#Enzyme #Bromelain Exhibits Genotoxic Activity in #Cancer's Nucleic Acid in Vitros, According to Studies

Genotoxicity is a process of destruction of a cell's genetic material (DNA, RNA), leading to gene mutation.
In other words, genotoxins are mutagens, substances that have been found to induce cancer. The most common genotoxins are radiation and toxic chemicals.

Believe it or not, besides causing cancer, genotoxins are found to induce various types of genetic diseases, by the damage the germ cell.

Some researchers suggested that the damage of DNA may be controlled if the genotoxins are identified through the use of anti-antimutagens/anti-clastogens that suppress or inhibit the mutagenesis process.

The most common causes of genetic damage or alternation are reactive oxygen species (ROS), the unstable and genotoxic molecules that exist everywhere in the environment and inside the human body.

By regulating the levels of free radicals in the body through the increased antioxidant enzyme inside the host or from the dietary sources, oxidative stress induced by ROS may be controlled. Thereby, reducing the risk of gene damage.


Dr. Mohamed SAKS, the lead scientist wrote in an article published in the Global Journal of Pharmacy & Pharmaceutical Sciences, "There are mainly two mechanisms of anti-mutagenesis:
a. Desmutagenesis in which the factors on the mutagens are somehow inactivated,

b. Bio-anti mutagenesis, in which the factors act on the process of mutagenesis or by repairing the damaged DNA that results in the decreased frequency of DNA mutation [15]. a number of cells have a DNA repair system by which they try to control the DNA mutations naturally".

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With the aim to find a potential treatment for cancer, researchers examined the genotoxic effect of
pineapple extract on oral cancer cell line by DNA fragmentation.

According to the DNA fragmentation assay, bromelain showed significant genotoxic activity by invading the cancer nucleus and causing damage to the nucleic acid.

In the examination of 3 different doses of bromelain incubated with oral cancer cells, DNA fragmentation occurred in all three concentrations.

In other words, bromelain inhibited oral cancer cell progression through genotoxic properties.

Furthermore, bromelain also inhibited the expression of MUC1, a glycoprotein associated with tumor cells' invasive, metastatic, and chemo-resistant properties in breast and pancreatic cancer cells.

Moreover, bromelain used a combination with either cisplatin or 5-FU also enhanced conventional medicine efficacy.

Additionally, in malignant peritoneal mesothelioma (MPM) expressed MUC1, administration of bromelain also affected the viability of MPM cells adversely.

Taken altogether, bromelain may be considered a supplement for the prevention and treatment of cancer through its genotoxic activity, pending the confirmation of the larger sample size and multicenter human study.


Intake of bromelain in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Prostate cancer is a medical and chronic condition caused by cell growth irregularly in the prostate gland. Most cases of prostate cancer are slow-growing and enlarged prostate and prostate cancer may be detected during Physical (rectum) exams.

Most cases of prostate cancer begin in the cells on the surface of the inner lining of the prostate tissue before penetrating into deeper layers to induce colonization.

At the advanced stage, the prostate cancer cells can spread to distant parts of the body, leading to secondary metastasis.

Prostate cancer can be diagnosed by* Blood tests
* Digital Rectal Examination (DRE)
In DRE, your doctor inserts a lubricated, gloved finger into the rectum to feel the surface of the prostate gland to check for any abnormality of the prostate.

* Transrectal Ultrasound (TRUS)
Ultrasound allows your doctor to visualize and assess the size of your prostate and to check for any abnormality and surrounding area with an image taken from the test. In transrectal ultrasound, a probe is inserted into the rectum to emit ultrasonic impulses against the prostate.

* Prostate-specific antigen (PSA) test
The test is to measure the levels of prostate-specific antigen (PSA), a protein produced by the prostate. The high levels of PSA may be an indication of a higher risk of the presence of prostate cancer.

* Prostatic Acid Phosphatase (PAP) test
Prostatic acid phosphatase (PAP) is a test to measure the level of prostatic acid phosphatase, an enzyme produced by the prostate. The higher levels of PAP are an indication of the disease progression.

* Prostate biopsy
In a prostate biopsy, samples of the prostate are taken by a thin, tube-like instrument to examine under microscopy to review the stage of cancer.

According to the statistics, prostate cancer is the most common cancer among men particularly in older over 65, after skin cancer. in 2019, approximately 174,650 men in the US will be diagnosed with prostate cancer.

The average age of diagnosis is 66.

Carotenoids are a class of mainly yellow, orange, or red fat-soluble pigments, including lycopene and carotene, found abundantly in ripe tomatoes, pumpkins, carrots, corn, and daffodils.

In finding a potential compound for the prevention and treatment of prostate cancer, researchers examined the effect of carotenoid levels on the risk of high-grade prostate cancer.

