DIM) Inhibits Epstein-Barr Virus (EBV)-Positive Burkitt's Lymphoma through Its Cytotoxicity Effects

Kyle J. Norton

Herpesvirus is a large family of DNA viruses that cause diseases, depending on the types of viral infection.
According to viral classification, there are 8 types of herpes virus, including human herpesvirus 1 that causes cold sores around the mouth. and 2 that causes genital herpes, herpes zoster that causes chickenpox and shingles, Epstein-Barr virus that causes infectious mononucleosis, cytomegalovirus (CMV) that causes mononucleosis, Human herpesvirus 6 and 7(HHV6 - 7) that causes roseola and Human herpesvirus 8 (HHV8) that causes cancer, such as Kaposi's Sarcoma.

According to statistics, in the United States, half of teens and adults under age 50 are still infected with the oral herpes virus, and about 1 in 8 have an infection with the genital herpes virus.

All herpes viruses like other viruses are nuclear-replicating, infecting healthy cells nuclei through replication of viral DNA and transcription of viral mRNA.

People infected may experience symptoms of painful sores in the genital area, anus, buttocks, thighs, itching, painful urination, vaginal discharge,..... and tender lumps in the groin.

Epstein-Barr virus (EBV)-positive is early in the course of the illness associated with VCA-IgM antibodies caused by EBV infection.

The Epstein–Barr virus (EBV), is also known as human gammaherpesvirus 4, one of the nine known human herpesvirus types in the herpes family.

Unlike other types of the herpes virus, symptoms of EBV may vary, including extreme fatigue, fever, sore throat, and swollen lymph nodes.

Conventionally, there is no specific treatment for EBV. The most common treatment is to improve quality of life by targeting symptoms such as reducing swelling of tissues with corticosteroids

In finding a potential plant with infectious diseases, researchers compared the effects of indole-3-Carbinol (I3C) a natural product found in broadly consumed plants of the Brassica genus, such as broccoli, cabbage, and cauliflower, effects on the various vegetablesEpstein-Barr virus (EBV)-positive

Based on the tested results, I3C efficiently induced apoptosis in cell lines derived from EBV-positive Burkitt's lymphomas (virus latency I/II).

However, I3C did not have any cytotoxic activity against EBV-negative Burkitt's lymphomas and immortalized EBV-infected lymphoblastoid cell lines (virus latency III).

Furthermore, The effect of I3C on cell viability. in EBV-positive Burkitt's lymphoma was attributed to C6-methylated derivative, but not other 3-substituted indoles.

Moreover, 3C treatment caused apoptosis characterized by loss of cancer cell mitochondria membrane potential and caspase activation by alternating the expression of proteins involved in the control of apoptosis and transcription regulation in EBV-positive Burkitt's lymphoma cell lines.

Particularly, I3C derivative 3,3'-diindolylmethane (DIM) prolonged survival and reduced tumor burden of mice xenotransplanted with EBV-positive Burkitt's lymphoma Daudi cells.

Based on the results, researchers said, "These results, together with previous reports from clinical trials indicating the lack of toxicity in humans of I3C and derivatives, support the use of these compounds as a new therapeutic approach for treating patients with endemic (EBV-positive) Burkitt's lymphoma".

Taken altogether, Brassica Genus may be considered anti the Epstein-Barr virus (EBV)-positive induced cancer remedy, pending the confirmation of the larger sample size and multicenter human study.

#RapeseedProtein Inhibits #LiverFibrosis Associated with Over expression of Oxidativestress, Acccording to Studies

Kyle J. Norton

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing them to other parts of the body.
The liver also produces cholesterol, a waxy substance which aids digestion, builds cell membranes, and produces vitamin D and steroid hormones.

Liver disease is a class of medical conditions characterized by reducing partly or completely the function of the liver, including liver failure.

Liver fibrosis is a condition characterized by overly exuberant wound healing causing excessive connective tissue builds up in the liver. In other words, hepatic fibrosis is a portion of the liver that was affected by scar formation.

The exact causes of liver fibrosis are unknown. However, certain risk factors such as repeated liver injury caused by a virus infection, and a blockage in the bile ducts are found to elaborate on the onset of liver fibrosis.

Long-term untreated liver fibrosis can lead to cirrhosis, a condition of severe scarring of the liver, and life-threatening liver disease, including liver cancer.

The most common scale to grade the stage of liver fibrosis is the METAVIR scoring system. By examing the grade and stage of liver fibrosis of patients, their doctors can now determine the degree of severity to which patients' liver is affected.

The severity of the grades ranges from A0 to A3:
A0: no activity
A1: mild activity
A2: moderate activity
A3: severe activity

The severity of fibrosis stages ranges from F0 to F4:
F0: no fibrosis
F1: portal fibrosis without septa
F2: portal fibrosis with few septa
F3: numerous septa without cirrhosis
F4: cirrhosis, the most severe stage of fibrosis

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of liver diseases, researchers examined the effect of the potential crucial pathways involved in ameliorating liver fibrosis of the rapeseed protein-derived peptide DHNNPQIR (RAP-8).

