#Cucumbers Inhibit #GastricCancer Involved in the Regulation of Cancer Cell Growth and Survival, According to Studies

Kyle J. Norton

Gastric or mouth cancer is a chronic medical condition characterized by abnormal cells growing in the tissues of the stomach.

The stomach-formed part of the digestive system is located between the esophagus and the small intestine and is a muscular, hollow of the digestive tract.

Most cases of gastric or mouth cancer start in the mucus-producing cells of the inside lining of the stomach due to the alternation of DNA. Adenocarcinoma is the most common type of gastric cancer.

At the early stage, due to the small size of the tumor, most patients with gastric cancer are asymptomatic.

However, as cancer continues to grow, it may induce general symptoms shared by all cancer and specific symptoms such as mild aches in the upper part of the body, difficulty swallowing, feeling full or bloated after a small meal, vomiting blood, or having blood in the stool.

At this stage, gastric cancer also induces secondary metastasis, by infecting other healthy tissues and organ far away from the original site.

Out of many risk factors associated with the onset of gastric cancer including diet, long-term inflammation of the stomach, smoking, and obesity, some researchers suggested low levels of gastric acid production by the stomach may have a strong implication in facilitating the onset of the condition.

Dr. Helge L. Waldum, the lead scientist in the analysis of the correlation between Helicobacter pylori and gastric acid, wrote, "Helicobacter pylori (Hp) is the main cause of gastritis, peptic ulcer disease, and gastric cancer. There are still unanswered questions related to the interaction between Hp and man, like what determines the susceptibility for the initial infection and the mechanisms for the carcinogenic effect".

And, "The initial infection seems to require a temporal gastric hypoacidity. For Hp to survive in the gastric mucous layer, some acidity is necessary. Hp itself is probably not directly carcinogenic. Only when inducing oxyntic mucosal inflammation and atrophy with hypoacidity, Hp predisposes for gastric cancer. Gastrin most likely plays a central role in the Hp pathogenesis of duodenal ulcer and gastric cancer".

The cucumber plant is a species of Cucumis Sativus, belongs to the family Cucurbitaceae and native to Western Asia. It is also a creeping vine with roots in the ground and grows up with the support of frames.

With an aim to find a potential compound for the treatment of gastric cancer, researchers examined the effect of Ecballium elaterium (L.) A. Rich (Cucurbitaceae) (squirting cucumber) on cell lines of gastric and esophageal origin namely called AGS (human gastric carcinoma) and KYSE30 (human esophageal squamous cell carcinoma).

The study included AGS and KYSE30 cancer cell lines incubated for 24, 48, and 72 h with an aqueous extract of the fruits of E. elaterium prepared and freeze-dried.

In AGS, at the IC(50) values of 2.5, 0.7, and 0.7 μg/ml, the aqueous extract for the cell line after 24, 48, and 72 h, respectively showed significant induction of cell apoptosis.

Similarly, KYSE30 cell line induced apoptosis was found at IC(50) values for KYSE30 cell line in doses of 500, 150, and 125 μg/ml after 24, 48, and 72 h, respectively.

Furthermore, the injection of the extract also showed a significant increase in cell density at the G2/M phase, according to cell cycle analysis.

Moreover, the efficacy of the extract against gastric cancer cell lines was attributed to several compounds in the induction of cytotoxicity.

Based on the findings, researchers said, "The results of the current study showed that the freeze-dried aqueous extract of E. elaterium fruit has a cytotoxic effect on gastric and esophageal cancer cell lines by means of apoptosis".

In order to reveal more information on cucumber anti-gastric cancer activity, researchers investigated the effects of Cucurbitacin B (CuB), a natural compound found in Cucurbitaceae in cisplatin (DDP)-resistant human GC cell line SGC7901/DDP.

According to the tested assays, CuB treatment significantly suppressed SGC7901/DDP cell proliferation and induced caspase-dependent apoptosis, and autophagy.

Interestingly, the activation of autophagy was mediated through CuB- inhibition of the protein (TORC1) involved in the regulation of cancer cell growth and survival.

In order to reveal more information of CuB anti-gastric cancer potency, scientists examined the CuB activity as a cancerous inhibitor of protein phosphatase 2A (CIP2A), a human oncoprotein that is overexpressed in multiple kinds of tumors including gastric cancer (GC).

