#Phytochemical 3,3'-#Diindolylmethane Inhibits Diseases Associated with Overexpression of #OxidativeStress, Researchers Find

Scientists may have found a bioactive ingredient for the treatment of diseases associated with oxidative stress with no side effects, according to studies.

Oxidative stress is a condition caused either by overexpression of free radicals or depletion of antioxidant enzymes produced by the host.

In other words, the condition of oxidative stress occurs only under an imbalance of the ratio of free radicals and antioxidants.

Antioxidants can be either produced by the body or injected into the body from outside sources such as the consumption of antioxidant-rich fruits and vegetables.

Free radicals are everywhere, including the air we breathe, the food we eat and the body daily functioning such as cell metabolism.

In other words, any chemical compound which has an oxygen molecule can become a free radical after entering our body.

Most healthy individuals can produce enough amounts of antioxidants endogenously to counter the bombards of free radicals. However, people with a weakened immune system such as the elderly, and immature immune systems such as babies may need the injection of antioxidants from outside sources.

More precisely, people with a weakened immune system do not produce enough antioxidant enzymes to inhibit the chain reaction of free radicals which occurs daily in the body due to the unstable nature of the molecules.
Believe it or not, our body needs to limit levels of some free radicals to function better, including lower blood pressure, and relax blood vessels to move blood, such as nitric oxide NO.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, kale, etc.

With an aim to find the natural ingredient for the treatment of diseases associated with oxidative stress, researchers investigated the effect of indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) in type 2 diabetes mellitus (T2DM).

The study included selected genetically modified mice (C57BL/6J mice) similar to that of metabolic abnormalities of the human disease after the administration of a high-fat diet (HFD).


Including indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) into the diet showed positively modulated glucose, insulin, and Hb and HbA1c levels in HFD mice against the risk of diabetes complications.

Compared to the HFD group, injection of I3C and DIM protected the infected rat against the formation of lipid peroxidation observed by levels of thiobarbituric acid reactive substances (TBARS), lipid hydroperoxides (LOOH), conjugated dienes (CD).


Furthermore, levels of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were also increased, an indication of the balance of the ratio of antioxidants and free radicals.

Interestingly, I3C and DIM also improved the free radical scavengers such as vitamin C, and vitamin E, and improved the reduced glutathione (GSH) levels in treated mice.

Based on the findings, researchers summarized, "From the study, it was clear that the antioxidant-scavenging action was accelerated in mice treated with DIM than the I3C treatment group which was comparable with the standard drug metformin".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and combined with primary medicine for the treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Scientists may have found a natural whole food that processes a significant activity in reducing the risk and treatment of acute coughing in children with no side effects, some studies suggested.
Coughing can be a natural and reflex reaction to expel phlegm from the airway or a result of viral and bacterial infection of the upper airway.

The study includes the search of available randomized controlled trials (RCTs) comparing honey, given alone, or in combination with antibiotics, versus nothing, placebo, or other over-the-counter (OTC) cough medications to participants aged from two to 18 years for acute cough in ambulatory settings.

Out of hundreds of studies reviewed by the researchers, only 2 studies were selected for the review.

Compared to dextromethorphan, diphenhydramine, and 'no treatment' on symptomatic relief of cough using the 7-point Likert scale. honey was a much better medicine than no treatment in reducing the frequency of cough and exerted similar or better effects to those of counter medicine.

However, mild adverse events were found in seven children (9.3%) from the honey group and two (2.7%) from the dextromethorphan group.

After taking into account co and confounders, Dr. Oduwole O. the lead scientist said, "There was no significant difference between honey versus dextromethorphan or honey versus diphenhydramine".

Furthermore, in the concerns of the side effects of honey and over-the-counter medicine, researchers examine the effects of two kinds of Iranian honey with diphenhydramine (DPH) on nocturnal pediatric coughs and the sleep quality of children and their parents.

A total of 87 patients included in the study were randomly assigned to one of three treatment groups: Group 1, Honey type 1 (Kimia Company, Iran) (n = 42), Group 2, Honey type 2 (Shahde-Golha, Iran) (n = 25), and Group 3, DPH (n = 20).