The study included the measurement of plasma levels of carotenoids and genotyped 20 single nucleotide polymorphisms (SNP) in SOD1, SOD2, SOD3, XRCC1, and OGG1 in 559 men with non-metastatic prostate cancer undergoing radical prostatectomy.

According to the results from tested assays, the serum of circulated carotenoid levels was inversely associated with the risk of high-grade prostate cancer, with odds ratios (OR) of 0.34 compared to the highest versus lowest quartiles.

In chemical analysis, the relative odds ratio of α-carotene,β-carotene, lycopene, and total carotenoids.was 0.31, .55, 34, and 0.37 respectively.

In other words, all major phytochemicals found in carotenoids work synergistically to enhance the efficacy of carotenoids against the risk of high-grade prostate cancer.

Furthermore, the presence of the SNPs rs25489 in XRCC1, rs699473 in SOD3, and rs1052133 in OGG1 genotypes also was found to modify the activity of α-carotene, β-carotene, and lycopene, respectively.

Believe it or not, the high serum of lycopene was found among men with a Gleason score ≤ 3 + 4 and lower FGA (P = 0.04).


More precisely, the efficacy of carotenoids reduced the risk of prostate cancer, however, the risk of high-grade invasive prostate cancer are depending on the levels of lycopene.

Dr. Nordström T, the lead scientist said, "Higher lycopene level was associated with less genomic instability among men with the low-grade disease which is novel and supports the hypothesis that lycopene may inhibit the progression of prostate cancer early in its natural history".

Taken altogether, carotenoids may be considered a remedy for the prevention and treatment of prostate cancer, pending the confirmation of the larger sample size and multicenter human study.

The intestinal microbiome is the community of bacteria living in the gut. Under normal conditions, the ratio of good and bad microbiomes is balanced.

Believe it or not, our GI tract harbors one of the most complex and abundant ecosystems colonized by more than 100 trillion microorganisms, including Firmicutes and Bacteroidetes.

The composition and functions of the microbiome also have been found to influence by a number of factors including genetics, diet, age, and medical treatments such as antibiotics.

Epidemiological studies suggested that an unbalanced ratio of bacterias in the GI tract due to the aforementioned factors also induce changes in microbiome structure, which links to inflammatory, functional, and metabolic disorders such as IBD, IBS, and obesity.

Dr. Ana M Valdes, the lead scientist in the examined the health benefits associated with intestinal microbiome suggested that gut microbes have a strong effect on human health including immune, metabolic, and neurobehavioral traits.

According to the tested results, Bronze-enriched chow showed no macroscopic effect on the IBD symptoms after 2 weeks of administration. However, the injection of bronze tomato exerted a significant effect on the production of anti-inflammatory cytokines and reduced levels of pro-inflammatory production in the same periods in ulcerative colitis mice.

Furthermore, the analysis of the microbial composition suggested that the Bronze enriched diet demonstrated a strong effect on the microbial composition of Winnie and control mice by increasing levels of good bacteria in the gut.

According to the chemical analysis, the effect of bronze tomatoes in improving the ratio of intestinal microbiome composition was attributed to the polyphenol-enriched diets in creating unfavorable conditions for bad bacterial species.

Another study conducted by the University of Arkansas at Little Rock examined the effect of tomato fruits derived from plants exposed to multi-walled carbon nanotubes (CNTs) on gastrointestinal epithelial barrier integrity and their impact on the human commensal intestinal microbiota, in vitro cell culture and batch human fecal suspension models also suggested that
* At 0.001 μg ml-1 concentration, CNT-containing fruits showed insignificantly activity on the model T-84 human intestinal epithelial cells.

* However, at 10 μg ml-1 concentration, CNTs showed a strong effect in penetrating the cell membrane and changing the gene expression profile of exposed tested cells.

The liver is the largest internal organ that not only filters the blood coming from the intestinal system but also protects our body against chemical and drug toxicity.
Additionally, the liver also produces cholesterol that plays an essential role to aid digestion, producing vitamin D and steroid hormones, and building strong cell membranes.

Believe it or not, the liver also produces antioxidant enzymes that inhibit free radical oxidation and interact with the hormone insulin to store glucose in the form of glycogen.

Hepatic steatosis is a condition caused by the accumulation of fat in the liver. Liver steatosis can be classified into 2 types

* Alcoholic liver steatosis is caused by over-consumption of alcohol over time.

* Non-Alcohol Steatosis (NASH) is a condition of accumulation of excess fat in the liver in people who drink little or no alcohol. ... Non-Alcohol Steatosis is considered a major cause of liver scarring and cirrhosis.

The most common symptoms of liver steatosis are jaundice, abdominal swelling (ascites), enlarged blood vessels just beneath the skin's surface, enlarged breasts in men, enlarged spleen, and red palms.