In cisplatin (DDP)-resistant human GC cell line SGC7901/DDP, CuB treatment exerted a similar effect as of that of the aforementioned study in the inhibition of SGC7901/DDP cell proliferation, induced caspase-dependent apoptosis, and autophagy.

Moreover, CuB inhibited mTORC1 as showed in the previous study via the inhibition of CIP2A which activates the protein phosphatase 2A (PP2A) in the induction of cancer cell proliferation.

Based on the results, researchers said, "CuB can inhibit the proliferation, induce caspase-dependent apoptosis, and autophagy of SGC7901/DDP cells by suppressing CIP2A/PP2A/mTORC1 signaling axis".

Taken altogether, cucumber processed with a high amount of Cucurbitacin B (CuB) as the main ingredient may be considered supplements for the prevention and treatment of gastric cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Cucurbitacin B (CuB) in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

#CranberryExtract Did not Prevent Urinary Tract Infection UTI in #MultipleSclerosis (MS) Patients, According to Studies

Kyle J. Norton

Infection is an inflammatory condition caused by a microbial invasion of body tissues
Most cases of bacterial infection are treated with antibiotics. However, bacteria are very adaptable, and overuse of antibiotics has been found to make them resistant to antibiotics.

The most common symptoms of bacterial infection include redness and heat, swelling, fever, and pain at the site of infection.

Urinary tract infection (UTIs) mostly occurred in women is an infection that affects the bladder and urethra caused mostly by bacterial strains Escherichia coli (E. coli), commonly found in the gastrointestinal (GI) tract.

UTIs have been found to limit the function of the bladder and induce kidney infection.

The most common symptoms of urinary infection are the persistent urge to urinate, urinary burning sensation, cloudy urine, and smelly urine.

In severe cases, patients may also experience symptoms of blood in the urine and bladder and pelvic pain.

The most common risk factors for urinary tract infection are gender, females are more like to have UTI comapred to males, sexual activity, use of birth control, and menopausal women.

Multiple sclerosis (MS) is a potentially disabling disease of the brain and spinal cord (central nervous system) casued by the immune system attacking the protective sheath (myelin) that covers nerve fibers.

MS causes communication problems between the brain and the rest of the body.

SHARON BEGLEY in examing the genetic preposition involved in MS wrote, " A newly discovered genetic mutation gives people who carry it a whopping 70 percent chance of developing multiple sclerosis, potentially opening the way toward new therapies for a debilitating neurological disorder whose ultimate cause has stumped researchers for decades".

The author continued, "The 70 percent chance of developing MS compares to a 0.1 percent risk in the general population, and the 100 or so previously known genetic variants increase the chance of MS to only about 0.3 percent at most, with no single gene packing much of a punch".

In other words, people having certain genes inherited from their parents are at a substantial risk of MS developing comapred to people who do not.

According to the statistics, in 2010/2011, approximately 93,500 Canadians were diagnosed with the disease, or 290 cases per 100,000 population. Believe it or not, MS is the most prevalent in Canada comapred to other parts of the world.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the prevention of urinary infections, researchers examined the effect of cranberry extract (CE) in multiple sclerosis (MS) patients suffering from urinary disorders.

The prospective, double-blind study included a total of 171 adult MS outpatients with urinary disorders presenting at eight centers that received either cranberry extract ((36 mg proanthocyanidins per day) or a matching placebo for a year.

According to the results of the follow-up, the was no difference between cranberry and placebo).
In other words, there was no difference in time to the first symptomatic UTI distribution across 1 year, with an estimated hazard ratio of 0.99, 95% CI [0.61, 1.60] (p = 0.97).

Furthermore, the secondary endpoints and tolerance also did not differ between groups.

Based on the findings, researchers aid, "Taking cranberry extract versus placebo twice a day did not prevent UTI occurrence in MS patients with urinary disorders".

Taken together, cranberry can not be considered a remedy for the prevention and treatment of urinary disorders in MS patients, pending the confirmation of the larger sample size and multicenter human study.

#Honey's #PropolisExtract Exhibits Considerable Ameliorative Effects Against #Asthma, According to Studies

Kyle J. Norton

Scientists may have found a natural alternative for the treatment of asthma, according to studies.Asthma is a chronic inflammation of the airway of the lung with recurring symptoms. Asthma, attacks triggered by the stimuli can induce airway swollen and inflamed, leading to muscle constriction.

During asthma attacks, patients may experience symptoms of difficult breathing, and severe wheezing in the breath, and coughs that won't stop.

