#Herbal #Anise Inhibits the #Nephrotoxicity Caused by Convention Medicnes Gentamicin (GM) Through Its Antioxidant Property

Anise may have a potential and substantial antioxidant effect in balancing the ratio of antioxidants and free radicals in the body, some scientists suggested.

Free radicals are unstable and uncharged molecules that are highly reactive by having an unpaired valence electron.

Some researchers suggested that due to the nature of free radicals' high reactivity, free radicals have the potential to be extremely harmful contaminants and extremely powerful chemical tools for the treatment of diseases through inducing cytotoxicity.

Free radicals exert their powerful reactivity through a chain reaction. Without inhibiting the unpaired electrons by antioxidants, the chain reaction can go on forever.

Radical chain reactions are processed in three distinct phases: initiation, propagation, and termination.

Contrastly, an antioxidant is a molecule with the function to inhibit oxidation, especially in counteracting oxidative stress caused by the overproduction of free radicals.

Researchers suggested that antioxidants found in the plant not only exert activity in the inhibition of free radicals but also stimulate the antioxidant enzymes in the host body.

Interestingly, in some cancer cells in vitro, the application of antioxidants found in the plant such as green tea not only protect the healthy cells of the host but also increase the function of free radicals in killing cancer cell through cytotoxic activity.

One theory suggested if we can maintain a healthy ratio of antioxidants and free radicals in the body, we may live a life of diseases free.

In the study of the effect of EtOAc soluble extract (PBet) isolated from Anise researchers found that at the dose of PBet (0.5-2mg/mL), Anise extract demonstrates a protective effect of Bel-7404 cells from free radical Hydrogen peroxide H(2)O(2) and prevent free radical in inducing cell damage through scavenging of intracellular ROS.


Further analysis also suggested that bioactive compounds such as β-sitosterol in the EtOAc soluble extract (PBet) improved the antioxidant status of the extract and stimulated the production of the host natural antioxidant profile in the inhibition of lipid peroxidation.

At a dose of 0,03 - 12g/kg BW, the extract expressed no acute toxic effect observed by the bone marrow polychromatic erythrocyte micronucleus test in determining the cytogenetic damage.


Dr. Lu J, the lead author, after taking into account other co-founders suggested that "PBet could be considered as a safe functional food with antioxidant activities".


Further evaluation of the expression of antioxidants in the inhibited free radical reactive activity in the development of degenerative diseases, researchers at the joint study led by the West Bengal State University, compared the antioxidant capacities of extracts of some commonly used in Indian spices (anise, cardamom, Ceylon cinnamon, and clove) along with their purified components (anethole, eucalyptol, cinnamaldehyde, and eugenol, respectively) indicated that anethole isolated from Anise displays a comparative antioxidant capacity in free radical scavenging activity against some ROS species.


Moreover, Anise spices as a whole are more potent antioxidants than their purified active components, and at higher concentrations, purified components are more toxic than their respective crude extract.

The above illustration was supported by the study in the analysis of the antioxidant and protective effects of Pimpinella anisum (P. anisum) against gentamicin (GM) in the induction of drug
nephrotoxicity.


A total of 40 male Wistar rats divided into four groups: control and sham administrated normal saline orally and intraperitoneally (i.p.), gentamicin (GM) group received 100 mg/kg bw/day i.p., GM and ethanolic extract of P. anisum with an oral dose of 300 mg/kg bw/day for 8 days, researchers suggested that application of Anise reduces kidney and liver toxicity and inhibits overexpression of reactive oxygen species in initiating chain reaction in oxidative stress the facilitation, and normalizes the levels of excretion of sodium and potassium induced by free radical expression, caused by an injection of gentamicin (GM)
All inhibitions were observed through the expression of levels of creatinine, associated with kidney damage and blood urea nitrogen (BUN), an indication of liver toxicity, and Malondialdehyde MDA, an oxidative stress marker, and the absolute excretion of sodium and potassium.