Each group received double doses of the respective treatments on two successive nights, researchers indicated that
* Honey type 1 was superior to DPH in improving all aspects of coughs, except the frequency,

* Honey type 2 was more effective than DPH in improving all aspects of coughs, except the sleep quality of the child.

* There was no significant difference between Honey type 1 and 2 in any aspects of cough relief.

After examing the standard validated questionnaire completed by the parents before and after treatment.

Dr. Ayazi P, the lead author wrote in the final report, "Honey may provide better cough relief than DPH in children and improve the sleep quality of children and their parents".

By comparing the above studies, there is no doubt that honey may be used for the treatment of acute cough in children.

Free radical scavengers are antioxidants which process a function to protect the body against the oxidative stress by inhibiting free radical oxidation before they can facilitate a chain reaction or by stopping the chain reaction before they can cause damage to the body.

Free radicals are highly reactive molecules. Internally, they are produced by our body cell metabolism. Externally, free radical can enter our body from the environment sources, such as the air we breathe, the food we eat and the water we drink.

More precisely, any chemicals compounds or molecules which process oxygen atom can be free radicals after chemical interaction.


Believe it or not, free radicals in moderate amounts has benefits the body functioning, such as NO for vascular relaxation.

However, without interfering with the antioxidant, overexpression of free radicals has been found epidemiologically to damage the lipid and protein and induce the alternation of healthy cell DNA, leading to chronic diseases, including arthritis, cardiovascular disease, and diabetes.

Oxidative stress is a result of the imbalanced ratio of the levels of free radicals and antioxidant enzymes in the body.

Out of all damages caused by the overexpression of free radicals, some researchers suggested that lipid peroxidation induced by free radicals has a strong implication in the pathogenesis of various disorders and diseases.

And, "Indeed, a number of preclinical and clinical studies suggest the involvement of lipid peroxidation products in numerous pathological conditions such as inflammation, atherosclerosis, diabetes, aging, neurodegenerative diseases, and cancer".

In other words, an antioxidant with free radical scavenging activity may protect the body against the damage of various cellular components, including proteins, lipids, and DNA in the initiation of diseases.

Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.

With an aim to find a potential compound for the treatment of diseases involved in oxidative stress, researchers examined the free radical-scavenging activity of indicaxanthin.

According to the tested analysis on the reaction of the phytochemical indicaxanthin with lipoperoxyl radicals generated in methyl linoleate methanol solution by 2,2'-azobis(2,4-dimethylvaleronitrile), and in aqueous soybean phosphatidylcholine unilamellar liposomes by 2,2'-azobis(2-amidinopropane) hydrochloride,

* Indicaxanthin incorporated in liposomes prevented lipid oxidation.

* The free radical scavenging property was found to be concentration-dependent, but not linearly related to the phytochemical concentration.

* In liposome, indicaxanthin showed a significant delay against oxidation, particularly in that of the lower concentration.

* However, in the corporation with alpha-tocopherol, Indicaxanthin exhibited cooperative antioxidant effects and reciprocal protective interactions.

Taken altogether, indicaxanthin processes strong free radical scavenging activity, pending to the confirmation of the larger sample size and multicenter human study.

At the 4th stage of cancer, malignant cells may metastasize to other organs a distance away from the original site.

A total of 24 male Sprague Dawley rats aged 5 weeks are included in the study.
Rats were divided into 4 groups: (G1) untreated group not induced with AOM, (G2) untreated group induced with AOM, (G3) treated group induced with AOM, and (G4) treated group not induced with AOM.
Injection of AOM (15 mg/kg) was via intraperitoneal route once a week for two subsequent weeks to induce colorectal cancer.
The treatment groups were given oral administration of KH (1183 mg/kg body weight) twice daily for 8 weeks.

AOM injected group showed a significant increase in the total number of aberrant crypt foci (ACF) and aberrant crypts (AC) and crypt multiplicity and damage to liver and kidney

After treatment of KH, AOM group displayed a reduction of crypts and chemotoxicity by restoring the liver and kidney function observed by blood profile.