According to the statistic provided by the Centers for Disease Control and Prevention (CDC), 1 in 13 people in the US has asthma.

In other words, more than 26 million Americans have asthma, with half of the patients are children.

If you experience symptoms of a cough which is often worse at night or early in the morning, wheezing in your breath, accompanied by chest tightness and shortness of breath, you may be at the early stage of asthma. Please check with your doctor to rule out the possibility.

Genetic and environmental factors are the most common risk factors associated with the onset of asthma. Some researchers suggested that the widespread obesity over the past few decades may also have a strong implication on the onset of the disease.

Dr. David A. Beuthe r, the lead scientist in the concern of the association of asthma and obesity said, "Cross-sectional and prospective cohort studies of humans have shown a modest overall increase in asthma incidence and prevalence in the obese, although body mass index does not appear to be a significant modifier of asthma severity".

And, "Studying the obesity-asthma relationship in large cohorts, in which self-reports are frequently used to ascertain the diagnosis of asthma, has been complicated by alterations in pulmonary physiology caused by obesity, which may lead to dyspnea or other respiratory symptoms but do not fulfill accepted physiologic criteria for asthma".

These results strongly suggested that if you are obese, your risk of asthma is increased substantially.

Honey is a miraculous product made by bees using nectar from flowers.

The rich golden liquid is considered one of the healthy sweet food for replacing the use of white sugar and artificial sweetener.

Scientists on finding a natural remedy for the treatment of asthma investigated the effect of aerosolized honey on airway tissues in a rabbit model of ovalbumin (OVA)-induced asthma.

The study included 40 New Zealand white rabbits sensitized twice with a mixture of OVA and aluminum hydroxide on days 1 and 14.

Honey treatments were given from day 23 to day 25 at two different doses (25% (v/v) and 50% (v/v) of honey diluted in sterile phosphate buffer saline.

During the study, the aerosolized honey treatment group restored the epithelium, mucosa, and submucosal regions of the airway infected by the application of OVA.

Furthermore, the aerosolized honey treatment was also found to reduce the number of airway inflammatory cells present in bronchoalveolar lavage fluid and inhibit goblet cell hyperplasia, an indication of airway remodeling in asthma.

The findings strongly suggested that aerosolized honey has a significant effect on the treatment and management of asthma in animal models.

Additional differentiation of the modulatory effect of honey, royal jelly, and propolis extract on peripheral blood leukocytes and lung inflammation in a mouse conalbumin-induced asthma model in mice model divided into* The mice in group I that were not sensitized or treated; they were kept as controls.

* The mice in group II were sensitized and challenged with conalbumin. Twenty-four hours after the first challenge with antigen.

* The mice in group III received 0.5 mg/kg of dexamethasone intraperitoneally per day for 18 consecutive days and were kept as positive controls.

* The mice in groups IV, V, and VI received 650, 1000, and 30 mg/kg of honey, royal jelly, and propolis (aqueous and ethanolic extract), respectively, once per day for 18 consecutive days.

At the end of the experiment, by comparing the blood sample before and after treatment, researchers showed
* The groups treated with propolis extract exhibited considerable ameliorative effects against asthma, probably due to the expression of flavonoids and phenolics that exert antioxidative effects

* Honey- or royal jelly-treated groups exhibited an increased incidence of asthma cascade events due to increased inflammatory cells, probably due to the antagonist to bronchial asthma cases.

The results clearly indicated that not all honey is the same. Some can be used for asthma while others can not, probably due to types of chemicals compounds found in the pollen

Taken altogether, honey and other bee products may be considered functional remedies for the prevention and treatment of asthma, pending the confirmation of the types of honey in a large and multicenter human study.

#Healthyfood #Blackbeans Induce #Osteocarcinoma Cells Death Associated with the Protein, RNA, and DNA Expressions, According to Studies

Kyle J. Norton

Intake of black bean regular may reduce risk and delay progression and treatment of osteosarcoma, an invasive form of bone cancer. Osteosarcoma is a condition of bone cancer caused by irregular cell growth in bone, affecting mostly adolescents and young adults.

Black beans, small roughly ovoid legumes with glossy black shells, genus Phaseolus, belongings to the family Fabaceae can be bought in most grocery stores all around the year in dried and canned forms. It is believed that the black bean was the first domesticated growth in South America.