Interestingly, compared to the control, the application of Anise at 300 mg/kg bw/day significantly reduced the plasma concentrations of renal function markers (serum creatinine) in the group receiving GM (P < 0.05) and protected them against GM in inducing tubule damage.


Dr. Changizi-Ashtiyani S, the lead author suggested the "potential therapeutic impact of P. anisum to attenuate GM-induced nephrotoxicity. Therefore, the simultaneous use of the ethanolic extract of P. anisum during GM administration is recommended to reduce its nephrotoxicity effects".

Taken together, the findings suggested that Anise may be considered a functional herb in the improved antioxidant status against ROS and drug-induced toxicity through its bioactive compounds by stimulating the production of natural antioxidants in the host.

However, intake of Anise extract in high concentration should be taken with exceptional care as liver acute toxicity has been reported by some medical literature.

Coffee intake may have a positive implication in reduced risk of early onset of Alzheimer's disease, a respectable joint study suggested.

The moderate-to-severe premenstrual syndrome in Chinese women studies also showed significantly greater improvement of symptoms with intake of vitex agnus castus (VAC, BNO 1095)group than placebo, including symptoms of insomnia.

Additionally, according to Dr. Döll M, the extract of chaste tree berry decreased the levels of prolactin serum during the first 3 weeks of the cycle of the premenstrual period, inducing stress involved expression of symptoms of PMS.

Eggplant may have a potential and substantial effect in reduced risk and treatment of hypercholesterolemia and hypertension, some scientists suggested.
Eggplant is a species of S. melongena with deep purple color, belongings to the family Solanaceae and native to India, cultivated in southern and eastern Asia since prehistory.

1. Hypercholesterolemia
Hypercholesterolemia is a common genetic disorder of cholesterol metabolism characterized by abnormal high low-density lipoprotein cholesterol (LDL-C) levels, one of the leading causes of cardiovascular disease.

Hyperlipidemia is a chronic condition of abnormally high levels of blood cholesterol.

According to statistics, approximately 78 million U.S. adults (nearly 37%) had abnormally high blood cholesterol levels in the US in 2012.

In the investigation of the blood lipid effect of four eggplant varieties on lipid indices in 108 adult albino rats for 4 weeks (30 days), assigned to 2 major groups (fed with 10% and 20% eggplant supplementation) with four sub-groups (Groups A-D) and remainder rat assigned to control group fed with commercial feed, at the end of the experiment, researchers found that all groups fed with eggplant supplement regardless to doses exhibit a significant lower blood cholesterol in compared to control group.

Further analysis indicated that administration of diet supplemented at doses of 10% and 20% of S. aetheopicum and S. gilo in the rat group showed a highest of 9.92 and 9.53 and lowest 7.65 and 5.95g weight gain respectively.

Levels of cholesterol concentration of (60.76±1.7mg/dl) also were found significantly lower in the 20% S. gilo supplemented diet rat group compared to control and levels of HDL concentration significantly increased in rats placed on 10% and 20% S. macrocarpon (oval) and S. gilo supplemented diet respectively relative to the control group.

The results finding indicated that all 4 varieties of eggplant exerted s strong effect in displayed weight reduction and improved hypolipidemic levels.

Although Solanum gilo and S. macrocarpon (oval) displayed higher hypocholesterolemia than the other 2 groups, overall, all groups increased HDL and reduced weight gain in tested subjects.

Dr. OSSAMULU, the lead author, after taking into account other con founders said, "(The findings suggested that eggplant) can be employed in the management of overweight and several cardiovascular diseases".

In the study to test the efficacy of dried powdered fruits of eggplant (Solanum melongena) used in traditional medicine in Brazil for the treatment of human hyperlipidemia, researchers at the joint study led by the Instituto de Endocrinologia de Maringá conducted a double-blind placebo-controlled study to test the effectiveness of oral Solanum melongena (SM) in a total of 41 hyperlipidemic volunteers allocated to active treatment (n= 21, received two capsules containing SM (450 mg) or placebo (n= 20, 450 mg) twice a day for 3 months, indicated that there was no significant difference of levels of serum triglycerides, HDL-c, VLDL-c, AST, ALT, gGT, glucose, and body mass index, in the group treated with SM in compared to placebo.