Furthermore, KH was not toxically identified by the KH-treated group not induced with AOM.

These results suggested that KH may have a potential effect on inhibiting the toxicity caused by chemotherapy and improving the chemo drug's activity without inducing any side effects.


Moreover, on finding a natural whole food to overcome 5-fluorouracil (5-FU) therapeutic resistance and adverse toxicity in colorectal cancer, researchers looked into Manuka honey (MH), a rich source of bioactive compounds to examine its anticancer effects on human colon cancer HCT-116 and LoVo cells.

Compared to 5-FU alone, MH synergistically enhanced the chemotherapeutic effects of 5-FU, by reducing cell proliferation through the suppression of enzymes and proteins associated with the function in stimulating cancer cell multicity and activation of pathways associated to cell death programming.

Combined with chemo drug 5-FU, MH promoted the expression of antibody production and production of ROS in the induction of cytotoxicity activity against cancer cells without causing harm to the nearby cells and tissue.

This result was observed by the reducing antioxidants produced by the cancer cells and increasing oxidative stress in the tumor.

Additionally, MH also induced cancer cell apoptosis through cell cycle arrest.

The findings clearly indicated MH could be a useful preventive or adjuvant agent in the treatment of colorectal cancer with 5-FU by inhibiting the toxicity and cancer cell resistance.

Taken altogether, KH and MH can be considered functional foods for the prevention and adjunct therapy combined with the primary medicine for the treatment of cancer without inducing chemotoxicity.

The liver is the largest internal organ that not only plays an essential role in cleansing the blood coming from the digestive system before passing them to other parts of the body but secrets cholesterol to aid digestive function, produce steroid hormones and vitamin D, and build strong cell walls.

Steatosis is a condition caused by abnormal retention of lipids within a cell or organ. Hepatic steatosis is a medical condition caused by the accumulation of fat in the liver.

In other words, if the fat of your liver is more than 5% of your liver weight, you are considered to have hepatic steatosis. Over time, untreated hepatic steatosis can lead to advanced forms of nonalcoholic fatty liver disease which have been found to induce negatively the function of the liver, particularly. in patients with obesity associated with alcohol intoxication and/or hepatic disorders.

There are several mechanisms involved in the accumulation of intrahepatic fat, including increased flux of fatty acids to the liver, increased de novo lipogenesis, and/or reduced clearance through β-oxidation or very-low-density lipoprotein secretion.

People who are obese, having the condition of insulin resistance or diabetes, dyslipidemia, hypertension, metabolic syndrome, rapid weight loss, use hepatotoxic medications total parenteral nutrition (TPN) are at an increased risk of hepatic steatosis.

Conventionally, there is no effective treatment for liver steatosis. However, treatment of liver steatosis in obese patients is focusing to reduce weight and symptoms and improve quality of life, totally depending on the severity of the disease.

Usually, weight loss through a combination of a healthy diet and exercise is also the first line of treatment. in patients with an advanced stage of liver fibrosis, (cirrhosis) liver transplantation may be an option.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a natural compound for the prevention of liver steatosis, researchers examined the alleviation effect of KBH-1, an herbal mixture of Saururus chinensis, Curcuma longa, and Polygala tenuifolia in obesity-induced hepatic steatosis.

The experiment included HepG2 cells, primary neuronal cells, and a high-fat diet (HFD)-induced obesity rat model in vitro and in vivo.

According to the tested assays, KBH-1 not only suppressed the lipid deposition in HepG2 cells exposed to free fatty acids (FFA) but also significantly inhibited major factors in lipogenesis associated with the formation of fat and promoted major factors in lipolysis involved in the breaking down of fats.

In the high fat-induced obesity model, KBH-1 improved hepatic steatosis by alleviating the effects on lipogenic genes and kinases similar to those in vitro.

Furthermore, KBH-1 significantly improved the leptin-mediated signals that regulate food intake, energy expenditure, and metabolism,.. impaired by obesity or FFA in the obesity model.


Moreover, according to the HPLC analysis, onji-saponin B and curcumin found in the formula have also potently suppressed the level of triglycerides.