If you have some of the aforementioned symptoms, please check with your doctor to rule out the possibility.

Conventionally, there is no effective treatment for liver steatosis. Weight loss and weight management through a combination of a healthy diet and exercise are the most recommended.

Some researchers suggested that the rise of liver steatosis in the US may be correlated to the promotion of a high-fat diet over the past few decades.

Dr. Arisqueta L, the lead scientist wrote, "High-fat diet (HFD) feeding or leptin-deficient mice are extensively used as models resembling features of human nonalcoholic fatty liver disease (NAFLD)".

And, "The concurrence of experimental factors as fat content and source or total caloric intake leads to prominent differences in the development of the hepatic steatosis and related disturbances".

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, chard, etc.

On finding a natural pigment for the prevention and treatment of liver steatosis, researchers examined whether betanin (from natural pigments, a member of betacyanins) had anti-oxidant properties against paraquat-induced liver injury in Sprague-Dawley rats.

Paraquat was injected intraperitoneally into rats to induce liver toxicity and randomly assigned to 4 groups: a control group, a paraquat group, and two groups received betanin at doses of 25 and 100mg/kg/day three days before and two days after administered paraquat.

Administration of betanin inhibited the elevated serum aspartate aminotransferase and alanine aminotransferase levels of the paraquat rats.

Betanin treated group also enhanced the production of antioxidant enzymes that counter the overexpression of free radicals-induced oxidative stress.

Furthermore, the betanin-treated group also increased the expression of an enzyme associated with detoxification.

Moreover, liver mitochondrial damage caused by paraquat injection also was reduced by the administration of betanin.

In other words, all parameters associated with the onset of liver damage that causes steatosis to develop were decreased substantially in the animals treated with betanin than in those who received paraquat alone.

Based on the results, researchers after taking other factors into account wrote, "Betanin had a protective effect against paraquat-induced liver damage in rats. The mechanism of the protection appears to be the inhibition of CYP 3A2 expression and protection of mitochondria".

Taken altogether, betacyanins processed abundantly bioactive ingredient betanin may be considered a remedy for the prevention and treatment of liver steatosis, pending the confirmation of the larger sample size and multicenter human study.

Intake of betacyanins in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

#GreenTea Improves the Gastrointestinal Phases of Digestion Against High Fasting Blood Sugar and Cholesterol Levels in Vivos

Green tea may have a therapeutic and positive effect in enhanced gastrointestinal function in food absorption, some scientists suggested.

According to Agriculture and Agri-Food Canada, native catechins found in green tea were not absorbed by the digestive system after intake. The polyphenol accumulated in the intestinal lumen inside the space of a tubular structure where they exhibited significant activity in ameliorated cellular glucose and cholesterol uptake, thus reducing the risk of overexpression of fasting glucose and lipid accumulation entering the bloodstream in the induction of syndromes of abnormally high blood sugar and cholesterol.

The pervaded catechins found at approximately, 2-15% in the intestinal levels significantly bind to 37% to plasma HDL by inhibiting the key enzymes involved in lipid biosynthesis and reducing intestinal lipid absorption, thus improving blood lipid profile action of cholesterol metabolism.

Further analysis of the unstable condition of polyphenols in the intestinal environment, also found that the transition from the gastric phase to the intestinal phase of green tea extract shows a 50% decrease in the antioxidant activity due to the degradation of polyphenols but the deceased extract still exert a strong effect in the gastrointestinal environment.

However, the intake of whole food milk and cheese in the definition of dairy matrices displayed a significant improvement of polyphenol stability in the intestinal phase and an increase of the antioxidant activity by 29% (cheese) to 42% (milk) compared to nongreen tea injection control.

Additionally, administration of green tea also demonstrated strong activity in decreased expression of the stomach secretes pepsinogen, in breaking down milk and cheese protein and fat intake in degraded into their component polypeptide or amino acid and glycerol and fatty acids, respectively in the gastric phase but had no effect on the intestinal phase, thus reducing glucose and fat being absorbed and presented in the bloodstream.

The evidence showed that green tea polyphenols intake together with dairy products improved antioxidant activity and enhanced the integrity of polyphenols during digestion.

Interestingly, in the evaluated rats fed with selenium-containing green tea (Se-GTE, 1.44 mg selenium/kg dry leaves) and China green tea(CH-GTE, 0.13 mg selenium/kg leaves) and China green tea(CH-GTE) effect in the intestinal micro bacterial activity researchers indicated that oral administration of rat group fed with Se-GTE extract for 6 days exerted a significant increase in caecal counts of lactobacilli and bifidobacteria while significantly reducing the caecal counts of Bacteroides and clostridial bacteria in compared to CH-GTE rat group in slight but not significant increase in the numbers of caecal lactobacilli and bifidobacteria but decreased significantly the numbers of Bacteroides (p < 0.05) and clostridia.