And both groups expressed the same levels of decreased of levels of serum total cholesterol, LDL-c, and LDL-c/HDL-c.

The result of the findings prompted Dr. Silvard Fruits), to require further clinical trials before being recommended to treat hyperlipidemia".

In contrast to the above study, the oral application of infusion of a powdered preparation of the eggplant fruit of 38 hypercholesterolemic human volunteers, receiving either S. melongena infusion (N = 19) or placebo (N = 19) participated in two clinical experiments, exhibited a strong effect in significantly reduced the blood levels of total and LDL cholesterol and of apolipoprotein B in compared to control, according to the Intraindividual analysis.

However, after taking into account dietary orientation, researchers suggested that no intra- or intergroup differences were seen for any of the parameters analyzed.

Dr. Guimarães PR, the lead scientist at the Instituto de Ciências Biológicas said, " S. melongena infusion had a modest and transitory effect, which was not different from that obtained with standard orientation for dyslipidemia patients (diet and physical activities)".


2, Hypertension

Interestingly, in the study to evaluate the cardiovascular action of Solanum melongena extract (SME) in vivo and in vitro, researchers found that application of SME displays a significant and produced dose-dependent effect in reduced hypertensive responses in normotensive albino rats through a modulated expression of β-adrenoceptor on cardiac and smooth muscle tissues in precipitated high blood pressure.

More precisely, researchers suggested that SME modulated high blood pressure through induction of vaso-relaxation and negative inotropy and mediation of the β-adrenergic and acetylcholine receptors in promoted vasodilation of the resistant vessels and a reduction in heart activity, respectively.

Conclusively, compared to Captopril, a hypertensive medicine, researchers also indicated that possibly, the hypotensive effect of SME could be attributed to SME influence on the activity of the renin-angiotensin system, which is a hormone system with function in regulated blood pressure and fluid balance.

In fact, the effect of eggplant in the regulated function of diuresis in the renin-angiotensin system may have resulted in an ameliorated risk of high blood pressure as diuretics were found to associate to the function of kidneys in removal of more sodium and water from the body, triggering relaxation of the blood vessel walls, thereby lowering blood pressure.

Taking together, although there is no universal confirmation, eggplant may have a potential and therapeutic effect in reduced risk and treatment of hyperlipidemia and Hypertension,

#GreenTea Inhibits #Fibromyalgia Through Its Anti #Oxidativestress Activity, Researchers Find

Green tea may have a therapeutic and positive effect in reducing symptoms and treatment fibromyalgia caused by oxidative stress, some scientists suggested.

Oxidative stress is a condition of imbalance of the ratio of antioxidants and free radicals in the body. In other words, the overproduction of free radicals accompanied by decreased levels of antioxidants may cause cytotoxicity in the induction of oxidative stress.

Some researchers suggested long-term suppression by oxidative stress can activate the expression of over 500 different genes, including those for growth factors, inflammatory cytokines, chemokines, cell cycle regulatory molecules,...


Truly, oxidative stress-activated inflammatory pathways can also lead to the transformation of a normal cell to a cancer cell, protecting tumor cell survival, proliferation, chemoresistance, radioresistance, invasion, and angiogenesis.


Dr. Khansari N, the lead scientist in the study ' Chronic inflammation and oxidative stress as a major cause of age-related diseases and cancer" said, " Overproduced free radicals react with cell membrane fatty acids and proteins impairing their function permanently. In addition, free radicals can lead to mutation and DNA damage that can be a predisposing factor for cancer and age-related disorders" and "chronic inflammation could have a serious role in a wide variety of age-related diseases including diabetes, cardiovascular and autoimmune diseases".

Green tea, a precious drink processes a number of health benefits known to almost everyone in Asia and the Western world.