According to the analysis of the fibrotic and RAP-8 treated mice by measuring cell cycle and oxidative stress in LX-2 cells and liver samples,

* RAP-8 treated mice showed a change of modules that were mainly enriched in cell division, nuclear division, and mitotic cell cycle process by alternating the Kyoto Encyclopedia of Genes and Genomes associated with enriched in the cell cycle.

* More precisely, the regulations of three core genes (Cenpp, Cyp2c55, Serpinh1) by RAP-8 were verified to target for treating liver fibrosis.

* RAP-8 induced cell cycle arrest and prevents oxidation stress in the induction of liver fibrosis

Based on the results, researchers wrote, " As a promising therapeutic candidate for hepatic fibrosis treating, RAP-8 exhibited anti-fibrotic effects via exerting cell cycle arrest and inhibiting oxidative stress."

And "However, because variability exists in the timing and methods of detection for SFN conjugate levels, it is difficult to compare systemic SFN levels across studies".

In other words, SFN protected the integrity of the neurons against neuron damage through its anti-neuroinflammatory and neuroprotective activities.

Taken together, rapeseed protein may be considered a supplement for the prevention and treatment of liver fibrosis, pending the confirmation of the larger sample size and multicenter human study.

Herbal #Angelica Inhibits Neuro-Inflammation Through Its Anti-ProinflammatoryCytokines Activity, Researchers Reveal

Kyle J. Norton

The central system is the complex of nerve tissues including the brain and spinal cord with functions to respond to the integrating sensory information.

Neuroinflammation is a process of the immune system that responds to harmful pathogens that induced oxidative stress in the brain.

Neuroinflammation is also the inflammation of nervous tissue associated with the immune response in the first phase of infection including infection, traumatic brain injury, toxic metabolites, or autoimmunity.

There are many risk factors associated with the onset of neuroinflammation similar to other tissue inflammation, including infection, traumatic brain injury, toxic metabolites, or autoimmunity.

Inflammation is a natural response of the immune system in which the macrophages of the first defense stimulates the production of a blood palette to cover the wound in case of injury and the production of proinflammatory cytokines to the site of infection to fence off the foreign pathogens.

Inflammation also can be triggered by the immune mistakenly attacking the body tissues in cases of autoimmune disease.

Neuroinflammatory disease can be acute or chronic. Chronic neuroinflammation that leads to the activation of glial cells and the recruitment of inflammatory cytokines into the brain is found to associate with neurodegenerative diseases.

Most cases of neuroinflammatory disease associated with the loss of neuronal structure and function can be measured by the typical inflammatory markers, including CSF Aβ1-42, tau, and p-tau181, and a panel of 37 neuroinflammatory markers in patients with Alzheimer's disease.

On finding a natural ingredient for the prevention and treatment of neurological disorders, scientists examined the effects of dangguishaoyao-san (DSS) (Angelica sinensis, Paeoniae radix, Rhizoma Ligusticum, Poria cocos, Rhizoma Atractylodis Macrocephalae, and Rhizoma Alismatis) on lipopolysaccharide (LPS)-stimulated BV-2 microglial cells.
Angelica or Danggui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen the heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the results of BV-2 cells pretreated with 0.58-1.16 mg/mL of DSS for 2 h and then treated with 1 μg/mL LPS for 24 h,
* DSS treatment at concentrations of 0.58-1.16 mg/mL did not induce significant cytotoxicity.

* DSS attenuated the release of pro-inflammatory factors, such as interleukin-1β (IL-1β), iNOS, and tumor necrosis factor-α (TNF-α) in LPS-induced BV-2 cells.

* DSS attenuated the mRNA expression of pro-inflammatory cytokines, TLR2, and TLR4 by decreasing TLR4 and TLR protein levels and decreased levels of phosphorylation of IκB which regulates the cellular response to inflammation in LPS-induced BV-2 cells.

* DSS also down-regulated the nuclear translocation of p65 in the initiation of cancer cell proliferation and survival.

Based on the results, researchers suggest, " DSS has a protective effect on neuroinflammation in LPS-induced BV-2 microglial cells through the TLRs/NF-κB signaling pathway".

Taken together, Agelica may be considered an antioxidant remedy for the treatment of neuroinflammation, pending the confirmation of the larger sample size and multicenter human study.

#TomatoJuice JC Inhibits the Onset of #PulmonaryDiseases Through Its Antixodant Activity, According to Studies

Kyle J. Norton

Pulmonary diseases are a class of diseases associated with the lungs, including asthma, COPD, influenza, pneumonia and tuberculosis, and lung cancer.
In other words, any diseases involved in breathing problems that reduce the function of the lung are pulmonary diseases.

The most common pulmonary diseases are

* Asthma
Asthma is a chronic inflammatory disease affecting the airway of the lung with recurring symptoms, such as wheezing, chest tightness, shortness of breath, and coughing. The disease affects people of all ages, and mostly starts during childhood.

* Pneumothorax
Pneumothorax is a condition of collection of air within the pleural cavity, from either the outside or from the lung.