According to the Chinese University of Hong Kong, hemagglutinin isolated from an edible legume, namely black bean, showed an anti-proliferative activity against tumor cell lines MG-63 and HepG2 but not the normal cell line.

Malignant osteosarcoma cells treated with the phytochemical also exerted a significant effect in shrinking tumor cells as well as inducing apoptosis.

The study also stated that hemagglutinin also induced cell death by interfering with the first stage of tumor cell division in the expression of protein, RNA, and DNA and inducing mitochondrial membrane depolarization in controlling the plasma membrane of a muscle or nerve cell

Furthermore, the study also suggested that the efficacy of hemagglutinin in causing cell death may be attributed to the function of protein in increased regulation of pro-inflammatory expressions.
Dr. Dan X, the lead author said, "A novel hemagglutinin from an edible legume (showed) a growth-inhibitory effect on osteosarcoma and liver cancer cells" and " The hemagglutinating activity of this hemagglutinin was demonstrated to be ion-dependent and stable over a wide range of temperature and pH values".

The findings suggested that black beans may be considered a functional food in improved bone health as well as reducing risk and treatment of osteosarcoma, but further studies are necessary to confirm its potency.

#Bilberon25, the Proportion of #Anthocyanins From Bilberry Protects the Skin Against Allergic Contact Dermatitis in Vivos, Researchers Say

Kyle J. Norton

Contact dermatitis is a skin condition with symptoms of a red, itchy rash caused by direct contact with a substance or an immune overreaction against contact allergens.

Many chemicals and nonchemical substances have been found to induce such abnormal reactions, including cosmetics, fragrances, jewelry, and plants.

Most cases of contact dermatitis are nonlife-threatening, however, the condition can induce certain discomforts, particularly in people with an allergic reaction to other conditions.

Contact dermatitis can be healed within a week or 2. The condition can be preventable if the cause is identified.

Besides the common symptoms of a red, itchy rash on the skin, some people may experience more serious symptoms including crusting swelling, burning, or tender skin.

Dr. Imbesi S, the lead scientist in the explanation of the immune involvement of the allergic contact dermatitis wrote, " an inflammatory skin condition due to pro-inflammatory and toxic factors able to activate the skin innate immunity (irritant contact dermatitis) or caused by a T-cell- mediated hypersensitivity reaction (allergic contact dermatitis)".

Bilberry is a species of low-growing shrub in the genus Vaccinium, belongings to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

In the urgency to find a natural ingredient for the treatment of chronic allergic contact dermatitis with no side effects, researchers examined the bilberry extract (Bilberon-25) in the alleviation of pruritus in a mouse model of chronic allergic contact dermatitis.

The study involved BALB/c mice with chronic allergic contact dermatitis induced by 3 weeks of repeated application of 2,4,6-trinitro-1-chlorobenzene (TNCB) following by the administration of Bilberon-25 orally for 3 weeks after sensitization with TNCB.

During the experiment, administered Bilberon-25 orally significantly attenuated the TNCB-induced increase in scratching behavior, compared to that of dexamethasone.

Both Bilberon-25 and dexamethasone inhibited the ear swelling.

Additional differentiation of the Bilberon-25 efficacy, the action of the herbal remedy inhibited symptoms of chronic allergic contact dermatitis by alleviating T helper cell type 2 (Th2) predominance of the lesional skin.

Based on the findings, researchers wrote in the final report, "Anthocyanins from bilberry might be beneficial for the treatment of chronic pruritus which can occur in patients with inflammatory skin diseases such as atopic dermatitis".

In order to reveal more information about anthocyanin activity against chronic allergic contact dermatitis, researchers at Chiba Univ compared the anti-pruritic effect of anthocyanin-rich quality-controlled bilberry extract and anthocyanidin-rich degraded extract in a mouse model of allergic contact dermatitis.

Once again, BALB/c mice with allergic contact dermatitis were induced by 4 weeks of repeated application of 2,4,6-trinitro-1-chlorobenzene (TNCB) followed by administration of Bilberon-25 orally for 4 weeks.

Compared with nonheated Bilberon-25, the proportion of anthocyanins in heated Bilberon-25 decreased, and the proportion of anthocyanidins was increased in a heated-time-dependent manner.

Treatment with non-heated Bilberon-25 significantly attenuated the TNCB-induced increased scratching behavior, whereas treatment with 2 h-heated Bilberon-25 did not.