Fibromyalgia (FM) is a chronic condition characterized by muscle and soft tissue pain affecting over 10 million (3.6% of the population) people in the US alone.

According to the study in the review of literature on the frequency and pattern of complementary and alternative medicine (CAM) of 289(95%) patients who completed the survey (263 women and 26 men) between February 2003 and July 2003, green tea was recommended at 24% for treatment of fibromyalgia (FM).

Some evidence in the medical literature suggested that the overproduction of ROS in the induction of oxidative stress may have a strong implication in the pathophysiology of FM.

In the investigation of the effect of green tea epigallocatechin-3-gallate (EGCG) in the protection of oxidative stress to cardiac cells cultured in the conditions of control, 400 μM H2O2 exposure for 30 min with and/or without 10 to 20 μM EGCG pre-treatment, researchers at the National Tsing Hua University found that treatment of green tea in H(2)O(2) group demonstrated a significant reduction of levels of reactive oxygen species through its antioxidant and its ability to induce production of natural antioxidant present in the cardiac cells, compared to overexpression of ROS in H2O2 treatment group without injection of EGCG.

Also, the application of green tea reduced levels of cytosolic Ca2+ overload in the H(2)O(2) group in induced oxidative stress in experiment cell apoptosis through the improved antioxidant protein.

Interestingly, EGCG ameliorated H(2)O(2) expression in the increased glycolytic protein in response to the degree of oxidative stress in the culture cells and α-enolase, a key glycolytic enzyme on the surface of several cell types in the contribution of impaired glycolytic activity through oxidative and nitrative stress.

Additionally, green tea EGCG also decreased levels of peroxiredoxin-4, an antioxidant with a function in the protection of cells against oxidative stress by detoxifying peroxides and mitochondrial proteins with a function of redox reactions of oxidative phosphorylation.

After taking into account confounders, researchers suggested that ingestion of green tea EGCG inhibited the damage of H(2)O(2) group through inhibition of the downstream signaling for Akt in expression glucose oxidation and cell apoptosis in cellular processes and loss of phosphorylation of GSK-3β and cyclin D1 depletion in oxidative stress facilitation.

These results postulated that green tea EGCG exerted a similar inhibited effect of those of GSK-3β inhibitor (SB 216763) in significantly improved H(2)O(2)-induced suppression on cell viability, phosphorylation of pAkt (S473) and pGSK-3β (S9), and level of cyclin D1 in cells.

The above differentiation was supported by the study conducted by the Universidad de Sevilla, in evaluating some evidence of green tea in reduced oxidative stress in the facilitated pathophysiology of FM in initiating signs and symptoms of muscular alteration and mitochondrial dysfunction.

Truly, in the primary rat model, administration of green tea EGCG in different concentrations for 24 h before being exposed to hydrogen peroxide (H(2)O(2)) for 2 h to induce oxidative stress, researchers found that pretreatment with 10, 25, and 50 µM EGCG significantly inhibited the expression of H2O2 in a reduced substantial decrease in cell viability.

Further analysis also showed that green EGCG application at a dose of 50 µM ameliorated the proportion of propidium iodide (PI)-positive cells increased in cultures caused by H(2)O(2) injection, thus reducing H(2)O(2) in the induction of cell death.

Taken together, green tea and its bioactive polyphenols EGCG may be considered a functional food for the treatment of fibromyalgia caused by overexpression of oxidative stress. However, the intake of green tea supplements should be taken with extreme care as acute liver toxicity has been reported in a number of medical literature.

#Lentils Inhibit Paraneters Associated with the Risk of #MetabolicSyndrome, Scientists Show



Daily and regular intake of lentils may have a potential and therapeutic effect in reduced risk and treatment of metabolic syndrome, some researchers suggested.
Lentil, a type of pulse and tiny legume that can be cooked quickly and is rich in protein and fiber is a bushy annual plant of genus Lens pf the Fabaceae family, native to the Middle East.

Metabolic syndrome is a set of conditions characterized by increased blood pressure, high blood sugar, excess body fat around the waist, and abnormal cholesterol or triglyceride levels which are considered risk factors for cardiovascular disease and diabetes.