Moreover, rats fed with CH-GTE and Se-GTE showed a 17.2% and 21.3% reduction in the activity of the bacterial enzyme β-glucuronidase, which has been found to contribute to biliary sludge and stone formation and increased risk for colorectal cancer, respectively.

More promisingly, treatment of both CH-GTE and Se-GTE also resulted in 19% and 25.5% increments in the activity of β-glucosidase in reduced beta-D-glucosides and oligosaccharides in released glucose, respectively

Taken together, green tea with abundantly major bioactive polyphenols may be considered a functional food in improved gastrointestinal function but reduced absorption of glucose and lipid in the induction of abnormal high fasting blood sugar and cholesterol levels through binding to fat and protein in the gastric phase of digestion.

Whole food(herbal medicine), linking health benefits in the prevention, management, and treatment of diseases has played an important role in human history over many centuries. The finding of whole food medication by renowned scientists all over the world to replace the single ingredient of Western medicine with little or no side effect has been difficult due to no commercial benefit and patent right to producers.

Garbanzo bean also known as chickpea is an edible legume of genus Cicer and the family Fabaceae, high in protein and minerals. It is one of the earliest cultivated vegetables, native to the Middle East.

Chickpeas may be the next generation of antioxidants in scavenging against the development of free radical H2O2, malondialdehyde (MDA) through the reduction of antioxidant stress.

In the study in the identification and characterization of antioxidant peptides from chickpea protein hydrolysates, researchers at the joint study led by Avda. Padre García insisted that main sequences, ALEPDHR, TETWNPNHPEL, FVPH, and SAEHGSLH, corresponded to legumin, the main seed protein, which has been shown to contain free radical scavenging and cellular antioxidant activities.

In support of the above study, researchers at Jiangnan University said, " This antioxidant peptide(found in chickpea) efficiently quenched the free radical sources 1,1-diphenyl-2-pycryl-hydrazyl (DPPH), hydroxyl, and superoxide free radicals. The Cu(2+) and Fe(2+) chelating activities were 76.92% and 63.08% at the peptide concentration of 50μgmL(-1), respectively. Furthermore, the inhibition of the Fra.7 on lipid peroxidation was greater than that of α-tocopherol".

Also in the investigation of free radicals scavenging activity, pectin (CAP) extracted from the husk of Cicer arietinum L. exhibited a dose-dependent free radical scavenging activity at a scavenging rate of 29% on DPPH radicals in 1.0 mg/mL

Therefore, the intake of. Chickpeas accompanied by a good diet can reduce the risk of free radical-induced certain diseases, such as cancers, arthritis, dementia, etc...

Epidemiological studies suggested that bean fiber may have the potential used as a therapeutically functional food in reducing the risk of hyperglycemia.

Hyperglycemia is a condition characterized by abnormally high blood sugar in the bloodstream.

According to the University of Wisconsin-Madison, dietary fiber intake daily and regularly not only lowers blood cholesterol levels but also normalizes blood glucose and insulin levels, thus reducing the risk and treatment of cardiovascular disease and type 2 diabetes.

Unfortunately, recommended intakes, 20-35 g/day for healthy adults and age plus 5 g/day for children, are not being met, according to the statistic.

In a randomized, single-blind, crossover design study (food intake: n=22; glycemia: n=10) with 5 liquid meals distributing similar amounts of macronutrients (C - control shake, OB - oat bran shake, F - flaxseed shake, WB - white bean extract shake, and UB - unripe banana flour shake) consumed in five non-consecutive days, returned questionnaire from participants after 15, 30, 45, 60, 90 and 120 minutes ingestion of each shake suggested that
1. Compared to the control, intake of white bean extract shake showed a significantly increased fiber component.
2. Bean extract exhibited strong glycemic control through the expression of incremental areas under the curves (iAUC).

Additionally, in the study of children completed a trial of 4 different breakfasts, given in random order, with 3 diets differed in fiber content and the fourth diet contained soya beans as part (38%) of the dietary fiber source were asked to exercise vigorously for an hour each morning and to rest for an hour, with no different mean diet contributed in the morning, after taking account of other risk factors, blood samples drawn from children reveal the following results.

1. There were no differences in blood glucose levels on the high-fiber diet compared to those on the medium-fiber diet
2. Children fed with a bean diet scored a significantly lowest mean blood glucose level and
3. The effect of glycemic control on the blood glucose of the bean diet was last until the hour before lunch.
Even with better control of blood glucose, the bean diet was found to be an unacceptable diet among children, compared to the likes of the high- and medium-fiber diets, which were as popular as the low-fiber diet.

Taking it all together, there is no doubt that bean fibers may have a positive effect on the attenuated risk of abnormally high blood glucose, a major risk factor in an increased risk of type 2 diabetes and cardiovascular disease.