In other words, KBH-1 activated the expression of AMP-activated protein kinase (AMPK) associated with the regulation of energy metabolism that stimulates glucose uptake and lipid oxidation to produce energy and suppressed the expression of PPARγ involved in hepatic steatosis... in obesity.


Based on the findings, researchers wrote, "These findings show the potential of KBH-1 as a functional food supplement or preventive agent in the treatment of obesity (.associated with hepatic steatosis)".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of hepatic steatosis in obese, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Green Tea and Its' Polyphenols (GTPs) May Process Anti-Melanoma and Non-Melanoma #SkinCancers Activities, According to Studies

Green tea, a precious drink processes a number of health benefits known to almost everyone in Asia and the Western world. However, as yin in nature herbal medicine or food, long-term injection of large amounts may obstruct the balance of yin-yang, inducing "yin excessive syndrome" or "yang vacuity syndrome" including weakened immunity and painful case of GERD,... according to traditional Chinese medicine's Yin-Yang theory.

Skin cancer is a medical condition characterized by irregular cell growth in the tissues of the skin. At the later stage, the cancerous cell may travel a distance away to invade other healthy tissues and organs.

According to statistic, approximately, 87,110 new cases of invasive melanoma are diagnosed in the U.S every year

The evaluation of the green tea effect in overexposure to environmental solar ultraviolet (UV) radiation (290-400 nm) which has been found to increase the risk of the development of melanoma and non-melanoma skin cancers in mice model showed that oral administration of green tea polyphenols (GTPs) in drinking water or the topical application of EGCG prevents UVB-induced skin tumor development in tested subjects.

Particularly, (-)-Epigallocatechin-3-gallate (EGCG), the major and most photoprotective polyphenolic component of green tea also exhibited a protective activity against UV in the induction of skin cancer, through induction of immunoregulatory cytokine interleukin (IL) 12 in differentiation of naive T cells into Th1 cells and acted as T cell-stimulating factor in stimulate the growth and function of T cells.

Moreover, interleukin (IL) 12 also demonstrated a significant effect as an antagonist against immunosuppression induced by solar/ultraviolet (UV) radiation by restoring immune responses and also stimulating DNA repair.

Further analysis also indicated that green tea (-)-Epigallocatechin-3-gallate (EGCG) also inhibited UV in the induction of skin tumors; angiogenetic effect in stimulated production of new blood vessels to
provide nutrients and oxygen for cancer cells proliferation and survival and initiated cytotoxic T lymphocyte cells in induced cancer and infectious and damaged cells apoptosis.


Interestingly, an additional study of green tea polyphenols (GTPs) in prevention of UVB irradiated (80 mJ/cm(2)) 3 times each week for 24 wk in induced skin cancer in mice divided into group consumed water and a group fed with water containing 2 g/L GTPs, showed that group treated with GTPs displayed a significant reduced UVB-induced tumor incidence by 35%, tumor multiplicity by 63%, and tumor growth by 55% in compared to the non-treatment group.

Group treated with GTPs+UVB also demonstrated a reduced expression of the matrix metalloproteinases (MMP)-2 and MMP-9, which are activator associated with progression from benign to advanced cancer and enhanced expression of specific endogenous tissue inhibitors of metalloproteinases (TIMPs), tissues inhibitor of MMP in skin tumors in compared to mice treated with UVB alone.

Furthermore, GTPs+UVB group also reduced expressions of CD31 and vascular endothelial growth factor, in the activated development of new blood vessel to nourish skin tumor cells proliferation and site expansion and inhibited expression of proliferating cell nuclear antigen, a key factor in DNA replication and cell cycle regulation in skin cancer in compared to UVB group.

More importantly, mice treated with the GTPs+UVB group also showed more cytotoxic CD8(+) T cells in the tumors compared to the UVB group by exhibiting greater activation of caspase-3 expression in induced apoptosis of the tumor cells.

The information findings suggested a potential and therapeutic effect of green tea and its bioactive polyphenols (GTPs) in attenuating the risk, progression, and treatment of melanoma and non-melanoma skin cancers.