Investigation of lentils' effect on the risk of metabolic syndrome was depending on a number of mechanisms through several aspects.

The testing of low-GLoad foods effect in metabolic syndrome occurrence with 131 male Nile rats aged 3 weeks to 15 months randomly assigned to 15 dietary exposures of varied Glycemic Index (GI, 36-88), GLoad (102-305/2000 kcal), and cumulative GLoad (Cum GLoad=days×GLoad, 181-537g total glucose), indicated that lentil diets with low GLoads (102, 202) displayed a significant effect in reduced onset and treatment of metabolic syndrome in compared to other food with higher glycemic index.

More specifically, the benefit from lentils exceeded dietary factors of combined macronutrient composition (%Energy from carbohydrate:fat: protein, between 70:10:20 to 40:40:20), total fiber (0-24%), or dietary caloric density (2.9-4.7 kcal/g) found in the legume.

Additionally, the experiment also found that the lentil diet even reversed the progression in metabolic syndrome and diabetes expression in the tested Nile rats.

According to the randomized assays, intake of lentils over 15 dietary exposures decreased factors of insulin resistance, and lower blood sugars, particularly in fasting glucose levels as well as improving levels of blood lipid and the ratio of cholesterol= high-density choleaterol#/ total cholesterol#.

Inspection of the total food intake of the tested rat also demonstrated a strong correlation between the cumulative GLoad (Cum GLoad=days×GLoad, 181-537g total glucose) and parameters of MetS and T2DM.

More profoundly, according to the University of Toronto, in the experiment to investigate the effects of frequent consumption (five cups/week over 8 weeks) of pulses (yellow peas, chickpeas, navy beans and lentils), compared with counselling to reduce energy intake by 2093 kJ/d (500 kcal/d), on risk factors of the MetSyn of two groups (nineteen and twenty-one subjects, respectively) of overweight or obese (mean BMI 32·8 kg/m2) adults, measurement of body weight, waist circumference, blood pressure, fasting blood parameters and 24 h food intakes at weeks 1, 4 and 8 and blood glucose, insulin, C-peptide, glucagon-like peptide-1 (GLP-1) and ghrelin were measured after a 75 g oral glucose load at weeks 1 and 8 indicated that frequent consumption (five cups/week over 8 weeks) of pulses displayed a significant and positive effect in reduced energy intake in ubction of weight loss, waist circumference, systolic blood pressure, glycosylated Hb (HbA1c), and glucose AUC and homeostasis model of insulin resistance (HOMA-IR) following the glucose load (P < 0·05) similar to those in energy group.

Glycosylated Hb (HbA1c), a measurement of three-month average plasma glucose concentration is used to determine the risk of diabetes in suspected subjects. There is a suggestion of a lower HbA1c level by 1% associated with a lower 19% risk of diabetes onset.

The homeostatic model is a test with an aim to assess the function of β-cell and insulin resistance (IR) from basal (fasting) glucose and insulin or C-peptide, a proinsulin molecule concentrations.

Further analysis also found that frequent consumption (five cups/week over 8 weeks) of pulses (yellow peas, chickpeas, navy beans, and lentils also increased HDL levels and fasting C-peptide and insulin Area under the curve (AUC) responses in compared to energy group. In deed, the influence of the above parameters was associated linearly and directly with types of diet intake.

More precisely, the pulse treatment group showed HDL and C-peptide increased by 4·5 and 12·3 %, respectively, compared to a decrease of 0·8 and 7·6 %, respectively, in the energy-restricted group.
Insulin AUC decreased in both females and males on the energy-restricted diet by 24·2 and 4·8 %, respectively, but on the pulse diet, it decreased by 13·9 % in females and increased by 27·3 % in males (P < 0·05).

Taken together, there is no doubt that frequent intake of lentils or legume products may have a substantial and direct influence on reduced risk, progression, and treatment of metabolic syndrome.