Particularly, 300 μg/mL nonheated Bilberon-25 showed significant inhibition of degranulation in RBL-2H3 mast cells.

The results suggested that the anthocyanin-rich but not anthocyanidin-rich bilberry extract inhibited skin diseases involving pruritic symptoms, such as chronic allergic contact dermatitis, atopic dermatitis, and rhinitis.

Taken altogether, bilberry-processed abundantly bioactive compound anthocyanoside may be considered a functioning remedy for the treatment of symptoms in patients with chronic allergic contact dermatitis, pending the validation of a larger sample size and multicenter human study.

#Q-Actin™, an Aqueous Extract of Cucumis Sativus #Cucumber Improves the Pain Scores in Patients with #Osteoarthritis

Kyle J. Norton

Osteoarthritis (OA) is a chronic condition affecting the joints in the hands, knees, hips, and spine mostly occurred in older adults.

More precisely, OA is a result in which the protective cartilage on the ends of your bones wears down over time due to repetitive movement of the same joints that cause the thinning cartilage, leading to bone rubbing on bone.

According to the statistics, in the US, over 25 million people are living with OA.

The precisive characteristics of osteoarthritis are aching pain(5), stiffness(6), or difficulty of moving the joint or joints(7). The pain usually gets worse in a change of weather, at night, and in advanced diseases, the pain can occur even at rest(8).

Today management of osteoarthritis (OA) focuses on pain relief and improved physical function through pharmacological, nonpharmacological, and surgical treatments(4).

Believe it or not, other causes of OA also include
1. Process of wear and repair
Osteoarthritis (OA), widespread degenerative disease of skeletal joints(34) and is often associated with senescence invertebrates. arising from long-term wear and tear, heavy long-term use, or specific injuries(35).

The improper repair process of injuring joints can also result in symptoms of Osteoarthritis (OA)(36) in old age(37), according to TCM.

2. Nutrient deficiency and overload
Poor nutritional conditions experienced early in life are linked to the greater prevalence of osteoarthritis (OA)(35)(38), such as vitamin D.

On the other hand, nutrient overload and metabolic surplus, such as obesity may contribute to the early onset of osteoarthritis (OA)(38).

3. Cartilage
Cartilage is a flexible connective tissue that cushioned the ends of bones in your joints and allowed the joints to move smoothly. Rough or wears down cartilage due to aging or damage can cause pain due to the bone in the joint rubbing against another bone(39)(40).

In other words, osteoarthritis (OA) may also be the result of injuring (35), overuse(15)(16), Rheumatoid Arthritis(41), etc.

The cucumber plant is a species of Cucumis Sativus, belongs to the family Cucurbitaceae, native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

With an aim to find a potential compound for the treatment of osteoarthritis, researchers evaluated the efficacy of Q-Actin™, an aqueous extract of Cucumis sativus (cucumber; CSE) against glucosamine-chondroitin (GC) in the management of moderate knee OA.

The randomized double-blind, parallel-group clinical trial took place in three different centers. included 122 patients (56 males and 66 females) aged between 40 and 75 years and diagnosed with moderate knee OA assigned to 2 groups (n= 61).

Among the groups, the GC group received orally the generally prescribed dose of 1,350 mg of GC twice daily and the CSE group received orally10 mg twice daily of CSE.

According to the assays, on days 30, 60, 90, 120, 150, and 180 compared to the baseline (Day 0), the WOMAC score in the CSE group was decreased by 22.44% and 70.29% on days 30 and 180, respectively, compared to a 14.80% and 32.81% respectively decrease in the GC group.

The pain scores also showed similar trends in flavor to the CSE group. There was no adverse effect on the course of the experiment.

In other words, CSE is more effective for the treatment of OA compared to that of GC.

Dr. the lead scientists wrote, "The use of 10 mg CSE, twice daily, was effective in reducing pain related to moderate knee OA and can be potentially used in the management of knee pain, stiffness, and physical functions related to OA".

Taken altogether, cucumber extract may be used for the treatment of OA, pending the confirmation of the larger sample size and multicenter human study.

#Fennel's #Anethole Suppresses #LipidPeroxidation and ROS Generation Associated with Acute and Chronic Inflammatory-Related Disease, Researchers Show

Kyle J. Norton

Fennel may process a strong anti-inflammatory activity in reducing the risk of tissue injury and cell apoptosis, some scientists suggested.