* Bronchitis
Bronchitis is a condition of an inflammation of the mucous membranes of the bronchi, the larger and medium-sized airways that carry airflow from the trachea into the lung parenchyma.

* COPD (chronic obstructive pulmonary disease)** Emphysema is a type of COPD with a long-term and progressive condition that causes shortness of breath, depending on the stage of lung function associated with the damage of tissues of the air sacs (alveoli) in the lungs.

** Chronic bronchitis
Chronic bronchitis is a chronic inflammation of the lung’s bronchi that cause the increased production of mucus in the lung associated with difficulty of breathing.

*Lung cancer is a condition of the abnormal growth of the cells in the lung's tissue. The most common form of primary lung cancers are derived from epithelial cells

* PneumoniaPneumonia is a condition of the inflammation of the lung caused by bacteria, such as bacterial infection, Streptococcus pneumonia, or influenza viruses in most cases.

* Pulmonary edema
Pulmonary edema is a condition of fluid accumulation in the air spaces and parenchyma of the lungs that lead to difficulty of breathing and respiratory failure.

Tomato is a red, edible fruit, genus Solanum, belonging to the family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find a potential ingredient for the treatment of pulmonary disease, researchers examined the dietary supplementation of tomato juice, TJ protection against the developing lung injury caused by breathing hyperoxic gas.

Neonatal mice (C57BL6/J) breathed either 65% O2 (hyperoxia) or room air from birth until postnatal day 7 (P7d)selected to the study underwent necropsy at P7d or raised in room air until adulthood (P56d). In a subset of both groups, drinking water was replaced with TJ (diluted 50:50 in water) from late gestation to necropsy.

In P7d, TJ injection increased pulmonary total antioxidants (TAC) and COL1α1 expression associated with pulmonary fibrosis and attenuated the hyperoxia-induced increase in production ROS without affecting the changes in lung structure.

Furthermore, in P56d, TJ increased TAC, decreased oxidative stress, and reversed the hyperoxia-induced increase in bronchiolar smooth muscle by ameliorating the production of pro-inflammatory cytokines.

Based on the findings, researchers said, "TJ supplementation during and after neonatal exposure to hyperoxia protects the lung from some but not all aspects of hyperoxia-induced injury".

In other words, TJ protects the lung against injury by stimulating the expression of circulation of antioxidant concentration.

Taken altogether, tomatoes may be considered supplements for the prevention and treatment of pulmonary diseases, pending the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#P-coumaricAcid (CA) from #Turnip Induces the Production of production of Occludin SAgainst the Risk of #Hypoxia, Researchers Suggest

Kyle J. Norton

Hypoxia is an urgent medical emergency of deficiency in the amount of oxygen reaching the tissues.
In other words, if your body does not get enough oxygen, you might get hypoxia. The condition has been found to induce brain, liver, and other organs damaged just minutes after symptoms starting.

The most common symptoms of hypoxia are associated with a lack of oxygen that reaches the organs and tissues, including changes in the color of the skin, ranging from blue to cherry red, confusion, cough, increased heart rate, rapid breathing, and shortness of breath.

The most common risk factors associated with the onset of hypoxia are damage to the lung due to trauma, medical conditions such as an asthma attack, lung diseases such as chronic obstructive pulmonary disease (COPD), emphysema, bronchitis, pneumonia, pulmonary edema, intake of certain medications that hold back breathing and people with severe anemia.

The most common treatment for hypoxia is to withdraw medications that cause the condition and give additional oxygen to the patient quickly. Almost all patients respond to additional oxygen supplied by a nasal cannula.

Tibetan turnip (Brassica rapa L.), native to the Tibet region, is a medical plant used in Tibet as a “tonic and anti-hypoxia” “heat-clearing and detoxification” and “alleviating fatigue” herbal medications.

In finding a potential compound for the treatment of hypoxia, researchers conducted a joint study led by Zhejiang University on an animal model.

Selected male ICR mice included in the study were assigned to the normoxia group and several hypoxia groups, given sterile water, p-coumaric acid (CA), or dexamethasone orally, once daily for four consecutive days.

Based on the results of the analysis in a hypoxic environment of 9.5% O2,, mice treated with 100 mg/kg body wt CA showed a significantly lower lung water content (LWC), and the protein content of bronchoalveolar lavage fluid (BALF) content than mice in the hypoxia vehicle group.

Where lower lung water content (LWC), and the protein content of bronchoalveolar lavage fluid (BALF)content are the symptoms of the deficient amount of oxygen in the lung.

Moreover, mice treated with CA also protect the lung functioning by showing an intact lung blood gas barrier, increased levels of plasma total NO,

Additionally, levels of preproendothelin-1 (PPET1), associated with vasoconstriction were also decreased in the CA treatment group.

More importantly, CA induced the production of occludin, a tight junction protein in maintaining the integrity of the lung epithelial barrier against hypoxia.

Taken altogether, turnip may be considered herbal medicine for the prevention and treatment of hypoxia, pending the confirmation of the larger sample size and multicenter human study.