Acute inflammation is a natural and systematic immune response to protect our body against the invasion of foreign bacteria and viruses by the initiated production of inflammatory cytokines to the site of infection.

However, overproduction of pro-inflammatory cytokines initiated by the immune system for a prolonged period of time may lead to cell damage and sometimes death of the healthy cells in the infectious site and nearby cells.

These complications can also cause improper healing for the forming of scars and damage to the tissue of the organ, affecting and limiting the organs' functioning, such as liver scars caused by hepatitis virus infection.

Badly enough, some proteins process the duo functions of pro and anti-inflammation. Under abnormal conditions, they can switch from their original action of anti-inflammatory cytokines to pro-inflammatory cytokines.

Furthermore, chronic inflammation occurred that lasts for months or years when the immune system responds to the site of infection failing to eliminate agents that cause acute inflammation.

An autoimmune disorder attacks normal healthy tissue, mistaking them for pathogens that can also cause chronic inflammatory disease.
Most cases of inflammation, are treated with NSAIDs, or non-steroidal anti-inflammatory drugs. However, in some people intake of these medications can cause common and serious side effects, including gastrointestinal issues, such as pain, constipation, diarrhea, gas, nausea, stomach ulcers, kidney problems, anemia,.... and swelling in the leg.

If you are presently taking anti-inflammatory medicine, please make sure that you discuss the adverse effects with your doctor.

Fennel (Foeniculum vulgare) is a plant species, belongings to Apiaceae (Umbelliferae), native to the Mediterranean.

Herbal medicine has been used in traditional and herbal medicine as a warming, carminative, antispasmodic, antidepressant agent and to stimulate the appetite, ease indigestion, soothe coughing, reduce intestinal spasms, regulate the menstrual cycle, and relieve PMS,...

According to a study of BALB/C mice were intraperitoneally administered anethole (62.5, 125, 250, or 500 mg/kg) 1 h before intratracheal treatment with LPS (1.5 mg/kg) and sacrificed after 4 h, researchers found that in the anethole treatment group exerts a significant increase of numbers of mobile white blood cells macrophages and neutrophils, the first immune response to the infectious site.

Furthermore, the application of LPS also induced the production of inflammatory mediators such as matrix metalloproteinase 9 (MMP-9), tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) all systematic inflammatory cytokines in response to cellular damage and tissue injury.

Moreover, LPS also exhibited the overproduction of nitric oxide (NO) observed in bronchoalveolar lavage fluid in facilitated cellular cytotoxicity.

Compared nontreatment group, the injection anethole group demonstrated a strong anti-inflammatory effect through decreased activities of proinflammatory factors without affecting the immune activity in protecting against infection.

Additional analysis also found that the treatment group reduced acute lung injury caused by the injection of LPS through its antioxidants and induced the production of host natural antioxidants in the body tissue.

The inhibited LPS results were attributed to the anethole effect in suppressing the activation of NF-κB in signaling the production of inflammatory cytokines by blocking the initiation of IκB-α degradation by inhibited TRAF2 in the induction of inflammation through TNF receptor superfamily members and NIK, a key signaling molecule of the noncanonical NF-κB.

Interestingly, further investigation of anethole as an inhibitor of TNF-induced NF-kB activation (an early response) as monitored by electrophoretic mobility shift assay, by suppression of IkBa phosphorylation and NF-kB reporter gene, researchers also indicated that

* Anethole also suppressed TNF-induced activation of the transcription factor AP-1 in regulated gene expression in response to a variety of stimuli, including LPS application.

*c-Jun N-terminal kinases (JNKs) with function in response to stress stimuli, such as cytokines,
And

* MAPKkinase in signals inflammatory response by transmitting information from a receptor on the surface of the cell to the DNA in the nucleus of the cell.

* Application of anethole also suppressed TNF-induced both lipid peroxidation and ROS generation, through the stimulated production of natural antioxidants in the host and itself, resulting in balancing the ratio of antioxidant and ROS expression.

The illustration and differentiation were elite by the experiment in the application of TNF (100 pM) with the cells pretreated for 240 and 120 min with anethole, NF-kB activation was almost completely inhibited.

However, the inhibition decreased gradually with decreased pre-incubation time.

Taken together, fennel and its bioactive anethole may be considered as a functional food in reduced risk and treatment in acute and chronic inflammatory-related diseases through the properties like a pro-antioxidant and anti-inflammatory agent.
